An indirect fluorescent antibody test was adapted for measuring serum anti-Treponema hyodysenteriae antibodies with a fluorometer. The immunofluorescence was recorded as fluorescent signal units. Cultures of T. hyodysenteriae and Treponema innocens were used as antigen. There was a significant (P less than 0.01) correlation between the immunofluorescence recorded with the fluorometer and that evaluated visually with a microscope. The swine exposed orally to swine dysentery infective inoculum and subsequently hyperimmunized by the intravenous inoculation of live cultures of T. hyodysenteriae had the highest average fluorescent signal unit, which was 104.5. There was a significant (P less than 0.01) correlation between the level of anti-T. hyodysenteriae antibody and the interval length between the last day of diarrhea and the day of bleeding. However, in measuring fluorescent signal units in serum from swine infected with nonpathogenic large spirochetes, (T. innocens), there was also a significant (P less than 0.01) correlation between T. hyodysenteriae and T. innocens as antigen. The coefficient of variation of the average fluorescent signal unit for a highly positive serum and a highly negative serum between 16 runs of assays were 5.7% and 19% respectively; the coefficient of variation of the average fluorescent signal unit for duplicate samples on 358 serum samples tested was 5.8%.  相似文献

Swine dysentery was induced in pigs and in ligated colonic segments by inoculation of pure cultures of, or colonic contents containing, Treponema hyodysenteriae. The mildest changes, best seen in ligated segments 48 or 72 hours after inoculation, were congestion and leucocytic margination in mucosal capillaries and depletion of mucigen from goblet cells lining the base of the crypts of Lieberkühn. Superficial mucosal necrosis and crypt cell hyperplasia were later changes. Perfusion studies with India ink did not demonstrate occlusive mucosal ischemia in acute swine dysentery. Mucosa with lesions of swine dysentery contained at least 10(5) colony forming units of T. hyodysenteriae per gram. Mucosa without lesions had 10(5) or fewer T. hyodysenteriae per gram. Segments with acute swine dysentery were distended with clear mucoid fluid with electrolyte composition indicative of net colonic secretion. No increase in the concentration of volatile fatty acids was detected in content from intact colons or colonic segments with lesions of acute swine dysentery.  相似文献

Broth cultures of Treponema hyodysenteriae and colonic content from pigs with swine dysentery were tested for cytotoxicity in cell cultures, erythrocyte suspensions and in ligated segments of pig colon. Live cells of T. hyodysenteriae attached to the surface of cells in all cultures tested but did not penetrate them nor cause morphologic change detectable by light microscopy. Only live T. hyodysenteriae caused erythrolysis. Broth cultures or colonic content sterilized by filtration or by disruption with ultrasound had no visible effect on the cell cultures, erythrocyte suspensions or the mucosa of ligated colonic segments.  相似文献

Transmission experiments were carried out in gnotobiotic pigs to determine whether lesions typical of swine dysentery could be produced by oral inoculation of Treponema hyodysenteriae in combination with Bacteroides vulgatus or Fusobacterium necrophorum, or both. Each of the organisms had been isolated from swine with early lesions of the disease. Lesions were not found in 6 pigs inoculated with T hyodysenteriae alone, in 4 pigs given F necrophorum and T hyodysenteriae, or in 4 pigs given B vulgatus and F necrophorum. Lesions typical of swine dysentery developed in 8 pigs given B vulgatus, F necrophorum, and T hyodysenteriae as well as in 3 of 4 pigs given B vulgatus and T hyodysenteriae. In both of these groups, the inoculated bacteria were recovered from the colon, and T hyodysenteriae was demonstrated in the colonic crypts, epithelium, and lamina propria. The pathogenicity of the T hyodysenteriae was shown by the development of characteristic signs and lesions of swine dysentery in 12 of 14 naturally farrowed pigs inoculated with T hyodysenteriae alone.  相似文献

Swine-ligated loops were used to demonstrate passive protection against swine dysentery. Loops inoculated with immune sera containing complement and with homologous Treponema hyodysenteriae were normal at necropsy. Loops inoculated with heat-inactivated immune sera and heterologous T hyodysenteriae were not protected. Loops inoculated with heat-inactivated immune sera and homologous T hyodysenteriae were partially protected. Positive control loops inoculated with isolate B204 (88%) or B234 (44%) T hyodysenteriae and normal sera developed lesions typical of swine dysentery, whereas negative control loops inoculated with nonexposed sera only were normal.  相似文献

