The effects of seed pretreatment and harvest date on the germination of European rowan (Sorbus aucuparia L) seeds were examined. In one experiment, seeds were subjected to drying, storage, soaking, warm and chilling treatments after harvesting in mid- and late August. In another experiment, fully imbibed (FI) seeds were given warm treatment for six weeks and then various durations of chilling (4?±?1°C) for up to 24 weeks. Thereafter, the seeds were adjusted to target moisture content of 35% and 30% or remained in the FI state and were then subjected to either subsequent chilling or freezing (?3°C) for up to 32 weeks. The treated seeds were allowed to germinate at a constant 15°C with 8 hours of lighting per day. Treatment effects were generally consistent for each harvest date. Seeds did not germinate unless they were chilled and germination rates were low unless the seeds were soaked. Warm treatment applied before chilling appeared to induce dormancy. The effect of storage varied with harvest date and storage treatment, with germination being highest for seeds harvested in late August. Mild freezing of ≥16 weeks duration following 16–24 weeks initial chilling resulted in >80% germination, more than could be achieved using chilling alone.  相似文献

Abstract

The response of common alder [Alnus glutinosa (L.) Gaertn.] and downy birch (Betula pubescens Ehrh.) seeds to germination temperature was examined following chilling and priming. Seeds of two seed lots of each species were subjected to combinations of chilling (4±1°C) and priming (20°C) treatments in fully imbibed (FI) state or a lower target seed moisture content (TMC) level (30% and 35% in alder and birch, respectively). After treatment, the seeds were allowed to germinate for 56 days at constant temperatures of 7.5, 10, 15, 20, 25 or 30°C. The response to temperature and pretreatment differed between species, but the effect was consistent in each seed lot within each species. In alder, the TMC seeds germinated well across the full range of temperatures, whereas there was an optimum temperature (22–23°C) for seeds given the FI pretreatment. Priming had no significant effect on the germination response of the TMC seeds in alder, but priming greatly improved germination in the FI seeds, especially at the lower germination temperatures (optimum 18–19°C). In contrast, in birch, the TMC seeds germinated better across the full temperature range, but the optimal germination temperature (15°C) was the same for all seed pretreatments. Priming improved germination in both the FI and TMC seeds in birch.  相似文献

The effects of seed moisture content (MC), gibberellic acid (GA3) concentration, chilling and priming pretreatments on the germination of common alder (Alnus glutinosa) and downey birch (Betula pubescens) seeds were examined. After treatment, the seeds were allowed to germinate for 42 days at 15 °C or 20 °C (dark)/30 °C (light). Treatment responses were similar at both temperatures and in both species. GA3 treatment of seeds in fully imbibed (FI) state for 30 days, or at the lower, target moisture content (TMC) for 30–90 days, significantly improved germination, but longer treatment periods reduced it (FI seeds) or had no effect (TMC seeds). Priming for two days improved germination in the FI seeds, but more than 4 days reduced it. Priming for up to 14 days had little effect on the germination of the TMC seeds.  相似文献

We investigated the effect of provenance variation on cone and seed morphology and germination behaviour under different pre- treatments in Picea smithiana. Three categories of cones were recognized: large (13.18 cm × 3.30 cm 15.42 cm × 4.35 cm), medium (10.85 cm × 3.93 cm 12.18 cm × 3.98 cm) and small (7.69 cm × 3.06 10.98 cm× 3.39 cm). Significant variation was observed for various cone and seed characteristics. Seeds moisture content varied from 44.48% to 56.91%, seed size from 0.64 cm × 0.31 cm (largest seed) to 0.49 cm × 0.10 cm (smallest seed), and the seed weight from 2.45 g per 100 seeds to 1.36 g per 100 seeds on fresh weight basis of P. smithiana. The highest seed germination (72.0±7.53%) at 10°C was observed under chilling treatment in Tapovan provenance, while the minimum (15.0±5.71%) seed germina- tion was recorded at 25°C under control set in Tapovan provenance. Further among all the sources Pandukeshwar consistently had the highest average (38%) percent germination across all treatments. Compared to other temperature regimes and pretreatments, seeds subjected to 10°C and chilling treatment had the highest germination and took the least time for germination irrespective of provenance effects. Altitude (provenance) had little or no relationship with germination following laboratory treat- ments. Seeds from Tapovan provenance subjected at 10°C following chilling treatment may be suggested for further multiplication of this species.  相似文献

