Twenty-eight horses with a residual burden of strongyle eggs in the faces after treatment with mebendazole (MBZ) paste were treated with a suspension of either MBZ or oxibendazole (OBZ). Fecal samples were collected before and 14 days after these treatments. The number of strongyle eggs/g (epg) of feces for each horse was estimated using the Cornell-McMaster dilution and the Cornell-Wisconsin double centrifugation procedures. The epg for each horse was transformed using log (× + 1) and in an analysis of variance of the reduction in egg count for each horse on the logarithmic scale, there was a highly significant difference between the treatments. The mean epg was increased in the MBZ-treated horses and reduced in the OBZ-treated horses, but the reduction was only by 82% with an upper confidence limit of 89%. Subsequently, the horses were retreated with MBZ and OBZ suspensions without significant reduction in the mean epg for OBZ-treated horses.  相似文献

A survey was conducted to estimate the prevalence of gastrointestinal nematodes in grazing dairy replacement heifers in western Canada. Fecal samples were collected from 566 animals on 34 farms distributed over the three prairie provinces. Fecal egg counts were performed using a modified Wisconsin centrifugation technique. The overall mean farm fecal egg count was 16.0 eggs per gram feces (range 0.7-78.1 epg). Gastrointestinal nematodes were demonstrated on every farm examined, and 93.3% of all individuals tested were positive.  相似文献

Several variables in a standard vial fecal gravitational flotation technique were investigated. These were the specific gravity of the sodium nitrate flotation solution, duration of flotation and mesh sizes of strainers. The number of eggs which floated and adhered to a coverslip were counted and estimates of the number of eggs remaining in the strained fecal suspension and in the feces trapped on the strainer were made. Eggs from hookworms, Trichuris vulpis and Toxocara canis in feces from dogs, Nematodirus spp. from sheep and Parascaris equorum from horses floated equally well in solutions with specific gravities (SpGr) ranging from 1.22-1.38. Taenia spp. from dogs had a slightly narrower range (SpGr 1.27-1.38) for best recovery. Eggs from Haemonchus contortus from sheep appeared to float best between SpGr 1.22- 1.32. Strongyles from one horse floated best with SpGr 1.27-1.32 and from another with SpGr 1.11-1.38. Coccidial oocysts from sheep floated best in a narrow range of SpGr from 1.22-1.27. However, as the SpGr of the solution was increased the recognition of eggs under the coverslip was increasingly difficult and especially so at SpGr 1.38 with sheep feces. This was due to the increasing amount of debris and the more rapid formation of crystals with evaporation with solutions of higher SpGr. It appeared, therefore, that solutions with SpGr of 1.22-1.35 would be best for routine laboratory use. At specific gravity 1.27, there appeared to be no difference in the number of eggs recovered for a four, eight and 12 min flotation period.

Only 3-7% of the eggs in 4 g of feces were counted under the coverslip. This poor efficacy resulted first because approximately 50% of the eggs were trapped in the feces and retained on the strainer. Secondly, only one half of the strained fecal suspension, containing approximately 25% of the eggs, was placed in the vial for examination. Thirdly, of those eggs in the vial only 16-29% were counted under the coverslip. When the second half of the strained fecal suspension was placed in another vial, the amount of debris and air bubbles adhering to the coverslip was much less than that for the first vial. Egg counts for both vials appeared similar and it may be that when debris is excessive the fecal examination should involve counts from a second vial. The use of strainers finer than the standard tea strainer and the addition of minimal amounts of detergent did not increase the egg count.

