Distribution pattern of fibre types was studied in the muscles of the soft palate (palatinus, levator veli palatini and tensor veli palatini muscles) in the dog. The fibrillar classification was based on using histochemistry and immunohistochemistry methods: myofibrillar adenosine thriphosphatase (mATPase) to different pH of pre‐incubation; nicotine adenine dinucleotide (reduced) tetrazolium reductase (NADH‐TR) and finally, application of specific monoclonal antibodies against myosin heavy chain isoforms I, IIa and IIx. In the palatinus and levator veli palatini muscles, pure type I fibres and the hybrid type IIax and IIc were shown, with a checkerboard distribution in the first and a clear predominance of hybrid fibre types (about 98% of the total population) in levator veli palatini muscle. On the other hand, in the tensor veli palatini muscle, type IIx and IIm fibres were identified (fast‐twitch fibres related to fast‐moving muscles and the powerful jaw muscles of carnivores). The tensor veli palatini muscle had a different distribution and fibrillar composition with predominantly type IIm fibres in its central zone, whilst the peripheral zone was primarily type I and IIx fibres.  相似文献

In this study, we compare the effects of pre‐ and post‐natal food deprivation on the relative proportion of fibre types and contractile responses in the extensor digitorum longus (EDL) muscle of female and male rats at different post‐natal ages. EDL muscles from undernourished male (UM) rats showed a higher proportion of Type IIB than IIA fibres and larger normalized twitch responses (with respect to muscle weight) than those of controls (CM). In contrast, EDL muscles from control (CF) and undernourished female rats (UF) showed no significant differences in their fibre type composition and normalized twitch forces at most of the ages analysed. Our data are indicative that the EDL muscles from undernourished males are more susceptible to the effects exerted by low food income than the EDL muscles from female rats. It is proposed that changes in the reactive oxygen species (ROS) concentration and hormonal factors, due to undernutrition, are involved in the alterations observed in the fibre type composition and force production of EDL muscles in undernourished male rats and that estrogens may have an antioxidant protective role on the undernourished EDL muscles in female rats.  相似文献

Protein profiles of whole homogenates of anconeus (slow twitch) and biceps femoris (fast twitch) muscles of clinically normal dogs and of Labrador Retrievers with hereditary myopathy (HM) were resolved on flat bed polyacrylamide isoelectric-focusing gels. Three methods of sample solubilization were performed. The solubilization buffer, with high concentrations of urea, precipitated the zwitterionic detergent, but use of the buffer containing 3% NP-40, 9.2M urea, and 0.1M arginine resulted in better resolution and stability of pH gradient. Gels of anconeus muscle from clinically normal dogs contained 2 protein bands specific to anconeus muscle, whereas gels of biceps femoris muscle from clinically normal dogs contained 3 protein bands amplified in biceps femoris muscle that were barely detectable in anconeus muscle. The staining intensity of protein bands in biceps femoris muscles from Labrador Retrievers with HM was decreased, relative to controls. The quantitative analysis of peak height ratios of biceps femoris muscle revealed significant (P less than 0.05) differences between profiles of clinically normal dogs and Labrador Retrievers with HM.  相似文献

In this study, the effect of testosterone on gastrocnemius muscle fibres in growing and adult rats (male and female) was examined using histochemical, morphometric and ultrastructural techniques. After physiological saline (PS), olive oil (OvO) or olive oil + testosterone (OvOT) injections on 72 rats (growing and mature, 36 male and 36 female), the sample tissues of fibre types of the gastrocnemius muscle taken were examined by histochemical [alkaline adenosine triphosphatase (alk-ATPase), acid ATPase (ac-ATPase)], morphometric and ultrastructural techniques. In PS-injected control groups, the gastrocnemius muscle of both sexes contained all the fibre types studied [slow-oxidative muscle fibres (type I), fast-oxidative glycolytic muscle fibres (type IIA) and fast-glycolytic muscle fibres (type IIB)]. The type I fibres had the smallest diameter, type IIA had a medium diameter and type IIB fibres had the largest diameter. In OvO-injected groups, it was observed that the OvO had little effect on the gastrocnemius muscles of either sex, although there was significant enlargement of type IIB fibres. After the injection of OvOT, hypertrophy of muscle fibres was determined by morphometric study. The biggest increase in diameter was on type I fibres. In addition, degenerations on some mitochondria, accumulation of lipid droplets on type I and type II fibres, an increase in glycogen particles, bifurcation of myofibrils, an increase in the number and diameter of units resembling T tubules and an increase in ribosomal content were also observed in the same group by transmission electron microscope. Consequently, it was determined that testosterone can induce protein synthesis in gastrocnemius muscle fibres, and induces changes in shape and size, and also can change the appearance and the number of fibres.  相似文献

