AIM: To study the correlation between phasic change of the relative quantity of major crystallins with aging in rats. METHODS: Six groups of SD rats (age 1 d, 8 d,2 weeks,8 weeks,8 months and 1.5 years) were raised routinely. Water soluble crystallins were extracted and separated by two-dimensional polyacrylamide gelelectrophoresis. After comassize blue staining,the crystallins patterns were scanned and analyzed. RESULTS: (1) Out of the eighteen water soluble major rat crystallins tested in each group, seven showed gradual phasic changes in relative quantity of crystallins, but there were no significant changes in total quantity of water soluble crystallins. (2) Phasic changes in these crystallins presented four different patterns: increasing (βB4、αB2、αA2、βA1), decreasing (β7、β8、γ2,3、γ5,6),relatively stable(βA3、βB5), and irregular. (3) The ratio of βB4 /αA2 increased gradually with the rat aging process. CONCLUSION: The gradual phasic changes in relative quantity of crystallins reflect the aging status of rat crystalline.  相似文献

AIM: To study the effect of crystallin βB2 on the aging of lens. METHODS: SD rats were maintained routinely and killed at 6 different ages (1 d, 8 d, 2 weeks, 8 weeks, 8 months and 1.5 years). Water-soluble crystallins were extracted and separated by two-dimensional polyacrylamide gel electrophoresis (IEF/SDS-PAGE). After Comassize blue staining, the crystallin patterns were screened and analyzed. βB2 crystallin and the main chaperone proteins (αA2, αB2) were identified and the relative quantity was measured. RESULTS: (1) The quantity of water-soluble crystalline βB2 increased in close relation to the aging of the rat. (2) αA2, αB2 chaperone proteins increased with the aging of the rat too. (3) The change of the quantity of water-soluble crystalline βB2 was closely related to the changes of αA2 , αB2 chaperone proteins. (4) Degraded and modified crystallins began to appear clearly in the lens after 8 months old. CONCLUSION: Based on our results, we infer that water-soluble crystalline βB2 increases with the aging dof the rat, which is helpful to maintain the structure and transparency of the lens.  相似文献

AIM: To study the role of the gene and protein expression of MIP-1α and RANTES in the bronchus of murine asthma. METHODS: 20 male BALB/C mice were randomly divided into two groups: the control group (A0 group) and asthma group (B0 group). In the experiment, the mice model of asthma was established by the ovalbumin (OVA) challenge methods. The protein expression of MIP-1α and RANTES were detected by immunohistochemistry methods. The gene expressions of MIP-1α and RANTES were detected by in situ hybridization methods. RESULTS: Immunohistochemistry showed that the expressions of MIP-1α protein and RANTES protein around the bronchus of group B0 were significantly higher than those of group A0 (P<0.01), the epithelial cells were the chief expression cells; (2) In situ hybridization showed that the expressions of MIP-1α gene and RANTES gene around the bronchus of group B0 were significantly higher compared to those of group A0 (P<0.01), the epithelial cells were the chief expression cells. CONCLUSION: MIP-1α and RANTES are high expression in the bronchus epithelial cells in experimental murine asthma.  相似文献

AIM: To explore the effects of lipoxin A4 on the expression of cyclooxygenase 2 (COX-2) in human bronchial epithelial cells (HBECs). METHODS: HBECs were incubated with various concentrations (0.1, 1 and 10 mg/L) of lipopolysaccharide(LPS) for 9 h, or 1 mg/L LPS for different time (3 h, 6 h and 9 h). The levels of COX-2 mRNA in HBECs and prostaglandin E2 (PGE2) in the culture supernatant were measured. In addition, the HBECs were exposed to lipoxin A4 at concentration of 0, 100 and 400 μmol/L after stimulated with LPS at concentration of 1 mg/L for 9 h, and the supernatant of the culture cells was collected for determining the content of PGE2 by ELISA. The cells were also harvested, and the mRNA and protein levels of COX-2 were analyzed by RT-PCR and Western blotting, respectively. RESULTS: LPS increased the mRNA expression of COX-2 and production of PGE2 in a dose and time dependent manners in HBECs. Induction of COX-2 mRNA and protein by LPS were inhibited by lipoxin A4 in a dose-dependent manner. Lipoxin A4 also significantly decreased LPS-induced production of PGE2. CONCLUSION: Lipoxin A4 down-regulates LPS-induced expression of COX-2 and consequently inhibits the production of PGE2 in HBECs.  相似文献

