Infectivity of resting spores ofPolymyxa betae in soil stored air-dry or moist was determined by assessing infection of bait plants that were exposed to the soil. Storage of soil under air-dry conditions at room temperature resulted in a delayed onset of germination of resting spores compared to germination in soil stored under moist and cool conditions, as inferred from the infection of the bait plants. Bait plants had to be exposed for more than 12 h to flooded infested soil before germination and infection had occurred. However, when soil was prewetted for 24 h before exposing bait plants, germination, infection and transmission of beet necrotic yellow vein virus (BNYVV) were accomplished within 12 h, but only with the moistly stored soil. When resting spores isolated from roots were stored for 4 and 8 weeks under dry conditions at 22°C, germination of viruliferous spores, as measured by detection of BNYVV in bait plants exposed for 48 h to the spores, was less than that of spores stored in moist soil at 22°C. Approximately 100% of bait plants were infected after exposure to resting spores that were frozen in demineralized water or stored cool (5°C) in water or moist soil for 42 weeks. Air-dry cool storage for 42 weeks resulted in a low percentage of infection. Storage conditions of soil influence the results of bioassays for detection of rhizomania when short baiting periods are applied, whereas differences in infectivity were not detected using a bioassay with long duration.  相似文献

Sclerotium cepivorum (the causal agent of white rot of onions) persists in soil in the form of sclerotia whose germination is triggered by volatile sulphides and thiols released into the soil from roots of the genus Allium. The effects of treatment of the soil with diallyl disulphide (DADS) on the germination of these sclerotia has been studied, because if the germination can be triggered in the absence of the host crop, control of the fungus could be achieved. Five isolates of the fungus responded similarly but marked effects of DADS concentration and of the time-of-year of application were discovered, poor results being obtained with summer applications of DADS. Promising results were obtained at other times of the year with DADS flooded on to land containing sclerotia. The effectiveness of DADS treatment was shown to be related to the rate at which the material disappeared from soil, which was itself dependent upon soil temperature.  相似文献

A total of 188 fungal isolates was obtained from the rhizosphere of Vicia faba grown in an Egyptian soil heavily infested with Orobanche species. Agar cultures of 58 isolates inhibited the germination of conditioned seed of Orobanche crenata exposed to the germination stimulant, GR24. Filtrates of inhibitory fungi grown in liquid medium for 9–15 days were also assayed and those of five isolates, which were morphologically similar, inhibited germination even when diluted 16-fold. The fungus was identified as Myrothecium verrucaria (Alb. & Schwein.) Ditmar by its morphology and the nucleotide sequence of the ITS1 and ITS2 regions of the ribosomal repeat unit. Purification of the inhibitor to homogeneity was accomplished by solvent partitioning, flash chromatography on silica gel, semi-preparative HPLC on a reversed phase C18 column, solid phase extraction and tlc on silica gel. The inhibitor was identified as verrucarin A by nuclear magnetic resonance spectroscopy and comparison of the spectra with those of an authentic sample of the compound. A preliminary experiment demonstrated that infection of V. faba by O. crenata could be prevented by addition of spores of the fungus to soil infested by the parasite.  相似文献

Seed transmission ofPyrenophora tritici-repentis, a common foliar pathogen of wheat, was investigated in soft white winter wheat cv. Frankenmuth and found to be non-systemic; the emerging coleoptile was infected externally by hyphal growth from the infected pericarp. Hyphae from the infected coleoptile then infected the first and second seedling leaves as they emerged. Coleoptile symptoms ranged from tiny brown streaks or spots to large, brown necrotic areas accompanied by cracking and distortion of the coleoptile. Small brown spots sometimes occurred on the first and rarely on the second seedling leaves, often accompanied by leaf distortion. Pseudothecial initials of the fungus were present within or on the seed remnants. Seed transmission efficiency was as high as 92%in vitro and 60% in potting soil outdoors. Seed infection did not affect germinationin vitro, but slightly reduced emergence in potting soil. Seedling weight and height were reduced significantly. In potting soil, seed infection also resulted in delayed plant growth and increased tan spot severity at later stages of plant development. Under controlled conditions, seed transmission efficiency and incidence of pseudothecia on seed were negatively correlated with seed germination temperature in the range of 9 to 21 °C, whereas frequency of recovery of the fungus from symptomatic coleoptiles and leaves was positively correlated with seed germination temperature.These results suggest that infected seed may serve as a source of inoculum for tan spot epidemics and for dispersal of strains of the fungus to new areas.  相似文献