The germination of common alder (Alnus glutinosa (L.) Gaertn.) and downey birch (Betula pubescens Ehrh.) seeds is often poor in bare-root nurseries. The effect of a variety of seed coverings and a few seed pretreatments on seedling emergence was examined in this study in an attempt to address this problem. Seeds of each species were sown in trays containing nursery soil, covered with grit, gravel, sand, combinations of these coverings, a hydromulch or a sealed plastic cloche and then incubated for 6 weeks at 17–20°C. The grit combined with sand or gravel, the hydromulch and the cloche increased seedling emergence when compared with the standard grit. In another experiment, seeds of each species were fully imbibed (FI) (>50% moisture content, MC), as per standard practice, or adjusted to target MC (TMC) (30–35% MC) levels, and then chilled to release dormancy. Some seeds of each MC treatment were primed at 20°C for 2 days following chilling, after which all seeds were evaluated in laboratory tests and a nursery trial. Germination potential of the FI seeds declined in the lab tests by the second test date, which was reflected in low seedling emergence in the nursery in birch. The primed FI seeds of alder germinated most rapidly in the nursery, but other effects were not significant. Seedling emergence was better in the nursery in response to the TMC than the FI pretreatment in birch.

Seeds of Strychnos nux-vomica have slow and erratic germination; thus different presowing treatments were applied to enhance the germination of its seeds collected from Tamaraikulam, Tamil Nadu, India. In addition, the effects of desiccation and different storage conditions on the germination of S. nux-vomica seeds were investigated. The results show that soaking in 500 ppm gibberellic acid (GA3) for 24 h, incubation of seeds at 40 °C for 3 days and alternate water soaking (16 h) and drying (8 h) for 14 days significantly increased the percentage germination compared to the control. Desiccation of seeds down to 10% moisture content resulted in better germination. Germination of S. nux-vomica seeds differed significantly between different storage periods, moisture contents of the seed and for first and second order interactions (p<0.001). The highest germination (92%) was achieved when seeds with 10% moisture content were stored at ambient temperature for 30 weeks. Evidence from the present study indicates that S. nux-vomica seeds possess physiological dormancy that can be broken effectively by after-ripening. As seeds of S. nux-vomica are found to be desiccation tolerant, dry seed (10% moisture content) can be hermitically stored at ambient temperature for 30 weeks without losing their viability.  相似文献

The effect of seed moisture content (m.c.) and seed storage conditions of bay laurel (Laurus nobilis L.) was investigated in relation to seed viability. In the first experiment, the effect of drying rate on seed moisture and seed germination was investigated. Fresh seeds, with their original moisture content displayed a germination percentage of 55.1%. When the seed moisture content was reduced by 2.0% in an oven, the germination percentage rose to 81.0%. When the seed moisture content was reduced even more by using the same method, the germination percentages decreased dramatically. Reducing the seed moisture content to 28.7 and 23.5% by drying the seeds in alternating room conditions resulted in an increase of seed germinability to 84.3 and 90.9%, respectively. The drying of the seeds for 45, 60 and 75 days reduced their seed germination to 66.8, 49.4 and 48.0%, respectively. Reducing seed moisture content below 15.0% resulted in practically nullifying seed germinability. The fact that bay laurel seeds cannot retain their germinability at lower moisture contents demonstrates that it is a species with recalcitrant seeds. In the second experiment, moist and dry storage conditions were tested under different temperatures and moisture contents. The storage experiment showed that the most effective way of conserving the bay laurel seeds is moist storage at 0 ± 1°C for 4 months without previous drying of the seeds.  相似文献

Ponderosa pine (Pinus ponderosa Dogl. ex. Laws.) is the most planted conifer species in the forest-steppe ecotone of Patagonia, Argentina, because of its adaptability and excellent growth rates. In spite of this, and the increasing demand for this species, local commercial seed lots showed low quality, making hazardous seedling production. Aiming to upgrade germinability of a local seed lot, we set an experiment to determine the duration of the prechilling period (0, 10, 20, 30, 40 and 60 days) that promoted the highest seed germination speed (GE) and percentage (GP). Moreover, part of that lot was IDS treated, in an attempt to separate empty and dead filled seeds from viable seeds. Results showed that after 40 days prechilling, GE reached 62%, and GP 70%, both higher than the values obtained under customary conditions. The application of the IDS technique, after 40 days prechilling, 8 h drying at ambient conditions (16 ± 2°C and 50% HR), and 25% seed moisture content (mc), increased GE and GP to 68% and 89%, respectively. Optimal prechilling periods and the application of the IDS technique successfully improved germinability of ponderosa pine seeds from Patagonian stands.  相似文献

