Summary Twenty-one accessions of 3 wild Lactuca species which could be hybridised with L. sativa, the cultivated lettuce, were inoculated at different stages of plant development with 3 multivirulent isolates of Bremia lactucae. Nineteen sources of resistance to B. lactucae, not attributable to the previously recognised resistance factors 1–11 were identified. Two lines of L. serriola showed similar resistance patterns as lines carrying R11. The resistance of some accession was incomplete particularly at the seedling stage and this phenomenon may be race specific.Tests on segregating F2 populations of crosses between 2 different L. serriola accessions and L. sativa cultivars showed that the resistance in one line (LSE/18) appears to be inherited as a single dominant gene, which is sometimes incomplete in expression and allelic to either Dm6 or R7. The segregation patterns for resistance in PI 281876 did not give readily interpretable ratios.To assess the frequency of occurrence in B. lactucae populations of virulence factors to overcome this novel resistance, 11 of the novel sources of resistance were inoculated with numerous collections of the pathogen from the UK, Czechoslovakia and elsewhere and found to show a high level of resistance.  相似文献

Summary More than 200 accessions of three wildLactuca species were screened in the laboratory for race-specific resistance toBremia lactucae. Only a fewLactuca entries showed resistance both as seedlings and in a leaf disc test. Accessions ofL. serriola andL. sativa had a low degree of resistance. The best entries were also compared under field conditions. TheL. saligna entries were totally free from disease in the field test. High resistance was also recorded in a fewL. serriola entries and in lettuce cultivars such as Saffier and Mariska.  相似文献

Summary The response of Lactuca sativa, L. serriola and L. saligna to saprol was studied. Spraying the whole plant or part of a leaf caused wilting of susceptible plants. A similar effect was observed when leaves were soaked in a solution containing saprol.Inheritance studies with L. sativa and L. serriola showed that resistance to the wilting reaction caused by saprol is controlled by a single recessive gene.Contribution no. 198-E, 1978 series.  相似文献

Summary A total of 1789 accessions of several lettuce collections was screened to find new major gene resistance to the downy mildew fungus Bremia lactucae Regel. The accessions belonged to the species Lactuca sativa (N=1288), L. serriola (N=399), L. saligna (N=52) and L. virosa (N=50). A total of 20 races of B. lactucae were used, 14 of which were NL-races, isolated from cultivated lettuce in the Netherlands. The other six races were isolated from wild L. serriola in Czechoslovakia. The accessions were initially screened with two races: NL1 and NL3. Accessions with resistance to one or both of these races were tested with the other races. Phenotypes with new resistance were found in accessions of all four Lactuca species. Of L. sativa, four accessions were found with resistance phenotypes that could not be explained by combinations of known major genes. Many accessions of L. serriola had resistance phenotypes that indicated the presence of unknown resistance genes. All interactions between accessions of L. saligna and races of B. lactucae were incompatible in leaf disc tests, except for four accessions, which showed some sporulation with race NL6. Several accessions of L. virosa were resistant to all races used. Other accessions of L. virosa gave a race-specific interaction with B. lactucae.  相似文献

