A common female founder individual of the Portuguese horse breeds Sorraia and Lusitano was found while conducting research on the variation of the Lusitano mitochondrial DNA lineages in relation to studbook information. We obtained 416‐bp control region sequences from 16 descendents of a female Sorraia founder (Pomba) still represented in the living population of the Lusitano, according to the most recent edition of this breed's studbook. The same haplotype was found for all analysed samples and belongs to the haplogroup described by several authors as having predominantly Iberian, South American and North African haplotypes bringing new insights on the relationship between the Sorraia and the other Iberian breeds. This work illustrates how weak the boundary of breed establishment can be, especially at the same geographical region. Using the same founders in different breeds is surely one of the explanations to frequently shared haplotypes among recent breeds, resulting in a lack of consistency between mtDNA sequences and breeds and/or geographical regions.  相似文献

为研究广西德保矮马、百色马和新疆伊犁马的生长激素(GH)基因多态性与体尺指标之间的相关性,本实验利用PCR-SSCP技术分析3个品种277匹马GH基因5′侧翼区、第3外显子、第4外显子、第5外显子的遗传多态性。结果表明:只有GH基因第5外显子出现多态性,表现为AA、BB和AB 3种基因型,其他片段未发现多态性。群体遗传学分析表明,等位基因A和B的基因频率相等;Hardy-Weinberg平衡检验显示,3个品种马均处于非平衡状态;它们的多态信息含量均为中度多态。统计分析表明,GH基因第5外显子的基因型与3个品种马体尺指标之间无显著相关性。  相似文献

Genetic variation at 7 blood-group and 10 biochemical genetic loci was examined in 30 horses from a feral herd from the Namib Desert of Namibia, Africa. The observed genetic variability was extremely low compared with that found in domestic horse breeds. The low variation was most probably a result of recent small population size and a small founding population size. Genetic comparison of the Namib horses, which were of unknown origins, to domestic horse breeds, showed that the Namib horses had the highest genetic similarity to Arabian type horses, although they did not closely resemble this type of horse in conformation.  相似文献

Summary We analysed a 610-bp mitochondrial (mt)DNA D-loop fragment in a sample of German draught horse breeds and compared the polymorphic sites with sequences from Arabian, Hanoverian, Exmoor, Icelandic, Sorraia and Przewalski's Horses as well as with Suffolk, Shire and Belgian horses. In a total of 65 horses, 70 polymorphic sites representing 47 haplotypes were observed. The average percentage of polymorphic sites was 11.5% for the mtDNA fragment analysed. In the nine different draught horse breeds including South German, Mecklenburg, Saxon Thuringa coldblood, Rhenisch German, Schleswig Draught Horse, Black Forest Horse, Shire, Suffolk and Belgian, 61 polymorphic sites and 24 haplotypes were found. The phylogenetic analysis failed to show monophyletic groups for the draught horses. The analysis indicated that the draught horse populations investigated consist of diverse genetic groups with respect to their maternal lineage.  相似文献

This study was the first to analyze the polymorphic characteristics of a wide range of genetic markers in three Lithuanian horse breeds. Genetic variation at seven red blood cell alloantigen (blood group) loci, 10 biochemical genetic loci and 16 microsatellites was examinated in 31 Zemaitukai, 30 large-type Zemaitukai and 24 Lithuanian Heavy Draught horse and compared to that of other domestic horse breeds. Genetic variability of the Zemaitukai breeds based upon blood groups and biochemical loci was higher than average for domestic horse breeds. For microsatellite loci, the variability of the large-type Zemaitukai and the Heavy Draught breeds also was high compared to the mean variation for domestic breeds; however, for the Zemaitukai breed, variation levels were very near the average for horses.  相似文献

The genetic variability within the Uruguayan Creole horse and its relationship to a group of geographically or historically related breeds (Spanish Pure-bred, Barb, Quarter horse, Paso Fino, Peruvian Paso, Arabian and Thoroughbred horse), was evaluated using 25 loci (seven of blood groups, nine of protein polymorphisms and nine microsatellites) analyzed on a total of 145 Uruguayan Creole horses. In this study, blood group and protein polymorphism variants that are considered to be breed markers of Spanish Pure-bred and Barb horses were detected in the Creole breed. Conversely, some microsatellites and protein polymorphisms alleles were found uniquely in the Creole horse. American horse breeds together with Barb and Arabian horses clearly formed a separate cluster from the Spanish pure-bred and Thoroughbred breeds, as shown by an UPGMA dendrogram based on Nei's standard genetic distance. Data in this study provided evidence for considerable genetic variation within Uruguayan Creole horses and of a distinctive breed profile. Both traits were most likely inherited from the XVIth century Spanish horses, more closely related to Barb than to Spanish Pure-bred.  相似文献

