The influence of three different environmental salinities (seawater, SW: 38 ppt salinity; brackish water, BW: 12 ppt; and low salinity water, LSW: 6 ppt salinity) on the growth, osmoregulation and metabolism of young gilthead sea bream (Sparus aurata L.) was studied over a period of 100 days. 480 inmature fish (20 g mean body weight) were randomly divided into six tanks of 2500 l (80 fish per tank) and maintained under three different salinities (38 ppt, 12 ppt and 6 ppt) in an open system. Every three weeks, 10 fish from each tank were anesthetized, weighed and lenghed. At the end of experiment, 10 fish from each tank were anesthetized, weighed and sampled for plasma, brain, gill and liver. Gill Na+, K+-ATPase activity, plasma osmolality, ions (sodium and chloride), glucose, lactate, protein and triglyceride, and hepatosomatic index were examined. In addition, levels of glycogen, lactate, ATP and activities of potential regulatory enzymes (hexokinase, pyruvate kinase, glycogen phosphorylase, and glucose 6-phosphate dehydrogenase) were assesed in liver, brain, and gill. BW-acclimated fish showed a better growth with respect to SW- or LSW-acclimated fish (12 > 38 > 6 ppt). The same relationship was observed for weight gain and specific growth rate. Osmoregulatory parameters in plasma (osmolality, Na+ and Cl levels) were similar in SW- and BW-acclimated fish but significantly higher than those of LSW-acclimated fish. Gill Na+, K+-ATPase activity showed lower values in intermediate salinity (6 > 38 > 12 ppt). No changes were observed in metabolic parameters analyzed in plasma, whereas only minor changes were observed in metabolic parameters of liver, gills and brain that could be correlated with the higher growth rates observed in fish acclimated to BW, which do not allow us to attribute the best growth rate observed at 12 ppt to lower metabolic rates in that salinity.  相似文献

Localization of Na+, K+-ATPase-rich cells in the gill and urinary system of Acipenser persicus fry was performed through immunofluorescence light microscopy using a mouse monoclonal antibody IgGα5 raised against the α-subunit of chicken Na+, K+-ATPase. Different types of epithelia were clearly identified in the gill epithelium: epithelia of branchial arch, interbranchial septum, filament and lamellar epithelium. The Na+, K+-ATPase-rich cells were found in the epithelia of branchial arch, interbranchial septum, filament, interlamellar region and also in the lamellae. Histologically, the urinary system is divided into head kidney, trunk kidney and short caudal kidney. The head kidney is composed of the pronephric tubules and the haemopoietic tissues, while the trunk kidney is composed of a large number of glomeruli and convoluted nephrons. Each nephron consisted of a large glomerulus and tubules (neck, proximal, distal and collecting tubules) which connected to ureters. Posteriorly, ureters extended and joined together to form a small urinary bladder. In the urinary system, no specific fluorescence staining was observed in the glomerulus, neck segment and proximal tubules. The distal tubule cells and collecting tubule cells showed a strong immunostaining of Na+, K+-ATPase. Epithelia of ureters and urinary bladder also showed several isolated immunofluorescent cells. Immunofluorescent cells were rich in Na+, K+-ATPase enzyme which is very important for osmoregulation.  相似文献

Abstract –  Downstream movement of a wild population of brown trout was examined in a small Danish stream in relation to morphological and physiological smolt status from March to May. Downstream movement was monitored in a Wolf-type trap covering all possible passage routes in the stream. Trout caught in the trap were classified as parr, pre-smolt or smolt based on morphological criteria and compared with trout randomly caught by electrofishing upstream of the trap. Representative gill samples from trap-caught and electrofished trout were analysed for gill Na+, K+-ATPase activity and used as a measure of physiological smolt status. Only a few parr occurred in the trap. Few pre-smolts occurred in the trap evenly in March and early April. In late April, pre-smolt movement peaked. By comparison, the main downstream movement of smolts occurred in distinct peaks through late March and April. The majority of fish caught in the trap were judged as pre-smolts or smolts based on morphological criteria's and they were characterised by relatively high gill Na+, K+-ATPase activity compared with trout judged as parr. Trout caught by electrofishing upstream the trap, were classified as parr, pre-smolts and smolts early in the season (March). During and after the main smolt-run in April the distribution of the remaining trout in the brook became skewed in favour of pre-smolt and parr. The study suggests that smolting trout initiate downstream movement once having reached a certain physiological smolt condition (judged by increased gill Na+, K+-ATPase activity).  相似文献

