Two new varieties of interspecific hybrids of Passiflora have been developed from the cross between P. gardneri versus P. gibertii, both registered under the Passiflora Society International. Twelve putative hybrids were analyzed. Hybridization was confirmed using RAPD and SSR markers. Primer UBC11 (5′-CCGGCCTTAC-3′) generated informative bands. Primer SSR Pe75 has amplified species-specific fragments and a heterozygote status was observed with two parent bands 300 and 350 bp. The molecular markers generated have been analyzed for the presence or absence of specific informative bands. Based on the morphological characterization, we have identified two hybrid varieties: P. ‘Gabriela’ and P. ‘Bella’. P. ‘Gabriela’ produced flowers in bluish tones, bluish petals on the adaxial and abaxial faces, light blue sepals on the adaxial and light green on the abaxial faces, corona with the base of filaments in intense lilac color and white apex. P. ‘Bella’ produced flowers in lilac tones, intense lilac petals on the adaxial and abaxial faces, dark lilac sepals with whitish edges on the adaxial and light green on the abaxial faces, corona with the base of filaments in intense lilac color and white apex. The cytogenetic analysis verified that the hybrids have the same chromosomal number as the parents (2n = 18); the formation of bivalents between the homeologous chromosomes (n = 9) was observad, leading to regular meiosis, which allows the sexual reproduction and use of these hybrids in breeding programs.  相似文献

Knowledge of ploidy level differences, genome size and genetic relationships between species facilitates interspecific hybridization in ornamentals. For Sarcococca (Buxaceae) only limited (cyto)genetic information is available. The aim of this study was to determine the genome size and chromosome number and to unravel the genetic relationships of a breeder’s collection using AFLP marker analysis. Based on these results, interspecific crosses were made and the efficiency and hybrid status was verified. Two groups of diploid plants (2n = 2x = 24) were observed, with either a genome size of 4.11–4.20 or 7.25–9.63 pg/2C. All the tetraploid genotypes (2n = 4x = 48) had genome sizes ranging from 7.91 to 8.18 pg/2C. In crosses between parents with equal ploidy level and genome size a higher crossing efficiency (on average 58% of the hybridizations resulting in fruits) and more true hybrids (on average 96% of the offspring) were obtained compared to crosses between plants with different genome size and ploidy level (on average 23% fruits and 24% hybrids, respectively). In none of the cross combinations, the ploidy level or genome size was found to be a complete hybridization barrier, although unilateral incongruity was found in some cross combinations. Distant genetic relationships did not hamper the hybridization within Sarcococca genotypes. Our findings will contribute to a more efficient breeding program and a faster achievement of hybrids with an added value.  相似文献

Nineteen accessions of the tuber-bearing species Solanum berthaultii, S. chacoense, S. leptophyes, S. microdontum, S. sparsipilum, S. sucrense, S. venturii, S. vernei and S. verrucosum were tested for their resistance to late blight in two years of field experiments. Plants were artifically inoculated with zoospores of race 1.2.3.4.5.7.10.11 and the development of the disease was followed. Resistance ratings, calculated as the areas under the disease progress curves (ADPC), demonstrated a high resistance in all accessions except in S. sparsipilum, S. leptophyes and their interspecific hybrid. Segregations suggest that major genes for resistance are present in S. sucrense and S. venturii, and may also play a role in S. verrucosum. It is not yet certain wether the resistance of the other accessions is comparable to the partial and durable resistance of S. tuberosum cultivars like Pimpernel, as inheritance and mechanism have yet to be established. However, segregations suggesting the presence of single major genes with complete dominance were not found in these other accessions. Tuber initiation in the field occurred in only one accession, S. tuberosum ssp. andigena, and maturity of the clones was not related to their resistance. In the other accessions maturity types could not be assessed, as the clones require short day conditions for tuber initiation.  相似文献

