Two ejaculates were harvested by electroejaculation on each of 3 d per week for 14 wk from 14, 12- to 24-mo-old Holstein bulls. Ejaculates during the first 2 wk served to stabilize sperm output. Data for the remaining ejaculates were used in a components of variance approach to determine the number of bulls per treatment and ejaculates per bull that would be needed to provide adequate sensitivity and precision for assessing treatment effects on seminal characteristics. The latter included the number of sperm (total and motile), volume and sperm concentrations of the sperm-rich fraction, and the percentage of progressively motile sperm in first and second ejaculates or per day. Replication requirements declined as the number of ejaculates per bull was increased to about 15 to 25 but declined minimally thereafter. The number of bulls needed per treatment varied in relation to the size of the treatment response to be detected but was much greater than the replication used in typical experiments. Replication for assessing sperm output in first ejaculates or per day was approximately one-half as great for experiments in which pretreatment information was used to adjust post-treatment data. Tables should be useful as a guide for designing efficient, cost-effective studies of known sensitivity and precision.  相似文献

Standard breeding soundness examinations plus measurement of scrotal surface temperature (SST), internal/scrotal testicular temperatures, testicular ultrasonographic echotexture, daily sperm production, and epididymal sperm reserves were conducted on 129, 16-month-old crossbred beef bulls. There were significant positive linear correlations between SST and internal scrotal/testicular temperatures, a positive linear regression (P < 0.06) of bottom SST with the incidence of secondary sperm defects, but a negative linear regression (P < 0.01) with the incidence of primary sperm defects. Testicular echotexture had a positive linear regression with daily sperm production (P < 0.002) and testicular tone had a negative linear regression (P < 0.008) with epididymal sperm reserves. Scrotal circumference had a positive linear regression (P < 0.04) with the percentage of progressively motile sperm, a negative linear regression (P < 0.1) with the incidence of primary sperm defects, and a positive linear regression (P < 0.0001) with epididymal sperm reserves. In addition to seminal quality and scrotal circumference, testicular ultrasonographic echotexture has considerable promise for augmenting breeding soundness examinations of bulls.  相似文献

Three dogs diagnosed as having asthenozoospermia were given three intramuscular injections of 50 mg testosterone(T)-depot plus 250 IU pregnant mare serum gonadotropin (PMSG) at 2-week intervals, and their plasma T and testicular transferrin (Tf) concentrations, testicular histology, and semen quality were examined during the period of hormone therapy. Plasma T concentrations temporarily increased, and there was a slight improvement in spermatogenesis. Increased Tf concentrations suggested that Sertoli cell function in all three dogs was promoted by hormone treatment. The results showed that semen quality, especially the percentages of motile sperm and abnormal sperm, were improved between 1 and 5 weeks after the start of T-depot plus PMSG treatment.  相似文献

As electroejaculation (EEJ) is prohibited for use on unanaesthetized animals in Sweden, there is a need for an alternative method of semen collection from bulls in the field. The aim of the present study was to evaluate the use of transrectal massage (TM) of the ampullae to collect semen from yearling beef bulls under field conditions in Sweden. Transrectal massage was performed on 52 yearling beef bulls. Volume of semen collected, duration of procedure, percentage progressively motile sperm, and sperm concentration were measured. Smears were prepared for sperm morphology examination. Semen samples were obtained from 47 of 52 bulls. Mean volume was 3.2 ml (SD +/- 3.7), mean duration of collection was 7.4 min (SD +/- 2.8), mean percentage progressively motile sperm was 43.5% (SD +/- 29.2) and mean concentration was 201.9 x 10(6) spermatozoa/ml (SD +/- 278 x 10(6)). Twenty-three of the 52 bulls were slaughtered 3-4 days after semen collection and aliquots of the cauda epididymal contents were collected for sperm morphology examination. The percentages of proximal droplets, abnormal tails and abnormal midpieces were significantly higher (p < 0.05) and the percentage of detached heads was significantly lower (p < 0.05) in the post-mortem samples compared with those in the TM samples. However, importantly there was no significant difference between the two sample types in the percentages of abnormal heads. This study demonstrates that semen can be collected from yearling beef bulls by TM. We think that TM constitutes a useful tool, when semen collection with EEJ or artificial vagina (AV) is not possible under field conditions, when included in the bull breeding soundness evaluation (BBSE) protocol. However, further studies are needed, and presently being carried out, to evaluate if semen samples collected by TM are comparable with semen samples collected by AV.  相似文献

