Genetic diversity of populations stored ex situ or in situ can be altered due to the management practices they are subjected to. In this paper, we compare populations of two common bean (Phaseolus vulgaris L.) landraces grown on farms with material collected from the same farms and now kept in two ex situ collections (CIAT and REGEN) with the purpose to monitor any changes that have occurred due to ex situ conservation. The diversity was measured using seven bean microsatellite markers. Further phenotypic and developmental traits were registered in a field experiment. Compared with the in situ populations, the ex situ ones had a lower level of gene diversity and we suggest that this is due to the regeneration process. Most of the phenotypic traits did not differ significantly between ex situ and in situ populations, although for yield and 100-seed weight, the CIAT material showed significant lower values. We assume that these populations have gone through an adaptational change. Overall, the conservation ex situ has been successful in maintaining the majority of the adaptations found in the landraces studied, however, the probable loss of genetic diversity that we have observed, suggest that protocols for the regeneration process must be carefully worked out if the majority of alleles are to be preserved for the future. This study also highlights the complementarity of ex situ and in situ conservation methods in order to preserve landrace adaptations and to capture new, useful diversity generated in in situ populations.  相似文献

The morphological diversity and relationship among 138 pear genotypes, including 11 commercial cultivars and 127 wild pear accessions from central Zagros mountains located in the west of Iran, based on 64 morphological characters were studied. The higher coefficient of variation indicated that high level of variation was belonged to the petiole stipule, fruit the relative area of the skin color, fruit relative area periphery rust dents end, fruit relative blight on an area of skin, fruit curved tail, fruit dents end, fruit depth dents end, fruit relative area periphery patina tail connection characters. Pyrus communis L. and Pyrus glabra Boiss. species were the highest and the shortest tree, respectively. According to the cluster analysis, 138 genotypes were clustered into nine main groups. Principal components analysis revealed that the first three components accounted for 68.77 % of the total variation in data. Moreover, 19 factors justified 72.57 % of the total variation based on factor analysis. The research results showed that the Zagros mountains are the centers of diversity of pear. Therefore, it is necessary to use the Iranian genotypes as the genetic resources for improving the morphological characters of pear.  相似文献

Cleome gynandra L. is a leafy vegetable native to sub-Saharan Africa and Asia and cultivated on dry areas. The plant plays an important role in the food and nutrition security of local communities. The objective of this study was to illustrate variation and diversity in the Cleome collection held at the World Vegetable Center. In total, 242 accessions were cultivated and morphologically characterized and analyzed. The gynophore and filament, both important organs of the flower, played a key role in taxonomical identification. High morphological variation was detected in traits including plant height, pod length, leaf size, flower color, and earliness. Distinct morphological differentiation was detected between Asian and African accessions. On average, the African accessions were larger, less uniform and later in flowering and seed maturation than the Asian accessions. The results were used to establish a core collection of 49 Cleome accessions. Gaps in the current collection were detected. High within-accessions diversity challenges the existing ex situ conservation system; to maintain diversity, seed should be collected from a large number of plants, both during the collection mission and in ex situ regeneration. The results are relevant for germplasm collection strategies and regeneration protocols for good genebank practices.  相似文献

Genetic diversity in 50 snake melon accessions collected from Palestine (West Bank) was assessed by examining variation in 17 phenotypic characters. These accessions belonged to four important landraces of Cucumis melo var. flexuosus: Green “Baladi” (GB), white Baladi (WB), green Sahouri (GS), and white Sahouri (WS). Principal component analysis (PCA) and a dendrogram were performed to determine relationships among populations and to obtain information on the usefulness of those characters for the definition of cultivars. PCA revealed that secondary fruit skin color, flesh color, primary fruit skin color, and secondary skin color pattern were the principal characters to discriminate melon accessions examined in the present study. According to the scatter diagram and dendrogram, landraces of C. melo var. flexuosus: GB, WB, GS, and WS formed different clusters. However, based on Euclidean genetic coefficient distances, GB and WB had the least degree of relatedness with GS and WS, indicating distantly related landraces (Baladi and Sahouri). On the other hand, the highest degree of relatedness was detected between WS on one hand, and both GS and WB on the other indicating closely related cultivars. Fruit traits variability among the different snake melon landraces was evaluated and discussed in this study. This evaluation of fruit trait variability can assist geneticists and breeders to identify populations with desirable characteristics for inclusion in cultivars breeding programs.  相似文献

