为探究干旱锻炼对小麦幼苗期抗氧化特性的影响,设置对照处理和干旱锻炼处理,在一心一叶时用PEG6000营养液进行干旱锻炼,之后恢复霍格兰氏全营养液,在三叶一心时分别进行干旱胁迫处理,观测幼叶中与抗氧化特性相关物质的动态变化.研究发现,随干旱程度及胁迫时间的增加,叶片SOD、POD活性、可溶性蛋白不同程度的减小,MDA、可溶性糖含量显著升高;与对照相比,干旱锻炼后叶片SOD、POD活性、可溶性蛋白及降低程度较小,可溶性糖含量升高程度较大,MDA升高程度较小;不同干旱程度下,25%PEG6000胁迫下干旱锻炼小麦的抗氧化能力较35%PEG6000胁迫下较高.由此说明,小麦经过一定浓度的干旱锻炼能够明显增强其对干旱环境的适应能力,有利于幼苗期的生长,也为小麦的抗逆性研究提供理论支持.  相似文献

2001年田间小区试验研究了转Bt基因棉和转双价基因(Bt+CpTI)棉对棉苗蚜及其天敌种群动态的影响;室内研究了转基因抗虫棉对两种棉蚜天敌的生物学影响.结果表明,转基因抗虫棉对棉蚜种群数量的影响不明显,未达显著水平.单作转基因棉田棉蚜发生期比麦套棉田提前5天左右,发生数量是麦套棉田的1.6倍,表明麦棉套作可有效的控制苗蚜的为害.室内研究表明,异色瓢虫对用Bt棉处理的的棉蚜的捕食量比对照增加22.0%,棉蚜茧蜂对用Bt棉处理的棉蚜的寄主率降低23.5%.  相似文献

本研究以采自陕西省柞水县野生大豆种子为供试材料,并模拟该地区土壤生境特点,分别采用聚乙二醇(PEG6000)、NaCl以及二者混合液三种方式浸种,研究PEG6000和NaCl浸种对野生大豆种子萌发及幼苗生长的影响。研究表明:三种浸种处理对野生大豆的种子萌发无显著作用;而PEG6000和NaCl浸种处理均显著增加了野生大豆幼苗可溶性糖含量、可溶性蛋白含量、SOD和CAT活力;但PEG6000和NaCl混合液浸种则降低了幼苗可溶性糖含量和SOD活力,不利于野生大豆的生长。推测干旱以及Na+含量较高的生境,不利于野生大豆幼苗的生长,可能是造成野生大豆濒危的原因之一。  相似文献

 对新疆棉区主栽的9个棉花品种进行了叶片形态特性、生化物质含量测定,结果表明,品种间抗蚜程度存在显著差异,新海21号抗蚜性最强,中棉所35和81-3感蚜。品种叶片的蜡质含量越高,对棉蚜的抗性越强,茸毛密度大,影响棉蚜的取食;游离棉酚和可溶性糖含量与抗蚜程度呈显著正相关;单宁含量与蚜害指数呈显著正相关;氨基酸中的谷氨酸含量越高,抗蚜性越强。逐步回归和相关分析发现,氮含量和氨基酸总量与抗蚜程度无明显相关性。品种的抗蚜性是多种特性综合作用的结果。  相似文献

以克新一号品种为试验材料,采用组织培养方法,研究了钾对NaCl胁迫下马铃薯脱毒苗脯氨酸、可溶性蛋白、可溶性糖含量、相对电导率和根系活力的影响。结果表明:NaCl胁迫下,不同浓度钾处理对马铃薯脱毒苗脯氨酸含量、相对电导率和根系活力影响显著,但对可溶性蛋白和可溶性糖含量的影响不显著。在25mmol/L和50mmol/LNaCl胁迫下,培养基中添加8mmol/L K2SO4,脯氨酸含量最高,根系活力最大,相对电导率最低。但在75mmol/L NaCl胁迫下,4mmol/L K2SO4处理,缓解效果最好。综合分析表明:钾具有缓解NaCl胁迫对马铃薯脱毒苗伤害的作用,但钾的添加量并非越多  相似文献

