Infection with potato virus S Andean (PVSA) and ordinary (PVSo) strains was found in potato breeder's selection No. 8163-511 imported from West Germany; the two PVS strains were differentiated by their reactions on Chenopodium quinoa Tests on potato leaf samples using enzyme-linked immunosorbent assay followed by inoculation to C quinoa were subsequently used to detect PVSA and PVSo in a large-scale survey of imported and domestic potato material. PVSA was detected in breeders' selections and cultivars imported from the Netherlands and West Germany, but not in domestic certified seed potato stocks or farmers' once-grown stocks. PVSo was found in both imported and domestic certified stocks, but infection was commoner in the imported ones. When plants of C. quinoa, C. amaranticolor, C. murale and Nicotiana debneyi were inoculated with four isolates of PVSA, one induced mild symptoms while the reactions of the others ranged from moderate to severe. When plants of different potato cultivars were inoculated with three isolates, the plants were mostly infected without symptoms. However, when tubers from some were grown on, the progeny plants of most of the different combinations of cultivar and isolate of PVSA developed one or more of the following symptoms: vein deepening, rugosity, interveinal chlorosis, premature senescence and early loss of lower leaves.  相似文献

苹果褪绿叶斑病毒生物学及生化特性研究   总被引:12,自引:1,他引:11  
 对从苹果和扁桃上获得2个苹果褪绿叶斑病毒的分离物ACLSV-C和ACLSV-B的主要生物学和生化特性进行了比较。人工接种5科19种草本植物,发现两者均能侵染苋色藜(Chenopodium amaranticolor)、昆诺藜(Ch.quinoa)和西方烟(Nicotiana occidentalis),产生局部侵染斑和系统褪绿斑。但症状反应存在差异,后者在这3种植物上引起叶片反卷等较强症状反应,还可潜伏侵染笋瓜(Cucurbita maxima cv.Buttercup Burgess)。经SDS-聚丙烯酰胺凝胶电泳结果显示,ACLSV-B衣壳蛋白的迁移率较ACLSV-C快。两者的RNA分子量及双链RNA数量无明显差异。根据已报道的核苷酸系列设计合成引物,采用PCR法检测ACLSV分离物,均获得特异性扩增产物。  相似文献

 从表现花叶症状的德国进口番红花上获得一线状病毒分离物SA,线状病毒粒子长度为700~800nm。其在人工接种的11科39种植物中能侵染6科14种植物,在克里芙兰烟上产生系统坏死,在小白菜、花椰菜等十字花科植物上产生系统花叶或为隐症,在昆诺藜等藜科植物上为局部侵染。在番红花病叶、球茎及克里芙兰烟病叶组织中观察到卷轴状和片层状聚集的风轮状内含体。免疫吸附和免疫修饰电镜观察结果显示,SA与芜菁花叶病毒((TuMV)具有紧密的血清学关系。根据这些特征将SA鉴定为TuMV的一个分离物。这是TuMV侵染番红花的首次报道。  相似文献

According to EC regulations, imported material of tuber-forming Solarium spp. has to be tested for the absence of defined quarantine viruses. In order to allow an efficient and timesaving application of the quarantine inspection procedure, antisera have been developed and scrutinized for their use by the ELISA technique. The viruses concerned were Andean potato latent virus (APLV), Andean potato mottle virus (APMV), arracacha virus B oca strain (AVB-O), potato virus T (PVT) and tobacco ringspot virus Andean potato calico strain (TRSV-Ca). The results show that all viruses can be reliably detected by double-antibody sandwich (DAS) ELISA. The detection limits were in the range ≤ 1–32 ng ml−1. The strain specificity by DAS-ELISA of the antisera for APLV and APMV was overcome by mixing antisera from the different strains and strain groups, respectively. Strains C and Lm of APMV seemed to be serologically identical. A comparison of the suceptibility of several wild species of tuber-forming Solanum with that of several cultivars of Solanum tuberosum showed a higher frequency of infection in the wild species.  相似文献

应用酶联免疫吸附法(ELISA)检测了浙江丽水和杭州的东方百合、亚洲百合等栽培品种上黄瓜花叶病毒(CMV),64个田间样品中有26个带毒,检出率为40.6%;dsRNA检测结果与已知CMV-FQS株系的电泳条带相同,但百合组织中CMVdsRNA含量较低;电镜观察,受CMV侵染的样品中大多含有线状病毒,复合侵染率为35.9%;寄主反应测定显示从百合植株上获得的CMV株系均不能通过汁液摩擦接种侵染昆诺藜、苋色藜、普通烟、心叶烟等6科10种指示植物。  相似文献

