Changes in the contents and composition of polyphenolics and resulting antioxidant activities of S. verbenaca by-products were investigated at three phenological stages (flowering, early fruiting and late fruiting stages). The highest accumulation of total phenolics was detected at the flowering stage (58.36 mg GAE/g DW). HPLC analysis of methanolic extracts showed the prevalence of methyl carnosate (821.45–919.82 μg/g DW) and rosmarinic acid (544.51–649.26 μg/g DW). Phenolic diterpenes (1056.90–1148.42 μg/g DW) was the most represented class of compounds. Three complementary tests namely, DPPH? (IC50 value, 49.22 μg/mL) and ABTS?+-radical scavenging assays (146.86 μM TE/mg) and FRAP reducing power test (188.93 mM Fe(II)/mg) were used to evaluate the antioxidant capacity and showed the best performance at the early fruiting period. The current study evidenced the significant effect of phenophase on antioxidants and contributed to valorize S. verbenaca extracts as a source of functional phenolic compounds.

The antioxidant activity in 70% aqueous acetone extracts of the herb of Ambrosia artemisiifolia L., Asteraceae was evaluated with regards to total polyphenol and flavonoid contents. Antioxidant activity has been assessed by two commonly used in vitro tests, based on determination of ferric-reducing antioxidant power (FRAP assay) and DPPH free radical scavenging ability (DPPH assay), against butylated hydroxytoluene (BHT), ascorbic acid and quercetin, which were used as positive control substances. The results of both antioxidant tests showed that the plant material expressed a considerable activity (DPPH IC50 = 27.6 μg/ml; FRAP value = 2.37 mmol Fe2+/g), attributed to both flavonoid aglyca and resembling glycosides, as verified by dot-blot TLC analysis.  相似文献

The objectives of this study were to examine the free radical scavenging activity and the protective effects against macromolecular oxidation as well as the cytotoxic activity of Aphanes arvensis aqueous and methanolic extracts. Free radical scavenging activity was determined by DPPH method. The methanolic extract showed a scavenging activity nearly equivalent to Trolox and Vitamin C and has an IC50 value of 4.54 μg/mL. Total antioxidant capacity was determined by CUPRAC method. The antioxidant capacity of aqueous and methanolic extract was 0.792 and 1.550 mmol TE/g DWE, respectively. The protective effect of A. arvensis extracts against lipid peroxidation was evaluated using a liposome oxidation system. The methanolic extract was more active than the aqueous extract. The aqueous extract possessed protective effect against protein oxidation in a dose dependent manner. Both extracts showed inhibitory effect on DNA oxidation as measured by plasmid relaxation assay. Results presented here indicate that A. arvensis possess strong antioxidant activity and protective effects with very little cytotoxic effect, and they can therefore be used as a natural additive in food, cosmetic and pharmaceutical industries.  相似文献

To search for dried plant seeds with potent anti-diabetes activity, we conducted a large scale screening for inhibitory activity on tumor necrosis factor-alpha and facilitating activity on adiponectin production in vitro. These activities in 3T3-L1 adipocytes were screened from ethanol extracts of 20 kinds of dried plant seed marketed in Japan. komatsuna (Brassica rapa var. perviridis), common bean (Phaseolus vulgaris L.), qing geng cai (Brassica rapa var. chinensis), green soybean (Glycine max), spinach (Spinacia oleracea L.) and sugar snap pea (Pisum sativum L.) markedly enhanced adiponectin production (11.3 ~ 12.7 ng/ml) but Japanese radish (Raphanus sativus), edible burdock (Arctium lappa L.), bitter melon (Momordica charantia) and broccoli (Brassica oleracea var. italica) did not (0.9 ~ 2.7 ng/ml). All adiponectin-production-enhancing seeds except spinach (2.7 pg/ml) and okra (Abelmoschus esculentus) (6.6 pg/ml) effectively decreased tumor necrosis factor-alpha levels (0.0 pg/ml). We further examined the effects on free radical scavenging activities in the dried seed extracts. Although scavenging activity correlated well with total phenolic content of samples, no correlation was observed with adiponectin production. These results point to the potential of dried seed extracts as a means to modify the activity of tumor necrosis factor-alpha for the adiponectin production.  相似文献