The wild lettuce species L. serriola, L. saligna, and L. virosa are important genitors in lettuce (L. sativa) breeding. Identifying these wild species can be problematic because in some cases they look very similar. Flow cytometry was tested for its reliability and general applicability as a tool to distinguish them. Three series of tests were conducted: (1) Tests with three accessions of L. sativa and one accession of each of the wild species, repeated three times throughout the year. In each repeat, the mean relative DNA amount of L. serriola was significantly higher than that of L. saligna, but significantly lower than that of L. virosa. The mean relative DNA amount of L. sativa did not differ from that of L. serriola.(2) Tests with each wild species represented by 10 accessions. Significant differences between the accessions within each species demonstrated the presence of intraspecific variation. Notwithstanding this intraspecific variation, the relative DNA amounts of all accessions of L. serriola were significantly higher than that of all L. saligna accessions, and significantly lower than that of all L. virosa accessions. Therefore, all accessions could be assigned to the appropriate species on the basis of their DNA amounts. (3) Tests with single plants from 10 accessions of each of the wild species. These test revealed that individual plants of L. serriola, L. saligna, and L. virosa can be reliably identified with flow cytometry, when aL. serriola sample of established identity is used as internal reference. Summary Many lettuce cultivars (Lactuca sativa L.) with high resistance to lettuce root aphid (Pemphigus bursarius L.) also carried the gene Dm-6 for specific resistance to downy mildew (Bremia lactucae Regel). Summary In common rice, Oryza sativa L. (n=12), the gene Am for non-glutinous is dominant over the gene am for glutinous. In African rice, O. glaberrima Steud. (n=12), no spontaneous glutinous strain has been found, but recently a glutinous strain of glaberrima was induced by EMS-treatment.The interspecific cytoplasm substitution line with sativa cytoplasm and glaberrima nucleus is male sterile. It has been confirmed that the complete restoration of pollen fertility in this male sterile line is attributed to a single dominant nuclear gene Rf j.Trial to transfer gene am from sativa to glaberrima was commenced with backcrosses of the F1 hybrid (glutinous sativa cv. Iwai-mochi × glaberrima ) to glaberrima type plants of the substitution line homozygous for Rf j,using the latter as the pollen parent. At the B1 step, highly fertile glaberrima type Am/am plants were obtained. Thereafter plants of this type were backcrossed to normal glaberrima as the recurrent pollen parent to complete the nuclear substitution. It was confirmed that the EMS-induced glutinous character of glaberrima was a monogenic recessive and that the same gene controls the expression of glutinous character in the different rice species, sativa and glaberrima.  相似文献

Summary None of the tested cultivars of lettuce was found resistant to Stemphylium leaf spot, a common disease in Israel. Within a Lactuca saligna population collected in wild lettuce in Israel, resistance was traced. Interspecific crosses of L. saligna x L. sativa were made and the mode of inheritance of resistance to this disease was studied. Summary Twenty-seven accessions of Hordeum species and interspecific hybrids were screened in a growth chamber for resistance against the aphid Rhopalosiphum padi. Resistance measurement was based on the aphids' population growth during a 16-day period. The mean number of nymphs produced per female was only about 14% of that on the control cv. Tellus. H. bogdani was the most resistant accession. As a group, interspecific F1-hybrids with common barley (H. vulgare × H. sp.) held an intermediate position. Differences in resistance between hybrids as well as between species (H. jubatum and H. lechleri) were observed. The most resistant species were all diploids. No effects of the host plant on formation of alatae or on the within-plant distribution of aphids were found.  相似文献

Summary The green peach aphid (Myzus persicae) is a serious pest of the tomato (Lycopersicon esculentum). Trichome-based host plant resistance of wild Lycopersicon species may offer an alternative to pesticides for management of this pest. Two wild species, Lycopersicon cheesmanii f. minor and L. pennellii, were crossed with L. esculentum and the types and densities of trichomes possessed by the F1 hybrids measured. The effects of these trichomes on M. persicae were assessed by placing nymphs on leaves and, 96 hr later, designating each nymph as either dead, alive, emigrating off the leaf or unaccounted for. A generalised linear model was used to detect relationships between the numbers of nymphs in each designation and the densities of trichomes and leaf area. Mortality of nymphs on L. pennellii hybrids was associated with high densities of type IV and low densities of type I trichomes. Mortality of nymphs on L. cheesmanii f. minor hybrids was associated with high densities of type III trichomes. An increase in the densities of type III trichomes on L. pennellii hybrids was associated with greater numbers of nymphs emigrating off the leaf whilst unaccounted nymphs on L. pennellii hybrids were associated with increased densities of type VII and type I trichomes. Further experimentation using excised leaflets found mortality on L. pennellii hybrids may be due to nymphs being hindered by type I trichomes and increased densities of type V trichomes were associated with greater numbers of unhindered nymphs. Implications of results are discussed in the context of introducing trichome-based host plant resistance into L. esculentum.  相似文献

