The effect of naturally acquired bacterial infection of the bovine udder on the activity of 5'-thyroxine monodeiodinase (5'-MD), and on the concentrations of the pro-inflammatory cytokines interleukin (IL)-6 and tumour necrosis factor (TNF)-alpha in milk, from healthy (control) and inflamed quarters, was determined. The diagnostic procedure included history and clinical examination of the udder, macroscopic evaluation of secretions, the Californian Mastitis Test, determination of somatic cell counts and bacteriological examination of milk. It has been found that the milk triiodothyronine (T3) content and the 5'-MD activity from inflamed quarters were decreased when compared with controls. The decrease in the milk T3 from subclinical mastitic quarters was manifested when somatic cell counts were > 10(6) ml(-1). TNF-alpha was on average 2-fold higher in infected milk, and the concentration of IL-6 was unchanged. These results suggest that the decreased T3 content in mammary secretions during naturally occurring mastitis is associated with the severity of inflammation, increased TNF-alpha concentration and impaired enzymatic activity of 5'-MD.  相似文献

The potential of near-infrared spectroscopy (NIR) in the region from 1,100 to 2,500 nm to measure somatic cell count (SCC) content of cow's milk was investigated. A total of 196 milk samples from seven Holstein cows were collected for 28, consecutive days, starting from 7th d after calving, and analyzed for fat, protein, lactose, and SCC. Three of the cows were healthy, and the remainder had periods of mastitis during the experiment. Near-infrared transflectance milk spectra were obtained using an InfraAlyzer 500 spectrophotometer. The calibration for logSCC was performed using partial least square (PLS) regression and different spectral data pretreatment. The best accuracy of determination was found for an equation that was obtained using smoothed absorbance data and 10 PLS factors. The standard error of calibration was 0.361, the calibration coefficient of multiple correlation was 0.868, the standard error of prediction for independent validation set of samples was 0.382, the correlation coefficient was 0.854, and the coefficient of variation was 7.63%. The accuracy of logSCC determination by NIR spectroscopy would allow health screening of cows and differentiation between healthy and mastitic milk samples. It has been found that SCC determination by NIR milk spectra is based on the related changes in milk composition. The most significant factors that simultaneously influenced milk spectra with the elevation of SCC were alteration of milk proteins and changes in ionic concentration of milk.  相似文献

Antibody levels against Ostertagia ostertagi in the milk are a promising parameter to identify dairy cows or herds with production losses due to gastrointestinal nematodes. However, milk antibody levels can be influenced by udder-related factors. In this study, the effect of an experimentally induced mastitis on the test results (ODR) of a milk O. ostertagi ELISA was investigated. Twenty-five cows that were naturally infected with gastrointestinal nematodes, were inoculated in their left udder quarters with Escherichia coli P4:O32 and quarter milk samples were collected at several intervals from 24 h before until 144 h after experimental infection. The effect of the contribution of milk from one or more infected quarters on the bulk tank milk ODR was estimated, based on a titration experiment. The mean O. ostertagi antibody levels of the infected udder quarters were significantly (P < 0.001) higher than those of the uninfected udder quarters at each sampling time post-infection. The largest difference was observed at 24 h post-infection with a mean difference of 0.251 ODR (95%CI: 0.172; 0.330). There was also a significant increase (P < 0.001) in total IgG levels with the largest difference being observed at 24 h post-infection. Highly significant (P < 0.005) correlation coefficients were observed between O. ostertagi ODR, total IgG ODR, Na+ and Cl- ion concentration and log transformed somatic cell counts at 24 h post-infection. The results demonstrate that an acute mastitis causes a flow of specific and non-specific antibodies from serum to milk with a subsequent increase in the O. ostertagi ODR values. The effect of the contribution of milk from infected quarters on the bulk tank milk O. ostertagi ODR was estimated to be minor if the relative number of infected quarters is small (< 3%).  相似文献

Two hundred and fifty-two teats from the udder quarters of 68 dairy cows were studied by bacteriological examination of quarter milk samples and microscopic examination of the teats. Of 252 quarters, 169 teats showed inflammatory changes, while 83 were of normal appearance. The inflammatory changes consisted of either neutrophil infiltrations, mononuclear cell infiltrations and fibrosis, or a combination of these changes. The reaction was restricted to the teat cisterns and/or Fürstenberg's rosettes, but very rarely also in the teat canal. In 136 of 169 cases, the inflammatory reaction was associated with bacteria in the milk, while in 33 cases no bacteria could be recovered from milk, in spite of inflammatory changes in the teat structures. Only 36 of 83 of normal teats were bacteriologically positive.  相似文献