Rice husks of the RD 6 variety were refluxed with water for 30, 60 and 120 min. The longest extraction time resulted in the greatest yield (0.089 ± 0.008%), radical scavenging activity (IC50 = 37.203 ± 0.421 μg/ml), and total phenolic and flavonoid contents (7.512 ± 0.008 g GAE/100 g extract and 36.886 ± 0.004 g QE/100 g extract). Fatty acids of cosmetic importance, such as hexanoic, heptatonic, octanoic, nonanoic, stearic, palmitic, oleic and linoleic acids, were quantified using the best antioxidant extract (166.004 mg/kg). The total phenolic content was related to the antioxidant activity and total fatty acid content (r = 0.997 and 0.864) in linoleic acid (r = 0.989), which displayed antioxidant properties (r = 0.996). The extract was non-cytotoxic in vero cells. Furthermore, it was compatible and stable in cosmetic formulations. Therefore, rice husks should be further studied for use in health promotion products.  相似文献

Betalains are water-soluble plant pigments that are widely used as food colorants, and have a wide range of desirable biological activities, including antioxidant, anti-inflammatory, hepatoprotective, anti-cancer properties. They can be produced from various plants, notably beetroot, but betalain products obtained in this way also have some undesirable properties and are difficult to standardize. A potentially attractive alternative is to use hairy root cultures. In the study reported here, we found that betalain extracts obtained from hairy root cultures of the red beetroot B. vulgaris cv. Detroit Dark Red also had higher antioxidant activity than extracts obtained from mature beetroots: six-fold higher 2,2-dyphenyl-1-picrylhydrazyl radical scavenging ability (90.7% inhibition, EC50 = 0.11 mg, vs 14.2% inhibition, EC50 = 0.70 mg) and 3.28-fold higher oxygen radical absorbance capacity (4,100 μM TE/g dry extract, vs 1,250 μM TE/g dry extract). The high antioxidant activity of the hairy root extracts was associated with increased concentrations (more than 20-fold) of total phenolic concomitant compounds, which may have synergistic effects with betalains. The presence of 4-hydroxybenzoic acid, caffeic acid, catechin hydrate, and epicatechin were detected in both types of extract, but at different concentrations. Rutin was only present at high concentration (1.096 mg.g−1 dry extract) in betalain extracts from the hairy root cultures, whereas chlorogenic acid was only detected at measurable concentrations in extracts from intact plants.  相似文献

Studies on Indian Green Leafy Vegetables for Their Antioxidant Activity   总被引:1,自引:0,他引:1  
To identify the potential of green leafy vegetables (GLV) as antioxidants, methanolic extracts of Amaranthus sp., Centella asiatica, Murraya koenigii and Trigonella foenum graecum were studied for their antioxidant activity in different systems at multiple concentrations. Total antioxidant activity assessed by phosphomolybdenum method, free radical scavenging activity by 1,1-diphenly-2-picryl hydrazyl (DPPH), reducing power and ferrous ion chelating activity were determined. The GLV were analyzed for ascorbic acid, total and β-carotene and total polyphenol contents. The ascorbic acid, total carotene, β-carotene and total phenolic content (tannic acid equivalents) of the GLV ranged between 15.18–101.36, 34.78–64.51, 4.23–8.84 and 150.0–387.50 mg/100 g GLV, respectively. The extracts were found to have significantly different levels of antioxidant activities in the systems tested. The total antioxidant activity was highest in Murraya koenigii (2,691.78 μmol of ascorbic acid/g sample) and least in Centella asiatica (623.78 μmol of ascorbic acid/g sample). The extract concentration causing 50% inhibition of DPPH (IC50) was determined (M. koenigii < C.asiatica < Amaranthus sp. < T. graecum). The maximum DPPH scavenging activity and reducing power was exhibited by Murraya koenigii. Multiple regression analysis showed that the relationship of total antioxidant activity, free radical scavenging activity, and reducing power with polyphenol and total and β-carotene was highly significant. The present study aimed to evaluate for the first time the phenols, flavonoids, carotenoids, capsaicin and dihydrocapsaicin content and the antioxidant and hypoglycemic properties of Capsicum annuum var. acuminatum small and C. annuum var. cerasiferum air-dried fruits. The ethanol extract of C. annuum var. acuminatum small, characterized by the major content of total poliphenols, flavonoids, carotenoids and capsaicinoids, showed the highest radical scavenging activity (IC50 of 152.9 μg/ml). On the contrary, C. annuum var. cerasiferum showed a significant antioxidant activity evaluated by the β-carotene bleaching test (IC50 of 3.1 μg/ml). The lipophilic fraction of both C. annuum var. acuminatum and C. annuum var. cerasiferum exhibited an interesting and selective inhibitory activity against α-amylase (IC50 of 6.9 and 20.1 μg/ml, respectively).  相似文献

