Objective

To characterize the cardiovascular effects of dexmedetomidine, with or without MK-467, following intravenous (IV) administration in cats.

Study design

Prospective Latin square experimental study.

Animals

Six healthy adult purpose-bred cats.

Methods

Cats were anesthetized with desflurane in oxygen for instrumentation with a carotid artery catheter and a thermodilution catheter in the pulmonary artery. One hour after discontinuation of desflurane, cats were administered dexmedetomidine (25 μg kg–1), MK-467 (600 μg kg–1), or dexmedetomidine (25 μg kg–1) and MK-467 (600 μg kg–1). All treatments were administered IV as a bolus. Cardiovascular variables were measured prior to drug administration and for 8 hours thereafter. Only data from the dexmedetomidine and dexmedetomidine–MK-467 treatments were analyzed.

Results

Dexmedetomidine produced significant decreases in heart rate, cardiac index and right ventricular stroke work index, and significant increases in arterial blood pressure, central venous pressure, pulmonary artery pressure and systemic vascular resistance index. Dexmedetomidine combined with MK-467 resulted in significant but transient decrease in blood pressure and right ventricular stroke work index.

Conclusion and clinical relevance

Following IV co-administration, MK-467 effectively attenuated dexmedetomidine-induced cardiovascular effects in cats. The drug combination resulted in transient reduction in arterial blood pressure, without causing hypotension.  相似文献

Objectives

1) To determine which peripheral artery commonly used for invasive arterial blood pressure (IBP) monitoring yields the least bias when compared with noninvasive blood pressure (NIBP) values obtained at the antebrachium of the dog, and 2) to identify and describe differences in systolic (SAP), mean (MAP) and diastolic arterial pressures (DAP) among different anatomical locations.

Study design

Prospective experimental study.

Animals

Twenty adult hound dogs weighing 24.5 ± 1.1 kg (mean ± standard deviation).

Methods

Four peripheral arteries—dorsal pedal, median caudal, intermediate auricular and superficial palmar arteries—were catheterized with 20 gauge, 3.8 cm catheters. One NIBP cuff was placed in the middle third of the antebrachium. Four sets of IBP and NIBP measurements were simultaneously collected every 2 minutes. A linear mixed model was performed to analyze the collected data.

Results

IBP values varied depending on the arterial catheterization site. The difference was greater for SAP. NIBP measured at the antebrachium had the best agreement with IBP measured at the median caudal artery.

Conclusion and clinical relevance

IBP varies among anatomical locations. The smallest bias and narrowest limits of agreement were obtained at the median caudal artery, providing the best overall agreement with the equipment studied. The median caudal artery may be the preferable anatomical location for clinical comparison studies between IBP and NIBP in dogs when the cuff is on the antebrachium.  相似文献

Objectives

To determine the feasibility of acquiring quality transesophageal (TEE), epicardial (EE), and intracardiac (ICE) echocardiographic images in ovine subjects and to discuss the merits of each technique with a focus on ICE image acquisition.

Animals

Eleven male castrated Dorset adult sheep.

Methods

Transesophageal echocardiography was performed under general anesthesia. Epicardial echocardiography was performed as part of an open chest (thoracotomy or sternotomy) experiment. Subjects were recovered with permanent jugular vein indwelling catheter and ICE from this approach was described. Feasibility of each technique was qualitatively assessed based on subjective image quality from three images for each image plane in each sheep.

Results

Transesophageal echocardiography was technically challenging and did not provide adequate image quality for consistent interpretation. Epicardial echocardiography and ICE had more favorable results with ICE demonstrating unique benefits for post-operative serial monitoring.

Conclusions

Epicardial echocardiography and ICE were effective imaging techniques. Epicardial echocardiography required the least specialized training but was considered to have limited feasibility due to its requirement for an open chest procedure. Even with the necessity for permanent indwelling jugular cannulation, ICE was the least invasive of the three imaging techniques and potentially the most practical approach for chronic studies by minimizing post-operative stress. Transesophageal echocardiography was not a feasible technique in this study.  相似文献

Objective

The evaluation of alfaxalone as a premedication agent and intravenous anaesthetic in pigs.