The purpose of the study was to estimate the extent to which one quarter could be inflamed relative to the other three quarters of a cow, given that knowledge only of the composite milk somatic cell count (SCC) was available. An algebraic relationship, which incorporated the parameters of composite milk SCC, production loss associated with composite milk SSC, SCC of milk from an inflamed quarter, and SCC from three non-inflamed quarters, was used to derive hypothetical estimates of the SCC in one inflamed quarter. A simple case was considered in which one quarter was mastitic, the SCC in milk of the noninflamed quarters was equal, and there was no production compensation by the noninflamed quarters. Previously published estimates were used for production loss associated with composite milk SCC. For moderate composite milk SCC (300,000-500,000 cells/mL) and low-to-moderate SCC in milk of noninflamed quarters (100,000 cells/mL), the SCC of milk from one inflamed quarter was predicted to be very high, ranging from about 1.26 million (log10 SCC = 6.1) to 6.3 million (log10 SCC = 6.8) cells/mL, and compatible with signs of clinical mastitis. These results suggest that in order to screen cows with clinical mastitis in only one quarter, composite milk SCC should be considerably lower than values presented previously by other investigators.  相似文献

应用超高压液相色谱-四级杆飞行时间质谱法(UPLC-QTOF)建立了筛查牛奶中55种药物的方法。以乙腈提取样品中的药物,提取液经过HLB固相萃取柱净化浓缩后检测。在不同添加浓度下获得了精确分子离子质量,质量偏差的绝对值低于9.9×10-6。55种药物的检出限为2~10μg/kg,在5~200 ng/mL范围内线性关系良好,相关系数r≥0.98。在牛奶中添加20~100μg/kg的药物,回收率在22.9%~114.3%之间。本方法操作简单、高效、杂质干扰少,适用于牛奶中55种药物的检测需要。  相似文献

A rapid method for the determination of urea in milk was evaluated. The method employed was a urease/glutamate dehydrogenase procedure where the decrease in absorbante of NADH is monitored spectrophotometrically at 340 nm. The repeatability expressed as a coefficient of variation, was 1.4 % and 0.8 % for whole milk and skim milk, respectively. The accuracy of the method for urea determinations in whole milk was satisfactory (coefficient of variation, 2.6 %). Adding the preservative bronopol (0.02%) to the milk or freezing of the milk samples for one week did not alter urea levels significantly (P > 0.05). The storage of preserved whole milk in a refrigerator for 14 days, however, increased milk urea levels measured by 3.1 % (P < 0.001).It was concluded that the method evaluated is simple, rapid (40–50 samples/h) and yields reliable results also when whole milk is used. Since the determination of milk urea may be useful in assessing the feeding of energy and protein, the method is suitable for integration in the existing milk recording system in Sweden.  相似文献

本试验旨在研究利用超高效液相色谱测定原料乳中羟脯氨酸(Hyp)含量的方法。在原料乳中羟脯氨酸测定过程中各种条件对测定结果的影响,并获得了较好的测定条件。结果发现本方法的回收率在94.7%~98.6%之间,在1.0~16.0 μg/mL标准曲线的回归方程为y=3286.5x+87.943,R2=0.9999,相对标准偏差(RSD)小于2.36%;最低检出限为1 mg/kg。该方法操作简便、色谱分离速度快、准确度高、精密度好,可用于原料乳中羟脯氨酸的定量测定,从而判定是否在原料乳中添加动物水解蛋白。  相似文献

The aim of this work was to study the relationship between the reactions obtained by indirect methods of testing milk samples and the cell content, determined by direct cell counting after Prescott & Breed (1910). The indirect tests used were the California Mastitis Test (CMT), Whiteside Test (WsT), Brabant Mastitis Reaction (BMR), and the Catalase Test (CT).The results of the investigations are summarized below.Samples with cell values lying within fixed limits do not neces¬sarily conform to particular indirect test reactions.All four indirect tests have wide ranges of variation of cell level within a given reaction with considerable overlapping between reac¬tions.The standard deviation for cell levels within a given reaction is large, although this can be partly explained by the fact that the cell values are estimated rather than real. Correction for this produces only an insignificant decrease, however.The coefficient of correlation is a little higher for the correlation CMT—CC than for the other indirect tests and CC.When sample groups with different mean cell levels (calculated from the whole material) are compared, the variation ranges (dif¬ference between the largest and the smallest observation) and standard deviations arc generally the same for the different CMT reactions. However, the mean cell values, for different reactions lie at higher levels when the mean cell content of the whole material is higher. Similarly they are lower when the mean of the whole material is lower. This is, however, not true for 0 reactions.The mean cell values for CMT 4- and for 4- 4- reactions varied considerably in different herds.It was not considered acceptable, for any of the four indirect tests, to substitute reactions by average cell values, either to determine the mean cell value or to decide whether a sample is inacceptable, if this means that the sample has a cell content above a certain value.Negative and 4- reactions for CMT and WsT could possibly be replaced by a mean cell level, since the range of variation is not so wide here and the majority of samples giving these reactions contain less than 3xl05 cells per ml.The possibility exists of combining indirect tests and direct cell counting to examine milk samples, so that samples with low reactions could be passed, while those with higher reactions would be examined by cell counting.The indirect tests differentiate milk at different cell levels. The possibility of using a particular indirect test depends on the cell level to be examined and on the degree of accuracy required.  相似文献