Muscle fibre recruitment was investigated during draught loaded exercise by studying glycogen depletion patterns from histochemical stains of muscle biopsies from the gluteus and semitendinosus muscles. Three Standardbred trotters performed several intervals of draught loaded exercise on a treadmill with 34 kp at a trot (7 m/sec) and with 34 and 80 kp, respectively at a walk (2m/sec). Exercise was continued until the horses were unwilling to continue. Glycogen depletion was seen in all three fibre types when trotting with 34 kp for 5 or 10 mins. When an equal weight resistance was pulled at a walk, glycogen depletion was first seen in type I fibres only, then followed by a small percentage of type IIA fibres after at least 1 h. When 80 kp was pulled at a walk both type I and IIA fibres showed glycogen depletion, and after at least 30 mins exercise a small percentage of type IIB fibres was also depleted. These results indicate that the muscle fibres are depleted, in order, from type I through IIA to IIB as the intensity or duration of draught work increases.  相似文献

This study was designed to determine the histochemical properties, size and composition of fibres in the diaphragm, intercostal and abdominal muscles of goats to clarify whether reported similarities in respiratory muscle physiology between goats and humans have a structural basis. Serial sections (10 µm) of muscular tissue from adult female goats were stained for myosin adenosine triphosphatase and reduced nicotinamide adenine dinucleotide dehydrogenase-tetrazolium reductase activities; the fibres were classified into types I, IIA and IIB; and their mean diameter and composition were determined. Abdominal and intercostal muscles contained types I, IIA and IIB fibres in the ratio 1:1:1, and the mean diameter of the fibres ranged from 49.2 to 62.2 µm. In contrast, the diaphragm contained 58.9% type I and 41.1% type II fibres, and the latter could not be differentiated into types IIA and IIB. Diaphragmatic fibres were also smaller (36.9 – 40.9 µm). These findings contrast with those in humans, where the diaphragm, intercostal and abdominal muscles contain >50% type I fibres and have fibres of identical diameter. The differences in fibre characteristics between the diaphragm, intercostal and abdominal muscles of goats and the differences between goats and humans need to be taken into consideration in interpreting the results from studies in respiratory muscle physiology.Abbreviations mATPase myosin adenosine triphosphatase - NADH-TR nicotinamide adenine dinucleotide dehydrogenase-tetrazolium reductase  相似文献

The distribution of muscle fiber types in rostral and caudal portions of the musculus digastricus (digastric muscle) was studied in 6 dogs. Staining procedures which stain specifically for type IIM fibers, a fiber type found in other muscles supplied by the trigeminal nerve, were used. Rostral and caudal portions of the muscle were compared because the rostral portion is innervated by the trigeminal nerve, and the caudal portion is innervated by the facial nerve. The musculus triceps brachii (triceps muscle), which contains fiber types I and IIA, and the musculus masseter (masseter muscle), which contains type IIM, were used as controls. Mean fiber diameters were calculated for each of the muscles. Both portions of the digastric muscle exhibited the same histochemical behavior, possessing types I and IIA myofibers. Neither portion contained type IIM fibers. Type I fibers in the masseter muscle were histochemically different from type I fibers in the other muscles studied. Type II fibers predominated in all 3 muscles, but there were significantly (P less than 0.001) more type I fibers in the triceps muscle than in either portion of the digastric muscle or in the masseter muscle. Type II fibers were significantly larger than type I fibers in the caudal digastric (P less than 0.01) and masseter (P less than 0.05) muscles. The middle gluteal muscle of five, two-year-old untrained trotters was investigated by repeated needle biopsy sampling over a training period of six months. A second group of five, three-year-old untrained horses was included to examine the effect of growth. After the training period increases were found in the relative distribution of slow twitch (ST) fibres from 18 per cent to 25 per cent and fast twitch (FTa) fibres from 36 per cent to 45 per cent, and a decrease in FTb fibres from 46 per cent to 30 per cent. A proportionally equal reduction (approximately 18 per cent) in the cross sectional area of all fibre types was observed after the first two months of training succeeded by an increase to approximately pretraining levels at the end of the period. The number of capillaries per fibre was enhanced from 1.7 to 2.4. Proliferation of capillaries occurred around fibres of all types. The metabolic adaptations showed increases in the activities of 3-hydroxy-acyl-CoA dehydrogenase (HAD) (50 per cent) and citrate synthase (31 per cent). Growth had no effect on the relative fibre type distribution nor the capillary per fibre ratio, but as the mean fibre area increased 36 per cent (primarily because of increases in FT fibres) the number of capillaries/mm2 was lower in the older untrained horses (350 capillaries/mm2) compared with the younger untrained ones (460 capillaries/mm2). Increase with growth was found in the activity of phosphorylase and HAD and a decrease was seen in the activity of hexokinase. It is concluded that the training programme exclusively induced alterations which improved the aerobic capacity of the muscle.  相似文献