Aging is a major risk factor for spermatogenesis deterioration. However, the influence of age on spermatogenic stem cells and their progenitors in bulls is largely unknown. Here, we report age-related changes in undifferentiated and differentiating spermatogonia in Japanese Black cattle with nearly constant sperm output, by using spermatogonial markers. The numbers of differentiating spermatogonia and more differentiated spermatogenic cells were significantly decreased in aged bovine testes compared with those in young testes. In contrast, the number of undifferentiated spermatogonia was maintained, and their proliferative activity did not differ significantly between young and aged bovine testes. Although severe calcification was only observed to a small extent in aged testes, fewer Sertoli cells and interstitial fibrosis were observed in noncalcified testicular regions. These results suggest that, even in old bulls with nearly constant sperm output, testicular spermatogenic activity declined whereas undifferentiated spermatogonia numbers were maintained. Thus, we propose that undifferentiated spermatogonia may be resistant to age-related changes in bovine testes. Because undifferentiated spermatogonia may contain stem cell activity, our findings highlight the potential utility of undifferentiated spermatogonia as an agricultural resource to produce spermatozoa beyond the natural bovine lifetime through transplantation and in vitro spermatogenesis in future animal production.  相似文献

The value of routine evaluation of bull semen was analysed for 117 AI bulls placed in two studs. Data from semen analysis of a total of 1635 ejaculates was compared statistically with the nonreturn rates of the bulls. The semen parameters which were significantly correlated to nonreturn rates were the motility of the freshly collected ejaculates (p = 0.0140) and post-thaw motility (p = 0.0075). The total number of motile sperm in the inseminate ranged from 10.9 to 19.3 × 106 and according to previous reports the effects of low motility should be fully 'compensated' when doses above 10 × 106 sperm/dose are used for insemination. In conclusion, the motility of freshly collected semen does not appear to be 'compensation' and a low percentage of motile sperm in an ejaculate may indicate other dysfunctions of the population of motile cells. Furthermore, post-thaw motility appears to correlate significantly with nonreturn rates. The largest proportion of the variation was explained by the breed of the bull and stud (42.2% of the variation), whereas the two motility parameters explained 10% of the total variation in nonreturn rates. Objective and precise evaluation of sperm motility in combination with other semen traits are needed to improve breeding efficiency. Although microscopic evaluation of sperm motility correlates with nonreturn rates of bulls, the methods are subjectively assessed and inaccurate and therefore do not allow a satisfactory prediction of fertility.  相似文献

Five mature stallions with poor semen quality were tentatively diagnosed as having testicular degeration of unknown cause. Testis samples from these five stallions, and from three mature stallions with normal semen quality, were obtained by castration and prepared for histomorphometry. Increased germ cell loss rates during late meiosis and spermiogenesis occurred in the stallions with idiopathic testicular degeneration. Poor semen quality, represented by a low percentage of morphologically normal, progressively motile sperm in ejaculates, appeared to be a good predictor of testicular degeneration in these stallions.  相似文献