ABSTRACT Immuno-capillary zone electrophoresis (I-CZE) is a technique that combines the specificity afforded by serological assays with the sensitivity, rapidity, and automation in detection provided by capillary zone electrophoresis. Cymbidium mosaic potexvirus (CymMV) and odontoglossum ringspot tobamovirus (ORSV) were detected in their purified forms as well as in the crude saps of infected Nicotiana benthamiana leaves and Oncidium orchid flowers. The two orchid virus-antibody complexes were resolved via the combined actions of electrophoretic migration and electro-osmotic flow along a buffer-filled, uncoated fused-silica capillary. The I-CZE fractions collected from both CymMV- and ORSV-antibody complex peaks, as well as the RNA purified from them, retained their infectivity upon inoculation onto Chenopodium quinoa. I-CZE assays were able to detect as little as 10 fg each of both CymMV and ORSV in their purified forms as well as in the crude saps of infected N. benthamiana and Oncidium orchid. As multiple samples can be analyzed rapidly, I-CZE offers an ideal diagnostic technique for routine mass-indexing programs such as virus-free certification, breeding for virus-resistant cultivars, plant quarantine, and germ plasm screening. This is the first report of the application of I-CZE for the detection of plant viruses.  相似文献

 电镜观察了葡萄扇叶病毒杭州分离物(Grapevine fanleaf virus Hangzhou isolate,GFLV-H)侵染苋色藜(Chenopodium amaranticolor)和昆诺藜(C.quinoa)的细胞超微结构变化。病毒粒子存在于叶肉组织薄壁细胞的细胞质内,常成单纵列排列在小管结构中,小管形成聚集体。在系统感染的昆诺藜细胞中位于管状结构中的病毒粒子穿过胞间连丝。2种感病寄主细胞质内膜结构增生,在核周围形成含细纤维状物质的小囊泡。液泡边缘存在少许小泡,有多泡体和髓鞘样结构伸入液泡,叶绿体和线粒体等细胞器也发生不同程度的变化。  相似文献

 以葡萄扇叶病毒(GFV)干u马铃薯Y病毒(PVY)外壳蛋白基因的重组质粒为模板,用聚合酶链式反膨技术(PCR)分别合成了长度为1.5kb和0.75kb的生物素标记双链cDNA探针,在斑点杂交反应中,探针的最适使用浓度为1/100,GFV探针检测GFV-RNA的灵敏度为10pg,检测提纯GFV的灵敏度为25pg,检测感病苋色藜的最大稀释倍数为10000倍;PVY探针检测PVY-RNA的灵敏度为30pg,检测提纯PVY灵敏度为500pg,感病烟草检测的最大稀释度为8000倍,阴性对照均无杂交信号出现。  相似文献

Mixed instars of Thrips tabaci were allowed to feed on test seedlings of cucumber, the thrips were removed and wounds caused by thrips feeding dusted with tobacco streak virus (TSV) carried in tomato pollen. Transmission of TSV occurred in three out of the four experiments conducted. When Chenopodium amaranticolor test seedlings were dusted with infective tomato pollen, and thrips introduced for varying periods before being killed with insecticide, TSV transmission occurred after exposure to thrips for 1 h, 5 h, 1 day or 2 days, but not 5 min. Pollen-washing experiments indicated that TSV was carried both internally and externally in pollen of tomato and several weed hosts. T. tabaci was able to transmit TSV from pollen of Nicotiana clevelandii and Nicandra physalodes , and TSV from tomato pollen was transmitted to several weed species. It is concluded that T. tabaci transmits TSV associated with tomato pollen by a mechanical mechanism whereby virus carried externally, or released from inside the pollen, infects feeding wounds.  相似文献

 本文从寄主范旧,蚜传特性、病毒颗粒形态、免疫电镜等方面对嫁接获得的甘薯羽状斑驳病毒廊坊分离物(SPFMV-LF)进行了鉴定,并和SPFMV-RC(美国株)进行了实验比较。SPFMV-LF易汁液摩擦传播和嫁接传播,并以蚜虫非持久性传播,SPFMV-LF系统侵染巴西牵牛(Ipomoea setosa)、牵牛(I.nil);不侵染Chenopodium quinoa、C.ama-ranticolor、Nicotiana benthamiana及其它烟草。尚未发现其局部斑寄主。SPFMV-LF及SPFMV-RC感染I.setosa后引起相似的症状,但感染I.nil后症状差别较大。病毒颗粒长860-830nm,经两次蔗糖垫超速离心从SPFMV-LF感染的I.setosa叶片提取病毒并在BALB/C小鼠中制备抗SPFMV-LF的腹水多克隆抗体,免疫电镜表明,SPFMV-LF的抗体和SPFMV-RC株系有反应。上述实验结果表明SPFMV-LE是SPFMV的一个株系,但不同于SPFMV-RC。  相似文献

