Aegilops tauschii Coss., the D-genome progenitor of common wheat (Triticum aestivum L.) includes two subspecies, tauschii and strangulata (Eig) Tzvel. Subspecies tauschii has a wide geographic distribution spreading westwards to Turkey and eastwards to Afghanistan and China, while ssp. strangulata has a narrower distribution occurring only in two disjoined regions, southeastern Caspian Iran and Transcaucasia. A collection of 56 Ae. tauschii accessions was screened at adult stage against a mixture of pathotypes of stripe rust prevalent in the current wheat production in China. The results for three crop seasons indicated that among the 38 ssp. tauschii accessions, 37 were susceptible and only one was resistant, while all the 18 ssp. strangulata accessions were resistant. These results indicated that stripe rust resistance was related to taxonomic origin. Further genetic analysis revealed the resistance of stripe rust in ssp. strangulata accession AS2388 was conferred by a single dominant gene.  相似文献

Triticum militinae Zhuk. et Migusch. belongs to timopheevii [Triticum timopheevii (Zhuk.) Zhuk.] group of wheats with 2n = 4x = 28 chromosomes and genome formula AtAtGG. Triticum militinae Zhuk. et Migusch. is known to carry resistance to fungal diseases including rusts and powdery mildew. Genes from timopheevii wheat can be incorporated into cultivated wheat by either direct hybridization or through development of amphiploids. Three T. militinae derived introgression lines (ILs) Triticum Militinae Derivative (TMD) 6-4, TMD7-5 and TMD11-5 were selected for the current study based on cytological stability. All three ILs showed resistance against wide spectrum of Indian pathotypes of leaf rust. More than 1200 SSR markers were used for genotyping of ILs and parental lines. The ILs showed variable and multiple introgressions in different chromosomes of A, B and D genome of wheat. The introgression points were distributed mostly in the distal regions though significant introgressions were also observed in proximal regions of some chromosomes. The extent of introgression in ILs TMD6-4, TMD7-5 and TMD11-5 was 2.8, 8.3 and 8.6% respectively. The set of ‘informative markers’ in the Molecularly Tagged Chromosome Regions (MTCR) of T. militinae origin can also be used in future for tagging of genes associated with traits of economic importance apart from leaf rust resistance. The transferability of Triticum aestivum L. SSR markers to T. militinae was 96.4% for A genome, 95.8% for B genome and 84.3% for D genome. Transferability of wheat SSR markers to T. militinae can be used in preparing genetic maps in timopheevii group of wheats.  相似文献

Seedlings of 41 emmer (Triticum dicoccon Schrank) and 56 durum (T. durum Desf.) wheat accessions were evaluated for their response to stem rust (Puccinia graminis f. sp. tritici) infection under greenhouse condition at Kulumsa Agricultural Research Center, Ethiopia. The objectives were to identify tetraploid wheat accessions that could serve as sources of resistance to stem rust, and postulate the stem rust (Sr) resistance genes through multipatotype testing. The test included screening of accessions for stem rust resistance and multipatotype testing. To ensure vigorous screening, a mixture of six isolates (Si-1a, Am-2, Ku-3, Dz-4a, Ro-4 and Na-22) that were collected from severely infected emmer, durum, and bread wheat (Triticum aestivum L.) varieties of major wheat growing areas of Ethiopia was used as inocula. Out of the tested accessions, 18 emmer and 6 durum accessions exhibited low infection types (0–2) response and hence selected as a source of resistance to stem rust infection. Multipatotype testing was done to postulate Sr genes in the selected accessions. In the test, 10 different stem rust races (A2, A9, A11, A14, A16, A17, B3, B7, B15, and B21), 33 stem rust differential lines, and a universal susceptible check variety, Morocco were used, The high (3–4) and low infection type reaction patterns of the tested accessions and differential lines were used to postulate the genes that exhibit gene-for-gene relationship. The presence of Sr 7b, 8b, 9a, 9b, 10, 14, 24, 27, 28, 29, 30, 31, 32 and Tt-3+10 genes were postulated in 16 selected emmer and 5 durum wheat accessions. Efforts to transfer these valuable Sr genes from cultivated tetraploid wheats could be rewarding to get stem rust resistant varieties and boost wheat production.  相似文献

