The objective of this study was to examine the health and meat production of cloned sows and their progenies in order to demonstrate the application of somatic cell cloning to the pig industry. This study compared the growth, reproductive performance, carcass characteristics and meat quality of Landrace cloned sows, F1 progenies and F2 progenies. We measured their body weight, growth rate and feed conversion and performed a pathological analysis of their anatomy to detect abnormalities. Three of the five cloned pigs were used for a growth test. Cloned pigs grew normally and had characteristics similar to those of the control purebred Landrace pigs. Two cloned gilts were bred with a Landrace boar and used for a progeny test. F1 progenies had characteristics similar to those of the controls. Two of the F1 progeny gilts were bred with a Duroc or Large White boar and used for the progeny test. F2 progenies grew normally. There were no biological differences in growth, carcass characteristics and amino acid composition among cloned sows, F1 progenies, F2 progenies and conventional pigs. The cloned sows and F1 progenies showed normal reproductive performance. No specific abnormalities were observed by pathological analysis, with the exception of periarteritis in the F1 progenies. All pigs had a normal karyotype. These results demonstrate that cloned female pigs and their progenies have similar growth, reproductive performance and carcass quality characteristics and that somatic cell cloning could be a useful technique for conserving superior pig breeds in conventional meat production.  相似文献

体细胞核移植技术可以成功用于克隆性能优良的种用家畜,理论上克隆个体与供体在遗传上是完全相同的。为验证该理论,本试验检测了克隆种公猪自身在出生后的生长发育指标以及克隆种公猪与供核种公猪精液品质各项指标、情期受孕率、窝产仔数、产活仔数、断奶活仔数、仔猪初生重、仔猪断奶重和平均日增重等指标,探讨了克隆种公猪与供核种公猪在繁殖性能以及遗传性能之间的关系。结果显示,克隆种公猪与供核种公猪在各检测指标之间均不存在显著差异(P>0.05),提示克隆种公猪与供核种公猪在繁殖性能和遗传性能上无显著差异,可以作为种公猪用于扩繁。  相似文献

旨在通过猪体细胞克隆技术,生产乌金猪火毛系,并分析生长发育能力及繁殖性能,为该技术在地方优良猪种保种上的应用提供理论依据。本研究采集了符合乌金猪种质特征的3头公猪和12头母猪的耳组织样品。公猪年龄分别为3、6、11月龄,母猪年龄差距较大,最小的2月龄,最大的10岁。其中3头母猪和1头3月龄公猪已被去势,10岁母猪已无繁殖能力。结果,成功建立了15头乌金猪的耳组织成纤维细胞系,分别选择1头3月龄乌金猪()和10岁乌金猪(♀)的成纤维细胞进行体细胞核移植,经胚胎移植入16头代孕母猪,共获得25头克隆猪,克隆猪生长发育正常,性成熟后进行自然交配共获得39头F1代活仔。本研究通过体细胞克隆技术成功克隆了乌金猪,具备正常的生长发育性能和繁殖性能,为地方猪种的保护研究提供了新的途径。  相似文献

The purpose of this experiment was to study the growth and developmental ability and reproductive performance of cloned Wujin pigs fire hair line by somatic cell cloning technology, and provide a theoretical basis for the application of somatic cell cloning technology in the conservation of local excellent pig breeds. In this study, the ear samples were collected from 3 male and 12 female pigs which met the characteristics of Wujin pigs. The 3 boars were 3, 6 and 11 months old, respectively. The sows had a large age gap, from 2 months old to 10 years old, the three sows and one boar of 3-month-old had been castrated and the sow of 10-year-old had no reproductive capacity. The ear fibroblast cell lines were successfully established from 15 Wujin pigs. Then the fibroblast cells from 3-month-old() and 10-year-old(♀) pigs were used as donor cells for somatic cell nuclear transfer. The cloned embryos were transplanted into 16 surrogate sows and 25 cloned pigs were obtained. The growth and development indexes of these cloned pigs were all normal. After sexual maturity, the male and female cloned pigs mated naturally and 39 live piglets (F1) were obtained. This study successfully cloned Wujin pigs by somatic cell nuclear transfer technology, and cloned Wujin pigs had normal growth and developmental ability and reproductive performance. The study result provides a new way for the conservation of local pig breeds.  相似文献

