Piscine reovirus (PRV) was common among wild and farmed salmonids in British Columbia, western Canada, from 1987 to 2013. Salmonid tissues tested for PRV by real‐time rRT‐PCR included sections from archived paraffin blocks from 1974 to 2008 (n = 363) and fresh‐frozen hearts from 2013 (n = 916). The earliest PRV‐positive sample was from a wild‐source steelhead trout, Oncorhynchus mykiss (Walbaum), from 1977. By histopathology (n = 404), no fish had lesions diagnostic for heart and skeletal muscle inflammation (HSMI). In some groups, lymphohistiocytic endocarditis affected a greater proportion of fish with PRV than fish without PRV, but the range of Ct values among affected fish was within the range of Ct values among unaffected fish. Also, fish with the lowest PRV Ct values (18.4–21.7) lacked endocarditis or any other consistent lesion. From 1987 to 1994, the proportion of PRV positives was not significantly different between farmed Atlantic salmon, Salmo salar L. (44% of 48), and wild‐source salmonids (31% of 45). In 2013, the proportion of PRV positives was not significantly different between wild coho salmon, Oncorhynchus kisutch (Walbaum), sampled from British Columbia (5.0% of 60) or the reference region, Alaska, USA (10% of 58).  相似文献

This research was initiated in conjunction with a systematic, multiagency surveillance effort in the United States (U.S.) in response to reported findings of infectious salmon anaemia virus (ISAV) RNA in British Columbia, Canada. In the systematic surveillance study reported in a companion paper, tissues from various salmonids taken from Washington and Alaska were surveyed for ISAV RNA using the U.S.‐approved diagnostic method, and samples were released for use in this present study only after testing negative. Here, we tested a subset of these samples for ISAV RNA with three additional published molecular assays, as well as for RNA from salmonid alphavirus (SAV), piscine myocarditis virus (PMCV) and piscine orthoreovirus (PRV). All samples (n = 2,252; 121 stock cohorts) tested negative for RNA from ISAV, PMCV, and SAV. In contrast, there were 25 stock cohorts from Washington and Alaska that had one or more individuals test positive for PRV RNA; prevalence within stocks varied and ranged from 2% to 73%. The overall prevalence of PRV RNA‐positive individuals across the study was 3.4% (77 of 2,252 fish tested). Findings of PRV RNA were most common in coho (Oncorhynchus kisutch Walbaum) and Chinook (O. tshawytscha Walbaum) salmon.  相似文献

A Jaundice Syndrome occurs sporadically among sea‐pen‐farmed Chinook Salmon in British Columbia, the westernmost province of Canada. Affected salmon are easily identified by a distinctive yellow discolouration of the abdominal and periorbital regions. Through traditional diagnostics, no bacterial or viral agents were cultured from tissues of jaundiced Chinook Salmon; however, piscine reovirus (PRV) was identified via RT‐rPCR in all 10 affected fish sampled. By histopathology, Jaundice Syndrome is an acute to peracute systemic disease, and the time from first clinical signs to death is likely <48 h; renal tubular epithelial cell necrosis is the most consistent lesion. In an infectivity trial, Chinook Salmon, Sockeye Salmon and Atlantic Salmon, intraperitoneally inoculated with a PRV‐positive organ homogenate from jaundiced Chinook Salmon, developed no gross or microscopic evidence of jaundice despite persistence of PRV for the 5‐month holding period. The results from this study demonstrate that the Jaundice Syndrome was not transmissible by injection of material from infected fish and that PRV was not the sole aetiological factor for the condition. Additionally, these findings showed the Pacific coast strain of PRV, while transmissible, was of low pathogenicity for Atlantic Salmon, Chinook Salmon and Sockeye Salmon.  相似文献