Seasonal variations in osmoregulatory and metabolic parameters were assessed in juvenile gilthead sea bream ( Sparus auratus ) cultured in earthen ponds under a natural photoperiod and temperature. Specimens were sampled, and the plasma, gill, kidney and liver were collected during winter 2005 and 2006 (January), spring 2005 (April), summer 2005 (July) and autumn 2005 (October). Plasma osmoregulatory parameters showed higher values in summer, while metabolic parameters presented different patterns of variations. Gill Na+,K+-ATPase activity decreased significantly in winter, while gill metabolite levels showed different patterns of variations among seasons. The enzymatic activities tested did not present a clear pattern of variation [(glutamate dehydrogenase (EC 1.4.1.2) (GDH) and hexokinase (EC 2.7.1.11) (HK)] or significant differences along seasons [glucose 6-phosphate dehydrogenase (EC 1.1.1.49)]. Kidney Na+,K+-ATPase activity decreased during summer and autumn. Different patterns of variation were observed in kidney metabolite levels while all the enzymatic activities assessed [lactate dehydrogenase-oxidase (EC 1.1.1.27) (LDH-O), HK and GDH] presented the highest values during summer. In the liver, metabolite levels and enzymatic activities did not show significant variations or present clear patterns of variation along different seasons. These results indicated seasonal variations in the osmoregulatory and metabolic parameters of different organs (blood, gill, kidney and liver) in earthen pond-cultured gilthead sea bream ( S. auratus ), which could be mainly attributed to seasonal changes in temperature.  相似文献

The purpose of this study was to determine the effect of clove oil (4.0 mg L−1) sedation, compared with non-sedation, on the primary (plasma cortisol), secondary (osmoregulation) and tertiary (mortality) stress responses in Atlantic salmon smolts during transport and transfer to sea. Clove oil sedation during on- and off-loading sufficiently reduced the primary stress response to lower mortality (2.1%) during transfer to sea compared with unsedated fish, which experienced a mortality rate above 12.2%. The unsedated fish experienced an acute mortality that stabilized only 6 days after the transport. None of the secondary stress responses measured in this experiment could contribute towards explaining this phenomenon, with the possible exception of plasma magnesium (Mg2+). Plasma Mg2+ differed between the groups; while plasma Mg2+ in the clove oil sedated group returned to pre-stress levels 72 h after transport, the unsedated group showed no such recovery even 1 week after transport, which may indicate a disturbance in the hydromineral balance, and provides a plausible explanation for the delayed mortality in this group. Eugenol-based anaesthetics appear to be promising as a stress-reducing sedative for Atlantic salmon smolts, and, if used properly, this chemical could improve animal welfare and survivability during and after common aquaculture-related incidents.  相似文献

A method for measuring protein synthesis has been developed for small juvenile fish. A flooding dose of [3H] phenylalanine (Houlihan et al. 1988b) was added to the water containing the animals. Using juvenile tilapia O. mossambicus of approximately 15 mg, stability of the phenylalanine free pool specific radioactivity was achieved within 10 h. A linear increase in the whole body specific radioactivity of protein-bound phenylalanine occurred 3 h after [3H] phenylalanine was added to the water. In larger animals the flooding dose was injected into the peritoneum, and stable free pool phenylalanine specific radioactivities were achieved over 2 h. In the small fish the rate of protein synthesis exhibited a diurnal pattern. Scaling relationships for growth, protein synthesis and protein degradation were similar. The energy cost of protein synthesis in small juvenile animals, determined from the difference before and after the addition of the protein synthesis inhibitor cycloheximide, was close to the theoretical minimal cost.  相似文献

