AIM: To investigate the feasibility of differentiation of bone marrow mesenchymal stem cells (MSCs) into cardiomyocytes and vascular endothelial cells in dilated cardiomyopathy (DCM). METHODS: 20 rabbits were randomly divided into two groups. MSCs were isolated from bone marrow and cultured. Adriamycin was applied to the two groups to create rabbit DCM models. At 3 weeks after the creation of DCM models, the experiment group animals received intramyocardial injection of autologous MSCs. 4 weeks after transplantation, the implanted sites were examined to identify the labelled cells and to investigate its differentiation through immunofluorescence. RESULTS: At 4 weeks after the MSCs transplantation, the implanted cells were found in the experiment group and some differentiated into cardiomyocytes and vascular endothelial cells, which was not founded in the control group. CONCLUSION: Autologous bone marrow mesenchymal stem cells can differentiate into cardiomyocytes and vascular endothelial cells in dilated cardiomyopathy.  相似文献

AIM: To establish the Wistar rat model of furazolidone-induced dilated cardiomyopathy (Fz-DCM). METHODS: The Wistar rat model of Fz-DCM was established by feeding the animals with furazolidone. The left ventricular dimension and cardiac function were detected by echocardiogram. Aortic and right atrial pressure were measured by invasive catheter. Left ventricular interior diameter and the thickness of left ventricular free wall were measured after the rats were killed. Myocardial collagen network remodeling was observed and collagen volume fraction (CVF) was calculated by Van Gieson stain. RESULTS: ①The total incidence rate of DCM was 66.6% (20/30) in DCM group. ②Compared the corresponding subgroups to control group, the left ventricular end-diastolic diameter (LVED), left ventricular end-systolic diameter (LVES), the right atrial pressure, the left ventricular interior diameter and the ratio of left ventricle weight and body weight were increased significantly. The fraction shortening (FS), the left ventricular ejection fraction (LVEF) and the thickness of left ventricular free wall were decreased significantly. ③In FZ-DCM rat, the myocyte hypertrophy and degeneration, interistial fibrous tissue hyperplasia, the quantity of typeⅠand type Ⅲ collagen fibers and the collagen volume fraction (CVF%) were increased significantly. CONCLUSIONS: The rat model of DCM can be induced successfully by feeding the animals with furazolidone. In the rats with Fz-DCM, there are left ventricular dilation, the thinness of ventricular wall, the interistial fibrous tissue hyperplasia, and the decrease in left ventricular contractic function, indicating that the Fz-DCM rat model represents the pathophysiological characters of dilated cardiomyopathy.  相似文献

AIM: To investigate the effects of atorvastatin on myocardium peroxisome proliferator-activated receptors (PPARs) expression, regression of left ventricular hypertrophy, and the possible mechanisms in spontaneously hypertensive rats (SHR). METHODS: 16 nine-week-old male spontaneously hypertensive rats were randomly divided into two groups: SHR received atorvastatin at dose of 30 mg·kg-1·d-1 by oral gavage once daily for 8 weeks (SHR-A, n=8), and SHR received vehicle (0.9% saline) as controls (SHR, n=8). Age-matched Wistar-kyoto rats received vehicle for 8 weeks were served as normaltensive controls (WKY, n=8). Systolic blood pressure was measured at the beginning, 2, 4 and 8 weeks of the experiment. At the end of the experiment, plasma lipid levels were measured. Left ventricular hypertrophy was accessed by pathological analysis. The expressions of PPARα and PPARγ were investigated by the method of Western blotting. RESULTS: There was not much difference of systolic blood pressure and plasma lipid levels between SHR-A and SHR group (P>0.05). Compared with SHR group, left ventricular weight mass index decreased significantly in SHR-A group (P<0.01). The myocardium PPARα and PPARγ expression increased significantly (P<0.01). CONCLUSION: Atorvastatin regresses left ventricular hypertrophy and increases myocardium PPARα and PPARγ expression in spontaneously hypertensive rats, which is independent of its lipid-lowering activity.  相似文献