为鉴定分枝期不同大豆品种的耐旱性,以2个杂交大豆品种和2个常规品种大豆为材料,以高分子聚乙二醇6000(PEG6000)作为水分胁迫渗透剂,通过测量叶片叶绿素、脯氨酸、可溶性糖、丙二醛含量和谷胱甘肽还原酶活性,探讨不同PEG浓度(0、5%、10%、15%)对分枝期(30d苗龄)大豆生理特性的影响。结果表明:干旱胁迫导致4个品种大豆叶片丙二醛含量上升、可溶性糖含量和脯氨酸含量下降;吉育72和杂交豆2号叶绿素含量和谷胱甘肽还原酶活性均下降,其余2个品种表现出相反趋势。根据干旱胁迫后4个品种大豆的表型特征和对上述5种生理指标的综合反应,可知分枝期杂交豆5号和吉育88比杂交豆2号和吉育72表现出更强的耐干旱胁迫能力。  相似文献

为探究外源CO缓解干旱胁迫下黄瓜种子萌发生长的内在机制,以黄瓜品种新津研四号为试材,高铁血红素(0. 01μmol/L)为CO供体,采用PEG-6000 (聚乙二醇)模拟干旱胁迫,研究外源CO对干旱胁迫下黄瓜种子萌发生长、养分吸收能力、质膜稳定性、渗透调节物质含量、水解酶活性及其同工酶表达水平的影响。结果表明,外源CO能够提高干旱胁迫下黄瓜种子的发芽势、发芽率和发芽后的根长、胚轴长及鲜质量,能够促进黄瓜种子的吸胀,改进根系活力,显著降低质膜透性和丙二醛(MDA)含量,增加可溶性蛋白和可溶性糖的含量,还能诱导增强淀粉酶和酯酶的活性及相应同工酶的表达水平。与干旱胁迫处理组(PEG)相比,胁迫前施用CO处理组(PEG+CO)的根系活力提高了12. 57%,可溶性蛋白和可溶性糖含量分别增加了5. 84%和32. 56%,淀粉酶和酯酶的活性分别增强了9. 87%和48. 87%,MDA含量降低了25. 81%。这些结果表明,外源CO可以通过增强黄瓜种子的养分吸收能力,保护细胞膜系统的稳定性,改善渗透调节能力,提升水解酶活性及其同工酶的表达水平来提高黄瓜种子对干旱胁迫的适应性,促进种子在干旱胁迫下的萌发和生长。  相似文献

以3个不同铃重的棉花基因型为材料,研究了土壤干旱对棉叶碳水化合物代谢的影响.结果表明,干旱胁迫使主茎叶片与不同部位内围果枝叶片可溶性糖含量均有不同程度的增加,而淀粉的积累受到明显抑制.虽然干旱条件下不同基因型棉叶的可溶性糖与淀粉含量的变化趋势一致, 但小铃基因型开花 (7月5日)后的主茎叶可溶性糖含量较中、大铃基因型变化小,淀粉含量则低于大铃基因型.上部内围果枝叶淀粉含量在棉铃发育过程中波动明显大于中、下部果枝,而小铃品种相对稳定.研究结果加深了对干旱胁迫下棉叶碳水化合物时空变化特征的认识.  相似文献

转盐地碱蓬SsDREB基因烟草的抗逆生理机制分析   总被引:1,自引:0,他引:1  
为了研究盐地碱蓬SsDREB基因在转基因烟草中的功能,探究转基因烟草的抗逆生理机制,我们构建了Ss-DREB基因的植物高效表达载体pCAMBIA2301-SsDREB,并通过农杆菌介导法将其导入烟草NC89中,通过Kan抗性、PCR及RT-PCR 筛选鉴定阳性苗;对转基因烟草苗进行高盐、干旱等抗逆性分析,同时测定在不同浓度 NaCl 和PEG6000处理后转基因烟草叶片的净光合速率( Pn )、气孔导度( Gs )、光能转化效率( Fv/Fm )、实际光量子效率(φPSⅡ)以及脯氨酸和可溶性糖的含量。结果表明:通过Kan抗性、PCR及RT-PCR筛选鉴定,最终获得12个株系转基因阳性苗。转基因植株的抗逆性与对照相比明显提高。在不同浓度NaCl和PEG6000处理下,对照和转基因苗的Pn和Gs随着处理浓度的提高均呈现逐渐下降趋势,但在相同浓度处理下转基因苗的Pn和Gs均比对照高;随着NaCl处理浓度的增大,对照和转基因植株的Fv/Fm和φPSⅡ均逐渐降低,但转基因苗的降低幅度比对照小;随着PEG处理浓度的提高,对照和转基因植株的Fv/Fm逐渐降低,但转基因苗的降低程度较对照小,而φPSⅡ下降趋势基本相同。在不同浓度NaCl和PEG处理后,转基因植株和对照植株脯氨酸和可溶性糖含量均随着NaCl处理浓度的增加而提高,但是转基因植株较对照植株增高更快。  相似文献