The variation in 20 morphological and agronomical traits has been evaluated in a set of chickpea genetic resources from four countries. Data indicated differences between accessions in leaf, flower, pod, and seed traits and characteristics, as well as in vegetation period. Multivariate analyses of these data segregated chickpeas into groups. Polymorphism in seed glutelines was absent, while variation in seed prolamines was very limited. DNA amplification patterns have been analyzed by semi-arbitrary primers and by specific microsatellite primers. Only some of semi-arbitrary primers generated usable DNA banding patterns, moreover interpretation of these patterns can be more or less difficult. On the contrary specific primers amplifying microsatellites at the specific loci generated unambiguous and reproducible differences between chickpeas.  相似文献

The Acacia senegal complex is formed by closely related species of Acacia senegal (L.) Willd. These species share several botanical characters, so from a morphological point of view, there is no clear discontinuity between some of them. A. dudgeoni Craib ex Holland is one species of the A. senegal complex that was formerly described as A. senegal ssp. senegalensis var. samoryana (A. Chev.) Rob. In order to differentiate Acacia senegal from A. dudgeoni, we analyzed a range of morphological traits such as tree height and diameter in natural stands, and, at the nursery stage, seedling height, number of branches, main root depth, biomass dry weight and leaf characteristics. Within addition, molecular polymorphism analyses were conducted using 11 microsatellite markers. Leaf characteristics and molecular markers appear to be the most effective tools to distinguish A. senegal from A. dudgeoni. These tools can improve our understanding of the relationship between two species belonging to the same species complex.  相似文献

Gossypium mustelinum Miers ex Watt is a native from Northeastern Brazil and belongs to the primary gene pool of the cultivated cotton. The unique places where the species was known to occur were visited to plan ex situ and in situ preservation. Two populations at Caicó became extinct, and only eleven individuals were found. At Jaguarari, although one population became extinct, a new one was localized with approximately 500 adult plants, where a fence avoided cattle feeding. The population from Macururé consisted of 28 plants protected from animals by thorns of Bromelia laciniosa Mart. ex Schult. f. Thirteen SSR primer pairs amplified 40 alleles, among which 30% were exclusive of one of the populations. The genetic differences between populations represented 58.3% of the total genetic variation observed. The high genetic distances are likely to be caused by geographical isolation as well as by the small number of individuals which contribute to new generations, and consequential genetic drift.  相似文献

Trigeneric hybrids may help establish evolutionary relationships among different genomes present in the same cellular-genetic background, and also offers the possibility to transfer different alien characters into cultivated wheat. In this study, a new trigeneric hybrid involving species from the Triticum, Psathyrostachys and Secale was synthesized by crossing wheat-P. huashanica amphiploid (PHW-SA) with wheat-S. cereale amphiploid (Zhongsi 828). The crossability of F1 hybrid was high with 35.13%, and the fertility was 41.95%. The morphological characteristics of F1 plants resembled the parent Zhongsi 828. The trigeneric hybrids pollen mother cells (PMCs) regularly revealed averagely 19.88 univalents, 9.63 ring bivalents, 3.97 rod bivalents, 0.60 trivalents and 0.03 tetravalents per cell. Multivalents consisted of trivalents and tetravalents can be observed in 52.7% of cells. A variation of abnormal lagging chromosome, micronuclei and chromosome bridge were formed at anaphase I and telophase II. The mean chromosomes number of F2 progenies was 2n = 46.13, and the distribution range was 42–53. GISH results revealed that most F2 plants had 6–12 S. cereale chromosomes, and only 0–2 P. huashanica chromosomes were detected. The results indicated that S. cereale chromosomes can be preferentially transmitted in the F2 progenies of trigeneric hybrid than P. huashanica chromosomes. A survey of disease resistances revealed that the stripe rust resistance from the PHW-SA were completely expressed in the F1 and some F2 plants. The trigeneric hybrid could be a useful bridge for the transference of P. huashanica and S. cereale chromatins to common wheat.  相似文献

Calaverita an endangered Mexican orchid is one of the most elegant and showy orchids of the genus Laelia and essential symbol within the local traditional celebrations of the Day of the Dead, in the region of Chilapa-Guerrero. We investigated morphological variation in flowers of la calaverita (Laelia anceps Lind. subsp. dawsonii (J. Anderson) Rolfe f. chilapensis Soto-Arenas, Orchidaceae) using the labellum of the specimens cultivated in traditional home gardens in the region of Chilapa, Guerrero, Mexico. The use of morphometric traits of the labellum is useful in appraising infra-subspecific morphological variation within the chilapensis form. Floral morphological variation exists among the specimens examined. These specimens, found exclusively in traditional home gardens, were grouped into four morphotypes: NAT, Topiltepec, Xulchuchío, and Terrero. The study of variation of la calaverita revealed the existence of four local morphotypes that originated from a long and complex process of selection of traits which responds to traditional preferences and knowledge about specific biotic and agroecological conditions. The cultural and aesthetic influence of traditional homegarden owners in the Chilapa region acts directly on selection, use and conservation of the morphological traits of la calaverita.  相似文献