Abstract— Feed intake, specific growth rate and changes in body composition were studied in age 1+ (140-190 g) brown trout ( Salmo trutta L.) reared at three temperatures (2, 4 and 6°C) under continuous light conditions. Feed in take increased from 35.7 kJ-kg−1.day−1 at 2°C to 95.7 kJ-kg−1-day−1 at 6°C, and the growth rate increased from 0.19%-day−1 to 0.42%-day−1 over the same temperature range. The estimated lower temperature limits for feeding and growth were slightly above 0°C. For all groups of fish, the majority (about 75%) of the weight gain comprised water and protein, but lipid deposition tended to increase with increasing temperature. The deposition of lipid accounted for about 50% of the body energy gain at all temperatures.  相似文献

The influence of dietary cation–anion difference (CAD, Na+ + K+– Cl, mEq kg–1) on energy metabolism and nitrogen losses in juvenile African catfish Clarias gariepinus (Burchell) was examined in fish exposed to different dietary CAD levels (–146, 116, 497, 713 and 828 mEq kg–1 diet). The experiment was conducted in open circuit balance respiration chambers over a 3-week period. Five 24-h monitoring periods were carried out at 3-day intervals during the experimental period with O2 consumption, ammonia and nitrate + nitrite (NOx) and CO2 production being measured at 5-min intervals for each chamber. The negative dietary CAD (–146 mEq kg–1) resulted in the highest energy expenditures (83 kJ kg–0.8· d–1). With increasing dietary CAD levels, heat loss gradually decreased to minimum values of 56 kJ kg–0.8 day–1 at a dietary CAD level of 713 mEq kg–1. Consequently, metabolizable energy utilization efficiency (MEU, percentage of retained energy over metabolizable energy) quadratically ( P  < 0.05) increased and reached a maximum at a dietary CAD of 713 mEq kg–1.  相似文献

Abstract  The distribution and overall biomass of an underfished vendace, Coregonus albula L., population in the mesotrophic Henne Reservoir (Germany) was studied using hydroacoustics and gill nets. Additionally, midwater trawling was carried out. Overall fish biomass, based on five hydroacoustic surveys (June to September 2002), ranged from 188 kg ha−1 in early August to 302 kg ha−1 in September 2002. The overall mean fish biomass was 256 kg ha−1 (±48 kg ha−1 SD). Biomass of fish smaller than 25 cm total length (mostly vendace) varied from 56 kg ha−1 in August to 99 kg ha−1 in September, with an overall mean fish biomass of 74 kg ha−1 (±17 kg ha−1 SD). The echograms showed temporal variation in fish distribution in Henne Reservoir. In June, fish were fairly evenly distributed over the whole reservoir but in September a dense aggregation of fish (mostly vendace) was found in the deeper water layers near the dam. The distribution of vendace stock, its impact on water quality and fisheries management, biomanipulation and effort for mass removal are discussed.  相似文献

ABSTRACT: To examine the involvement of chloride cells in the uptake of Ca2+ in freshwater (FW) killifish, chloride cell morphology was compared in fish acclimated to different defined FW environments with Ca2+ concentrations of either 0.1 mM, 0.5 mM, or 2.5 mM. Numerous chloride cells were detected in whole-mount preparations of the gill filaments, which were stained with an antiserum specific for Na+, K+-ATPase. Chloride cells, located mostly on the afferent–vascular edge of the filaments, were larger at lower Ca2+ concentrations. Electron microscopic observations showed that in the 0.1 mM and 0.5 mM Ca2+ experimental groups, the apical membrane of chloride cells were flat or slightly projecting and equipped with numerous microvilli. In the 2.5 mM Ca2+ group, some chloride cells formed an apical pit, whereas other cells were similar to those observed in the 0.1 mM and 0.5 mM Ca2+ groups. Plasma osmolality decreased with decreasing ambient Ca2+ concentration, suggesting that environmental Ca2+ affects the permeability of the body surfaces. Gill Na+, K+-ATPase activity in the 0.1 mM and 0.5 mM Ca2+ groups were significantly higher than that in the 2.5 mM Ca2+ group, implying the involvement of the Na+–Ca2+ exchanger in Ca2+ uptake in the gills. These findings suggest that chloride cells function as the site for Ca2+ uptake in killifish acclimated to low Ca2+ environments.  相似文献