AIM:To sieve matrix metalloproteinases (MMPs) and the tissue inhibitors of matrix metalloproteinases (TIMPs) closely associated with ventricular remodeling of human heart failure using antibody chip technology.METHODS:We performed cytokine-specific antibody array analysis using individual left ventricular myocardial samples from 6 patients with heart failure due to arrythmogenic right ventricular cardiomyopathy (ARVC) undergoing transplantation and matched samples from 6 non-failing subjects to screen differentially expressed MMPs and TIMPs associated with the ventricular remodeling of heart failure.The results were further validated by ELISA and immunohistochemical analysis.RESULTS:We identified high expression of MMP-7 and MMP-10 and low expression of TIMP-4 in ARVC failing hearts compared to non-failing hearts by hybridization with the cytokine-specific antibody arrays containing 17 MMPs and 4 TIMPs on the chips.ELISA and immunohistochemical analyses further confirmed that differentially expressed levels of MMP-7, MMP-10, and TIMP-4 were observed not only in ARVC failing heart, but also in failing hearts due to ischemic (ICM) and dilated cardiomyopathy (DCM).CONCLUSION:Highly expressed MMP-7 and MMP-10 and lowly expressed TIMP-4 may be involved in the ventricular remodeling of heart failure derived from cardiomyopathy of different etiology.  相似文献

AIM: The study was designed to explore the autoimmune mechanism of myocardial injury and ventricular remodeling after acute myocardial infarction (AMI). METHODS: An experimental animal model of AMI was adopted in Wistar rats. After 6 weeks, splenocytes were freshly transferred to syngeneic inbred rats. Four weeks later, these recipient rats were anesthetized for hemodynamics analysis by catheter technique. Serum antibody against cardiac myosin heavy chanin (MHC) was screened by ELISA. Histopathological studies were performed on all hearts. The phenotypes of T lymphocytes in myocardium were analyzed by histocytochemistry stain. RESULTS: Histopathological studies showed the lymphocytes infiltration in non-infarction myocardium in AMI rats and the organ specific inflammation of myocardium in all succedent recipient (AMI-T) rats. Histocytochemistry stain revealed the predominant CD4+T cells infiltration in myocardium. The antibody against MHC was examined in 8/22 cases of AMI rats and AMI-T rats, but none in sham-T rats. The left ventricular dysfunction was found in AMI-T rats, which was characterized by slight decline of +dp/dtmax. CONCLUSIONS: The study showed inflammatory response of non-infarction myocardium in AMI rats and demonstrated the lymphocytes-mediated myocardial injury and cardiac dysfunction by adoptive transfer of splenocytes of AMI rats. The autoimmune-mediated myocardial injury might be a novel mechanism of ventricular remodeling after AMI.  相似文献

AIM: To study the mechanism of diabetic cardiomyopathy and abnormality of oxygen free radicals. METHODS: The contents of myocardial cytosolic cytochrome C, mitochondria cytochrome C, mitochondrial calcium, NO, MDA and the activity of SOD and NOS were determined in diabetic rats induced by STZ. The pathological changes were observed under transmission electron microscope. RESULTS: Compared to the normal and ganoderma group, the levels of mitochondrial NO, iNOS, MDA, calcium and plasma Cyt-C in rat myocardium were higher (P<0.05), while mitochondrial Cyt-C and SOD were lowered in model group (P<0.05). The bouncary indistinct, disorganization, a focal loss of muscular fibril, myocardium mitochondria swelling, pulmonary vascular endothelial cellular swelling and obstructed lumen of the capillary were also observed under transmission electronic microscope. CONCLUSION: The findings indicate that oxyradical and lipid peroxidation might be associated with the damage of myocardial mitochondria in NIDDM rats. Cyt-C and mitochondrial calcium is also involved in the process.  相似文献