A potyvirus (eggplant mottle virus, EMoV) causing mosaic mottling in eggplant ( Solanum melongena ) was characterized on the basis of biological, serological and partial nucleotide sequence properties. EMoV infected Chenopodium amaranticolor and members of the Solanaceae. Polyclonal antiserum against EMoV showed antigenic relationship with henbane mosaic potyvirus (HMV) and potato Y potyvirus (PVY). Virus-specific antibodies directed to the N-terminal region of EMoV cross-reacted only with PVY. Determination and comparison of nucleotide sequence of the coat protein (CP) and the 3'-untranslated region (UTR) of EMoV with other potyviruses showed that the level of homology was highest with PVY isolates. Comparative sequence analyses of the CP amino acid and 3'-UTR sequences with distinct PVY isolates placed EMoV within the PVYO subgroup.  相似文献

In Brazil plants of Pfaffia glomerata with mosaic symptoms were found to be infected with a previously undescribed potyvirus, Pfaffia mosaic virus (PfMV). Virus particles were long and flexuous, c.  10 × 700–800 nm, and cylindrical inclusions typical of potyviruses were present in cells of infected tissue. Partial host-range studies revealed that in addition to P. glomerata , PfMV infected only Chenopodium amaranticolor and Chenopodium quinoa . It was efficiently transmitted by the aphids Aphis gossypii and Myzus persicae . Polyclonal antiserum produced against the PfMV coat protein (CP) reacted with Potato virus Y (PVY), but not with four other potyviruses in PTA-ELISA. The similarity of the nucleotide sequence of the PfMV coat-protein gene ( CP ) varied from 7 to 76% when compared with other members of the family Potyviridae . Similarity of the 3' NTR sequence varied from 4 to 23%. In both cases the highest similarity was with PVY. These data indicate that PfMV is a new species in the genus Potyvirus .  相似文献

A virus having tubular particles (124, 142 and 167 nm) was manually transmitted from and to Anthoxanthum odoratum (sweet vernal grass). Other experimental hosts were Chenopodium quinoa, C. amaranticolor, Zea mays, Avena sativa and Hordeum vulgare (barley); winter cultivars of barley were more often infected than spring cultivars. In tests which simulated mechanical injury, the virus was transmitted from infected to healthy A. odoratum but not to healthy barley. The virus particles, which were seen as aggregates in the cytoplasm of A. odoratum leaf cells and in anther walls, reacted strongly in immunosorbent electron microscopy tests with antisera to strains of barley stripe mosaic hordeivirus. Nevertheless, the virus could be distinguished from the Type strain of barley stripe mosaic virus by reciprocal serological tests and by symptoms in infected barley cv. Black Hulless. Using an antiserum produced against the virus, a survey of A. odoratum in the field revealed one of 72 tested plants to be naturally infected. The virus is probably that described by Catherall & Chamberlain (1980) and named anthoxanthum latent blanching virus.  相似文献

为了防止南芥菜花叶病毒(Arabis mosaic virus,ArMV)传入我国,采用血清学、分子生物学、电镜观察及生物学接种等方法对从荷兰进口的郁金香种苗进行了ArMV检测。结果表明:ArMV血清学反应为强阳性;RT-PCR反应扩增出370bp的特异性目标条带;RT-PCR产物与已报道的3个ArMV部分外壳蛋白基因的核苷酸序列同源性为91.00%~93.24%,氨基酸序列同源性为99%~100%;免疫电镜观察到叶片病汁液中含有直径约30nm的球状病毒粒体;该病毒在黄花烟、白肋烟、矮牵牛和昆诺藜等鉴别寄主上引起坏死和褪绿等典型症状,经DAS-ELISA检测,这些接种寄主均呈ArMV血清学阳性反应。故将该病毒鉴定为南芥菜花叶病毒。  相似文献