Seeds of Juniperus procera collected from five provenances across its geographic range in Ethiopia were subjected to cold-moist stratification at 5°C or 10°C for 6–12 weeks. The effect of aqueous smoke solution in overcoming the light requirement for germination, and the potential of visible (VIS) and near infrared reflectance (NIR) spectroscopy for sorting sound and insect-damaged seeds were also investigated. Highly significant differences in germination were detected among provenances (P < 0.0001) and stratification periods (P < 0.0001), but not between temperature regimes (P=0.111). Seeds from the south and southeast distribution ranges had higher percentage germination after 6 weeks of stratification than seeds collected from north, northwest and central ranges of distribution. The smoke treatment did not affect germination regardless of whether the seeds were exposed to light. Exposure to light increased germination capacity three fold. Sound and damaged seeds were distinguished with 90% accuracy using VIS + NIR spectroscopy. It can be concluded that dormancy in juniper seeds varies with provenances, and cold stratification for 6 weeks alleviates dormancy in some seed lots. Tentatively, smoke treatment seems ineffective in overcoming photo-dormancy in juniper seeds. VIS + NIR spectroscopy has demonstrated a great potential for sorting damaged seeds, thereby upgrading seed lot purity.  相似文献

The effect of harvest time on seed germination, desiccation tolerance and level of dormancy is not known in Sorbus mougeotii (Soyer-Will. & Godron). Dry weight (DW), moisture content (MC) and leakage conductivity (LC) were tested on seeds from six harvest times. Chilling requirement and germinability were tested on seeds without drying and after drying and rehydration. The effect of postharvest ripening of seeds with or without drying was also tested. The DW was stable during the last 6 weeks of maturation, whereas MC initially decreased but later increased simultaneously with an increase in LC. Seeds were desiccation tolerant, deeply dormant and after a chilling treatment germinated to a high percentage at all harvest times. The chilling requirement decreased significantly with later harvest, suggesting a decrease in level of dormancy. Drying and postharvest ripening reduced and synchronized the chilling requirement.  相似文献

Acer velutinum Boiss is a valuable tree species native to Iran, and its seeds possess physiological dormancy that hampers seedling production in the nursery for large-scale reforestation efforts. The aim of this study was to determine the optimal dormancy breaking treatments for A. velutinum seeds. We conducted a factorial experiment involving six seed lots collected along an elevation gradient from 300 to 1800 m at 300 m interval and four cold-moist stratification periods (0, 4, 8 and 16 weeks) at 4℃ and 70% relative humidity. The result shows that the germination of cold-moist stratified seeds was significantly (p < 0.0001) higher than the control for all seed lots. The highest germination capacity was recorded after 16 weeks of cold-moist stratification for all seed lots (68% 88% depending on the seed lot) except those collected from mid altitude sites (600 and 900 m) that germinated equally well (≥ 75%) after 4and 8-week of clod-moist stratification compared to the other seed lots. The mean germination time was significantly shorter (12 to 19 days, depending on the seed lot) for seeds stratified for 16 weeks than for untreated seeds. It can be concluded that: (1) cold-moist stratification for 16 weeks is the best pre-sowing treatment for breaking dormancy in A. velutinum seeds; and (2) seeds should be collected from mid altitude sites (600 and 900 m) to get more than 80% germination within 15 days, and these seed lots even required shorter cold-moist stratification period(eight weeks) than other seed lots.  相似文献

The aim of this study was to determine the efficiency of methods for the artificial scarification of black locust seeds based on the influence of the air and liquid nitrogen temperatures. Three populations were selected for the analyses: a managed tree stand, a selected seed stand and a seed orchard. Samples of 200 seeds (4 × 50) were randomly selected. Before scarification, the seeds were stored at ?70 °C for 24 h. Nine different scarification treatments were evaluated. To calculate the germination capacity and the dynamics of germination, observations were made on days 3, 5, 7, 10 and 14 after treatment. For statistical analyses, we used repeated measures analysis of variance. The obtained results indicated that the highest scarification efficiency for black locust seeds occurred in the treatment in liquid nitrogen and in an air temperature range of 85–90 °C. The provenance of seeds had a significant effect on the dynamics of germination. Thermal methods and the use of liquid nitrogen can be alternatives to the labour intensive methods of cutting the seeds or using measures that require specialised equipment.  相似文献