AIM: To establish a method of isolating,culturing the adipose tissue-derived mesenchymal stem cells in vitro and to investigate the possibility of exogenous transplanting the adipose tissue derived mesenchymal stem cells for treatment of rat acute myocardial infarction.METHODS: 18 male rats were separated randomly into 3 groups: sham surgery group (control,n=6),acute myocardial infarction control group (AMI,n=6) and myocardial infarction plus cell transplantation group (AMI+cell,n=6).The infarcted hearts were made by occlusion of left coronary artery.The mesenchymal stem cells were isolated from the rats’ peritoneum by using digestion methods and reproduced in vitro,then the cells were labeled with BrdU and implanted into the infarcted heart of the rats.Heart functions were measured 4 weeks after implantation.The hearts were also harvested for pathological and histoimmunochemical observations to determine the survival and location of the implanted cells.RESULTS: Plenty of mesenchymal stem cells were obtained from the adipose tissue of rats’ peritoneum.Compared with the AMI group,the left ventricular systolic pressure in the cell therapy group was increased significantly (P<0.01),the left ventricular end-diastolic pressure was decreased (P<0.01),and the ratio of the left ventricular pressure rise and decay (±dp/dt) was decreased (P<0.05).The number of blood vessels was increased at the boundary of infarction site by pathological observation.The labeled cells were founded in the infarcted myocardium and the blood vessel wall.CONCLUSION: The adipose tissue is a new optional stem cell source.The methods of exogenous transplantation of adipose tissue derived mesenchymal stem cells for treatment of AMI is effective and feasible.  相似文献

AIM:To investigate the expression of F-box/WD repeat-containing protein 7 (FBXW7) in the myocardium of type 1 diabetic rats and to clarify its role in the development of diabetic cardiomyopathy. METHODS:All rats were randomly divided into non-diabetic (ND) group, 4-week diabetes mellitus (DM) model group, 8-week DM mo-del group and 12-week DM model group. The DM model was prepared by intraperitoneal injection of streptozotocin (60 mg/kg). The change of myocardial pathological structure was investigated by HE staining. The FBXW7 expression level in the myocardium was determined by Western blot and immunohistochemistry methods. RESULTS:DM induced cardiomyocyte degeneration and necrosis as shown by cardiac histological analysis. Both Western blot and immunohistochemistry results showed that the expression level of FBXW7 was significantly increased in 4-week, 8-week and 12-week DM groups compared with control group (P<0.01). However, The FBXW7 expression level in 12-week DM group was decreased compared with 4-week and 8-week DM groups. CONCLUSION:With the development of diabetes, the expression of FBXW7 in the myocardium of the rats with diabetic cardiomyopathy shows a tendency to increase first and then decrease, suggesting that it plays some roles in the development of diabetic cardiomyopathy.  相似文献

AIM: To investigate the ultrastructural changes of islet microvascular endothelial cells in STZ-induced type 1 diabetic mice. METHODS: BALB/c mice were randomly divided into diabetic group and control group. The expression of insulin and platelet-endothelial cell adhesion molecule-1 (CD31) in islet microvessels was detected by immunohistochemical staining. The ultrastructural changes of islet β cells and islet microvessels were observed under transmission electron microscope. RESULTS: Compared with control group, the number of islet β cells, ratio of β cells/α cells, average number of secretory granules in β cells and insulin expression area per islet in diabetic group were significantly decreased (P<0.01). Besides, diabetic group had fewer microvessels with lower expression of CD31 (P<0.01). Mitochondria in islet microvascular endothelial cells and pericytes in diabetic group were swelling. The basement membrane of islet microvessels became thicker in diabetic group (P<0.01). CONCLUSION: Islet microvascular endothelial cells were impaired in type 1 diabetic mice.  相似文献