The middle Missouri River (MMR; Fort Randall Dam, SD to Gavins Point Dam, NE‐SD) is stocked with hatchery‐reared pallid sturgeon, Scaphirhynchus albus (Forbes and Richardson), from upper Missouri River broodstock to aid recovery of this federally endangered species. Emigration of these fish through Gavins Point Dam restores genetic connectivity that likely existed pre‐impoundment but could lead to outbreeding depression in the future. Recapture data of hatchery‐reared pallid sturgeon stocked in the MMR were evaluated to improve understanding of pallid sturgeon emigration. From 2004 to 2015, 219 emigrants were caught: 4 stocked at age ≥2 years and 215 stocked at age ≤1 year. Emigration of the 2001‐2007 year classes stocked at age 1 was a consistent phenomenon and appeared higher than emigration of year classes stocked at ages 2–3. Little evidence suggested emigration was associated with an unusually high‐water event in 2011. The annual emigration probability of individuals stocked at age 1 estimated from multi‐state mark–recapture models was 0.05 [95% confidence interval = 0.04–0.06] for fish ages ≥1 year. This study suggests that alterations to stocking practices (e.g. stocking age) may affect emigration rates and, therefore, connectivity among pallid sturgeon populations.  相似文献

Non‐native predators may interfere with conservation efforts for native species. For example, fisheries managers have recently become concerned that non‐native brown trout may impede efforts to restore native salmon and trout in California's Trinity River. However, the extent of brown trout predation on these species is unknown. We quantified brown trout predation on wild and hatchery‐produced salmon and trout in the Trinity River in 2015. We first estimated the total biomass of prey consumed annually by brown trout using a bioenergetics model and measurements of brown trout growth and abundance over a 64‐km study reach. Then, we used stable isotope analysis and gastric lavage to allocate total consumption to specific prey taxa. Although hatchery‐produced fish are primarily released in the spring, hatchery fish accounted for most of the annual consumption by large, piscivorous brown trout (>40 cm long). In all, the 1579 (95% CI 1,279–1,878) brown trout >20 cm long in the study reach ate 5,930 kg (95% CI 3,800–8,805 kg) of hatchery fish in 2015. Brown trout predation on hatchery fish was ca. 7% of the total biomass released from the hatchery. Brown trout only ate 924 kg (95% CI 60–3,526 kg) of wild fish in 2015, but this was potentially a large proportion of wild salmon production because wild fish were relatively small. As large brown trout rely heavily on hatchery‐produced fish, modifying hatchery practices to minimise predation may enhance survival of hatchery fish and potentially reduce the abundance of predatory brown trout.  相似文献

Unusually large returns of several stocks of fall Chinook salmon (Oncorhynchus tshawytscha) from the U.S. Northwest commonly occurred during the late 1980s. These synchronous events seem to have been due to ocean rather than freshwater conditions because natal rivers of these stocks were geographically disconnected. We examined year‐to‐year variability in cohort strength of one of these stocks, Upriver Bright (URB) fall Chinook salmon from the Columbia River Hanford Reach for brood years 1976–99 (recovery years 1979–2002). We used the ocean recovery rate of coded‐wire‐tag (CWT) fish as an index of cohort strength. To analyse year‐to‐year variability in the ocean recovery rate, we applied a log‐linear model whose candidate explanatory variables were ocean condition variables, fishing effort, age of recovered fish, and fish rearing type (hatchery versus wild). Explanatory variables in the best model included fishing effort, and the quadratic term of winter sea surface temperature (SST) measured from coastal waters of British Columbia, Canada during the fish's first ocean year. The coefficient of the quadratic term of SST was significantly negative, so the model shape was convex. Our findings can be used to infer year‐to‐year variability in cohort strength of other fall Chinook salmon whose life history and ocean distributions are similar to the URB fish.  相似文献

Kudoa thyrsites (Myxozoa) encysts within myocytes of a variety of fishes. While infected fish appear unharmed, parasite‐derived enzymes degrade the flesh post‐mortem. In regions of British Columbia (BC), Canada, up to 4–7% of fillets can be affected, thus having economic consequences and impacting the competitiveness of BC's farms. K. thyrsites was monitored in two farms having high (HP) or low (LP) historical infection prevalence. At each farm, 30 fish were sampled monthly for blood and muscle during the first year followed by nine samplings during year two. Prevalence and intensity were measured by PCR and histology of muscle samples. In parallel, fillet tests were used to quantify myoliquefaction. Infections were detected by PCR after 355 and 509 degree days at LP and HP farms, respectively. Prevalence reached 100% at the HP farm by 2265 degree days and declined during the second year, whereas it plateaued near 50% at the LP farm. Infection intensities decreased after 1 year at both farms. Blood was PCR‐positive at both farms between 778 and 1113 degree days and again after 2000 degree days. This is the first monitoring project in a production environment and compares data between farms with different prevalence.  相似文献

