AIM: To observe the direct effect of LPS on expressions of ET-1, eNOS, and iNOS mRNA in human umbilical vein endothelial cells, and further research the molecular mechanism of effect of LPS on production of ET-1 and NO. METHODS:The third passage of cultured human umbilical vein endothelial cells was incubated with low concentration (100 μg/L) of LPS for 6 h. Total RNA was extracted. The expressions of ET-1, eNOS, and iNOS mRNA were analyzed by semi-quantitative RT-PCR method. RESULTS: ET-1 mRNA experession increased significantly, while expression of eNOS mRNA decreased significantly, and there was no significant change in expression of iNOS mRNA. CONCLUSION: In human umbilical vein endothelial cells, low concentration of LPS enhanced the expression of ET-1 mRNA, inhibited the expression of eNOS mRNA, and had no significant effect on the expression of iNOS mRNA.  相似文献

AIM: To observe the function and morphological change of human umbilical vein endothelial cells (HUVECs) treated with lipopolysaccharide (LPS) and composite salviae dropping pills (DSP). METHODS: HUVECs were cultured and incubated within 10 mg/L LPS for 12 hours. Different final concentrations of composite salviae dropping pills (1 g/L, 0.5 g/L, 0.25 g/L, 0.1 g/L) were added before and after LPS treatment. Cell viability, NO, NOS, ET-1 and intracellular calcium were measured. The cells were observed under inverted microscope, inverted phase contrast microscope, laser confocal scanning microscopy and transmission electron microscope. RESULTS: When given after LPS treatment, different final concentrations of composite salviae dropping pills played a protective role (P<0.05), and the concentration of 0.5 g/L was the most effective (P<0.01). When given before LPS treatment, 0.5 g/L and 0.25 g/L of composite salviae dropping pills protected the human vascular endothelial cells (P<0.05), but 1 g/L and 0.1 g/L didn't play a protective role (P>0.05). The HUVECs injured by LPS underwent apparent morphological change after treatment with composite salviae dropping pills. CONCLUSION: Composite salviae dropping pills have an evident protective effect on human umbilical vein endothelial cells exposed to LPS.  相似文献

AIM:This experiment is to investigate the effects of LPS on the organization and localization of VE-cadherin and F-actin in cultured human umbilical endothelial cells.METHODS:The human umbilical vein endothelial cell lines ECV-304 were incubated with LPS at different concentrations for 30 min. VE-cadherin was detected by immunofluorescence with primary mAb of VE-cadherin and FITC-conjugated secondary antibody. F-actin was detected with fluorescence staining with rodamine-phalloidin.RESULTS:At high concentration, LPS could induce reorganization of VE-cadherin with the formation of serrata cellular border and enlargement of intercellular gaps, which were apparently different from that in normal conditions with the high fluorescence intensity at cell-cell junction. F-actin depolymerization could also be induced by LPS at high concentration with the formation of stress fiber and filopodia.CONCLUSION:LPS(300 μg/L) could induce reorganization of VE-cadherin and F-actin in human umbilical vein endothelial cells.  相似文献

AIM: To study the protective effect of anti-aging Klotho protein on human umbilical vein endothelial cells (HUVECs) treated with high glucose (HG).METHODS: HUVECs were cultured in vitro, and divided into PBS control group, 5.5 mmol/L glucose group, 33.3 mmol/L glucose group, 0.1 μmol/L Klotho+33.3 mmol/L glucose group, 1 μmol/L Klotho+33.3 mmol/L glucose group, and 10 μmol/L Klotho+33.3 mmol/L glucose group. The viability of the HUVECs was measured by MTT assay. The content of malondialdehyde (MDA), and the activities of lactate dehydrogenase (LDH), superoxide dismutase (SOD) and glutathione (GSH) in cell culture supernatants were observed. The production of reactive oxygen species (ROS) in HUVECs was analyzed by flow cytometry. The levels of nitric oxide (NO), endothelin (ET-1), intercellular adhesion molecule-1 (ICAM-1) in HUVEC culture medium were detected by ELISA. The protein expression of nuclear factor-kappa B (NF-κB) in the HUVECs was determined by Western blot. RESULTS: Compared with PBS control group, 33.3 mmol/L glucose significantly decreased the HUVEC viability, increased ROS, LDH and MDA levels, reduced the activities of SOD and GSH, decreased the NO secretion, and induced the ET-1 and ICAM-1 secretion and the protein expression of NF-κB in HUVECs. When HUVECs were treated with Klotho protein at different concentrations combined with 33.3 mmol/L glucose, the cell viability was increased significantly, the ROS, LDH and MDA levels were decreased significantly, the antioxidant SOD and GSH activities were significantly increased, the secretion of NO was increased, but ET-1 and ICAM-1 releases and protein expression of NF-κB were significantly reduced.CONCLUSION: Anti-aging Klotho protein promotes the viability of HUVECs treated with HG, reduces the oxidative damage and ROS production, and restores the normal secretory function of HUVECs, thus playing a protective role in vascular endothelial cells through reducing the protein expression of NF-κB.  相似文献

