Summary

The course of post‐mortem breakdown of glycogen and ATP in turkey pectoralis major muscle was markedly influenced by several ante‐ and peri‐mortem variables. Application of a proper stunning procedure was highly effective in preventing peri‐ and post‐mortem muscle stress reactions.

The physiological level of glycogen and ATP was not significantly affected by road transportation covering 260 km. Birds which rested for 24 hrs following transportation had lower glycogen and ATP levels at the moment of slaughter than non‐rested birds.

According to the changes in the rate and extent of post‐mortem biochemical reactions, several meat characteristics such as water‐holding capacity, colour, and tenderness were significantly changed.

Furthermore, the results also indicate that turkey breast muscle is susceptible to a PSE‐like condition as described in pork.  相似文献

1. A standard (FG, fast-growing), a black local or 'label', type (SG, slow-growing) turkey line, and the crossbreed between these two lines were compared for muscle post-mortem metabolism and related meat quality traits. 2. Ninety male turkeys (30 of each genetic type) were raised under the same experimental conditions until slaughter at 16 weeks of age. 3. Live weights at 16 weeks of age differed significantly (7.8, 6.0 and 4.2 kg, for the FG, crossbred and SG lines, respectively). Collagen content of Pectoralis superficialis (PS) muscle was higher in SG birds than in the other two types. 4. The rate of post-mortem glycogen depletion and lactate accumulation in PS and Ilio tibialis (IT) muscles were similar in the 3 lines, as were the rate and extent of post-mortem pH fall in PS muscle. In IT muscle, however, SG birds showed a slight but significantly faster pH decline. 5. Colour measurements indicated a paler breast muscle and a higher degree of myoglobin oxidation in SG birds at 24 h post mortem, than in both other lines. But these differences had disappeared after 4 and 7 d post mortem 6. SG birds showed higher drip loss and instrumentally-assessed toughness in breast muscle, compared with crossbred and FG birds. FG birds, however, had the lowest yield of breast meat after curing-cooking. 7. No marked differences in post-mortem metabolism were found between the three lines. However, differences in water-holding capacity of fresh and cured-cooked meat suggest that factors other than the rate and extent of post-mortem pH fall may contribute to the respective characteristics of these lines.  相似文献

Treadmill training of pigs was shown to retard the pH-drop of longissimus dorsi biopsy samples. Training similarly had an inhibiting effect on the post-mortem pH, ATP and glycogen, and on fluid loss (expressible juice) of the gracilis muscle.  相似文献

为探讨复方丁氨丙磷溶液对动物耐力和能量代谢的影响。采用小白鼠耐寒、耐热、耐缺氧和负荷游泳试验以及用经典蒽酮显色法和反相高效液相色谱法分别测定肝脏和骨骼肌中的糖原含量和3种磷酸腺苷含量,观察复方丁氨丙磷溶液对小白鼠耐力和能量代谢的影响。结果表明:复方丁氨丙磷溶液能明显提高小白鼠耐寒、耐热和耐缺氧能力。明显延长小白鼠负荷游泳时间;能显著提高肝糖原和肌糖原含量以及肝组织和肌肉组织的三磷酸腺苷(ATP)、二磷酸腺苷(ADP)含量。而对肝组织一磷酸腺苷(AMP)含量无明显影响,对肌肉组织一磷酸腺苷含量却有一定的降低作用。说明复方丁氨丙磷溶液是通过提高肝糖原和肌糖原储备以及促进ATP和ADP的合成代谢来增强动物的耐力和能量代谢。  相似文献

1. Early post-mortem pH was used to identify PSE-like turkey meat, characterised by a low water-holding capacity and pale colour. In a commercial turkey processing facility, the pH value of turkey breast muscle was measured at 0, 9, 10, 14, 154, 231, and 246 min post mortem. 2. At 14 min post mortem, the carcases were separated into 3 pH classes: low (<5.70), medium (5.70 to 6.18), and high (>6.18). Low pH carcasses (<5.70) had higher drip loss(1.75%) than medium (0.81%) and high pH carcases (0.75%). Furthermore, high pH carcases (>6.18) had a lower L* value (darker) than low and medium pH carcases. 3. The effects of using CO2 'snow' on turkey breast muscle quality were also investigated. Light CO2 'snow' treatment (57 g snow/kg breast) and heavy CO2 treatment (113 g snow/kg breast) resulted in higher drip losses than the control no CO2 snow) group.  相似文献