DNA from female and male hemp (Cannabis sativa L.) plants belonging to nine different varieties were screened with180 RAPD primers in a search for sex-associated DNA markers. About 1500bands were produced in total, nine primers were found yielding one or two DNA bands amplified in all nine male DNA bulks and absent in all female DNA bulks. These putatively male-associated markers were then scored in three different F1progenies, deriving from a cross between a common male parent and three different female plants. The sex of the progeny was accurately scored on the basis of the floral phenotype, and the presence of the nine male-associated markers was verified by RAPD analysis. In all three progenies examined, all the male plants showed the DNA markers previously identified by bulk segregant analysis (BSA) on the hemp varieties, while all the female plants lacked them. The fact that the association between these markers and the staminate phenotype is found when examining male plants of distantly related varieties, and that such linkage is never broken when different progenies are examined, strongly supports the hypothesis that the markers found are physically located on the Y chromosome, in a region excluded from recombination during meiosis. Another marker was shown to be present in the male parent, in all the male plants of each progeny, and in 50% of the female progenies, while it was absent in the female parent; the possible occurrence of markers deriving from multiple amplification sites of the genome is discussed. Summary Gene action and heritability of groat protein percentage were determined in F1, F2, and F3 generations of nine crosses between three Avena sativa L. cultivars and three A. fatua L. selections. Relationships among groat protein percentage, grain yield, and 100-seed weight also were evaluated. The three A. sativa parents were Dal (high grain yield and intermediate groat protein percentage), Goodland (low grain yield and high groat protein percentage), and Stout (high grain yield and low groat protein percentage). The three A. fatua parents were chosen for the study on the basis of vigorous plant growth and high groat protein percentage. The study was conducted at Madison, Wisconsin in 1979 and 1980.There was partial dominance towards low groat protein percentage. Narrow sense heritability estimates for groat protein percentage were low in Dal and Goodland crosses and intermediate in Stout crosses. In the F2 generation, groat protein percentage was significantly higher in shattering than in nonshattering plants in 1979, but not in 1980. There were significant, positive correlations between groat protein percentage, 100-seed weight, and grain yield in F1 and F2 generations, but they were not large numerically. Relationships among these traits were either negative or nonsignificant in the F3 generation. Although our results indicated that selection for higher groat protein percentage is possible when a low protein A. sativa cultivar is used, most of our simple cross progenies from A. sativa x A. fatua crosses had weak straw and were susceptible to crown rust (Puccinia coronata Cda. var. avenae Fraser and Led.).  相似文献

Thirty-one accessions of four Lactuca species (L. serriola, L. saligna, L. aculeata, L. squarrosa) and one L. serriola×L. sativa hybrid were screened for field resistance to Bremia lactucae. In the L. serriola group, significant differences in the level of field resistance were distinguished. The accessions PI 281876 and PI 253467 were free of infection. Summary Recurrent backcrossing has been carried out with a view to transfer a gene for non-glutinous endosperm from two strains of O. glaberrima (Wx g /Wx g ) to glutinous japonica and indica varieties (wx/wx) of Oryza sativa. In the course of backcrosses Wx g /wx segregants were crossed with each of the two glutinous varieties of sativa as the respective recurrent male parent. The wx/wx and Wx g /wx segregants in the successive generations were consistently fully fertile and semi-sterile, respectively. The semi-sterility of Wx g /wx plants was attributable to abortion of most of the pollen grains carrying the gene wx. The nucleus but not cytoplasm was related to the semi-sterility. The Wx g /Wx plants having the gene for non-glutinous endosperm of a glaberrima strain and a japonica variety of sativa were also semi-sterile. Both wx- and Wx-megaspores in the plants heterozygous for the gene Wx g were deleteriously affected. The results could be explained by assuming that a factor tightly linked with the gene Wx g of glaberrima sterilizes gametes not carrying it in the heterozygotes and that the gametocidal action is exerted when combined with the sativa nucleus by the recurrent backcross method.  相似文献

