Metabolic and pathophysiologic changes of secondary ketosis were studied. Plasma samples were obtained from a group of 8 mature, lactating dairy cows before, during, and after a 48-hour fast. These samples were analyzed for glucose, free fatty acids (FFA), lactic acid, ketone bodies, glucocorticoids, insulin, and growth hormone concentrations. In the prefasting period, metabolites and hormones remained constant. Lactic acid and glucocorticoids also remained stable during the entire experiment, except on the 1st day after termination of the fast, when glucocorticoids increased significantly (P less than 0.05). Glucose values decreased significantly (P less than 0.01) with the onset of fasting, began to rise halfway through the fast, and were elevated following termination of the fast. Plasma glucose concentration returned to normal by the end of the experiment. Plasma FFA concentrations increased during the early portions of the fast and decreased thereafter. Plasma ketone body concentration responded similarly, but the change occurred approximately 12 hours after the plasma FFA changes. A value determined as ketotic was reached during the fast (14.6 +/- 1.3 mg/dl). Insulin concentrations remained low during the fast and increased after fasting. Plasma growth hormone concentration increased to a new plateau in response to fasting and then decreased somewhate after fasting was ended.  相似文献

Transition from pregnancy to lactation in dairy cows involves considerable metabolic adaptation. Additional stress is incurred during infections such as periparturient mastitis. Multiparous Holstein-Friesian cows kept under normal production conditions (n = 15) were used to evaluate changes in circulating metabolite and hormone concentrations from 5 days before to 5 days after calving. Insulin-like growth factor binding protein (IGFBP) profiles were also monitored. Marked time-related changes were observed for plasma thyroid hormone, IGF, cortisol, insulin, beta-hydroxybutyrate (BHB) and non-esterified fatty acid (NEFA) concentrations but not for plasma leptin. A decrease in IGF-II concentration and maximal intensity of the putative IGFBP-1 band occurred at parturition. When compared with the five healthy cows,low IGF-II levels were prolonged to day 2 post-partum in five cows with Escherichia coli-associated mastitis. However, marked decreases in IGFBP-2 band intensity were evident only in two of the four cases examined. Individual total ligand (IGF-I + IGF-II) concentration and IGFBP pattern prepartum were largely regained 5 days post-partum in all cows. Hormone and metabolite concentrations in the two cows with Staphylococcus aureus-associated mastitis were very similar to those in the five healthy cows. Plasma thyroxine (T4) was lower 2 days prepartum in the cows, which later developed Gram-negative mastitis. Multiregression analysis showed that variance in T4 concentration was significantly and independently associated with triiodothyronine (T3) and IGF-I positively and with cortisol negatively (R2 = 0.648). This study confirms the close inter-relationship between the thyroid hormone and IGF axes in cattle and indicates possible effects of Gram-negative mastitis infection on IGF-II metabolism.  相似文献

对某奶牛场16头乳牛产前10d至产后56d血样的瘦素、酮体、葡萄糖、脂肪水平及其动态特征和相关性进行了检测。结果表明,血浆瘦素、血酮、血脂含量在不同乳牛和不同时间点之间均有显著差异。酮体在产后14~49d出现高峰,瘦素和血脂从产后第0d到产后第56d内逐渐升高。瘦素水平与血脂水平呈极显著正相关,血酮与血糖呈极显著负相关。亚临床酮病组乳牛的瘦素和血脂水平分别极显著或显著低于血酮正常组,其瘦素、血酮和血糖在试验期内的波动频率明显减少,跨度变长,瘦素在试验期内仅出现一个两端接近0、波峰不超过1.5ng/mL的波。证实,产后乳牛瘦素、血酮、血糖含量的低频率波动变化和产后8周内血浆瘦素跨时持久的单峰动态变化与乳牛亚临床酮病的发生有关。  相似文献

