The level of resistance of carnation ( Dianthus caryophyllus ) cultivars to wilt caused by Fusarium oxysporum f.sp. dianthi was compared in root-dip-inoculated plants grown in pots (filled with tuff or sandy soil) in a greenhouse and plants grown in a field where the soil was artificially infested with the fungus. In the field, wilt symptoms appeared first in susceptible and subsequently in resistant cultivars; none was immune. Variations in the level of resistance were expressed either by different percentages of wilted plants (i.e. disease incidence) or by delayed disease progress as compared to a susceptible cultivar. The range of disease severity in the field, ranked on a scale from 0 to 4, was highly and significantly correlated with the percentage of diseased plants. The greenhouse test was unreliable as a predictor of the degree of resistance observed in the field. Similar wilt levels in the greenhouse and the field were found only in susceptible cultivars.  相似文献

The response of carnation (Dianthus caryophyllus L.) cultivars toFusarium oxysporum f.sp.dianthi (F.o.d.) was evaluated in an artificially infested field from 1988/9 to 1991/2. Disease incidence was highly correlated with disease severity, indicating that disease incidence may be used to estimate the impact ofF.o.d. on the host. Based on the results, the following stepwise procedure was developed for characterizing the response of carnation cultivars toF.o.d. First, the general response of the tested cultivar was classified as resistant or susceptible on the basis of disease incidence values recorded 180 days after planting. Empirical analysis of the data revealed that a disease incidence level of 75% may be taken as a reliable cut-off point for separation of cultivars into the two groups. Within each group, cultivars were then subjected to a more explicit classification: in the resistant group the records of actual disease incidence were used for classification, while in the susceptible group the linear rearrangement of the disease progress curve was calculated according to the Gompertz function, and the value of the intercept was used for classification.Contribution No. 3539-E series, from the Agricultural Research Organization.  相似文献

The RAPD fingerprinting procedure was used in combination with pathogenicity assays on differential cultivars to characterize a representative collection of 72 Fusarium spp. isolates of different geographic origin collected from diseased carnation. In F. oxysporum f. sp. dianthi, isolates were grouped according to the physiologic race: group 1 included isolates of race 4; group 2 was formed by isolates of race 2 and single representatives of races 5 and 6; group 3 included isolates of races 1 and 8. No correlation was found between RAPD data and geographic origin of the isolates tested: representatives of race 2 isolated in Italy, Israel and Japan had the same amplification profile. Three isolates which showed a low level of pathogenicity on all carnation cultivars tested shared an identical amplification pattern and are probably saprophytic F. oxysporum. Finally, two F. redolens isolates from Japan and seven non-pathogenic isolates of F. proliferatum collected from diseased carnation in Italy, Israel and The Netherlands were clearly distinguishable according to their RAPD fingerprint. The results are discussed in relation to previous studies on the genetic diversity of F. oxysporum f. sp. dianthi and to the development of forma specialis- and pathotype-specific diagnostic tools.  相似文献

Fusarium wilt is now a major disease of carnation crops worldwide. Methyl bromide, which is used to remedy it, is environmentally unsafe. An alternative approach integrated into biological control is to grow crops in suppressive media. Suppressiveness of seven plant growth media to Fusarium oxysporum f. sp. dianthi was evaluated in bioassays with carnation (Dianthus cariophyllus) cv. Medea. These media were: (1) grape marc compost, (2) cork compost, (3) olive oil husk + cotton gin trash composted and mixed with rice husk, (4) spent mushroom compost mixed with peat, (5) coir fibre, (6) light peat and (7) vermiculite. In order to look for carnation Fusarium wilt suppressiveness indicators, growth medium pH and β-glucosidase activity were evaluated. Furthermore, F. oxysporum populations were measured in plant growth media at the beginning and end of bioassays. The compost media showed a range of suppressiveness in comparison with peat. Grape marc compost was the most effective plant growth medium in suppressing carnation Fusarium wilt. On the other hand coir fibre, peat and vermiculite were conducive for this disease. β-glucosidase activity and pH were positively correlated with disease severity as in other reports for tomato. Therefore, these two parameters are good indicators for carnation Fusarium wilt suppressiveness, and possibly for other F. oxysporum pathosystems. All composts showed similar F. oxysporum populations at the end of the bioassays to peat and vermiculite.  相似文献