Spongospora subterranea, causal agent of powdery scab of potatoes and vector of potato mop-top furovirus, survives in the soil as balls of resting spores (cystosori). So far, the factors affecting longevity, germination and infectivity of cystosori have not been investigated. A rapid and versatile bioassay with tomatoes as bait plants has been developed to quantify the infectivity of cystosorus inoculum or infested soil. The intensity of root infection, as a measure of infectivity, was determined by evaluating the quantity of zoosporangia present in epidermal cells and root hairs of the whole, stained root system. A correlation was obtained between the intensity of root infection and the cystosorus inoculum density in nutrient solution. Sterile soil suppressed the inoculum potential of pure cystosori. Infectivity of untreated soil decreased with increasing time of storage. Root infection was not influenced by the pH level of the nutrient solution.  相似文献

The effects of different inocula of the mycoparasite Coniothyrium minitans on carpogenic germination of sclerotia of Sclerotinia sclerotiorum at different times of year were assessed. A series of three glasshouse box bioassays was used to compare the effect of five spore-suspension inocula of C. minitans , including three different isolates (Conio, IVT1 and Contans), with a standard maizemeal–perlite inoculum. Apothecial production, as well as viability and C. minitans infection of S. sclerotiorum sclerotia buried in treated soil, were assessed. Maizemeal–perlite inoculum at 107 CFU per cm3 soil reduced sclerotial germination and apothecial production in all three box bioassays, decreasing sclerotial recovery and viability in the second bioassay and increasing C. minitans infection of sclerotia in the first bioassay. Spore-suspension inocula applied at a lower concentration (104 CFU per cm3 soil) were inconsistent in their effects on sclerotial germination in the three box bioassays. Temperature was an important factor influencing apothecial production. Sclerotial germination was delayed or inhibited when bioassays were made in the summer. High temperatures also inhibited infection of sclerotia by C. minitans . Coniothyrium minitans survived these high temperatures, however, and infected the sclerotia once the temperature decreased to a lower level. Inoculum level of C. minitans was an important factor in reducing apothecial production by sclerotia. The effects of temperature on both carpogenic germination of sclerotia and parasitism of sclerotia by C. minitans are discussed.  相似文献

The effect of metham-sodium on Orobanche aegyptiaca Pers. was tested in the laboratory and in soil columns. The laboratory experiment was carried out on O. aegyptiaca seeds placed in Petri dishes and germinated with GR24, a synthetic strigol analogue. In soil columns, metham-sodium was applied by application of the chemical through the irrigation water to three different soils and its dissipation determined in three soil layers by gas chromatography, by a lettuce bio-assay to check the herbicide's phytotoxicity, and with a flax bioassay to check its effect on O. aegyptiaca. Results of the germination experiments showed an exponential decrease in O. aegyptiaca germination, parallel with the increase of metham-sodium concentration, with an average effective concentration (EC50) of 18 mg L?1. In a soil column, methylisothiocyanate (MIT, the metham active product) rapidly disappeared from the upper soil level (0–10 cm) within 24 h. Seven days after application only traces of MIT remained in all soil layers in all soils, except for the sandy Rehovot soil that contained low concentrations in the lower soil layer (20–30 cm). Flax bioassay confirmed the chemical analysis, showing that O. aegyptiaca tubercles developed only on plants grown in the upper soil layer of all three soils.  相似文献

