Calcium salts have been reported to play an important role in the inhibition of postharvest decay of apples and in enhancing the efficacy of postharvest biocontrol agents. Therefore, the present study was conducted in order to examine and compare the effects of calcium and magnesium salts on the germination and metabolism of the postharvest pathogens Botrytis cinerea and Penicillium expansum , and to determine the effects of these salts on the biocontrol activity of two isolates (182 and 247) of the yeast Candida oleophila. Increasing concentrations of CaCl2 (25–175 mM) resulted in decreased spore germination and germ-tube growth of both pathogens. The greatest effect was observed in the case of B. cinerea. The inhibitory effect could be overcome by the addition of glucose to the germination medium. MgCl2 (25–175 mM) had no effect on germination or germ-tube growth of either pathogen, indicating that the calcium cation rather than the chloride anion was responsible for the inhibition. The pectinolytic activity of crude enzyme obtained from the culture medium of both pathogens was also inhibited by 25–175 mM CaCl2, with the greatest effect on the crude enzyme from P. expansum. Biocontrol activity of isolate 182 was enhanced by the addition of 90 or 180 MM CaCl2, whereas there was no effect on the biocontrol activity of isolate 247. This was apparently due to the inability of isolate 247 to proliferate in apple wounds. It is postulated that enhanced biocontrol activity of isolate 182 of the yeast C. oleophila in the presence of Ca2+ ions is directly due to the inhibitory effects of calcium ions on pathogen spore germination and metabolism, and indirectly due to the ability of isolate 182 to maintain normal metabolism in the presence of"toxic" levels of calcium.  相似文献

The efficacy of the commercial glyphosate [( N -phosphonomethyl) glycine] formulations Roundup Ultra, Touchdown and Engame were compared for the control of prickly sida ( Sida spinosa L.), morningglory ( Ipomeae hederacea var. integriuscula Gray), sicklepod ( Senna obtusifolia L.) and purple nutsedge ( Cyperus rotundus L.). Engame is a new formulation of glyphosate that contains glyphosate acid and 1-aminomethanamide dihydrogen tetraoxosulfate (AMADS), a proprietary mixture of sulfuric acid and urea, other than glyphosate salt and surfactants. Injury by Engame differed from Roundup Ultra and Touchdown in that necrotic lesions formed on leaves several hours after treatment. Leaves of very susceptible species, such as prickly sida, were rapidly, although incompletely, desiccated and then became chlorotic and died in a manner typical of other glyphosate formulations. Engame was 2–3 times more active to growth inhibition than either the Roundup Ultra or Touchdown formulations, based on GR50 comparisons expressed on an acid equivalent basis. The GR50 estimates did not change over the 3 week evaluation period for prickly sida and purple nutsedge, and after 2 weeks after treatment for morningglory. The GR50 estimates for sicklepod decreased over the 3 week evaluation period indicating a slower response to glyphosate. The application of AMADS alone caused minute necrotic lesions on sicklepod and purple nutsedge, and lesions up to 3 mm in diameter on prickly sida and morningglory. Further injury from AMADS was not noted and plants resumed growth without apparent delay. At glyphosate rates above 1120 g ha−1, greater than 80% control was achieved at 7 days after treatment. These results demonstrate that glyphosate efficacy can be further enhanced by formulations that apparently improve uptake and translocation.  相似文献

Genetic variation of the barley powdery mildew fungus ( Erysiphe graminis f.sp. hordei ) was estimated in three Danish local populations. Genetic variation was estimated from the variation amongst clones of Egh , and was therefore an estimate of the maximum genetic variation in the local populations. The average gene diversity, Ĥ , was estimated as 0.84. The effective population size was estimated as: log10 ( N^ e ) = 0.64 − log10(μ), or 4.4 × 109, assuming a nucleotide mutation rate ( μ) of 10−9 per base per generation. The sensitivity of 22 major crops, pastures and weeds from the north-east grain region of Australia to atrazine and chlorsulfuron residues was determined in a glasshouse using a soil-free bioassay system. A logistic equation was fitted to the seedling fresh weights as a function of the logarithm of herbicide concentration by non-linear regression and used to calculate the doses for 10%, 30% and 50% inhibition of seedling growth (ID10, ID30 and ID50). The ID50 for atrazine ranged from 0.03 to 0.04 mg a.i. L–1 for Salvia reflexa Hornem. and barley to 1.47 mg a.i. L–1 for sorghum. The ID50 for chlorsulfuron ranged from 0.19 to 0.21 μg a.i. L–1 for lucerne and snail medic to 102 μg a.i. L–1 for wheat. Based on ID50 values measured, the predicted responses of each species to a range of concentrations of atrazine and chlorsulfuron were classified into four categories ranging from no damage to severe damage. These sensitivity data will assist in planning cropping sequences in soils previously treated with atrazine or chlorsulfuron.  相似文献

