Abstract

After an epizootic of amyloodiniosis (caused by the protozoan Amyloodinium ocellatum) in a commercial aquaculture facility for hybrid striped bass (female striped bass Morone saxatilis × male white bass M. chrysops), sera from these fish, as well as from others that had been experimentally immunized with the parasite, were evaluated by an enzyme-linked immunosorbent assay for antibody specific for the parasite. Titers were similar between the fish infested in culture and the experimentally immunized fish, and were significantly higher in both the cultured and artificially exposed fish than in unexposed fish. These results suggest that an infestation of A. ocellatum can stimulate the production of humoral antibodies to the parasite, providing further evidence that natural infestations of the parasite may confer protective resistance in fish that survive the initial parasitic infestation.  相似文献

Abstract

An outbreak of amyloodiniosis in pondcultured hybrid striped bass × white bass (female Morone saxatilis × male M. chrysops) is the northernmost documented outbreak caused by an endemic source in fish of the western Atlantic. Dinospores produced by this isolate of Amyloodinium ocellatum were anteroposteriorly flattened, as contrasted with the gymnodinoid dinospores of the A. ocellatum-type species, suggesting that more than one strain or species of A. ocellatum may exist.  相似文献

Abstract

Commercial Vibrio anguiliarum-V. ordalii bacterin was used to vaccinate hybrid striped bass (Morone saxatilis ♀ × M. chrysops ♂) to test the vaccine efficacy against vibriosis. Vaccination by direct immersion of fish in diluted Vibrio vaccine for 20 s resulted in increased protective immunity. The relative percent survival of hybrid striped bass challenged 35 d after vaccination was 66.7% for those challenged by 1-h immersion exposure to 7.03 × 107 V. anguillarum cells/mL, 75.0% for those challenged by injection with 3.51 × 105 cells/fish, and 86.7% for those challenged by injection with 3.51 × 104 cells/fish.  相似文献

Abstract

Intense infections of the gill pathogen Dermocystidium salmonis were associated with mortality of prespawning chinook salmon Oncorhynchus tshawytscha in several Oregon rivers in 1988. The occurrence of the pathogen in returning adult chinook salmon was monitored in several coastal Oregon stocks from 1989 to 1993. Although the prevalence of the pathogen was high in these fish (up to 66.6%), infection intensities were generally low, and no mortality attributable to D. salmonis was observed. In 1988, the pathogen was associated with a lethal epizootic among juvenile chinook salmon smolts at the Trask State Fish Hatchery near Tillamook, Oregon. Histological examination of gills from heavily infected fish revealed hyperplasia of gill epithelium and fusion of gill lamellae. When naturally infected smolts were transferred from fresh to salt water, the most heavily infected fish died within 10 d, and the number of D. salmonis cysts declined and disappeared from previously infected salmon after 21–42 d.  相似文献

Abstract

Specific-pathogen-free channel catfish Ictalurus punctatus were exposed to sediment and mud from a pond containing channel catfish with proliferative gill disease. In one experiment, fish were to exposed to mud and sediment for 2 months in water maintained at 19°C. Fish were necropsied weekly, and certain tissues were examined histologically and ultrastructurally. Four trials were conducted with sediment samples from different epizootics of proliferative gill disease. In a second experiment, fish were exposed to sediment for 7 d in water maintained at 16, 19, or 26°C; the fish were then moved to clean water held at 16, 19, or 26°C. Fish were necropsied before transfer to clean water and weekly thereafter for 2 months. Channel catfish held at 19°C developed proliferative gill disease within 2 d of exposure to sediment. Primary cells of a uninucleate myxosporean parasite were present in the gills at the base of lamellae. These developed into plasmodia with numerous secondary cells, and some primary cells disintegrated, releasing their internal secondary cells. Similar development was observed in internal organs 1 week after appearance of the parasite in gills. Complete sporogony did not occur over the 2 months of this study. Plasmodia became necrotic and were not detected after 60 d. In fish exposed to sediment for 7 d at 16, 19, and 21°C, similar organisms were detected, but clinical disease occurred only at 19 and 26°C. Proliferative gill disease may be attributed to extrasporogonic stages of a myxosporean resembling Sphaerospora spp.  相似文献