Piscine orthoreovirus (PRV) is a common and widely distributed virus of salmonids. Since its discovery in 2010, the virus has been detected in wild and farmed stocks from North America, South America, Europe and East Asia in both fresh and salt water environments. Phylogenetic analysis suggests three distinct genogroups of PRV with generally discrete host tropisms and/or regional patterns. PRV-1 is found mainly in Atlantic (Salmo salar), Chinook (Oncorhynchus tshawytscha) and Coho (Oncorhynchus kisutch) Salmon of Europe and the Americas; PRV-2 has only been detected in Coho Salmon of Japan; and PRV-3 has been reported primarily in Rainbow Trout (Oncorhynchus mykiss) in Europe. All three genotypes can establish high-load systemic infections by targeting red blood cells for principal replication. Each genotype has also demonstrated potential to cause circulatory disease. At the same time, high-load PRV infections occur in non-diseased salmon and trout, indicating a complexity for defining PRV's role in disease aetiology. Adult sockeye salmon, Oncorhynchus nerka (Walbaum), migrating upstream in the Fraser River, British Columbia, are exposed to the myxozoan parasite Parvicapsula minibicornis when they enter the river from the ocean. Infections are initially localized in the kidney but have recently been associated with branchitis in one population. Adult fish from five locations in the watershed were sampled to determine whether branchitis was widespread. P. minibicornis infections in kidney glomeruli were prevalent in all samples except for a sample of fish that had just entered the Fraser River from the ocean. For fish captured in spawning streams, parasites were observed in the renal tubules and gill, and branchitis was observed in 70% of fish. Plasma osmolality was negatively correlated with the number of parasites in the kidney tubules, which we hypothesize to be caused by the breach of glomerular membranes as the parasite leaves the fish. Plasma lactate values increased with increasing levels of pathology in gills. These findings support the hypothesis that P. minibicornis impacts the physiology of migrating fish, which may in turn affect the likelihood that adults will be able to migrate and spawn successfully.  相似文献

Distinguishing hatchery‐reared fishes from wild conspecifics can be required to quantify the success of augmentation programmes. This study estimated the probability of identifying calcein‐marked, hatchery‐reared Colorado pikeminnow Ptychocheilus lucius Girard from external and internal structures. Both control and marked fish held in the laboratory were correctly identified 80% of the time after 300 days. A fluorometer was more accurate 180 days post‐marking, whereas visual observation was more discriminating at 300 days. There were no differences in detection rates among structures in laboratory fish, and for most structures of fish held outdoors, detection rates were <20%. Overall, a strong negative effect of light on mark persistence was observed. Also, an initial positive effect of fish size on mark fluorescence and a negative effect of fish growth were detected. These results suggest the potential use of calcein as a batch‐marking tool would be aided by future studies that better quantify light exposure and calcein mark persistence in both field and experimental settings.  相似文献

Many aquaculture hatchery practices are detrimental to the long-term viability of restocking and selective improvement programs. Small effective broodstock population sizes, differential broodstock contribution, differential larval/juvenile survival during metamorphosis and size-based grading, all have the potential to drastically reduce the level of genetic variation remaining in hatchery populations. Monitoring levels of genetic variation and maintaining detailed pedigrees on progeny is the key to circumventing these problems. In this study we used microsatellites, coupled with DNA parentage analyses, to track the loss of genetic diversity in two independent commercial barramundi (Lates calcarifer) hatcheries over three mass spawning events, where up to two females and seven males had the opportunity to participate. Initial broodstock contributions were observed to be highly skewed, with significant differences observed in both the level of contribution by females to each mass spawning, as well as in the number of males participating and subsequently contributing to the genetic composition of cohorts. Effective population sizes were around half that of census sizes. We then examined whether differential family survival through metamorphosis (27 days post-hatch) and/or first size grading further influenced the retention of genetic diversity levels initially sampled during spawning. Parentage analyses indicated that some families that had been initially represented in cohorts had been lost, or that the contribution by particular broodstock had changed. In one cohort, as many as 55% of progeny were found to be sired by a single male individual. Size grading was also found to potentially impact on genetic diversity, with data suggesting that family representation in each of the grades was non-uniform and that some families were on average faster or slower growing than others. These results illustrate that hatchery management practices have the potential to significantly impact on the retention of genetic diversity in this species.  相似文献