Scrotal insulation and dexamethasone treatment were used as a model to compare the effect of testicular heating and stress on spermatogenesis. Insulation was applied to the scrotum of eight bulls (insulated) for a period of four days, eight bulls were treated daily for seven days with 20 mg dexamethasone injected intramuscularly, and four bulls were untreated controls. Semen from four bulls in each group was collected and evaluated over a six-week period after treatment. Blood samples for testosterone analysis were taken hourly for eight hours at the beginning and the end of the six-week period from the control bulls and before and after treatment from the four insulated and four dexamethasone-treated bulls that were not used for semen collection. At the end of the last blood sampling period, the four bulls in each group were castrated for the collection of testicular tissue for the determination of testosterone concentrations. Basal, peak episodic, and mean serum testosterone concentrations among control bulls, pre and postinsulated bulls, and pretreatment samples of dexamethasone-treated bulls were not different (p > 0.05); however, bulls that had received dexamethasone treatments had significantly lower basal, peak episodic, and mean testosterone concentrations (p < 0.05). Tissue concentrations of testosterone in control, insulated, and dexamethasone-treated bulls were not significantly different but tended to be lower in dexamethasone-treated bulls (p > 0.13). The spermiograms of the control bulls varied insignificantly over the six-week sampling period; however, there was a marked increase in sperm defects in insulated and dexamethasone-treated bulls. The types of sperm defects and the temporal relationships of rises and declines of sperm defects were quite similar for both treatments. All bulls recovered to approximately pretreatment levels of sperm defects by six weeks after the initiation of treatment. Results indicate that two of the most common types of insults to spermatogenesis in bulls, heat and stress, result in similar spermiograms.  相似文献

A limiting factor in canine artificial insemination (AI) is the low number of insemination doses obtained per ejaculate. In this study, semen was collected from dogs (n = 28) either once and frozen directly after collection or the same dogs were submitted to a dual semen collection with a 1-hr interval and the two ejaculates were combined for cryopreservation. We hypothesized that combining two ejaculates increases semen doses per cryopreservation process without negative effects on semen characteristics. Total sperm count was lower in semen from a single semen collection in comparison with the combination of the first and second ejaculate of a dual semen collection (p < .001). The percentage of motile and membrane-intact spermatozoa determined by computer-assisted sperm analysis (CASA) in raw semen did not differ between single and combined dual ejaculates and was reduced (p < .001) by cryopreservation to the same extent in single (motility 73.7 ± 1.8%, membrane integrity 65.6 ± 2.2%) and combined dual ejaculates (motility 72.7 ± 2.3%, membrane integrity 64.6 ± 2.5%). The percentage of spermatozoa with morphological defects increased after cryopreservation (p < .001) but was similar in single and combined dual ejaculates. The CASA sperm velocity parameters decreased with cryopreservation (p < .001) but did not differ between single and combined dual ejaculates. The number of insemination doses increased from 2.7 ± 0.4 for single to 4.7 ± 0.8 for combined dual ejaculates (p < .01), based on 100 million motile spermatozoa per frozen-thawed semen dose. In conclusion, combining two ejaculates collected at short interval for one cryopreservation process increases the number of AI doses without compromising semen quality.  相似文献

In the present study variability of bull sperm concerning percentages of sperm with intact plasma membranes (PMI), high mitochondrial membrane potential (HMMP) and positive acrosomal status (PAS) before and after cryopreservation (vKK; nKK) between bulls and between ejaculates within bulls was examined. Studies were performed on 4 semen samples each of 20 Deutsche Fleckvieh bulls. VKK-Values were 76.5% +/- 9.6% (PMI) 68.3% +/- 8.9% (HMMP) and 9.8 +/- 5.1% (PAS) and nKK-values were 38.1 +/- 14.0% (PMI), 38.2 +/- 14.0% (HMMP) and 30.9 +/- 12.1% (PAS). After freezing, variabilities in sperm parameter values between bulls (nKK: PMI: 49.8%, HMMP 52.1% and PAS: 56.6%) were nearly quite as high or higher than variabilities between ejaculates (nKK: 50.2%, 47.9% and 43.4%). VKK-values of PMI, HMMP and PAS were only fairly to moderately related (0.36 < r < 0.53; P < 0.05) to nKK-values. The results show that PMI, HMMP and PAS did not only vary between bulls, but also between ejaculates within bulls. As there are no high relationships in these sperm parameters between times before and after cryopreservation, each ejaculate should be examined after cryopreservation in order to receive a reliable information about the quality of cryopreserved sperm.  相似文献