Most forage cultivars released for the genus Paspalum belong to a section named Plicatula. The species of Plicatula are mostly apomictic and consequently the genetic diversity is locked for their genetic improvement. The objectives were to evaluate the crossability, hybrid fertility, heterosis, and genetic distances between apomictic accessions and a sexual genotype of species of Plicatula group of Paspalum. Crosses were made using 22 apomictic tetraploid accessions belonging to 12 different species as pollen donors, and a sexual tetraploid genotype induced by colchicine from a sexual diploid accession of P. plicatulum. Crossability varied between 0 and 16% among crosses. Viable hybrid offspring were recovered from 15 out of 22 crosses. The most successful crosses involved P. guenoarum, P. plicatulum, P. chaseanum, and P. oteroi. Fertility of the sampled hybrids varied between 1.6% for the cross involving P. lenticulare, and 40.1% for an intraspecific cross (P. plicatulum, accession Hojs388). The genetic distance between parents was estimated using amplified fragment-length polymorphism, and it varied between 0.34 and 0.53. There was no correlation between genetic distances and crossability or fertility of the hybrids. Hybrids from the most numerous families were classified for mode of reproduction using flow cytometric seed analysis. The ratio between sexual and apomictic hybrids varied between 0.6:1 and 1.6:1. A selected group of apomictic hybrids were evaluated for several agronomic traits in the field. Heterosis was observed for frost tolerance and cattle preference. The results indicated that gene transfer via hybridization is possible among several species of Plicatula. Superior hybrids for specific traits can be generated and fixed by apomixis.  相似文献

The germplasm of valuable for breeding wild allotetraploid potato species Solanum stoloniferum is rarely used because of pre- and postzygotic reproductive barriers with cultivated potatoes. One of the factors that complicate crosses between S. stoloniferum and S. tuberosum is unilateral incompatibility (UI). Here, we present the results of application of S. verrucosum and S v S v -lines for overcoming UI in crosses with S. stoloniferum and of generating male fertile hybrids derived from this species. S v S v -lines are F2 S. tuberosum dihaploid × S. verrucosum that are male fertile and have D/γ-type cytoplasm. Since they are homozygous for S v gene from S. verrucosum, they were expected to have the same ability for elimination of prezygotic incompatibility as this species. Three accessions of S. verrucosum and seven S v S v -lines were pollinated by 26 accessions of S. stoloniferum. The crosses with S. verrucosum failed or had low efficacy (1.5–2.4 seeds per pollination). On the other hand, use of S v S v -lines was more efficient: 15.8 seeds per pollination. In spite of low percentage of germination (1.9%), 40 seedlings of interspecific hybrids were produced. The experiment on hybridization between S v S v -lines and S. stoloniferum has been reproduced with the accession PI 205522 of the wild species, which had DNA markers of PVY and LB resistance genes and W/γ cytoplasm: 950 hybrid seeds and 12 viable seedlings were produced. The genome of the seedlings was doubled by colchicine treatment, which generated hexaploids that formed highly fertile pollen and set seeds from self-pollination. In most sugarcane cultivation areas, sugarcane brown rust (SBR), caused by Puccinia melanocephala, is an economically important fungal disease that leads to severe yield loss in susceptible cultivars. Bru1, which is the major dominant SBR resistance gene, has been widely used in the prediction of brown rust resistance in sugarcane. In this study, three panels of sugarcane germplasms, the major varieties approved over the past 10 years and new elite clones in the current national regional trial, together with one panel of Saccharum spontaneum, were employed in estimating the possibility of SBR epidemic and to assess the efficiency of 9O20-F4-HaeIII in eliminating false positives. Among the current top five varieties used as sucrose feedstock, accounting for more than 68.9% of the total cultivated area, all were highly resistant to SBR, although only three harboring Bru1. Two major varieties Yuetang60 and Guitang46 without harboring Bru1 were highly susceptible to SBR, together with highly susceptible Funong41, which need prudent promotion. Approximately 60.5% of the 38 new elite clones were Bru1 positive. Considering the susceptibility of Liucheng03-1137, which exhibits a strong promotion momentum, together with Funong41, Guitang46, Yuetang60, and Yunzhe06-47, four were favored by the enterprise due to their superior sucrose content and good stalk yield, despite their high susceptibility to SBR, and additional Yuetang93-159, one current top five varieties with declining resistance, which results in a potential risk for brown rust epidemic. Furthermore, low frequency of the wild germplasm of S. spontaneum from five different countries was Bru1 positive. In addition, a perfect molecular diagnostic result was observed in all modern sugarcane clones using two dominant markers, and HaeIII can prevent the occurrence of false positive results when the 9O20-F4 PCR products of S. spontaneum are digested by RsaI. The prevalent chewing cane Badila without Bru1 is highly resistant to SBR. Our results provide valuable information for the extension of sugarcane varieties and a batch of novel SBR resistance sources with superior comprehensive characters for crossbreeding, and for SBR-resistant gene pyramiding by crossing or through mining and using of new SBR-resistant genes.  相似文献