In this study, we investigated the toxic effects of ammonia‐N on the plasma and haematological parameters and histological structure of blunt snout bream (Megalobrama amblycephala) juveniles. The fish (initial weight, 14.79 ± 0.01 g) were randomly sorted into six tanks (200 L), and each tank was stocked with 40 fish for culture. The juveniles were exposed to two ammonia‐N levels—0 mg/L (control group) and 25 mg/L (experimental group)—and sampled at 0, 6, 12, 24, 48 and 72 hr, and then they underwent 96 hr of post‐exposure recovery. The results showed that ammonia‐N had significant effects on the plasma and haematological parameters. The treatment group showed increased cortisol, plasma ammonia and haematocrit levels and white blood cell count with increasing exposure time, up to 24 hr, and then the levels and count decreased. A significantly higher plasma glucose level was observed in the treatment group at 12 hr. After 96 hr of post‐exposure recovery, all parameter levels decreased to the control levels. The fish displayed histopathological alterations in the gills, liver and kidney. The results indicate that the severity of the lesions clearly differed among the organs, with the liver showing the most extensive damage, followed by the gills and kidney. Adverse effects to physiological indicators and histological structure increased with increasing exposure time before 24 hr. The fish showed self‐regulation; however, the histological structure could not recover fully, the gill tissue showed irreversible changes and the kidney tissue exhibited the worst recovery ability.  相似文献

It is advantageous for fish to choose different substrate types and brightness based on their current life cycle stage, foraging requirements, predator avoidance and water conditions. We determined the substrate type and brightness preference of hatchery‐reared and wild juvenile Schizothorax wangchiachii and Percocypris pingi under laboratory conditions. Individuals and groups were exposed to four kinds of substrate types (sand: diameter < 1 mm, small gravel: diameter approximately 2–3 cm, large gravel: diameter approximately 10–15 cm, sand and medium gravel mix: diameter approximately 4–7 cm) and two kinds of substrate brightness (black and white). The results showed that hatchery‐reared and wild S. wangchiachii and P. pingi significantly preferred large gravel and black substrates regardless of the number of fish (p < .05). P. pingi had a significantly higher preference for black substrate than S. wangchiachii both in hatchery‐reared and wild individuals (p < .05). Hatchery‐reared and wild individuals of the two species shared a preference for large gravel and black substrates suggesting that substrate preferences might be genetically based. The findings in the present study could be used to improve the rearing conditions for the two species in hatchery and thereafter to enhance its adaptability after releasing to the wild.  相似文献

Ichthyophthirius multifiliis, more commonly known as ‘white spot’ is recognised as one of the most pathogenic diseases of wild and cultured freshwater fish. In trout hatchery systems infections can quickly establish and result in high mortality if left unregulated. Current control centres around the use of regular bath treatments with formalin, however, treatments are not always effective and are labour intensive. Environmentally safe, non-chemical alternatives are required. This paper describes the performance of a mechanical system developed to remove cysts from commercial trout raceways. The system consists of two parts: a specially designed suction head connected to a pump that is used to vacuum the bottom of hatchery raceways, and a low-adhesion polymer raceway lining. Following tests on a series of potential linings, the most efficacious product in preventing settlement of cysts was used to line three raceways in a commercial rainbow trout (Oncorhynchus mykiss) hatchery. Over a period of three months, lined raceways were vacuumed on a daily basis. The three control raceways were maintained according to the farms normal husbandry procedures. Approximately thirty fish were sampled from each raceway at fortnightly intervals, and the abundance of the parasite in each raceway determined. The mechanical system led to a significant reduction in the abundance of the parasite in the test raceways, with a mean abundance of 361.88 ± 260.75 parasites per fish in control raceways compared with 1.64 ± 1.59 parasites per fish in the test raceways at the peak of the infection. Additionally fish survival was significantly higher in test raceways over control, with a mean of 84.5% of the stock surviving in the test raceways compared with only 70.6% in the controls by trial end.  相似文献