At the beginning of the breeding season an eight-year-old standardbred stallion had semen with virtually zero sperm motility and an approximately 90 per cent incidence of midpiece and tail defects. The motility of the sperm improved to 7 per cent when semen was collected daily but its morphology did not improve. Electron microscopy revealed that the defects consisted mainly of a loss of microtubules in the axoneme and of disorganised midpieces. A pregnancy rate of 24 per cent per cycle and 44 per cent for the season was achieved in 32 mares after the insemination of whole ejaculates collected from the stallion frequently. The fertility was much higher than would have been expected from the characteristics of the semen. It is concluded that this sperm defect, reminiscent of the 'Dag defect' in bulls and the defect in T-locus mice, does not render the animal infertile.  相似文献

The morphology of sperm in raw semen was compared with that of the live sperm in semen which had been frozen and thawed. The thawed semen was stained with 6-carboxyfluorescein diacetate and propidium iodide and examined by fluorescence microscopy; smears of the raw semen were stained with eosin and nigrosin. Thirty-four ejaculates from 24 bulls of various breeds were examined. There were fewer abnormal heads, detached heads, coiled tails and proximal cytoplasmic droplets/pseudodroplets in the thawed semen than in the raw semen, there was no change in the number of bent tails, but the number of distal cytoplasmic droplets/pseudodroplets increased. There were no significant differences in morphology between ejaculates which passed or failed the osmotic resistance test after thawing, but failed batches tended to have more distal cytoplasmic droplets/pseudodroplets.  相似文献

Young beef bulls (n = 27) were used in a trial to study the effect of dihydrostreptomycin sulfate (DHS) or oxytetracycline (OTC) hydrochloride on spermatogenesis, epididymal sperm maturation, and freezability of sperm. Nine of the bulls were given a 22 mg/kg dose of DHS twice, 12 hours between doses. Nine other bulls were treated with OTC--1 dose of 26.4 mg/kg of body weight, and then 6 more doses each of 17.6 mg/kg, ca 12 hours between doses. The remaining 9 bulls were nontreated controls. The treatment regimens with the 2 antibiotics were without effect on spermatogenesis. These treatments also were without effect on seminal pH, ejaculate volume, percentage of motile spermatozoa, rate of spermatozoal motility, or concentration of spermatozoa in ejaculates harvested on day 3 or 7 of the study (day 0 = 1st day of treatment). There was a treatment-by-day effect on spermatozoal concentration; the number of sperm per milliliter was markedly increased on day 3 for OTC-treated bulls. The increased spermatozoal concentration in the OTC-treated group was associated with an influence of the antibiotic on ejaculation. All bulls given this antibiotic ejaculated without palpable penile engorgement or erection on day 3. On day 7 the rate of spermatozoal motility was increased in the 2 treatment groups compared with the rate in the control bulls. Flow cytometry of testicular and sperm cells was used to evaluate effects of pre-weaning zeranol implants on spermatogenesis. Forty five Angus-Simmental bulls were randomly assigned to three treatment groups of 15 bulls each: no implant, one implant at 30 d of age and two implants, one at 30 and the second at 120 d of age. Prior to slaughter at approximately 15 mo, semen was collected from 30 bulls, 10 of each group. Following slaughter, testes were weighed, and testicular biopsies and vas deferens sperm obtained from the same 30 bulls. Testicular and sperm cells were stained with acridine orange and measured by flow cytometry. Proportions of testicular haploid, diploid and tetraploid cells were determined by relative amounts of green (DNA) and red (RNA) fluorescence. Treatment of sperm at low pH prior to acridine orange staining potentially induces partial denaturation of DNA, detectable by the metachromatic shift from green (native DNA) to red (single-stranded DNA) fluorescence. The effect of this shift was quantified by alpha-t [alpha t = red/red + green) fluorescence]. Nonimplanted bulls had heavier (P less than .01) testicular weights than treated bulls. The proportion of haploid cells was greater (P less than .02) and diploid cells less (P less than .03) in testes of nonimplanted bulls. Sperm from implanted bulls had altered chromatin structure, indicated by higher (P less than .05) alpha t values. Flow cytometry is an effective means for detecting changes in testicular cell subpopulations and chromatin structure of sperm.  相似文献