Intact seeds from freshly harvested fruits of Myrica rubra (Sieb et Zucc.) were dormant and required 8 weeks of warm stratification followed by 12 weeks of cold stratification for germination. Exogenous application of gibberellic acid (GA(3)) to intact fresh seeds was effective in breaking dormancy, with > 70% of seeds germinating when treated with 5.2 mM GA(3) and incubated at a day/night temperature of 30/20 degrees C for 20 weeks. Removing the hard endocarp or endocarp plus seed coat of fresh seeds promoted germination, and addition of GA(3) to the embryo accelerated germination. The gibberellins GA(1) and GA(4) were more effective than GA(3) in promoting germination of seeds with the endocarp removed. Endogenous contents of GA(1), GA(3), GA(4), GA(7) and GA(20) were quantified by gas chromatography-mass spectrometry-selected ion monitoring in the endocarps, seed coats and embryos of fresh seeds treated with 5.2 mM GA(3). The content of GA(3) decreased in the endocarp during incubation, whereas GA(1) contents increased in the endocarp and seed coat. A high GA(1) content was detected in the endocarps and embryos of newly germinated seeds. We speculate that GA(3) was converted to GA(1) during incubation and that GA(1) is involved in seed germination. Endogenous abscisic acid (ABA) contents were measured in fresh seeds and in warm and cold stratified seeds. The ABA content in fresh seeds was distributed in the order endocarp > seed coat > embryo, with the content in the endocarp being about 132-fold higher than in the seed coat and embryo. Total ABA content of seeds subjected to warm or cold stratification, or both, was 8.7- to 14.0-fold lower than that of fresh seeds. Low contents of endogenous GA(1), GA(3), GA(7) and GA(20), but elevated contents of GA(4), were found in the seed coats and endocarps of warm plus cold stratified seeds and in the seed coats and embryos of newly germinated seeds. These observations, coupled with the finding that GA stimulated germination of dormant Myrica seeds, provide evidence that endogenous ABA inhibited release of dormancy and that endogenous gibberellins, especially GA(4) or GA(1), or both, are involved in germination.  相似文献

Mass maturity (end of the seed-filling phase) occurred at about 72 days after flowering (DAF) in developing seeds of Mimusops elengi, at which time seed moisture content had declined to about 55%. The onset of ability to germinate was detected at 56 DAF and seeds showed 98% germination by 84 DAF. Tolerance of desiccation to 10% moisture content was first detected at 70 DAF and was maximal by 84 DAF. Delaying collection by a further 14 days to 98 DAF, when fruits began to be shed, reduced seed viability, particularly for seeds first dried to 10% moisture content. Hence the best time for seed collection appears to be about 14 days before fruits shed. In a separate investigation with six different seed lots, desiccation below about 8–12% moisture content reduced viability (considerably in some lots). The viability of dry seeds (below about 10% moisture content) stored hermetically was reduced at cool temperatures (5 °C and below), and none survived storage at sub-zero temperatures. The results suggest that Mimusops elengi shows intermediate seed storage behaviour and that the optimal hermetic seed storage environment is about 10% moisture content at 10 °C, while short-term, moist, aerated storage at high (40%) moisture content is also feasible.  相似文献

本研究设置干燥、布袋与保鲜袋包装以及系列温度处理,开展了3a西南桦种子贮藏试验。结果表明:布袋与保鲜袋处理对西南桦种子贮藏影响极不影响;在常温常规条件下,西南桦种子贮藏3个月即失去生活力,而在常温干燥条件下,西南桦种子贮藏10个月种子发芽率尚未显著下降;在15℃条件下贮藏10个月几乎丧失发芽能力,非常有趣的,西南桦种子在10、5、0℃和-5℃各温度条件下贮藏3a,效果十分理想,10℃似乎是西南桦种子低温贮藏的临界温度。本研究促进了西南桦的扩大栽培和基因资源的保存。  相似文献