From 1973 to 1977, a survey of internal parasites in Wyoming cattle was conducted via fecal analyses of 1,490 beef cattle. Sugar flotation techniques were used, with a factor of 2 times the actual egg counts. The prevalence of internal parasites of beef cattle in this survey was compared with that in a previous survey conducted on Wyoming beef cattle from 1957 to 1961. Results of analyses indicated no true change in mean eggs per gram of feces (epg) as follows: calves, 14 epg in 1961 and 20 epg in 1977; yearlings, 29 epg in 1961 and 19 epg in 1977; and adults, 22 epg in 1961 and 21 epg in 1977.  相似文献

Treatment with 3 formulations of cambendazole (paste, pellets, and suspension) was compared with thiabendazole treatment in 181 Quarter Horses (females and intact and altered males) of different ages in 2 experiments. The mean output of strongyle eggs, as measured by eggs per gram of feces (epg), was reduced by at least 95% by the 3 formulations of cambendazole compared with pretreatment epg and epg in simultaneously nonmedicated horses. Eggs of Parascaris equorum were seen in 19 of the younger horses in experiment I. Parascaris eggs were not seen in postmedication fecal samples from 14 horses treated wtih cambendazole. Of the 5 horses treated with thiabendazole, 2 had postmedication Parascaris egg counts.  相似文献

Haemonchus contortus eggs were extracted from sheep feces and known numbers were added to helminthologically sterile bovine feces to provide samples with seven, 30 and 60 eggs per gram (epg). At 60 epg, dilution techniques (modified Cornell-McMaster and modified McMaster) tended to overestimate the number of eggs and more eggs were recovered (mean of 121 and 88% respectively) with these techniques than with centrifugal concentration procedures (modified Cornell—63% and Wisconsin— 69%). At 30 epg, all techniques were comparable (modified Cornell-McMaster 67%, modified McMaster 63%, modified Cornell and Wisconsin 64%). At 7 epg, the Wisconsin (61%), modified Cornell (60%) and Cornell-McMaster (94%) techniques were comparable and better than the modified McMaster technique (16%). At all levels of epg, the modified Cornell and Wisconsin techniques recovered eggs from 100% of the samples. The Cornell-McMaster and modified McMaster techniques recovered eggs from 90 and 100% of samples at 60 epg; 40 and 100% at 30 epg; and 21 and 11% at 7 epg. With a gravitational concentration procedure, the Standard Vial, no more than 16% of the eggs at any level of epg were recovered and at 7 epg eggs were recovered from only one-half of the samples. Five gravitational concentration techniques were assessed over 66 to 490 epg. The Ovassay, Fecalyzer and modified Standard Vial techniques were comparable in efficiency (28%, 25% and 24% respectively), but the Standard Vial technique was less efficient (11%).

Introduced into diagnostic parasitology was the concept of predictive values which is the proportion of samples that a technique correctly identifies as being negative for parasite eggs. At 7 epg this was calculated to be zero for the modified Cornell-McMaster, modified McMaster and Standard Vial techniques and 100 for the Wisconsin and modified Cornell techniques.

Fresh fecal specimens from deer were examined for nematode eggs (primarily Haemonchus and Ostertagia), using a flotation technique (sugar, sp gr = 1.27), and then were reexamined for up to 200 days after storage in 2.5, 5, or 10% formalin, absolute methyl alcohol, or 70% ethyl alcohol at room temperature (20 C) or after storage without preservative at 4, 0, or -20 C. For long-term storage, 10% formalin was the best fixative for recovery of eggs (compared with the rate of recovery of eggs from fresh feces). Approximately 50% of the strongyle eggs were detected in feces stored in formalin for 200 days. However, between days 3 and 10 of storage, the recovery rate was low (less than 50%), presumably due to ion binding. Alcohols were unsuitable for preservation, and storage at 0 or -20 C resulted in an egg recovery rate of less than 50%. Storage at 4 C for 50 days resulted in approximately 90% recovery of nematode eggs. Number of Parelaphostrongylus tenuis larvae recovered from fecal specimens stored in 10% formalin for 24 days was greater than that recovered from fresh fecal specimens.  相似文献

A variety of procedures are available to detect parasite eggs or oocysts in feces. This study compared the efficacy of simple flotation, a commercial assay, and various centrifugation techniques and three common flotation solutions. Results indicate that centrifugation consistently recovered more eggs than other methods. Proper technique is critical, including ensuring that the specific gravity of the flotation solution is correct and allowing the sample to stand for a sufficient amount of time before examining the coverslip. Because of the zoonotic health risks of many companion animal parasites, veterinarian and their staff should better utilize fecal examinations in their routine diagnostic plan.  相似文献