According to the results of bioassay studies, 50 p.p.m. and higher dosages of fentin acetate inhibited the growth of Phytophthora citrophthora in situ. Zoospore movement of P. citrophthora varied between 3 and 15 min at 0.5, 1, 10, 100 and 500 p.p.m. fentin acetate. The fungicide also inhibited fruit infections caused by P. citrophthora zoospores at 5 p.p.m. concentration, and protected citrus seedling roots against infection by the fungus at 20–50 p.p.m. concentrations in water culture. Fruit infection was significantly lower in fentin acetate plots than in those without fungicide applications. Stem infection was 15.5 and 8.3% when fentin acetate was applied one or three times respectively as a soil drench in a lemon orchard, but reached 74.9%) in control plots. Fentin acetate treatment decreased inoculum potential to 1/2 and 1/128 in the root zone from the first to the third application.  相似文献

The mechanism of the biofungicide Prestop® (Clonostachys rosea) was investigated for control of clubroot (Plasmodiophora brassicae) on canola. The key product components were partitioned and assessed for their effect on pathogen resting spores, root hair infection (RHI) and disease development using light microscopy, quantitative PCR and different application treatments during infection. The whole product of Prestop was consistently more effective than the C. rosea conidial suspension or product filtrate alone in reducing RHI and clubroot development. This biofungicide showed little effect on germination or viability of resting spores. Two‐application treatments at seeding and 7–14 days after seeding achieved greater clubroot control than a single application of the biofungicide at either seeding or post‐seeding stage. This may indicate the need to maintain a high biofungicide dose in the soil during primary and secondary infection. This biocontrol fungus colonized the rhizosphere and interior of canola roots extensively, and possibly induced plant resistance based on up‐regulation of the genes that are involved in jasmonic acid (BnOPR2), ethylene (BnACO) and phenylpropanoid (BnOPCL, BnCCR) biosynthetic pathways. It is concluded that the biofungicide Prestop suppressed clubroot on canola at least via root colonization and induced systemic resistance (ISR), and the latter may be through the modulation of phenylpropanoid and jasmonic acid/ethylene metabolic pathways elicited by the fungus.  相似文献

Mannose-binding rice lectin (MRL), which is almost identical to the salt-induced protein SalT, binds to mannose and glucose residues. Expression of the MRL gene in response to infection with Magnaporthe oryzae, the rice blast fungus, was stronger in the incompatible interaction than in the compatible. Transgenic rice plants that constitutively over-expressed MRL strongly suppressed the growth of invasive hyphae of the fungus on leaf sheaths and the development of typical susceptible-type lesions on leaf blades, but did not affect penetration by the fungus in comparison with the wild-type. On a polycarbonate plate, purified recombinant MRL inhibited conidial attachment and appressorium formation but not conidial germination. These results suggest that MRL may play an essential role in disease resistance by suppressing development of M. oryzae in situ.  相似文献

Carboxin was toxic to the fungus Deuterophoma tracheiphila. Uptake and translocation of carboxin (2,3-dihydro-6-methyl-5-phenylcarbamoyl-1,4-oxathiin) in lemon seedlings were assessed in plant extracts by a bioassay technique. Root uptake and translocation to stem and leaves were demonstrated from both an aqueous solution and drenched soil. Leaf uptake was lower than root uptake, and could be enhanced by removing the epicuticular wax. Chromatographic tests suggested that the compound responsible for fungitoxicity within plant tissues may have been carboxin itself.  相似文献

A quantitative bioassay for estimation of photosynthetic inhibitors in soils A bioassay procedure for the quantitative determination of photosynthetic inhibitors in soil samples, hydroponic and staple fibres is described In detail. Garden cress (Lepidium sativum L.) has proved very suitable as test plant. Monolinuron and methabenzthiazuron were used to demonstrate the capability of the method for estimating herbicidal activity and plant availability, and for monitoring the total residues of herbicides in different soils. The method was found capable of assaying quantitatively concentrations as low as 0,04 ppm monolinuron. Photosynthetic inhibitors did not affect germination or seedling growth during the first 5 days after sowing: retardation of growth occurred only after food reserves in the endosperm were consumed. Therefore the content of food reserves in the cress seeds was determined in a dark-germination test and substracted from all yield results to give the dry matter yield under herbicide stress.  相似文献