ABSTRACT In order to elucidate the origin of Fusarium oxysporum f. sp. dianthi in Argentina, the genetic diversity among pathogenic isolates together with co-occurring nonpathogenic isolates on carnation was investigated. In all, 151 isolates of F. oxysporum were obtained from soils and carnation plants from several horticultural farms in Argentina. The isolates were characterized using vegetative compatibility group (VCG), intergenic spacer (IGS) typing, and pathogenicity tests on carnation. Seven reference strains of F. oxysporum f. sp. dianthi also were analyzed and assigned to six different IGS types and six VCGs. Twenty-two Argentinean isolates were pathogenic on carnation, had the same IGS type (50), and belonged to a single VCG (0021). The 129 remaining isolates were nonpathogenic on carnation and sorted into 23 IGS types and 97 VCGs. The same VCG never occurred in different IGS types. Our results suggest that the pathogen did not originate in the local populations of F. oxysporum but, rather, that it was introduced into Argentina. Given the genetic homogeneity within Argentinean isolates of F. oxysporum f. sp. dianthi, either IGS type or VCG can be used for the identification of the forma specialis dianthi currently in Argentina.  相似文献

A field experiment was conducted to assess the progress in time and spread in space of powdery mildew (caused by Erysiphe pisi ) in pea ( Pisum sativum ) cultivars differing in resistance to the disease. Disease severity (proportion of leaf area infected) was measured in 19 × 23 m plots of cultivars Pania and Bolero (both susceptible) and Quantum (quantitatively resistant). Inoculum on infected plants was introduced into the centre of each plot. Leaves (nodes) were divided into three groups within the canopy (lower, middle, upper) at each assessment because of the large range in disease severity vertically within the plants. Disease severity on leaves at upper nodes was less than 4% until the final assessment 35 days after inoculation. Exponential disease progress curves were fitted to disease severity data from leaves at middle nodes. The mean disease relative growth rate was greater on Quantum than on Pania or Bolero, but it was delayed, resulting in an overall lower disease severity on Quantum. Gompertz growth curves were fitted to disease progress on leaves at lower nodes. Disease progress on Quantum was delayed compared with Pania and Bolero. The average daily rates of increase in disease severity from Gompertz curves did not differ between the cultivars on these leaves. Disease gradients in the plots from the inoculum focus to 12 m were detected at early stages of the epidemic, but the effects of background inoculum inputs and the rate of disease progress meant that these gradients decreased with time as the disease epidemic intensified. Spread was rapid, and there were no statistically significant differences between cultivar isopathic rates (Pania 2.2, Quantum 2.9 and Bolero 4.0 m d−1).  相似文献

Fusarium oxysporum f.sp.dianthi causes severe wilting in carnation (Dianthus caryophyllus L.) worldwide. The pathogen is present in the soil profile in which carnation roots are distributed and may infect the plants at any time during the growing season. To minimize the losses induced by Fusarium wilt, growers use carnation cuttings free ofFusarium spp. and fumigate the soil with methyl bromide prior to planting. The severity of epidemics and the resulting losses are governed by the main and interacting effects of the three components of the disease syndrome: the host, the pathogen and the environment. Host variables include the type and the degree of cultivar resistance (i.e., complete, partial or tolerance); pathogen variables include the race, its virulence and infectivity, and the amount of initial inoculum; environmental variables include solar radiation intensity, photoperiod, temperature and the growth substrate. In the present review the information available on the effect of the host, the pathogen and the environment, and their interactions, on Fusarium wilt in carnation is summarized.  相似文献

Auxotrophic mutants were used to determine vegetative relatedness among isolates of Fusarium oxysporum f.sp. dianthi (F.o.d.) , the vascular wilt pathogen of carnation. At the first stage, different nitrate-non-utilizing (nit) mutants were produced from 11 isolates of F.o.d. collected in Israel. Complementation (heterokaryon) tests showed that all the isolates belonged to a single vegetative compatibility group (VCG), and two mutants were chosen as its testers. Additional isolates of Fusarium from carnation, collected during 1986-88, were analysed for pathogenicity and vegetative compatibility with the testers. A total of 170 Fusarium isolates, obtained from 42 cultivars at 40 sites, were tested. All the nit mutants of all the 132 pathogenic isolates formed heterokaryons with the testers, indicating that they belonged to the same VCG. None of the 38 non-pathogenic isolates was vegetatively compatible with the testers. The nit mutants retained pathogenicity to carnation. The F.o.d. testers were not compatible with testers of five other formae speciales of F. oxysporum. Thus, F.o.d. appears to constitute a distinct genetic population within the F. oxysporum complex.  相似文献