The antioxidant capacity as oxygen radical absorbance capacity (ORAC) and in vitro chemopreventive effect of phytochemicals extracted, partitioned and isolated from the decorticated bran of black Shawaya sorghum (Sorghum bicolor L. Moench) was studied. The chemopreventive effect was evaluated as the induction effect of quinone reductase (QR) using murine hepatoma cells Hepa 1c1c7. The crude methanol extract with an antioxidant capacity of 3.7 μmol Trolox equivalent (TE)/mg was partitioned into 120 fractions with fast centrifugal partition chromatography (FCPC). Fractions with partition coefficients between 0.1 and 0.26 exerted more than 50% of the antioxidant activity of the original crude extract and contained the highest concentration of anthocyanins. No correlation was found for ferulic acid content and ORAC or chemoprevention. A derivative of 7-methoxyapigeninidin at a concentration of 1 μg/ml increased by 31% the QR activity while the original extract reached a maximum of 27% at 40 μg/ml.  相似文献

Corn bran was microfluidized through a 200-μm channel in the pressure range of 124.1–158.7 MPa for 1–5 passes following the central composite experimental design. Physicochemical properties and antioxidant properties of microfluidized bran samples were measured and fitted to the second order polynomial model. The response surface equations obtained showed that all the properties examined had a positive linear relationship with pressure and a negative quadratic relationship with number of passes except for ABTS radical scavenging activity which was quadratically related to both processing parameters. The number of passes generally had a more pronounced effect on the examined properties compared with pressure. Within the experimental range, the maximum values of swelling capacity, water-holding capacity, and oil-holding capacity were respectively 10.62 ml/g d.w. (at 158.7 MPa), 5.49 g water/g d.w. (at 158.7 MPa), and 4.61 g oil/g d.w. (at 124.1 MPa); the maximum values of surface reactive phenolic content, DPPH and ABTS radical scavenging activities were 148.80 mg/FAE g d.w. (at 158.7 MPa), 50.02 μmol TE/g d.w. (at 158.7 MPa), and 47.90 μmol TE/g d.w. (at 145.9 MPa), respectively. All maximum values of the properties occurred at 5 passes.  相似文献

Leaves of Lycium barbarum are widely used as medicine vegetables and functional tea in China. The main flavonoids present in the leaves were separated and identified by high performance liquid chromatography (HPLC), liquid chromatography-atmospheric pressure chemical ionization mass spectrometry (LC-(APCI) MS) and ultraviolet-visible spectra with shift additives. The predominant flavonoid was identified as rutin. Leaves are the rutin-rich parts (16.03–16.33 mg/g). In the wild and cultivated L. barbarum fruits, contents of rutin were determined very low (0.09–1.38 mg/g). The contents of total flavonoids (21.25 mg/g) of cultivated L. barbarum leaves were much higher than those in the wild L. barbarum leaves (17.86 mg/g), so cultivated barbarum leaves are a suitable source for medicine vegetables and functional tea.  相似文献

Porophyllum tagetoides is an annual warm-weather herb that has an intense typical smell. Its leaves are commonly used in soup preparation and traditional medicine for treatment of inflammatory diseases. Its volatile compounds and antioxidant properties were evaluated in crude, aqueous and ethanol leaf extract and an oil emulsion using different antioxidant assays in vitro, such as: DPPH radical scavenging activity, redox potential, polyphenol content, reducing power and optical density. A high antioxidative activity was found when comparing leaves with stems. The crude extract from leaves showed a very high reducing power (2.88 ± 0.20 O.D.) and DPPH radical-scavenging activity (54.63 ± 4.80%), in concordance with a major concentration of vitamin C (23.97 ± 0.36 mg/100 g). Instead, the highest polyphenol content (264.54 ± 2.17 mg GAE/g of sample) and redox potential (561.23 ± 0.15 mV) were found by the ethanol and aqueous extract, respectively. Aldehydes and terpenes such as nonanal, decanal, trans-pineno, β-myrcene and D-limonene were the major volatiles found. This study suggests that Porophyllum tagetoides extracts could be used as antioxidants.  相似文献

