Wholegrain consumption is associated with several health benefits, in contrast to the consumption of refined grains. This can partly be related to the antioxidant compounds in the outer parts of the grain kernel. The bioaccessibility of these antioxidant compounds from the wholegrain matrix during gastrointestinal digestion is crucial for their absorption and bioavailability. In the current study, the bioaccessible compounds from aleurone, bran and flour were obtained from a dynamic in vitro model of the upper gastrointestinal tract. They were collected at 1 h time intervals to assess their antioxidant capacity (TEAC assay) and also their anti-inflammatory effect (TNF-α reduction in U937 macrophages stimulated with LPS). The bioaccessible compounds from aleurone had the highest antioxidant capacity and provided a prolonged anti-inflammatory effect, shown by the TNF-α reduction of a relatively late time-interval (3–4 h after start of digestion). The contribution of ferulic acid to those effects was minor due to its low bioaccessibility. Aleurone seems a promising wheat fraction for cereal products with a healthy added value.  相似文献

The development of anin vitroenzymic batch method for pretreatment of cereal samples prior toin vitrofermentation is presented. A statistical experimental design is described and the importance of bile and pepsin for the digestibility of starch was investigated. Day-to-day variation was studied by measuring glucose released and total starch in the digestion residue of rye flour. Four samples, wholemeal rye flour and bread, wholemeal and white wheat breads, were tested to evaluate the performance of the digestion procedure. Pepsin and bile enhanced the hydrolysis of slowly digestible starch. The extent of starch hydrolysis in rye and wheat breads (93–95%) was reproducible and about the same as using thermostablealpha-amylase. More soluble fibres were released from the wholemeal rye bread than from the wholemeal wheat bread. The procedure is an efficient method for the removal of starch and protein under physiological conditions prior toin vitrofermentation. The method can also be used as a tool for studying the solubility of dietary fibre (DF).  相似文献

Red beetroot has been ranked among the 10 most potent antioxidant vegetables, although only extraction-based methods have been used to evaluate its total antioxidant capacity. Therefore, the present study aims at comparing the traditional extraction-based method with two more recent approaches (QUENCHER -QUick, Easy, New, CHEap and Reproducible- and GAR -global antioxidant response method), in order to establish their suitability in the case of beetroot. Our results indicate that the total antioxidant capacity of beetroot would be underestimated when using extraction-based procedures, since both QUENCHER and GAR methods resulted in a higher total antioxidant capacity. The effect of a thermal treatment on the total antioxidant capacity of beetroot varies among the methods evaluated and our findings suggest different compounds responsible for the total antioxidant capacity detected in each pre-processing method. Remarkably, the present study demonstrates that the traditional extraction-based method seems useful to screen for (changes in) the “bioavailable” antioxidant potential of the root.  相似文献

In this study, bread crust-like systems were prepared by heating (5, 15 and 30 min at 200 °C) freeze-dried fermented doughs obtained using different cereal grains. The flours of whole wheat, refined wheat, whole einkorn, whole rye, whole oat and whole maize were used. The effects of fermentation and heating on bound ferulic acid concentration and total antioxidant capacity were determined. Besides, high molecular weight fractions, containing also melanoidins formed in bread crust-like systems during heating were analyzed for their bound ferulic acid content and total antioxidant capacity. Increasing heating time at 200 °C also increased the amounts of high molecular weight fractions in the bread crust-like systems for all cereals. The total antioxidant capacity of bread crust-like samples increased significantly with heating time (p < 0.05). However, the opposite was true for the high molecular weight fractions of bread crust-like samples. Mean bound ferulic acid concentrations of different cereal flours were found to range between 78.3 mg/kg (for refined wheat) and 667.7 mg/kg (for whole einkorn). Heating had no significant effect on bound ferulic acid contents of bread crust-like systems. For all cereals, the highest concentrations of bound ferulic acid were detected in the high molecular weight fractions of bread crust-like systems heated at 200 °C for 5 min. However, bound ferulic acid concentrations significantly decreased in high molecular weight fractions as heating time increased (p < 0.05).  相似文献