在单因素试验的基础上确定正交实验的因素水平,采用L9(34)正交实验,以多糖提取率为指标,对浸提温度(A)、料液比(B)、浸提次数(C)进行优化研究。结果表明:影响"洛阳红"牡丹叶片多糖提取率因素的主次关系为B>C>A,最佳提取条件为A2B3C2,即料液比为1 g:30 mL,浸提温度70℃,浸提2次,浸提时间为30 min。试验优选结果稳定可行。  相似文献

选择耐盐性不同的6份砧用南瓜(Cucurbita moschata)自交系为材料,采用完全双列杂交法对杂交后代耐盐性的配合力进行分析,研究砧用南瓜耐盐性遗传规律。结合各配合力的分析,在36个配制组合中,选出较为优良的配制组合18C0077×18C0005、18C0077×18C0046、18C0077×18C0049、18C0024×18C0046、18C0024×18C0049、18C0024×18C0026。同时,以耐盐性强的砧用南瓜自交系18C0077(P_1)和耐盐性弱的砧用南瓜自交系18C0005(P_2)配制得到的6个世代(P_1、P_2、F_1、B_1、B_2和F_2)为材料,利用数量性状遗传模型进行砧用南瓜耐盐性的遗传分析。结果表明,砧用南瓜耐盐性的遗传符合"2对加性显性-上位性主基因+加性-显性多基因"模型。砧用南瓜各世代耐盐性主基因遗传率在34.62%~62.08%之间,以主基因遗传为主,且主基因遗传力在B1中最高,适合在早期世代进行选择。  相似文献

AIM: To investigate the effect of human amniotic fluid on the release of thromboxane A2 (TXA2), prostaglandin I2 (PGI2) and Leukotriene C4 (LTC4) from blood cells. METHODS: 1 mL human amnioticfluid and 10 mL oneself blood collected from 38-41 weeks with cesarean section were cultured at 37 ℃ for 30 min, and then centrifuged. The supernatants were taken and stored at -70 ℃. TXB2 and 6-Keto-PGFof the superntants were determined by radioimmunoassay and LTC4 by enzyme immunoassay. RESULTS: It was found that the levels of TXB2 and LTC4 in blood were elevated from (63.5±52.0)ng/L and (40.1±39.2) ng/L to (189.1±102.0) ng/L and (293.5±206.1) ng/L, respectively (P<0.01), after cultivation of oneself amniotic fluid with blood, the concentrations of TXB2 and LTC4 were higher in meconium-stained group than those in clear amniotic fluid group, but the concentration of PGI2 only elevated slightly from (27.4±11.6) ng/L to (33.9±10.6) ng/L (P>0.05). CONCLUSION: Amniotic fluid might stimulate the release of TXA2 and LTC4 from blood, it might affect the balance of TXA2 and PGI2 in blood, which might play an important role in the pathogenesis of amniotic fluid embolism.  相似文献

AIM: To investigate the mechanisms by which paeoniflorin (Pae) attenuates lipopolysaccharide(LPS)-induced acute lung injury in mice. METHODS: Male BALB/c mice were randomly divided into 4 groups: control,LPS, Pae+LPS, and Pae. Mice were administered intragastrically with double distilled water or Pae (20 mg/kg) once a day for 3 days. One hour after intragastrical treatment on the third day, LPS (20 mg/kg) or normal saline was injected intraperitoneally. Twelve hours after LPS challenge, the histological changes of the lung were observed, and histology score was also assessed. The myeloperoxidase (MPO),cytosolic phospholipase A2 (cPLA2) and phosphorylated cytosolic phospholipase A2 (phospho-cPLA2) in lung tissues were detected by Western blotting.RESULTS: LPS challenge resulted in acute lung injury, activated cPLA2 and increased MPO content in lung. Pretreatment with paeoniflorin significantly attenuated lung injury induced by intraperitoneal injection of LPS. The levels of MPO and phospho-cPLA2 in the lung tissues of the mice in Pae+LPS group were lower than those in LPS group (P<0.05).CONCLUSION: Pretreatment with paeoniflorin remarkably reduces LPS-induced acute lung injury through inhibiting phosphorylation of cPLA2 and decreasing neutrophil infiltration in the lung. These findings provide a new strategy for the prevention and treatment of LPS-induced acute lung injury.  相似文献