A series of investigations evaluated the ability of different testing methods - a swing-head centrifugation technique using two flotation solutions (1.18-specific gravity zinc sulfate and 1.27-specific gravity Sheather's sugar solution), a passive commercial flotation technique, and the SNAP Giardia Test Kit from IDEXX Laboratories - to identify Giardia-positive dogs and recover the eggs of other intestinal parasites. It was determined that the SNAP Giardia test can improve a practice's ability to identify Giardia-infected dogs. Because of its higher specific gravity, the sugar solution was better for recovering heavy parasite eggs, such as Taenia spp, and thus is the flotation solution of choice when conducting routine centrifugation fecal examinations.  相似文献

Clinical trials using fecal egg count reduction tests and coproculture were conducted with yearlings and mares on a farm in 1997. Fecal samples were taken from each horse to estimate the number of strongyle eggs/g feces with Cornell-Wisconsin centrifugal flotation and Cornell-McMaster dilution techniques. Eleven of 15 yearlings, which had been on a daily feeding of grain with pyrantel tartrate for 66 d were found with strongyle eggs in feces. This was the first time the in-feed medication had been used on the farm. Nine yearlings were randomised into three groups; continuation of daily pyrantel tartrate or one treatment with pyrantel pamoate or moxidectin. Two of three yearlings given pyrantel tartrate or pamoate had no reduction in the eggs/g feces. These six yearlings were then given moxidectin and in all yearlings the eggs/g feces was reduced to zero. The 66 d of pyrantel tartrate use was an inadequate time for development of resistant cyathostomes and a hypothesis was the resistance was due to extensive use on the farm over many years of pyrantel pamoate at twice the label dose for control of tapeworms. That hypothesis was tested with 12 mares with strongyle eggs in the feces randomised into two treatment groups: pyrantel pamoate at label dose or moxidectin. Five of six mares given pyrantel had <80% reduction in egg/g feces. These mares were then given moxidectin and in all mares the eggs/g feces was reduced to zero. Only cyathostomes were found on culture and apparently there was side resistance among the pyrantel salts.  相似文献

Several faecal examination techniques have shown variable sensitivity in demonstrating Spirocerca lupi (S. lupi) eggs. The objective of this study was to determine which faecal examination technique, including a novel modified centrifugal flotation technique, was most sensitive to diagnose spirocercosis. Ten coproscopic examinations were performed on faeces collected from 33 dogs confirmed endoscopically to have spirocercosis. The tests included a direct faecal examination, a faecal sedimentation/flotation test, 4 direct faecal flotations and 4 modified faecal centrifugal flotations. These latter 2 flotation tests utilised 4 different faecal flotation solutions: NaNO3 (SG 1.22), MgSO4 (SG 1.29), ZnSO4 (SG 1.30) and sugar (SG 1.27). The sensitivity of the tests ranged between 42% and 67%, with the NaNO3 solution showing the highest sensitivity in both the direct and modified-centrifugal flotations. The modified NaNO3 centrifugal method ranked 1st with the highest mean egg count (45.24 +/- 83), and was superior (i.e. higher egg count) and significantly different (P < 0.05) compared with the routine saturated sugar, ZnSO4 and MgSO4 flotation methods. The routine NaNO3 flotation method was also superior and significantly different (P < 0.05) compared with the routine ZnSO4 and MgSO4 flotation methods. Fifteen per cent (n = 5) of dogs had neoplastic oesophageal nodules and a further 18% (n = 6) had both neoplastic and non-neoplastic nodules. S. lupi eggs were demonstrated in 40% of dogs with neoplastic nodules only and 72.9% of the dogs with non-neoplastic nodules. The mean egg count in the non-neoplastic group (61) was statistically greater (P = 0.02) than that of the neoplastic group (1). The results show that faecal examination using a NaNO3 solution is the most sensitive in the diagnosis of spirocercosis. The modified centrifugal flotation faecal method using this solution has the highest egg count. The study also found that dogs with neoplastic nodules shed significantly fewer eggs than dogs with non-neoplastic nodules.  相似文献

