我国常规稻主栽品种的遗传变异分析   总被引:10,自引:1,他引:9  
 采用40个SSR标记,分析了329份我国近50年来常规稻主栽品种的遗传变异。结果显示,39个SSR标记具有多态性,在多态性位点共检测到223个等位基因,每个位点2~11个,平均57个;平均Nei基因多样性指数(He)为0632。籼粳亚种间的SSR变异差异明显,籼稻平均等位基因数(Na)和Nei基因多样性指数(Na = 54,He = 0440)均高于粳稻品种(Na = 44,He = 0397)。Nei遗传相似系数表明总体样本具有较小的遗传相似度(I = 0366),而骨干亲本具有较高的遗传相似度(籼:I = 0590;粳:I = 0590)。这导致了籼粳亚种内较高的遗传一致性(籼:I = 0558;粳:I = 0600)。早、中、晚稻各类型遗传相似度差异明显,晚籼和早粳类型具有较高的遗传变异。籼粳稻品种尤其是粳稻的聚类结果显示出较强的季节型和地域特征。这些均提示育种家应选择更广泛的亲本源以拓宽选育品种的遗传基础。  相似文献

大麦亲本材料SSR标记遗传多样性及群体结构分析   总被引:2,自引:0,他引:2  
为探明我国大麦亲本材料的遗传背景和群体结构特点,提高大麦种质材料的利用效率,选用50对多态性好的SSR引物对63份大麦亲本材料进行遗传多样性和群体结构分析。结果表明,50对引物共检测到119个等位变异,平均每个位点的等位变异数为2.38个,变异范围为2~5;平均有效等位变异数为1.75,有效等位变异所占比重为74.16%;Shannon’s信息指数和多态信息含量(PIC)的变幅分别为0.082~1.383和0.031~0.701,平均为0.59和0.308。遗传相似系数(GS)变异范围为0.652~0.990,聚类分析表明63个大麦亲本材料在GS值为0.694水平上聚为3个大类,基于数学模型的群体结构可分为4个亚群。本研究涉及的大部分材料亲缘关系较近,需要引入新种质来拓展亲本的遗传基础。  相似文献

川渝地区地方和育成大豆品种SSR标记多样性分析?   总被引:1,自引:0,他引:1  
利用基本均匀分布于大豆20条染色体的135对SSR标记,对232份包括6个地方品种地域亚群和1个育成品种亚群进行全基因组扫描。结果表明:所有的标记都有多态性,所有检测到的位点都是纯合基因型,说明所选用品种高度纯合,每个标记存在2~4个等位变异,平均2.66个。亚群多态信息含量变异范围0.2751-0.3165,整个群体为0.3208;亚群内Nei遗传距离变异范围0.325 8~0.359 4,整个群体为0.3711,说明川渝地区大豆遗传变异较小。亚群间的遗传一致度(GI ≥ 0.8862)较高,亚群间遗传距离(GD ≤ 0.1208)较小,地方品种亚群间遗传差异更小,育成品种亚群与自然地域亚群的遗传差异相对较大。亚群间基因分化系数(Fst)平均为 0.0722,基因流(Nm)平均为 3.214,说明不同亚群之间存在一定的基因交流。主坐标分析表明第一、二和三主成分分别解释总变异的4.97%、3.54%和3.33%。来自同一区域的品种资源基本聚集在同一亚群,聚类分析同样表明同一自然地域亚群品种资源虽不能完全聚集到同一个遗传类群中,但具有一定的聚集效应,说明川渝大豆品种资源遗传变异与地理位置有一定的关系。分子方差分析表明亚群内变异占总变异的97%,亚群间变异仅占总变异的3%。Mantel收敛分析表明地方品种自然地域亚群的遗传距离与所处的地理位置距离(纬度和海拔)呈显著的正相关关系(R2=0.723)。川渝地区大豆种质资源群体遗传丰富度不高,当前的育成品种未蕴含本地区所有遗传变异。  相似文献