Summary Restriction fragment length polymorphism (RFLP) analysis has several advantages over traditional methods of genetic linkage mapping, one of these being the starting point for map-based cloning. The recent development of an RFLP map of cowpea (Vigna unguiculata L. Walp) has allowed the investigation of associations between genes of interest and RFLP markers. A cross between an aphid (Aphis craccivora Koch) resistant cultivated cowpea, TT84S-2246-4, and an aphid susceptible wild cowpea, NI 963, was screened for both aphid phenotype and RFLP marker segregation. One RFLP marker, bg4D9b, was found to be tightly linked to the aphid resistance gene (Rac 1) and several flanking markers in the same linkage group (linkage group 1) were also identified. The close association of Rac 1 and RFLP bg4D9b presents a real potential for cloning this insect resistance gene.  相似文献

Summary After steam sterilizing of certain glasshouse soils a surplus of exchangeable manganese (SEM) occurs. Great differences exist between lettuce genotypes for sensitivity to SEM.Research on the inheritance of insensitivity was carried out by intererossing five lettuce genotypes: Lactuca sativa cys. Neptune (sensitive). Plenos. Troppo and Celtuce (all insensitive) and an accession of L. scrriola (insensitive). From analysing various populations (as F1's. F2's and B1's) it appeared that different numbers of genes for insensitivity were present in the parents varying from one gene in Plenos and Troppo, two genes in L. serriola to possibly four genes in Celtuce. Genes in the three first mentioned parents were linked in repulsion phase and a chromosome map for these genes was made.  相似文献

Summary Thirty-five rice (Oryza sativa L.) varieties, including 18 japonica, 5 javanica and 12 indica subspecies and 12 lettuce (Lactuca sativa L.) varieties were identified taxonomically, using PCR with originally designed 21 RAPD (Random Amplified Polymorphic DNA) primers and 8 sequence-specific primers, used for amplifying four specific DNA fragments. Use of these primers revealed polymorphisms among varieties in rice and lettuce and facilitates DNA fingerprinting. Dendrograms of both species based on polymorphisms were constructed and genetical relationships were established. In rice, half the number of amplified bands were polymorphic and almost all varieties differentiated. However, differentiation of minor genetic alterations among somaclonal variants or mutants and their mother varieties was not feasible. In L. sativa, 47% of the amplified fragments were polymorphic and all 12 varieties were differentiated. Some of the PCR fragments were variety or type specific, which could be used for indicators for type-selection. The dendrogram obtained showed differentiated clusters of crisphead, leaf and butterhead type, findings in good accord with the classification based on the genetic background.  相似文献

Summary A reliable impression of the resistance of lettuce plants to the leaf aphid Myzus persicae can be obtained via aphid honeydew production. Under controlled temperatures, the number of honeydew droplets produced by these aphids per plant in 180 minutes with five plants per genotype offers a good criterion of this resistance.  相似文献

Summary The possibilities to transfer important traits and in particular resistance to the beet cyst nematode (Heterodera schachtii, abbrev. BCN) from Raphanus sativus to Brassica napus were investigated. For these studies B. napus, R. sativus, the bridging hybrid ×Brassicoraphanus (Raparadish) as well as offspring of the cross ×Brassicoraphanus (Raparadish) ×B. napus were used. Reciprocal crosses between B. napus and R. sativus were unsuccessful, also with the use of embryo rescue. Crosses between ×Brassicoraphanus as female parent and B. napus resulted in a large number of F1 hybrids, whereas the reciprocal cross yielded mainly matromorphic plants. BC1, BC2 and BC3 plants were obtained from backcrosses with B. napus, which was used as the male parent. F1 hybrids and BC plants showed a large variation for morphology and male and female fertility. Cuttings of some F1 and BC1 plants, obtained from crosses involving resistant plants of ×Brassicoraphanus, were found to possess a level of resistance similar to that of the resistant parent. These results and indications for meiotic pairing between chromosomes of genome R with those of the genomes A and/or C suggest that introgression of the BCN-resistance of Raphanus into B. napus may be achieved.  相似文献
