Shoot multiplication using seedling materials was achieved by subculture on Murashige and Skoog salts with Gamborg's B5 vitamins (MSB5) medium containing a combination of 5 M 6-benzyladenine (BA), 5 M thidiazuron (TDZ), and 1 M indole-3-butyric acid (IBA) with three green ash (Fraxinus pennsylvanica) clones, SD1009 (South Dakota origin), SD2002 (South Dakota origin), and KA2018 (Kansas origin). Shoots were rooted using in vitro and ex vitro methods. For in vitro rooting studies, elongated shoots were transferred to rooting plugs supplied with liquid MSB5 medium containing a 3×3 factorial arrangement of two different auxins, -naphthaleneacetic acid (NAA) and IBA, at three concentrations (0, 5, and 10 M). The most effective treatment for in vitro root number, root length, and shoot height was 5 M IBA. The three clones also were tested for ex vitro rooting using a quick dip in 1 mM NAA, 1 mM IBA, or control (no auxin). The maximal ex vitro rooting response occurred when shoot explants of the three clones were dipped in 1 mM IBA. Significant clonal differences were noted in response to in vitro and ex vitro rooting treatments. Rooted plantlets were acclimated to the greenhouse.  相似文献

Aims

This study aims to evaluate the effects of wood ash application on nutrient dynamics and soil properties of an acidic forest soil (Arenosol).

Methods

Treatments were loose and pelleted ash application (11?Mg?ha?1), alone or together with N fertiliser, and control treatment in a lysimeter experiment. Nutrient leaching was followed during a 2-year period and soil chemical and biological properties were evaluated at the end of the experiment.

Results

Wood ash increased leaching of total N, NH 4 + -N, base cations and P, mainly during the first months, the effect being more pronounced for the loose formulation. At the end of the study period, a positive effect on soil nutrient availability and soil acidity reduction was seen. The application of loose and pelleted ash alone decreased N leaching and increased N microbial biomass at the end of the experiment. The C dynamics was weakly affected.

Conclusion

Wood ash can be used to improve nutrient availability and balance nutrient exported by tree harvesting in acid forest soils, the effects at short-term being stronger for loose than for pelleted ash. However, their application should be carried out when vegetation is established to minimise nutrient losses at short-term and reduce the potential risk for water bodies. In N-limited soils, wood ash should be applied with N fertilisers to counteract N immobilisation.  相似文献

Chlorantraniliprole and novaluron, two reduced-risk insecticides (IRRs), are currently used to control codling moth (Cydia pomonella) in apple orchards in southeastern Canada. Reproduction of Harmonia axyridis (Pallas), an invasive ladybeetle, and the indigenous Coleomegilla maculata DeGeer, both important generalist predators, might be compromised after exposure of adults to these IRRs. The aim of the study was to compare the effects of chlorantraniliprole and novaluron on fecundity and fertility between the two ladybeetles in a laboratory setting. Reproductive mates were given chlorantraniliprole- or novaluron-treated prey for 24 h, and reproductive performance was followed during 15 days. Chlorantraniliprole did not affect reproduction of ladybeetles. However, novaluron-reduced fecundity, egg hatch, and fertility of the invasive ladybeetle H. axyridis, by 29, 43, and 78.7%, respectively, contrasting with no significant effect on the indigenous C. maculata. Additionally, H. axyridis oviposition pattern was altered by the same insecticide. This resulted in more egg masses containing fewer eggs, compared to the control. No such changes were detected for C. maculata. Chlorantraniliprole does not disturb reproduction of H. axyridis or C. maculata. Treatments of novaluron should have an important effect on populations of the invasive H. axyridis, and consequently on the impact of the invader on native ladybeetle species and on its dominance, and finally potentially on biocontrol services provided by the assemblage of ladybeetles.
