Insect bite hypersensitivity (IBH) is a seasonal allergic skin disease in horses caused by bites of certain Culicoides spp. The aim of our study was to investigate the maternal effect on IBH and to estimate the heritability and repeatability of IBH in the Dutch Friesian horse population. Data consisted of 3,453 Dutch Friesian broodmares with 3,763 visual observations on IBH clinical symptoms scored by 12 inspectors during organized foal inspections in 2004 and 2008. Nine percent of the mares (n = 310) were scored in both years. Mares descended from 144 sires and 2,554 dams and 26.2% of the dams (n = 669) had more than 1 offspring in the data set (range: 2 to 6). Insect bite hypersensitivity was analyzed as a binary trait with a threshold animal model with and without a maternal effect, using a Bayesian approach. Observed IBH prevalence in Dutch Friesian broodmare population was 18.2%. Heritability on the liability scale was 0.16 (SD = 0.06); heritability on the observed scale was 0.07; and repeatability was 0.89 (SD = 0.03). Maternal effect was 0.17 (SD = 0.06) and significantly differed from zero, although the animal model without a maternal effect fitted the data better. These results show that genetic and permanent environmental factors affect IBH in Dutch Friesian horses. The dam affected the IBH development of her offspring through an additive genetic influence but also by being part of their rearing environment.  相似文献

The aim of the study was to determine the situation of equine arteritis virus (EAV) infections in hucul horses. A total of 176 horses (154 mares and 22 stallions) from the biggest hucul horse stud in Poland were tested. Antibodies against EAV were detected in 97 (55.1%) horses. The EAV seroprevalence among mares was 53.2% while in stallions - 68.2%. The percentage of positive mares increased with their age, thus amongst the mares of less than 2 years of age the percentage was 32.5%, while in the group of 3-5 years old increased to 59.4% and in the mares in the age of 6-10 years and older than 10 years 89.5% and 95% were seropositive, respectively. Among 11 seropositive stallions five were supposed to be shedders of EAV with their semen. It is likely that those persistently infected stallions were the reservoirs of the virus in the stud. Genetic studies using of ORF5 gene showed high homology between the viruses detected in the semen of those stallions what suggested lateral transmission between the stallions sharing the same stable. Persistent infection in an immature stallion, which has not yet been used for breeding, was established as a result of infection via respiratory route. Phylogenetic analysis confirmed that all hucul viruses shared the same ancestor and as most of EAV strains dominating in Polish horse population belonged to the European origin EAV subgroup (EU-1).  相似文献

The horses studied were of the Swiss Warmblood breed and most were ELA-typed to assess a possible association of dermal hypersensitivity to insect bites with the major histocompatibility complex. Firstly, the occurrence of the condition was examined in 304 half-siblings sired by six stallions (A to F). Fourteen cases of dermal hypersensitivity were recognized and all were in the 153 offspring of Stallions C, E and F. Most animals of this group were also investigated for chronic hypersensitivity bronchitis: none of the sires displayed clinical signs of dermal hypersensitivity, but Stallions D, E and F were affected by chronic bronchitis. Among the animals investigated for both conditions only one horse showed coincidence of the two diseases as can be expected when the diseases are not correlated. The frequency of manifest dermal hypersensitivity and/or chronic hypersensitivity bronchitis varied in the half-sibling groups of individual sires. These findings suggest that the allergic conditions are independent entities. Secondly, the occurrence of dermal hypersensitivity was studied in three generations of horses at a stud at which Stallion C had exerted a particularly strong influence. A total of 302 animals, all born and raised at this stud, were surveyed over a period of 12 years. The descendants of Stallion C showed a significantly higher incidence (P less than 0.01) of dermal hypersensitivity (two daughters out of 19; eight second generation offspring out of 103; one third generation offspring out of 85) than did the controls of the same age classes but unrelated to Stallion C at the same stud (0 out of 95).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Insect bite hypersensitivity (IBH) is an allergic dermatitis of horses caused by IgE-mediated reactions to bites of insects of the genus Culicoides. IBH does not occur in Iceland due to the absence of Culicoides. However, Icelandic horses exported to mainland Europe as adults (1st generation) have a > or =50% incidence of developing IBH. In contrast, their progeny (2nd generation) has a <10% incidence of IBH. Here we show that peripheral blood mononuclear cells (PBMC) from Icelandic horses born in mainland Europe and belonging either to the IBH or healthy subgroup produce less interleukin (IL)-4 after polyclonal or allergen-specific stimulation when compared with counterparts from horses born in Iceland. We examined a role of IL-10 and transforming growth factor (TGF)-beta1 in down-regulation of IL-4 in healthy 2nd generation Icelandic horses. Supernatants of PBMC from 2nd generation healthy horses down-regulated the proportion of IL-4-producing cells and IL-4 production in stimulated cultures of PBMC from 1st generation IBH. This inhibition was mimicked by a combination of IL-10 and TGF-beta1 but not by the single cytokines. Cultures of stimulated PBMC of healthy 2nd generation horses produced a low level of IL-4, but IL-4 production was increased by anti-equine IL-10 and anti-human TGF-beta1. This shows for the first time that in horses, IL-10 and TGF-beta1 combined regulate IL-4 production in vitro. It is suggested that in this naturally occurring IgE-mediated allergy, IL-10 and TGF-beta1 have a role in the down-regulation of IL-4-induced allergen-specific Th2 cells, thereby reducing the incidence of IBH.  相似文献