ABSTRACT: The present study used squid gill as a source of transglutaminase (TGase) because it has extremely high TGase activity compared with other tissues. The enzyme was purified using successive chromatographies of Sephacryl S-300 and hydroxyapatite columns. The yield and purification-fold of the enzymatic activity was 12.6% and 14.1-fold, respectively. The molecular mass of the purified enzyme was estimated to be 94 kDa by using sodium dodecylsulfate–polyacrylamide gel electrophoresis analysis. Enzyme activity was enhanced 15-fold with an increase in NaCl concentration. Although the activity was dependent on Ca2+ concentration, it was not sufficiently activated even by 50 mM CaCl2 in the absence of NaCl, but could be fully activated with 10 mM CaCl2 in 0.7 M NaCl. However, in the absence of substrates, the enzyme was rapidly inactivated. The pH and temperature optima of the enzyme were approximately pH 8.0 and 20°C, respectively. It was stable in the absence of Ca2+ at pH 7.5–9.0 and had a rate constant (K D ) of 1.6 × 10–5 s–1 for thermal inactivation at 50°C. These results in which squid gill TGase could be activated at higher concentrations of Ca2+ and NaCl than at a physiological concentration, suggest that contact with seawater or body fluid seems to activate the enzyme if the tissue is disrupted.  相似文献

Five iso-nitrogenous (300 g crude protein kg−1 diet) semi-purified diets with graded levels of carbohydrate at 220 (D-1), 260 (D-2), 300 (D-3), 340 (D-4) and 380 (D-5) g kg−1 diet were fed ad libitum to Puntius gonionotus fingerlings (average weight 0.59±0.01 g) in triplicate groups (20 fish replicate−1) for a period of 90 days to determine the effect of the dietary carbohydrate level on the growth, nutrient utilization, digestibility, gut enzyme activity and whole-body composition of fish. Fifteen flow-through cement tanks of 100 L capacity with a flow rate of 0.5 L min−1 were used for rearing the fish. The maximum weight gain, specific growth rate, protein efficiency ratio, RNA:DNA ratio, whole-body protein content, protease activity, protein and energy digestibility and minimum feed conversion ratio (FCR) were found in the D-2 group fed with 260 g carbohydrate kg−1 diet. The highest protein and energy retention was also recorded in the same group. However, from the second-order polynomial regression analysis, the maximum growth and nutrient utilization of P. gonionotus fingerlings was 291.3–298.3 g carbohydrate kg−1 diet at a dietary protein level of 300 g kg−1 with a protein/energy (P/E) ratio of 20.58 −20.75 g protein MJ−1.  相似文献

The effect of hydrogen peroxide (H2O2) treatment on the clearance of formalin-killed Flavobacterium branchiophilum from the gills of rainbow trout was examined. In untreated control fish, clearance was characterized by a rapid initial phase, with 50% reduction in bacterial antigen in the first 12 h after exposure. The bacteria then cleared more gradually, with total clearance being achieved by 40 h. Treatment of fish with 100 p.p.m. H2O2 did not influence bacterial clearance compared to untreated controls. Exposure for 1 h to H2O2 concentrations ≤ 100 p.p.m. caused no detectable clinical signs or evidence of ultrastructural damage to the respiratory epithelium. However, levels in excess of this caused significant gill damage.  相似文献

ABSTRACT:   In the present study, the role of carbonic anhydrase (CA) and Na/K-ATPase in the gill and epidermal tissues in the giant freshwater prawn Macrobrachium rosenbergii was examined as a function of molting stage. CA activity levels in the front and back gills were low at the intermolt stage C0, but increased significantly at premolt stage D3, and then decreased after molting. In the epidermal tissue, activity levels decreased gradually towards premolt to a minimum level at stage D3, but became elevated at postmolt stages A and B. Na/K-ATPase levels in the front and back gills did not change significantly during the molt cycle. CA in the gill is possibly involved in supplying counter-ions for ion uptake, while CA in the epidermal tissue may play a role in mineralizing the exoskeleton after ecdysis. Na/K-ATPase in the gills may function in ion uptake from the ambient medium; however, since its activity was not influenced by the molt cycle, it probably does not have a major role in osmoregulation in the freshwater environment.  相似文献