Twenty-two cotton varieties were screened for resistance to cotton leaf curl disease (CLCuD), a disease of viral origin, using three procedures: field evaluation, whitefly transmission assay and graft inoculation. Viral infection of cotton varieties was determined by visual symptom assessment as well as dot-blot and multiplex PCR diagnostic techniques. Crosses were made between the most susceptible variety (S-12) and highly resistant varieties (CP-15/2, LRA-5166 and CIM-443). All F1 plants of these crosses were resistant, showing dominant expression of the resistance as well as the absence of extrachromosomal inheritance. The F2 plants of the crosses CP-15/2 × S12, LRA-5166 × S-12 and CIM-443 × S12 exhibited a ratio of 13 resistant (symptomless) to three susceptible (with symptoms). Screening of the F2 generation for virus infection by multiplex PCR further subdivided the resistant class into those exhibiting a high level of resistance (HR; PCR-negative) and those exhibiting resistance (R; symptomless, yet showing virus replication by PCR analysis). Hence, the final ratio was 3:10:3 (HR:resistant:susceptible). The F3 progeny of susceptible F2 plants segregated for resistance, indicating the probable presence of a suppressor gene ( S ). These findings are consistent with three genes being involved in G. hirsutum resistance to CLCuD, two for resistance ( R 1CLCuDhir and R 2CLCuDhir ) and a suppressor of resistance ( S CLCuDhir ).  相似文献

In glasshouse and laboratory experiments, calcium alginate gel, produced in situ on wounds by successive dipping in solutions of sodium alginate and CaCl2 gave significant protection against infection by pathogenic isolates of Agrobacterium tumefaciens on carrot discs, tomato cuttings and chrysanthemum cuttings. Gel protection was sometimes equal to that given by strain K84 of A. radiobacter and also operated against a pathogenic isolate insensitive to control by strain K84. Sodium alginate or CaCl2 alone gave little or no protection, suggesting that calcium alginate acted by physically excluding the pathogen from wound surfaces. The use of the method described is discussed for delivery of biological or chemical control agents to plant surfaces whilst also providing protection in its own right.  相似文献

The experiment was conducted under a controlled environment to study the effects of different temperature regimes (15/10°C, 20/15°C, and 25/20°C day/night) and sowing depths (0, 2, 4, and 6 cm) on the seedling emergence and early growth (height gain) of wheat (cv. Marvdasht) and wild barley ( Hordeum spontaneum ). The cumulative emergence and plant height gain over time were modeled with the use of a logistic function. For a particular temperature regime, the maximum percentage emergence (E max ) of wheat was higher than that of wild barley across all sowing depths. The maximum and minimum E max values for both species occurred at 20/15°C and 25/20°C, respectively. The time taken to reach 50% of the E max (i.e. E50) increased with the sowing depth in both species under all temperature regimes. The E50 of wild barley was greater than that of wheat for all temperature regimes, with maximum differences observed at 20/15°C. The greatest maximum plant height (H max ) was observed at the surface planting for both plants. The H max was reduced at temperatures either lower or higher than 20/15°C, with a more notable reduction in wild barley. At all temperature regimes, the time taken to reach 50% of the H max (i.e. H50) increased linearly with the sowing depth but, at higher temperatures, the accelerated growth rate reduced the H50. The wild barley seedling emergence and height gain rate, as expressed relative to those of wheat, revealed the highest superiority of wheat over wild barley at 25/20°C and the sowing depth of 4 cm.  相似文献

Transfer of factors for resistance to white blister disease caused by Albugo candida between Brassica species involving two genotypes each of B. juncea and B. rapa was studied in hybrids. More hybrids were obtained by in vivo than in vitro techniques, although an in vitro phase was a prerequisite for the establishment of in vivo hybrids. Hybrids were identified by PCR-based inter-simple sequence repeat (ISSR) markers with both male and female species-specific bands being identified. There was a positive correlation between disease severity and number of days after sowing ( r  > 0·93), the highest being towards pod formation and plant maturity at 110 days after sowing. The plants from F2 and BC1 progeny showed higher resistance to A. candida than either of the parents. Plants of B. juncea and B. rapa with high field resistance (disease index < 1·0) were selected from BC2 and F2BC1 generations. The frequency of plants classified as resistant in BC2 progeny ranged from 4·5 to 39·0% in cross-combinations involving B. juncea genotypes as female parent, compared with 100% in the reciprocal cross involving B. rapa as female parent.  相似文献