AIM:To observe the effects of angiopoietin 4 (Ang-4) on lipopolysaccharide (LPS)-induced injury of human umbilical vein endothelial cells (HUVECs). METHODS:The EnVision immunohistochemical method was used to identify the HUVECs. After pre-treated with different doses of Ang-4 for 0.5 h, HUVECs was exposed to LPS at concentration of 10 mg/L for 24 h. The cell viability was evaluated by MTT assay. The content of tumor necrosis factor-alpha (TNF-α) in the supernatant and the concentrations of intracellular and supernatant von Willebrand factor (vWF) were detected by ELISA. The mRNA levels of Toll-like receptor 4 (TLR4), NF-κB p65 and TNF-α were determined by real-time PCR. RESULTS:Factor Ⅷ in the cytoplasm was positive in the HUVECs.Compared with normal group, LPS reduced the cell viability (P<0.01), and significantly increased the secretion of TNF-α and vWF (P<0.01). The mRNA expression of TLR4, NF-κB p65 and TNF-α also increased (P<0.01). Ang-4 at concentration of 100 μg/L enhanced the cell viability (P<0.01), reduced the content of vWF and TNF-α, and inhibited the LPS-induced increases in the mRNA levels of TLR4, NF-κB p65 and TNF-α (P<0.01). CONCLUSION: Ang-4 antagonizes LPS-induced damage in HUVECs by inhibiting TLR4-NF-κB p65-TNF-α signaling pathways.  相似文献

AIM: To investigate the effects of rapamycin on apoptosis, proliferation, migration ability and tumor related apoptosis inducing ligand(TRAIL) in cultured human umbilical vein endothelial cells(HUVECs). METHODS: Cultured HUVECs were treated with rapamycin at the concentrations of 0, 1, 10 and 100 μg/L for 24 h. The cell proliferation was measured by CCK-8 method. The cell migration ability was detected by Transwell chambers and wound healing test. The apoptotic index of HUVECs was quantitatively determined by measuring the activation of caspase-3. The morphological changes of the apoptotic cells were observed by DAPI staining. The expression of TRAIL was detected by Western blotting. RESULTS: A 24 h-incubation with rapamycin(1-100 μg/L) caused significant cell loss associated with the increase in apoptosis, as quantified by the determination of caspase-3 activity(P<0.01) in HUVECs. Obvious apoptotic morphology was observed by DAPI staining in HUVECs incubated with rapamycin. Rapamycin at the concentrations of 1-100 μg/L also impaired the migration ability of HUVECs(P<0.01). In addition, rapamycin(10-100 μg/L) inhibited the proliferation of HUVECs, whereas rapamycin at 1 μg/L had no such effect(P<0.01). Rapamycin(10-100 μg/L) also induced TRAIL expression in a dose-dependent manner(P<0.01). CONCLUSION: Rapamycin induces apoptosis, and inhibits the proliferation and migration of HUVECs. The up-regulation of TRAIL might be related to the injury of vascular endothelial cells caused by rapamycin.  相似文献