AIM: To evaluate the effects of atorvastatin on nitric oxide(NO), endothelin-1(ET-1)and myocardial no-reflow in a rabbit model of acute myocardial infarction and reperfusion(AMI/R). METHODS: Twenty-four rabbits were randomized into 3 groups: 8 in AMI/R group, 8 in atorvastatin-treated group(5 mg·kg-1·d-1)and 8 in sham-operated group. Animals in the former two groups were subjected to 60 min of coronary occlusion followed by 120 min of reperfusion. Data on haemodynamics were collected. NO in blood sample, and in normal, and in infarcted reflow and no-reflow myocardium were evaluated respectively by nitrate reductase method. The levels of ET-1 in blood sample, and in normal, infarcted reflow and no-reflow myocardium were determined by radioimmunoassay. RESULTS: (1)Compared to the baselines, the heart rate(HR), systolic blood pressure(SBP), diastolic blood pressure(DBP), left ventricular systolic pressure(LVSP), maximal rate of increase and decline in left ventricular pressure(±dp/dtmax)and cardiac output(CO)in AMI/R and atorvastatin-treated groups were significantly declined, whereas left ventricular end-diastolic pressure(LVEDP)was increased after 60 min of coronary occlusion and 120 min of reperfusion(P<0.05 or P<0.01). However, in atorvastatin-treated group, LVSP, LVEDP, ±dp/dtmax and CO at the time point of 120 min of reperfusion recovered more significantly than those at the time point of 60 min of coronary occlusion(P<0.01), which was more significant than those in AMI/R group(P<0.05 or P<0.01). Compared to AMI/R group, the SBP and DBP were significantly heigher in atorvastatin-treated group(P<0.01).(2)In atorvastatin-treated group, the levels of ET-1 in blood sample were significantly lower than those in AMI/R group(P<0.01), and the levels of NO were significantly higher(P<0.01). Moreover, the levels of NO or ET-1 in infarcted reflow myocardium were significantly lower than that in AMI/R group(P<0.05 or P<0.01).(3)Atorvastatin could ameliorate myocardial function. CONCLUSION: Atorvastatin is effective in increasing NO and reducing ET-1 in blood plasma and local myocardium, and in protection of endothelial cells. Atorvastatin also has a beneficial effect on improving left ventricular function during acute myocardial infarction and reperfusion in rabbits.  相似文献

AIM:To study the effects of Ginkgo biloba extract (EGB) on myocardial TGF-β1 and collagen expression and interstitial fibrosis in type I diabetic cardiomyopathy rats. METHODS:Thirty male SD rats were randomly divided into normal control group (CON), diabetes mellitus group (DM) and EGB treatment group (EGB). Streptozocin was intraperitoneally injected into the animals in the latter 2 groups to induce type I diabetic rat model. The rats in EGB group were intraperitoneally injected with EGB. At the end of the 12th week, the body weight of each rat and its left ventri-cular weight, blood glucose, glycosylated hemoglobin and serum insulin concentration were measured. The left ventricular end-diastolic volume (LVEDV), the left ventricular end-systolic volume (LVESV), the left ventricular ejection fraction (LVEF) and the stroke volume (SV) were determined by echocardiography. The content of collagen in left ventricular myocardium, and the expression of transforming growth factor β1 (TGF-β1), procollagen type I and collagen type III were assayed by Sirius red staining, immunohistochemical staining and RT-PCR, respectively. Left ventricular myocardial cells of the neonatal SD rats were isolated and cultured in vitro with low-glucose culture medium (LG group), high-glucose culture medium (HG group) or high-glucose culture medium plus EGB (HG+EGB group). The mRNA levels of TGF-β1, procollagen type I and collagen type III were detected by RT-PCR. RESULTS:Compared with CON group, blood glucose, glycosylated hemoglobin, left ventricular weight index, the content of collagen, and the expression of TGF-β1, procollagen type I and collagen type III in left ventricular myocardial tissues of DM group were significantly increased, while the levels of blood insulin, LVEDV and SV were significantly decreased. However, compared with DM group, blood glucose, glycosylated hemoglobin, left ventricule weight index, the content of collagen, and the expression levels of TGF-β1, procollagen type I and collagen type III in the left ventricular myocardial tissues of EGB-treated rats were significantly decreased, while the levels of blood insulin, LVEDV and SV were significantly increased. Compared with LG group, the mRNA expression levels of TGF-β1, procollagen type I and collagen type III were significantly increased. However, compared with HG group, the mRNA expression levels of TGF-β1, procollagen type I and collagen type III were significantly decreased after treated with EGB. CONCLUSION: EGB retards the process of myocardial fibrosis and improves the cardiac functions in type I diabetic cardiomyopathy rats by down-regulating the expression of TGF-β1, reducing the synthesis and deposition of collagen type I and collagen type III.  相似文献