Conditioned seeds of Striga asiatica (L.) Kuntze release ethylene, which elicits germination. We investigated the activity of 1-aminocyclopropane-1-carboxylate (ACC) oxidase and respiration during conditioning. Seeds incubated in vivo with ACC, the substrate for ACC oxidase, produced negligible ethylene at the beginning of conditioning or if they were dormant (i.e. would not germinate after conditioning and treatment with stimulant). Non-dormant seeds produced 3000 ηL of ethylene/600 seeds/24 h after 12 days of conditioning. In vitro ACC oxidase activity at day 0 of conditioning produced 500 ηL of ethylene/μg protein/h and 8000 ηL of ethylene/μg protein/h after 12 days of conditioning. Incubation of seeds in strigol before protein extraction did not enhance enzyme activity. Seeds released 4000 μL/L CO2 in the first 24 h of conditioning, with the rate increasing to 15 000 μL/L/24 h on day 4 and then remaining roughly unchanged. Maximum in vitro activity of ACC oxidase required ACC, catalase, O2, Fe2+, ascorbate and CO2. In vivo activity of ACC oxidase required ACC and/or germination stimulant(s), suggesting that stimulants may be involved in providing substrates for the ACC oxidase. No difference was observed in the separation of extracted proteins, which suggests that ACC oxidase is activated during conditioning, perhaps as a result of changes in co-factor concentration. Application of these findings to Striga control is discussed.  相似文献

The genetics of avirulences towards barley mildew resistances were analysed in crosses of the Ervsiphe graminis f.sp. hordei isolate DH14 with CC107 and with CC138. Nine avirulences, Av ra9, Avr a10, Avr a11, Avr a12, Avr Ab, Avr CP, Avr h, Avr k and Avr La, segregated as single genes in one or other cross. However. F1 segregation data were consistent with avirulence matching the Mla7 resistance gene being controlled by two genes, designated Avr a7 1 and Avr a7 2. Infection types of avirulent isolates differed on varieties in which Mla7 had been derived from each of the four known sources of that resistance. Linkage was detected between Avr a7 1 and Avr h in the cross CC107 × DH14, and between Avr a10 and Avr k, Avr a11 and Avr La, and Avr h and the triadimenol response gene Tdl2 in CC138 × DH14.  相似文献

Populations of F1, F2, F1× parent 1 (BC1), F1× parent 2 (BC2), and F3 resulting from two crosses between four susceptible varieties of barley used in national and international breeding programmes were tested at the seedling stage for their resistance to an isolate of Pyrenophora teres which is virulent to the parental varieties. Infection type, average lesion size, and number of lesions per unit leaf area were used to assess disease reaction. In the two crosses, F2 and especially F3 generations (produced by self pollination of selected resistant F2 plants) showed more resistance expressed by infection type and average lesion size than either parent. The frequency distributions of F2 and F3 generations derived from these crosses were continuous and showed transgressive segregation for resistance. The results indicated that the four cultivars used in this study possessed additive genes for resistance to P. teres. Thus useful resistance could be obtained following crossing of susceptible cultivars and selection in succeeding generations.  相似文献

In 2002, the Malaysian government had banned the use of the hazardous herbicide, paraquat. Most growers perceive that paraquat is the most effective herbicide and provides the fastest mode of action to control weeds. An experiment was conducted at MAB Agriculture-Horticulture, Sepang, Selangor, Malaysia, from February 2004 to February 2005 to evaluate the efficacy and ability of the less hazardous herbicides, glufosinate ammonium and glyphosate, as an alternative to the hazardous herbicide, paraquat, in controlling weeds in immature oil palm (<3 years old). The results showed that paraquat needed high rates, 600 and 800 g ha−1, to control weeds effectively. However, lower rates of glufosinate ammonium (200 g ha−1) and glyphosate (400 g ha−1) gave excellent weed control. The results showed that the efficacy of glufosinate ammonium and glyphosate were much better than paraquat. The results also showed that, with no direct contact with the plants, paraquat, glufosinate ammonium, and glyphosate had no adverse effect on the vegetative and generative growth of oil palm in this study. These results proved that the less hazardous herbicides, glufosinate ammonium and glyphosate, could be used as an alternative to paraquat to control weeds in immature oil palm.  相似文献