Study design

Prospective, clinical trial.

Animals

Nine healthy, 6–8-week-old female Landrace pigs weighing 22.2 ± 1.0 kg, undergoing epidural catheter placement.

Methods

All pigs were premedicated with 4 mg kg?1 alfaxalone, 40 μg kg?1 medetomidine and 0.4 mg kg?1 butorphanol administered in the cervical musculature. Sedation was subjectively scored by the same observer from 1 (no sedation) to 10 (profound sedation) prior to induction of anaesthesia with alfaxalone intravenously to effect. All pigs were maintained on alfaxalone infusions with the rate of administration adjusted to maintain appropriate anaesthetic depth. Quality of induction was scored from 1 (poor) to 3 (smooth) and basic cardiorespiratory variables were recorded every 5 minutes during anaesthesia. Results are reported as mean ± standard deviation or median (range) as appropriate.

Results

Sedation scores were 9 (7–10). Inductions were smooth in all pigs and cardiovascular variables remained within normal limits for the duration of anaesthesia. The induction dose of alfaxalone was 0.9 (0.0–2.3) mg kg?1. Three pigs did not require additional alfaxalone after premedication to facilitate intubation.

Conclusions and clinical relevance

Intramuscular alfaxalone in combination with medetomidine and butorphanol produced moderate to deep sedation in pigs. Alfaxalone produced satisfactory induction and maintenance of anaesthesia with minimal cardiovascular side effects. Appropriate monitoring of pigs premedicated with this protocol is required as some pigs may become anaesthetized after intramuscular administration of this combination of drugs.  相似文献

Objective

To evaluate the onset and duration of hematological changes and the use of Doppler ultrasound (spleen) in dogs sedated with acepromazine or xylazine.

Study design

Clinical study.

Animals

A total of 24 mixed breed dogs aged 1–4 years and weighing 15–25 kg.

Methods

Dogs were randomly distributed into two groups: acepromazine group (AG) which were administered acepromazine (0.05 mg kg?1) intramuscularly and xylazine group (XG) administered xylazine (0.5 mg kg?1) intramuscularly. Sonographic evaluations (morphologic and hemodynamic splenic vascularization) and hematologic tests were performed before drug administration (baseline) and 5, 15, 30, 60, 120, 240, 360, 480 and 720 minutes after drug administration.

Results

A significant reduction occurred in erythrogram variables in AG at 15–720 minutes corresponding with a significant enlargement of the spleen. In XG, a significant reduction was observed in the erythrogram variables at 30–60 minutes without a significant enlargement of the spleen. Hilar diameter did not change over time in either group. Flow alterations were found only in the splenic artery in AG, with a decreased final diastolic velocity observed at 60–120 minutes.

Conclusions

Administration of acepromazine resulted in decreased red blood cell count, hemoglobin, packed cell volume and an increased diameter of the spleen. Xylazine administration resulted in similar hematologic changes but of smaller magnitude and duration and without splenic changes. The absence of significant changes in the Doppler flow parameters of the splenic artery and vein and the hilar diameter suggests that the splenomegaly that was observed in AG was not due to splenic vasodilation. No splenic sequestration occurred after xylazine administration.

Clinical relevance

The results indicate that acepromazine decreases the erythrocyte concentrations by splenic erythrocyte sequestration and concomitant splenomegaly. Xylazine can cause slight hematologic changes, but without splenic changes.  相似文献

Objective

To describe a transorbital approach to the maxillary nerve block in dogs and compare it with a traditional approach.

Study design

Prospective, randomized controlled study.

Animals

Heads from 17 euthanized dogs (10 Greyhounds, three Border Collies and four of mixed breed).

Methods

A volume of 1 mL of methylene blue dye was injected by each of two techniques, a traditional percutaneous approach and a transorbital approach to the maxillary nerve block. Both techniques were used on each head, alternating the left and right sides after random assignment to the first head. The heads were dissected to reveal the maxillary nerve and the length of nerve stained was measured.