Three clinical trials with fecal egg count reduction tests and coproculture were conducted on 2 standardbred farms in Ontario. On Farm A, the treatment groups were mebendazole and ivermectin in trial 1, and fenbendazole and moxidectin in another. On Farm B, treatment groups were mebendazole and ivermectin. All horses treated with mebendazole or fenbendazole were subsequently treated with ivermectin or moxidectin. Strongyle eggs/g feces were estimated pre- and post-treatment using the Cornell-McMaster dilution and Cornell-Wisconsin centrifugal flotation techniques. After treatment, there was no change in the arithmetic mean eggs/g feces for horses given mebendazole, and a reduction of only 49.1% for those given fenbendazole. All horses receiving ivermectin or moxidectin had their egg counts reduced to 0. Only cyathostomes were found on culture. On both farms the benzimidazole resistant strains appeared to have persisted for at least 10 years. Development of and monitoring for anthelmintic resistance are briefly discussed.  相似文献

为了解规模化舍饲湖羊消化道寄生虫感染情况,本研究应用饱和蔗糖溶液漂浮法、饱和盐水漂浮法、卢戈氏碘液染色法、离心沉淀法和麦克马斯特氏计数法等对采自河南部分地区规模化全舍饲湖羊养殖场共计553份粪便样品进行了调查。调查结果显示:寄生虫总感染率高达97.47%,75.23%的样品混合感染,样品混合感染的寄生虫种类最多为5种;共查到球虫、隐孢子虫、贾第虫、阿米巴、鞭虫、圆线虫和绦虫7种寄生虫,感染率分别为90.42%、0.90%、4.88%、65.64%、12.48%、42.13%和4.88%;感染强度最大的为球虫,每克粪便的卵囊数(OPG)最高达652 000,其次为圆线虫,每克粪便的虫卵数(EPG)最高为7 000;湖羊消化道寄生虫感染无明显的年龄、性别差异(P>0.05);季节流行动态显示,春、夏、秋三季的寄生虫感染率与冬季相比有较大差异。以上结果说明,湖羊消化道寄生虫感染较为普遍,应采取有效的综合防控措施,以保障羊群的健康发展。  相似文献

旨在探究不同粪菌液处理方法对其菌群活性及存活菌群物种组成的影响,为猪粪菌移植(FMT)的优化应用提供参考。本研究共设置7个试验组制备粪菌液:静置制备新鲜粪菌液组(FS)、静置制备粪菌液于-80℃冷冻1周组(FFS)、离心制备新鲜粪菌液组(FC)、离心制备粪菌液于-80℃冷冻1周组(FFC)、液氮冷冻粪便24 h静置制备粪菌液组(NS)、甘油+液氮冷冻粪便24 h静置制备粪菌液组(GNS)和液氮冷冻粪便24 h静置制备粪菌液于-80℃冷冻1周组(NFS),在37℃厌氧培养箱中平板培养48 h后,进行菌落计数。从FS、FFS、FC和FFC 4组中根据菌落形态随机挑取87个单菌落纯化后进行16S rRNA基因全长测序。结果表明,静置和离心两种方法制备新鲜粪菌液时,粪菌活性存在显著差异,离心组显著低于静置组(P<0.05)。但冷冻粪便使用静置和离心两种方法制备的菌悬液,其粪菌活性无显著差异(P>0.05)。无论是静置还是离心方法制备粪菌液,新鲜和冷冻两种不同处理方式下的粪菌活性均无显著差异(P>0.05),且对活菌的组成也无显著影响。使用静置法制备粪菌液时,FS、NS、GNS和NFS组间粪菌活性无显著差异(P>0.05)。综上表明,静置方法制备粪菌液时,超低温冷冻保存对粪菌活性及其活菌组成均无显著影响,这为大样本、长距离猪粪菌移植技术的应用提供了重要试验参考。  相似文献