Stems of Chenopodium album . and Sinapis arvensis . and leaves of Lolium perenne . were cut with a CO2 laser or with a pair of scissors. Treatments were carried out on greenhouse-grown pot plants at three different growth stages and at two heights. Plant dry matter was measured 2 to 5 weeks after treatment. The relationship between dry weight and laser energy was analysed using a non-linear dose–response regression model. The regression parameters differed significantly between the weed species. At all growth stages and heights S. arvensis was more difficult to cut with a CO2 laser than C. album . When stems were cut below the meristems, 0.9 and 2.3 J mm−1 of CO2 laser energy dose was sufficient to reduce by 90% the biomass of C. album and S. arvensis respectively. Regrowth appeared when dicotyledonous plant stems were cut above meristems, indicating that it is important to cut close to the soil surface to obtain a significant effect. When cutting L. perenne plants with 2-true leaves at a height of 2 cm from the soil surface with a laser, the biomass decreased significantly compared with plants cut by scissors, indicating a delay in regrowth. This delay was not observed for the dicotyledonous plants nor for the other growth stages of L. perenne .  相似文献

The susceptible rose cv. Madelon and the partially resistant cv. Sonia both responded with reduced development of rose powdery mildew when they were treated with the synthetic inducer 2,6-dichloroisonicotinic acid (INA). The EC50 for number of colonies cm−2 was approximately 0.4 mg L−1 in both cultivars when treated 4 days prior to inoculation. However, conspicuous differences were observed with respect to number of spores per cm2. For sporulation, the EC50 was 0.37 mg L−1 in cv. Madelon and only 0.08 mg L−1 in cv. Sonia. A comparison with the pathosystems cucumber/ Sphaerotheca fuliginea and red cabbage/ Peronospora parasitica is made and the importance of the observed phenomenon for the selection of parents in a breeding programme for (partial) resistance is discussed.  相似文献

Paddy herbicides have the potential to cause adverse effects on non-target plants. Susceptibilities of some aquatic ferns ( Azolla japonica Franch. et Savat., Isoetes japonica A. Braun, Marsilea quadrifolia L. and Salvinia natans All.) and duckweeds ( Lemna minor L. and Spirodela polyrhiza Schleid.) to paddy herbicide bensulfuron methyl (BSM) were evaluated with a 20 day exposure experiment using 200 cm2 pots. The BSM concentrations in the surface water of monitoring pots with no plants dissipated exponentially with half lives of 3.5 and 3.9 days at application rates of 15 and 150 g ha−1, respectively. The BSM concentrations in the surface water 1 day after application in the culture pots were comparable among plant species, and were lower than those in the monitoring pots. Bensulfuron methyl reduced the plant growth in all species. I . japonica showed the lowest intrinsic relative growth rate (RGR) and the lowest susceptibility with an effective dose resulting in 50% growth inhibition (ED50) of 21 g ha−1. Except for I . japonica , the RGR of the duckweeds was similar to the ferns, and ED50 for the duckweeds was higher than the ferns. ED50 for Sa . natans , A . japonica and M . quadrifolia were 1.1, 1.8 and 1.2 g ha−1, respectively, which were smaller than 1/20 of the recommended field dose (51–75 g ha−1) and ranged from 1/2 to 1/6 of ED50 for L . minor and Sp . polyrhiza (6.5 and 3.2 g ha−1, respectively). These results suggest that BSM application in paddy fields and its runoff in some localities is expected to have adverse effects on the growth of Sa . natans , A . japonica and M . quadrifolia .  相似文献

The reactions of several tomato cultivars and the genetic basis of resistance to an isolate of Alternaria alternata f. sp. lycopersici were examined. Among the 105 cultivars tested only two, Regal and Toper, were susceptible, while the remainder were highly resistant. The resistant cultivar ACE 55 VF (P;) was crossed with the susceptible cultivar Earlypak No. 7 (P1)- When F1, F2, F3, BC1, BC1 selfed, F2 X P2 and BC1P2 x P1 progenies were inoculated at the two-leaf stage with a suspension of spores of the pathogen, the ratios of resistant to susceptible plants indicated that resistance was conferred by a single dominant gene.  相似文献