Despite extensive genetics and breeding research, effective control of leaf rust caused by Puccinia triticina Eriks. and an important foliar disease of wheat, has not been achieved. This is mainly due to the widespread use of race-specific seedling resistance genes, which are rapidly overcome by new virulent races. There is increased emphasis now on the use of race-nonspecific adult plant resistance (APR) genes for durable control of leaf rust. The objective of this study was the evaluation of Aegilops tauschii Coss. (the D-genome donor of bread wheat) for APR, previously known to be a rich source of seedling resistance genes to leaf rust. A geographically diverse collection of A. tauschii maintained by the Wheat Genetics Resource Center was evaluated for APR in the field with a leaf rust composite culture of predominant races. Out of a total of 371 A. tauschii accessions, 50 with low to moderate levels of disease severity were subsequently tested at the seedling stage in the greenhouse with four races and one composite culture of leaf rust. Nine accessions displayed moderate resistance to one or more races of leaf rust at the seedling stage. The remaining 41 seedling-susceptible accessions are potential sources of new APR genes. Accessions from Afghanistan only displayed APR whereas both seedling resistance and APR were common in the Caspian Sea region (Iran and Azerbaijan). The APR in these newly identified A. tauschii accessions will be further characterized for novelty, effectiveness, and race-specificity.  相似文献

Six new amphiploids, Triticum kiharae Dorof. et Migusch. (2n?=?6x?=?42, AtAtGGDD), are described in this study. They were developed by the chromosome doubling of F1 hybrid crosses between Triticum timopheevii Zhuk. (AtAtGG) with high resistance to stripe rust and Aegilops tauschii Cosson (DD) by colchicine treatment. These amphiploids showed a high level of fertility of 68–80% and exhibited relatively normal chromosome pairing in meiotic metaphase I. Individual chromosomes of T. kiharae could be identified by multicolor fluorescence in situ hybridization using the combination of oligonucleotides probes Oligo-pSc119.2-1, Oligo-pTa535-1, and Oligo-pTa71-2. T. kiharae exhibited high resistance to predominant stripe rust races CYR34, CYR31, CYR32, CYR33, and SY11-4 both during the seedling and adult stages. However, high molecular weight glutenin subunits from Ae. tauschii parents were only partially expressed in the T. kiharae background. These T. kiharae lines provide novel materials to widen the genetic diversity of the common wheat gene pool.  相似文献

Field and controlled environmental tests indicated that the 49 accessions of closely related species and 12 landraces of wheat (Triticum aestivum L. em. Thell.) from the National Gene Bank of China showed different reactions to powdery mildew (Blumeria graminis (DC.) E. O. Speer. f. sp. tritici) and stripe rust (Puccinia striiformis Westend f. sp. tritici) at adult and seedling stages. Unknown Pm genes or alleles were postulated with Triticum baeoticum Boiss. accessions BO 3 and Triticum monococcum L. MO 4 and MO 5 when inoculated with 21 powdery mildew isolates at seedling stage. Fourteen accessions of T. baeoticum, T. monococcum, Triticum durum, and wheat landraces were inoculated with 30 stripe rust isolates at seedling stage. Unknown Yr genes or alleles were postulated with T. baeoticum Boiss. accession BO 5, as well as wheat landraces Xiaobaimai, Laomangmai, and Shaanxibai. Heterogeniety in reaction to powdery mildew isolates and stripe rust races were observed in related species and landraces of wheat.  相似文献