Epidemiological studies have linked whole grain consumption to prevention of several chronic diseases. Whole grain is a source of important phytochemicals, such as ferulic acid (FA). FA is the most abundant phenolic and major contributor to the in vitro antioxidant capacity of wheat grain. Several studies have reported highly variable results on FA bioavailability (0.4–98%). The binding of FA to polysaccharides may limit its bioavailability. Therefore, our study aimed at monitoring release features of FA during gastrointestinal (GI) transit. This was termed bioaccessibility. The bioaccessibility of FA was studied from different wheat fractions and breads with the use of a dynamic in vitro system that simulates the upper GI transit and digestion. The results showed low bioaccessibility of FA from the wheat fractions and breads (<1%). However, the bioaccessibility was high when free FA was added to flour (∼60%). The bioaccessibility of FA appeared to be determined by the percentage of free FA. In wheat grain, most of FA is bound to arabinoxylans and other indigestible polysaccharides restricting its release in the small intestine. New processing developments should be considered to increase free FA in the cereal matrix in order to improve its bioavailability and systemic health effect.  相似文献

Sets of triticale (X Triticosecale Wittmack) lines derived from the cv. Presto with HMW glutenin allele Glu-D1d (subunits 5+10) translocated from bread wheat (Triticum aestivum L.) chromosome 1D to chromosome 1R were evaluated for agronomic and grain quality characteristics in 2002–2005. Two different translocation types were used: (a) single translocation 1R.1D5+10-2 where the long arm of 1R carries the wheat segment from 1DL with the Glu-D1d replacing a secalin locus Sec-3, (b) double translocation Valdy where the long arm of 1R has the translocation 1R.1D5+10-2 and the short arm has a segment from 1DS carrying wheat loci Gli-D1 and Glu-D3. The presence of Glu-D1d was determined by polyacrylamide gel electrophoresis (PAGE-ISTA) and DNA markers. The tested lines of triticale were compared with the check triticale cv. Presto and with wheat cultivars of different bread making quality (E-C quality classes). Single translocation 1R.1D5+10-2 reduced grain yield by 16% and Valdy translocation by 24% as compared with cv. Presto. The Valdy translocation had substantially shortened spike length and reduced specific weight in comparison with check cv. Presto. Wet gluten content (according to the Perten method) was 12% in both translocation types, 8% in check Presto and on average 24% in wheat. Translocations increased the Zeleny sedimentation value (Valdy — 27 ml, 1R.1D5+10-2 – 25 ml, cv. Presto — 23 ml). Triticale had a very low Hagberg falling number (FN) of 62–70 s without significant differences, while wheat had on average 301 s. The translocations did not significantly increase loaf volume; however, they improved loaf shape (height/width ratio): Valdy — 0.61, 1R.1D5+10-2 – 0.56, cv. Presto 0.44, wheat on average 0.70. The dough was non-sticky in Valdy, slightly sticky in 1R.1D5+10-2 and sticky in cv. Presto. Problems with a low FN for improving bread making quality of triticale are discussed. Higher bread making quality can be influenced by appropriate combination with donors of low α-amylase activity.  相似文献

Depolymerization of mixed linkage cereal β-glucans has been observed during the mixing and fermentation of multigrain breads. To maintain the bioactivity of β-glucans in bread and other foods, control of β-glucanase activity during production is needed. β-Glucanase activity in wheat and dairy ingredients was determined by adding them to β-glucan solutions and following the change in the molecular weight of β-glucan at 50 °C for 60 min. The activities in six wheat and six dairy ingredients ranged from undetectable to a 98% reduction in molecular weight. Calcium propionate, potassium sorbate, and sodium benzoate are salts of organic acids commonly used as food preservatives. Low concentrations of these additives (0.1–1.0%) were found to have an inhibitory effect on the β-glucanases found in wheat flour and whey protein isolate. The depolymerization rate of β-glucan was reduced by up to 76% by the three additives.  相似文献

Grain hardness is one of the most important quality characteristics of cultivated bread wheat (Triticum aestivum L.). A large deletion in the puroindoline a (Pina) gene or single nucleotide polymorphisms (SNPs) in the puroindoline b (Pinb) gene results in hard grain texture. So far, nine Pina alleles (Pina-D1aPina-D1b, Pina-D1kPina-D1q) and seventeen Pinb alleles (Pinb-D1aPinb-D1g, Pinb-D1pPinb-D1ab) have been identified in bread wheat. The major Pina and Pinb alleles identified in hard wheat cultivars are Pina-D1b, Pinb-D1b, Pinb-D1c and Pinb-D1d. In this study, a three-primer PCR system was employed to develop nine co-dominant STS markers for genotyping Pina-D1a and Pina-D1b, whereas temperature-switch (TS) PCR was used to develop six co-dominant SNP markers for genotyping the Pinb-D1a, Pinb-D1b, Pinb-D1c and Pinb-D1d alleles. These STS and TS-PCR markers were used to verify the grain hardness genotype of 100 wheat cultivars. The reliability and genotyping accuracy of TS-PCR markers were confirmed through sequencing of PCR products and a comparison with previously published results. Therefore, STS and TS-PCR markers offer a simple, cost-effective and reliable method for high-throughput genotyping Pina and Pinb alleles to select grain hardness in wheat quality breeding programs and for wheat market classification.  相似文献