Twenty-four Bouviers with dysphagia were examined between October 1986 and October 1988. The type of dysphagia was characterised by the results from the clinical examination, the videofluorographic examination and the electromyographic recordings from the oral, pharyngeal, and esophageal muscles. Electromyography indicated neurogenic as well as myogenic causes of dysphagia. Tissues from 10 dogs were available for histopathologic examination. In nine dogs there was a progressive muscular degeneration of the pharyngeal and/or esophageal muscles, resembling muscular dystrophy. In two of these dogs the same abnormalities were also noticed in the masseter and temporalis muscles and in the intrinsic laryngeal muscles. In one dog small areas with hyalin degeneration and fragmentation of muscle fibres were found in the cricopharyngeal muscle. No abnormalities in nerve tissue were found. Muscular dystrophy is a hereditary disease. The mode of transmission in these Bouviers is not yet known.  相似文献

The compositional characteristics of the three basic types of myofiber, namely type I (slow‐twitch oxidative), type IIA (fast‐twitch oxidative glycolytic) and type IIB (fast‐twitch glycolytic), are clarified in the skeletal muscles of Japanese Black cattle. The myofiber composition, which is characteristic of the muscles of Japanese Black cattle, markedly changes during their growth, when some type IIA myofibers are transformed into type I or IIB, depending on the different muscles. Independent of these changes with growth, inter‐ and intramuscular variations of myofiber type distribution is evident. The small extensor muscles in deep regions around bone contain a lot of type I myofibers, whereas the large muscles at surface regions have many type II myofibers. Japanese Black cattle have typical white muscles such as the Longissimus thoracis and Semitendinosus, containing half the myofibers as red (type I + IIA). The muscles of Japanese Black cattle show a tendency to contain a higher percentage of type I myofibers than other breeds over an intrabreed variation of the myofiber type composition. In the big muscles such as the Longissimus thoracis and Biceps femoris, a great diversity of myofiber type composition is observed among the different regions. When fattened, heifers produce Longissimus thoracis and Biceps femoris muscles of smaller weight than steers, but in heifers the myofiber size in each type is rather larger. In the Psoas major, Vastus lateralis and Serratus ventralis muscles, heifers contain a higher frequency of red (type I + IIA) myofibers with no differences in myofiber size. Among the several muscles of fattened Japanese Black steers, the percentage distribution of type I myofibers has a positive correlation with the percentage amount of intramuscular fat. From these results, the high potential of Japanese Black cattle to produce marbled beef could be based on the histochemical properties of myofibers in their skeletal muscles.  相似文献

This study characterised muscle fibres in trunk, forelimb and hindlimb muscles of three bat species: little Japanese horseshoe (Rhinolophus cornutus), greater horseshoe (Rhinolophus ferrumequinum) and Egyptian fruit (Rousettus aegyptiacus). Twenty-seven muscles from trunk, forelimb and hindlimb were dissected, weighed and analysed by immunohistochemistry and sodium didecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and determined their cross-sectional areas (CSA). Results showed that Type IIa and Type IIa/x made the highest proportion of total muscle mass. Moderate proportion was formed by Type IIb. Type I and IIx appeared at very low levels in all bats. Type IIb was the only fibre type detected in patagial muscles in wing membrane of greater horseshoe while other fibre types were not observed. Type I muscle fibres were very few and appeared infrequently in fifteen muscles of Egyptian fruit and in only one muscle in each, greater horseshoe and little Japanese horseshoe. Type IIx was also detected in three muscles in greater horseshoe and only one muscle in Egyptian fruit but none in little Japanese horseshoe. The highest average CSA μm2 was detected in Type IIb and values were 734.2μm2 for LHB; 1537.9μm2 for GHB and 1,720.9μm2 for EFB. Lowest and undetermined values were observed for Type I and IIx. These data demonstrate that Type IIa, IIa/x and IIb form significant proportion of adult bat muscle mass and Type IIb is the largest fibre type. The distribution pattern is suggestive of specialised functions of the fibres in relation to orientation and speed of bats during flight.  相似文献