Lamproglena clariae attaches to gill filaments of Clarias gariepinus, penetrates the gill tissue and consumes blood. Hence, the purpose of this study was to investigate the mechanical damage caused by this parasite on gill filaments of the host. Infestation intensity and haematocrit values of fish blood were determined and correlated. Fish were killed, gills dissected out and observed with a dissection microscope. Gill filaments with adult females in situ were fixed in a solution of alcohol, formaldehyde and acetic acid and preserved in 70% ethanol. Fixed specimens were studied by histological examination following staining with Heidenhein’s azan solution. Gross morphology revealed varying degrees of swelling of gill filaments near the attached specimens of L. clariae. Epithelial hyperplasia, resulting in fusion of gill lamellae and necrosis of the host tissue in the vicinity of the parasite were revealed through histological examination. No correlation was evident between the infestation intensity and fish haematocrit values. Although results showed that L. clariae causes localized infestation, high intensity of infestation may be detrimental to the fish. Hyperplasia of epithelial tissue resulting in fusion of lamellae would result in restriction of air passages and thus hinder the process of respiration of the host.  相似文献

This study aimed to determine the cellular alterations in the gill of Labeo rohita exposed to lethal temperature maxima (LTMax) and lethal temperature minima (LTMin) by means of transmission electron microscopy (TEM). Acclimation of advanced fingerlings of L. rohita was carried out at 26°C for 30 days. Acclimated fish were subjected to a constant rate of increase or decrease in temperature (0.3°C/min) until the LTMax and LTMin values were reached. Dissected gills were processed for TEM, both at the end of acclimation period at ambient temperature (26°C) and at lethal temperatures. Results indicated that at ambient temperature, the gill tissues appeared normal. However, significant changes were observed at lethal temperatures. The gill tissues at lethal temperature maxima showed severely damaged lamellae, with more vacuolated space. At lethal temperature minima, gill tissues showed increased density of mitochondria. Our prima‐facie report indicated that L. rohita exposed to lethal temperatures exhibited marked ultrastructural changes in the gills.  相似文献

Abstract

Largemouth bass virus (LMBV) is an iridovirus that was isolated from wild adult largemouth bass Micropterus salmoides in the southeastern United States in 1994. Although originally isolated from moribund wild fish, its virulence to juvenile largemouth bass is uncertain. To help clarify this point, two LMBV titrations were made in juvenile largemouth bass. Titers of LMBV in fathead minnow cells were 104.8 and 105.8 tissue culture infectious doses—50% cytopathic endpoint (TCID50) per milliliter, respectively. Tenfold serial dilutions of LMBV employed in each cell culture titration, injected intraperitoneally (0.1 mL/fish) into largemouth bass produced calculated lethal dose—50% mortality endpoints (LD50s) of 282 (102.45) and 288 (102.46) infectious doses in two consecutive infectivity trials. Virus yield of assayed infected fish averaged 108.5 TCID50/g and 107.7 TCID50/g in viscera of moribund and dead fish in the two trials and 106.5 TCID50/g in surviving exposed fish 14 d after infection. In a second experiment, largemouth bass had 100% mortality 5 d after injection while virus immersed fish had a significantly (P ≤ 0.005) lower mortality of 17% at 14 d. Similarly treated juvenile striped bass Morone saxatilis suffered 63% mortality after injection and significantly (P ≤ 0.005) lower mortality of 10% after immersion. In a third study of 25 d, 100% of injected largemouth bass died by 5 d after injection, and all of them were virus-positive. Injected striped bass had a significantly (P ≤ 0.005) lower mortality of 24%; all three fish were virus-positive initially, two fish were virus-positive at 18 d, and none were positive at 25 d. Juvenile largemouth bass were highly susceptible to LMBV injection and striped bass were moderately susceptible, but both species were only mildly susceptible when exposed by immersion.  相似文献