Results

There was no significant difference (p = 0.67) in the proportion of nerves stained for a length >6 mm by either technique (88.2% transorbital versus 82.3% percutaneous). The mean length of nerve stained did not differ significantly between the techniques (p = 0.26).

Conclusions and clinical relevance

The transorbital approach to the maxillary nerve block described here presents a viable alternative to the traditional percutaneous approach. Further study is required to confirm its efficacy and safety under clinical conditions.  相似文献

Objective

To determine the degree of agreement between arterial and venous blood gases in anesthetized lambs using a point-of-care analyzer.

Study design

Prospective experimental study.

Animals

A group of 12 female Dorset cross-bred lambs, weighing 37.3 ± 7.4 kg (mean ± standard deviation).

Methods

Lambs were anesthetized with isoflurane for catheterization of the jugular vein and femoral artery, and blood samples were collected simultaneously for analysis of pH, PCO2, PO2, base excess (BE), bicarbonate (HCO3?), total carbon dioxide (tCO2), oxygen saturation (SO2), ionized calcium (iCa) and potassium (K+) using the Vet Scan i-STAT handheld analyzer. The Bland–Altman method was used to calculate agreement between arterial and venous measurements.

Results

Strong agreement was identified between arterial and venous pH (bias = 0.04; 95% limits of agreement = 0.02–0.06), K+ (bias = –0.1 and 95% limits of agreement = –0.8 to 0.5) and iCa (bias = 0.04; 95% limits of agreement = –0.08 to 0.0003). There was poor agreement for PO2 (bias = 168.0; 95% limits of agreement = 77.4–258.7) and SO2 (bias = 9.1; 95% limits of agreement = 2.8–15.3), as these values were higher in arterial blood than in venous blood. Moderate agreement was present for BE (bias = 1.7; 95% limits of agreement = –2.7 to 6.1), PCO2 (bias = –2.1; 95% limits of agreement = –7.5 to 3.4), HCO3? (bias = 1.1; 95% limits of agreement = –3.1 to 5.3) and tCO2 (bias = 1.0; 95% limits of agreement = –3.3 to 5.3).

Conclusions and clinical relevance

Venous blood can be used for the measurement of pH, K+ and iCa in anesthetized Dorset cross-bred lambs. Arterial blood is required for accurate measurement of PO2, SO2, PCO2, HCO3?, tCO2 and BE to assess systemic blood oxygenation and pulmonary function in anesthetized Dorset cross-bred lambs.  相似文献

Objectives

To determine the prevalence of mitral valve regurgitation (MR) in asymptomatic Swedish Norfolk terriers.

Animals

Seventy-nine privately owned Norfolk terriers.

Materials and methods

A prospective observational study was conducted where dogs were recruited via the Swedish Norfolk terrier club. All dogs were examined using the same protocol including physical examination and Doppler echocardiography.

Results

Fifteen dogs (19%) had a murmur at the time of the examination. A total of 35 dogs (44%) had MR, including 23 dogs (29%) with both MR and tricuspid valve regurgitation and 12 dogs (15%) with MR only, identified on Doppler echocardiography. In addition, 7 dogs (9%) had tricuspid valve regurgitation only. The prevalence of MR increased with increasing age (p < 0.0001).

Conclusions

Mitral valve regurgitation is common in asymptomatic Norfolk terriers with and without murmurs and the prevalence increases with age. The impact of MR in this breed on survival remains to be elucidated by a longitudinal study.  相似文献

Background

False tendons (FTs) are string-like structures in the left ventricle. A FT might produce focal thickening at its insertion region of the left ventricle, which could be mistaken for focal hypertrophic cardiomyopathy.

Objectives

To perform a prospective, echocardiographic follow-up examination of feline FTs and compare the wall thickness at the FT insertion region and a normal region without FTs at both examinations.

Animals

One hundred twenty-eight cats with one or multiple FTs without other cardiac abnormalities or systemic disease.