AIM:To observe the protective effect of delivery of acidic fibroblast growth factor (aFGF) to myocardium by ultrasound-targeted microbubble destruction (UTMD) on left ventricular function in diabetic cardiomyopathy (DCM) rats and to investigate the possible mechanisms. METHODS:Twenty-four rats were intraperitoneally injected with streptozocin to induce DCM and were randomly divided into DCM group and aFGF treatment group. Twelve healthy rats served as normal controls. The rats in aFGF treatment group were infused with SonoVue-aFGF mixed fluid through tail vein and UTMD was simultaneously performed. Four weeks after intervention, all rats underwent cardiac catheterization to mea-sure left ventricular end-systolic pressure (LVESP), left ventricular end-diastolic pressure (LVEDP) and the maximal increase/decrease rate of left ventricular pressure (LV±dp/dtmax). The microvessel density (MVD) of rat myocardial tissues was measured by immunohistochemical staining for CD31. The myocardial collagen volume fraction (CVF) was determined by improved Masson staining. The apoptotic index (AI) was detected by TUNEL method. RESULTS:Four weeks after intervention, the LVESP and LV±dp/dtmax in aFGF treatment group were significantly increased compared with DCM group (P<0.01), while the LVEDP in aFGF treatment group was significantly lower than that in DCM group (P<0.01). The MVD in aFGF treatment group was significantly increased compared with DCM group (P<0.01), but the CVF and AI in aFGF treatment group were significantly lower than those in DCM group (P<0.01). CONCLUSION: Delivery of aFGF to diabetic myocardium by UTMD could improve the left ventricular function of DCM rats and may be a new feasible therapeutic method for DCM.  相似文献

AIM: To investigate whether and how AT1 receptor blocker, valsartan, attenuates left ventricular remodeling and failure in a rat model of adriamycin(ADR)-induced dilated cardiomyopathy. METHODS: Weight-matched adult male Wistar rats were randomly divided into 3 groups as follows: 1) the ADR group, in which 2.5 mg/kg of ADR was weekly injected via a tail vein for 10 weeks (n=25); 2) concomitant AT1 receptor blocker valsartan and ADR, in which valsartan was administered by daily gavage at a dose of 30 mg·kg-1·d-1 (n=10); 3) control group (n=10). Hemodynamics and echocardiographic measurements were obtained at 12 weeks after treatment. Finally, left ventricle (LV) samples were collected at 12 weeks. The hydroxyproline content was determined by the methods of chloramines T. The expression of MMP-2, MMP-9 and tissue inhibitors of metalloproteinase-1 (TIMP-1) were measured by Western blotting. MMP-2 and -9 gelatinolytic activities were measured by gelatin zymography. RESULTS: Mortality was significantly lower in valsartan -treated rats than that in ADR rats (20% versus 40%, P<0.01). The dilatation of LV cavity was significantly attenuated in ADR-induced dilated cardiomyopathy rats given valsartan. Valsartan partially normalized LV contractile function, which was significantly reduced in ADR rats. The hydroxyproline content was increased in ADR-DCM group and significantly reduced by valsartan treatment (P<0.01). The protein levels of LV MMP-2 and MMP-9 were increased in ADR rats and attenuated by valsartan treatment (both P<0.01). However, no change in TIMP-1 was observed (P>0.05). The activities of LV myocardial MMP-2 and -9 gelatinolytic were increased significantly in ADR rats (both P<0.01) and attenuated by valsartan treatment (both P<0.01). CONCLUSION: Pretreatment with AT1 receptor blocker valsartan attenuates left ventricular remodeling and failure in a rat model of adriamycin-induced dilated cardiomyopathy.  相似文献

