Neutrophils were purified from blood of dexamethasone-treated (0.04 mg/kg of body weight) and untreated calves. Cells were untreated (controls) or cultured in media containing 5 or 10 ng of bovine recombinant granulocyte-macrophage colony-stimulating factor (rbGM-CSF)/ml for 10 to 12 hours before being tested for various functions. Dexamethasone treatment of calves decreased luminol-dependent chemiluminescence, decreased phagocytosis of Pasteurella multocida and several Staphylococcus spp by various degrees, and decreased antibody-dependent cell-mediated cytotoxicity against bovine herpesvirus-infected cells by 26 to 32%. The percentage phagocytosis of coagulase-positive S aureus and S intermedius was higher than that of coagulase-negative S epidermidis for neutrophils from all calves. Culture of neutrophils with rbGM-CSF significantly increased (P less than 0.05) all of the aforementioned functions, compared with control neutrophils; however, rbGM-CSF-induced increases in function tended to be higher in neutrophils from dexamethasone-treated calves than in neutrophils from untreated calves.  相似文献

Twenty calves, heifers of the Holstein-Friesian breed and crossbreds with the Slovak Pied breed, were divided into two groups at the average age of 19 days. The trial group was kept outdoors in wooden hutches and the control group was housed in an insulated building. Blood was sampled at the age of 20, 33, 48 and 60 days at the outdoor temperatures of 3 degrees C, -2 degrees C, -5 degrees C and -8 degrees C. The calves kept in hutches where temperatures were always lower than in the insulated calf-house had the higher level of nonesterified fatty acids in all observations. The largest, highly significant difference was determined at the age of 60 days at the outdoor temperature of -8 degrees C (271 mumol/l vs. 224 mumol/l), and the significance of differences was also observed in the first and third blood samplings at the temperatures of 3 degrees C and -5 degrees C. The differences were highly significant in the first group between the first and fourth, and second and fourth samplings. In the calves kept in the insulated building the difference was significant between the first and fourth observation because the content of free fatty acids was also gradually increasing in this case (Tab. I). Glycaemia values were also higher in the calves kept in hutches (Tab. II). The most noticeable (significant) difference was determined at the age of 48 days at the outdoor temperature of -5 degrees C (4.3 mmol/l vs. 3.9 mmol/l). Significant differences within the group were recorded only in calves from the trial group kept in hutches. Insulin concentrations increased gradually with the older age of animals (Tab. II). At the age of 20 days the values were identical in fact in both groups and the highest concentrations were recorded at the age of 60 days. The differences between the groups were not significant, the largest difference was observed at the end of milk feeding period at the age of 60 days (19 microUI/l in calves from hutches and 15.6 microUI/l in calves from the insulated building). Triiodothyronine concentrations decreased from the starting values of 0.8 nmol/l and 0.76 nmol/l in both groups at the age of 33 days to the values of 0.61 and 0.62 nmol/l.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Two groups of four calves were exposed to a poly-disperse aerosol of 1.3 X 10(4) to 16 X 10(4) colony forming units (CFU) of nalidixic acid resistant Pasteurella haemolytica type A1 litre-1 of air with a mass median aerodynamic diameter of 2.6 +/- 0.8 microns. One group of calves was kept at 5 degrees C, 72 per cent relative humidity (RH) and the second was subjected to an abrupt change in climate directly after aerosol exposure from 5 degrees C, 75 per cent RH to 13 degrees C, 84 per cent RH. Clearance of the organism from the respiratory tract of the calves was monitored over the subsequent 23 hours by a method of tracheal and nasopharyngeal swabbing. Clearance measured at the trachea in all calves in both groups was not a continuous, uninterrupted process but exhibited a temporary decline between eight and 14 hours. Calves subjected to an abrupt change in climate after aerosol challenge had raised respiratory rates eight to 14 hours later and P haemolytica proliferated in the nasopharynx over the entire 23 hours. There was no apparent effect of climate on P haemolytica in the trachea. It is suggested that rapid growth of P haemolytica accompanying a change in climate may be an important aetiological factor in pneumonic pasteurellosis of calves.  相似文献