Grains of 80 accessions of nine species of wild Triticum and Aegilops along with 15 semi-dwarf cultivars of bread and durum wheat grown over 2 years at Indian Institute of Technology, Roorkee, were analyzed for grain iron and zinc content. The bread and durum cultivars had very low content and little variability for both of these micronutrients. The related non-progenitor wild species with S, U and M genomes showed up to 3–4 folds higher iron and zinc content in their grains as compared to bread and durum wheat. For confirmation, two Ae. kotschyi Boiss. accessions were analyzed after ashing and were found to have more than 30% higher grain ash content than the wheat cultivars containing more than 75% higher iron and 60% higher zinc than that of wheat. There were highly significant differences for iron and zinc contents among various cultivars and wild relatives over both the years with very high broad sense heritability. There was a significantly high positive correlation between flag leaf iron and grain iron (r = 0.82) and flag leaf zinc and grain zinc (r = 0.92) content of the selected donors suggesting that the leaf analysis could be used for early selection for high iron and zinc content. ‘Chinese Spring’ (Ph I ) was used for inducing homoeologous chromosome pairing between Aegilops and wheat genomes and transferring these useful traits from the wild species to the elite wheat cultivars. A majority of the interspecific hybrids had higher leaf iron and zinc content than their wheat parents and equivalent or higher content than their Aegilops parents suggesting that the parental Aegilops donors possess a more efficient system for uptake and translocation of the micronutrients which could ultimately be utilized for wheat grain biofortification. Partially fertile to sterile BC1 derivatives with variable chromosomes of Aegilops species had also higher leaf iron and zinc content confirming the possibility of transfer of required variability. Some of the fertile BC1F3 and BC2F2 derivatives had as high grain ash and grain ash iron and zinc content as that of the donor Aegilops parent. Further work on backcrossing, selfing, selection of fertile derivatives, leaf and grain analyses for iron and zinc for developing biofortified bread and durum wheat cultivars is in progress. Simple sequence repeats (SSRs), highly dispersed nucleotide sequences in genomes, were used for germplasm analysis and estimation of the genetic relationship of the D-genome among 52 accessions of T. aestivum (AABBDD), Ae. tauschii (DtDt), Ae. cylindrica (CCDcDc) and Ae. crassa (MMDcr1Dcr1), collected from 13 different sites in Iran. A set of 21 microsatellite primers, from various locations on the seven D-genome chromosomes, revealed a high level of polymorphism. A total of 273 alleles were detected across all four species and the number of alleles per each microsatellite marker varied from 3 to 27. The highest genetic diversity occurred in Ae. tauschii followed by Ae. crassa, and the genetic distance was the smallest between Ae. tauschii and Ae. cylindrica. Data obtained in this study supports the view that genetic variability in the D-genome of hexaploid wheat is less than in Ae. tauschii. The highest number of unique alleles was observed within Ae. crassa accessions, indicating this species as a great potential source of novel genes for bread wheat improvement. Knowledge of genetic diversity in Aegilops species provides different levels of information which is important in the management of germplasm resources.  相似文献

This study aimed to investigate the effect of inoculation with plant growth-promoting Rhizobium and Pseudomonas species on NaCl-affected maize. Two cultivars of maize (cv. Agaiti 2002 and cv. Av 4001) selected on the basis of their yield potential were grown in pots outdoors under natural conditions during July. Microorganisms were applied at seedling stage and salt stress was induced 21 days after sowing and maintained up to 50% flowering after 120 days of stress. The salt treatment caused a detrimental effect on growth and development of plants. Co-inoculation resulted in some positive adaptative responses of maize plants under salinity. The salt tolerance from inoculation was generally mediated by decreases in electrolyte leakage and in osmotic potential, an increase in osmoregulant (proline) production, maintenance of relative water content of leaves, and selective uptake of K ions. Generally, the microbial strain acted synergistically. However, under unstressed conditions, Rhizobium was more effective than Pseudomonas but under salt stress the favorable effect was observed even if some exceptions were also observed. The maize cv. Agaiti 2002 appeared to be more responsive to inoculation and was relatively less tolerant to salt compared to that of cv. Av 4001.  相似文献