Abstract

Proliferative gill disease (PGD) was first reported in channel catfish Ictalurus punctatus at commercial farms in 1981 and is caused by the myxozoan parasite Henneguya ictaluri. The disease affects the gills and is characterized by severe branchial inflammation, epithelial hyperplasia, lamellar fusion, and lysis of chondrocytes. Presumptive diagnosis is based on the presence of lytic areas in the cartilage of the primary lamellae on microscopic examination and is confirmed histologically by the presence of the organism. In these trials, PGD was induced by exposing channel catfish fingerlings to fresh or aged infectious water collected from a pond containing fish diagnosed with severe PGD. The severity of disease was graded by histological scoring and microscopic examination of wet mounts to determine the percentage of gill filaments containing chondrolytic lesions. Exposure of fish to infectious pond water was shown to produce pathological lesions consistent with PGD, and the percentage of gill filaments containing chondrolytic lesions was positively correlated with histological scoring of gill pathology. The number of trophozoite stages in the gills was shown to increase with the severity of the disease. In most cases, however, parasitic cells were not observed in tissue samples with chondrolytic lesions during the early stages of infection. These observations indicate that pathology and lysis of chondrocytes can occur prior to detection of the organism by histopathology. Exposing fish to infectious pond water that was aged for 1 d produced negligible gill pathology and implies that the infectivity of the H. ictaluri actinospore stage is short lived. Removing fish from the source of infection promoted repair of damaged gill tissue; within 14 d of fish transfer to clean water, gill pathology associated with the acute infection was negligible.  相似文献

A series of histochemical techniques have been employed to localize alkaline phosphatase, acid phosphatase, non-specific esterase, catalase and peroxidase; and to visualize and characterize glycoprotein (GPs) moieties in the epithelium of gill arch, gill filaments and secondary lamellae of an angler catfish Chaca chaca. The epithelium of gill arch and gill filament shows strong alkaline phosphatase activity in the deeper layer epithelial cells; strong non-specific esterase activities in the outer layer epithelial cells; and weak acid phosphatase activity throughout the epithelium. The activity of these enzymes in the secondary lamellae is weak. The catalase and peroxidase show strong activities in the blood cells of the secondary lamellae. Various classes of GPs have been identified and characterized in the mucous secretions of the gill epithelium of C. chaca. These include—GPs with oxidizable vicinal diols, GPs with sialic acid residues without O-acyl substitution and GPs with O-sulphate esters. The functional significance of different enzymes in gill epithelium and the GPs in the mucus secreted on the surface has been discussed with the physiology of the gills in relation to the characteristic habit and habitat of the fish.  相似文献

Abstract

Tissue residues were measured in striped bass Morone saxatilis that had been injected intraperitoneally with 50 mg oxytetracycline (OTC)/kg or had been fed a ration containing 75 mg OTC/kg of fish daily for 10 d. Concentration of OTC in muscle of fish fed medicated feed was below the quantitation limit in 67% of the fish 12 d after treatment and was below the quantitation limit in all fish after 16 d at 23°C. The calculated terminal elimination rate constant (β) was 0.20/d (range, 0.15–0.37/d), and the half-life was 2.7 d (range, 1.9–4.5 d). The concentration of OTC in muscle after intraperitoneal injection was below the quantitation limit after 24 d at 23°C. The β-value was 0.34/d and half-life was 2.0 d in fish injected with OTC intraperitoneally.  相似文献

Abstract

Prebiotics recently have been shown to increase immune responses and disease resistance in certain fish species; therefore, the current study was conducted to evaluate the commercially available dairy–yeast prebiotic, GroBiotic-A, for use with juvenile goldfish Carassius auratus. The study consisted of two 10-week feeding trials in which juvenile goldfish were fed practical diets that were either unsupplemented or supplemented with the dairy–yeast prebiotic at 2% by dry weight. Juvenile fish were sorted by size and stocked into 12 units within each of two culture systems: one indoor system supplied with recirculated well water and one system located outdoors with a continuous flow of pond water to provide a source of phytoplankton and zooplankton. Both diets were fed to fish in six units within each system at the same fixed percentage of body weight twice daily. Culture system (i.e., presence or absence of phytoplankton and zooplankton) was the primary factor influencing (P < 0.0001) percent weight gain, feed efficiency, and survival of goldfish during the feeding trials. No dietary effect was detected, although there was a significant (P < 0.05) interaction between culture system and diet, with supplementation of the dairy–yeast prebiotic tending to improve weight gain and feed efficiency of fish in the presence of phytoplankton/zooplankton. During a controlled disease challenge with an intraperitoneally administered dose of Aeromonas hydrophila that was equivalent to a predetermined LD50 (dose lethal to 50% of test fish), average survival values ranged between 67% and 83% for fish that previously had access to phytoplankton/zooplankton compared with 17–33% for fish that had no access to phytoplankton/zooplankton. The dairy–yeast prebiotic, however, did not enhance resistance of goldfish to the bacterial pathogen and did not greatly alter microbiota of the anterior or posterior gastrointestinal tract based on denaturing gradient gel electrophoresis analysis. In conclusion, the dairy–yeast prebiotic did not improve feed efficiency in goldfish or resistance to a bacterial pathogen as previously observed in golden shiners Notemigonus crysoleucas and hybrid bass (white bass Morone chrysops × striped bass M. saxatilis).  相似文献