At the constant temperature of 25 degrees C and relative humidity (RH) of 84%, the average pre-oviposition period of Hyalomma lusitanicum was 47 days, the oviposition lasted an average of 26 days and the total egg production was an average of 6320 per female. At 16 degrees C the females did not lay eggs at all, but those which survived for 1 year and were transferred thereafter to 25 degrees C and 84% RH laid viable eggs. At 35 degrees C, the oviposition was identical at all levels of RH tested (25, 62 and 93%). At 25 degrees C, the pre-oviposition period was shorter at 93% RH, and the number of eggs laid was fewer at 25% RH. The eggs hatched in 32-40 days, the hatching percentage being lower in batches of eggs laid at the beginning and at the end of the oviposition period. The larval and nymphal moultings were not influenced by the type of host. As the temperature increased, the pre-moult period became shorter. The engorged larvae were more sensitive to the low RH than the engorged nymphs, whose moulting percentage was always greater than 72 in all regimes. Low temperature and high humidity had a favourable effect on the survival of unfed nymphs. The female-to-male ratio was 1:2. Hyalomma lusitanicum always behaved as a 3-host tick. The adults did not engorge on rabbits. The female ticks engorged on calves weighed an average 543 mg. Ticks maintained at 25 degrees C and 84% RH and engorged on calves completed the life cycle in 138-196 days, which does not include the period of chitinization of about 30 days. Functional activities (phagocytosis and killing) of neutrophil leucocytes (NL) and immunoglobulin G concentrations were evaluated in six healthy foals from birth to 6 months of age. Peripheral blood NL were reacted with Streptococcus equisimilis in 20 per cent pooled equine serum for 30, 60 and 90 mins and functional activities of NL were determined using a fluorochrome microassay. Values for foal NL function were compared with those of healthy adult horses (n = 28). Foal neutrophil function was influenced by age. Killing capacity of NL decreased, whereas phagocytic capacity increased, until 113 days of age, after which a reversal in trends became apparent. Immunoglobulin G concentrations changed significantly over time and were lowest at 29 to 56 days of age. All foal values for NL function fell within the range of normal values established for healthy adult horses.  相似文献

Forty-three calves of various age from the day of birth to the age of three months were subjected to the 2-hydroxyethylmethacrylate (HEMA) particle phagocytosis test. Significant differences in the phagocytic activity of blood leucocytes were recorded, all depending on age. New-born calves had very low phagocytic activity: only 16.5% of the leucocytes were found to be phagocytic, the phagocytic index being 0.39. A rapid increase in phagocytic activity was recorded during the subsequent days. At the age of three days 43% of the leucocytes of the calves were phagocytic. A decrease in phagocytic activity was found on the sixth to seventh day, and another increase was recorded only from the age of three to four weeks of age. No such changes in the development of phagocytic activity were observed in the calves reared without colostrum administration. Low values were maintained for a longer time after birth without the typical increase on the third to fifth day. Phagocytic activity increased in the zero-colostrum calves experimentally infected with rotavirus.  相似文献

Effects of source and washing of RBC on quantitative growth and hemolytic zone sizes of common bacterial pathogens of the bovine mammary gland were evaluated. Blood samples used to prepare the blood agar media were obtained from 10 adult dairy cows, 10 dairy calves, and 10 sheep. Hemolytic zone sizes produced by Staphylococcus aureus were significantly (P less than 0.01) larger on blood agar prepared with washed RBC than on blood agar prepared with nonwashed RBC, regardless of RBC source. With the exception of Corynebacterium bovis, growth of all bacteria was equivalent or significantly higher on medium prepared with washed RBC, compared with that on medium prepared with nonwashed RBC, regardless of RBC source. Significantly higher numbers of C bovis (P less than 0.01) and Streptococcus agalactiae (P less than 0.01) were isolated on medium prepared with washed cow RBC. Significantly higher numbers of Str uberis (P less than 0.01) and S aureus (P less than 0.05) were isolated on medium prepared with washed sheep RBC and washed calf RBC, respectively. Growth of Escherichia coli was not affected by the RBC source. Seemingly, RBC used in the preparation of medium should be washed. The source of RBC, as well as inter-animal variation, also should be considered in the quality control of medium.  相似文献