REASONS FOR PERFORMING STUDY: Insect bite hypersensitivity (IBH) is an IgE-mediated allergic dermatitis caused by bites of Culicoides and Simulium species, and improved means of diagnosis are required. OBJECTIVES: The cellular antigen simulation test (CAST) with C. nubeculosus and S. vittatum extracts was assessed in a population of IBH-affected and healthy horses. Variations in test results over a one year period and possible cross-reactivity between different insect extracts was studied. METHODS: A total of 314 mature horses were studied using the CAST. Influence of severity of clinical signs, gender and age were evaluated, and 32 horses were tested repeatedly over one year. The kappa reliability test was used to assess agreement of the test results with different insect extracts. RESULTS: Horses with IBH had significantly higher sLT release than controls with C. nubeculosus and S. vittatum. The highest diagnostic sensitivity and specificity levels were attained when using adult C. nubeculosus extracts with the CAST (78% and 97%, respectively), suggesting that most horses with IBH are sensitised against Culicoides allergens. A proportion of IBH-affected horses was found to be sensitised to allergens of Simulium spp. in addition to those of C. nubeculosus. The CAST with C. nubeculosus had positive and negative predictive values > or = 80% for a true prevalence of IBH of 12-52%. In the follow-up study, the proportion of IBH-affected horses with a positive test result ranged from 90% in November to 68% in March. Severity of clinical signs or age did not influence test results significantly. However, IBH-affected males achieved significantly more positive test results than IBH-affected females. CONCLUSIONS: The CAST with adult C. nubeculosus has high specificity and good sensitivity for diagnosis of IBH. Horses with IBH are mainly sensitised to Culicoides allergens, and some horses are additionally also sensitised to allergens in Simulium spp. POTENTIAL RELEVANCE: The CAST is likely to be a useful test for diagnosis of IBH, even allowing the identification of IBH-affected but asymptomatic horses. This test may also help in further characterisation of allergens involved in this condition.  相似文献

Insect bite hypersensitivity (IBH) is a seasonal recurrent allergic reaction of horses to the bites of certain Culicoides spp. and is found throughout the world. The aim of our study was to estimate the heritability and repeatability of IBH in the Dutch Shetland pony population. A total of 7,924 IBH scores on 6,073 mares were collected during foal inspections in 2003, 2005, and 2006. Mares were scored for clinical symptoms of IBH from June until February by 16 inspectors. Of all mares, 74.4% (n = 4,520) had a single observation, 20.7% (n = 1,255) had 2 observations, and 4.9% (n = 298) had 3 observations in different years. The overall mean IBH prevalence was 8.8%. Heritability was 0.08 (SE = 0.02) on the observed binary scale and 0.24 (SE = 0.06) on the underlying continuous scale. Repeatability was 0.30 (SE = 0.02) and indicates that including repeated observations of the clinical symptoms of IBH will improve the accuracy of breeding values for IBH. We conclude that IBH, based on clinical symptoms, is a heritable trait in the Dutch Shetland pony population. Therefore, the IBH prevalence in this population can be decreased by selection.  相似文献