The effects of the density and proportion of both volunteer barley ( Hordeum vulgare L.) and Stellaria media (L.) Vill. (common chickweed) on the seed yield of both species was investigated in linseed ( Linum usitatissimum L.) and autumn-sown field bean ( Vicia faba L.). A model was created to estimate these effects. It was a combination of two models. The first was a simple linear model relating weed seed number m2 to weed dry weight m2. The second was an inverse linear model relating weed dry weight per plant to weed density. A very good relationship existed between barley dry weight and number of seeds m2 and between S. media dry weight and number of seed capsules m2. This relationship was relatively consistent between experiments. The inverse linear model provided a good estimation of the relationship between weed density and weed dry weight per plant for both barley and S. media . Model variables, however, differed between experiments. Using the combined model it was estimated that, in the absence of other weed species and at a density of 800 plants m2, S. media would produce between 4000 and 48 000 seeds m2, whereas volunteer barley, at a density of 400 plants m2, would produce between 2000 and 8000 seeds m2. The presence of barley always reduced S. media seed yield and a barley population of 100 plants m2 could reduce S. media seed yield by up to 79%. The presence of S. media reduced barley seed yield in only one of three experiments, in which a S. media density of 800 plants m2 reduced barley seed yield by up to 68%. A survey was made on the occurrence of soilborne Phytophthora species in 35 oak stands on a range of geologically different sites in Bavaria. The most widespread species were P. quercina , P. cambivora and P. citricola . Seven other Phytophthora species were isolated infrequently. The fine root systems of 106 healthy and 111 declining mature trees of Quercus robur and Q. petraea were intensively investigated. The results indicate that, depending on the site conditions, at least two different complex diseases are referred to under the name 'oak decline'. On sites with a mean soil pH (CaCl2)  3·5 and sandy-loamy to clayey soil texture Phytophthora spp. were commonly isolated from rhizosphere soil, and highly significant correlations existed between crown transparency and various root parameters. Oaks with P. quercina or other Phytophthora spp. in their rhizosphere had markedly higher levels of fine root damage than oaks without Phytophthora spp., and were subject to a relative risk of severe crown symptoms of 2·1 and 2·8, respectively. In contrast, in stands with sandy to sandy-loamy soils and a mean soil pH  3·9, Phytophthora spp. were not found. In these stands, correlations between crown transparency and various root parameters were either less significant or not significant. It is concluded that Phytophthora species are strongly involved in oak decline on sandy-loamy to clayey sites with a mean soil-pH (CaCl2)  3·5.  相似文献

Field experiments were conducted at five locations in the major wheat production regions of Iran to evaluate the efficacy of sulfosulfuron in controlling weed barley species (including Hordeum spontaneum , Hordeum murinum , Hordeum distichon , and Hordeum vulgare ) in the 2004–2005 and 2005–2006 growing seasons. Sulfosulfuron was applied either postemergence (POST) or preplant-incorporated (PPI) at 0, 20.25, 30.75, 40.5, 51.0, 60.75 or 71.25 g ai ha−1 to plots arranged in a randomized complete block design with four replications. Sulfosulfuron at the recommended rate (20.25 g ai ha−1) failed to provide acceptable control of the weed barley species. However, the level of control increased with the application rate, particularly at rates >51.0 g ai ha−1. Generally, PPI-applied sulfosulfuron resulted in markedly greater control levels than those of a POST application and complete control of H. murinum and H. vulgare was achieved with PPI-applied sulfosulfuron at all rates >20.25 and 30.75 g ai ha−1, respectively. In most cases, the wheat yield increased with the application rate without any crop injury. The highest yield increase (186%) was obtained with a PPI application of 71.25 g ai ha−1.  相似文献

A total of 651 isolates of cucumber corynespora leaf spot fungus ( Corynespora cassiicola ) collected from cucumber in Japan, either with (438 isolates) or without (213 isolates) a prior history of boscalid use, were tested for their sensitivity to boscalid by using a mycelial growth inhibition method on YBA agar medium. Additionally, seven isolates of C. cassiicola obtained from tomato, soybean, eggplant (aubergine) and cowpea in different locations in Japan were tested before boscalid registration. Minimum inhibitory concentration (MIC) and 50% effective concentration (EC50) values for 220 isolates from crops without a prior history of boscalid use ranged from 0·5 to 7·5 μg mL−1 and from 0·04 to 0·59 μg mL−1, respectively. Two hundred and fourteen out of 438 isolates collected from ten cucumber greenhouses in Ibaraki Prefecture, Japan, which received boscalid spray applications showed boscalid resistance, with MIC values higher than 30 μg mL−1. Moreover, resistant isolates were divided into two groups: a moderately resistant (MR) group consisting of 189 isolates with EC50 values ranging from 1·1 to 6·3 μg mL−1, and a very highly resistant (VHR) group consisting of 25 isolates with EC50 values higher than 24·8 μg mL−1. MR isolates were detected from all ten greenhouses, but VHR isolates were detected from only three. As a result of fungus inoculation tests which used potted cucumber plants, control failures of boscalid were observed against resistant isolates. Efficacy of boscalid was remarkably low against VHR isolates in particular. This is the first known report on boscalid resistance in Japan.  相似文献