The distribution of genetic diversity and the genetic relationships among western Mediterranean horse breeds were investigated using microsatellite markers. The examined sample included seven Spanish and three Italian local horse breeds and populations, plus a Spanish Thoroughbred outgroup. The total number of animals examined was 682 (on average 62 animals per breed; range 20–122). The microsatellite marker set analysed provided 128 alleles (10.7 alleles per locus). Within‐breed genetic diversity was always high (>0.70), with breeds contributing about 8% of the total genetic variability. The mean molecular coancestry of the entire population examined was 0.205, Losino being the breed that contributed most. In addition to Nei's standard and Reynolds’ genetic distances, pair‐wise kinship distance and molecular coancestry were estimated. Remarkably similar breed rankings were obtained with all methods. Clustering analysis provided an accurate representation of the current genetic relationships among the breeds. Determining coancestry is useful for analysing genetic diversity distribution between and within breeds and provides a good framework for jointly analysing molecular markers and pedigree information. An integrated analysis was undertaken to obtain information on the population dynamics in western Mediterranean native horse populations, and to better determine conservation priorities.  相似文献

The evaluation of the genetic structure of four native Norwegian horse breeds was investigated using 35 genetic markers including nine biochemical loci and 26 microsatellites. The proportion of genetic variation measured as heterozygosity and the number of alleles indicated high variation in Fjord Horse and Coldblooded Trotter, but somewhat reduced variation in Døle Horse. No clear signs of bottleneck effects were found in either of the breeds, but significant inbreeding estimates in Nordland/Lyngen Horse may indicate subdivision of this breed. Significant population differentiation was detected between all breeds, also between the recently diverged Døle Horse and Coldblooded Trotter. The principal component analysis supports the close genetic relationship between these two breeds.  相似文献

To assess genetic markers able to distinguish between five Greek breeds of horses, the random amplified polymorphic DNA (RAPD) technique was used. A total of 40 primers were initially screened in all breeds to identify primers that consistently revealed well-amplified, polymorphic bands. Despite the variability that was found within breeds, no specific RAPD marker for the discrimination of the breeds was detected. This is probably due to the extensive cross-breeding that has taken place over the years, resulting in a mixing of breeds. The high phenotypic divergence of the pony of Skyros from all the other Greek breeds of horses could be explained in terms of micro-evolutionary processes, such as natural selection owing to differential fitness. In addition, to determine the genetic affinities between breeds, all samples were screened with five primers. These primers produced a total of 93 bands, 51 of which were polymorphic. Data analysis for genetic polymorphism revealed a degree of divergence between breeds. According to the discriminant analysis (DA) classification, 100% of the individuals examined in this study could be classified correctly into their breeds. The results of this study show that RAPDs could be a powerful tool for the classification testing of individual horses, as well as for the determination of the genetic variation found among breeds, which is important for a proper management policy designed to protect this species.  相似文献

选用来源于牛和绵羊的27个微卫星DNA标记,对山东省4个地方山羊品种进行遗传多样性分析,通过计算等位基因频率、多态信息含量、不同标记的平均杂和度、总群体杂合度、亚群体杂合度、群体杂合度、基因分化系数、不同群体的F-统计量、基因流动数、不同群体间的基因流动个数和遗传距离并进行聚类分析,评估其种内变异和种间变异的关系,以群体平均杂合度、F-统计量和遗传分化系数为基础,结合四个山羊种群的实际生存状况,提出避免近交和种群间杂交符合山东山羊种群实际状况的保种模式。研究结果可为山东地方种质特性研究提供基础数据,为山东地方山羊种群的合理保护利用提供科学依据。  相似文献

利用随机扩增多态DNA技术研究了我国主要地方山呼吸主部分国外品种共计23个山羊品种251个山羊个体的随机扩增多态DNA。结果表明:(1)总群体平均遗传多样性指数(Hsp)为0.7084,群体遗传分化指数为0.8583,山羊品种间平均遗传距离指数(AIGD)(0.0810-0.2044)明显大于品种内的AIGD(0.0444-0.1112),不同山羊品种的遗传多样性指数(Ho)除安哥拉山羊和承德无角山羊大于50%外,其它各品种均低于50%,上述结果说明所研究山羊群体不仅具有较为丰富的遗传多样性,而且山羊核基因组遗传变异主要存在于品种间,我国地方山羊品种间遗传分化比较明显。(2)中卫山羊(0.3410)和辽宁绒山羊(0.2589)的Ho值明显低于各品种的平均值(0.3881);雷州山羊(0.0566)、中卫山羊(0.0564)和辽宁绒山羊(0.0444)的AIGD值也明显低于各群体的平均值(0.0749),说明辽宁绒山羊、中卫山羊和雷州山羊核基因组的遗传多样性相对其它品种来说较为贫乏,应作为重点保护对象。  相似文献