本试验旨在研究谷胱甘肽过氧化物酶6(glutathione peroxidase 6,GPx6)在大白公猪附睾细胞中的表达和定位,并探究其与公猪繁殖性能的相关性。选取15月龄的公猪和母猪各3头,取睾丸、附睾、前列腺、尿道球腺、精囊腺、卵巢和输卵管,提取蛋白,通过蛋白免疫印迹和免疫组化(IHC)检测组织和细胞中GPx6蛋白的表达和定位;根据评定大白公猪受精能力的数学模型,按照公猪配种胎次≥20胎、3次配种公猪为同一头的标准,筛选并采集符合要求的20头公猪精液,同时,统计相对应的1 279头母猪的生产成绩,计算得到公猪繁殖性能指标(包括窝产总仔数、窝产活仔数、分娩率和繁殖力)。提取精子蛋白和精清蛋白,通过BCA和ELISA检测精子和精清中GPx6蛋白的含量。使用SPSS软件的独立样本t检验及单因素方差分析(one-way ANOVA),进行差异显著性分析,用双变量Pearson进行相关性分析,P<0.05表示差异或相关显著。结果表明,GPx6蛋白在附睾中高表达,IHC结果显示,GPx6蛋白在附睾的顶细胞、基底细胞、晕细胞、主细胞和精子中表达,在肌样细胞中不表达;精清蛋白中GPx6的含量是精子蛋白的7倍,精液中GPx6蛋白的含量与窝产活仔数、窝产总仔数呈负相关关系。结果提示,GPx6在附睾的顶细胞、基底细胞、晕细胞、主细胞和精子中表达,且其在精液中的含量会影响公猪的繁殖性能,这为GPx6对公猪受精能力影响的研究奠定了理论和试验基础。  相似文献

Somatic cell nuclear transfer (SCNT) is a useful technique for creating pig strains that model human diseases. However, production of numerous cloned disease model pigs by SCNT for large-scale experiments is impractical due to its complexity and inefficiency. In the present study, we aimed to establish an efficient procedure for proliferating the diabetes model pig carrying the mutant human hepatocyte nuclear factor-1α gene. A founder diabetes transgenic cloned pig was generated by SCNT and treated with insulin to allow for normal growth to maturity, at which point epididymal sperm could be collected for cryopreservation. In vitro fertilization and intrafallopian insemination using the cryopreserved epididymal sperm resulted in diabetes model transgenic offspring. These results suggest that artificial reproductive technology using cryopreserved epididymal sperm could be a practical option for proliferation of genetically modified disease model pigs.  相似文献

To assess the performance of boars derived by somatic cell cloning, we analyzed various aspects of their reproductive characteristics and the expression of two imprinted genes. Cloned boars (cloned Duroc × Jinhua) were analyzed for birth weight, growth rate, age at first ejaculation, semen characteristics and fertility, in comparison with naturally bred control boars of the same strain. The expression of imprinted genes was analyzed using the microsatellite marker SWC9 for the paternally expressed gene insulin‐like growth factor ‐2 (IGF2) and with single nucleotide polymorphisms (SNPs) for the gene maternally expressed 3 (MEG3). The cloned boars had high production of semen and were nearly equal in level of fertility to conventional pigs; they showed similar characteristics as naturally bred boars of the same strains. The expression of IGF2 was partially disturbed, but this disturbed expression was not linked to a change in developmental fate or reproductive performance. These results indicate that use of cloned boars could be highly effective for proliferation of pigs with desirable characteristics, preservation of genetic resources and risk reduction against epidemic diseases, such as foot‐and‐mouth disease, through storage of somatic cells as a precautionary measure for use in regenerating pig populations after a future pandemic.  相似文献