Wilt caused by the fungus Fusarium oxysporum f. sp. ciceris adversely affects the productivity of cultivated chickpea. For the management of this disease, seed and soil application formulations developed from another fungus, Trichoderma species, were evaluated. In pot experiments, T. harzianum-based formulations Pusa 5SD for seed dressing and Pusa Biopellet (PBP) 10G and Pusa Biogranule (PBG) 5 for soil application, and T. viride-based formulations Pusa 5SD for seed dressing and PBP 4G and PBG 4 for soil application, were found to be highly effective against the disease. A combination of PBP 4G (T. viride) for soil application and Pusa 5SD (T. harzianum) for seed treatment together with a fungicide, carboxin, provided the highest seed germination, shoot and root lengths and grain yield with the lowest incidence of wilt in chickpea under field conditions. Individually, soil application of PBP 4G, and seed treatment with Pusa 5SD were effective in reducing the incidence of wilt and increasing the grain yield of chickpea, but their effectiveness was greater when applied as a combination. Thus, combined application of the formulations of two different species of Trichoderma in two modes of application is recommended for the management of chickpea wilt.  相似文献

The effects of host plant exudates, light and temperature on germination of oospores of Peronospora viciae f.sp. pisi in vitro were investigated. Seed and root exudates did not increase percentage germination, whereas light inhibited germination. The first germ tubes appeared after 4, 7, and 14 days of incubation at 15, 10 and 5 °C, respectively. The eventual level of germination was highest and had similar values at 5 and 10 °C. At 20 °C germination was poor and at 25 °C no germination was observed. Oospores placed on membrane filters were incubated on soil. When oospores were retrieved from the membrane filters after six days and placed in water at 10 °C, they germinated within 2 days. On soil significantly less oospores germinated than in water. Germinability of oospores stored in the dark at 5 or 20 °C at 30 or 76% RH was studied over a two-year period. Germinability generally increased over time, but fluctuations were observed indicating the occurrence of secondary dormancy. Time courses of germinability were generally similar for oospores stored at several temperatures and humidities. No effect of light on time course of germinability was found when oospores were exposed to alternating light-dark periods or stored in continuous dark for 140 days. Percentage germination observed in a germination assay was correlated with percentage infection determined in a bioassay.  相似文献

在辽宁省发現一种寄生于柞蚕寄生蝇蛹体上的食虫菌,鑑定为赤色穗状菌[Spicaria fumoso-rosea (Wize) Vassilijevsky],是我国首次詳細观察記载。此菌容易培养,在馬鈴薯、甘薯块上,以及馬、羊粪混和腐植土上都可以生长发育良好。致病力強,自然寄生率为3.4—20.3%;接种感染寄生率为10—100%。此菌除寄生于柞蚕寄生蝇蛹外,还可寄生于粘虫、苹果食心虫及家蝇等。本菌孢子发芽所需相对湿度为98—100%,在98%以下不能发芽;土壤绝对含水量15—55%时发生良好,尤以35%为最适宜。最适温度为20—24℃。小区接种試驗結果:在地面撒菌粉让寄蝇蛆爬行钻入,寄生率为72.98%;在土深3与5厘米处撒菌粉,寄生率为40.24%和43.98%。  相似文献

Colletotrichum gloeosporioides f. sp. malvae, isolate Biomal®, ATCC 20767, was originally isolated from round-leaved mallow (Malva pusilla) and developed as a weed biocontrol agent. Ribosomal DNA sequence analysis was recently used to re-classify this fungus as C. orbiculare, which is an aggregate species with a number of formae speciales. Several morphological features of ATCC 20767 were examined that were consistent with those described for C. orbiculare, and inoculation of a number of Nicotiana species and several cultivars of N. tabacum showed that this fungus was pathogenic to many of these previously undescribed hosts. Spore germination and appressorium formation were higher on tobacco than previously observed on round-leaved mallow. The pathogen produced melanized appressoria on N. tabacum leaves that formed preferentially at the anticlinal epidermal cell wall. A symptomless phase of infection persisted for 72–96 h postinoculation, during which time the fungus first produced a spherical infection vesicle from an infection peg, and then large primary hyphae which grew through the epidermal cells. The large primary hyphae were highly constricted at the points of penetration of the host cell walls. Thin secondary hyphae appeared at 96–120 h postinoculation coinciding with the appearance of light green, water-soaked spots and the formation of acervuli. The infection of tobacco by C. orbiculare ATCC 20767 is not a non-specific interaction but appears to follow an intracellular hemibiotrophic infection process that is very similar to that established for the C. orbiculare infection of round-leaved mallow, cucurbits and beans.  相似文献