Farmed fish are usually exposed to routine procedures which have strong effects on stress responses. Rodlet cells may represent an useful biomarker for studies on the presence of stressors in aquaculture. This work focused on the localization of rodlet cells by light and electron microscopy in gills of sea bass (Dicentrarchus labrax) subjected to different conditions of overcrowding. In general, a significant increase in number of rodlet cells has been observed in all animals subjected to overcrowding stress. In gills of control group rare rodlet cells were detected at the level of both primary and secondary lamellae, whereas in stressed group clusters of rodlet cells have been found in the epithelium of primary and secondary lamellae indicating that these cells are influenced by stocking density.  相似文献

Abstract

Wild striped bass Morone saxatilis in Chesapeake Bay are experiencing a high prevalence of mycobacteriosis, which produces granulomatous lesions of the skin and visceral organs. Culture-based studies have indicated that the newly described species Mycobacterium shottsii and M. pseudoshottsii are the dominant isolates from diseased fish. The classical fish pathogen M. marinum is also found, albeit at much lower frequencies. Both M. shottsii and M. pseudoshottsii are extremely slow-growing on standard selective media, and up to 12 months may be required for isolation and characterization. Epidemiological studies of mycobacteriosis in Chesapeake Bay would therefore benefit from rapid molecular assays with which to detect these species in fish. In this paper, we describe the development of polymerase chain reaction–restriction fragment length polymorphism (PCR–RFLP) assays capable of detecting M. shottsii, M. pseudoshottsii, and, in most instances, coinfections thereof in striped bass tissues. In addition, PCR–RFLP assays were designed to detect M. marinum and other as-yet-undescribed Mycobacterium spp. present in Chesapeake Bay striped bass. Comparison of these molecular assays with culture-based techniques using splenic tissue from wild striped bass yielded generally concordant results and demonstrated the applicability of these techniques to the study of wild fish.  相似文献

Abstract

Proliferative gill disease (PGD), a condition not previously reported in wild fish, was found in two channel catfish Ictalurus punctatus sampled from the Tennessee-Tombigbee Waterway in Mississippi during June and July 1989. The parasite thought to cause PGD was observed in only one of the fish, but the distinctive lesions associated with this disease were prominent in both of the channel catfish. Organisms resembling the PGD parasite were also found in the gills of 4 of 18 largemouth bass Micropterus salmoides and 6 of 20 bluegills Lepomis macrochirus, but there was little or no host response to these parasites.  相似文献

Abstract

This study was conducted to investigate the effects of garlic and ginger oils on hematological and biochemical health characteristics of sea bass Dicentrarchus labrax. Fish were exposed to garlic oil (0.01 or 0.02 mL/L), ginger oil (0.01 or 0.02 mL/L), or a combination of the two oils (each oil at a concentration of 0.005 or 0.01 mL/L) for 96 h via bath immersion. Results showed that the red blood cell count, hematocrit (%), hemoglobin (Hb) concentration (g/dL), mean corpuscular volume (μm3), mean corpuscular Hb (pg), and mean corpuscular Hb concentration (%) were not significantly affected by herb oil exposure. However, some changes in biochemical variables were observed. Sea bass exposed to the 0.005-mL/L garlic oil–ginger oil mixture exhibited a significant increase in serum glucose. Serum total protein and albumin levels decreased in sea bass that were exposed to a garlic oil–ginger oil mixture (0.005 or 0.01 mL/L) or to garlic oil at 0.02 mL/L. Serum globulin levels decreased and triglyceride levels increased in sea bass exposed to 0.02-mL/L garlic oil or to the 0.01-mL/L mixture. The serum lipase level decreased and the cholesterol level increased in fish that were exposed to 0.02-mL/L garlic oil. In summary, ginger oil at 0.01–0.02 mL/L can be used without negative effects, while the garlic oil or garlic oil–ginger oil mixture should be applied at a concentration below 0.005 mL/L for bath immersion of sea bass. This is the first study to examine how garlic oil and ginger oil exposure via bath immersion affects the hematological and biochemical status of sea bass.