AIM: To study the effects of cladribine on growth and secretion activity of human umbilical vein endothelial cell line EA.hy926, and to investigate the mechanism of its anti-tumor effect by inhibiting endothelial cells. METHODS: The effects of cladribine at different concentrations on the cell viability were detected by CCK-8 assay. Apoptosis and cell cycle distribution were examined by flow cytometry. The protein expression levels were determined by Western blot. The levels of tumor necrosis factor-α (TNF-α), transforming growth factor-β1 (TGF-β1) and vascular endothelial growth factor (VEGF) secreted by EA.hy926 cells with cladribine treatment for 48 h were analyzed by ELISA. The nitric oxide (NO) production was measured by Gries method. RESULTS: Cladribine at 0.4~1 μmol/L inhibited the viability of EA.hy926 cells in time-and dose-dependent manners. The IC50 was about 3.644 μmol/L. The results showed 43.74% cells in S phase when the concentration of cladribine was 0.4 μmol/L, and 77.23% cells in S phase when the concentration of cladribine was 1 μmol/L. The apoptosis was not induced by cladribine at 0.4~10 μmol/L. The protein expression of Bax and caspase-3 did not change. The expression of p21 increased and the p53 decreased (P<0.05). The levels of TNF-α and TGF-β1 secreted by EA.hy926 cells increased after cladribine treatment for 48 h. The levels of VEGF and NO decreased. CONCLUSION: Cladribine obviously inhibits the viability of EA.hy926 cells. The mechanism is related to the cell cycle arrest. Cladribine promotes the secretion of TNF-α and TGF-β1 by EA.hy926 cells and inhibits the secretion of VEGF and NO.  相似文献

AIM: To study the protective effect of brain-derived neurotrophic factor (BDNF) on vascular endothelial cells with H2O2-induced oxidative injury. METHODS: Human umbilical vein endothelial cells (HUVECs) were cultured in vitro, and the oxidation injury model of HUVECs was established by treatment with H2O2. The oxidatively injured HUVECs were cultured with different concentrations (1, 10 and 100 μg/L) of BDNF. At the same time, the control group (no injury), PBS treatment after H2O2 injury group and TrkB inhibitor group (with 100 μg/L BDNF and 1: 1 000 TrkB inhibitor) were also set up. The viability of the HUVECs was detected by MTT assay. The levels of LDH, MDA, SOD and GSH were measured. The releases of NO, ET-1 and ICAM-1 were analyzed by ELISA. The changes of ROS production and cell apoptosis were evaluated by flow cytometry. The protein levels of TrkB, p-TrkB, cleaved caspase-3, Bcl-2 and Bax were determined by Western blot. RESULTS: Compared with uninjured control group, in H2O2 oxidative injury plus PBS treatment group, the viability of the cells was decreased significantly, the LDH and MDA levels were increased significantly and the activities of SOD and GSH were decreased significantly. The NO secretion was decreased, and the ET-1 and ICAM-1 concentrations were increased significantly. The ROS content and apoptotic rate were increased significantly. The protein levels of cleaved caspase-3 and Bax were increased but Bcl-2 protein expression was decreased significantly. Compared with PBS treatment group, in H2O2-injured HUVECs treated with different concentrations of BDNF, the cell viability was gradually increased, the LDH and MDA levels were decreased and the activities of SOD and GSH were increased gradually. The secretion of NO was increased but ET-1 and ICAM-1 were decreased gradually. The ROS content and apoptotic rate were decreased significantly. The TrkB and p-TrkB levels were significantly increased significantly, the protein expression of cleaved-caspase 3 and Bax was decreased gradually and the Bcl-2 protein expression increased gradually. The role of BDNF was inhibited by TrkB inhibitor. CONCLUSION: BDNF protects HUVECs from oxidative injury by binding with TrkB to activate the BDNF-TrkB signaling pathways.  相似文献

AIM: To evaluate the effect of exogenous hydrogen sulfide (H2S) on the expression of NLRP3 inflammasome in hepatocytes.METHODS: The hepatocytes L02 and SMMC-7721 were used to establish the model of inflammation by stimulating with lipopolysaccharide (LPS) at different concentrations in vitro. The expression of NLRP3 inflammasome in the hepatocytes was detected by Western blot and the cell viability was measured by MTT assay for determining appropriate concentration of LPS. The hepatocytes were divided into 4 groups:the cells in control group were incubated with normal medium for 18.5 h; the cells in LPS group were incubated with normal medium for 0.5 h followed by 100 μg/L LPS for 18 h; the cells in LPS+H2S group and H2S group were incubated with 200 μmol/L sodium hydrosulfide hydrate (NaHS) for 0.5 h followed by 100 μg/L LPS or normal medium for 18 h, respectively. The protein expression of NLRP3 and caspase-1 in the cells of every group was determined by Western blot. RESULTS: Compared with control group, the protein expression of NLRP3 and caspase-1 increased significantly in LPS group (P<0.05) and had no significant change in H2S group. Compared with LPS group, the protein expression of NLRP3 and caspase-1 in LPS+H2S group decreased significantly (P<0.05). CONCLUSION: In hepatocytes, exogenous H2S suppresses the expression of NLRP3 inflammasome.  相似文献