The objective of this study was to investigate the effect of the different concentrations of the seed film coating salicylic acid on the cold tolerance of cotton seedlings using the sand culture method. The physiological response of cell membrane permeability, protective enzyme activity and osmotic adjustment of cotton seedling were measured before and after at low temperature stress and at normal temperature. The results showed that 1-10 mmol·L-1 of seed film coating with salicylic acid seed coating treatment could effectively alleviate damage to cotton seedlings at low temperature, and the accumulation of the relative electrical conductivity and malondialdehyde were reduced on cotton seedlings. The activity of superoxide dismutase, peroxidase, catalase, and soluble protein, soluble sugar and proline contents increased sharply. These results indicate that film coating with salicylic acid can alleviate the effects of low temperature stress on seedling growth inhibition and improve the cotton seedlings by providing chilling tolerance, and the optimum treatment concentration salicylic acid is 5 mmol·L-1.  相似文献

以棉花幼苗为试材,叶面分别喷施外源调节物质水杨酸(SA)、壳聚糖(CTS)、水杨酸+壳聚糖(SCM)、水杨酸+壳聚糖+硝酸钙(SCCa M)后,在5℃低温下进行处理,分别于0、24 h、48 h及恢复处理24 h时测定相对电导率、MDA含量、叶绿素含量,SOD、POD、CAT的活性和可溶性蛋白、可溶性糖、脯氨酸的含量。结果表明:低温胁迫前用0.7 mmol·L-1水杨酸、0.6 mmol·L-1壳聚糖及复配制剂处理棉花幼苗,其叶片的SOD、POD、CAT活性和叶绿素、可溶性蛋白、可溶性糖、脯氨酸含量与对照相比均有不同程度的提高,相对电导率和MDA含量有所降低;复配制剂处理效果优于水杨酸和壳聚糖单独施用的效果,而水杨酸、壳聚糖、硝酸钙三元复配的效果比水杨酸和壳聚糖两元复配的效果好。表明水杨酸、壳聚糖和硝酸钙在诱导棉花幼苗耐寒性中具有协同效应。  相似文献

[Objective] The aim of this study is to clarify the competition of the absorption and accumulation of nutrients in cotton by the Bolboschoenus planiculmis (F. Schmidt) T. V. Egorova. [Method] In this experiment, the population densities of B. planiculmis were set at 0, 20, 40, 60, 80, 100 plant·m-2. The contents of phosphorus, potassium, nitrate nitrogen, soluble sugar and soluble protein in cotton seedling stage, bud stage and flowering and boll stage were detected to study the effect of population density of B. planiculmis on the nutrient absorption and accumulation of cotton. [Result] At the cotton seedling stage, when the population density of B. planiculmis was 20 plants·m-2, the soluble protein content of cotton leaves increased significiantly (P<0.05) compared with the CK, but there was no significant difference in other indicators. However, the higher population density of B. planiculmis could reduce the soluble protein content of cotton leaves and increase the soluble sugar content of cotton leaves. At the cotton bud stage, when the population density of B. planiculmis was lower than 40 plants·m-2, the phosphorus content in cotton leaves increased significantly. At the cotton boll stage, the population density of all B. planiculmis populations could reduce the content of potassium and soluble sugar in cotton leaves. When the population density of B. planiculmis was 20 plants·m-2, no significant effects were found on cotton yield. But under other population densities, the cotton yield was significantly reduced. [Conclusion] When the population density of B. planiculmis was higher than 20 plants·m-2, significant effects were found on the absorption and accumulation of cotton nutrients and cotton yield.  相似文献