The information of ploidy, karyotype and genetic relationship is useful for interspecific hybridization in ornamental plants. For Tulipa species native to China, very limited cytological information is available now. The objective of this study was to verify the chromosome number, karyotype and genetic relationship of the eight Tulipa species: T. edulis, T. schrenkii, T. iliensis, T. thianschanica, T. altaica, T. sinkiangensis, T. heterophylla and T. buhseana. And the interspecific crosses were made between T. altaica and ten tulip cultivars to obtain novel germplasm. The ovary-swelling, fruit-setting and bulblet formation rates were surveyed when different ploidy cultivars were used as female parents. This work confirmed that all eight species collected in China were diploid (2n?=?2x?=?24), among which chromosome numbers of T. thianschanica, T. sinkiangensis and T. heterophylla were firstly reported and the karyotypes of all any other species except for T. edulis were determined for the first time. The karyotypes of eight Tulipa species were classified as 3A, 4A or 3B. The results of interspecific hybridization showed significant difference when different ploidy cultivars were used as female parents. The highest fruit-setting rate was obtained when diploid cultivars were used as female parents crossed with diploid T. altaica, whereas the ovary swelling was observed in two out of four triploid cultivars as female parents, and no seeds were harvested when tetraploid cultivars were used as female parents. Our findings provided an effective means of cultivar improvement in tulip.  相似文献

    In attempts to intercross Lolium perenne and L. multiflorum with Festuca pratensis in a glasshouse, 13,284 emasculated flowers of diploid, triploid and tetraploid Lolium plants were provided with pollen of diploid and tetraploid Festuca plants. Only the combinations 2n\sx4n and 3n\sx4n produced hybrids, viz. on average 5.3 and 1.0 mature hybrid plants per 100 pollinated flowers. In a previous investigation on the reciprocal difference of interspecific hybridization between three different flower colors of Iris dichotoma and Iris domestica in the F1 offspring from crosses where I. domestica was a maternal parent were similar in morphological and cytological characters to their maternal parent. This could be evidence of apomixis; however, matroclinal progeny with complete morphological similarity to the maternal parent could be attributed to the heterozygosity for these characters in the pollen parent. The F1 plants were investigated in order to identify apomixis in I. domestica. Four matroclinal plants were randomly selected from each F1 population produced from Iris domestica × Iris dichotoma that had three different colors of flowers and were allowed to self-pollinate to establish an F2 population. All of the F2 plants had no segregation to I. domestica in their morphological characters. In addition, 13 reciprocal F1 plants were detected by 25,719 single nucleotide polymorphism (SNP) markers. When I. dichotoma plants with three different flower colors were used as maternal parents, all the progenies were genuine hybrids. When I. domestica were used as maternal parents, all the F1 plants were apomictic progenies. Apomixis of I. domestica was successfully identified and SNP markers identified F1 hybrids derived from six interspecific crosses between I. dichotoma and I. domestica, which provides a reference for authenticating offspring identity during Iris cross breeding in the future.  相似文献

The genus Psidium includes important fruit crops. However, there are very few studies focusing on its reproductive biology, which limits the establishment of breeding programs. The present work investigated the reproductive biology of Psidium guajava and Psidium cattleianum in terms of compatibility of crossings between these two species aiming at interspecific hybridization because the latter species is an important source of resistance against the nematode Meloidogyne enterolobii. Several types of crosses were performed to understand the reproductive biology of these species, including the compatibility of intra- and interspecific crossings, using assisted in vivo germination of pollen grains on the stigma. In addition, the in vitro germination of both Psidium species was studied at different stages of fruit development to rescue young seeds to improve the chances of obtaining the hybrids. No fruits of 270 pollinations were obtained on guava buds at the pre-anthesis stage, regardless of the source of the pollen grain and the cultivar used as female genotype. Microscopic analyzes demonstrated the germination of pollen grains and pollen tube growth at crosses between guava cv. ‘Pedro Sato’ (P. guajava) and Psidium cattleianum. High germination percentages of Psidium cattleianum seeds were obtained in MS medium without sucrose or containing 15 g/L of this carbohydrate.  相似文献