AIM: To investigate the effect of quercetin on endothelin-1-induced T-type calcium channel(TCC) expression in primary cultured human umbilical arterial smooth muscle cells for exploring the protective role of quercetin in cardiovascular system. METHODS: Human umbilical arterial smooth muscle cells were verified by immunocytochemistry. The cells in 2-3 passages were used and randomly divided into control group, quercetin alone group, model group and experimental group. The cells in control group were cultured without any drugs for 24 h. The cells in quercetin alone group were cultured with 80 μmol/L quercetin for 24 h. The cells in model group were cultured with ET-1 at the concentration of 100 nmol/L for 24 h. The cells in experimental groups were pretreated with quercetin for 1 h, then coincubated with 100 nmol/L ET-1 for 24 h. The concentrations of quercetin used in this study were 20, 40and 80 μmol/L, respectively. The expression of α1G, a TCC major subunit, was assayed at mRNA and protein levels by RT-PCR and Western blotting, respectively. The TCC currents(IcaT) were detected by the technique of whole-cell patch-clamp. RESULTS: Compared with control and experimental group, ICaT density (P<0.01) and the expression of α1G at mRNA (P<0.05) and protein (P<0.01) levels in model group were significantly increased. No significant difference in the results of quercetin alone group and control group was observed. CONCLUSION: The protective roles of quercetin in cardiovascular functions are related to the depressive effects of quercetin on ET-1-induced increase in both ICaT density and the expression of α1G at mRNA and protein levels in cultured human vascular smooth muscle cells.  相似文献

AIM: To investigate the effects of simvastatin on cigarette smoke extract (CSE)-induced expression levels of soluble endothelial cell protein C receptor (sEPCR) and membrane-associated endothelial cell protein C receptor (mEPCR ) in human umbilical vein endothelial cells (HUVECs). METHODS: Cultured HUVECs at passage 4 to 6 were randomly divided into control group, 5% CSE group, simvastatin groups and simvastatin+CSE groups. In simvastatin groups, HUVECs were incubated with simvastatin at the concentrations of 50, 100 and 200 μmol/L for 24 h. In simvastatin+CSE groups, the cells were treated with simvastatin at the concentrations of 50, 100 and 200 μmol/L for 2 h, and then exposed to CSE for 24 h. The protein level of sEPCR in the culture supernatants was measured by ELISA. The cells were collected for determining the mRNA expression of mEPCR by real-time PCR. RESULTS: Compared with control group, the protein level of sEPCR was significantly increased, and the mRNA expression of mEPCR was significantly decreased in 5% CSE group (both P<0.05). The protein levels of sEPCR were significantly increased, and the mRNA expression of mEPCR was significantly decreased in 100 μmol/L and 200 μmol/L simvastatin groups. However, the protein levels of sEPCR were lower, and the mRNA expression of mEPCR was significantly higher in 100 μmol/L and 200 μmol/L simvastatin groups than those in 5% CSE group. Compared with 5% CSE group, the protein levels of sEPCR in simvastatin+CSE groups were significantly decreased, but higher than those in control group and simvastatin group with corresponding concentration. On the contrary, the mRNA expression of mEPCR in simvastatin+CSE groups was significantly increased, but lower than that in control group and simvastatin group with corresponding concentration (all P<0.05). CONCLUSION: Simvastatin obviously increases the mRNA expression of mEPCR, decreases the protein level of sEPCR, and attenuates the CSE-induced endothelial injury in vitro .  相似文献