 为了揭示云南元阳哈尼梯田两个不同时期种植的水稻地方品种的遗传多样性,以元阳哈尼梯田过去种植(20世纪70年代收集于元阳县,现保存于种质库)的72个和当前种植的76个品种为材料,采用48个SSR分子标记进行多态性、遗传相似性和聚类分析。平均每个标记检测到的等位基因数、有效等位基因数、位点多态信息含量和基因型多样性指标在过去与当前的品种间,差异均未达显著水平。共有24个SSR标记的52个等位基因位点在过去与当前种植的品种中存在差异,其中,有18个SSR标记的32个等位基因位点在当前的品种中未检测到,同样,有16个SSR标记的20个等位基因位点在过去的品种中未检测到。基于48个SSR分子标记的平均遗传相似性系数,当前种植的品种为03340,过去为03313,过去与当前的品种分别在遗传相似性系数0203和0174处分为籼、粳两个亚种群。表明元阳哈尼梯田水稻地方品种经过30多年的自然与人工选择,虽有部分等位基因位点发生了变异,但仍保留了原有的遗传多样性;元阳哈尼梯田水稻地方品种存在明显的籼、粳分化。元阳哈尼梯田及其水稻地方品种是研究稻作籼粳演化的理想基地和材料。  相似文献

本研究以返回式卫星搭载花生品种粤油7号种子为材料,经过3年地面种植和选择,获得21个变异株系。选用110对SSR引物对21个变异株系进行SSR多态性分析,有5对引物的扩增在变异株系与对照品种之间表现出多态性。说明太空能够诱导花生种子发生基因变异,空间诱变可作为花生种质资源创新和品种选育的方法之一。  相似文献

19个亚洲国家大麦种质材料的遗传多样性分析   总被引:1,自引:0,他引:1  
为了解大麦种质材料的遗传多样性,并为合理选择育种亲本及保护与利用大麦种质资源材料提供依据,采用21对SSR荧光标记引物对来自19个亚洲国家的175份大麦种质材料进行遗传多样性分析,筛选出的19对SSR引物共检测到134个等位变异,平均每对引物检测到7.05个等位变异,其中SSR17位点对亚洲大麦基因组DNA变异检测最有效,其多态性信息含量指数(PIC)和标记指数MI均最高,分别为0.9和10.77;基于SSR数据,对19个亚洲国家175份大麦材料进行UPGMA聚类分析,结果聚为三组:组群Ⅰ共包含19个国家的161个品种;组群Ⅱ共包含4个国家的12个品种;组群Ⅲ只包括来自土库曼斯坦的2个品种。根据按国家划分的19个群体的遗传一致度数据,聚类分析可以分为3组群(其中组群1可分为4个亚组),主坐标分析可分为3个组群和7个亚组。本研究表明,聚类法和主坐标分析法结果基本吻合,均表明亚洲大麦种质资源存在丰富的变异,但主坐标分析法更精细。  相似文献

槟榔是海南重要的热带特色经济作物,具有很高的药用价值,被列为“四大南药”之首。但其基础研究落后,缺乏对种质资源的系统研究,遗传多样性尚不明确。利用SSR分子标记技术对海南岛58份栽培槟榔资源进行遗传多样性分析,旨在明确槟榔栽培种的亲缘关系,为槟榔杂交育种、功能基因的挖掘提供理论依据。结果表明:从500对SSR引物中筛选出32对多态性好、条带清晰的引物,在58份种质资源中共检测到79个等位基因,每个SSR位点2~5个,平均2.4688个,其中有效等位变异占观测等位变异的58.92%。各引物PIC值变幅为0.0169~0.5969,平均值为0.2254,Shannon信息指数(I)为0.0496~1.2552,平均值为0.4496;Nei’s遗传多样性指数(Nei)变幅为0.0171~0.6492,平均值为0.2596,说明所选SSR引物在供试样品中检测等位基因遗传多样性偏低。利用UPGMA构建58份栽培槟榔的遗传聚类树状图,第V组材料遗传多样性最丰富,其他分组群体遗传多样性较低,聚类结果说明海南栽培槟榔品种无明显的区域划分,表明海南槟榔不同地区间的种苗交流频繁,总体遗传多样性水平较低,该研究为槟榔遗传育种亲本选择提供依据,也为今后引进种质增补差异资源提供参考。  相似文献