Isolation of Treponema hyodysenteriae from sources other than swine   总被引:3,自引:0,他引:3  
Fecal samples were collected from animals and environments on 3 swine farms and cultured for Treponema hyodysenteriae. Each farm was a farrow-to-finish operation and, at the time of sampling, swine dysentery was enzootic among 8- to 22-week-old pigs. Pathogenic T hyodysenteriae was isolated from pigs on all 3 farms. On farm A, nonpathogenic T hyodysenteriae was isolated from a sample of lagoon water. On farm B, pathogenic T hyodysenteriae was isolated from a waste-holding pit. On farm C, a dog was observed to be eating feces of pigs that had swine dysentery. The dog was diarrheic and a fecal sample yielded a pathogenic isolant of T hyodysenteriae. Further isolation attempts were unsuccessful after the dog was removed from the infected premises. Isolation of pathogenic and nonpathogenic organisms from waste-holding systems emphasizes the need for cultural techniques in detecting pathogenic T hyodysenteriae.  相似文献

An attenuated strain of Treponema hyodysenteriae was used to immunise 18 pigs in three experiments. Live attenuated spirochaetes were dosed orally and injected intra-peritoneally, and killed spirochaetes were injected intramuscularly with adjuvant. The vaccinated pigs, which developed high serum agglutination titres against T hyodysenteriae, and 18 unvaccinated litter-mates were repeatedly challenged with virulent T hyodysenteriae. Nine vaccinated pigs and 16 control pigs developed typical swine dysentery.  相似文献

Using an indirect fluorescent antibody test, immunofluorescence of large spirochetes was observed with serum from swine that had recovered from swine dysentery. The spirochetes were obtained from scrapings of the colonic mucosa on the first day of diarrhea which was the time when the spirochete population was observed to be the highest. Of 29 exposed nonmedicated swine which developed and recovered from a diarrhea characteristic of swine dysentery 27 had antispirochete serum titers which ranged from 1:2 to 1:16. None of the 50 nonexposes swine developed a titer. Of 19 swine with a serum titer and reexposed with infective swine dysentery inoculum, 18 did not develop a diarrhea and were presumed to be immune. Considering these findings it is possible that this test could be used to detect antispirochete antibody in unknown swine serum.  相似文献

When 47 pigs were dosed orally with cultures of Treponema hyodysenteriae, 44 (94 per cent) developed swine dysentery. Of those which recovered and were rechallenged, nine of 21 (43 per cent) showed clinical signs, as did one of 10 (10 per cent) challenged on a third occasion. Clinical disease was associated with development of specific IgG, IgA and IgM antibodies in serum and the local production of IgA in gut mucosal tissues. The appearance of antibody was not directly related to protection but rather indicated either prolonged exposure (in the case of serum IgG) or recent exposure to T hyodysenteriae (for secretory IgA). Infection also resulted in the appearance of IgG and IgA memory cells in gut-associated lymphoid tissue. However, these studies indicated that humoral immunity alone is not responsible for the onset of a protective response to T hyodysenteriae in the colon.  相似文献

Swine dysentery did not recur during a nine week period after withdrawal of medication in swine fed ronidazole at a level of 60 parts per million of feed for ten weeks or fed either carbadox at 55 ppm or lincomycin at 110 ppm of feed for six weeks. During this period swine dysentery was neither transmitted to accompanying sentinels after the withdrawal of the above medication or was Treponema hyodysenteriae isolated and cultured or observed in stained smears from rectal swabs and feces or from colonic scrapings at necropsy. Beginning three weeks after the withdrawal of medication, all swine were fed sodium arsanilate at a concentration of 220 ppm of feed for three weeks in an attempt to excite the carrier of swine dysentery into developing a swine dysentery diarrhea. A swine dysentery diarrhea did recur during the feeding of sodium arsanilate in swine previously fed ronidazole at a level of 60 ppm of feed for only six weeks. It was concluded: that swine dysentery was probably eliminated with the feeding of ronidazole for the longer duration and with the feeding of carbadox and lincomycin and that sodium arsanilate was of value in identifying the carrier state.  相似文献

When pure cultures of Treponema hyodysenteriae were orally inoculated into pigs, severe disease characteristic of swine dysentery developed. Less severe lesions were produced by oral inoculation of infective minced colon. Noninoculated pigs were used as controls. Inoculations of surgically isolated porcine colonic loops with either pure cultures or infective minced colon produced lesions only in the injected loop; the adjacent noninjected colon remained normal. Pigs and other experimental animals, including rabbits, guinea pigs, and mice, inoculated with washed cultures by various parenteral routes remained normal.  相似文献