So far, data on absorption and metabolism of steryl ferulates from edible sources is scarce. Therefore, the impact of enzyme-aided baking and in vitro digestion was examined in this study. Wheat flours and wheat breads were subjected to a static in vitro digestion model and changes in the contents of steryl ferulates and free sterols (possible hydrolysis products of steryl ferulates) were monitored. Baking degraded steryl ferulates at a similar rate in all types of breads (43–47%) compared to the corresponding flours, while baking induced changes in free sterols showed no clear pattern. In vitro digestion provoked five folds lower content of steryl ferulates in flours than in breads and it also resulted in significant free sterol accumulation. Interestingly, bioaccessibility (0.01–0.25%) was not influenced by the cereal matrix. The four steryl ferulate species, which were detected in wheat, showed similar hydrolysis rates during digestion. As baking had a significant impact on the steryl ferulate content of wheat, we suggest that both raw and processed sources should be considered further in vitro, animal or human trials, when studying the metabolic fate of steryl ferulates.  相似文献

Volunteer winter cereals are found sporadically in maize (Zea mays L.) fields across southern Ontario. Seven field trials were conducted over a two-year period (2006 and 2007) at four locations to determine the efficacy of five acetolactate synthase (ALS)-inhibiting herbicides for the control of volunteer cereals applied at two post-emergence application timings (2–4 and 4–7 maize leaf tips). The volunteer cereals were a hard red winter wheat (Triticum aestivum L.) (‘Hyland AC Morley’), soft red winter wheat (‘Pioneer 25R47’), soft white winter wheat (‘Pioneer 25W41’), and a autumn rye (Secale cereale L.) (‘FR’) cultivar. Volunteer cereal competition in maize resulted in a yield reduction of up to 44%. Foramsulfuron, nicosulfuron, nicosulfuron/rimsulfuron provided greater than 70% control of the volunteer cereals at 56 days after treatment (DAT), while primisulfuron and rimsulfuron provided greater than 60% control. Volunteer cereal control with early and late application was greater than 82 and 61%, respectively. Hard red winter wheat was the most sensitive to the ALS-inhibiting herbicides with control of 84–93%. Soft red and soft white winter wheat cultivars were intermediate in sensitivity with control of 76–87%, while autumn rye was the least sensitive with control of at 56–71% control at 56 DAT. Maize yields were improved when volunteer cereals were controlled with the use of herbicides compared to the weedy control, but were lower than the weed-free control. Early herbicide application resulted in improved control of volunteer cereals and higher maize yield.  相似文献

Wheat bread was lyophilised, ground, extracted and centrifuged. The supernatants were analysed for gluten content by RP-HPLC and a commercial sandwich ELISA. Prolamin extraction solvents contained tris(2-carboxyethyl)phosphine (TCEP; 1, 2, 5, 10, 20, 50 mmol/L), guanidine hydrochloride (GUA; 0 or 2 mol/L) and a buffered salt solution. A commercial cocktail solution (250 mmol/L mercaptoethanol (ME), 2 mol/L GUA, buffered salt solution) as well as 60% (v/v) ethanol were used as control solvents. Wheat flour was the control for the extractability of the native prolamins. 60% ethanol only extracted 37% of the prolamins from wheat bread (cocktail = 100%). When ME was replaced by TCEP the protein yield increased from 35% at the lowest TCEP-level to 95% when 20–50 mmol/L TCEP were used. The use of GUA was essential to extract prolamins quantitatively. Comparative protein analysis using RP-HPLC and ELISA showed that both methods provided comparable prolamin (gliadin) concentrations of the wheat flour (40.3–45.7 mg/g), when 60% ethanol was used as extraction solvent. The extraction yields from bread were considerably lower (16.7–24.7 mg/g). Cocktail and TCEP extracted almost the same amount of protein from flour and bread with TCEP showing slightly lower yields. Total extractable protein (gliadin + glutenin) as determined by RP-HPLC was 70.5–75.3 mg/g, and total gliadin as determined by ELISA was 42.7–44.2 mg/g. Thus, the study has shown that TCEP in combination with GUA extracts proteins from heated, gluten-containing foods as effective as the commercial cocktail solution.  相似文献