 用20对SSR引物对原产于云南的16份地方稻种和2份选育品种进行单个品种群体内的遗传多样性分析。结果表明,87.5%的地方品种群体内SSR多态性高于选育品种,而12.5%的地方品种群体内SSR标记多态性与选育品种相近。81.2%的地方稻种群体内的等位基因数(Na)和Nei基因遗传多样性指数(He)高于选育品种,而18.8%的地方品种群体内Na和He与选育品种相同或略小。水稻地方稻种群体内He的差异较大,其变异范围为0.0146~0.5117,其中,黄板所 1(1980年收集)、黄板所 2(2007年收集)、麻线谷 1(1980年收集)、麻线谷 2(2007年收集)的群体内遗传多样性较高,分别为0.2327、0.4214、0.5117和0.4489。87.5%地方品种的杂合度(Ho)明显高于选育品种;地方品种群体间遗传多样性差异很大,其中1/4的遗传差异来源于地方稻种群体内,差异极显著。RM333、RM257和RM180在供试云南地方稻种群体内的多态性、等位基因数、多样性指数和变异百分率均较高,适用于云南地方稻种群体内遗传多样性检测。  相似文献

为了探讨利用小麦亲本间分子遗传距离预测F1杂种优势的可行性,以河南省8个不同类型的小麦品种(系)为材料,分别于2003年和2004年按不完全双列杂交组配28个组合,分析与产量相关的9个性状的F1杂种优势。分别选用均匀分布于小麦21条染色体上的287对SSR引物和33个二核苷酸重复ISSR引物,计算8个亲本间的分子遗传距离.进而分析亲本间分子遗传距离与F1杂种优势表现的相关性。结果表明,SSR和ISSR两种标记估算的遗传距离间相关极显著(相关系数为0.832),表明用两种分子标记对小麦品种进行遗传差异分析的结果是一致的。除株高外,利用两种分子标记计算的亲本间分子遗传距离与F1杂种优势在全部28个组合中相关不显著,而在12个极端组合中相关系数得到较大提高,并在株高、主茎千粒重和千粒重3个性状上相关达到显著或极显著水平,且两年的研究结果一致。这说明亲本间分子遗传距离与F1杂种优势的相关性与材料的选择有关。因此认为利用小麦亲本间的SSR和ISSR遗传距离预测F1的杂种优势还需进一步研究。  相似文献

将水稻(鄂香1号)与甜高粱[Sorghum dochna (Forssk.) Snowden]远缘杂交后,经过13代连续自交筛选,获得一批性状相对稳定的变异株系。对其中11个(编号为DC1~DC11)有代表性的变异株系进行表型、遗传背景和应用研究。结果表明,各变异株系生育期(122~141 d)、株高(118.9~228.9 cm)、单株产量(27.5~40.4 g)、结实率(49.7%~89.6%)、垩白度(0.3%~14.4%)、整精米率(53.9%~68.6%)等性状差异明显。利用水稻差异性鉴定的48对标准SSR引物对受体亲本及变异株系进行鉴定,发现该11个株系与鄂香1号差异引物个数为8~15个,表明这11个株系是不同于受体亲本的新材料,该11个株系之间的差异引物个数为2~15个,表明多数变异株系之间也存在差异。利用受体亲本和变异株系与不育系C6S配制杂交组合并进行综合评价,发现部分变异株系所配杂交组合与对照品种丰两优四号相比,在产量和米质方面具有一定的优势。表明通过远缘杂交创造的部分变异株系具有潜在的应用价值。  相似文献

The phenotypically uniform indica variety IR64 was chosen for study of the source and inheritance of within cultivar residual variation using a set of SSR markers.Residual heterogeneity in IR64 was identified on the short arm of chromosome 2 involving at least 5 SSR loci spanning nearly 30 cM.The SSR variations originated from the parental lines of IR64(IR5657-33-2/IR2061-465-1-5-5)and were segregating in the selfed bulk seed stock in a Mendelian manner for more than 20 years.This study verified that the within cultivar variations of SSR in a morphologically uniform variety IR64 of a selfing crop came from its parental lines,which has immediate and commercial applications including test of hybrid seed purity,varietal fingerprinting,and curation and propagation of germplasm collections.  相似文献