The location of carbonic anhydrase III (CA-III) in frozen sections of biopsies of Thoroughbred horse skeletal muscle was studied. Fibre types were determined by ATP-ase and succinate dehydrogenase staining. CA-III isozyme was detected using a peroxidase conjugated anti-CA-III antibody. CA-III was found to be localised in slow twitch oxidative fibres (ST), but was also present in fast twitch oxidative (FTH) fibres in small amounts. Fast twitch glycolytic (FT) fibres were stained lightly compared with control sections. The concentrations of CA-III in muscle and liver were 70 micrograms/mg protein and 4 micrograms/mg protein, respectively. CA-I and CA-II were not found in muscle extracts by the double immunodiffusion method.  相似文献

In order to obtain broader insights into the equine musculoskeletal system, we studied the fibre type composition of 2 locomotory muscles in biopsies from Dutch Warmblood foals taken at 3 different ages in the first year postpartum. The muscle fibre types were determined histochemically as well as immunohistochemically. ATPase-characterised IIB fibres appear to express either IId or type lIa plus IId myosin heavy chain (MHC). A high percentage of fibres classified as IIA with ATPase expressed both fast types of MHC. The type I classification by the 2 methods matched almost completely. There was an increase with age of fibres expressing I and IIa MHC in the gluteus medius. At the same time, there was a decrease of fibres expressing IId MHC and fibres co-expressing MHC IIa and IId. MHC expression of the semitendinosus muscle did not change over time at first, but from age 22-48 weeks there was a decrease in the percentage of type IId fibres. In general, the gluteus medius contained more type I fibres but fewer type IId fibres compared to the semitendinosus. At most ages the fibre type compositions of both muscles correlated with one another. To examine the effect of exercise, one-third of the foals were given box rest, one-third received training and one-third kept at pasture during the first 22 weeks of life. The 3 exercise groups differed in their fibre type composition; however, these differences could not be attributed to the effect of exercise.  相似文献

The microstructure of two type of muscles was studied in a selection experiment conducted with Dutch Large White pigs (boars and gilts) selected for either low backfat thickness (L-line) or fast growth (F-line). Second- and fourth-generation pigs were used to determine effects of selection on fiber type composition, fiber area, and capillary density in the longissimus lumborum (LL) and biceps femoris (BF) muscles. Immediately after slaughter samples were taken from the LL and BF muscles. The latter was divided into an inside (BFi) and outside (BFo) portion, which refer to the red and white portions of the biceps femoris. Serial sections were stained for ATPase (pH 4.60), succinate dehydrogenase (SDH), and alpha-amylase-periodic acid shiff (PAS) to determine fiber type and capillary density. The LL and BFo muscles had predominantly type IIBw fibers, whereas the BFi muscle had a 2 to 4 times higher amount of type I fibers. In most muscles there were more type I and fewer type IIBw fibers in F- than in L-line pigs (P < .05), except in the LL muscle of second-generation pigs and in the red part of the BF muscle of fourth-generation pigs. In both selection lines lower type I and higher type IIBw percentages were found in muscles from gilts than in those from boars (P < .05). Capillary density and fiber area of L- and F-lines showed minor differences, which could be explained by differences in weight and age of the pigs of both lines. The results suggest that selection for low backfat thickness in pig breeding compared with increased growth rate resulted in fewer oxidative and more glycolytic muscle fibers. The magnitude of the effect depended on muscle type and duration of the selection period.  相似文献

The effect was investigated of the Ralgro preparation with the active substance zeranol on histological and histochemical properties of bull muscles. The anabolic effect was displayed by a trend toward greater thickness of muscle fibres in m. longissimus thoracis, m. semitendinosus and m. triceps brachii, whilst differences between the muscles of experimental and control animals were not statistically significant. The bulls administered zeranol had the higher proportions as well as the higher relative volumes of white (aW) muscle fibres, but neither were these differences statistically significant in comparison with the control. The growth and development of testicles are inhibited by zeranol. The inhibition is significant and persists during 30 days after the last administration. Later on, the rate of development and growth are increased with the testicles reaching the weight of the sexual glands of control animals in 90 days after the last administration; the coiled seminiferous tubules grow and spermiogenesis occurs.  相似文献

Following a study of fibre population fibre size was determined in three muscles of different functional purpose in four breeds of dog with different locomotory capacities. Samples were taken from specific sites in the tibialis cranialis (extensor), flexor digitalis medialis (flexor) and pectineus (adductor) muscles of 27 dogs of four different breeds—German shepherd, Spanish greyhound, Spanish mastiff and Iberian hound—. Samples were subjected to histochemical analysis using ATPase (pH 9.4) and fibre size was assessed using an IBAS-2 Kontron image analyser.  相似文献