The genus Aegilops L. is a very important genetic resource for the breeding of bread wheat Triticum aestivum. Therefore, an accurate and easy identification of Aegilops species is required. Traditionally, identification of Aegilops species has relied heavily on morphological characters. These characters, however, are either not variable enough among Aegilops species or too plastic to be used for identification at the species level. Molecular markers that are more stable within species, therefore, could be the alternative strategy towards an accurate identification. Since the chloroplast DNA has a lower level of evolution compared to the nuclear genome, an attempt was made in this study to investigate polymorphism in the chloroplast DNA among 21 Aegilops species (including Ae. mutica that is now known as Amblyopyrum muticum) and between the latter and T. aestivum to generate markers for the diagnosis of all targeted species. Cleaved amplified polymorphic sequence (CAPS) applied on 22 coding and non-coding chloroplast regions using 80 endonucleases and sequencing of two of those regions revealed little polymorphism between T. aestivum and the various Aegilops species examined and to a less extent was the variation among Aegilops species. Polymorphism observed among species analysed allowed the discrimination of T. aestivum and 12 Aegilops species.  相似文献

Trigeneric hybrids may help establish evolutionary relationships among different genomes present in the same cellular-genetic background, and also offers the possibility to transfer different alien characters into cultivated wheat. In this study, a new trigeneric hybrid involving species from the Triticum, Psathyrostachys and Secale was synthesized by crossing wheat-P. huashanica amphiploid (PHW-SA) with wheat-S. cereale amphiploid (Zhongsi 828). The crossability of F1 hybrid was high with 35.13%, and the fertility was 41.95%. The morphological characteristics of F1 plants resembled the parent Zhongsi 828. The trigeneric hybrids pollen mother cells (PMCs) regularly revealed averagely 19.88 univalents, 9.63 ring bivalents, 3.97 rod bivalents, 0.60 trivalents and 0.03 tetravalents per cell. Multivalents consisted of trivalents and tetravalents can be observed in 52.7% of cells. A variation of abnormal lagging chromosome, micronuclei and chromosome bridge were formed at anaphase I and telophase II. The mean chromosomes number of F2 progenies was 2n = 46.13, and the distribution range was 42–53. GISH results revealed that most F2 plants had 6–12 S. cereale chromosomes, and only 0–2 P. huashanica chromosomes were detected. The results indicated that S. cereale chromosomes can be preferentially transmitted in the F2 progenies of trigeneric hybrid than P. huashanica chromosomes. A survey of disease resistances revealed that the stripe rust resistance from the PHW-SA were completely expressed in the F1 and some F2 plants. The trigeneric hybrid could be a useful bridge for the transference of P. huashanica and S. cereale chromatins to common wheat.  相似文献

A set of wheat cv. Chinese Spring (CS)-Lophopyrum elongatum addition and substitution lines has been analyzed for its potential to offer new sources of stripe rust resistance for wheat. The adult plants of these lines, together with CS-L. elongatum amphiploid and CS, were inoculated by new physiological races CYR-30 and CYR-31 of stripe rust in China. The resistance investigation indicated that chromosome 7E1 of L. elongatum was responsible for the adult resistance in CS background. Moreover, the expression of the adults plant resistance derived from chromosome 7E1 in wheat background was dependent on the wheat genotype. In addition, the different rust resistance and glutenin composition were observed in 2 CS-L. elongatum amphiploids, indicating the genetic diversity existed in different origins of L. elongatum.  相似文献

Cowpea landraces belonging to Vigna unguiculata (L.) Walp. subsp. unguiculata cv.-gr. unguiculata and cv.-gr. sesquipedalis collected from part of the Deccan Plateau and West Coast of India were evaluated to (i) identify the diverse source(s) of variation for improved characters like pods/peduncle and seed index (ii) study the response of landraces for adaptation to drought and heat stress and (iii) understand the breeding value of a landrace in the genetic improvement of a popular cowpea cultivar. Landraces were evaluated for various morphological characters, pods/peduncle, seed index and other economically important agronomic traits, rust resistance and drought and heat tolerance in different years and environments. Landraces were found as an important source of genetic variability for pods/plant, pods/peduncle, better pod filling ability (seed index), grain yield/plant as well as drought and heat tolerance and rust resistance. Hybridization between C 152 (cv.) and DWDCC 016 (landrace) resulted in release of new variation not present in the two parents. Thus the landrace, DWDCC 016, can be utilized to improve cultivated varieties by transferring to them the economically valuable traits like pods/peduncle and seed index thereby enhancing realisation of sink potential and ultimately grain yield in a sustainable way.  相似文献