The aim of this study was to evaluate the effects of breed and season on semen quality parameters of zebu bulls. Data (1,632 registers) of semen production from Gir (n?=?4) and Nelore (n?=?15) bulls were collected between October 2005 and November 2009. The ejaculates were collected twice a week during various seasons (summer, fall, winter, and spring) and evaluated for the following semen parameters: ejaculate volume, sperm concentration, sperm motility, forward progressive motility (FPM), and sperm morphology. Factor analysis was used to determine the relationship among variables. The effect of breed (Gir and Nelore) and season and their cross effect on each parameter and extracted factor were tested using ANOVA. A negative correlation (P?<?0.05) was observed between FPM and proximal droplet, as well as with abnormal loose head, abnormal small head, pouch formation, abnormal mid-peace, and strongly folded tail. Gir bull sperm showed more major defects, detached acrosome, and minor FPM (P?<?0.01), whereas Nelore bulls showed a higher number of sperm with normally loose head.  相似文献

The predictability of semen quality of mature sires from measurements at an early age is not well established. The aim of the present study was to determine age-dependent changes in the quality of bull semen from six Estonian Holstein (EHF) bulls, processed when the sires were 3, 5 and 7 years old. Fertility data such as 60-day non-return to oestrus rates (60d-NRRs) were available for 3-year-old bulls. From each batch, semen straws were analysed immediately after thawing [i.e. post-thaw (PT)] (controls) and after a swim-up (SU) procedure. The analyses comprised subjective and computerized measurements of sperm motility using computer-assisted sperm analysis (CASA) as well as estimations of sperm concentration, morphology and membrane integrity. There was a significant (p < 0.05) increase in the percentage of sperm motility (SU), membrane integrity (PT, SU) and normal tail and acrosome morphology (SU) with an increase in the age of the sires. The percentage of total motile spermatozoa PT measured by CASA correlated between 3- and 7-, and between 5- and 7-year-old bulls (p < 0.05). In addition, the proportion of head abnormalities tended to correlate between all three age groups both PT and after SU (p < 0.1). The sperm parameters correlating with fertility were average path velocity (VAP) (p < 0.001), total motility as measured by CASA (p < 0.01), linearly motile spermatozoa (p < 0.05) and CASA-assessed numbers of motile spermatozoa (p < 0.05), all after SU selection. The results showed that overall semen quality examined at 3 years of age is related to the semen parameters later in bulls' life. Moreover, CASA-assessed motility after SU seems to be a reliable marker for semen quality assessment as it shows correlation not only between the ages, but also to field fertility.  相似文献