Juvenile yellow perch Perca flavescens were fed semipurified diets with varying protein to metabolizable energy ratios (PME, g protein MJ−1 metabolizable energy) and nutrient densities in three experiments to determine recommended dietary protein and energy concentrations. Experiment 1 fish (18.6 g) were fed diets containing 450 g crude protein kg−1 dry diet and 14.5–18.8 MJ ME kg−1 dry diet for 10 weeks. No differences were found in the growth of experiment 1 fish fed the different diets. Experiment 2 fish (21.9 g) were fed diets containing 15.7 MJ ME kg−1 dry diet and 210–420 g crude protein kg−1 dry diet for 8 weeks. Fish fed the diet containing 340 g kg−1 protein (diet PME = 22) exhibited the greatest weight gain. Experiment 3 fish (27.1 g) were fed diets with a PME of 22 and varying nutrient density (yielding 205–380 g crude protein kg−1 dry diet) for 8 weeks. No differences were found in the growth of experiment 3 fish. Yellow perch fed the semipurified diets exhibited increased liver fat content, liver size and degree of liver discoloration compared with fish fed a commercial fish meal-based diet. Liver changes may have resulted from high dietary carbohydrate levels. We conclude that a protein level of 210–270 g kg−1 dry diet is suitable for juvenile yellow perch provided that the dietary amino acid profile and carbohydrate content are appropriate for yellow perch.  相似文献

An increase in salinity of freshwater can affect the physiology and metal uptake in fish. In the present study, Nile tilapia Oreochromis niloticus were exposed to copper (1.0 mg/l) in increased salinities (2, 4, and 8 ppt) for 0, 1, 3, 7, and 14 days. Following the exposures, the activities of Na+/K+-ATPase, Mg2+-ATPase, and Ca2+-ATPase were measured in the gill, kidney, and intestine to evaluate the changes in osmoregulation of fish. Results showed that increases in salinity and Cu exposure of fish significantly altered the ATPase activities depending on the tissue type, salinity increase, and exposure durations. Salinity-alone exposures increased Na+/K+-ATPase activity and decreased Ca2+-ATPase activity. Na+/K+-ATPase activity decreased following Cu exposure in 2 and 4 ppt salinities, though the activity increased in 8 ppt salinity. Ca2+-ATPase activity decreased in the gill and intestine in all salinities, while the activity mostly increased in the kidney. However, there were great variations in Mg2+-ATPase activity following exposure to salinity alone and salinity+Cu combination. Cu accumulated in the gill and intestine following 14 days exposure and accumulation was negatively correlated with salinity increase. Data indicated that ATPases were highly sensitive to increases in salinity and Cu and might be a useful biomarker in ecotoxicological studies. However, data from salinity increased freshwaters should carefully be handled to see a clear picture on the effects of metals, as salinity affects both metal speciation and fish osmoregulation.  相似文献

Two trials were conducted to assess the effects of repeated prophylactic formalin treatments on the gill structure of salmonids. In trial 1, which involved Atlantic salmon, Salmo salar L., approaching smoltification in a commercial facility, fish were treated with either 167 or 250 mg l-1 formalin for 90 min every 2 weeks for 12 weeks. Formalin-treated salmon had slight, but not significant, increases in the frequency of lamellar fusion, numbers of lamellar mucous cells, and numbers of an endemic gill ciliate, Trichophyra piscium, after 6 and 12 weeks of treatment. In trial 2, which involved juvenile rainbow trout, Oncorhynchus mykiss (Walbaum), fish were treated with 200 mg l-1 formalin for 60 min twice weekly for 12 weeks. Significant effects were limited to an increase in the numbers of mucous cells present on gill lamellae. In both trials, there was no evidence of lamellar oedema or necrosis of lamellar epithelial cells.  相似文献

Four practical diets containing different levels of soybean cake (0, 155, 320, 490 g kg−1) were prepared to investigate the effect of replacement of fishmeal by soybean cake on the Chinese longsnout catfish ( Leiocassis longirostris Günther) during a 62-day growth trial. The diets were isonitrogenous, isoenergetic and contained about 430 g kg−1 crude protein and 20 MJ kg−1 gross energy. With increasing levels of dietary soybean cake, growth rate and feed utilization decreased; feeding rate and the contents of dry matter, fat and energy of fish body were not significantly affected while body protein showed lowest value in the fish fed the diet containing 320 g kg−1 soybean cake.  相似文献