Phoma koolunga is a recently recognized pathogen in the ascochyta blight complex of field pea (Pisum sativum). Unlike the other three ascochyta blight pathogens, survival of P. koolunga is poorly understood. Survival of this fungus was examined on field pea stubble and as pseudosclerotia on the surface of, and buried in, field soil. Pseudosclerotia were formed in plates containing potato dextrose agar (PDA) mixed with sand or amended with fluorocytocin. After 1 month, P. koolunga was recovered on amended PDA from 93% of stubble sections retrieved from the soil surface, 36% of buried stubble sections and 100% of pseudosclerotia buried in field soil, pasteurized or not. The frequency of recovery of P. koolunga decreased over time and the fungus was not recovered from stubble on the soil surface at 15 months, nor was it recovered from stubble buried in soil at 11 months or later, or from pseudosclerotia buried for 18 months. In a pot bioassay, most ascochyta blight lesions developed on plants inoculated with stubble retrieved from the soil surface after 1 month. Infectivity of the inoculum decreased over time. Disease on plants inoculated with stubble that had been buried or left on the soil surface for up to 6 and 5 months, respectively, and pseudosclerotia retrieved at 14 months and later from field soil did not differ from the non‐inoculated control. These results suggest that field pea stubble may play a role in survival of P. koolunga, especially if it remains on the soil surface. In addition, pseudosclerotia were able to persist in soil and infect field pea plants into the next season.  相似文献

This paper deals with honey root rot of apricots, caused by the fungus Armillariella mellea (Vahl.) Karst. =Armillaria mellea (Vahl.) Quelet. Symptoms of the disease are described, both on the root system and on the aerial parts, where the action of the fungus mostly provokes a sudden wilting or apoplexy. The infection process is elaborated and the importance of the subterranean rhizomorph in the infection cycle is emphasized. Conditions favourable and unfavourable to the disease are briefly discussed and it is shown that presence of woody remains in the soil is the most important factor in the spread of the disease. Some isolation and inoculation methods are described. Discussion of the methods of control places emphasis mainly on preventive measures, such as use of tolerant rootstocks, destruction of potential nutrient sources and soil disinfection by fumigation.  相似文献

玉米丝黑穗病是白城地区的重要病害,一般发病率为3—5%,重者高达30%以上,是生产上急待解决的问题。其症状以正常型为主,有时出现矮化型、矮化丛生型和多分蘖型等特异症状。病株以雌穗为黑穗、雄穗为健穗及雌雄穗皆为黑穗者居多。病穗上自然脱落的厚垣孢子萌发率高,病穗内的萌发率明显降低。此病为系统侵染病害,种子及土壤均可带菌传病,以土壤带菌为主,属土传病害。厚垣孢子在土壤里能存活三年,土壤带菌量越多,菌土覆盖种子越厚,发病率越高。丝黑穗病菌侵染玉米幼苗时间较长,从种子萌芽到四叶期最易侵染,五叶期后,侵染明显下降,八叶期后不再侵染。在幼苗叶片上接菌不能侵染致病。玉米丝黑穗病菌不能侵染高梁。高梁丝黑穗病菌能侵染玉米,但侵染率甚低。侵染玉米后的病菌,不再侵染高梁,只侵染玉米,且致病力明显增强。两菌是同一菌种的不同生理型。用来自九个省、市的丝黑穗病菌接种于不同抗性的自交系鉴定寄主,其致病力不同,陕西和新疆的致病力弱,辽宁和吉林的致病力强。在田间人工接种条件下,鉴定了二百余份自交系和杂交种的抗病性,品种间有明显差异,高抗的近于免疫,高感的发病率在90%以上。抗病性遗传规律表现为双亲高抗的,其后代亦抗,以母本的抗病性起主导作用。选百余种农药处理种子,筛选结果:个别药剂防效有高达70%以上者,但年度间效果不稳定,多数药效稳定在50%左右。  相似文献