AIM:To explore the effects of oxidized low density lipoprotein (OxLDL) and one of its component— lysophosphatidylcholine (LPC) on cholesterol efflux from mouse macrophage foam cells.METHODS:(1) Cholesterol efflux induced by apoAI from mouse peritoneal macrophage foam cells loaded with OxLDL or acylated LDL(AcLDL) was measured. (2) Cholesterol efflux induced by LPC and apoAI from macrophage foam cells separated from normal or apoE gene deficient (E0) mouse loaded with AcLDL were measured.RESULTS:(1) When the macrophage foam cells were incubated with apoAI, cholesterol efflux from AcLDL-induced macrophage foam cells increased significantly compared to that of OxLDL-induced macrophage foam cells. (2) LPC promoted cholesterol efflux from macrophage foam cells in relation to both dosage and time. When LPC was incubated with E0 mouse macrophage foam cells, the released cholesterol mass was significantly lower than that of normal mouse macrophage foam cells. It was also found that cholesterol efflux induced by apoAI normally occurred in E0 mouse macrophage foam cells.CONCLUSIONS:(1) OxLDL accumulated cholesterol in macrophages and impair cholesterol efflux. (2) LPC induced cholesterol efflux from macrophage foam cells, which may occur via apoE pathway.  相似文献

AIM: To investigate the influence of spirolactone (SPI) on mouse pulmonary macrophage subtype switching induced by silicon dioxide (SiO2). METHODS: C57BL/6 mice were divided into normal saline (NS) group, SiO2 group, SiO2+SPI group and SiO2+NS group. A mouse model of silicosis was developed with crystalline SiO2 particles (40 mg/kg, via oropharyngeal instillation). SPI (20 mg/kg) or (NS) was delivered daily by oral gavage after SiO2 administration. The mice were sacrificed on days 3, 14 and 28. Alveolar washing, total cell counting, differential cell counting and flow cytometry analysis were performed. The right lower lobe of lavaged lungs was collected to prepare single-cell suspension for flow cytometry analysis. RESULTS: Compared with SiO2 group, SPI treatment significantly alleviated SiO2-induced inflammatory cell accumulation in brochoalveolar lavage fluid on day 3 and 14. Compared with NS group, the M1 alveolar macrophages and M1 pulmonary interstitial macrophages in SiO2 group switched to M2 subtype dramatically, while SPI treatment reversed the switching effectively.CONCLUSION: SPI treatment alleviates SiO2-induced inflammatory cell accumulation, which may be related to reversing macrophage subtype switching.  相似文献

AIM: To observe the kinetic alteration of nitric oxide formation in the lungs in the development of pulmonary fibrosis in the rat. METHODS: The contents of hydroxyproline in the lungs, NO2-/NO3- (nitrite/nitrate) in out-flowing and in-flowing pulmonary blood (OPB, IPB) were assayed on the day 7, 14, 21, 30 and 70 after intratracheal administration of bleomycin A5 . The content of NO2-/NO3- in supernatants of culture of the alveolar macrophages (AMs) and the amount of iNOS positive stain cells in lung tissue section were also observed in the rat on 14th day after-bleomycin A5 administration. RESULTS: The content of lung hydroxyproline had no change on the 7th day, increased on the 14th day (P<0.05), increased significantly on the 21th day, 30th day and 70th day post-bleomycin A5 compared with control rats. On the 7th day and 14th day, the content of NO- 2 /NO3- increased in OPB and decreased in IPB (P<0.01). On the 21th day, the content of NO2-/NO3- abated in OPB (P>0.05) but still decreased in IPB (P<0.01). On the 30th day and the 70th day, the NO2-/NO3- level recovered both in OPB and IPB. AMs from rats on the 14th day post-bleomycin A5 showed significant elevation (P<0.01) in NO2-/NO3- level. The amount of iNOS positive stain cells increased in rats on the 14th day post-bleomycin A5. CONCLUSION: The amount of NO in the lungs was high in the initial phase of fibroproliferative reaction induced by bleomycin A5 ,and these might be associated with the enhanced ability of AMs to release NO and the increased amount of iNOS.  相似文献

AIM:To study the relationship between the activity of phospholipase A2 (PLA2) and chronic hypoxic pulmonary hypertension. METHODS:29 healthy SD rats were randomly divided into normal control group, chronic hypoxic group and hypoxia plus Polidatin (PD) group. The model of rat chronic hypoxic pulmonary hypertension was made by method of intermittent isobaric hypoxia for 21 days. The mean pulmonary arterial pressure (mPAP) was measured by inserting a microcatheter into the pulmonary artery. RESULTS:After exposing hypoxia for 21 days, the mPAP, R/L+S, the PLA2 activity, TXB2, MDA in plasma and lung homogenate increased significantly, while 6-k-PGF, SOD decreased significantly. Pretreatment with PD could relieve the changes mentioned above.CONCLUSION:PLA2 plays an important inducing role through its metabolic products and the interactional radicals in the formation of chronic hypoxic pulmonary hypertension.  相似文献