AIM: To study the electrical heterogeneity of transient outward potassium current (Ito) in left and right ventricular myocytes of cardiomyopathy rat. METHODS: The rats were peritoneally injected with L-thyroxine 0.5 mg/kg for 10 d to establish the model of ventricular hypertrophy. The right and left ventricular parts of the heart were separated and the ventricular myocytes were prepared by step digestion using enzyme solution. Ito was recorded by using whole cell patch clamp technique. The change of the electrical heterogeneity was determined. RESULTS: The electrical heterogeneity of Ito existed in the normal myocytes of left and right ventricles. In the myocytes of left and right ventricles isolated from the cardiomyopathy rats, the electrical heterogeneity was enhanced obviously and showed statistical difference. At +40 mV depolarizing test potential, the current density of Ito in the myocytes of right ventricle was increased from (9.23±0.84) pA/pF to (11.19±1.73) pA/pF, while the current density of Ito in the myocytes of left ventricle was decreased from (6.99±1.14) pA/pF to (4.95 ±1.84) pA/pF and the dispersion was increased. The V1/2 of right ventricle steady inactivation was increased significantly [from (-68.85±1.37) mV to (-49.86±0.69) mV]. The time constant τ of de-inactivation changed significantly [τleft=(79.16±7.04) ms,τright=(53.19±3.72) ms]. CONCLUSION: Enhanced electrical heterogeneity of Ito in the left and right ventricular myocytes of cardiomyopathy rat may represent one of the important ionic mechanisms for some arrhythmia caused by myocardial hypertrophy.  相似文献

AIM: To determine the relevance of NADPH oxidase subunit p22hox and the expression of superoxide anion on ventricular remodeling in myocardial infarction (MI) rats. METHODS: MI of Sprague-Dawley rats were established by left anterior descenting coronary artery ligation. 8 weeks after MI, Doppler echocardiography, hemodynamic study and histomorphometry were performed to analyze the ventricular remodeling. The level of thiobarbituric acid reactive substance in plasma and myocardium were measured, and the distribution of superoxide anion was observed with laser scanning confocal microscope. The expression of p22phox mRNA and protein level was detected by RT-PCR and immunohistochemistry. RESULTS: The left ventricular remodeling was significant in MI rats, also the level of thiobarbituric acid reactive substance increased in the plasma and non-infarcted myocardium. The expressions of p22-phox mRNA and protein levels, and superoxide anion increased in infarcted and non-infarcted myocardium in MI rats. CONCLUSION: Our results suggest that the expression of NADPH oxidase and its derived superoxide anion may take part in left ventricular remodeling through oxidative stresss after MI.  相似文献