A programme of selective anthelmintic therapy was used in a herd of 31 horses. Faecal egg counts were done during the months of September, November, January, March, May and the following September. Horses with greater than or equal to 100 eggs per gram (epg) were treated with ivermectin, and those with less than 100 epg were not treated. The criteria for adequate internal parasite control in the herd was a median herd faecal egg count of less than or equal to 100 epg. Effectiveness of selective therapy was assessed by faecal egg count after nine months of treatment and was determined to be adequate when a median herd egg count of 0 epg was obtained. However, on returning from pasture the following September, median herd egg count had risen to 325 epg. A statistically significant correlation was seen in the paired September faecal egg counts of the horses in that initial September faecal egg count was predictive for the following September. Initial September faecal egg count was related to the number of anthelmintic treatments required during the period of selective therapy, whereas age of horse was not. We propose that faecal egg counts be incorporated into strategic anthelmintic programmes as an economical tool for identifying and targeting herd members predisposed to shedding elevated numbers of helminth eggs.  相似文献

Faecal egg count patterns and clinical signs associated with gastro-intestinal (GI) nematodes of 107 zoo ruminants were monitored at fortnightly intervals for 1 year. The ruminants in this study were kept under different husbandry conditions at two sites of the Royal Zoological Society of Antwerp, the Antwerp Zoo and the Animal Park Planckendael. Artiodactylids involved were Arabian oryx (Oryx leucoryx), scimitar-horned oryx (Oryx dammah), bongos (Tragelaphus euryceros isaaci), sitatungas (Tragelaphus spekii gratus), common eland (Taurotragus oryx), impala (Aepyceros melampus), slender-horned gazelles, (Gazella leptoceros), blue wildebeest (Connochaetes taurinus taurinus), Kordofan giraffes (Giraffe camelopardalis antiquorum) and okapi (Okapia johnstoni). Nematode eggs were recovered from 586 of 1606 (36.5%) individual faecal samples, using flotation techniques. Infection levels were distinctly low at Antwerp Zoo, probably due to zero grazing and daily dung removal. At Planckendael, the herds of Arabian oryx, scimitar-horned oryx and slender-horned gazelles showed markedly higher egg counts than the other herds, with more than 10% of the faecal egg counts having more than 100 eggs per gram (epg) and maximum faecal egg counts of 600, 750 and 1350 epg, respectively. Faecal egg counts increased during the mid-grazing season (July) and peaked at the end of the grazing season (October). No clinical signs, such as loss of faecal consistency, could be correlated with faecal egg counts (P > 0.05). With the exception of significantly more Nematodirus spp. eggs that were present in juvenile eland, no differences in faecal egg counts could be found between the sexes and different age groups. Abomasa and intestines of 17 animals that died during the survey were available for total worm counts. In one Arabian oryx, four slender-horned gazelles and one sitatunga low burdens ranging from 200 to 14,300 were found. Nematode species recovered were Camelostrongylus mentulatus from the abomasa and Trichostrongylus retortaeformis, Nematodirus fillicollis, Capillaria spp. and Trichuris spp. from the intestines. Our findings suggest different nematode infection levels between herds, which are mainly due to husbandry conditions but to a lesser extent to species- or individual susceptibility. Identification of ungulates that are highly infected and knowledge of the seasonal variation of their helminths can contribute greatly to a well-adjusted species-specific management and helminth control program.  相似文献