An Internet-based questionnaire among horse owners was carried out to identify factors affecting the incidence of insect bite hypersensitivity (IBHI) among horses in the Netherlands. Information was obtained for 794 horses of various breeds, but the breed distribution was not representative for the Dutch horse population. Of the horses for which information was available, 56% suffered from IBH and 44% did not. The most common clinical symptoms were pruritus, scaling, and hair loss, occurring mainly at the base of the tail and along the mane. Breed, age, region (and local habitat), stabling, type of bedding in the stable, use of the horse, deworming frequency, and season were associated with significant differences in IBH incidence. Knowledge of the factors influencing the incidence of IBH may make it possible to reduce the number of animals affected and help alleviate symptoms in affected horses.  相似文献

There is substantial international influence on the Swedish Warmblood (SWB) sport horse population. The SWB Association suspects that imported horses are pre-selected based on their anticipated performance level, which could lead to biased estimated breeding values (EBVs) for stallions. This study examined different strategies to handle records for imported horses in the genetic evaluation. The stallions were evaluated for 10 traits using 3 different EBV estimation methods that were compared based on ranking comparisons of stallions, accuracy, correlations and absolute differences between EBVs. The results showed that the stallions’ EBVs were affected by imported offspring which, as a group, had higher average scores than horses born in Sweden. Sire EBVs differed by up to 0.6 genetic standard deviation of the trait between methods for stallions with >50% imported offspring. Excluding imported offspring lowered the accuracies and caused larger re-ranking compared with including a fixed effect of origin in the model.  相似文献

The Franches-Montagnes is an indigenous Swiss horse breed, with approximately 2500 foalings per year. The stud book is closed, and no introgression from other horse breeds was conducted since 1998. Since 2006, breeding values for 43 different traits (conformation, performance and coat colour) are estimated with a best linear unbiased prediction (BLUP) multiple trait animal model. In this study, we evaluated the genetic diversity for the breeding population, considering the years from 2003 to 2008. Only horses with at least one progeny during that time span were included. Results were obtained based on pedigree information as well as from molecular markers. A series of software packages were screened to combine best the best linear unbiased prediction (BLUP) methodology with optimal genetic contribution theory. We looked for stallions with highest breeding values and lowest average relationship to the dam population. Breeding with such stallions is expected to lead to a selection gain, while lowering the future increase in inbreeding within the breed.  相似文献

The objective was to investigate whether it is possible to improve the quality of stallion semen, with respect to sperm morphology and chromatin integrity, both of which have been linked to fertility, using either density gradient centrifugation (DGC) or a new method, hereby named single layer centrifugation (SLC). The two methods of colloidal centrifugation were evaluated using 38 ejaculates from 10 stallions. Sperm morphology, subjective motility and sperm chromatin integrity were compared in uncentrifuged samples and in centrifuged sperm preparations. The proportion of morphologically normal spermatozoa varied between stallions (p < 0.001) and was increased by both methods of colloidal centrifugation (median value before centrifugation 67.5%; after SLC 78%; after DGC 77%; p < 0.001). The incidence of certain abnormalities was reduced, e.g. proximal cytoplasmic droplets were reduced from 12.9% to 8.8% (p < 0.001), and mid-piece defects from 5.3% to 1.4% (p < 0.05). Similarly, sperm motility and chromatin integrity were significantly improved (p < 0.001), with no difference between the two centrifugation methods. Centrifugation through colloids can enrich the proportions of stallion spermatozoa with normal morphology and normal chromatin structure in sperm preparations. The new method, SLC, was as effective as DGC in selecting motile stallion spermatozoa with normal morphology and intact chromatin. SLC, being simpler to use than DGC, would be appropriate for routine use by stud personnel to improve stallion sperm quality in insemination doses.  相似文献