The rapid reaction of the diphenylamine agent with DNA was used for the determination of the counts of somatic cells in cow's milk, using the DNA filter method. The method is based on the filtration of a warmed (65-70 degrees C) mixture of milk with Triton X-100 through the Synpor nitrocellulose membrane filter, pore size 2 to 5 microns, and subsequent DNA determination of the collected somatic cells by the colour reaction of diphenylamine. A 2ml quantity of distilled water and 4 ml of diphenylamine reagent were added to the membrane filters with somatic cells. The mixture is warmed in water bath at 90 to 100 degrees C for 20 min., then it is cooled, centrifuged (3500 X g, 15 min.), and the optical density is measured at 595 nm. The relation 8 micrograms = 1 million cells was used for the conversion of DNA content to the counts of cells. The average variation coefficient of the determination was 5.9% and the coefficient of correlation between the diphenylamine DNA filter method and the direct microscopy of the somatic cells on membrane filters was r = 0.997. Using the diphenylamine DNA filter method, the counts of somatic cells can also be determined from milk samples stored in frozen condition or from the filters with collected cells kept at the temperature of 4 degrees C (10 days) or 25 degrees C (3 days). Milk stabilized with formaldehyde can also be used for the determination if stored at 4 degrees C.  相似文献

The aim of the present study was to investigate whether milk composition and milk yield are changed in relation to a moderate increase in milk somatic cell count (SCC) in separate udder quarters. During a period of 13 weeks, 4158 bulk quarter milk samples from 68 cows were collected and analysed for milk SCC and milk composition. The sampling was done twice weekly. The cows were in different stages of lactation and in different lactation numbers. For calculations, three groups of cows were formed according to their SCC value. Group 1 cows, where all quarters had an SCC <100,000 cells/ml at all sampling occasions, were considered to be non-affected. Group 2 cows had one udder quarter with an increased SCC >100,000 cells/ml and 1.5-fold higher than the opposite quarter at one sampling occasion. For group 3 cows, the increase in SCC remained for several consecutive sampling occasions. Data from group 1 cows revealed that front and rear quarters were similar when compared with each other. For group 3 cows, the lactose content in milk decreased significantly, simultaneously with the increase in SCC and remained decreased for two sampling occasions after the initial increase in SCC. It was concluded that deviations in lactose content within front and rear quarters, respectively, may be a useful tool for detection of moderately increased SCC in separate udder quarters.  相似文献

In the present study, the prevalence of S. aureus in mammary gland quarters of dairy cows in Switzerland was estimated and a risk factor analysis was carried out. Dairy cows were selected by one-step-cluster sampling with stratification by herd size. Forty-seven of 50 randomly chosen farms participated in the study, resulting in 603 cows and 2388 quarter samples. Milk samples were collected in all herds on two occasions two weeks apart. In 6% of cows (95% CI: 2.7-9.3%) at least one milk sample was positive for S. aureus and from 2% (0.8-3.2%) of all quarters, S. aureus was cultured at least once. In four quarters a latent S. aureus infection (agent detected and somatic cell count (SCC) <100,000cell/ml) was diagnosed. Multivariable hierarchic logistical regression analysis yielded five significant risk factors for observing S. aureus in a milk sample: high SCC, a S. aureus-positive neighbouring quarter, a palpable induration in the quarter, and a wound, scar tissue or crush injury affecting the teat. The type of housing (P=0.1596) was also a factor that remained in the model. The mentioned risk factors must be considered during the evaluation of herds with S. aureus problems. The occurrence of latent S. aureus infections emphasises that not only quarters with a high SCC but all quarters of all cows must be cultured for control measures to be effective.  相似文献