A collection of 164 Aegilops tauschii accessions, obtained from Gatersleben, Germany, was screened for reaction to leaf rust under controlled greenhouse conditions. We have also evaluated a selection of synthetic hexaploid wheats, produced by hybridizing Ae. tauschii with tetraploid durum wheats, as well as the first and second generation of hybrids between some of these resistant synthetic hexaploid wheats and susceptible Triticum aestivum cultivars. Eighteen (11%) accessions of Ae. tauschii were resistant to leaf rust among which 1 was immune, 13 were highly resistant and 4 were moderately resistant. Six of the synthetic hexaploid wheats expressed a high level of leaf rust resistance while four exhibited either a reduced or complete susceptibility compared to their corresponding diploid parent. This suppression of resistance at the hexaploid level suggests the presence of suppressor genes in the A and/or B genomes of the T. turgidum parent. Inheritance of leaf rust resistance from the intercrosses with susceptible bread wheats revealed that resistance was dominant over susceptibility. Leaf rust resistance from the three synthetics (syn 101, syn 701 and syn 901) was effectively transmitted as a single dominant gene and one synthetic (syn 301) possessed two different dominant genes for resistance.  相似文献

In a Robinia-pseudoacacia-dominated coastal forest in Tottori prefecture Japan, the growth and survival of Pinus thunbergii seedlings and the natural regeneration of P. thunbergii was disturbed by R. pseudoacacia. In order to improve the growth of P. thunbergii seedling in the Tottori sand dune, we tried to find a mycorrhiza helper bacteria (MHB) from P. thunbergii mycorrhizosphere in a Tottori sand dune. Two MHB, Ralstonia sp. and Bacillus subtilis, were selected from the nine bacterial species isolated from the mycorrhizosphere of P. thunbergii. The bacterial effect on the ectomycorrhizal fungus Suillus granulatus was investigated by confrontation assay and a microcosm experiment. The confrontation assay showed that Ralstonia sp. promoted the hyphal growth of S. granulatus. Moreover, the S. granulatusP. thunbergii symbiosis was significantly stimulated by Ralstonia sp. and B. subtilis. Ralstonia sp. and B. subtilis were regarded as MHB associated with P. thunbergii. This is the first report of Ralstonia sp. as an MHB.  相似文献

The aims of this study are to determine the geographical and ecological distribution of nine Aegilops species in Republic of Armenia and to make an assessment of their IUCN Red List status, using the IUCN Red list categories and criteria, in order to develop an in situ conservation strategy for wild relatives of wheat in Armenia. Ecogeographic surveys of nine Aegilops species were undertaken over 2 years in Armenia. They included a herbarium survey followed by extensive ground-truthing field surveys where targeted Aegilops species occur. The study showed that of the nine Aegilops species studied, four are threatened and of these, Ae. mutica and Ae. crassa are critically endangered. The latter species may even be extinct in Armenia. Ae. neglecta and A. biuncialis are endangered. Additional studies are required to assess the threat status of Ae. umbellulata. Ae. columnaris was assessed as near threatened, while the remaining species (Ae. triuncialis, Ae. cylindrica and Ae. tauschii) are of least concern. There has been a dramatic decline in the genetic resources of Aegilops species during recent years in Armenia as a result of adverse human impacts such as expansion of agriculture, urbanization and uncontrolled grazing. Several species, especially Ae. mutica and Ae. crassa, should be prioritized in conservation activities in Armenia. Efforts should be made to conserve genetic diversity of crop wild relative species both in situ and ex situ, bearing in mind that their germplasm carries potentially valuable information (traits) that can improve adaptability and productivity of cultivated wheat varieties.  相似文献