The quality of bread made from frozen dough is diminished, and staling rate is increased by changes that occur during freezing and storage. New cultivars of waxy wheat flour (WWF), containing higher levels of amylopectin, may help improve the quality of baked products. Bread quality and staling were investigated for bread containing 0–45% WWF and 55–65% water after freezing and 90-day frozen storage. The specific volume was highest with 15% WWF substitution and 60% water in bread made from both unfrozen and frozen dough. With higher levels of WWF and lower water content, bread staling rates decreased. Bread with higher levels of WWF were darker and had greater color variation. 1H NMR studies showed that bread with greater WWF and water had higher transverse relaxation (T2) times (9–11 ms), but less change in T2 during storage. This research demonstrated that specific combinations of WWF and water produced a better quality of bread after dough freezing.  相似文献

The study presents a protocol for the preparation of phenolics-saponins rich fraction (PSRF), a new active nutraceutical from defatted rice bran followed by the determination of its antioxidant properties. PSRF was prepared by employing a simple alcoholic fractionation procedure on the crude alcoholic extract (CAE) of defatted rice bran. PSRF was found to be significantly higher in the contents of total phenolic, saponin, and steroidal saponin than CAE and its counterpart, aqueous fraction (AqF) (p < 0.05). Except for iron chelating activity, PSRF exhibited notably higher activity than CAE and AqF in all antioxidant activity assays performed (p < 0.05). HPLC-DAD analysis revealed that PSRF contained substantially higher amounts of gallic acid, 4-hydroxybenzoic acid, caffeic acid, p-coumaric acid, and ferulic acid than CAE and AqF (p < 0.05). In conclusion, alcoholic fractionation of CAE simultaneously concentrated the phenolic compounds and saponins into PSRF, thus contributed to its higher antioxidant activity. Due to its elevated antioxidant properties, PSRF may be recommended for investigation as an active ingredient in the nutraceutical, functional food, and natural food preservative formulations. This is also the first report suggesting defatted rice bran as a potential and sustainable source of saponins.  相似文献

Antioxidant properties of amaranth extracts isolated sequentially by acetone and methanol/water from defatted plant leaves, flowers, stems and seeds were assessed by ABTS+?, DPPH?, ORAC and total phenols content (TPC) assays. In addition, antioxidant properties of solid plant material were evaluated by the direct QUENCHER method using the same assays. Leaves and flowers of amaranth as well as their extracts possessed the highest antioxidant activities. Radical scavenging capacity in ABTS+? assay for leaves, flowers, stems and seeds evaluated by QUENCHER method were 144.24?±?2.41, 112.33?±?7.45, 19.05?±?1.13 and 21.82?±?1.06 μmol trolox equivalents in 1 g of dry weight, respectively. On-line HPLC-DPPH? assay was used to determine the activity of separated compounds and it was observed that rutin was the main radical scavenger in amaranth extracts. Preliminary screening of extract composition was performed by UPLC/ESI-QTOF-MS and rutin, nicotiflorin, isoquercitrin, 4-hydroxybenzoic and p-coumaric acids were identified by measuring their accurate mass and retention time.  相似文献

Rising prevalence of hypertension is pushing food industry towards the development of innovative food products with antihypertensive effects. The aim was to study the effect of reduced sodium content and 21 % addition of wholemeal wheat sourdough (produced by Lactobacillus brevis CECT 8183 and protease) on proximate composition, γ-aminobutyric acid (GABA) and peptide content of wheat bread. Angiotensin converting enzyme I (ACE) inhibitory and antioxidant activities were also evaluated. Sodium replacement by potassium salt did not affect chemical composition and biological activities of bread. In contrast, GABA and peptides <3 kDa contents in sourdough bread (SDB) were 7 and 3 times higher, respectively, than the observed in control. ACE inhibitory and antioxidant activities of the peptide fraction?<?3 kDa from SDB was 1.7 and 2.6-3.0 times higher than control. Therefore, the combination of reduced sodium content with enriched concentrations of bioactive compounds in bread making may provide interesting perspectives for development of innovative breads towards blood pressure reduction.