AIM: To investigate the effect of oxidized LDL (ox-LDL) on the expression of gap junction protein connexin43 in cultured human umbilical vein endothelial cells (HUVECs) in vitro. METHODS: Human umbilical vein endothelial cells cultured in normal condition were divided into blank control group, 50 mg/L,100 mg/L and 200 mg/L ox-LDL intervention groups. The mRNA expression of connexin43 in cultured HUVECs was detected with RT-PCR method; while the protein level of connexin43 was determined by the method of immunocytochemistry in the control and 100 mg/L ox-LDL intervention groups 24 h after ox-LDL was given. RESULTS: Different concentrations (50 mg/L, 100 mg/L, 200 mg/L) of ox-LDL up-regulated mRNA expression of connexin43 in cultured HUVECs after 24 h intervention (P<0.01). The protein level of connexin43 in cultured HUVECs intervened with 100 mg/L Ox-LDL for 24 h was up-regulated as compared to the control cells (P<0.01).CONCLUSION: Ox-LDL may up-regulate the expression of connexin43 at mRNA and protein levels in cultured human umbilical vein endothelial cells within short time, indicating that connexin43 plays an important role in the pro-atherosclerotic effect of Ox-LDL.  相似文献

AIM:To study the effect of activated protein C (APC) at different concentrations on apoptosis of human umbilical vein endothelial cells (HUVECs) induced by lipopolysaccharide (LPS).METHODS:The HUVECs were induced by LPS (1.0 mg/L) as apoptotic model that was administered by different concentration of APC (10 μg/L or 50 μg/L). Meanwhile, the control group and induced apoptosis group induced by LPS (1.0 mg/L) stimulation were also set up. The changes of cellular ultrastructures were observed under electron microscope. The DNA ladder and TUNEL fluorescent staining were measured in cells. Annexin-Ⅴ/PI double staining was used to measure the cell apoptosis rate by flow cytometry. Cell survival rate was measured by MTT assay. The proliferating cell nuclear antigen (PCNA) expression levels in cells were also measured by Western blotting to reflect the proliferation of the cells.RESULTS:There were significant apoptotic changes in the cells induced by LPS, but the apoptotic changes were reduced and apoptosis rates were decreased in the cells treated with APC. Meanwhile, cell survival rate and the protein levels of PCNA were increased after APC treatment, particularly at the concentration of 50 μg/L, which showed difference when compared with those induced apoptosis group by LPS (P<0.05).CONCLUSION:APC can inhibit HUVECs apoptosis induced by LPS and promote cell proliferation, thus protect the cells from injury.  相似文献

AIM:
To investigate the levels of aldolase A (ALDOA), carcinoembryonic antigen (CEA) and lactate dehydrogenase (LDH) in malignant pleural effusion (MPE) from patients with lung cancer and tuberculous pleural effusion (TBPE) from patients with tuberculous pleurisy, and to explore the effects of ALDOA on the proliferation, migration and invasion of human lung adenocarcinoma A549 cells. METHODS:Pleural effusion samples including 65 cases of MPE and 35 cases of TBPE were collected, and the levels of ALDOA, CEA and LDH were detected by ELISA and chemiluminescence assay. After A549 cells were treated with different concentrations of ALDOA, the proliferation, migration and invasion of the cells were investigated by MTT assay, scratch test, Matrigel assay and Transwell invasion assay. RESULTS:The levels of ALDOA, CEA and LDH in MPE were (46.8±21.4) μg/L, (82.2±56.6) μg/L and (755.8±382.5) U/L, respectively, which were significantly higher than those in TBPE [(23.9±17.2) μg/L, (12.6±9.7) μg/L and (388.4±163.9) U/L, respectively; P<0.01]. The concentration of ALDOA in MPE from adenocarcinoma patients [(71.7±32.1) μg/L] was significantly higher than that in MPE from squamous-cell carcinoma patients [(21.3±14.6) μg/L, P<0.05]. The concentrations of ALDOA in MPE and TBPE were positively correlated with the concentrations of CEA and LDH (P<0.01 or P<0.05). ALDOA enhanced the proliferation, migration and invasion of A549 cells in a concentration-dependent manner. CONCLUSION:The expression level of ALDOA in MPE is significantly higher than that in TBPE, especially in MPE from lung adenocarcinoma patients. There are highly positive correlations between ALDOA and CEA, ALDOA and LDH in pleural effusion. ALDOA concentration-dependently promotes the proliferation, migration and invasion of A549 cells.  相似文献