ABSTRACT Apple latent spherical virus (ALSV) expressing yellow and cyan fluorescent proteins (ALSV-YFP and ALSV-CFP) was used to investigate the distribution of identical virus populations in coinfected plants. In Chenopodium quinoa plants inoculated with a mixture of ALSV-YFP and ALSV-CFP, fluorescence from YFP and CFP was always distributed separately in both inoculated and upper uninoculated leaves. Inoculation of each ALSV-YFP and ALSV-CFP to different leaves of a C. quinoa plant resulted in the separate distribution of each virus population among different upper leaves. When C. quinoa leaves were first inoculated with ALSV-CFP and then ALSV-YFP was reinoculated into the same leaves at various times after the first inoculation, ALSV-YFP infected only tissues where ALSV-CFP infection had not been established. The spatial separation was also found in Nicotiana benthamiana leaves coinoculated with Bean yellow mosaic virus (BYMV)-YFP and BYMV-CFP. In contrast, both YFP and CFP fluorescence signals were observed in the same tissues of N. benthamiana leaves mixed infected with ALSV-YFP and BYMV-CFP. YFP fluorescence from ALSV-YFP in mixed-infected leaves was brighter and longer than in leaves infected with ALSV-YFP singly.  相似文献

A potyvirus that induced stunting and a characteristic bushy appearance at the apical region, due to proliferation of terminal branches with narrowed, reduced and deformed leaflets, was isolated from chickpea in India. The virus was sap-transmissible to 14 species of Chenopodiaceae, Leguminosae, Solanaceae and Malvaceae; Chenopodium amaranticolor was a good local lesion host. Virus particles, trapped by immunosorbent electron microscopy and stained with uranyl acetate, were 710 ×10 nm long. Purified virus preparations contained a single polypeptide species of 32,500 Da and one nucleic acid species of 3.1 · 106 Da. The virus was serologically related to soybean mosaic, azuki bean mosaic and peanut mottle viruses but not to clover yellow vein, pea seed-borne mosaic and bean yellow mosaic viruses.
On the basis of these properties, the virus was identified as a previously undescribed potyvirus in chickpea, for which the name chickpea bushy dwarf virus is proposed.  相似文献

A range of selected PVY isolates that induce superficial necrosis on potato tubers, originating from several countries, were compared with standard strains of PVA, PVV and PVY. Biological properties (e.g., host range, aphid transmissibility and relationships based on cross-protection between virus isolates) were studied. PVYNN isolates differ from the normal PVYC, PVYN and PVO strains by their ability to infect Capsicum annuum but not Chenopodium amaranticolor and C. quinoa. All PVYNN isolates are transmissible by Myzus persicae , without any significant differences from one standard strain. These additional data confirm that these tuber-necrosing isolates belong to PVY. However, they could be ranged in a homogeneous and distinct group inside the PVYN group, based on the differences revealed in the host range, in addition to the specific ability naturally to induce necrosis on tubers.  相似文献

在昆明地区栽培的香石竹上发现了香石竹斑驳病毒,人工接种可局部侵染苋色藜、墙生藜、千日红及番杏,可系统侵染昆诺阿藜和美国石竹,分别造成褪绿斑、斑驳或坏死斑。经二轮PEG(分子量6000)沉淀,病毒提纯量约220mg/kg鲜组织。电镜观察病毒颗粒直径为28nm,等轴对称。提纯物紫外扫描呈典型病毒核蛋白吸收峰,A260/A280值为1.56。提纯病毒经4次免疫家兔,所得抗血清经试管沉淀测定效价为1∶4096,琼脂糖双扩散效价为1∶1024。  相似文献

The aim of this work was to correlate the appearance of the symptoms, multiplication and spread of virus after mechanical inoculation of potato (Solanum tuberosum L.) cultivars showing different levels of susceptibility and sensitivity to Potato virus YNTN (PVYNTN). The potato cultivars used were the resistant cultivar Sante and susceptible cultivars Igor, Pentland squire and Désirée. The spread of the virus PVYNTN in infected plants was monitored using different methods: DAS-ELISA, tissue printing, immuno-serological electron microscopy and real-time PCR. In all three susceptible cultivars, the virus was detected in the inoculated leaves 4–5 days after inoculation. From there virus spread rapidly, first into the stem, then more or less simultaneously to the upper leaves and roots. Real-time PCR was shown to be very sensitive and enabled viral RNA to be detected in non-inoculated leaves of susceptible cultivar Igor earlier than other methods. Therefore, for exact studies of plant–virus interaction, a combination of methods which detect viruses on the basis of their different properties (coat protein, morphology or RNA) should be used to monitor the spread of viruses.  相似文献