Interspecific hybrids were developed between Trifolium alexandrinum cultivar Wardan × Trifolium vesiculosum and T. alexandrinum cultivar BL1 × T. vesiculosum through embryo rescue, as the crosses failed to set seed under natural conditions. Trifolium vesiculosum was used as a donor/male parent in this study as it is reported to possess tolerance to stem rot and high forage yield. Fertilization in crossed florets of the crosses was manifested from the recovery of swollen ovaries (< 7.80%) and confirmed from the presence of one degenerated ovule in most (> 93.00%) of the swollen ovaries. The hybrid embryos at various developmental stages (heart, torpedo and cotyledonary) were rescued at a frequency of 2.56% from Wardan × T. vesiculosum and 6.12% from BL1 × T. vesiculosum. Differentiation occurred only in the cotyledonary stage embryos, resulting in 17 putative interspecific hybrid plantlets. The assessment of plantlet hybridity through SSR markers (for the alleles inherited from the donor parent), micromorphological leaf traits (leaf texture and stomata) and morphological characters (plant height, leaflet length and width) confirmed production of two interspecific hybrids designated as AV1 and BV3 representing both the crosses. AV1 displayed moderate resistance and BV3 was resistant to stem rot.  相似文献

Cultivar pedigrees from two sugarcane origins, 9 Argentine (AR) and 7 American (AM) have been reconstructed, and their genetic similarities (based on coefficient of parentage, COP, estimates) show an average of 0.206 ± 0.054. CP clones that enter the pedigrees of AM cultivars are parents or grandparents of AR cultivars, demonstrating that these genotypes have a strong genetic lineage in common. On average, AR pedigrees are smaller and contain less number of founding species than AM pedigrees. However, the lower height of the former is not explained by the different participation of founding species in the pedigrees. The presence of founding species in AR and AM pedigrees increases with the year of cultivar selection, indicating that more founding species entered the pedigree in recently selected cultivars than in older ones. The ancestry of the 16 cultivars trace back to 3 founding species: Saccharum officinarum, S. barberi, and S. spontaneum, with S. officinarum showing the greatest percentage of participation. As S. sinensis participates in 13 pedigrees and S. robustum in 3, the 5 founding species are present in 2 out of 16 pedigrees. Interestingly, the genus Sorghum enters the pedigree of LCP 85-384 pedigree. Industrial parameters assayed indicate that AR and AM cultivars were preferentially selected for their relatively high sucrose content and sugar yield in accordance with two facts: (i) the genetic background and the sucrose genes of sugarcane hybrids provided by the 68% of participation S. officinarum clones, and (ii) no clones of high fiber S. spontaneum have entered the pedigrees in earlier generations.  相似文献

Pre-harvest sprouting (PHS) causes significant yield loss and degrade the end-use quality of wheat, especially in regions with prolonged wet weather during the harvesting season. Unfortunately, the gene pool of Triticum durum (tetraploid durum wheat) has narrow genetic base for PHS resistance. Therefore, finding out new genetic resources from other wheat species to develop PHS resistance in durum wheat is of importance. A major PHS resistance QTL, Qphs.sicau-3B.1, was mapped on chromosome 3BL in a recombinant inbred line population derived from ‘CSCR6’ (Triticum spelta), a PHS resistant hexaploid wheat and ‘Lang’, a PHS susceptible Australian hexaploid wheat cultivar. This QTL, Qphs.sicau-3B.1, is positioned between DArT marker wPt-3107 and wPt-6785. Two SCAR markers (Ph3B.1 and Ph3B.2) were developed to track this major QTL and were used to assay a BC2F8 tetraploid population derived from a cross between the durum wheat ‘Bellaroi’ (PHS susceptible) and ‘CSCR6’ (PHS resistant). Phenotypic assay and marker-assisted selection revealed five stable tetraploid lines were highly PHS resistant. This study has successfully established that PHS-resistance QTL from hexaploid wheat could be efficiently introgressed into tetraploid durum wheat. This tetraploid wheat germplasm could be useful in developing PHS resistant durum cultivars with higher yield and good end-use quality.  相似文献