Little is known about the antimicrobial potency and specificity of polymorphonuclear leucocytes which actually appear at the sites of bacterial invasion in tissues. In the present work we have compared inflammatory leucocytes induced by intraperitoneal injection of casein in rabbits with autologous peripheral blood cells in killing Escherichia coli serotype 01 and Staphylococcus aureus 502A. The results indicate that inflammatory leucocytes differ significantly from their virgin blood ancestors. While the blood leucocytes were only able to suppress the growth of the gram-negative bacteria, autologous exudative cells killed more than 95% of the test organisms within 1 h of incubation at 37 degrees C. The enhanced microbicidal activity of the inflammatory cells however, was only specific for the gram-negative bacteria, as evidenced by the failure of leucocytes to kill Staph. aureus to the same extent as the peripheral blood cells. In association with the enhanced gram-negative microbicidal activity the inflammatory cells produced chemiluminescence and released two to three times more O2-anions than the peripheral cells. We interpret these observations to mean that chemotactic factors such as casein activate inflammatory cells to increase their oxidative metabolism. Since microbicidal action of leucocytes is thought to proceed in part through oxygen-dependent reactions, the inflammatory leucocytes would be expected to effectively kill bacteria that are highly susceptible to these lethal oxygen metabolites. It cannot therefore be assumed that assessment of the functional capacity of the virgin peripheral blood PMNs would provide information on the functional characteristics of activated leucocytes which actually migrate to and accumulate at inflammatory sites.  相似文献

The time course of phagocytosis and intracellular killing of serum-opsonized Escherichia coli K12 and Staphylococcus aureus SG511 by glass-adherent bovine peripheral blood polymorphonuclear leukocytes (PMNLs) and cultured monocytes (macrophages) was monitored by fluorescence microscopy of single cells using the acridine orange (AO)/crystal violet (CV) technique. After interaction of glass-adherent leukocytes (20, 40, 60 min, 37 degrees C) with opsonized bacteria, cells were stained with the fluorescent dye AO. Living bacteria stained green, dead bacteria stained orange. The addition of CV to AO-stained bacteria quenched the fluorescence of extracellular bacteria only. CV does not penetrate living bovine PMNLs which allows the discrimination of ingested (fluorescent) and extracellular (nonfluorescent) bacteria during attachment and phagocytosis of bacteria by adherent PMNLs. We investigated quantitatively phagocytosis and intracellular killing of serum-opsonized bacteria by bovine PMNLs from 22 bulls of 4 different Swiss dairy breeds. Within 60 min maximum uptake (approximately 12 bacteria/PMNL) and killing (approximately 80%) of serum-opsonized Escherichia coli K12 and Staphylococcus aureus SG511 were achieved. The AO/CV technique was also used to quantify the uptake and intracellular killing of serum-opsonized Escherichia coli K12 by cultured monocytes (macrophages). Within 60 min maximum uptake of bacteria (approximately 16/MO) was achieved; approximately 83% of bacteria were killed.  相似文献

Bovine adenovirus type-3 infection in feedlot calves   总被引:1,自引:0,他引:1  
The frequency of bovine adenovirus type-3 (BA3) infection in 84 of 165 light feeder calves and the possible role of BA3 in inducing febrile disease and affecting weight gain performance were studied for 56 days after the calves entered a feedlot. Samples of blood for serotesting were obtained periodically, and samples for virus isolation were collected on days 28 and 56. At the time the calves entered the feedlot, 17.8% (15 of 84) had serum virus-neutralizing antibodies to BA3. During 56 days, 54.8% (46 of 84) seroconverted to BA3. Of these seroconversions, 87% took place during the first 28 days. Bovine adenovirus type-3 was isolated from 2 calves (1 isolate each on days 28 and 56). Pyrexia (greater than or equal to 39.2 C) was observed more often in BA3-infected calves than in noninfected calves. However, there was no significant (P less than 0.05) difference in weight gain between the BA3-infected and noninfected calves.  相似文献