Net electrolyte and water transport and unidirectional Na+ fluxes were examined in ligated colonic loops of clinically normal pigs and in pigs with swine dysentery (etiologic agent Treponema hyodysenteriae) in the presence or absence of theophylline. In normal pigs, theophylline abolished net Na+ absorption via a reduction in the lumen-to-blood flux, decreased Cl- absorption, and increased HCO3- accumulation in the lumen. In infected pigs, all net ion transport was abolished, with the addition of theophylline producing little effect. The absence of net Na+ absorption in infected pigs was also the result of a decreased lumen-to-blood flux. Seemingly, colonic malabsorption may be the primary transport alteration in swine dysentery. Concentrations of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) were measured in samples of colonic mucosa from normal and infected pigs after in vitro exposure to a Ringer's solution containing 0 or 20 mM theophylline. Basal values of cAMP or cGMP did not increase in infected colonic mucosa. There was a diminished capacity of the infected mucosa to respond to theophylline. Alterations in ion transport in conjunction with measurements of cAMP and cGMP indicated that the pathogenic mechanism(s) in swine dysentery were not similar to those of Salmonella, Shigella, Vibrio cholerae, or Escherichia coli diarrhea.  相似文献

Twenty-six specific-pathogen-free pigs were fed pure cultures of Treponema hyodysenteriae. Five untreated pigs were controls. Distribution of this large spirochete in pigs with swine dysentery was shown by the indirect fluorescent antibody technique. Findings by this method were compared with those from dark-field examination of colonic mucosal scrapings and from tissue sections. The cultures caused mucohemorrhagic colitis which by 10 days after inoculation was indistinguishable from the colitis of swine dysentery. Control pigs remained normal. Pigs killed when spirochetes were first seen in the feces had normal colonic mucosa with only a few spirochetes. At the first sign of diarrhea, however, the colonic mucosa was thicker than normal and had many spirochetes. T. hyodysenteriae was confined to regions of hypertrophy and exudation of the large intestine mucosa throughout the course of disease.  相似文献

Sodium arsanilate was fed to nondiarrhetic swine, previously exposed to and treated for swine dysentery, for the purpose of inducing them into developing a swine dysentery diarrhea. From 40 to 100% of these swine in each pen had previously had a swine dysentery diarrhea. The isolate of Treponema hyodysenteriae in the diced colon which was used to expose the swine was resistant to sodium arsanilate. After an interim of no treatment for swine dysentery, sodium arsanilate was fed at a level of 220 parts per million for 21 days. Of the 14 pens containing swine fed sodium arsanilate, ten pens had one or more swine that developed a swine dysentery diarrhea while being fed sodium arsanilate. This was significantly (P less than 0.05) greater than the three pens that each had one pig that developed a swine dysentery diarrhea of 13 pens containing similar swine not fed sodium arsanilate during a comparable period. In the 14 pens containing swine fed sodium arsanilate, 14 swine were the first to develop a swine dysentery diarrhea since in four pens, two swine in each pen developed diarrhea within 24 hours of each other. This also was significantly (P less than 0.01) greater than the three swine in the ten pens not fed sodium arsanilate. From these results, it was theorized that sodium arsanilate excited the nondiarrhetic carrier into developing a swine dysentery diarrhea and that this phenomenon may have potential in identifying the carrier state.  相似文献

An experiment was conducted to study the effect of diets with contrasting fermentability in the large intestine on experimental infections with Brachyspira hyodysenteriae, the causative agent of swine dysentery, and the whip worm, Trichuris suis, in pigs. Two diets with organically grown ingredients were composed. Both diets were based on triticale and barley and supplemented with either rape seed cake (Diet 1) or dried chicory root and sweet lupins (Diet 2). The study had a three-factorial design, with eight groups of pigs receiving Diet 1 or Diet 2, +/-B. hyodysenteriae, and +/-T. suis. Pigs fed Diet 2 and challenged with B. hyodysenteriae did not develop swine dysentery and B. hyodysenteriae was not demonstrated in any of the pigs during the study. In contrast, 94% of the B. hyodysenteriae challenged pigs fed Diet 1 showed clinical symptoms of swine dysentery and all the pigs were shedding B. hyodysenteriae in faeces at some points in time during the experiment. The number of T. suis was lower in pigs fed Diet 2 compared to pigs fed Diet 1, but the differences were not significant. Pigs on Diet 1 and challenged with both pathogens showed clinical symptoms of SD for a longer period than pigs inoculated with B. hyodysenteriae only. The study showed that diets supplemented with highly fermentable carbohydrates from dried chicory roots and sweet lupins can protect pigs against developing swine dysentery, but do not have any significant influence on T. suis.  相似文献