AIM: To investigate the differential expression of short-chain acyl-CoA dehydrogenase (SCAD) in cardiac hypertrophy induced by hypertension or exercise training. METHODS: Spontaneously hypertensive rats (SHR) were used as the model of pathological cardiac hypertrophy. The swim-trained rats were used as the model of physiological cardiac hypertrophy. The systolic pressure, cardiac hypertrophy parameters, echocardiogram parameters, free fatty acid in serum and cardiac muscle, and the expression and activity of SCAD in the left ventricle were measured. RESULTS: Compared with the control rats, trained rats developed an athletic heart, of which cardiac function was enhanced, whereas SHR developed hypertensive cardiac hypertrophy, of which cardiac function was deteriorated. Compared with the control rats, the ratios of left ventricular weight to body weight were both increased in trained rats and SHR, showing that the degrees of cardiac hypertrophy were similar in the 2 models. Compared with the control rats, the decrease of free fatty acid both in serum and myocardium indicated that the fatty acid utilization was increased in the left ventricle of trained rats. Meanwhile, the expression and activity of SCAD in the left ventricle of trained rats were increased. However, free fatty acid both in serum and myocardium were increased, indicating that the fatty acid utilization was decreased in the left ventricle of SHR. Furthermore, SHR had the decreased expression and activity of SCAD in the left ventricle. CONCLUSION: The changes of SCAD are different in cardiac hypertrophy induced by hypertension and exercise training, indicating that SCAD may be used as a molecular marker of physiological and pathological cardiac hypertrophy, and a potential therapeutic target of pathological cardiac hypertrophy.  相似文献

AIM: To investigate changes of calcitonin gene-related peptide(CGRP) in myocardial stunning rats. METHODS: Rat in vivo myocardial stunning model was used. CGRP content in plasma and myocardium were determined by radioimmunoassay. RESULTS: Plasma level of CGRP increased significantly (P<0.01), but in left ventricular myocardium CGRP decreased obviously (P<0.05) in myocardial stunning group compared with the control group. CONCLUSION: CGRP content in the left ventricular myocardium was negatively correlated with plasma CGRP.  相似文献

AIM:To explore the effect of Xinshuaikang on myocardial autophagy in the rats with chronic heart failure and its relationship with the MAPK/ERK1/2 signaling pathway. METHODS:The rats were divided into sham group, model group (rat model of chronic heart failure was established by ligation of anterior descending branch of left coronary artery), low-, middle-, and high-dose Xinshuaikang treatment (TL, TM and TH) groups and captopril group (treated with captopril as positive control), with 12 in each group. Doppler echocardiography was used to evaluate the cardiac function. The morphological changes of the myocardium were observed by HE staining. TUNEL staining was used to detect cardiomyocyte apoptosis. The expression of microtubule-associated protein 1 light chain 3-Ⅱ (LC3-Ⅱ) in the myocardium was detected by immunofluorescence labeling. The protein levels of p-ERK, p-p38 MAPK, LC3-Ⅱ, beclin-1 and p62 in the myocardium were determined by Western blot. RESULTS:Compared with sham group, left ventricular end-diastolic dia-meter (LVEDD) and left ventricular end-systolic diameter (LVESD) in model group were increased, while left ventricular posterior wall thickness at end-diastole (LVPWTd), left ventricular posterior wall thickness at end-systole (LVPWTs), left ventricular ejection fraction (LVEF), cardiac output (CO), left ventricular diastolic pressure (LVDP), left ventricular systolic pressure (LVSP) and maximum rate of rise/decrease of left ventricular pressure (+dp/dtmax/-dp/dtmax) were decreased (P<0.05). The myocardial cells were deformed and necrotic, and the myocardial fibers were broken, with inflammatory cell infiltration. The apoptotic rate, the positive rate of LC3-Ⅱ, and the protein levels of p-ERK, p-p38 MAPK, LC3-Ⅱ/LC3-I and beclin-1 were increased, and the protein expression of p62 was decreased (P<0.05). Compared with model group, the levels of LVEDD and LVESD were decreased, LVPWTd, LVPWTs, LVEF, CO, LVSP, LVDP, +dp/dtmax and -dp/dtmax were increased in Xinshuaikang groups and captopril group (P<0.05). The morphological changes of myocardial cells were gradually returned to normal, and inflammatory cell infiltration, the apoptotic rate and the positive rate of LC3-Ⅱ were decreased. The protein levels of p-ERK, p-p38 MAPK, LC3-Ⅱ/LC3-I and beclin-1 were decreased, and the protein expression of p62 was increased (P<0.05). CONCLUSION:Xinshuaikang inhibits myocardial auto-phagy to play a role of cardiac protection in the rats with chronic heart failure, and its mechanism may be related to inhibition of MAPK/ERK1/2 signaling pathway.  相似文献