Wild abortive (WA)-type cytoplasmic male sterility (CMS) has been exclusively used for breeding three-line hybrid indica rice, but it has not been applied for generating japonica hybrids because of the difficulties related to breeding japonica restorer lines. Determining whether the major restorer-of-fertility (Rf) gene used for indica hybrids can efficiently restore the fertility of WA-type japonica CMS lines may be useful for breeding WA-type japonica restorer lines. In this study, japonica restorer lines for Chinsurah Boro II (BT)-type CMS exhibited varying abilities to restore the fertility of ‘WA-LiuqianxinA’, which is a WA-type japonica CMS line. Additionally, Rf genes for WA-type CMS were identified in the BT-type japonica restorers. Meanwhile, ‘C9083’, which is a BT-type japonica restorer, exhibited a limited ability to restore the fertility of WA-type japonica CMS lines, and a genetic analysis revealed that the fertility restoration was controlled by one locus. The Rf gene was mapped to an approximately 370-kb physical region and was identified as Rf4. Furthermore, Rf gene dosage effects and the temperature influenced the fertility restoration of WA-type japonica CMS lines. This study is the first to confirm that Rf4 has only minor effects on the fertility restoration of WA-type japonica CMS lines. These results may be relevant for the development of WA-type japonica hybrids.  相似文献

Strawberry cultivars showed limited cold resistance in the Northeast of China, while we obtained a synthetic dodecaploid strawberry hybrid ‘YH15-10’ (2n = 12x = 84) which showed sufficient cold resistance in this area. The reciprocal crosses between F. × ananassa cv. ‘Allstar’ (2n = 8x = 56) and ‘YH15-10’ (2n = 12x = 84) were carried out to select cold resistant strawberry in this study. The 134 seedlings were obtained from the cross of Allstar × YH15-10, while failed in its reciprocal cross. The 30 randomly selected seedlings were examined in terms of morphological characters, chromosome numbers and cold resistance. Most morphological characters were widely separated among F1 progeny with a high broad-sense heritability, which showed that these variations mainly resulted from genetic effect. Some hybrids exhibited heterosis, especially in growth vigor and runner production. Among the 30 tested hybrids, 28 decaploids (2n = 10x = 70), one octoploid (2n = 8x = 56) and one enneaploid (2n = 9x = 63) were observed. The 63.3% hybrids demonstrated higher cold resistance than that of ‘Allstar’ at P < 0.05. These high polyploidy strawberries have potential values in commercial production and modern cultivar improvement.  相似文献

Tomato is affected by a large number of arthropod pests, among which the whitefly (Bemisia tabaci) is considered to be one of the most destructive. Several accessions of the wild species of Solanum galapagense, including accession LA1401, are considered resistant to whitefly (B. tabaci). This resistance has been associated with the presence of type IV glandular trichomes on the leaf surface. Our research aimed to study the inheritance of type IV glandular trichome density and its association with resistance to whitefly (B. tabaci biotype B) in populations derived from the interspecific cross Solanum lycopersicum × S. galapagense ‘LA1401.’ High estimates for both broad-sense and narrow-sense heritabilities of type IV glandular trichome densities suggest that inheritance of this trait is not complex. Whitefly resistance was associated with high density of type IV glandular trichomes. F2 (S. galapagense × S. lycopersicum) population plants selected for the highest densities of type IV glandular trichomes showed similar levels of resistance to those found in the donor of resistance LA1401.  相似文献

Cotton although is an autogamous species could be cross pollinated under favorable climate conditions and/or in the presence of pollinators. The coexistence of cotton with Malvaceae species raises questions on the possibility pollen to be exchanged among Malvaceae species and on the resulted consequences. The present work was undertaken to evaluate the in situ response of cotton flowers (G.hirsutum L, G. barbadense L.) and their F1 interspecific hybrids when are artificially pollinated with Malva sylvestris L., Hibiscus syriacus L. and Abelmoschus esculentus Moench. Furthermore, an in vitro protocol was attempted to support embryos’ growth in order to produce viable progenies originating from crosses between cotton and the aforementioned Malvaceae species. The obtained results gave evidence that pollen from the above Malvaceae species stimulated cotton ovaries without successful hybridization. The interaction between pollen and cotton’s stigmas was higher at early stages when M. sylvestris was used as pollinator but in crosses with H. syriacus and A. esculentus more ovules were activated as revealed by the percentage of carpodesis and life-time of bolls onto maternal plants. Only crosses between cotton with okra produced cotton seeds under in situ conditions. The in vitro embryo-ovule culture protocol, used, increased the number of regenerated cotton plants, especially in crosses among F1 interspecific cotton hybrids and A. esculentus. In this case, regenerated plants were recombinant aneuploids, combining traits from both cotton species. This novel cotton germplasm possessing unique chromosome rearrangements, at aneuploid level could be proved useful after cytogenetic, molecular or QTL genetic analysis referring to important agronomic traits.  相似文献