The skeleton of the carnation phytoanticipin, acetophenone, was detoxified by Fusarium oxysporum f.sp. dianthi , the main fungal parasite of carnation. This process consisted of the reduction to ethanol of the acetyl group, leading to the formation of phenylethanol, which has lower fungitoxic activity than the parent molecule. The conversion took place through the activity of an adaptive fungal oxidoreductase, which was NADH-dependent and was released by the fungus as two enzymatic forms within the culture substrate in the presence of acetophenone. Reduction was stereospecific and gave rise to only one of the two possible enantiomeric forms.  相似文献

DNA restriction fragment length polymorphisms (RFLPs) among 46 isolates of Fusarium oxysporum from Dianthus spp., representing the known range of pathogenicity in carnation, were determined using total DNA digested with the restriction enzyme Hind III and a previously described probe, D4. Distinct multiple band RFLP patterns were found, which delineated RFLP groups as follows: (i) F. oxysporum f.sp. dianthi races I and 8; (ii) F. oxysporum f.sp. dianthi races 2, 5 and 6; (iii) F. oxysporum f.sp. dianthi race 4; (iv) a recently described race of F. oxysporum f.sp. dianthi (wilt-causing isolates from D. caryophyllus formerly classified as F. redolens); (v) wilt-causing isolates from D. barbatus formerly classified as F. redolens and (vi), (vii) and (viii), three further recently described races of F. oxysporum f.sp. dianthi. Isolate groups derived from analysis of RFLPs were consistent with existing and recently described vegetative compatibility groups (VCGs) in F. oxysporum f.sp. dianthi , but not in all cases with races. Isolates of F. oxysporum and F. proliferatum not associated with wilt disease had simpler RFLP patterns (with one exception) that were not associated with VCGs.  相似文献

The incidence/severity of soil-borne plant diseases is often reduced when composts are used as growth media. However, much less information is available about the effects of composts on the development of foliar diseases. Here we studied the suppressive capacity of five composts (from olive marc-cotton gin trash, grape marc, cork, spent mushroom and municipal organic and yard wastes) as growth media against Botrytis cinerea disease in cucumber plants. We also examined the putative correlations of several biotic and abiotic factors involved in disease suppression. The suppressive capacity of the growth media was studied by comparing disease incidence/severity in plants grown in composts with that occurring in plants grown in commercial peats, which are conducive to most soilborne diseases. Correlations were made between the occurrence of disease and leaf nutrient status, as well as electrical conductivity (EC) and microbial activity (measured as β-glucosidase activity) in the growth media. Cucumber plants grown in the peats showed greater severity of B. cinerea during the bioassay than those grown in composts. Mo, Ca and Si content in leaves showed negative correlations with this disease. A negative correlation with disease severity was observed for EC and microbial activity in the growth media. The noticeable reduction in B. cinerea in plants grown in composts was related to the supply of specific chemical elements, a certain degree of salt stress, and the high microbial activity of composts.  相似文献

ABSTRACT Strains of the carnation wilt pathogen, Fusarium oxysporum f. sp. dianthi, can be distinguished by DNA fingerprint patterns, using the fungal transposable elements Fot1 and impala as probes for Southern hybridization. The DNA fingerprints correspond to three groups of F. oxysporum f. sp. dianthi strains: the first group includes isolates of races 1 and 8; the second group includes isolates of races 2, 5 and 6; and the third group includes isolates of race 4. Genomic DNAs flanking race-associated insertion sites of Fot1 (from races 1, 2, and 8) or impala (from race 4) were amplified by the inverse polymerase chain reaction (PCR) technique. These regions were cloned and sequenced, and three sets of primers overlapping the 3' or 5' end of the transposon and its genomic insertion were designed. Using fungal genomic DNA as template in PCR experiments, primer pairs generated amplification products of 295, 564 and 1,315 bp, corresponding to races 1 and 8; races 2, 5, and 6; and race 4, respectively. When multiplex PCR was performed with genomic DNA belonging to races 1 and 8, 2, or 4, single amplimers were generated, allowing clear race determination of the isolate tested. PCR was successfully performed on DNA extracted from susceptible carnation cv. Indios infected with isolates representative of races 1, 2, 4, and 8.  相似文献