China is one of the most diverse countries, which have developed 88 indigenous pig breeds. Several studies showed that pigs were independently domesticated in multiple regions of the world. The purpose of this study was to investigate the origin and evolution of Chinese pigs using complete mitochondrial genomic sequences (mtDNA) from Asian and European domestic pigs and wild boars. Thirty primer pairs were designed to determine the mtDNA sequences of Xiang pig, Large White, Lantang, Jinhua and Pietrain. The phylogenetic status of Chinese native pigs was investigated by comparing the mtDNA sequences of complete coding regions and D-loop regions respectively amongst Asian breeds, European breeds and wild boars. The analyzed results by two cluster methods contributed to the same conclusion that all pigs were classified into two major groups, European clade and Asian clade. It revealed that Chinese pigs were only recently diverged from each other and distinctly different from European pigs. Berkshire was clustered with Asian pigs and Chinese pigs were involved in the development of Berkshire breeding. The Malaysian wild boar had distant genetic relationship with European and Asian pigs. Jinhua and Lanyu pigs had more nucleotide diversity with Chinese pigs although they all belonged to the Asian major clade. 旨在通过测定基因组选择选留的大白公猪的后裔生产性能,探究基因组选择实际育种效果。本研究选用913头大白猪构建参考群体,利用ssGBLUP对新出生的823头大白公猪在去势前进行第一次基因组评估,待生产性能测定后进行第二次基因组评估,最终选留10头性能差异显著的公猪留种,比较其后代生长性状表型和育种值及综合选择指数差异。结果表明,两次基因组遗传评估,达100 kg体重日龄、100 kg活体背膘厚和总产仔数3个性状基因组育种值(GEBV)估计准确性分别由0.56、0.67和0.64提高至0.73、0.73和0.67,两次基因组选择基因组母系指数相关系数为0.82,表明在去势前进行公猪基因组选择具有较高的准确性,可实现种猪早期选择。根据各性状GEBV和基因组母系指数,10头公猪被划分为高、低生产性能组,后裔测定成绩表明,两组公猪后代100 kg体重日龄表型均值之差为2.58 d,育种值之差为3.08 d,100 kg活体背膘厚表型均值之差为1.15 mm,育种值之差为1.03 mm,综合母系指数均值之差为9.3,除后代100 kg体重日龄表型均值之差外,其他差异均达到极显著水平。本研究证明,在基因组评估中具有显著差异的公猪其后代在表型值和育种值等方面均存在显著差异,通过基因组选择能够挑选出优秀种公猪,可将其遗传优势传递给后代。  相似文献

This study aimed to evaluate the actual genetic improvement effect of genomic selection in Large White boars through progeny testing in production performance. Nine hundred and thirteen Large White pigs were used to construct a reference group, and 823 new-born Large White boars were used to implement the first genomic selection through ssGBLUP before castration. The second genomic selection were carried out after performance testing, then 10 boars with significant difference in production performance were selected and their offsprings were compared in phenotypic values, estimated breeding values of growth traits and selection index. The results showed that the accuracies of genomic prediction on age at 100 kg body weight, 100 kg backfat thickness and total number born increased from 0.56, 0.67 and 0.64 in the first genomic selection to 0.73, 0.73 and 0.67 in the second genomic selection, respectively. The correlation coefficient of maternal selection index between the two genomic selection before castration and after performance testing was 0.82, which indicated that the first genomic selection before castration was accurate enough to make early selection on boars. According to the genomic breeding values and maternal selection index of 10 selected boars, two groups with high and low production performance were set up. The progeny testing showed that the difference of average phenotypic value between groups was 2.58 days, and the difference of average evaluated breeding value(EBV) between groups was 3.08 days in age at 100 kg body weight, those were 1.15 mm and 1.03 mm in 100 kg backfat thickness, respectively, and the difference in the mean of the comprehensive maternal index was 9.3, all the differences(except age at 100 kg body weight) were extremely significant. This study prove that the offspring of boars with significant differences in genomic evaluation have significant differences in phenotypic values and breeding values, which indicate that, through genomic selection, excellent breeding boars can be selected and their genetic superiority can be passed to their offsprings.  相似文献

试验旨在研究前列腺素内过氧化物合酶2(prostaglandin-endoperoxide synthase 2,PTGS2)基因的单核苷酸多态性,并将其与大白猪的繁殖性状进行关联分析,为猪分子遗传标记提供依据。以美系及法系纯种大白猪基因组DNA为模板,构建DNA混合池,通过克隆测序比对,检测猪PTGS2基因的遗传变异;采用PCR-RFLP技术对多态性位点进行基因分型,并与目标性状进行关联分析。结果显示,在猪PTGS2基因外显子2上存在1个g.86A>G碱基突变,并具有Msp Ⅰ酶切位点多态性,且在大白猪群体中检测到AA、AG和GG 3种基因型;哈迪-温伯格平衡检验结果显示,在法系大白猪群体中,PTGS2基因g.86A>G多态性分布达到遗传平衡状态(P>0.05)。关联分析结果表明,在美系大白猪群体中,AA基因型初产母猪弱仔数显著少于AG基因型(P<0.05),AA基因型经产母猪初生窝重显著高于AG基因型(P<0.05);在法系大白猪群体中,GG基因型个体总产仔数、健仔数和初生窝重均高于AA和AG基因型,但未达到显著水平(P>0.05)。PTGS2基因外显子2 g.86A>G多态性位点显著影响初生窝重和弱仔数,可作为猪分子标记辅助选择育种的潜在位点。  相似文献