Pure cultures of Treponema hyodysenteriae given orally to conventional pigs resulted in the development of swine dysentery, whereas identical cultures given to gnotobiotic pigs did not produce the disease. Oral inoculation of gnotobiotic pigs with Vibrio coli and/or a peptostreptococcus in addition to T. hyodysenteriae did not result in dysentery. Neutralization of gastric secretions with NaHCO3 immediately prior to inoculation with T. hyodysenteriae increased the period during which treponemes were evident in the feces, as did the inoculation of this organism via the intracecal route. None of the gnotobiotic pigs with a persistent fecal Treponema population developed signs of dysentery. Factors other than those investigated in this work must play a part in the etiology of swine dysentery.  相似文献

A direct fluorescent antibody test was developed for the identification of large spirochetes which are considered to be the cause of swine dysentery. Sera from swine which had recovered from swine dysentery and had been hyperimmunized by the intravenous and intraperitoneal injection of filtered spirochetes were used for conjugation with fluorescein isothiocyanate. A bright greenish fluorescence of large spirochetes was observed with the conjugated serum from hyperimmunized pig No. 1 when diluted 1:8 and hyperimmunized pig No. 2 when diluted 1:2. Pig No. 1 had developed a serum titer of 1:64 using the indirect fluorescent antibody test for large spirochetes. The conjugated serum from the three swine which had recovered from swine dysentery fluoresced spirochetes only when undiluted. The conjugated serum from the two swine treated while having a hemorrhagic diarrhea did not fluoresce spirochetes. No immunofluorescence of Vibrio spp. was observed.  相似文献

This study tried to evaluate the effect of highly fermentable fiber on the incidence and severity of swine dysentery (SD) after experimental oral infection with pure cultures of Brachyspira (B.) hyodysenteriae. Forty eight growing pigs were allocated to two groups and treated until slaughter as follows: Group 1 (n = 24): infected with B. hyodysenteriae and fed with a food containing 9.6% highly fermentable neutral detergent fiber. Group 2 (n = 24): infected with B. hyodysenteriae and fed with a food containing 6.1% low fermentable neutral detergent fiber. Pigs of each group were intragastrically inoculated on each of three consecutive days with pure culture of 1.8 x 10(10) B. hyodysenteriae. All pigs were monitored daily until slaughter. Faecal shedding of B. hyodysenteriae by polymerase chain reaction, antibody response by IFA, clinical signs, growth performance and extents of gross and microscopical lesions specific for swine dysentery were determined. Faecal shedding of B. hyodysenteriae and antibodies specific for B. hyodysenteriae were detected at day 30 post infectionem. Significant (p < 0.05) milder clinical signs typical for swine dysentery were detected in group 1, fed with 9.6% high fermentable fiber compared to group two fed with a food containing 6.1% low fermentable neutral detergent fiber. Daily weight gain differed significantly (p < 0.05) between the groups (group one 780 g vs. group two 760 g). Food conversion efficiency showed in group one a significant (p < 0.05) better (3.28) result than in group two (3.38). Feed consumption presented significantly (p < 0.001) better results in group one compared to group two (2.38 kg vs. 2.25 kg). From our experimental findings we conclude that in production units suffering of swine dysentery high levels of highly fermentable fiber in diet may increase health and performance.  相似文献

The diarrhea of swine dysentery receded in swine treated with 60 or 45 mg of tiamulin/L of drinking water (60 or 45 ppm). However, within 2 to 10 days (average 4.1 days) after drug withdrawal, diarrhea recurred. Tiamulin (22.5 mg/L in drinking water) did not markedly reduce the diarrhea during medication, and tylosin (66 mg/L in the drinking water) was not effective. In swine treated with 120 mg of dimetridazole/L of drinking water, there was no recurrence of diarrhea. After the recurrence of diarrhea in swine, repeated medication with tiamulin in drinking water reduced the severity of diarrhea and prevented deaths. After 1 to 3 retreatments, swine were immune to exposure with swine dysentery inoculum, and there was a significant (P less than 0.05) increase in their serum anti-Treponema hyodysenteriae antibodies. Seemingly, drug withdrawal permitted the occurrence and recurrence of diarrhea that was necessary to stimulate immunity.  相似文献