Blood samples were obtained sequentially from 10 dairy cows around the time of parturition to assess plasma fluctuations in estradiol-17β (E2) levels in association with those of several bone resorption markers. Plasma E2 concentration increased sharply a few days prepartum and decreased quickly after parturition. In terms of bone resorption markers, the plasma level of tartrate-resistant acid phosphatase isoform 5b (TRAP5b) rose significantly, commencing 1 week prepartum, and was maintained at this level to a few days postpartum. The plasma concentration of carboxyterminal collagen cross-links of type-I collagen (CTx) increased significantly after parturition. These observations suggest that osteoclast-mediated bone resorption was activated after parturition when plasma E2 concentrations decreased.  相似文献

The effect of cracked corn grain supplementation (3.5 kg/day) during 3 weeks before the expected calving date on milk production and composition, body condition score (BCS), metabolic and hormonal profiles and length of postpartum anoestrus was evaluated in multiparous Holstein dairy cows under grazing conditions (Energy supplemented group, n = 10; Control group, n = 10). Body condition score was weekly recorded during the peripartum period, from days −21 to +35 (parturition = day 0). Non-esterified fatty acids, β-hydroxybutyrate, cholesterol, urea, insulin, insulin-like growth factor I (IGF-I), leptin, thyroxine (T4) and 3,3'5-triiodothyroinine (T3) were weekly determined in plasma from days −21 to +35. The reinitiation of ovarian cyclicity was twice weekly determined by ovarian ultrasonography and confirmed by plasma progesterone concentrations. Cows fed energy concentrate prepartum had higher BCS during the prepartum and postpartum and produced more milk. Non-esterified fatty acids plasma concentrations were significantly higher in the energy group, while cholesterol was higher in the control group. Treated cows had higher levels of plasma insulin, IGF-I and leptin pre-calving. IGF-I, leptin and T4 were diminished during the early postpartum period in both groups. Insulin levels were also diminished in the control group, but levels remained high in the energy-supplemented group. Treated cows ovulated sooner after parturition than controls. We conclude that Energetic supplementation prepartum in cows under grazing conditions increased milk production and reduced the reinitiation of ovarian activity, consistent with a better EB (BCS), higher prepartum levels of IGF-I, leptin and insulin, and higher insulin levels during early postpartum.  相似文献

Plasma concentration stability of glucose, free fatty acids, ketone bodies, growth hormone, insulin were determined in lactating dairy cows. Concentrations of these metabolites and hormones were measured during a 36- to 48-hour period in 3 normal, mature dairy cows in the 2nd month of lactation. Samples were taken at 30-minute intervals; also, intensive sampling (every 10 minutes) was done at varying times in relation to feeding and milking. Of the 5 components measured, glucose concentration was the most stable, easiest to assay, and most reliable for use as a diagnostic aid in assessing metabolic carbohydrate disturbances in dairy cattle.  相似文献

To investigate the effect of hoof trimming before the dry period, the hooves of 10 cows (trimmed group) were trimmed at 79.6 +/- 8.6 days before parturition and the hooves of 52 cows were left untrimmed (control group). Blood biochemistry and hormone concentrations were investigated for 6 cows from each group. The daily milk yield after parturition in the trimmed group tended to be higher than that of the control group. Between 0 and 30 days after parturition, the levels of non-esterified fatty acids were significantly lower stet, and the plasma glucose and glucose disposal rates were significantly higher in the trimmed group. The plasma cortisol concentration was stable before and after parturition in the trimmed group. Hoof trimming before the dry period appears to reduce stress and maintain the nutritional conditions of perinatal dairy cows.  相似文献