Methods

Measurements of the interventricular septum at end-diastole at a region with and without FT insertion were performed using two-dimensional echocardiography at both examinations and compared statistically using a Student's t-test.

Results

The follow-up interval ranged from 5 to 110 months (mean, 33 months). Myocardial wall segments with FT insertions were significantly thicker compared with neighboring wall regions in the long axis, but not in the short-axis views obtained. Comparing the wall thickness of follow-up examinations with the initial examination, revealed a significant growth of both FT and non-FT segments. However, differences in growth between the FT region and region without FTs were not statistically different.

Conclusions and clinical importance

Many normal cats have FTs, associated with focal thickening compared with neighboring regions. This thickening can increase over time, proportionate to growth in other (non-FT) segments. The association of such thickening with an FT and the absence of disproportionate growth in this segment over time suggests that these segments are simply thicker related to FT insertion.  相似文献

Objective

To describe a novel ultrasound-guided posterior extraconal block in the dog.

Study design

Prospective experimental cadaveric study.

Animals

A total of 13 Beagle Cross cadaver heads.

Methods

After describing the ultrasound bony landmarks and posterior extraconal local regional technique in one head, 12 heads were used to evaluate the spreading of contrast and evaluate potential complications. A 5–8 MHz microconvex ultrasound probe was positioned caudal to the orbital ligament, with the beam orientated transversely, and then tilted caudally until the orbital fissure was visualized. After identifying the bony structures consistent with the orbital fissure, a needle was advanced using an in-plane technique and 0.5 mL of a 50:50 mixture of iohexol and methylene blue was injected. Computed tomography (CT) and dissection were used to evaluate successful injections and potential complications. The injection was considered successful if radiopaque contrast medium was 5 mm from the orbital fissure. Potential complications were defined as the presence of radiopaque contrast within the globe or the intracalvarial tissues.

Results

The CT images confirmed contrast at the target site in 15/24 (63%) of the injections. Only two injections were found in the temporalis muscle; the rest of the injections were located in the extraconal space. No potential complications such as intracranial spreading of contrast, intravascular or intraocular injection were found.

Conclusions and clinical relevance

The technique can deliver contrast close to the main nerves which provide sensory and motor innervation to the eye. Further studies are needed to evaluate this technique in clinical cases.  相似文献

Introduction

The aim of this study was to establish echocardiographic reference values for the equine species using allometric regression equations based on body weight (BW) and thoracic circumference (TC).

Animals

A total of 239 horses or ponies were studied, including 65 warmbloods, 33 Standardbreds, 41 Thoroughbreds, 32 Arabian horses, 28 draft horses, and 40 ponies aged from 1 day to 30 years, weighing from 18 to 890 kg, with no evidence of cardiac disease.

Methods

For each horse or pony, a two-dimensional and M-mode echocardiography was performed. Within each breed, the relationships between BW or TC and echocardiographic dimensions were examined using power regression equations. Predictions and their 95% prediction intervals were calculated for the echocardiographic measurements.

Results

Within each breed, all echocardiographic measurements showed a significant and positive relationship with a high coefficient of determination for the estimation of the regression equations using BW and TC as the main explanatory variables. Breed-specific power regression equations as well as the 95% prediction intervals were calculated for each echocardiographic measurement as a function of BW and TC.

Conclusions

In the future, the body size-corrected and breed-specific echocardiographic reference values calculated in the present study could be used to discriminate between normal and abnormal values in a given animal.  相似文献

Objective

To investigate the pharmacokinetics of buprenorphine and its main active metabolite, norbuprenorphine, after administration of an intravenous loading dose followed by constant rate infusion (CRI) in dogs.

Study design

Prospective, clinical study.

Animals

A total of seven healthy dogs undergoing elective ovariectomy.