Criollo horse populations descend from horses brought from the Iberian Peninsula over the period of colonization (15th to 17th century). They are spread throughout the Americas and have potentially undergone genetic hybridization with other breeds in the recent past. In this study, 25 autosomal microsatellites were genotyped in 50 horse breeds representing Criollo populations from 12 American countries (27 breeds), breeds from the Iberian Peninsula (19), one breed each from France and Morocco and two cosmopolitan horse breeds (Thoroughbred and Arabian). The genetic relationships among breeds identified five clusters: Celtic; Iberian; North American with Thoroughbred influence; most Colombian breeds; and nearly all other Criollo breeds. The group of “all other Criollo breeds” had the closest genetic relationship with breeds originating from the Iberian Peninsula, specifically with the Celtic group. For the whole set of Criollo breeds analysed, the estimated genetic contribution from other breeds was approximately 50%, 30% and 20% for the Celtic, Iberian and Arab-Thoroughbred groups, respectively. The spatial distribution of genetic diversity indicates that hotspots of genetic diversity are observed in populations from Colombia, Ecuador, Brazil, Paraguay and western United States, possibly indicating points of arrival and dispersion of Criollo horses in the American continent. These results indicate that Criollo breeds share a common ancestry, but that each breed has its own identity.  相似文献

In this study, genetic analyses of diversity and differentiation were performed on five horse breeds raised in Algeria (Barb, Arab‐Barb, Arabian, Thoroughbred and French Trotter). All microsatellite markers were highly polymorphic in all the breeds. A total of 123 alleles from 14 microsatellite loci were detected in 201 horses. The average number of alleles per locus was the highest in the Arab‐Barb horses (7.86) and lowest in the thoroughbred breed (5.71), whereas the observed and expected heterozygosities per breed ranged from 0.71 (Thoroughbred) to 0.752 (Barb) and 0.71 (Thoroughbred) to 0.77 (Arab‐Barb), respectively. The genetic differentiation between the breeds was significant (p < 0.01) based on the infinitesimal model (FST). Three different approaches for evaluating the genetic relationships were applied. Genetic distances, the factorial correspondence analysis and structure analysis showed that a significant amount of genetic variation is maintained in the native horse populations and the other breeds. The Barb and Arab‐Barb breeds seem to be the most genetically related and support the decision to consider the breeds as same population.  相似文献

This article described the relationship among six Chinese indigenous sheep breeds.There were six microsatellite loci which used to analyze the genetic diversity.Based on the allele frequency of six sheep variations, we calculated the mean heterozygosity (Ho), which were from 0.5446 to 0.7140;The polymorphism information content (PIC) in six sheep breeds were from 0.5211 to 0.7732.Genetic variations within breed were analyzed in the study, and we clustered the UPGMA tree according to the DA genetic distance.The results showed that six breeds had relatively abundant genetic diversity, and MXB had the highest genetic variation, while SW had the lowest genetic variation.The inbreeding coefficient of SW was higher than other breeds, which revealed a higher inbreeding degree in this breed.Phylogenetic tree was constructed using UPGMA, which showed that LZD and MXB were classified as a class, HZK and GF were classified as a class, SIH and SW were classified as a class.The results revealed the genetic structure of breeds correlated with geographical distribution and history of domestication.  相似文献

试验通过9个微卫星座位对贵州矮马和伊犁马2个品种共84匹马进行了遗传多样性检测,以8%非变性聚丙烯酰胺凝胶电泳分离PCR扩增产物,并用银染法显色,计算各微卫星位点的等位基因频率、有效等位基因数(Ne)、杂合度(H)和多态信息含量(PIC)。结果表明,9个微卫星座位中COR059检出的变异最大,AHT040的变异最小;伊犁马的Ne、H和PIC的平均值高于贵州矮马;2个群体的平均遗传分化系数为0.041。说明2个品种都有丰富的遗传多样性,贵州矮马的基因较伊犁马更为纯合。  相似文献

为研究中国地方绵羊品种之间的遗传多样性,利用6个微卫星位点,对中国6个绵羊品种群体进行遗传检测,计算了各品种群体的平均杂合度(He)和平均多态信息含量(PIC),同时分析了各品种群体内的遗传变异,进行了UPGMA聚类。结果表明,6个绵羊品种Ho为0.5446~0.7140,PIC为0.5211~0.7732;6个绵羊品种都具有较丰富的遗传多样性,其中岷县黑裘皮羊的遗传变异最高,小尾寒羊的遗传变异最低;小尾寒羊的近交系数最高,揭示该群体内近交程度可能较高。利用UPGMA方法构建了系统发生树,兰州大尾寒羊与岷县黑裘皮羊为一类,哈萨克羊与贵德黑裘皮羊为一类,泗水绵羊与小尾寒羊为一类,表明群体遗传结构与地理分布及其驯化历史相关。  相似文献