Leukocytosis (34,600 WBC/microliter of blood) was detected in an apparently healthy 7-day-old Holstein heifer. Analysis of blood samples obtained over the next 41 days revealed chronic progressive neutrophilia, which peaked at greater than 85% neutrophils and exceeded 100,000 WBC/microliter. In vitro assessment of isolated blood neutrophils obtained from the heifer at 38 and 45 days of age revealed selected functional abnormalities. Endocytosis of immunoglobulin-opsonized Staphylococcus aureus and killing of this test organism by the calf's neutrophils were significantly diminished, as were phagocytosis-associated superoxide generation, chemiluminescence activity, and myeloperoxidase-catalyzed iodination. Diminished H2O2 elaboration by the calf's neutrophils was evident during ingestion of opsonized zymosan or on exposure to phorbol myristate acetate. Extracellular release (secretion) of elastase during ingestion of zymosan was also diminished, although total cell content of elastase was normal, compared with that of neutrophils from age-matched calves, and granular or other morphologic abnormalities of the calf's neutrophils were not evident by ultrastructural examination. Abnormalities of random migration were inconsistently detected, and normal or high degree of antibody-dependent cytotoxicity or natural killing by the calf's neutrophils was observed. Similar in vitro assessment of neutrophils obtained from the calf's dam revealed no functional abnormalities. The calf died at 48 days of age, with persistent fever and chronic diarrhea, despite administration of antibiotics. Histologic examination at necropsy revealed large numbers of intravascular neutrophils in most tissues, including massive neutrophil sequestration in spleen. However, a striking lack of extravascular neutrophils was evident in inflamed submucosa adjacent to intestinal ulcers heavily contaminated with enteric microorganisms. Bone marrow examination revealed diffuse myeloid hyperplasia, but no other abnormalities.  相似文献

Nasopharyngeal swabs were taken from four groups of veal calves at intervals throughout their growth and the aerobic bacteria cultivated from the swabs counted. The calves were kept under three different husbandry systems; naturally ventilated straw-yards, fan-ventilated crates and crates with a controlled climate. The numbers of bacteria isolated varied in a complex manner; however, in one group of calves a significant proportion (P less than 0.01) of the variation in weekly bacterial counts was associated with the changes in vapour pressure and temperature which took place between two and four days previously. In calves kept at a constant temperature of 16 degrees C, the bacterial populations in the nasopharynx were at a minimum between 65 and 75 per cent relative humidity and tended to rise at humidities outside this range. There was a temporal relationship between nasopharyngeal bacteria and lung lesions. In three groups the numbers of bacteria in calves at nine weeks old were positively correlated (P less than 0.05) with lung damage observable at 16 to 18 weeks old.  相似文献

Phagocytic and oxidative metabolic activities of bovine blood neutrophils were determined in the presence of glycolytic (NaF) and cytoskeletal (colchicine, cytochalasin B, and prostaglandin E1) inhibitors. Phagocytosis and post-phagocytic oxidative metabolic activity, measured by nitroblue tetrazolium reduction, were determined using zymosan, Escherichia coli, Staphylococcus aureus, or Streptococcus agalactiae. Sodium fluoride (1.25 microM to 1.25 mM concentrations) did not significantly (P greater than 0.05) inhibit phagocytosis of S aureus and Str agalactiae, whereas phagocytosis of zymosan and E coli was significantly (P less than 0.05) inhibited only at 1.25 mM concentration. Colchicine at 1.25 nM to 1.25 microM concentrations significantly inhibited phagocytosis of zymosan and E coli, but not of S aureus and Str agalactiae. Cytochalasin B at 1.25 nM to 1.25 microM concentrations significantly inhibited phagocytosis of zymosan and all 3 bacteria, whereas prostaglandin E1 was noninhibitory at similar concentrations. Nitroblue tetrazolium reduction, in general, was not significantly affected by NaF and cytoskeletal inhibitors.  相似文献