 研究了转基因、非转基因等6个不同棉花品种以及棉叶中可溶性糖、可溶性蛋白、棉酚含量对扶桑绵粉蚧生长发育、繁殖特性的影响,结果表明:扶桑绵粉蚧在各品种棉花上都能危害,且在发育历期、产卵量和存活率等方面均存在明显差异。中棉所41上扶桑绵粉蚧产卵量、寿命、净增殖率R0、内禀增长率rm与周期增长率λ数值最大,种群加倍时间DT值最小,为该虫最适宜棉花品种;而中棉所59上产卵量、寿命、R0、rm、λ值最小,死亡率与DT值最大,为该虫最不适宜品种。嫩叶棉酚含量与2龄若虫历期成极显著正相关,棉酚含量与扶桑绵粉蚧寿命、产卵量显著负相关,表明棉酚对扶桑绵粉蚧生长发育有重要影响,并对该虫繁殖、存活能力有一定抑制作用。  相似文献

2013―2014年采用盆栽试验研究了不同种类及浓度的铜基营养保护剂对棉花生长、产量及防病效果等的影响。结果表明:4种铜基营养保护剂在喷施质量浓度为1 g·L-1和2 g·L-1时,与喷施清水相比,皮棉产量分别增加4.22%~13.38%和21.89%~38.36%;增加了棉株地上部干物质质量,最高达23.96%;降低了棉花病情指数26.75%~40.91%和22.78%~37.78%,但处理间差异不显著。因此,在棉花上喷施铜基营养保护剂既可促进棉花生长,增加棉花产量,又可提高棉花的防病效果。  相似文献

随着棉花基因组学、转录组学等重要学科的快速发展,针对候选基因进行序列多样性分析,开发基于候选基因的分子标记,将能够极大推进棉花分子育种研究。本研究利用高通量的测序技术对耐盐陆地棉品种Miscott 7913-83和盐敏感品种苏棉12盐胁迫前后根、叶总RNA的混样进行了转录组测序,对不同盐处理时期Miscott 7913-83和苏12根和叶分别进行表达谱分析。获得3232条盐胁迫下差异表达基因;基于差异表达基因利用生物信息学手段大规模开发了SSR以及In Del等分子标记;随机选择了部分SSR及In Del引物进行验证,进一步证实了分子标记的准确性。本研究为快速开发陆地棉品种间多态性分子标记提供了高效可行的分析方法,通过开发陆地棉耐盐相关功能标记,以期用于分子标记辅助育种改良陆地棉耐盐性。  相似文献

通过设置0、80、120 mmol·L-1 3种NaCl浓度,研究了3种盐分水平下2-氯-6-(三氯甲基)吡啶浸种对棉种发芽的影响,分析了棉种萌发过程中可溶性蛋白、可溶性糖、抗氧化物酶及丙二醛含量对2-氯-6-(三氯甲基)吡啶浸种的生理响应.结果表明,盐分胁迫显著抑制了棉种的萌发,而2-氯-6-(三氯甲基)吡啶浸种能明显减轻盐分胁迫对种子发芽的抑制.在3个盐分水平(0、80、120 mmol·L-1)下,2-氯-6-(三氯甲基)吡啶浸种处理的发芽率、发芽势及种子活力指数均比清水浸种有显著提高,其增幅分别为11.5%~58.4%、16.8%~65.0%和42.9%~82.5%;同时,2-氯-6-(三氯甲基)吡啶浸种可显著提高棉种SOD、POD、CAT的活性,分别比清水浸种增加了57.2%~282.7%、8.3%~139.3%和6.4%~15.1%,且显著降低了电解质外渗率与MDA含量.在盐分胁迫下,利用4.1×10-3 mmol· L-1的2-氯-6-(三氯甲基)吡啶浸种可提高棉种抗氧化物酶活性,促进其萌发过程中可溶性有机物的转化,显著提高棉种发芽率和发芽势.  相似文献