Somatic cell cloning is expected to be a valuable method for conserving genetic resources in pigs. In this study, we compared the reproductive and growth performance of Jin Hua cloned pigs with that of naturally bred Jin Hua pigs. In addition, we generated offspring from the cloned sows and examined the productivity and quality of meat in the progeny. The birth weights and growth rates of somatic cell-cloned pigs were similar to those of Jin Hua pigs. The cloned pigs reached puberty very early, and this is typical of the Jin Hua breed. Furthermore, reproductive performance, in terms of traits such as gestation period, litter size, and raising rate in the cloned pigs were similar to Jin Hua pigs. Although the offspring of the cloned (OC) pigs had lower birth weights than the Jin Hua breed, the daily weight gain of the OC pigs was significantly higher, especially at the finishing stage. The carcass quality of the OC pigs had similar characteristics to the Jin Hua breed, namely thick back fat and a small loin area. Furthermore, the meat qualities of the OC pigs were similar to those of Jin Hua pigs in terms of intramuscular fat content and tenderness. These results demonstrate that cloned pigs and their offspring were similar to the Jin Hua breed in most of the growth, reproductive, and meat productive performances. This strongly suggests that pigs cloned from somatic cell nuclei have the potential to be a valuable genetic resource for breeding.  相似文献

The aim of the study was to determine the relation between the semen quality, frequency of sperm defects, sperm dimensions and shape, and the ejaculate volume of Large White and Landrace boars. A total of 648 ejaculates collected from 31 Large White and 30 Landrace boars were divided into three groups according to the criterion of the ejaculate volume. In this study Landrace boars produced ejaculates with higher volume, sperm concentration, and total numbers of spermatozoa than Large White boars. Landrace boars also showed a lower frequency of sperm with morphological abnormalities (P < 0.05). Landrace boars sperm had larger heads, which were by 0.15 μm longer, and by a larger perimeter and area (P < 0.05). Landrace boar spermatozoa also had a longer flagellum and were generally larger and by 2.07 μm longer than Large White boar sperm (P < 0.05). Significant differences were also found in the shape of sperm of the two breeds (P < 0.05). Landrace boars sperm had more elongated heads, and the ratio of head size to flagellum length was lower than in Large White boars sperm (P < 0.05). Sperm from ejaculates with low volume had a shorter flagellum and a greater head length/flagellum length ratio than sperm from medium- and high-volume ejaculates (P < 0.05).  相似文献

本研究以中国优良地方品种梅山猪为材料,采用胰酶消化法获得猪胎儿成纤维细胞,通过使用Y染色体SRY基因引物SRY-1、SRY-2进行PCR扩增,结果发现,雄性胎儿样本能扩增出250 bp的Y染色体特异性基因片段,而雌性胎儿样本未扩增出此条带;G1、G2、G3、G4和G5代的融合效率差异不显著(P>0.05),但G5代作为供体细胞核移植的重组胚胎卵裂率显著高于G1、G2、G3和G4代(P<0.05);使用G5代的胎儿成纤维细胞作为供核细胞,通过手术移植的方法,将重构胚胎移植到二元后备母猪体内,结果成功地获得了体细胞克隆梅山仔猪,并且仔猪全部为雄性胎儿,说明该性别鉴定方法不但操作简单,而且准确度高;经微卫星DNA 多态性鉴定,确定克隆猪来自供核细胞,与代孕母猪无亲缘关系。本研究将为中国优质猪品种改良、保种、性别控制及建立人类疾病模型等研究提供有效可行的方法,为批量生产克隆优秀种猪提供了坚实的理论基础。  相似文献