Plum pox virus (PPV) causes serious damage in apricots grown in the Czech Republic and other countries where it is present. To study PPV resistance in apricot, three backcrosses between apricot cultivars or selection resistant to PPV and apricot cultivars or selections susceptible to PPV ('LE-3218' × 'Stark Early Orange', 'LE-3241' × 'Vestar' and 'LE-3246' × 'Vestar') were performed. The seedlings were repeatedly inoculated with PPV-M strain by an infected chip. The resistance of the plants was evaluated by visual inspection for symptoms and ELISA in three consecutive growth periods. The Chi-square (χ2) test was used to analyse the data. It was found that two independent dominant complementary genes conditioned PPV resistance in apricot in the case of the backcrosses. The mode of inheritance for resistance to PPV in cv. Harlayne was determined from F1 progenies. Resistance to PPV in 'Harlayne' was controlled by three independent complementary dominant genes. This knowledge will help us in effective planning of apricot breeding programmes.  相似文献

Powdery mildew and leaf rust caused large yield losses in spring barley grown near Christchurch, New-Zealand, in two seasons. Disease present during early growth stages was as damaging to yield as disease late in the season. Moderate leaf rust severities after anthesis were most damaging when combined with earlier mildew epidemics. Later growth did not compensate for reduced yield potential induced by early infection. This was attributed, at least in part, to an effect on leaf size, and therefore on green leaf area, at later growth stages. There was a closer relationship, by regression analysis, of yield to green leaf area than to disease severity in three cultivars.
The three cultivars. which differed in yield potential and disease resistance, were not equally sensitive to disease. It is proposed that high yielding cultivars may be the most sensitive to yield constraint by disease.  相似文献

Cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis ( Xam ) is a destructive disease occurring in most cassava growing-areas. Although Colombian isolates of Xam differ in DNA polymorphism and pathogenicity, no suitable host differentials have been identified to demonstrate physiological specialization. A set of 26 Xam isolates from three edaphoclimatic zones (ECZs) in Colombia was selected for inoculation on a set of 17 potential cassava differentials. Leaf inoculation and stem puncture were used in order to detect possible specific interactions between cultivars and isolates. Cultivar × isolate interaction was highly significant ( P  < 0·001) after stem inoculation, but not after leaf inoculation. The stem inoculation technique was selected as a method for resistance screening of cassava cultivars for bacterial blight resistance. A highly significant interaction was also detected when cultivar behaviour was rated as area under the disease progress curve (AUDPC) after stem inoculation. Different pathotypes were defined among the 26 isolates and differential cultivars were proposed to define the pathotypic composition of Xam populations in three ECZs in Colombia. The results should help to improve selection of sources of resistance to cassava bacterial blight.  相似文献

The effect of light duration on potato black dot was examined in plants grown under controlled conditions and in plantlets in tissue culture. A short-day (SD) photoperiod of 8 h light enhanced disease levels caused by Colletotrichum coccodes compared with a long-day (LD) photoperiod of 16 h light. Four cultivars (Agria, Cara, Désirée and Nicola) exhibited higher disease levels under SD conditions. Fungal colonization rates, level of sclerotia on roots, and crown rot symptoms were significantly higher under SD conditions in all cultivars. Parameters related to plant development (height and shoot weight) were reduced in plants grown under SD conditions in all cultivars tested, regardless of inoculation with the fungus. Similar results were also obtained with tissue-culture plantlets of cvs Cara, Désirée, Nicola and Alpha grown in an aseptic system under SD and LD conditions. Fungal colonization indices, level of sclerotia on roots, and crown rot symptoms on inoculated plantlets were significantly more severe in all cultivars tested when grown under SD conditions. Plant height and shoot weight of cvs Cara, Désirée and Nicola were significantly affected by both day length and inoculation with C. coccodes , with inoculated plantlets being shorter and smaller under SD conditions. These findings support the hypothesis that light duration plays an important role in C. coccodes infection, and may be one of the major factors responsible for the severe expression of black dot and yield losses observed in the Israeli autumn season, as may also occur in other potato-production areas in the Mediterranean region.  相似文献