摘要:【目的】棉花宿生栽培对于雄性不育系的多年生保持和杂交制种,固定杂种优势、加速杂种优势利用和保存种质资源具有重要意义;探讨一年生陆地棉宿生栽培叶片和茎枝N/P/K、可溶性糖含量,为制订栽培措施提供理论依据。【方法】采用随机区组设计,以4个陆地棉品种为供试材料,以一年生为对照,对三年生、二年生宿生陆地棉叶片和茎枝N/P/K、可溶性糖含量进行测定分析。【结果】各处理叶片N/P/K、可溶性糖平均含量分别为3.13%~3.50%、0.49%~0.67%、1.47%~1.78%、4.23%~5.48%,茎枝分别为0.56%~0.82%、0.21%~0.28%、1.14%~1.42%、0.95%~2.15%;不同宿生年限的植株叶片N/P/K含量无显著差异,多数品种一年生植株叶片可溶性糖含量显著高于宿生棉的;高产品种N/P/K含量适中,可溶性糖含量稍低,而低产品种N含量低,P/K、可溶性糖含量较高;随生育进程,叶片全N含量出现由高到低、可溶性糖含量出现由低到高的动态变化趋势,而叶片P/K含量的变化趋势较平稳。【结论】宿生陆地棉与其一年生棉比较,植株叶片N/P/K含量无明显变化,但宿生栽培的中后期应增施N肥,促进营养生长,延缓衰老。  相似文献

[Objective] This study investigates the effects of exogenous proline on the growth, physiological responses, and proline metabolism of cotton (Ekang No.8) seedlings treated with varying boron (B) concentrations. [Method] We conducted a randomized experiment with six treatments using the hydroponic method in a greenhouse at Huazhong Agricultural University. We applied three different concentrations of boron, including 0.02 μmol·L-1 (low-concentration B), 2.5 μmol·L-1 (medium-concentration B), and 100 μmol·L-1 (sufficient-concentration B). The first two are boron-deficient treatments. We also applied an exogenous proline treatment at 20 μmol·L-1(0 μmol·L-1 as control). When any significant difference among the treatments were observed, the related indicator was measured. [Result] The results showed that exogenous proline inhibited the growth of cotton seedlings under sufficient-concentration B treatment while promoting the absorption of B by roots under low-concentration B treatment. The application of exogenous proline to the seedlings under low-concentration B treatment reduced the contents of proline and H2O2 in leaves but increased the accumulation of MDA and H2O2 in roots. The activities of SOD and antioxidant enzymes (APX) in the roots and leaves were dramatically enhanced. Conversely, POD activity reduced significantly and there was no significant change in CAT activity relative to low-concentration B treatment. More importantly, we found that the application of exogenous proline under B deficiency increased the activities of P5CS, P5CR, OAT (synthetase), and PRODH (degrading enzyme) in proline metabolic pathways. [Conclusion] Applying exogenous proline under sufficient-B concentration inhibits growth. The application of proline to seedlings under low-B concentration promotes the absorption of B by roots, increases the activity of APX, and decreases the membrane lipid peroxidation in leaves. A B deficiency leads to proline accumulation in plants. The addition of proline under low-B concentration can reduce the proline content in leaves, which is caused by affecting the critical synthetase and catabolic enzyme activities in the proline metabolism pathway (Glu and Orn pathway). The main reason for this occurrence is the significant increase of proline dehydrogenase (PRODH) activity.  相似文献

[Objective] The purpose of this study was to expand upland cotton genotypes suitable for regeneration via somatic embryogenesis, so as to provide excellent receptor materials for the genetic transformation of cotton. [Method] Six different hormone combinations were studied to establish a somatic embryo regeneration system for hypocotyl explants of Xinluzao 45, a widely planted variety in Xinjiang Province. [Result] Faint yellow callus was induced on a medium consisting of hormone combinations C1 (0.1 mg·L-1 acetic acid dichlorobenzene oxide [2, 4-D] + 0.1 mg·L-1 kinetin [KT]) or C5 (0.1 mg·L-1 2, 4-D + 0.5 mg·L-1 indo-3-butyric acid + 0.1 mg·L-1 KT). The generated callus was able to successfully induce embryogenic callus on hormone-free medium containing doubled KNO3. The embryogenic callus then developed into somatic embryos and regenerated plants. Maximum rates of callus induction and regeneration were 92.2% and 40.3%, respectively. Although no significant difference was observed in the rate of callus induction of Xinluzao 45 hypocotyls between hormone combinations C1 and C5, the incidence of induced embryonic callus was higher on C1 than on C5. [Conclusion] The somatic embryo regeneration system developed in this study provides technical support that will facilitate Agrobacterium-mediated transformation of upland cotton Xinluzao 45.  相似文献