Emmer wheat (Triticum dicoccon Schrank, 2n = 4x = 28) consists in a hulled wheat; its cultivation has been drastically reduced during the last century as a consequence of its low yield. Recently, its agronomic and nutritive values, as well as the increase of popularity of organic agriculture, have led to a renewed interest making its cultivation economically viable in the marginal lands with an increase of the cultivated areas. In Italy, it mainly survives in few marginal lands of central and southern Italy, where local varieties, adapted to the natural environment from where they originate, are used; moreover, some selected lines have also been developed. In the present work, agro-morphological and qualitative traits, together with molecular analyses of 20 emmer accessions consisting of Italian landraces, breeding lines, and cultivars, were performed. The field experiments were conducted for two consecutive years (2001/2002–2002/2003) in two locations: Viterbo in central Italy, and Foggia in south Italy. The analyzed emmer wheat accessions showed a good amount of genetic variability for both evaluated agro-morphological and molecular traits. This study illustrates an increase in earliness, GY, TW, TKW, and YI going from landraces, breeding lines to cultivars, while the variability does not change proportionally.  相似文献

Stripe rust is one of the most serious diseases of wheat in China. Two new stripe rust physiological strains CYR30 (Inter. name: 175 E 191) and CYR31 (Inter. name: 239 E 175) have become the dominant and epidemic physiological strains since 1994 in China. Resistance to these strains of 48 Ae. tauschii accessions was evaluated at the seedling and adult stages. Out of 48, 28 accessions displayed seedling and adult resistance. All of the Chinese Ae. tauschii accessions were susceptible at seedling and adult stages. Seedling resistance was highly related to adult resistance in Ae. tauschii. The new genetic resources of resistance to CYR30 and CYR31 could be incorporated into commercial wheat varieties for wheat resistance breeding by direct hybridization.  相似文献

The genus Arachis is divided into nine taxonomic sections. Section Arachis is composed of annual and perennial species, while section Heteranthae has only annual species. The objective of this study was to investigate the genetic relationships among 15 Brazilian annual accessions from Arachis and Heteranthae using RAPD markers. Twenty-seven primers were tested, of which nine produced unique fingerprintings for all the accessions studied. A total of 88 polymorphic fragments were scored and the number of fragments per primer varied from 6 to 17 with a mean of 9.8. Two specific markers were identified for species with 2n = 18 chromosomes. The phenogram derived from the RAPD data corroborated the morphological classification. The bootstrap analysis divided the genotypes into two significant clusters. The first cluster contained all the section Arachis species, and the accessions within it were grouped based upon the presence or absence of the ‘A’ pair and the number of chromosomes. The second cluster grouped all accessions belonging to section Heteranthae.  相似文献

Leaf rust caused by Puccinia recondita f. sp. tritici belongs to the most important diseases of wheat worldwide. The progress of the disease is highly dependent on meteorological conditions. One of the parameters affecting the disease progress is latency period driven mostly by temperature. Examination of the influence of predicted temperature changes on the monthly sum of the daily latency period expressed as rate (MSLPR) in western Poland was the aim of our study. Experiments in quasi-natural conditions were performed with the aim of validating models describing the influence of temperature on leaf rust latency period in two wheat varieties, Turnia and Ostroga. Both models used for assessment of latency period duration on cv. Turnia and cv. Ostroga performed well. The influence of climate changes on leaf rust latency period was then simulated using meteorological data from three weather stations located in western Poland and virtual data obtained after transformation of the recorded data to reflect a temperature changes under RCP2.6 and RCP8.5 scenarios according to giss_e2_r climate model. The study shows that without mitigation of greenhouse gas emission latency period of wheat leaf rust is going to decrease which may result in increase of wheat leaf rust incidence.  相似文献