In this study a morphological and anatomical variation analysis was carried out with leaves and fruits of Sechium edule, collected in the central region of Veracruz, Mexico. The collected fruits were classified in eight groups according to their typical characteristics. The results showed that the phenotypical distinction of the studied infraspecific S. edule complex is related to morphological and anatomical changes in order to improve the adaptive specialization of the different chayote types with respect to the environment, and suggests that cultivated chayotes have followed different routes in the process of co-evolution with man, showing stable variation patterns in the so-called “green” and “yellow” groups. Considering the phenotypic variability existing in the analyzed characters, a certain prediction may be proposed about the direction the variation patterns may follow facing the transference of chayotes to environments of lower altitudes than mountain cloud forest.  相似文献

Variation in response to climatic and edaphic factors has resulted in the geographic differentiation and specific adaptation of lentil germplasm to its environment. The germination of lentil seeds can be affected by waterlogging caused by relay sowing into standing rice crops. Consequently, there is a demand for waterlogging-adapted lentil genotypes, particularly at germination. This experimentation investigated the effects of waterlogging at germination on a germplasm collection. Firstly, two contrasting genotypes, ATC 70876 and Nugget, were waterlogged for six durations (0, 3, 6, 9, 14 and 21 days) in a pot soil system in the glasshouse. Six days of waterlogging was the duration for germination where genotypes showed the largest contrast in germination rate; therefore, this duration was applied to a larger number (127) of genotypes from 11 countries in a screening trial. Finally, all 127 genotypes were grown in pots in the open to phenotype morphological traits (seed size, above-ground biomass, time to flower and maturity). There was significant variation among genotypes, and those originating from Bangladesh had the highest germination at 21.2 %. The heritability of germination on waterlogged soil among genotypes was intermediate at H2 = 44.8 %. Correlations between morphological and physiological traits showed that the capability of seeds to germinate on waterlogged soil was negatively correlated to time to maturity (r = ?0.643). It was concluded that genotypes from Bangladesh are adapted to waterlogging at germination. With an intermediate heritability for germination under waterlogging conditions, selection for relay sowing through waterlogging tolerance at germination is practicable.  相似文献

Thaumatococcus daniellii is a perennial African wild edible plant. Fruit arils from natural populations are the source of the protein sweetener, thaumatin. There are relatively few documented examples of its cultivation, however it has great potential as an intercrop for cultivation under rubber or cocoa. Across the species range there is notable diversity in leaf and fruit morphology. In this study, T. daniellii populations from four provenances in Ghana and Cameroon (separated by the Dahomey Gap) were compared for leaf and fruit morphology in situ and in an experiment using plants grown from sampled rhizomes planted under rubber in a single location in Cameroon. When collected rhizomes from four provenances were planted at one location, the samples from the local provenance produced the largest plants and fruits, and there was less morphological variation between samples from different provenances than was recorded in situ. In situ morphological differences were notable between north–south separated provenances in each country. Fruit collected in situ was analysed for aril thaumatin content and significant variation was found between provenances (1.08–2.00%). Amplified fragment length polymorphism (AFLP) was used to assess genetic diversity among the four provenances and 54% of loci were polymorphic. Cluster analysis of AFLP data separated samples of T. daniellii according to geographical origin. Maximum genetic diversity was found between provenances separated across the Dahomey Gap.  相似文献

Weedy rice (Oryza sativa f. spontanea Baker) is one of the most serious weeds in direct-seeded rice production; adversely affecting rice areas and yield in Thailand. The objectives of this study were to evaluate the level of morphological and agronomical variation and the level and pattern of genetic diversity and population structure of weedy rice populations in Thailand. Sixty-three weedy rice populations collected from three rice production areas of Thailand were sampled over four rice cultivation seasons to study phenotypic characteristics, genetic diversity and population structure. All analysis included comparison with seven cultivated rice varieties and seven local common wild rice populations. The results demonstrated considerable genetic and morphological variation, population structure, and changing dynamics of Thai weedy rice populations, both within and between regions and growing seasons. Phenotypic and genotypic variations and population structure of weedy rice populations changed temporally, with a trend over five years toward greater similarity to companion crop rice varieties. The ongoing gene flow, combined with ecological conditions created by farmers’ selection and agronomic practices, may enhance fitness and adaptability of weedy rice populations and stimulate the evolution of invasiveness in weedy rice plants. The adaptive evolution of weedy rice tended to parallel cultivated rice for high adaptation to the agronomic practices, possibly including selection for crop mimicry, which has led to the convergence of weedy populations phenotypically and genetically with the companion crop rice.  相似文献