The Gulf Killifish (Fundulus grandis) is a popular baitfish and a candidate for commercial‐scale production. For inland production, ponds may need to be stocked at different times and densities to meet market demands. Therefore, in Experiment 1, juveniles were stocked into freshwater ponds at a density of 25,000 fish/ha, with two cohorts (mean total length (TL) = 3.92; 3.52 cm) stocked 1 month apart and evaluated for 12 weeks. Cohort 1 had a lower overall survival (40%) than cohort 2 (79%); with no difference between mean final TL (8.46; 8.00 cm); reaching a market size of 6 cm in 22 days and 32 days respectively. In Experiment 2, juveniles (mean TL = 3.16; 3.23 cm) were stocked into freshwater ponds at two densities (25,000 fish/ha and 50,000 fish/ha). Mean final TL was greater in the lower stocking density (8.68 cm) than the higher stocking density (8.20 cm); with no differences in survival between treatments (75; 72%) respectively. Lower stocking and higher stocking densities reached a market size of 6 cm in 29 and 31 days respectively. These results suggest that multiple cohorts can be stocked and grown to market size in one season at a stocking density of 50,000 fish/ha.  相似文献

Slovenia has no history of health problems related to proliferative kidney disease (PKD) either in farmed or in wild fish. However, due to the past molecular evidence for the presence of Tetracapsuloides bryosalmonae DNA in tissues of some fish from open waters, a survey was conducted on wild salmonids that were primarily sampled for other purposes. In winter 2010–2011, specimens from a total of 244 rainbow trout, Oncorhynchus mykiss (Walbaum), and brown trout, Salmo trutta L., from 30 bodies of fresh water were examined for T. bryosalmonae using a PCR method. The sampled fish showed no clinical signs or gross pathological lesions characteristic of PKD. Nineteen (7.8%) fish from seven (23.3%) fresh waters were positive for T. bryosalmonae. The identity of PCR amplicons was confirmed by sequencing. With one exception, all the positive fish were found in waters from the regions where the average yearly temperatures and the environmental pollution are higher. This preliminary countrywide survey provided the first insight into the situation regarding T. bryosalmonae infection of wild salmonids in Slovenia.  相似文献

The effect of nocturnal and diurnal releases on survival and migration of wild and hatchery‐reared Atlantic salmon, Salmo salar, L., smolts (n = 82) was investigated by releasing acoustically tagged smolts at the lower end of the River Vosso, Norway. Hatchery smolts was registered in the estuary within hours of their release, whereas wild smolts migrated over a prolonged period. The time of estuary exit was affected by river discharge but not by time of release. Progression rates were slow through the estuary (0.25 BL/s ± 0.18 SD) and fast through the fjord (1.80 BL/s ± 0.69 SD), and they were not affected by the time of release or origin. Survival to the fjord was low (0%–15%). Survival was not affected by body length but was lower for wild smolts than for hatchery smolts, and survival of the former was lower when the fish were released in daylight.  相似文献

One hundred farmed Atlantic salmon, Salmo salar L., were examined for the presence of nematodes by digestion of tissue in HCl–pepsin solution. All fish were sampled from one cage in a fish farm on the Norwegian south‐west coast. Fifty harvest quality salmon, that is, salmon for human consumption (mean 5.4 kg, variation 3.0–7.6 kg), were sampled at the processing line while 50 salmon runts (mean 1.1 kg, variation 0.4–1.8 kg), discarded due to poor performance, were sampled from the discard bin after the grading station. Runts are individual fish with clear signs of poor performance over time and abnormal appearance and are thus not processed for human consumption. No nematodes were found in the musculature or viscera of the 50 harvest quality salmon. In total, 75 nematodes were found in 10 (20%) of the runts; 53 nematodes in the viscera and 22 in the musculature. Nematodes in the musculature were identified as Anisakis simplex (Rudolphi, 1809 det. Krabbe, 1878), while nematodes in the viscera were identified as A. simplex and Hysterothylacium aduncum (Rudolphi, 1802).  相似文献