The objective of the investigation was to describe moisture conditions and germination of Scots pine (Pinus sylvestris L.) seeds in different seedbed substrates (feather mosses, mineral soil, humus and ground organic material) with and without irrigation during 2 weeks, in a clearcut in northern Sweden. Without irrigation no seeds germinated, which could be explained as an effect of the low moisture conditions of the seedbed substrates. This was shown in the seeds' low moisture content, with highest values in mineral soil, 10-20%, and for the other seedbed substrates 5-15%. The moisture conditions were highest during the night and lowest in the afternoon. With irrigation, moisture conditions improved (seed moisture content 20-50%) and diurnal changes were moderated. Seeds germinated in all substrates with most seedlings in mineral soil (60% of sown germinable seeds). The moisture conditions of the seedbed substrates proved to be in poor agreement with the seeds' requirement for germination, especially during warm weather with no precipitation.  相似文献

We performed desiccation and storage trials to better under- stand storage behavior of Knema attenuata seeds. Mature seeds with moisture content (MC) of 31% exhibited 73% germination. During the period of desiccation (open lab condition) seeds with MC 23% showed 40% germination. After further drying to MC 21% germination was reduced to 16%. Complete loss in viability resulted when seed moisture was reduced to 18%. The seeds stored at -10°C, 0°C, 10°C and 28±2°C (open lab condition) lost their viability within 10 days. Seeds stored in sealed polythene bags and moist sand retained viability for more days than did seeds stored under all other storage conditions. Sensitivity of seeds to lower temperature and desiccation suggest that the storage be- havior of K. attenuata seeds is recalcitrant. Seeds stored in moist condi- tions can, at best, be stored for a period of two months.  相似文献

The germination of freshly collected Juniperus polycarpos (K. Koch) seeds is very low and attributed to the large proportion of nonviable seeds in the seed lots. Thus, the aim of this study was to improve seed lot quality by removing nonviable seeds using two flotation techniques: incubation-drying-separation (IDS) and modified specific gravity (MSG) separation. We examined different IDS conditions (the specific incubation time, subsequent drying time and sorting media) for effectively sorting out nonviable seeds; and tested the feasibility of modified SG separation, which involved soaking seeds in water for a certain period before sorting in different concentrations of sucrose solution. Viable seeds were expected to absorb and metabolically bind more water during soaking than dead seeds could, and hence be sorted effectively depending on the viscosity of the sucrose solution. The viability of the seeds that floated or sank was determined in a topographical tetrazolium chloride (TTC) test. For the IDS trial, 7 days incubation followed by 9 h of drying and sedimentation in pure water or 200 g·L?1 sucrose solution identified, respectively, 75 and 82 % of the seeds as viable (sunken) seeds. For the MSG trail, 77 % viable seeds were recovered in the sunken fraction when seeds were soaked for 48 h then immediately sorted in 600 g·L?1 sucrose solution. In both cases, the loss of viable seeds in the discarded floating fraction was only 4 %. The results demonstrate that both IDS and MSG separation techniques substantially improved seed lot viability, but MSG separation is simple and needs no modern seed handling facility, and its efficacy relies on seed mass (due to the initial soaking) and the specific density and viscosity of the flotation medium.  相似文献

We examined the effect of thermal treatment at two temperatures (60 s of 90°C dry heat and 60 min in 40°C hot water) versus control conditions on the germination of oriental spruce (Picea orientalis (L.) Link.) seeds originating from different subalpine provenances. Cutting test results showed that there was wide variation (between 10% and 90%) in filled seed rates. The results revealed that highest germination yield was attained via the hot water treatment. The maximum germination percentages were observed in seeds from “tree 3” in the first tree collective of provenances 1 (85.33 ± 5.77 control; 88.67 ± 7.02 via the hot water treatment; 82.00 ± 10.58 via the dry heat treatment) and 3 (70.67 ± 17.24 control; 88.00 ± 4.00 via the hot water treatment; 86.67 ± 11.37 via the dry heat treatment). The mean germination percentages were significantly different (p-value < .05): percentage values were 32.57% in provenance 1, 23.22% in provenance 3, 12.25% in provenance 4, and 2.82% in provenance 2. Mean germination time of seeds from different provenances and experiencing various pretreatments showed significant variation. The best pretreatment increase in germination rates was achieved via the wet heat method (60 min in 40°C hot water).  相似文献