本文通过研究温度对紫外分光光度法测定生乳中亚硝酸盐含量的影响,建立采用紫外分光光度法测定生乳中亚硝酸盐含量的方法。结果表明,此方法测定生乳中亚硝酸盐含量可以获得良好的线性关系,相关系数R2>0.99991,其拟合度较高,在加热温度95 ℃的条件下,3 个(0.1 mg/L、0.2 mg/L、0.3 mg/L)不同水平的加标平均回收率为91.00%、90.50%、92.17%,重复性试验RSD<5%。该方法操作简便,分析快速,亚硝酸盐提取速度快,适合生乳中亚硝酸盐含量的检测。  相似文献

为探讨湖北地区荷斯坦奶牛产奶量、乳成分的相关性,本研究选择2016年湖北省3个牧场的DHI数据,运用SPSS17.0中的双变量相关分析和偏相关分析,分析产奶量与乳成分的相互关系。结果表明:3个牧场产奶量与乳脂率、乳蛋白率、体细胞数、乳尿素氮呈负相关(P0.01);乳脂率与乳蛋白率、体细胞数、乳尿素氮均呈正相关(P0.01);乳蛋白率与体细胞数呈正相关(P0.01),与乳尿素氮呈负相关(P0.01);体细胞数与乳尿素氮均呈负相关(P0.01);产奶量与体细胞数简单相关系数-0.003(P0.05),偏相关系数-0.054(P0.01);体细胞数与乳尿素氮含量简单相关系数0.003(P0.05),偏相关系数-0.134(P0.01)。在产奶量与乳成分间关系研究中简单相关分析与偏相关分析结果不一致,在多个变量相关性研究中,偏相关分析更能真实反映2个变量间本质联系。  相似文献

The objectives of this study were to determine 1) the effect of intramammary infection on calf weaning weight, milk somatic cell count, and milk composition, and 2) the effect of parity on percentages of infected cows, infected quarters, and blind quarters. The number of infected quarters, milk somatic cell counts, milk components, and intramammary infection were studied at weaning in 164 beef cows. The percentage of infected cows ranged from 61.9% at first parity to 66.7% at fifth to ninth parities. Cows with three or four infected quarters had higher (P < .01) milk somatic cell counts than cows with zero, one, or two infected quarters. Among bacterial isolates, Staphylococcus aureus-infected quarters had the highest (P < .01) milk somatic cell count. Percentages of butterfat and lactose were lower (P < .01) in milk from infected quarters than from uninfected quarters. Infections by S. aureus and coagulase-negative staphylococci were the most common and accounted for 67 to 78% of the infections. Percentages of infected quarters and infections caused by S. aureus increased with parity (P < .01). Intramammary infections did not affect (P > .10) calf weaning weight. In conclusion, intramammary infection had no effect on calf weaning weight but increased milk somatic cell count and decreased the percentage of protein, lactose, solids-not-fat, and butterfat. The number of infected and blind mammary quarters increased with parity.  相似文献

Quarter foremilk samples were taken at 2–3 weekly intervals for several years in a experimental herd comprising about 45 cows. The samples were submitted to bacteriological analysis and somatic cell counting. The most prevalent quarter infections from 1982 to 1988 were by coagulase-negative staphylococci (15–20% of all the quarters sampled). Most of these (75.6%) persisted until drying-off Dry cow therapy eliminated 86.5% of these infections. Comparison of udder quarters within cows, involving 775 samples from pairs of non-infected quarters and quarters infected by coagulase-negative staphylococci, yielded geometric means of somatic cell counts of 210 000 and 420 000 cells/ml, respectively. The correlation (r=0.87) between the herd bulk milk somatic cell count (SCC) and its estimation from the quarter milk somatic cell count performed on the same day allowed us to evaluate the contribution of the different categories of quarters, according to their infection status, to the herd bulk milk SCC. Quarters infected by a major pathogen (8.5% of samples) gave rise to 46.6% of the total number of cells, while quarters infected by coagulase-negative staphylococci (17.8% of samples) gave rise to 18.1%. Although coagulase-negative staphylococci represented only a secondary source of somatic cells as compared to major pathogens, they were not a negligible source considering the threshold of 300 000 somatic cells advocated for herd milk of good quality.  相似文献