Abstract

The biflagellated, single-celled parasite Ichthyobodo necator can cause significant losses among fish populations, particularly those cultured in tanks. Treatments of KMnO4 and CuSO4 were evaluated against a naturally occurring I. necator infestation on sunshine bass (female white bass Morone chrysops × male striped bass M. saxatilis) raised in tanks. Four-hour static treatments with 3 mg of KMnO4/L of water (2.5 mg/L above the determined KMnO4 demand) or 2 mg of CuSO4/L of water (total alkalinity = 207 mg/L; total hardness = 95 mg/L) were randomly applied to 4 tanks/treatment (23 fish/tank); the same treatments were reapplied 2 d later. Four tanks were used as positive controls. By 2 d posttreatment (after the second treatment), only 17.4% of the untreated control fish survived, and a sample of the remaining fish was heavily infested with I. necator. All remaining control fish were dead by 5 d posttreatment. The KMnO4 treatment significantly curtailed the initial mortality (survival = 92.4%) and slightly reduced the high parasite loads at 2 d posttreatment. However, fish mortalities increased dramatically over the next 3 d (survival at 5 d posttreatment = 37.5%), and parasite loads from sampled fish remained high. The CuSO4 treatment was effective in significantly lowering the parasite load (almost eliminating I. necator) and maintaining a high fish survival (87.5%) by 5 d posttreatment. The findings in this study clearly demonstrate that CuSO4 is a viable treatment for ichthyobodosis in tanks.  相似文献

The fine structure of the principal cell types found in the gill filaments and secondary, respiratory lamellae of the hybrid striped bass (Morone saxatilis x M. chrysops) was studied by transmission electron microscopy. Most of the cell types generally resembled those observed at this level of resolution in other euryhyaline, teleostean species, and the mucus cells, epithelial cells, and cartilage cells were non-distinctive. Pillar cells were atypically flattened within the lamellae. The extensive cytoplasmic tubular system of the chloride cells was more densely distributed and was less branched than reported for many other teleosts. The vascular endothelial cells of the marginal, lamellar blood vessels were particularly striking because of their prominent, electron-dense cytoplasmic granules. Although the physiological function and chemical composition of these granules remain obscure, their presence suggests an important role of secretion into the vascular lumen. These gill data from normal specimens of this sport and commercial, hybrid species constitute a useful basis for interpreting gill changes in diseased striped bass.  相似文献

Abstract

The effectiveness of praziquantel (Droncit) against yellow grubs Clinostomum complanatum and unidentified, encysted, larval trematodes was tested in infected sunshine bass Morone chrysops female × M. saxatilis male. Praziquantel treatments significantly reduced the total number of live grubs in the fish in most treatment regimes tested. Treatment at 0.25 mg/L for 24 h was as effective as one at 8 mg/L for 8 h. Mature dead and dying metacercaria may have released toxins that killed host fish in some treatments.  相似文献

The aim of the present study was to describe the gill morphology of the gilthead sea bream and sea bass fishes that showed the same feeding habits. Our study focused on the gross morphological, scanning and light microscopical features of the gills, where a many differences in the gill arches and gill rakers surface in both species were explained. Our results marked that; there was an interbranchial median elevation appeared in sea bream. The gill arches length and the gaps between decreased medial wards in both species. The gill rakers had similar arrangement and varied number in both species. The first lateral rakers row was the longest one in sea bass. However the gill filaments appeared in double rows. Gill filaments were long at middle and short at extremities of gill arch in both species. Scanning electron microscopy revealed the surfaces of gill arch covered by a mosaic of pavement cells, varied sized pores of chloride cells and many pointed spines on the rakers ventral border in both species. Two types of taste buds marked on the gill arches and rakers in sea bass. Many lamellae appeared on filaments in both species.  相似文献