The Lanyu pig is an indigenous miniature pig breed on Lanyu Islet near Taiwan, with a mitochondrial DNA genetic lineage remote from Asian and European pig breeds. The unknown population genetic structure and increased inbreeding among the small population of conserved Lanyu pigs is now of great conservation concern. Additionally, the presence for more than a century of exotic pig breeds in Taiwan has made gene introgression from exotic pig breeds into Lanyu pigs very possible. The present study thus aimed to investigate nuclear genetic variation within the conserved Lanyu pigs and the phylogenetic relationship and possible genetic introgression between Lanyu and exotic pig breeds by determining the polymorphism of 19 microsatellite loci. In the neighbor-joining tree constructed from 7 pig breeds based on Cavalli-Sforza and Edward chord genetic distances, 3 major clades were recognized, in which the Asian and European breeds were separately clustered into 2 clades with a 93.0 and 99.9% bootstrap confidence value, respectively. All individuals of the Lanyu breed formed a unique subclade within the Asian clade based on the distance of the proportion of shared alleles, -ln(ps), suggesting that the Lanyu breed possesses a unique nuclear genetic structure and that no nuclear gene introgression from exotic breeds into the conserved Lanyu pigs has occurred in recent history. Fifteen of 19 microsatellite loci deviated from Hardy-Weinberg equilibrium (by Wright's statistic), suggesting a significant loss of heterozygosity in the conserved population. The valuable nuclear genetic structure and phylogenetic information should assist future conservation and population management of Lanyu pigs.  相似文献

Genetic variability at 7 blood-group and 10 biochemical genetic loci was examined in 3 South African horse breeds, the Nooitgedacht, Boerperd and Basuto Pony. Observed heterozygosity for these breeds was intermediate for domestic horses, with the highest heterozygosity in the Boerperd and the lowest in the Basuto Pony. The 3 breeds show greater genetic similarity to each other than to other domestic horse breeds. Compared to other breeds, the South African breeds show greater genetic similarity to breeds such as the Thoroughbred, Holstein, Trakehner and Hanovarian and also to North American breeds such as the Saddlebred, Standardbred and Morgan Horse.  相似文献

旨在揭示康西草原马匹的品种来源以及群体遗传结构。本研究采集了当地60匹健康状况良好、体重适中、3~6岁之间不区分公母马的血液样品,试验分为6个种群并以纯血马等4个国外马品种、1个我国地方马品种作为对照, 通过设计引物并提取基因组DNA结合微卫星扩增法对染色体上12个微卫星位点的等位基因进行检测,并对各种群遗传多样性参数、群体分化指数、瓶颈效应以及群体结构进行分析。试验按照群体个数为依据分为6组,每组1个重复,每个重复以该组样本量为标准。分析6个种群马的遗传参数结果表明,其中共检测到221个等位基因,平均Shannon指数为1.691;有效等位基因数(Ne)为3.649~5.397;期望杂合度(He)为0.681~0.798;观测杂合度(Ho)为0.632~0.780;平均多态信息含量(PIC)为0.643~0.772。这些结果表明,6个群体都具有很高的遗传多样性。通过微卫星DNA位点的连锁不平衡及哈迪-温伯格平衡分析发现, 大多数位点的P值都大于校正值(P=0.006 5),表示较多位点都处于哈迪-温伯格及连锁平衡状态。各群体间群体分化系数(Fst)表明,群体间分化程度较低(Fst<0.15)。基于Nei’s遗传距离与地理距离之间Mantel相关性检验显示二者之间呈现正相关性(R=0.502),但未达到显著水平(P=0.310);各马匹种群瓶颈效应分析表明,P值极显著(P(IAM)<0.01)说明该地区的马群体受到过瓶颈效应的影响且该马群体历史上数量有大规模的减少。通过 FCA因子分析、基于Nei’s标准遗传距离UPGMA树状图以及Structure贝叶斯聚类分析发现,该地区马种群与纯血马和温血马聚为一类,说明该种群最有可能主要来源于温血马和纯血马。这些结果揭示,康西草原地区马匹具有较高的遗传多样性,该地马群体的血统与纯血马和温血马相近,这为当地马遗传资源评估以及开发利用打下了坚实的基础。  相似文献