The present study was aimed to evaluate the influence of flotation solution, sample dilution, and the choice of McMaster slide area (volume) on the reliability of the McMaster technique in estimating the faecal egg counts of gastrointestinal (GI) strongyles and Dicrocoelium dendriticum in a composite sample of faeces from naturally infected sheep. Fourteen flotation solutions having densities between 1.200 and 1.450, and six sample dilutions, 1:10, 1:15, 1:20, 1:30, 1:40 and 1:50 were used. Each of the six dilutions was divided into 70 aliquots in order to have five replicates of each of the 14 flotation solutions at each of the six dilutions. For each McMaster slide, the GI strongyle and D. dendriticum egg counts were performed under one grid (McM 0.15 ml), two grids (McM 0.3 ml), one chamber (McM 0.5 ml), and both chambers (McM 1.0 ml). Mean eggs per gram (EPG) of faeces of GI strongyles and D. dendriticum were calculated and statistical analyses were performed on the resulting data. The type of flotation solution used significantly influenced the EPG in the GI strongyles and in the D. dendriticum egg counts. All the sucrose-based solutions at density between 1.200 and 1.350 floated more GI strongyle eggs than the others. With respect to D. dendriticum, only six solutions were capable of floating eggs and the potassium iodomercurate solution (density 1.440) floated more eggs than the others. The reliability of the McMaster technique regarding sample dilution was high for both GI strongyle and D. dendriticum EPG at 1:10 and 1:15, and then progressively decreased with increasing dilution. The reliability of the McMaster technique regarding the choice of the McMaster slide area (volume) was high for both GI strongyle and D. dendriticum EPG at the McMaster slide area (volume) of 1.0 ml, i.e. the total area of the McMaster slide. The EPG counts resulting from choosing any of the other three McMaster slide areas (volumes), i.e. McM 0.15 ml, McM 0.3 ml, or McM 0.5 ml, produced unreliable over-estimates. The findings of the present study show that the highest reliability of the McMaster technique for estimating GI strongyle and D. dendriticum egg counts in faeces from pastured sheep is obtained when using flotation solutions based on sucrose for GI strongyles, and potassium iodomercurate for D. dendriticum, dilutions which do not exceed 1:15, and the McMaster slide area (volume) of 1.0 ml.  相似文献

Fifty horses from a herd known to have benzimidazole-resistant small strongyles were treated with febantel (6 mg/kg), combinations of febantel (6 mg/kg) and piperazine citrate (25 or 55 mg base/kg), thiabendazole (44 mg/kg), or placebo (0.6 ml of water/kg). Pretreatment and 7-day posttreatment fecal examinations were done. Fecal cultures, strongyle egg per gram (epg) counts, sugar flotation fecal examinations, and in vitro testing for benzimidazole resistance were performed. Results of fecal examinations before treatment were similar in all horses, and results of testing were positive for benzimidazole resistance. Horses treated with febantel and piperazine at all dosages had significantly lower mean strongyle epg counts and greater percentage reduction in mean strongyle epg counts (99.7% to 99.9%) 7 days after treatment, compared with those determined for horses treated with febantel, thiabendazole, or placebo. Adverse reactions to treatment were not observed.  相似文献

The present egg isolation method is both a rapid and simple technique for recovering large numbers of Fasciola hepatica eggs (1 X 10(7) eggs/gradient) from bovine bile. Bile from infected cattle was first passed through a 45 micron screen sieve. The F. hepatica eggs were collected from the surface of the screen by backwashing with a jet of distilled water. The resultant egg suspension was layered on a 60% to 100% (v/v) linear Percoll gradient prepared in distilled water. Centrifugation at 450 g for 20 min resulted in the formation of 2 visible bands and a pellet. The top band (density of 1.075 g ml-1) contained viscous debris and crystallized bile pigments. The second visible band (density of 1.093-1.099 g ml-1) consisted of a relatively pure population of F. hepatica eggs (greater than 93%) while the pellet contained only F. hepatica egg shells.  相似文献

The prevalence of parasitic infections in backyard flocks was surveyed using litter samples from 74 pens located on 12 farms in central lower Michigan. Eight species of birds were represented. Two methods of litter examination (a sucrose flotation technique and a multiple washing/ZnCl2 flotation) were compared; the sucrose flotation technique was found to be more useful and was used in the survey. The following parasites eggs/oocysts were observed; ascarid-type eggs (in 34 pens from nine farms), Capillaria eggs (in 30 pens from nine farms), Strongyloides eggs (in nine pens from five farms), Syngamus eggs (in five pens from four farms), and coccidial oocysts (in 40 pens from 10 farms). Contamination of litter with ascarid-type eggs, Capillaria eggs, and coccidial oocysts was commonly found, irrespective of bird species. The contamination level in pens with more than one bird species was lower than in pens with a single species. The relatively high contamination rate may be an indication of the risk of parasitic disease in birds that are not raised under controlled conditions in confinement.  相似文献