Antioxidant properties of amaranth extracts isolated sequentially by acetone and methanol/water from defatted plant leaves, flowers, stems and seeds were assessed by ABTS+?, DPPH?, ORAC and total phenols content (TPC) assays. In addition, antioxidant properties of solid plant material were evaluated by the direct QUENCHER method using the same assays. Leaves and flowers of amaranth as well as their extracts possessed the highest antioxidant activities. Radical scavenging capacity in ABTS+? assay for leaves, flowers, stems and seeds evaluated by QUENCHER method were 144.24?±?2.41, 112.33?±?7.45, 19.05?±?1.13 and 21.82?±?1.06 μmol trolox equivalents in 1 g of dry weight, respectively. On-line HPLC-DPPH? assay was used to determine the activity of separated compounds and it was observed that rutin was the main radical scavenger in amaranth extracts. Preliminary screening of extract composition was performed by UPLC/ESI-QTOF-MS and rutin, nicotiflorin, isoquercitrin, 4-hydroxybenzoic and p-coumaric acids were identified by measuring their accurate mass and retention time.  相似文献

Organic solvent (methanol, ethanol, and acetone) extracts and water extracts of cherry (Prunus serrulata var. spontanea) blossoms were prepared, and antioxidant activities of the extracts were evaluated. Methanolic CBE (100 μg/ml) showed the highest total phenol content (104.30 μM), radical scavenging activity (34.2%), and reducing power (0.391). The effect of CBE on DNA damage induced by H2O2 in human leukocytes was evaluated by Comet assay. All CBE was a potent dose dependent inhibitor of DNA damage induced by 200 μM of H2O2, methanolic CBE showed the most strong inhibition activity. The methanolic CBE of 500 μg/ml showed 38.8% inhibition against growth of human colon cancer cell line HT-29. These results indicated that cherry blossoms could provide valuable bioactive materials.  相似文献

Chemical composition and the effects of domestic processing and storage were studied in organically cultivated or wild growing berries, harvested in certificated area in Serbia. The total phenolics content in fresh berries ranged from 380 to 1660 mg GAE/100 g. The phenolic content decreased during the processing to jams, and storage in almost all samples, except in black currant. Among fresh berries, the highest amount of total anthocyanins was found in bilberry (0.61 ± 0.03 %). Processing and storage caused decrease in anthocyanin content. The most ubiquitous anthocyanin aglycone was found to be cyanidin. Ellagic acid was detected in fresh and frozen black raspberries and related jams, and its content increases during thermal processing to jam. Among fresh berries, bilberry showed the strongest DPPH radical scavenging activity with an IC50 value of 0.82 mg/ml. However, frozen as well as processed berries in our study still showed comparable high values of radical scavenging activity. After 9 months of storage all investigated jams were microbiologicaly correct. Regardless of the losses in the amount of total phenolics, total anthocyanins and radical scavenging activity, frozen berries and related jams are still excellent sources of nutritional substances with antioxidant potential.  相似文献

The antioxidative activities of water extract (WE) and crude hot-water soluble polysaccharide (PS) from Ficus carica L. fruit were investigated using various assays in vitro, including scavenging abilities on DPPH, superoxide and hydroxyl radicals and reducing power. The immunity activities of PS were evaluated using the carbon clearance test and serum hemolysin analysis in mice. In addition, total phenolics and flavonoids contents were also determined. Both WE and PS have notable scavenging activities on DPPH with the EC50 values of 0.72 and 0.61 mg/ml, respectively. The PS showed higher scavenging activity than WE on superoxide radical (EC50, 0.95 mg/ml) and hydroxyl anion radical (scavenging rate 43.4% at concentration of 4 mg/ml). The PS (500 mg/kg) also has a significant increase in the clearance rate of carbon particles and serum hemolysin level of normal mice. The results indicate that both WE and PS might be applicable in healthy medicine and food industry.  相似文献