Canary seed (Phalaris canariensis) is used to feed birds but it has been recently considered a promising cereal with nutraceutical potential for humans. The aim of this work was to analyze the protein fractions from canary seed flour and from milk substitutes (prepared by soaking the seeds in water 12 and 24 h), and to evaluate antioxidant and antihypertensive capacity of peptides obtained after in vitro digestion. Prolamins were the major protein fraction, followed by glutelins. After digestion, albumins and prolamins fractions from milks presented higher levels of peptides than flour, globulins showed more peptides in flour and glutelins were found in similar concentrations in all samples; 24 h milk prolamins had the highest concentration of peptides. Purification by high performance liquid chromatography (HPLC), sequencing of peptides, in vitro antioxidant ABTS (2,2′-azino-bis, 3-ethylbenzothiazoline-6-sulphonic acid) and DPPH (2,2-diphenyl-1-picrylhydrazyl) assays, and antihypertensive capacity (angiotensin converting enzyme (ACE) assay), indicated that peptides from canary seed prolamins were the most efficient compounds with antioxidant and antihypertensive activity. Canary seeds may be considered an accessible and cheap source to prepare milk substitutes with high contents of bioactive peptides with remarkable functional properties to promote better human health and healthy ageing.  相似文献

The application of technical enzymes is a potential tool in modulating the dough and baking quality of cereal products. No endogenous amylases (α- and β-forms) are present in mature wheat grains; they may be synthesized or activated during germination. Hence, microbial α-amylases are added to the dough, being resistant to the endogenous α-amylase/trypsin inhibitors. Here, we report on the initial identification of two technical enzymes from a commercial sample based on an in-gel tryptic digestion coupled with MALDI-MS analysis. The primary component of the protein fraction with 51.3 kDa was α-amylase from Aspergillus species. A second major protein with 24.8 kDa was identified as endo-1,4-xylanase from Thermomyces lanuginosus. In the following experimental work up, a targeted proteomics approach utilizing the combination of specific proteolytic digestion of the added amylase and xylanase in wheat flour, dough or baked products, solid phase extraction of released peptides and their detection using LC-MS/MS was optimized. The targeted (MRM) MS/MS peptide signals showed that the peptide “ALSSALHER” (MW = 983) originating from amylase and “GWNPGLNAR” (MW = 983) from xylanase can be used to identify the corresponding technical enzymes added. Consequently, locally available baked products were tested and found to contain these enzymes as supplementary ingredients.  相似文献

This study compared the concentration of angiotensin-converting enzyme (ACE) inhibitory peptides at different stages of the bread-making process, including kneading, proofing, and final products. Steamed bread, baked bread, and soda crackers were produced with 3–20% addition of rye malt sourdoughs to assess products differing in their thermal treatment. Eight tripeptides with known or predicted ACE-inhibitory activity were quantified by LC/MS in multiple reaction monitoring (MRM) mode. In wheat sourdough and rye-malt gluten sourdough, IPP was the predominant tripeptide at 58 and 473 μmol kg−1, respectively, followed by LQP, IQP, and LPP. During the bread-making process, peptide concentrations were modified by enzymatic conversions at the dough stage and by thermal reactions during baking. The concentrations of IPP, LPP and VPP remained stable during dough preparation but decreased during thermal treatment; the concentrations of other peptides were changed at the dough stage but remained relatively stable during baking. The cumulative concentration of 8 ACE-inhibitory peptides in steamed bread and bread crumb exceeded 60 μmol kg−1, while soda crackers contained less than 3 μmol kg−1. The peptide levels in bread thus likely meet in vivo active concentrations.  相似文献

From a nutritional perspective, rice flour is one of the most valuable flours and it is suitable for preparing food for people suffering from wheat allergy. However, bread made from rice flour is very difficult to bake because it lacks gluten. We found that pre-fermenting of rice flour using Aspergillus oryzae facilitated a better formulation of gluten-free rice bread. Bread swelling was remarkably improved with a longer pre-fermenting period at 55 °C. The specific loaf volume (SLV) without polymeric thickeners after a 12 h fermentation was approximately 2.2-fold (2.98 ml/g) higher than that after 0 h (1.36 ml/g). An enzymatic assay of the batter indicated that protease activity during the pre-fermentation period increased from 0.38 to 1.44 U/ml and this activity correlated with bread swelling. Furthermore, a commercial protease from A. oryzae also produced similar results with an adequate SLV of 3.03 ml/g. Rheological analysis showed that batter treated with protease had an increased batter viscosity and decreased flour settling behavior because of the aggregation of flour particle after partial cleavage of storage proteins. These results indicated that the improved SLV was mainly because of an A. oryzae protease, which affected the batter rheology thereby improving gas retention before baking.  相似文献