Golden Retriever (GR) muscular dystrophy is an inherited degenerative muscle disease that provides an excellent model for Duchenne muscular dystrophy in humans. This study defined the histopathologic lesions, including the distribution of type I and II muscle fibers (FTI and FTII), in 12 dystrophic and 3 nondystrophic dogs between 7 and 15 months of age. The authors were interested in studying the influence on disease phenotype from crossing the base GR breed with Yellow Labrador Retrievers. The dystrophic dogs were divided according to breed: GRs and Golden Labrador Retrievers (GLRs). On hematoxylin and eosin staining, histopathologic lesions were more severe in GRs than GLRs. Six of eight GR muscles (75%) had a severe lesion grade (grade 3). In contrast, seven GLR muscles (87.5%) had mild lesions (grade 2), and only one had severe lesions (grade 3). Changes in fiber-type distribution were more pronounced in GRs versus GLRs. FTI:FTII ratio inversion was observed in three dystrophic GRs but only one GLR. The mean diameter of FTI and FTII was smaller in GRs and GLRs than in nondystrophic dogs (P < .01). The FTI of five GR muscles (62.5%) were larger than those of GLRs, whereas only one GLR muscle was larger (P < .05). The differential was less pronounced for FTII, with four GR muscles being larger and three GLR being larger. Observations indicate that crossing the base GR breed with Labrador Retrievers lessened the severity of the GR muscular dystrophy phenotype.  相似文献

1. Developmental states of the collagen content, distribution and architecture in the pectoralis (PT), iliotibialis lateralis (ITL) and puboischiofemoralis (PIF) muscles of male Red Cornish x New Hampshire (RN, 80 d, body weight 2.9 kg) and normal (3.1 kg) broilers were evaluated. 2. In PT muscle the total amount of collagen was significantly greater in RN broilers (3.33 mg/g) than in normal ones (1.71 mg/g). This higher collagen content in RN broilers was based mainly on the closer mesh sizes of endomysial honeycomb. The collagen structures in the perimysia also differed between broiler types, when more collagen fibres were observed in RN broilers. 3. ITL muscle contained total collagen of 4.10 to 5.00 mg/g. Types I and III collagens were distributed on the perimysia at higher percentages in RN broilers (31.6%, 37.2%) than normal (15.6%, 30.8%), respectively. The thick bands of tough collagen fibres characteristic of ITL muscle perimysium in cockerels had not yet developed in these broilers. 4. Total collagen was 4.63 to 6.29 mg/g in PIF material with fascia. In PIF muscle the perimysial collagen fibres had not yet attained their full growth but consisted of densely packed fibrils. PIF muscle was characterised by the earlier maturing collagen structure. 5. These results show that a perimysial collagen structure in broilers is still in an undeveloped state. It is supposed that tenderness of broiler meat is attributed mainly to characteristics of the collagen distribution, in which the majority of types I and III collagens is distributed on the closer mesh of endomysial honeycomb.  相似文献

The purpose of this study was to determine whether there were differences in skeletal muscle properties in the hindlimb muscles of different types of dogs. Muscle samples were obtained from the gracilis, sartorius cranial head, sartorius caudal head and tibialis anterior muscles of mixed-breed and hound-type dogs and Beagles. Fiber type, fiber size and capillary morphometry determinations of each muscle from each dog were made from sections stained for myofibrillar ATPase activity. Individual animals were bilaterally symmetric for all measured variables. Fiber type, fiber size and capillary geometry varied between dogs of a given type and muscles within a given dog. There were no differences between dog types for fiber type or fiber size; significant variation in log(muscle)/log(body) mass ratios between dog types was observed for all muscles. The results indicate that for a given muscle, significant variation can occur in skeletal muscle characteristics between different types of dogs and that these differences can be independent of differences in exercise history.  相似文献

1. Histochemical properties of M. pubo‐ischio‐femoralis pars medialis (PIF muscle) were compared in 7 breeds of cocks. This muscle was largely composed of Type I fibres and their transitional form (Type I tr). Type IIA fibres were observed in the cranial part.

2. The weight and cross‐sectional area of the PIF muscle increased with increasing body weight. However, the relative muscle development to body weight differed among the 7 breeds.

3. A quarter of Type I fibres were of the transitional variety in bantam fowls. Conversely, few, if any, Type I tr fibres were observed in the large breeds where the muscle was poorly developed.

4. As the histochemical properties of Type I fibres made an effective response to the different body weights and the relative PIF muscle development among breeds, it was concluded that PIF muscle performed an important function in supporting the body weight and maintaining posture.  相似文献