This study was designed to examine the in vitro antimicrobial and antioxidant activities and the amount of total phenolics of the methanol extracts of Ballota rotundifolia L. and Teucrium chamaedrys C. Koch. In the case of antimicrobial activity tests, polar sub-fractions of the methanol extracts of plant species exhibited weakest antimicrobial activity when compared with the non-polar ones. While, non-polar sub-fraction of B. rotundifolia showed moderate activity against A. lwoffii, C. perfringens and the yeasts, T. chamaedrys performed excellent activity pattern against all of the tested microorganisms. The sub-fractions were also screened for their possible antioxidant activities by two complementary tests, namely DPPH free radical-scavenging and β-carotene/linoleic acid assays. Non-polar extracts of the plant species remained inactive in both test systems. On the other hand polar extracts showed remarkable antioxidant activities. In DPPH system, free radical scavenging effect of T. chamaedrys was measured as 18.00 ± 1.42 μg.mg−1. It is extremely important to point out that, polar sub-fraction of T. chamaedrys is found as effective as the positive control BHT. Non-polar sub-fraction of T. chamaedrys found to have the highest total phenolic amount (97.12 ± 1.28 μg/mg). Results obtained from this experiment confirm the relationship between the amount of phenolics and biological activities.  相似文献

The aim of this investigation was to study the antioxidant and anticancer activities of young sprouts of some traditional Korean salad plants. Total phenolics, antioxidant and anticancer activities of the methanol extracts from young sprouts of 11 salad plants were determined. The highest amount of phenolics was found in methanol extracts of Euonymus alatus (235.7 mg kg−1), followed by Hypericum ascyron (197.1 mg kg−1), Zanthoxylum piperitum (194.1 mg kg−1) and Zanthoxylum schinifolium (142.5 mg kg−1). Methanol extracts of E. alatus, H. ascyron, and Z. piperitum at 63 mg kg−1 exhibited the highest dose-depend DPPH radical scavenging activity by 91.2, 91.2 and 83.9%, respectively. According to the MTT results, the methanol extracts from Stellaria aquatica, Eleutherococcus sessilifolrus and Z. schinifolium showed the highest anticancer activities against Calu-6 (IC50 < 25.0 μgml−1) and from S. aquatica—the highest anticancer activities against SNU-601 (153.3 μgml−1), following by E. sessilifolrus (196.7 μg ml−1) and Amaranthus mangostanus (303.1 μgml−1). Total phenolics were highly correlated with the DPPH, suggesting that they contribute to the antioxidant properties of the studied plants. In conclusion: young sprouts of Korean salad possess antioxidant and anticancer properties and could be used as a supplement to proper drugs.  相似文献

Spices are extensively used to enhance the taste and flavor of foods and are known to possess several medicinal properties. Myristica fragrans, Parmelia perlata, Illicium verum, Trachyspermum copticum and Myristica malabarica, the commonly used spices in India were assessed for antidiabetic activity in streptozotocin induced diabetic rats. In the in vitro insulin secretion studies on isolated islets of Langerhans, M. fragrans, T. copticum and M. malabarica showed dose dependent insulin secretion. At 1 mg/ml, P. perlata showed significant in vitro alpha-glucosidase inhibitory activity with IC50 value of 0.14 mg/ml followed by M. malabarica (0.64 mg/ml), I. verum (0.67 mg/ml), M. fragrans (0.85 mg/ml) and T. copticum (0.92 mg/ml). The DPPH free radical scavenging activity of the extracts at a concentration of 1 mg/ml was as M. malabarica (90.45%), M. fragrans (89.89%), I. verum (87.22%), P. perlata (76.70%) and T. copticum (38.14%). P. perlata showed the highest phenolic content (i.e., 118.5 mg gallic acid equivalents/g) followed by M. malabarica (84.13 mg gallic acid equivalents/g). M. malabarica showed the highest flavonoid content (i.e., 38.35 mg quercetin equivalents/g). Regular use of these spices may prevent postprandial rise in glucose levels through inhibition of intestinal alpha-glucosidase and may maintain blood glucose level through insulin secretagogue action.  相似文献