The accession Solanum peruvianum PI 126944 has been reported as resistant to different biotic and abiotic stresses. In previous works of the group, advanced generations were derived from two interspecific hybrids, including up to pseudo-F6 generations and backcrosses to the cultivated species of some of the pseudo-Fn generations. Moreover, resistance to Tomato yellow leaf curl virus (TYLCV) and Tomato spotted wilt virus (TSWV) was confirmed in some of the plant materials obtained. Here we describe the development of some advanced backcross generations between the cultivated tomato and S. peruvianum. Three backcross assays were carried out. In the first assay, 16 plants belonging to pseudo-F4, pseudo-F5 and pseudo-F6 generations were backcrossed as male parents to cultivated tomato. Immature seeds from the 408 fruits obtained were platted and 77 plants were regenerated. A total of 21 descendants obtained in the first assay were backcrossed as male parents in the second assay, which allowed the obtaining of 599 fruits and regeneration of 365 plants. The 19 male parents used in the third assay were obtained either in the first or in the second one. Seventeen of them were obtained in the first assay and two in the second. A total of 339 plants were regenerated from 519 fruits. A high number of the plants obtained in these assays has been acclimated. Some plants set fruits with viable seeds.  相似文献

Gluten protein determines the processing quality of both durum wheat and bread wheat. The glutenin subunits compositions and associated quality traits of 20 Ethiopian durum wheat varieties were systematically analyzed using SDS-PAGE and Payne numbers. A total of 16 glutenin patterns were identified. At the Glu-A1 locus, all varieties scored the null allele. The predominant glutenin alleles at the Glu-B1 locus were Glu-B1b (7+8) and Glu-B1e (20). In Glu-3, the most abundant glutenin subunits were Glu-A3a and Glu-B3c. Based on the Payne scores, the varieties Yerer, Ginchi, Candate, and Foka were identified to have allelic composition suitable for pasta making. The cluster analysis using agglomerative hierarchical clustering (AHC) method classified the varieties into four similarity classes. Based on the findings of this experiment, suggestions were made for allelic composition improvement through introgression of superior alleles from known Glu-1 and Glu-3 sources.  相似文献

The root lesion nematode Pratylenchus thornei is widely distributed in Australian wheat (Triticum aestivum) producing regions and can reduce yield by more than 50%, costing the industry AU$50 M/year. Genetic resistance is the most effective form of management but no commercial cultivars are resistant (R) and the best parental lines are only moderately R. The wild relatives of wheat have evolved in P. thornei-infested soil for millennia and may have superior levels of resistance that can be transferred to commercial wheats. To evaluate this hypothesis, a collection of 251 accessions of wheat and related species was tested for resistance to P. thornei under controlled conditions in glasshouse pot experiments over two consecutive years. Diploid accessions were more R than tetraploid accessions which proved more R than hexaploid accessions. Of the diploid accessions, 11 (52%) Aegilops speltoides (S-[B]-genome), 10 (43%) Triticum monococcum (A m -genome) and 5 (24%) Triticum urartu (A u -genome) accessions were R. One tetraploid accession (Triticum dicoccoides) was R. This establishes for the first time that P. thornei resistance is located on the A-genome and confirms resistance on the B-genome. Since previous research has shown that the moderate levels of P. thornei resistance in hexaploid wheat are dose-dependent, additive and located on the B and D-genomes, it would seem efficient to target A-genome resistance for introduction to hexaploid lines through direct crossing, using durum wheat as a bridging species and/or through the development of amphiploids. This would allow resistances from each genome to be combined to generate a higher level of resistance than is currently available in hexaploid wheat.  相似文献