AIM:To observe the expression of vascular cell adhesion molecule-1 (VCAM-1) in cultured human umbilical vein endothelial cells (HUVEC) by lipid peroxidation injury induced by exposure to diamide.METHODS:Expression of VCAM-1 mRNA and protein in HUVEC was determined by in situ hybridization and a cell enzyme-linked immunosorbent essay (cell ELISA), respectively.RESULTS:The HPIAS-1000 image analytic system in situ hybridization detected that the mean absorbance values in experiment groups(1, 5 and 10 μmol/L diamide for 8 hours) were 0.147±0.013, 0.292±0.020 and 0.396±0.022, which were 1.91-fold, 3.79-fold and 5.14-fold as much as that of the control group (0.077±0.011), respectively. There was significant statistical difference between groups (P<0.01). The cell ELISA showed that the mean absorbance values of VCAM-1 proteins in experiment groups were 0.158±0.008, 0.220±0.017 and 0.321±0.023, which were 1.53-fold, 2.12-fold and 3.09-fold as much as that of the control group (0.104±0.016), respectively. The analysis of variance proved the significant difference between groups (P<0.01).CONCLUSION:The results suggest that the increased expression of VCAM-1 is integral in promoting adhesion of monocytes to the vascular endothelium.  相似文献

AIM:To investigate effects of OX-LDL and VitE on the levels of IL-6,IL-8 and TNF-α in human umbilical vein endothelial cells(HUVEC).METHODS: Human umbilical vein endothelial cells were obtained by in vitro culture. HUVEC treated with or without Vit E was incubated with OX-LDL, and the levels of IL-6, IL-8 and TNF-α were determined by enzyme-linked immunosorbent assy technique. RESULTS:50 μg/L,100 μg/L, 200 μg/L OX-LDL induced the release of IL-6,IL-8 and TNF-α by HUVEC in a dose-dependent manner. Compared with the control group , the levels of IL-6 and IL-8 were significantly increased at 6-12 h of stimulation with OX-LDL . Maximal levels of IL-6 and IL-8 occurred after 24-36 h, reaching a plateau maintained for at least 48 h. TNF-α rose after 2-6 h in HUVEC, and reached a maximum after 12 h. In contrast to IL-6 and IL-8, TNF-α declined after 48 h. However, when VitE (50 mg/L,100 mg/L,200 mg/L)was added, it can significant inhibited the release of IL-6, IL-8 and TNF-α in a dose-dependent manner, and after 48 h these cytokines have no diference between OX-LDL+VitE groups and OX-LDL groups. CONCLUSION: OX-LDL can obviously stimulate the production of IL-6,IL-8 and TNF-α in vascular endothelial cells, which can significantly be inhibited by VitE in a short time.  相似文献

AIM: To investigate the effects of eicosapentaenoic acid(EPA) on the expression of nuclear factor kappa B(NF-κB) and release of vascular endothelial growth factor(VEGF), IL-1α and IL-6 in cultured human umbilical vein endothelial cells(HUVECs) stimulated by lipopolysaccharide(LPS).METHODS: HUVECs were obtained from cell strain and cultured in vitro. HUVECs were divided into 4 groups: control group, LPS group, 0.030 g/L EPA treatment group and 0.050 g/L EPA treatment group. The cells were cultured with LPS alone in LPS group and incubated with EPA for 1 h in the EPA pretreatment groups at the concentrations of 0.030 g/L and 0.050 g/L before LPS stimulation. Twenty-four hours after stimulated by LPS, the protein expression of NF-κB p65 in HUVECs were assessed by Western blotting analysis at different time points. The production of VEGF, IL-1α and IL-6 in cultured HUVECs was evaluated by ELISA. The effects of EPA on the protein expression of NF-κB p65 and the production of VEGF, IL-1α and IL-6 in HUVECs challenged by LPS were also determined.RESULTS: Compared with control group, the protein expression of NF-κB p65 was significantly increased in HUVECs induced by LPS and was inhibited by EPA. Compared with control group, the protein expression of VEGF, IL-1α and IL-6 was dramatically increased in HUVECs induced by LPS and most of the increase was inhibited by EPA.CONCLUSION: LPS enhances the protein expression of NF-κB and the release of VEGF, IL-1α and IL-6. EPA inhibits the protein expression of NF-κB, and the production of VEGF and the inflammatory cytokines in cultured HUVECs stimulated by LPS, indicating that EPA may be useful for preventing and treating neovascular and inflammatory diseases.  相似文献