A total of 334 introgression lines (INLs: BC3-derived lines) derived from crosses between a recurrent parent of Indica rice cultivar IR64 and 10 donor parents, including new plant type (NPT) lines IR65600-87-2-2-3, IR65598-112-2, IR65564-2-2-3, IR69093-41-2-3-2, IR69125-25-3-1-1, Hoshiaoba, IR66215-44-2-3, IR68522-10-2-2, IR71195-AC1, and IR66750-6-2-1, have been developed. These INLs with IR64 genetic background were characterized for eight agronomic traits: days to heading, culm length, leaf width, leaf length, panicle length, panicle number, 100-grain weight, and total spikelet number per panicle at the International Rice Research Institute from 2005 to 2007. To identify introgressed segments from the donor parents, genotypes of the 334 INLs were detected using more than 200 polymorphic simple sequence repeat markers. These segments detected on chromosomes 1, 2, 4, 5, and 6 were commonly introgressed across the INLs from more than four donor varieties. Based on the data of phenotype and genotype for the 334 INLs, associations between agronomic traits and introgressed chromosomal segments in the 334 INLs were investigated. A total of 54 regions for the eight traits were detected: seven regions for days to heading, eight regions for culm length, eight regions for leaf width, four regions for leaf length, six regions for panicle length, three regions for panicle number per plant, seven regions for 100-grain weight, and 11 regions for total spikelet number per panicle. Among them, the region on the long arm of chromosome 4 was associated with characteristics of the NPT such as long leaf, broad leaf, and high spikelet number. The developed 334 INLs with the IR64 genetic background will be useful materials for genetic analysis of agronomic traits.  相似文献

青海省几个主要春性甘蓝型油菜杂交种的SSR分析   总被引:2,自引:0,他引:2  
为了探明青海省主要甘蓝型油菜品种的遗传变异,建立准确高效的种子纯度鉴定方法,本研究选取5个青海省甘蓝型油菜主栽品种,利用140对简单重复序列(Simple sequence repeat,简称SSR)引物对其进行分子标记分析,从中筛选出26对能清晰扩增出多态性主带的引物,进一步对包括这5个品种及其中4个杂交品种的7个亲本系进行扩增后,经6%聚丙烯酰胺凝胶电泳分离和带型分析,成功构建出5个青海省甘蓝型油菜主栽品种及其中4个杂交种亲本系的指纹图谱,并鉴定出了这4个杂交种的共显性标记。这些标记不仅可用于亲本材料和杂交品种真伪的鉴别,还可为杂交种纯度的鉴定提供可靠方法,对亲本知识产权的保护及杂交种推广具有重要的意义。  相似文献

Five super hybrid rice combinations, i.e. HYS-1/R105, Pei‘ai 64S/E32, Liangyoupeijiu (Pei‘ai 64S/9311), 88S/0293, and J23A/Q611, and their parental lines were tested by means of SSR analysis. A total of 144 SSR primer pairs distributed on 12 rice chromosomes were used, out of which 47 detected polymorphism among the tested rice lines. Among all these primers, RM337 and RM154 produced polymorphic patterns in four or more of the tested experimental materials respectively, and they could distinguish among most rice genotypes tested. Twenty-four primer pairs, two on each rice chromosome, were selected to make a reference SSR marker-based fingerprinting for the rice lines. For most of the primer pairs, F1 hybrids mainly showed complementary pattern of both parents, which could be very useful to distinguish the F1 from its parental lines. In addition, 5 primer pairs were selected as special primer pairs for five hybrid rice combinations respectively. By combining the rapid, simple method on DNA extraction, it is suggested that SSR technique has wide prospective in variety authentication and purity identification.  相似文献

比较了籼、粳两个糯性突变品种与原亲本品种淀粉的理化特性。糯性突变后除了直链淀粉含量显著降低外,淀粉的糊化温度变高、胶稠度增大、RVA谱(rapid viscosity analyzer)参数下降。RVA谱参数降低的幅度从高到低依次是消减值>回复值>冷胶粘度>热浆粘度>最高粘度>崩解值,表明控制直链淀粉含量的Wx位点对淀粉糊化特性和粘滞性是有影响的,存在多效性。同时,两个糯稻品种之间的RVA谱差异较大,其中崩解值和最高粘度差值均大于糯性突变对它们的影响,说明除直链淀粉外,支链淀粉也影响淀粉的理化特性。对Wx位点鉴定的结果显示扬辐糯4号wx基因第一内含子剪切位点+1位的碱基由原品种的G突变成了T,扬糯2号与其原品种相同(均为T);两个糯稻突变品种wx基因前导区的(CT)n微卫星重复序列与原品种相比都发生了变化。  相似文献

We compared intra-varietal variation of two rice varieties (Ashoka 200F and Ashoka 900F) produced by a very simple bulk breeding method (mass selection with no line selection at any stage) with one line-selected variety (Ashoka 228), all derived from the same cross. Their parents, the upland variety Kalinga III and the irrigated transplanted medium-lowland variety IR64, both originated through line selection and were used as control varieties.