Furazolidone (FZ) at 700 ppm was added to feed mixtures fed turkey poults two weeks posthatching to induce acute experimental cardiomyopathy. Poults in the control pen received the same ration but without FZ. Four of the control poults developed spontaneous round heart disease. From EKG data and blood samples obtained at weekly intervals, poults were selected for sacrifice at 5 weeks of age. Tissue samples from the left myocardial wall, liver, and pectoralis major and tibialis anterior muscles were analyzed for glycogen by biochemical assay. Blood glucose was determined with the Technicon autoanalyzer. Deposition of glycogen increased significantly (p less than 0.05) in the myocardium of all affected poults and in the liver of all FZ-treated poults. Glycogen levels of the pectoralis major and tibialis anterior muscles were not affected by FZ, but a significant increase (p less than 0.05) was apparent in the pectoralis major muscle of spontaneous round heart poults. It was concluded that FZ influences glycogen metabolism, probably by enzyme inhibition, and that it tends to magnify effects seen in the spontaneous round heart syndrome. Glycogen infiltration of tissues such as the heart and white skeletal muscle suggests that the round heart syndrome may be a manifestation of the glycogen storage disease, idiopathic generalized glycogenosis. Lack of significant differences in the blood serum glucose levels of all poults indicates that these levels are not a reliable clinical parameter for monitoring development of the round heart syndrome.  相似文献

Forty-two pigs (German Landrace and commercial hybrids) were subjected to harpoon biopsy. A possibility of using the biochemical (adenosine triphosphate, creatine phosphate, glucose-6-phosphate, glucose, lactate, glycogen) and biophysical (pH value, R-value, water-binding capacity) parameters of muscle (m. longissimus dorsi) for forecasting the quality of pork was demonstrated. The post-mortem processes were simulated in muscle bioptate (1 hour of incubation at 39 degrees C). The highest correlations with meat quality in slaughtered animals (pH) were recorded in adenosine triphosphate, creatine phosphate, R-value, pH value, lactate and glycogen. No significant differences were found between the left and right side of the muscle (m. longissimus dorsi) in the studied parameters.  相似文献

Small samples of liver and m. adductor (AD) were collected within 45 minutes of death from the carcasses of a total of 604 pigs killed at three bacon factories. Glycogen concentrations were measured and part of the muscle sample allowed to complete post-mortem glycolysis in order to estimate ultimate pH (pHu) values. Liver glycogen levels were also used to predict overall food withdrawal times. Muscle glycogen concentrations ranged from 2.2 to 15.3 mg/g, liver glycogen from 0.01 to 50.7 mg/g and pHu in the AD from 5.51 t 6.76. The overall average predicted fasting time was 16.5 +/- 0.54 (SEM) hours and 22% of pigs had pHu values in the AD greater than 6.2 indicative of potentially dark, firm, dry meat. Muscle glycogen concentration was positively correlated with liver glycogen (r = 0.27, P less than 0.01) and negatively correlated with predicted fasting time (r = -0.30, P less than 0.001). The pHu in the AD was negatively correlated with liver glycogen (r = -0.34, P less than 0.001) and positively correlated with predicted fasting time (r = 0.37, P less than 0.001). Therefore, pigs which had reduced concentrations of glycogen in their livers, indicative of longer food withdrawal times before slaughter, tended to have less glycogen in their muscles and a higher pHu in the meat.  相似文献

To assess muscle breakdown during avian coccidiosis, the level of the non-metabolizable amino acid 3-methylhistidine (3MH) was determined in muscle and plasma from turkey poults that received an infection with a field isolate containing a mixture of Eimeria species. The effect of increased levels of parasitism was evaluated at 6 days postinoculation (DPI) in birds receiving 2.5 x 10(4), 1 x 10(5), or 2 x 10(5) oocysts each. The changes in 3MH levels during recovery from acute infection were assessed at 6-29 DPI in animals given 1.9 x 10(5) oocysts per bird. In some experiments, uninoculated birds given the same amount of feed as infected birds (pair fed) were used to determine the impact of feed deprivation on weight loss and 3MH levels. Infected birds had significantly elevated plasma and muscle 3MH at 6 DPI after a single dose of Eimeria oocysts. The plasma and muscle 3MH returned to control levels after 14 DPI. The 3MH levels increased with increased dose of oocysts. Plasma and muscle 3MH levels were well correlated, and an inverse curvilinear relationship between weight gain and plasma 3MH concentrations levels was observed. Plasma and muscle 3MH levels were significantly elevated in pair-fed birds, but 3MH levels in infected birds were increased by 30% over pair-fed birds. The results suggested that muscle breakdown, as assessed by plasma and muscle levels of 3MH, increased during the acute stage of Eimeria infection in turkey poults.  相似文献