OBJECTIVE: To compare plasma total calcium, phosphorus, magnesium, nonesterified fatty acids (NEFA), beta hydroxy butyrate (BHB), and glucose concentrations in parturient dairy cows that were fed an anionic prepartum diet between those with and without retained fetal membranes (RFM) at 24 hours after parturition. ANIMALS: 152 Holstein cows that calved during October through December of 1997 PROCEDURE: All cows were fed an anionic prepartum diet. Blood sample was taken within 6 hours after parturition from randomly selected cows. Thirty-nine cows had a diagnosis of RFM at 24 hours after parturition; 113 were not affected with RFM. At calving, body condition score (BCS; 1 [thin] to 5 [obese]), parity, and calving difficulty score were recorded. Plasma calcium, phosphorus, magnesium, NEFA, BHB, and glucose concentrations were compared between cows with or without RFM. RESULTS: Cows with RFM had significantly lower plasma calcium concentration soon after calving, compared with cows without RFM. Cows with a parity of > or = 3 had significantly lower plasma concentrations of calcium and higher concentrations of magnesium, compared with cows with a parity of 1 or 2. Cows with a BCS of > or = 3.25 at calving had significantly higher plasma concentrations of BHB than cows with a BCS of 2.75 to 3.0. Cows with dystocia had significantly higher plasma concentrations of glucose, compared with cows without dystocia. CONCLUSIONS AND CLINICAL RELEVANCE: In parturient cows fed a prepartum anionic diet, those with RFM have lower plasma calcium concentrations than cows without RFM, although this association does not prove a cause-effect relationship.  相似文献

Although our previous report demonstrated that adiponectin and AdipoR1 gene expressions changed among different lactation stages in the bovine mammary gland, its in vivo kinetics remain unclear in ruminant animals. In this study, we investigated the changes in circulating concentrations of adiponectin, as well as other metabolic hormones and metabolites, (i) during the periparturient period and (ii) among different lactation stages, in Holstein dairy cows. In experiment 1, serum adiponectin concentrations increased after parturition. Serum insulin concentrations were lower in the postpartum than prepartum period, whereas serum growth hormone (GH) concentrations increased in the postpartum period. Serum nonesterified fatty acids (NEFA) levels were increased during the postpartum period and were dependent on the parity. In experiment 2, there was no significant difference in plasma adiponectin concentrations among lactational stages. Plasma insulin concentrations tended to be lower in early lactation while plasma GH levels tended to be higher. Plasma NEFA concentrations were significantly lower in mid‐ and late‐lactation stages than non‐lactation stages. These findings indicate that elevation of serum adiponectin might be involved in energy metabolism just around parturition, and might exert its action through regulation of receptor expression levels in target tissues in each lactational stage in Holstein dairy cows.  相似文献

试验旨在揭示围产期奶牛血清代谢物和相关激素水平的动态变化规律,选取临床健康经产奶牛 20 头,分别于产前 14、7 d 及产后 0、7、14 d 采血并分离血清,检测血清葡萄糖、甘油三酯、非酯化脂肪酸、β-羟丁酸等代谢物浓度和胰岛素、生长激素、脂联素和瘦素等激素水平,并根据相应公式计算胰岛素敏感指数。结果表明:奶牛分娩当天的血糖浓度较高,而后逐步恢复至产前水平;奶牛分娩后甘油三酯浓度下降,非酯化脂肪酸和β-羟丁酸浓度升高;胰岛素浓度在产前 14 d 至产后 7 d 各时间点无显著变化;产前 7 d 时的生长激素浓度高于其他时间点(P<0.01);脂联素浓度在整个试验期无显著变化;瘦素水平在产前 7 d 和产后 0 d升高(P<0.01),但产后 7 d 时下降(P<0.01);胰岛素敏感性指数在分娩当天显著降低,随后逐步恢复。结果表明,奶牛产后体脂动员明显,可能与生长激素、瘦素等激素的调控有关。  相似文献

The effect of selection for milk yield on lactation yield, net energy balance, and on plasma growth hormone, insulin, prolactin, nonesterified fatty acids and glucose was studied in two groups of primiparous Holstein cows of differing genetic merit. Net energy balance was calculated and serial blood samples were collected for a 7 hr period at 0, 45, 90 and 180 days postpartum. Growth hormone releasing factor (.2 μg/kg BW) was administered after 2.5 hr at 0, 45 and 180 days postpartum, while epinephrine (.7 μg/kg BW) was administered at 90 days postpartum, Milk yield was greater, net energy balance was decreased and plasma growth hormone was greater in genetically superior selection group cows compared to control cows.