Panicle-to-row progenies of all the varieties were evaluated in eastern India under irrigation in the dry season and in the rainy season using two water regimes; entirely rainfed or with supplementary irrigation. Intra-varietal variation for quantitative traits, irrespective of the method of breeding, was low. Only Ashoka 200F had significant variation for grain yield, days to flowering and plant height but only in the dry season. This season was not the target of the breeding programme and selection would be unlikely to produce a worthwhile response for the rainy season.

At 43 SSR loci there was more genetic variation between lines within the bulk-selected varieties than within Ashoka 228. Kalinga III was the least variable variety while IR64 had less heterozygosity but greater heterogeneity than the two bulk-selected varieties.

Despite the greater simplicity of the method, mass selection in bulk populations produced varieties that met the distinctness, uniformity and stability (DUS) criteria for seed certification in India. Such uniformity was achieved because of the high selection pressures applied to the bulk. We conclude that this very simple bulk-population breeding approach is highly cost-effective and produces sufficient seed for wide testing earlier than any alternative method.  相似文献

利用SSR标记对64个玉米自交系进行了杂种优势群的划分。20对SSR引物在供试材料中共检测出96个等位基因的变异,每对引物检测出等位基因数2~9个,平均4.36个。多态信息量变化范围为0.22~0.86,平均多态信息量为0.61。UPGMA聚类分析结果表明,供试自交系可分为8大类群,分类的结果与系谱关系基本一致。  相似文献

The cytoplasmic-genetic male sterility is the most widely used system for hybrid rice (Oryza sativa) seed production. It requires three lines: the two hybrid parents (A and R lines) and a third line (maintainer (B line)), which maintains the sterility of the female parent during seed increase. Studies were undertaken to identify maintainers and restorers for introduced cytoplasmic male sterile (CMS) lines using simple sequence repeat (SSR) markers and testcrosses. Molecular screening with SSR marker RM6100 grouped the assembled inbred lines into 30 potential restorers and 55 potential maintainers. One maintainer (WAS 127 -12-1-6-3-1) and 16 restorers were confirmed by testcrosses. WAS 127-12-1-2-3 had higher grain yield per plant in crosses with CMS1 (IR69625A) than the local checks (AgraRice and Jasmine 85) and could be used as a restorer to develop a locally adapted, high-yielding hybrid. Conversion of WAS 127 -12-1-6-3-1 into a local CMS line was stopped because of sterility instability in the BC1F1 generation. Continued testcrosses to the set of identified potential maintainers could help identify a stable one that can successfully be converted into a locally adapted CMS line.  相似文献

为了比较SSR标记法和醇溶蛋白法检测小麦种子纯度的准确度,以参加冬小麦区域试验的8个品系为试材,利用SSR标记法和醇溶蛋白法分别检测8个品系中的杂株,以田间表型鉴定结果为对照.结果表明,在800个单株(每个品系取100个单株)中,醇溶蛋白法和SSR标记法分别检出44和55个杂株,田间表型鉴定出49个杂株.这49个杂株中,有41株同时被SSR标记法和醇溶蛋白法鉴定出来,另有8株仅被SSR标记法鉴定出来.证明SSR标记法检测小麦种子纯度较醇溶蛋白法更加准确.  相似文献

选用8个亲本按NCⅡ配制16个组合,研究了F1-F4:5代产量性状和F4种子品质性状的遗传变异情况,测定了F4:5384个株系、56个品系的产量。结果表明:①各性状的一般配合力变异大于特殊配合力变异,杂种均数的遗传分量中以加性效应为主,这与前人研究结果一致。②亲本产量配合力在杂种世代间表现不一致,早代测定以F3较为适宜。③亲本性状值与一般配合力效应呈正相关,表明亲本性状优劣对杂种后代表现有一定影响。④优良品系的分布很集中,主要分布在母本963-4-1和父本88-1202的序列组合中,显示出优良亲本在育种效果上的突出优势。⑤963-4-1和88-1202是很好的高产育种亲本,904-79和佐治亚的育种利用价值也较高。  相似文献