The major pathogens causing mastitis were evaluated by multiplex-polymerase chain reaction (M-PCR) with self-designed primers in four quarters of the first, third, and fifth parities in industrial, semi-industrial, and traditional dairy cattle farms in Iran. With the incidence of infection in the quarters by Staphylococcus aureus and Streptococcus agalactiae, the mean log somatic cell count (log SCC) increased from 5.06 to 5.77. The smallest changes occurred with Escherichia coli. Contagious pathogens, when compared with environmental pathogens, were more prevalent and common and created more profound quantitative and qualitative changes in SCC profiles. The second part of the study surveyed the diversity of contaminating pathogens and their effect on quantitative and qualitative profiles of somatic cells. M-PCR was used to determine the absence (M-PCR(-)) and presence of one (M-PCR(+1)), two (M-PCR(+2)), and three (M-PCR(+3)) major pathogens in raw milk samples. Quarter log SCC increased from 5.06 (for M-PCR(-1)) to 5.5 (for M-PCR(+1)), 5.7 (for M-PCR(+2)), and 6 (for M-PCR(+3)). AIM: To compare clinical and bacteriological cure rates of clinical mastitis following treatment with intramammary preparations containing either lincomycin and neomycin or penicillin and dihydrostreptomycin. METHODS: Cases of clinical mastitis were sourced from four seasonal-calving dairy herds in the central Waikato region of New Zealand during the first 120 days of lactation. Affected quarters were infused three times at 12 h intervals with either 333 mg lincomycin plus 100 mg neomycin (lin/neo; 197 glands),or 1,000 mg penicillin plus 500 mg dihydrostreptomycin (pen/DHS; 207 glands). Milk samples were collected for bacteriology from each quarter immediately before and approximately 21 days after initiation of treatment. Additionally, a composite milk sample from each cow was collected, on average, 54 days after enrolment for assessment of milk yield, composition and somatic cell count (SCC). The probability of bacterial cure was initially analysed using Chi-squared analysis, and factors that were associated (p<0.2) were offered to a reverse stepwise logistic regression model. Continuous variables (e.g. milk solids production and log10 SCC) were analysed using general linear models. RESULTS: A total of 404 quarters diagnosed with clinical mastitis, from 282 cows in the first 120 days of lactation, were included. Streptococcus uberis, coagulase-negative staphylococci and Staphylococcus aureus were isolated from 56.5%, 18.8% and 10.0% of the bacteriologically positive quarters. There was no difference in the bacteriological cure rate (76.7% vs 76.7%, OR=0.94; p>0.8), the log10 SCC (2.1, SE 0.1, vs 2.0, SE 0.1; p>0.3) or milk production (1.2, SE 0.1, vs 1.2, SE 0.1, kg milksolids/cow/day; p>0.7) between lin/neo vs pen/DHS treatments, respectively. However, the proportion of cows re-treated following initial treatment was higher for the lin/neo compared to pen/DHS-treated group (16.3% vs 5.2%, OR=3.46; p<0.05). CONCLUSIONS: No difference in bacteriological cure rate, milk production or SCC was evident between lin/neo and pen/DHS intramammary treatments for clinical mastitis in dairy cows during the first 120 days of lactation. In a study involving 56 quarters (14 cows), it was seen that standing estrus did not affect circulating neutrophil and milk somatic cell concentrations, neutrophil phagocytosis, milk production, or occurrence of clinical mastitis. The subcutaneous injection of 17beta-estradiol (E2) (0.05 mg/kg of body weight 2 times daily for 2 consecutive days) did not affect neutrophil phagocytosis or milk somatic cell values. However, a significant increase in circulating neutrophils and a significant decrease in milk production were observed. The injection of E2 was followed by clinical mastitis in 4 uninfected quarters, in 7 quarters infected with Corynebacterium bovis, and in 1 quarter infected with Streptococcus uberis and Escherichia coli.  相似文献

Mastitis was induced in 4 lactating cows by inoculation of Klebsiella pneumoniae (10(7) organisms/ml) via the teat canal. Sterile isotonic saline solution (1 ml) was instilled into designated control quarters via the teat canal. Changes in milk leukotriene B4 and C4 (LTB4, LTC4) concentrations, milk somatic cell counts, and milk bovine serum albumin concentration were monitored over a 24-hour postinoculation period. Milk LTB4 concentration before inoculation in control quarters and quarters later to be infected was 376 +/- 45 and 326 +/- 56 pg/ml of milk, respectively. A significant (P less than 0.05) increase in milk LTB4 concentration in the infected quarters was first observed at postinoculation hour 6, and milk LTB4 concentration in infected quarters generally remained significantly high through postinoculation hour 14. Thereafter, milk LTB4 concentration in infected quarters was not significantly different from the concentration in control quarters. Measurable amounts of LTC4 were not detected in the milk of either control or infected quarters. Milk bovine serum albumin concentration in the infected quarters generally was high throughout the study, as were milk somatic cell counts. The results of this study suggested that LTB4 contributes to the pathogenesis of bovine mastitis.  相似文献