Growth hormone showed similar increases in both genetic groups in response to growth hormone releasing factor, while prolactin, insulin and glucose were not altered. Epinephrine stimulated an increase in plasma nonesterified fatty acid, glucose and insulin, but responses did not differ between genetic groups. Results indicate differences exist in production efficiency, net energy balance and plasma growth hormone concentration among dairy cattle as a result of selection for milk yield and suggest that selection pressure may act to alter homeorhetic control of nutrient metabolism.  相似文献

The effect of an acid or alkali diet was investigated in 14 mature dairy cows during the last 28 days of pregnancy. The acid diet reduced the incidence of parturient hypocalcaemia compared with the alkali diet and was associated with higher blood ionised calcium and plasma chloride concentrations and lower blood pH and acid-base excess before parturition. Plasma 1,25-dihydroxyvitamin D concentrations before parturition were increased by the acid diet but the concentrations of parathyroid hormone and intact 1–86 parathyroid-hormone-related peptide were unaffected. The estimated fractional calcium absorption and calcium mobilisation rate during an ethylenediamine tetraacetic acid infusion were increased by the acid diet 14 days before parturition. Cortical bone remodelling occurred in all the animals during late pregnancy but was particularly evident in the cows given the acid diet. The data suggest that an acid diet ameliorates parturient hypocalcaemia by enhancing calcium mobilisation before parturition by increasing calcium absorption and bone resorption, these increases possibly being mediated by increases in the plasma concentration of 1,25-dihydroxyvitamin D.  相似文献

Ingestions of a moderately ketogenic silage twice daily were followed by transient increments in plasma insulin and ketone bodies and decreases in plasma glucose. Ketone bodies and glucose were negatively correlated throughout the day, but the insulin elevations culminated before the maximal effects on ketone bodies and glucose were established. Cows with varying glucose levels before morning feeding reacted to a highly ketogenic silage by decreasing their glucose level uniformly to about 3 mmol/l, in spite of a widely varying feeding-induced insulin increment. Hay-feeding caused insulin increments of the same magnitude as silage-feeding, but the glucose decrease and the ketone increment was much smaller. The results indicate some direct action of ketone bodies on blood sugar regulation, in addition to effects mediated by insulin. The role of ketone bodies as the insulinotropic factor was not confirmed. The insulin level after feeding seems to be determined by the carbohydrate status of the animal before feeding. No significant changes in plasma glucagon were observed after feeding, and no consistent differences in plasma levels of this hormone were found when non-ketonemic, ketonemic, and clinically ketotic cows were compared. The plasma level of enteroglucagon (GLI) was positively correlated to the relative amount of concentrates consumed, but no relation to plasma glucose was found.  相似文献

Plasma concentrations of prolactin (PRL), growth hormone (GH), insulin, glucagon, glucose, urea and free fatty acids (FFA) were measured in Holstein calves, yearlings, bred heifers and primiparous cows, either sired by bulls with high predicted differences (PD) for milk (selection group) or by bulls from an unselected random bred control population (control group; n = 6). Serial blood samples were collected before and after feeding for an 8-h period from 0900 to 1700 h. All animals were fed a complete feed at 1100 h and administered insulin (.6 IU/100 kg body weight) at 1400 h. Mean plasma PRL was greater in control animals after feeding and insulin administration, while GH was greater overall in selection cattle. Insulin remained elevated longer in selection animals after exogenous administration, and plasma glucagon was increased in the control group. While plasma glucose and urea were unaffected by genetic group, plasma FFA were elevated in selection group calves and primiparous cows compared with the control group. All hormones and metabolites differed among the pre- and post-feeding and insulin administration periods and also with age. Mean PRL and GH increased after feeding, while glucagon decreased after exogenous insulin. Plasma FFA declined after feeding, while urea and glucose were similar before and after feeding. Mean PRL increased and glucagon decreased with advancing age and plasma GH and insulin showed inverse relationships at different ages. Plasma FFA changes closely followed GH changes with age, while plasma glucose more closely followed insulin changes with age. Results indicate that all hormones measured and FFA responded to genetic selection for milk, and increases in GH are uniformly associated with increased genetic potential for milk yield.  相似文献