观赏海棠品种群开花进程中色素组分动态特征研究   总被引:3,自引:0,他引:3  
 以58个观赏海棠(Malus sp.)品种为研究对象,对大蕾期(S1)、盛开期(S2)、末花期(S3)花瓣的色素组分进行了测定,研究了海棠品种群之间色素组分关系及动态规律,为挖掘高花青素含量及高稳定性的海棠特异种质和进行花色改良育种提供参考。基于UV-2450紫外—可见分光光度计的实际含量测定和基于Unispec-SC光谱分析仪的反射光谱指标测定,分别构建了花青素、类胡萝卜素和叶绿素3大类色素组分含量与反射光谱指标之间的耦合函数:yAnth = 0.014x + 0.0012(R2 = 0.9974);yCar = 0.0038x0.3772(R2 = 0.9902);yChl =–0.1007x2 + 0.3221x–0.2169(R2 = 0.8179)。采用Origin7.0软件构建了观赏海棠品种群3大类色素组分含量和权重的三维动态图,结果表明:花青素为海棠花瓣中的主导色素,类胡萝卜素和叶绿素为次要色素;在开花进程中,海棠品种群花瓣中的花青素、类胡萝卜素和叶绿素含量整体呈大幅度降低趋势,但色素组分权重却保持着相对稳定性,尽管花青素的权重呈现小幅下降趋势,而类胡萝卜素和叶绿素的权重却呈现小幅上升趋势。基于花青素含量和权重的聚类分析表明:在遗传距离2.936处,58个海棠品种可划分为5个类群,即白色系(A)、粉色系(B)、浅紫红色系(C)、紫红色系(D)和深紫红色系(E),其中C、D、E色系构成泛紫红色系。各色系/子色系类群之间具有明显不同的色素组分动态特征。尽管粉色系之B2子色系在大蕾期具有较高的花青素含量[(0.090 ± 0.038)mg · g-1],但由于色素稳定性差,在S1–S2–S3开花进程中,花青素含量分别下降了83.2%和93.3%,其“红艳”观赏期仅限于盛花期之前;粉色系之B1子色系的花青素含量及稳定性皆显著高于B2子色系,在S1、S2和S3阶段,B1子色系的花青素含量分别为B2子色系的1.7、3.3和3.2倍,显著提高了色彩观赏性。与粉色系相比,泛紫红色系具有更高的花青素含量及稳定性,整个花期皆保持紫红色,显著延长了该类群的“红艳”观赏期,在S1、S2和S3阶段,D、E两个色系的花青素含量分别为粉色系均值的1.8 ~ 2.0倍、3.9 ~ 6.5倍和8.1 ~ 10.0倍,甚至在S3阶段仍达到B色系在S1阶段的花青素含量水平;C色系的花青素含量及稳定性皆逊色于D、E色系,但仍显著优于B色系,即使在S3阶段,C色系的花青素含量仍达到B色系在S1阶段的43.2%和S2阶段的152.5%。  相似文献

AIM:To study the effect of human complement C5b~9 complex on nitric oxide(NO) synthesis of glomerular mesangial cells (MC). METHODS: First, the human complement C5b~9 complexes were isolated and glomerular MC of rats were cultured. Second, the MC were stimulated with C5b~9 complex and changes of metabolism products of NO(NO3- and NO2) in MC culture supernatant at 6,24 and 48 hours after C+5b~9 stimulating were detected. Moreover, cGMP levels in cultured MC were also measured. RESULTS: NO3-/NO2- contents from culture supernatant and cGMP levels in MC were increased parallelly after C5b~9 complex stimulation. Further, NO synthesis was inhibited by L-NG-nitro-arginine-methylester(L-NAME). CONCLUSION: NO synthesis of rat glomerular MC was incerased by human complement C5b~9 stimulation.  相似文献

Chiku fruits (Achras sapota L.) were found to be climacteric with the respiratory peak occurring at the same time, or 1–2 days after peak ethylene production. The optimum storage temperature was near 20° C. Storage life at this temperature could be increased by removing ethylene (C2H4) and adding 5–10% (v/v) carbon dioxide (CO2) to the storage atmosphere. Extreme relative humidities or high concentrations of CO2 impaired the quality of stored fruits. Ripening was not affected by treatment with oxygen, C2H4 or indoleacetic acid. Ascorbic acid and glucose levels increased with ripening but ascorbic acid decreased when the fruit became over ripe. Recommended storage conditions are about 20° C with 5–10% CO2 coupled with the complete removal of C2H4 from the storage atmosphere. Short-term holding of the fruit at lower temperatures is also possible.  相似文献