The effects of two mixtures of resistant and susceptible apple cultivars on the development of scab caused by Venturia inequalis were observed in an experimental orchard over four years, initially for two years without fungicides against scab, and subsequently for two years with a moderate fungicide schedule. The row-by-row and within-row mixtures included a susceptible cultivar and a resistant cultivar in equal proportions. Without fungicides, the results showed a significant reduction of disease incidence over both years (7·3 to 21·3%), and severity in the second year (35·4%) in the within-row mixtures, compared to the monoculture of the susceptible cultivar. The best results were obtained when the within-row mixture was associated with moderate fungicide treatments; in this case the reduction in disease incidence reached 75·1% on leaves and 69·7% on fruits during the growth phase. The characteristics of the Venturia inaequalis / Malus  ×  domestica pathosystem and the results obtained in this experiment suggest a moderate but not negligible ability of cultivar mixtures for reducing epidemics of the disease.  相似文献

通过鉴定发现引起甘肃省康乃馨锈病的病原为石竹单胞柄锈菌(Uromyces dianthi Niessl.);分别采用孢子萌发法和田间喷雾法、灌根法对防治康乃馨锈病的药剂进行了室内筛选和田间药效试验.结果表明,12.5%烯唑醇可湿性粉剂、25%嘧菌酯悬浮剂等7种药剂对康乃馨锈病夏孢子的萌发抑制率均超过50%;对筛选出的7种药剂进一步进行田间药效试验,发现采用喷雾法时12.5%烯唑醇可湿性粉剂、25%嘧菌酯悬浮剂和10%苯醚甲环唑水分散粒剂三种药剂的防效高于其他几种药剂,分别达70.2%、68.3%和65.0%,而灌根法对康乃馨锈病的防效并不理想,最高防效仅为8.1%.  相似文献

Lettuce cultivars adapted to Californian growing conditions were screened for resistance to fusarium wilt caused by Fusarium oxysporum f.sp. lactucae in order to determine if differences in susceptibility among currently grown cultivars might contribute to management of this disease. Based on a preliminary evaluation of 46 cultivars, eight that were among the most resistant of their horticultural type (iceberg, romaine or leaf) were selected for further testing. The relative susceptibility of these cultivars was assessed by: (i) root-dip inoculation, (ii) sowing seeds into infested potting mix and (iii) transplanting seedlings into an infested field. Evaluations of disease severity showed that both methods (i) and (ii) produced cultivar rankings that were significantly correlated with rankings from field trials [method (iii)]. Two romaine and two leaf cultivars were highly resistant to fusarium wilt (mean disease severity rating of ≤1·3 on a 1–4 scale) under all test conditions. Other romaine and leaf cultivars, however, were highly susceptible in root–dip tests, so there was no consistent association between cultivar type and susceptibility to fusarium wilt. Likewise, there was considerable variation in susceptibility to wilt among iceberg cultivars, but all were significantly more susceptible than the most resistant romaine and leaf cultivars.  相似文献

Methods to assess light leaf spot ( Pyrenopeziza brassicae ) on winter oilseed rape cultivars were compared in laboratory, controlled-environment and field experiments. In controlled-environment experiments with seedling leaves inoculated at GS 1,4, the greatest differences in percentage area affected by P. brassicae sporulation were observed with inoculum concentrations of 4 × 103 or 4 × 104 spores mL−1, rather than 4 × 102 or 4 × 105 spores mL−1, but older leaves had begun to senesce before assessment, particularly where they were severely affected by P. brassicae . In winter oilseed rape field experiments, a severe light leaf spot epidemic developed in 2002/03 (inoculated, September/October rainfall 127·2 mm) but not in 2003/04 (uninoculated, September/October rainfall 40·7 mm). In-plot assessments discriminated between cultivars best in February/March in 2003 and June in 2004, but sometimes failed to detect plots with many infected plants (e.g. March/April 2004). Ranking of cultivar resistance differed between seedling experiments done under controlled-environment conditions and field experiments. The sensitivity of detection of P. brassicae DNA extracted from culture was greater using the PCR primer pair PbITSF/PbITSR than using primers Pb1/Pb2. P. brassicae was detected by PCR (PbITS primers) in leaves from controlled-environment experiments immediately and up to 14 days after inoculation, and in leaves sampled from field experiments 2 months before detection by visual assessment.  相似文献