1. The effect of light intensities from 10 to 700 lux on the performance of 5 to 18 week-old turkey males was studied in 2 trials. 2. Body weight of 18 week-old turkeys, in both experiments, was highest under the lowest light intensity This coincided with higher weight gain and lower food intake, which resulted in significantly better food conversion efficiency 3. Light intensity affected heart muscle weight but not weight of breast muscle, abdominal fat or testis as proportions of body weight. 4. The decline in plasma T3 concentration with age differed from other treatments at the low light intensity, which resulted in a significantly higher T3 concentration in turkeys exposed to 10 lux at the age of 10 to 15 weeks. 5. It is concluded that light intensity significantly affects food conversion efficiency in turkey males. This is likely to be related to differential investment of energy expenditure for maintenance.  相似文献

The aim of the present study was to investigate whether muscle glycogen stores in slaughter pigs could be decreased through strategic finishing feeding before slaughter. Moreover, preliminary meat quality traits were measured to see whether such a regulation of muscle glycogen stores affected ultimate pH, color, and tenderness in the meat. The strategic finishing feeding was carried out the last 3 wk prior to slaughter. Seven experimental groups with eight animals per group were fed diets low in digestible carbohydrates. A control group with four animals was fed a traditional grower-finishing diet. The muscle glycogen stores were reduced in longissimus muscle (LM) 11 to 26% at the time of slaughter in pigs that were fed the experimental diets compared with the control group. Meat quality measured as ultimate pH and color on LM muscle in half the pigs 24 h postmortem showed that ultimate pH in LM was not affected by the reduction in glycogen stores in the muscles from pigs fed any of the experimental diets. However, the meat from pigs fed the experimental diets was darker than the meat from pigs that were fed the control diet with two of the experimental diets, resulting in significantly lower L* values. Activities of key enzymes in the glycolytic pathway, glycogen phoshorylase a and b, phosphofructokinase, and the fatty acid oxidative pathway, beta-hydrozyacyl-CoA-dehydrogenase, were not affected by the strategic feeding. In contrast, the activity of the proteolytic enzyme calpain as well as its inhibitor calpastatin was influenced by the strategic feeding. Lower activity of mu-calpain and greater activity of calpastatin in the muscle samples from the strategically fed pigs indicate a lesser muscle protein degradation in the muscles compared with muscles of control animals. The present study showed that the muscle glycogen stores in slaughter pigs can be reduced at the time of slaughter through strategic finishing feeding with diets low in digestible carbohydrate without compromising growth rate.  相似文献

The aim of the study was to investigate how feeding-induced changes in muscle glycogen stores affect the ratio of between the glycogen pools, pro- and macroglycogen. Pro- and macroglycogen content were determined in longissumus muscle from slaughter pigs subjected to a feeding strategy known to reduce total glycogen stores. Furthermore, early postslaughter glycolysis of the two glycogen forms was determined. The feeding strategy involved a diet (GLYRED diet) with a low digestible carbohydrate (5%)/high fat (18%) content, which was fed to the pigs the last 3 wk before harvest. A control group was fed a standard pig diet (49% digestible carbohydrate/5% fat). Total glycogen was reduced by 48 micromol/g dry weight (d.w.) in GLYRED pigs during the 3-wk feeding period. This was mainly due to a reduction in macroglycogen of 42 micromol/g d.w. During postmortem glycolysis the proglycogen appeared to be degraded in favor of macroglycogen. Moreover, total glycogen was degraded to a larger extent in muscle from the control pigs compared with muscle from GLYRED pigs. This difference was due to a significantly greater degradation of proglycogen in the control pigs. In conclusion, the results support earlier studies suggesting that proglycogen and macroglycogen are different glycogen pools that have different functions. Furthermore, the results show that the muscle glycogen pools can be manipulated through diet and that proglycogen is degraded in favor of macroglycogen under the anaerobic conditions postmortem.  相似文献