The influence of supplemental phenothiazine (P) on growth and physiological criteria was studied in parasite-controlled calves consuming endophyte (Acremonium coenophialum)-infected tall fescue (TF). In Exp. 1, nine Angus heifer calves (312 kg) were supplemented with 227 g corn-mineral (CM) mix twice daily and allowed ad libitum access to either high-endophyte (HE) G1-307 (greater than 90% infected) or low-endophyte (LE) Kenhy (less than 1% infected) tall fescue hay, or HE G1-307 plus 2 g/d P in the daily supplement. Calves were kept in temperature-controlled rooms for 12 d at 21 degrees C followed by 7 d at 34 degrees C. In Exp. 2, 48 Angus steer calves (312 kg) were assigned to treatment groups consisting of calves grazing HE Kentucky-31 (57% infected) or LE Johnstone (less than 1% infected) TF, and supplemented daily with either .91 kg of a control CM mix or .91 kg of the CM mix containing 2 g P. The 112-d experiment was initiated on May 4 with BW and rectal temperature (RT) measurements and blood collected at 28-d intervals. In both experiments, calves receiving HE TF had lower (P less than .01) serum prolactin concentrations (PRL) at elevated ambient temperature and lower (P less than .01) serum alkaline phosphatase activities (AP) but higher (P less than .01) RT than calves consuming LE TF regardless of ambient temperature.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

As a feed additive, ascorbic acid has been shown to have a protective effect against bacterial and viral diseases and to reduce the impact of detrimental stress in chickens. This study examined the effect of ascorbic acid treatment on in vitro heterophil function by examining random migration and phagocytosis and bacterial killing of Staphylococcus aureus. Heterophils were evaluated in broiler chickens ranging from 5 to 16 wk of age, and age differences were seen. Significant increases in bacterial killing were found in heterophils treated with ascorbic acid, and this difference tended to be greater in chickens from 5 to 10.5 wk of age. No significant differences were found in phagocytosis or random migration, but ascorbic acid tended to decrease random migration. The most significant effect on in vitro heterophil function was an increase in bacterial killing.  相似文献

To examine the effect of parity on polymorphonuclear neutrophils (PMN) function, phagocytic and bactericidal activity of the PMN isolated from blood and milk against Staphylococcus aureus was compared between groups of 6 primiparous and 6 multiparous healthy dairy cows during early lactation using bacteriological and PMN-pathogen interaction assays. Latex-stimulated luminol-amplified chemiluminescence (CL) and viability of these PMN were also investigated. The phagocytosis and killing of S. aureus by blood were remarkably higher than those of milk PMN. Similarly, the CL and viability in blood PMN were markedly higher than in milk PMN. Both in blood and in milk the phagocytosis of S. aureus by PMN in primiparous cows was substantially higher than in multiparous cows. The killing activity of blood PMN against S. aureus was 42.3+/-3.4% and 23.2+/-1.7% in primiparous and multiparous, respectively. Milk PMN killed only 20.7+/-2% S. aureus in primiparous and 10.2+/-1.3% in multiparous cows. Blood and milk PMN CL and milk PMN viability were significantly higher in primiparous cows. The pronounced reduction in phagocytic and bactericidal activity in blood and milk-resident PMN from multiparous cows, in part, resulted from the pronounced decrease of PMN viability and free radicals production capacity; this suggests that heifers' udders could be more protected against S. aureus, which remains to be tested in the field.  相似文献

Seventeen newborn Holstein-Friesian bull calves were cold-stressed by total body immersion in water at 15 to 17 degrees C until the core body temperature was lowered by 10 degrees C. Nine additional calves (noncold-stressed) were immersed in water at 35 to 37 degrees C. Eight of the cold-stressed calves were euthanatized soon after removal from the water while the others (n=9) were allowed to recover in a cold room at 4 degrees C for 72 hours. Noncold-stressed calves were kept at 25 degrees C for 72 hours. Sympathoadrenal an adrenal hormonal responses of calves were determined by analysis of plasma for glucose, corticosteroids, and catecholamines. Plasma concentration of glucose and corticosteroids rapidly increased in cold-stressed calves soon after immersion and remained higher (P less than of equal to 0.05) than concentrations in noncold-stressed calves during immersion and most of recovery. There was a threefold increase (P less than or equal to 0.05) in concentration of catecholamines in plasma of cold-stressed calves and only a slight increase in noncold-stressed calves during immersion. Catecholamine concentrations remained elevated in cold-stressed calves during most of recovery. Results provide direct evidence that sympathoadrenal and adrenal hormonal responses to cold are well developed in newborn calves and that changes in concentrations of glucose, corticosteroids, and catecholamines in plasma of these animals are sensitive indicators of their ability to respond to cold stress.  相似文献