AIM: To detect the changes of S-adenosyl-L-homocysteine hydrolase (SAHH) signaling pathway and related proteins under the intervention of fimasartan (FIM) in rats with diabetes mellitus (DM), and to explore the pathological mechanism of the occurrence and development of diabetic cardiomyopathy (DCM). METHODS: The type 2 diabetic rat model was established by injection of streptozocin after 5-week high-fat diet. The rats were randomly divided into control group, DM group, and DM+FIM group. Fasting blood glucose (FBG), total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and fasting insulin (FINS) levels were tested. M-mode echocardiography was performed for determining the heart functions. High-performance liquid chromatography (HPLC) was used to detect homocysteine (Hcy), S-adenosylmethionine (SAM) and S-adenosyl-L-homocysteine (SAH). The alteration of SAHH in myocardium was determined by Western blot. RESULTS: The success rate of type 2 diabetic rat modeling was 84%. Compared with DM group, the body weight of the rats in DM+FIM group increased significantly (P<0.05), while cardiac index, left ventricular index, FBG and LDL-C were significantly reduced (P<0.05). The results of echocardiography showed that ejection fraction increased (P<0.05) in DM+FIM group. HPLC detection showed that the level of Hcy and the ratio of SAM/SAH were significantly reduced (P<0.05). The results of Western blot showed that the expression of SAHH in DM group was increased compared with control group, while that in DM+FIM group declined (P<0.01). CONCLUSION: The expression of SAHH, Hcy and other related factors may be important during the occurrence and development of early DCM, and FIM may play a role in this process as an inhibitor of SAHH.  相似文献

AIM: To study changes of medium chain acyl-CoA dehydrogenase (MCAD), muscle carnitine palmitoyltransferase I (M-CPT-I) and colligin mRNA/protein expression, to elucidate molecular mechanism of the recapitulation of fetal energy metabolism and ventricular remodeling and the effects of carvedilol during the development of pressure overload-induced left ventricular hypertrophy in rats. METHODS: Male Wistar rats of hypertrophy induced by constriction of abdominal aorta (CAA) were randomized into 2 groups (n=12, each group): 4-week group (CAA4 weeks group) and 12-week carvedilol intervention group (CAR group). Additional rats (n=12) underwent abdominal cavity incision without ligation to serve as age-matched sham operated controls (SH). Hemodynamics, ventricular remodeling parameters and free fatty acid (FFA) both in blood serum and myocardium were measured. RT-PCR analysis of the expression of mRNA of M-CPT-I, MCAD and collagen binding protein (colligin) were investigated. The protein expression of colligin was analyzed by Western blotting in the experimental animals and sham operation.RESULTS: LVM/BW and MAP in CAA group were increased more significantly than in sham group. There were progressive increases in FAA both in blood serum and myocardium in CAA group than in sham group, accompanied with downregulation of gene expressions of M-CPT-I and MCAD and colligin mRNA/ protein upregulation in LV in CAA group, while changes of all of these parameters in CAR group were attenuated.CONCLUSIONS: (1) The down-regulated expression of cardiac FAO enzyme genes (M-CPT-I and MCAD) in the hypertrophied heart may be responsible for “the recapitulation of fetal energy metabolism” during the development of pressure overload-induced left ventricular hypertrophy in rats. (2) Carvedilol attenuates the reversion of the metabolic gene expression back towards fetal type. (3) Carvedilol is effective in regressing the left ventricular remodeling by inhibiting colligin protein expression. A molecular mechanism by which carvedilol may confer cardioprotective effects in heart failure may be, in part, via preserving of the adult metabolic gene regulation and regressing left ventricular remodeling.  相似文献