In peripartum dairy cows, insulin resistance (IR) increases to adjust the direction of energy to lactation after calving. To investigate the effect of prepartum IR on postpartum reproductive performance, the insulin tolerance test (ITT) was applied to 15 cows at 3 weeks (Pre21) and 10 days (Pre10) before the predicted calving date. Blood glucose area under the curve (AUC(glu)) within 120 min after administration of 0.05 IU/kg-BW insulin was calculated. The occurrence of first ovulation, days to first artificial insemination (AI) and first AI conception rate were recorded. Nutritional status postpartum was evaluated by blood chemical analysis. Based on AUC(glu) changes from Pre21 to Pre10, cows were classified into either the AUC-up group (AUC(glu) increase, n=5) or the AUC-down group (AUC(glu) decrease, n=10). There was no difference in the decrease in blood glucose at 30 min after insulin injection between groups, although glucose recovery from 30 to 60 min during the ITT was slow at Pre10 in the AUC-up group. The AUC-up group had a higher number of days to first AI and high glucose, total protein, globulin, γ-glutamyltransferase, triacylglycerol levels and a low albumin-globulin ratio at the 14th day postpartum. The present study infers that prepartum slow glucose recovery rather than insulin sensitivity might increase the potential for subclinical health problems postpartum and thus suppress reproductive performance. During the prepartum transition period, glucose dynamics in the ITT can be considered as a new indicator for the postpartum metabolic status and reproductive performance of dairy cows.  相似文献

The study was performed in eight high-pregnant cows in which medium to high ketonuria was repeatedly detected. The control group consisted of six cows free from ketonuria before parturition. Blood and urine were sampled from the cows two to eight weeks before parturition, on the day of parturition, 24 hours, three to five days and two to six weeks after parturition. The concentrations of oxidated ketone bodies and beta-hydroxybutyrate were determined by the distilling method. Further, the values of clinico-biochemical parameters in the urine and blood were determined within the range of the preventive diagnosis system. No marked changes in the health condition were found by the current clinical examination. The average concentration of total ketone bodies in the blood of the cows two to eight weeks before parturition was 29.2 mg per litre; the respective values recorded in the control cows were lower by about a half. The concentration of ketone bodies in the blood of the cows in the experimental group decreased on the day of parturition. The decrease was followed by a gradual rise of the total ketone bodies content in the blood up to 50.6 mg per litre two to six weeks after parturition. Alterations testifying to a disorder in the function of liver were observed in the cows of both groups throughout the period under study. The disorder was due to excessive administration of crude protein to the high-pregnant cows combined with the use of low-quality silage. After parturition the feed ration was not differentiated according to performance and was deficient in the basic nutrients and vitamins. Ketonuria was obviously due to the simultaneous intensification of hepatal and alimentary ketogenesis, caused by hepatopathy in combination with the intake of large amounts of poor-quality silage containing butyric acid.  相似文献