Methods

Buprenorphine was administered as a loading dose (intravenous bolus of 15 μg kg?1) followed by CRI (2.5 μg kg?1 hour?1 for 6 hours). Moreover, intraoperative analgesia was supplemented by an intramuscular carprofen (4 mg kg?1) injection, administered prior to surgery, and by lidocaine, administrated through subcutaneous infiltration and through a splash on the ovarian vascular pedicle during surgery. Pain and sedation were scored for all animals throughout the 24-hour study period and rescue analgesia was administered when a visual analogue scale score was > 40 mm. Blood samples were collected from a jugular catheter at regular intervals, and plasma concentrations of buprenorphine and norbuprenorphine were determined by a validated liquid chromatography–tandem mass spectrometry method.

Results

Buprenorphine showed a two-compartment kinetic profile. Maximum concentration was 23.92 ± 8.64 ng mL?1 at 1 minute (maximum time); elimination half-life was 41.87 ± 17.35 minutes; area under the curve was 486.68 ± 125.66 minutes ng?1 mL?1; clearance was 33.61 ± 13.01 mL minute?1 kg?1, and volume of distribution at steady state was 1.77 ± 0.50 L kg?1. In no case was rescue analgesia required. Norbuprenorphine resulted below the lower limit of quantification in almost all samples.

Conclusions and clinical relevance

The results suggest that a buprenorphine CRI can be a useful tool for providing analgesia in postoperative patients, considering its minor side effects and the advantages of a CRI compared to frequent boluses. The negligible contribution of norbuprenorphine to the therapeutic effect was confirmed.  相似文献

Introduction

Transthoracic echocardiography (TTE) is the primary tool for the assessment of cardiac structure and function in dogs but is challenging in English bulldogs due to dorsoventral compression of the thorax, obesity, and narrow intercostal spaces. Multi-detector computed tomography angiography (CTA) may overcome the conformational obstacles of cardiac imaging in this breed.

Animals

Eleven client-owned English bulldogs.

Materials and Methods

Prospective clinical trial with paired analysis of TTE and CTA studies.

Results

Eight of the 25 linear cardiac dimensional measurements were significantly different between TTE and CTA (p<0.033). Intraobserver agreement was strong with average coefficients of variation (CV) of 5.34% for TTE and 2.50% for CTA. Interobserver agreement CV averaged 6.5% for TTE and 8.75% CTA. Ejection fraction, stroke volume, and end-systolic volume were significantly different between modalities (all p<0.002). No significant difference was present between end-diastolic volume for TTE compared with CTA.

Discussion

High-quality cardiac angiographic studies were accomplished using CTA without the use of general anesthesia in English bulldogs. Multi-detector computed tomography angiography and TTE are not interchangeable modalities in the clinical setting.

Conclusion

Multi-detector-CT ECG-gated cardiac angiography is possible in sedated, non-intubated English bulldogs. Differences were found between some cardiac dimensions as measured by TTE in the awake dog and compared with sedated CTA, indicating the two methodologies are not equivalent. Sedated, non-intubated CTA yielded high-quality imaging with strong intraobserver and interobserver measurement repeatability in English bulldogs.  相似文献

Objective

To compare electrical velocimetry (EV) noninvasive measures of cardiac output (CO) and stroke volume variation (SVV) in dogs undergoing cardiovascular surgery with those obtained with the conventional thermodilution technique using a pulmonary artery catheter.

Study design

Prospective experimental trial.

Animals

Seven adult Beagle dogs with a median weight of 13.6 kg.

Methods

Simultaneous, coupled cardiac index (CI; CO indexed to body surface area) measurements by EV (CIEV) and the reference pulmonary artery catheter thermodilution method (CIPAC) were obtained in seven sevoflurane-anaesthetized, mechanically ventilated dogs undergoing experimental open-chest cardiovascular surgery for isolated right ventricular failure. Relationships between SVV or central venous pressure (CVP) and stroke volume (SV) were analysed to estimate fluid responsiveness. Haemodynamic data were recorded intraoperatively and before and after fluid challenge.