The purpose of the present study was to evaluate the nature of ossification of the cartilages in the front feet of young, about 2-year-old Norwegian coldblooded horses, and to compare offspring of different sires in this respect. Dorsopalmar radiographs of the front feet of 392 horses (187 female and 205 male) were evaluated for ossification at the base of the cartilage and for separate centres of ossification. The horses were offspring of 45 different sires. Ossification extending above the navicular bone and separate centres of ossification were considered as significant. Minimal to mild ossification at the base of the cartilages was commonly seen, and significant ossification was present in one or more of the cartilages in 11.5% of the horses. The lateral compared to medial cartilages had more ossification and females had more ossification and more separate centres of ossification than males. The prevalence of horses with significant ossifications was significantly higher (46.3%) among offspring of one frequently used stallion than in the group consisting of offspring of 4 other popular stallions (3.5%) and in another group consisting of offspring of other, less frequently used stallions (9.6%). Ossification of the cartilages is considered to have a hereditary background in Norwegian coldblooded horses.  相似文献

SUMMARY Three Quarter Horse stallions and 5 of their 11 tested progeny were diagnosed as affected with the inherited autosomal dominant defect hyperkalaemic periodic paralysis in Victoria in 1992. The diagnoses were based on the appearance of clinical signs and associated increased plasma potassium concentrations in response to oral potassium loading. All affected horses were descendants of the American Quarter Horse Impressive. Indirect evidence indicates that at least 3 other affected Quarter Horse stallions have stood or are standing at stud in Australia. The clinical details of the affected horses are described and the size of the affected population in Australia is discussed.  相似文献

The stationary performance testing (SPT) of stallions as breeding candidates in the Dutch Warmblood riding horse population is evaluated. Genetic and phenotypic parameters of traits scored during SPT and the genetic correlation of these traits with performances in dressage and jumping competition from offspring of breeding stallions are estimated.Data from 1978–1988 are used, covering scores from 337 3-year-old stallions. Eight subjectively scored traits are considered. These traits are: walk; trot; canter; riding ability; show jumping; free jumping; cross country; character. SPT lasts for a period of 100 days. Data from SPT are analysed using an animal model. The relations between SPT of stallions and performances in jumping and dressage competition are analysed with an animal model for SPT data and a sire model for competition data. Variance and covariance components are estimated by restricted maximum likelihood (REML) procedures. Estimates of heritability are high (0.64) for gaits and riding ability, intermediate (0.41) for cross country and medium-high (0.31) for jumping. Estimated genetic correlation between show jumping scored during SPT and jumping in competition from offspring of breeding stallions is 0.84; for dressage this relation is 0.83. Some possible bias due to selection and the subjectivity of scoring is discussed. It is indicated that selection on SPT of stallions before entering breeding service is an effective tool to breed for ability of performance in competition.  相似文献

Gene contributions of foreign populations as well as coefficients of inbreeding and relationship were evaluated in 699 Black Forest Draught horses of Baden-Württemberg actually registered in the year 2002. Based on nearly complete 5-generation-pedigrees and after taking into account the remaining incompleteness, the mean coefficient of inbreeding for the total population was 6.5%. The recently by incrossing with different breeds newly established lines of stallions showed significantly lower mean coefficients of inbreeding. High rates of inbreeding of about 1.6% in the last five generations could also be faced by incrossing stallions of foreign coldblooded populations what resulted in a decrease of inbreeding in the last generation. In the total population the mean degree of relationship was 16%. The mean degree of relationships within lines of stallions was between 18.3 and 26.8%. The coefficients of relationships between lines of stallions varied between 5.1 and 16.6%. Especially, the newly established lines of stallions showed a lower mean degree of relationships to the other different lines of stallions. The proportion of purebred Black Forest Draught horses in the total population was nearly 70%. Assuming that most animals of unknown origin were purebred, the proportion of purebred Black Forest Draught horses reached about 90%. Austrian Noric, Swiss Freiberg and South German Coldblood stallions were the most important contributors to the Black Forest Draught horse population.  相似文献

Reproductive Performance of Thoroughbred Mares in Sweden   总被引:1,自引:0,他引:1
During 1997–2001, a total of 430 Thoroughbred mares were mated by one of the two Thoroughbred stallions on a well‐managed stud farm in the central part of Sweden. On arrival, a thorough gynaecological examination of all mares was performed. An early pregnancy examination was performed on days 14 and 15 after ovulation. The overall conception rates for the two stallions were 92.2 and 88.8%, and the corresponding live foal rates were 82.6 and 75.2%, respectively. The mean number of served oestrous cycles was 1.60 per mare per conception and 1.86 per live foal. The first and second cycle conception rates are reported for different age groups, mare categories and month of mating. The age of the mares had a significant influence on the live foal rate, being lower for mares >13 years of age. Resorption and abortion occurred in higher percentages among mares >8 years of age. The highest embryonic death occurred among mares with a history of being barren, or with having resorbed or aborted foetuses during the previous season. The month of mating had no significant influence on the first cycle conception rate or the second cycle conception rate. The frequency of twinning was 10.5% and the results of manual crushing of one of the conceptuses was successful in 88.9% of all cases, according to the ultrasound scanning 2 days later.  相似文献