Six calves, aged 24 to 58 days and not previously exposed to bovine viral diarrhoea virus (BVDV), were infected with this agent by nose-to-nose contact with a persistently BVDV viraemic calf. The study was conducted in two trials, using 3 calves in each. All 6 calves showed a peak interferon level in serum at 4 days post infection (dpi), and they seroconverted to BVDV at 16-21 dpi. The calves in trial 1 had diarrhoea for 2 or 3 days between 2 and 6 dpi and one calf again from 9 to 11 dpi. During the periods of fever, the calves were slightly depressed. Those in trial 2 were more depressed and their oral and nasal mucous membranes were reddened but they never had diarrhoea. In both trials, fever (up to 41.3 degrees C) was a prominent symptom at 8 to 9 dpi and 2 calves showed a diphasic fever course. Respiratory affection was mild and no medical treatment was required. Haematological assessment demonstrated a transient but significant leukopenia and lymphopenia at 4 dpi (P less than 0.01 and P less than 0.05 respectively) and 11 dpi (P less than 0.05 and P less than 0.01 respectively). A significant decrease in thrombocyte count was seen at 4 dpi (P less than 0.05, n = 3). This study has demonstrated that nose-to-nose contact is an effective way of transmitting BVDV from persistently infected to susceptible cattle.  相似文献

Pneumonia caused by Pasteurella (Mannheimia) haemolytica was induced in weaned beef heifer calves, approximately 6 months of age. Calves were treated at 20 h after challenge with therapeutic doses of danofloxacin or tilmicosin. Peripheral blood neutrophils were collected at 3, 24 and 48 h after treatment. The ex vivo effects on neutrophil function, neutrophil apoptosis, and hematological parameters were examined, as was the effect on percentage lung consolidation. Neutrophil function assays included random migration under agarose, cytochrome C reduction, iodination, Staphylococcus aureus ingestion, chemotaxis, and antibody-dependent and antibody-independent cell-mediated cytotoxicity. Apoptosis was determined using a cell death detection kit. Killing was performed at 72 h after treatment. Statistical comparisons were made among the three groups of challenged-treated animals: saline, danofloxacin, and tilmicosin. Comparisons were also made between nonchallenged nontreated animals (NCH) and challenged saline-treated animals. There were no significant differences for any of the neutrophil function assays or neutrophil apoptosis among the challenged-treated groups. This suggests that danofloxacin and tilmicosin have no clinically significant effects on neutrophil function or apoptosis. There were also no significant differences in percentage lung consolidation among the challenged-treated groups. Significant differences were found between the NCH calves and the challenged saline-treated calves in several neutrophil assays, which were attributed to effects of P. haemolytica infection.  相似文献

BACKGROUND: Although adequate colostrum intake and properly used antibiotics can provide much protection for the bovine neonate, increased antibiotic scrutiny and consumer demand for organic products have prompted investigations of natural immunomodulators for enhancing calf health. One plant-based immunomodulator, Morinda citrifolia (noni) fruit, is a well-recognized natural product that has a broad range of immunomodulatory effects. HYPOTHESIS: Neonatal calves fed noni puree would demonstrate whole blood phagocytic capacity in Gram-negative and Gram-positive in vitro assays. ANIMALS: Blood samples from 18 neonatal Holstein bull calves. METHODS: Calves were divided into 2 groups: Group 1 comprised control calves, whereas Group 2 received 30 mL of noni puree twice a day in milk replacer. Day 0 blood samples were obtained between 36 and 48 hours of age before the first feeding of puree. Ethylenediaminetetraacetic acid anticoagulated blood was collected from each calf on days 0, 3, 7, and 14. Bactericidal assays were performed to estimate the percentage killing of Escherichia coli and Staphylococcus epidermidis. RESULTS: Blood samples from noni puree-fed calves displayed significantly more E. coli bacterial killing than did controls on day 14, and although differences were not significant on days 0, 3, and 7, bacterial killing progressively increased over time. There was no significant difference between the groups for S. epidermidis killing. CONCLUSIONS AND CLINICAL IMPORTANCE: The immunomodulatory effect of noni puree may prove valuable in the future as production animal antibiotic use becomes more restricted. Additional clinical trials are warranted to investigate the clinical application of noni puree in promoting calf health.  相似文献