Results

Bland–Altman analysis of 332 matched sets of CI data revealed an overall bias and precision of – 0.22 ± 0.52 L minute?1 m?2 for CIEV and CIPAC (percentage error: 30.4%). Trend analysis showed a concordance of 88% for CIEV. SVV showed a significant positive correlation (r2 = 0.442, p < 0.0001) with SV changes to a volume loading of 200 mL, but CVP did not (r2 = 0.0002, p = 0.94). Better prediction of SV responsiveness (rise of SV index of ≥ 10%) was observed for SVV (0.74 ± 0.09; p = 0.014) with a significant area under the receiver operating characteristic curve in comparison with CVP (0.53 ± 0.98; p = 0.78), with a cut-off value of 14.5% (60% specificity and 83% sensitivity).

Conclusions and clinical relevance

In dogs undergoing cardiovascular surgery, EV provided accurate CO measurements compared with CIPAC, although its trending ability was poor. Further, SVV by EV, but not CVP, reliably predicted fluid responsiveness during mechanical ventilation in dogs.  相似文献

Objective

To test whether neurotoxic effects of a bupivacaine liposome injectable suspension differ from those of a standard formulation of bupivacaine hydrochloride (HCl) after intraneural injection into the sciatic nerves in pigs.

Study design

Prospective, randomized study.

Animals

Fifteen pigs, hybrids of Landrace and Large White.

Methods

After the National Ethics Committee approval, 15 pigs were randomly allocated to three groups (n = 5/group) to receive intraneural injections of 4 mL of 1.33% bupivacaine liposome injectable suspension, 0.5% bupivacaine HCl or normal saline. Serial neurologic examinations were conducted to detect sensory and motor response to noxious stimuli using a modified Thalhammer’s scale at 2 hour intervals for the first 12 hours after injection and daily thereafter for 2 weeks. Fiber characteristics (density) of the harvested sciatic nerves were measured during histomorphometric analysis. Inflammatory response was studied using immunohistochemical analysis. Data were tested using analyses of variance; p values for paired comparisons were Bonferroni adjusted.

Results

Compared with bupivacaine HCl, bupivacaine liposome injectable suspension provided longer sensory (11.2 ± 1.8 hours versus 3.2 ± 1.1 hours, respectively, p < 0.0001) and motor (10.0 ± 2.0 hours versus 4.0 ± 1.4 hours respectively, p < 0.0001) blockade. Histomorphometric parameters were similar among the groups. No changes in axonal density or myelin structure indicative of injury to the sciatic nerves were observed in any of the groups. Number of immunopositive cells did not differ between the bupivacaine liposome injectable suspension (23 ± 6 cells per mm2) and the bupivacaine HCl groups (21 ± 4 cells per mm2), p > 0.90.

Conclusions and clinical relevance

Intraneural injections of bupivacaine liposome injectable suspension or bupivacaine HCl in our porcine model did not result in evidence of neurotoxicity.  相似文献

Objective

To evaluate motor and sensory blockade of combining dexmedetomidine with ropivacaine, administered perineurally or systemically, for femoral and sciatic nerve blocks in conscious dogs.

Study design

Randomized, controlled, experimental study.

Animals

Seven healthy Beagle dogs, aged 3.3 ± 0.1 years and weighing 11.0 ± 2.4 kg.

Methods

Dogs were anesthetized with isoflurane on three separate occasions for unilateral femoral and sciatic nerve blocks and were administered the following treatments in random order: perineural ropivacaine 0.75% (0.1 mL kg–1) on each nerve and intramuscular (IM) saline (0.2 mL kg–1) (GCON); perineural dexmedetomidine (1 μg mL–1) and ropivacaine 0.75% (0.1 mL kg–1) on each nerve and IM saline (0.2 mL kg–1) (GDPN); and perineural ropivacaine 0.75% (0.1 mL kg–1) on each nerve and IM dexmedetomidine (1 μg mL–1, 0.2 mL kg–1) (GDIM). Nerve blocks were guided by ultrasound and electrical stimulation and dogs were allowed to recover from general anesthesia. Sensory blockade was evaluated by response to clamp pressure on the skin innervated by the saphenous/ femoral, common fibular and tibial nerves. Motor blockade was evaluated by observing the ability to walk and proprioception. Sensory and motor blockade were evaluated until their full recovery.