试验旨在研究催乳素受体(prolactin receptor,PRLR)和卵泡刺激素β(follicle-stimulating hormone β,FSHβ)基因的多态性及其与母猪繁殖性状的关联分析。采用PCR-RFLP方法对大白猪、杜洛克猪和长白猪3个猪种共487头母猪进行了PRLR和FSHβ基因多态性检验,并采用单因子方差分析、LSD法分析不同基因型与母猪总产仔数、产活仔数、断奶窝重和断奶窝仔数等繁殖性状的相关性。结果表明,PRLR基因在大白猪和长白猪中B等位基因均为优势基因,基因频率分别为0.717和0.548,大白猪BB基因型总产仔数显著高于AA基因型(P<0.05),长白猪BB基因型断奶窝重显著高于AB基因型(P<0.05);FSHβ基因在大白猪、杜洛克猪和长白猪3个猪种中B等位基因均为优势基因,基因频率分别为0.804、0.760和0.789,长白猪BB基因型总产仔数和产活仔数均显著高于AB基因型(P<0.05),呈现BB>AA>AB趋势。因此,PRLR和FSHβ基因对荣昌猪场母猪繁殖性能有一定影响,可作为母猪繁殖性能分子选育的候选参考基因。  相似文献

Monomeric Plum (Plum), a far-red fluorescent protein with photostability and photopermeability, is potentially suitable for in vivo imaging and detection of fluorescence in body tissues. The aim of this study was to generate transgenic cloned pigs exhibiting systemic expression of Plum using somatic cell nuclear transfer (SCNT) technology. Nuclear donor cells for SCNT were obtained by introducing a Plum-expression vector driven by a combination of the cytomegalovirus early enhancer and chicken beta-actin promoter into porcine fetal fibroblasts (PFFs). The cleavage and blastocyst formation rates of reconstructed SCNT embryos were 81.0% (34/42) and 78.6% (33/42), respectively. At 36–37 days of gestation, three fetuses systemically expressing Plum were obtained from one recipient to which 103 SCNT embryos were transferred (3/103, 2.9%). For generation of offspring expressing Plum, rejuvenated PFFs were established from one cloned fetus and used as nuclear donor cells. Four cloned offspring and one stillborn cloned offspring were produced from one recipient to which 117 SCNT embryos were transferred (5/117, 4.3%). All offspring exhibited high levels of Plum fluorescence in blood cells, such as lymphocytes, monocytes and granulocytes. In addition, the skin, heart, kidney, pancreas, liver and spleen also exhibited Plum expression. These observations demonstrated that transfer of the Plum gene did not interfere with the development of porcine SCNT embryos and resulted in the successful generation of transgenic cloned pigs that systemically expressed Plum. This is the first report of the generation and characterization of transgenic cloned pigs expressing the far-red fluorescent protein Plum.  相似文献

为了解云南热带雨林地区野猪的生物学特性与驯化技术,探讨野猪的规模化养殖与开发利用。通过野外实地调查和收集2~4个月体重约5~8 kg的16头野猪,按试验要求与同等体重家猪人工混养驯化2年时间,将试验中观察的野猪部分生物学特性和驯化方法进行总结,整理成可供推广应用的野猪驯化管理模式。结果表明:西双版纳热带雨林地区野猪适应性强,喜集群活动,驯化后性情和食性会发生改变,可全年发情,提高繁殖性能和生长速度。在增加防护措施和户外运动场所的基础上,可参照家猪饲养方法,对纯种野猪进行家养或将纯种公野猪与母家猪进行杂交利用,开发生产无公害优质野杂猪。  相似文献

随着规模化养猪业的发展及人工授精技术的普及,公猪繁殖性能已是影响猪场经济效益的重要因素。如何准确评定公猪的繁殖能力,保证人工授精效果,进而保证母猪的繁殖性能的发挥,是养猪人工作中的重要环节。生产实践中最常用的是通过显微镜对猪精细胞水平进行检测。科研人员也在一直研究影响人工授精效果的因素,将精液的检测从细胞水平上升至分子水平,再用分子标记手段将公猪繁殖性能由检测提前至预测,最大限度地保证公猪的繁殖性能,保证优良公猪的性能得到最大程度的发挥。文章从精液品质、人工授精效果、分子水平检测、标记物等几个方面介绍公猪繁殖性能评价时需要衡量的指标,为公猪繁殖性能评定提供参考。  相似文献