The hypocalcaemia caused by parturition and onset of lactation in high-production dairy cows was mimicked by subjecting cows to starvation periods before and after partus. The changes in plasma calcium, phosphate and magnesium were followed and compared with immunoreactive parathyroid hormone (iPTH) in 2 cows. During the starvation periods before partus, the cows developed hypocalcaemia with no or only small changes in the plasma concentration of magnesium. After the onset of hypocalcaemia, the concentration of iPTH increased on the average 3–4-fold and the raised hormone levels lasted about 24 h after start of refeeding. An increase in plasma phosphate occurred somewhat later than the rise in iPTH and lasted longer. After partus hypocalcaemia developed, together with smaller increases in iPTH concentration (about 2-fold). The post-partum starvation period again resulted in hypocalcaemia and raised iPTH concentrations.In conclusion, starvation and parturition induced inverse changes in plasma calcium and iPTH in dairy cows. The increases in plasma iPTH were reversible and considered secondary to the hypocalcaemia. Through the effect of paratyroid hormone, plasma calcium was normalized and phosphate concentration increased. Therefore, fatal hypocalcaemia which may occur following the course of parturition and onset of lactation is not due to impaired PTH secretion.  相似文献

For cows on d 137 (n = 6), 180 (n = 8), 226 (n = 9) and 250 (n = 5) of gestation (Exp. 1), concentrations of insulin and glucose were two- to three-fold less (P less than .01) in fetal venous plasma than in uterine arterial plasma. Concentrations of growth hormone, conversely, were 10- to 20-fold greater (P less than .01) in fetal venous than in uterine arterial plasma. Concentrations of insulin and glucose in maternal and fetal plasmas and concentrations of growth hormone in maternal plasma did not vary with stage of gestation. Concentrations of growth hormone in fetal venous plasma, however, were greater on d 226 and 250 than on d 137 and 180. For cows (n = 6) on d 198 of gestation (Exp. 2), concentrations of insulin and glucose in maternal and fetal plasmas and of growth hormone in maternal plasma remained relatively constant in samples collected every 30 min for 3 h. In contrast, growth hormone concentrations in fetal venous plasma were highly variable and appeared to be episodic, with pulses of 10 to 60 ng/ml in amplitude. No significant correlations were found among concentrations of insulin, glucose and growth hormone in fetal venous plasma. When samples were collected every 15 min for 4 h from cows (n = 5) on d 198 of gestation (Exp. 3), episodes of growth hormone in fetal venous plasma were irregular in amplitude and frequency.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The possibility that alterations in liver function may occur during late pregnancy as well as after calving has been investigated in healthy dairy cows and the results compared with those from non-pregnant non-lactating cows. There were significant alterations in plasma total bilirubin, glucose, total ketone body and urea concentrations, in bromsulphthalein clearance and in plasma aspartate aminotransferase and creatine kinase activities in periparturient cows compared with non-pregnant non-lactating cows. Of these, only the alterations in glucose, ketone bodies and urea concentrations and bromsulphthalein half-time and retention were markedly different before calving. The degree of fatty infiltration of the liver was significant two weeks before as well as two weeks after calving which differed from previous reports. It seems likely that the changes in the liver in dairy cows are functional and reversible and related to the metabolic demands of late pregnancy and early lactation. The results suggest that such changes in the liver occur well before calving.  相似文献

We investigated the changes in the plasma concentrations of oxidative stress markers in periparturient dairy cows in hot conditions. This study was performed on 10 cows from day ?5 to day 10 after parturition. The experimental duration was divided into three periods: period 1 (5 days before parturition); period 2 (5 days after parturition); and period 3 (from day 6 to day 10 after parturition). The average rectal temperature in the cows during the experimental period was more than 39°C. Plasma aspartate aminotransferase activities in periods 2 and 3 were higher than that in period 1 (P < 0.05). Blood urea nitrogen concentration in period 3 was lower than those in periods 1 and 2 (P < 0.05). The total ascorbic acid concentration in the plasma in period 2 was greater than those in periods 1 and 3 (P < 0.05). Thiobarbituric acid reactive substances (TBARS) concentrations in periods 2 and 3 were greater than that in period 1 (P < 0.05). TBARS concentration was positively correlated with aspartate aminotransferase activity (P < 0.05). These results suggested that oxidative stress increased in cows after parturition under hot climatic conditions.  相似文献