Results

No significant differences in onset time to motor and sensory blockade were observed among treatments. Duration of motor blockade was not significantly different among treatments; however, duration of tibial sensory blockade was longer in the GDPN than in the GDIM treatment.

Conclusions and clinical relevance

Although a longer duration of sensory blockade was observed with perineural dexmedetomidine, a significant increase compared with the control group was not established. Other concentrations should be investigated to verify if dexmedetomidine is a useful adjuvant to local anesthetics in peripheral nerve blocks in dogs.  相似文献

Objective

To determine the dose of cis-atracurium needed to produce a moderate neuromuscular blockade (NMB) in pigs.

Study design

Prospective experimental study.

Animals

Seven pigs [five females and two males; median (range) body weight: 47 (36–64) kg].

Methods

Pigs were premedicated with intramuscular midazolam (0.3 mg kg?1) and ketamine (7 mg kg?1). Anaesthesia was induced with intravenous (IV) propofol 3 (1–4) mg kg?1 and maintained with isoflurane in oxygen. Based on a preliminary study, the subjects were administered 0.3 mg kg?1 cis-atracurium followed by 0.48 mg kg?1 hour?1 constant rate infusion (CRI) IV. A moderate NMB was defined as a train-of-four (TOF) count of ≤2 by acceleromyography. When the TOF count was >2, 0.1 mg kg?1 cis-atracurium was administered and the CRI was increased. The cis-atracurium CRI was decreased when the TOF count was under 2 for more than 15 minutes. The total dose of cis-atracurium required to maintain a moderate NMB was calculated as the total amount of cis-atracurium used (both CRI and supplementary boluses) divided by the administration time.

Results

The cis-atracurium CRI lasted for 87 (76–151) minutes. To induce and maintain a moderate neuromuscular blockade, the initial dose of cis-atracurium was 0.3 (0.3– 0.5) mg kg?1 and the CRI was 0.71 (0.37–0.98) mg kg?1 hour?1.

Conclusions and clinical relevance

The doses described in our study may help researchers obtain a moderate NMB using cis-atracurium in pigs.  相似文献

Objective

To determine whether an ultrasound (US)-guided femoral nerve block using a ventral suprainguinal approach could be successfully achieved in sedated dogs; to measure the time to execute the nerve block, onset time, duration, and complete block rate in sensory and motor nerves; and to examine any differences between two volumes for injection.

Study design

Blinded crossover experimental study.

Animals

A total of 10 clinically healthy adult Beagle dogs.

Methods

The femoral nerve of the right pelvic limb was infiltrated with 0.5% bupivacaine at 0.4 (treatment 0.4B) or 0.2 mL kg?1 (treatment 0.2B), or saline at 0.4 mL kg?1 (control) in sedated dogs. The sensory and motor nerve functions were scored on a scale of 0 (complete blockade) to 2 (normal). The onset time and duration of the sensory and motor nerve blockade were compared between treatments 0.4B and 0.2B using a Wilcoxon signed rank test. Sensory and motor nerve function scores for each of the three treatments were compared at multiple time points using a nonparametric multiple comparisons test.

Results

The time to execute the nerve block was 2.5 ± 0.9 minutes (n = 30). For both 0.4B and 0.2B treatments, the onset times of both the sensory and motor nerve blockades were 15 minutes. The durations of the sensory nerve blockade for 0.4B and 0.2B were 9.9 ± 1.4 and 10.0 ± 1.2 hours, respectively, and those of the motor nerve blockades were 10.5 ± 1.3 and 10.2 ± 1.3 hours, respectively. No adverse effects were noted. No significant difference was observed between 0.4B and 0.2B.

Conclusions and clinical relevance

A US-guided femoral nerve block using a ventral suprainguinal approach demonstrated a short onset and long duration with 0.5% bupivacaine 0.2 mL kg?1 and can be performed under sedation in dogs.  相似文献