Optimal sperm cryopreservation is a prerequisite for the sustainable commercial application of frozen-thawed boar semen for AI. Three experiments were performed to identify factors influencing variability of postthaw sperm survival among 464 boar ejaculates. Sperm-rich ejaculate fractions were cryopre-served using a standard freezing-thawing procedure for 0.5-mL plastic straws and computer-controlled freezing equipment. Postthaw sperm motility (assessed with a computer-assisted semen analysis system) and viability (simultaneously probed by flow cytometry analysis after triple-fluorescent stain), evaluated 30 and 150 min postthaw, were used to estimate the success of cryopreservation. In the first experiment, 168 unselected ejaculates (1 ejaculate/boar), from boars of 6 breeds with a wide age range (8 to 48 mo), were cryopreserved over a 12-mo period to evaluate the predictive value of boar (breed and age), semen collection, transport variables (season of ejaculate collection, interval between collections, and ejaculate temperature exposure), initial semen traits, and sperm quality before freezing on sperm survival after freezing-thawing. In Exp. 2, 4 ejaculates from each of 29 boars, preselected according to their initial semen traits and sperm quality before freezing, were collected and frozen over a 6-mo period to evaluate the influence of interboar and intraboar ejaculate variability in the survival of sperm after cryopreservation. In Exp. 3, 12 ejaculates preselected as for Exp. 2, from each of 15 boars with known good sperm cryosurvival, were collected and frozen over a 12-mo period to estimate the sustainability of sperm cryosurvival between ejaculates over time. Boar and semen collection and transport variables were not predictive of sperm cryosurvival among ejaculates. Initial semen traits and sperm quality variables observed before freezing explained 23.2 and 10.9%, respectively, of the variation in postthaw sperm motility and viability. However, more that 70% of total variance observed in postthaw sperm quality variables among ejaculates was explained by boar. This indicates that boar is the most important (P < 0.001) factor explaining the variability among ejaculates in sperm cryosurvival, with most (14 of the 15 boars in Exp. 3) showing consistent (P > 0.05) sperm cryosurvival over time.  相似文献

Contents: Repeatability and heritability coefficients were estimated for ejaculate volume, mass movement, individual motility, sperm concentration, first and second post-freezing control of motility, number of doses frozen, total number of spermatozoa and total number of motile spermatozoa per ejaculate in the first ten ejaculates of 175 Swiss Holstein and 673 Simmental young bulls (200 pure Simmental, 473 Simmental x Red Holstein crosses). Data were analysed using the univariate individual animal model with effects of interval between consecutive collections, age of bull at collection, season of collection, breed group of Simmental bulls according to percentage of Red Holstein genes, bull (within breed group) and permanent environment. Crossbred bulls had a better performance in all traits. Repeatability coefficients ranged from 0.28 to 0.46. They indicate that the prediction of semen quality in young bulls of the Swiss Holstein and the Simmental breed is possible to some extent. Heritabilities in Swiss Holstein bulls were generally very low and did usually not exceed 0.10, but standard errors of these estimates were high. Relatively high heritabilities (0.18–0.30) were found for Simmental bulls, Differences in heritabilities between these two breed groups were probably a consequence of the low number of Swiss Holstein bulls.  相似文献

This study was designed to investigate the effects of feeding‐protected conjugated linoleic acid (CLA) on the semen production and sperm freezability in Holstein bulls. Twelve bulls were randomly assigned to two groups (n = 6 per group). Bulls received the normal diet (control group) or the normal diet top‐dressed with 50 g of CLA (treated group) for 10 weeks. The control group received 40 g/day calcium soap of fatty acid. Fresh and post‐thaw semen quality was assessed on ejaculates collected at the 0, 4, 6, 8 and 10 week of supplementation. Semen evaluations including sperm concentration, motion characteristics (subjective and computer‐assisted), viability (Eosin–Nigrosin), membrane integrity (hypo‐osmotic swelling test) and abnormality were conducted. Semen volume, sperm concentration and total sperm output were not affected by dietary treatment (p > .05). The proportion of spermatozoa with abnormal morphology in fresh semen significantly increased (p < .05) in the CLA‐fed group compared to control group. Also, in CLA‐fed group, the proportion of post‐thaw spermatozoa with abnormal morphology at week 10 of trial was significantly higher in CLA than control group (p < .05). Progressive motility tended to be increased in the CLA‐fed group, although dietary supplementation did not affect other CASA parameters or viability in fresh and frozen‐thawed sperm. In this study, CLA supplementation had little positive effect on fresh or post‐thaw sperm quality of Holstein bulls.  相似文献