A data set constituting a total of 310,109 Hanoverian warmblood horses was analyzed to ascertain the genetic variability, coefficients of inbreeding, and gene contributions of foreign populations. The reference population contained all Hanoverian horses born from 1980 to 2000. In addition, Hanoverian stallions born from 1980 to 1995 and Hanoverian breeding mares from the birth years 1980 to 1995 with registered foals were analyzed for the same genetic parameters. The average complete generation equivalent was approximately 8.43 for the reference population. The mean coefficient of inbreeding was 1.33, 1.19, and 1.29% for the reference population, stallions, and breeding mares, respectively. The effective number of founders was largest in stallions (364.3) and smallest in the reference population (244.9). The ratio between the effective number of founders and the effective number of ancestors was 3.15 for the reference population, 3.25 for the stallions, and 3.06 for the breeding mares. The effective population size in the Hanoverian warmblood reference population was 372.34. English Thoroughbreds contributed nearly 35% of the genes to the Hanoverian reference population and even slightly greater contributions (39%) to the stallions. Trakehner and Arab horses contributed approximately 8 and 2.7%, respectively, to the Hanoverian gene pool. The most important male ancestors were Aldermann I from the A/E line, Fling from the F/W line, and Absatz from the Trakehner line, whereas the breeding mare Costane had the greatest contribution to the reference population, stallions, and breeding mares. From 1996 onward, the stallions Weltmeyer and Donnerhall had the largest genetic impact on the Hanoverian horse population.  相似文献

The hypothesis that fowl adenovirus (FAV) and chicken anemia virus (CAV), transmitted vertically and simultaneously, induce the inclusion body hepatitis (IBH)/hydropericardium (HP) syndrome in progeny chickens was tested. Thus, 35-wk-old light brown layer breeders, showing absence of antibodies against FAV and variable titers against CAV, were intramuscularly singly infected with the FAV serotype 4 isolate 341 or dually infected with CAV (isolate 10343) and FAV. All hens (groups A [FAV alone], B [FAV + CAV], and C [noninfected]) were clinically healthy throughout the experimental period. Both infectious viruses FAV and CAV were isolated from progenies obtained as early as 5 days after infection of their breeders. Hematocrit, serum proteins, and aspartate-aminotransferase values showed a few statistical differences between the progeny groups. Most of these differences were detected in the progeny chickens of group B. However, almost all values met reference values for the species. The pathologic findings showed that progeny chickens obtained from both singly and dually infected breeders developed macroscopic and histopathologic changes of IBH/HP. The pathologic findings shown by progeny chickens of group A (FAV) were not expected because neither synergism nor prior immunodepression by CAV was concurrent. Chickens of group B (CAV + FAV) also developed IBH/HP. Although not many differences in the evaluated parameters between groups A and B were statistically significant, most pathologic findings of group B indicated a more severe manifestation of the disease. However, because FAV alone did reproduce the syndrome, the results shown by group B would not allow a definitive confirmation of the hypothesis that the association of FAV and CAV is necessary for the successful induction of the IBH/HP syndrome in chickens when transmitted vertically.  相似文献

Plasma concentrations of LH, FSH and testosterone are reported in stallions exhibiting a variety of reproductive problems. Stallions with poor libido were found to have low LH and FSH concentrations, while testosterone concentrations appeared normal. Stallions with good libido but experiencing ejaculatory disorders had normal concentrations of LH, FSH and testosterone. Older stallions experiencing a marked reduction in fertility had elevated FSH concentrations which were accompanied by increased LH concentrations in some cases, however, testosterone concentrations appeared normal in such stallions. Two young stallions which had experienced poor fertility (40 to 60% conception rates) from the beginning of their stud careers were found to have normal FSH and testosterone concentrations while LH concentrations were consistently low in one and normal in the other.  相似文献