Carbohydrate (CHO) stored in the form of skeletal muscle glycogen is the main energy source for glycolytic and oxidative ATP production during vigorous exercise in mammals. In man, horse and dog both short‐term high intensity and prolonged submaximal exercise deplete muscle glycogen. In horses, however, muscle glycogen synthesis is 2–3‐fold slower than in man and rat, even when a diet high in soluble CHO is fed. There appear to be significant differences in CHO and glycogen metabolism between horses and other mammals, and it is becoming increasingly clear that many conclusions drawn from human exercise physiology do not apply to horses. This review aims to provide a comprehensive, comparative summary of the research on muscle glycogen synthesis in horse, man and rodent. Species differences in CHO uptake and utilisation are examined and the issues with feeding high soluble CHO diets to horses are discussed. Alternative feeding strategies, including protein and long and short chain fatty acid supplementation and the importance of rehydration, are explored.  相似文献

Eight standardbred horses trotted on a treadmill for 55 mins at a sub-maximal speed of 5m/sec and subsequently performed an exercise test consisting of 2 min intervals at increasing speed. Heart (HR) and respiratory (Rf) rates and venous blood samples were obtained before, during and for 5 mins after exercise. Gluteus medius muscle biopsies and rectal temperatures were taken before and after exercise. The mean HR was 132/min and the mean Rf was 156/min during the 5m/sec trotting. With 5m/sec exercise, plasma free fatty acids (FFA), glucose, creatinine and cortisol concentrations increased markedly. Blood lactate increased slightly and plasma potassium increased initially and then decreased with a lengthened duration of trotting. Within 5 mins post exercise plasma FFA, glucose and cortisol concentrations continued to rise, whereas creatinine and lactate levels declined slightly and potassium concentrations declined rapidly to below resting values. The mean intramuscular (im) glycogen utilisation was 86 mmol/kg, no significant changes occurred in creatine phosphate (CP), adenosine triphosphate (ATP) and glucose-6-phosphate (G-6-P) concentrations and muscle lactate decreased significantly. During the second exercise test mean HR was 215/min and Rf 126/min at top speed. No significant change was seen in plasma glucose whereas cortisol levels rose to a lesser extent, and creatinine lactate, ammonia and potassium concentrations to a greater extent, compared to 5 m/sec trotting. Post exercise, these parameters continued to increase except for creatinine which declined slightly and potassium which decreased rapidly. Isometric contraction was elicited in cattle by electrical stimulation (electrical immobilization) and was used as a means of investigating the interaction between muscle contraction and epinephrine-induced muscle glycogen degradation. At 0.5 and 24 hours after a 15-minute period of continuous muscle contraction, glycogen content of longissimus muscle specimens collected via needle biopsy was not different from precontraction concentrations. Epinephrine (13.2 mg/kg of body weight) given subcutaneously resulted in a 30% to 35% reduction of muscle glycogen. Reduction of muscle glycogen was slightly greater when epinephrine was used in conjunction with isometric contraction, compared with epinephrine treatment alone. Muscle glycogen increased, and free glucose, lactate, and glucose-6-phosphate decreased, with increases in body weight. In younger, lighter cattle (370 kg), epinephrine decreased muscle glycogen and lactate concentrations and generally decreased muscle glucose concentrations. Muscle-free glucose and lactate concentrations were increased in older, heavier cattle (446 kg) by contraction and epinephrine injection when the animals were immobilized for 30 minutes, with intermittent periods of muscle relaxation. In these cattle, muscle glycogen concentration slightly decreased with isometric contraction. Data indicate that reports of increased glycogenolysis observed in cattle subjected to stress by mixing strange animals or exercise is due to dynamic muscle contraction and not due to isometric muscle contraction.  相似文献

Convalescent serum given to 1-day-old poults delayed clinical signs of turkey coryza by several days and reduced mortality on infected farms. Turkey breeders immunized with cell-culture-adapted infectious bursal disease virus (IBDV) or turkey infectious bursal disease virus (TIBDV) had a marked increase in virus-neutralization (VN) antibody titers. The VN antibody titer was significantly higher in progeny poults than in poults from unimmunized breeders. Clinical turkey coryza and mortality was considerably less in poults from IBD- or TIBD-vaccinated breeders than in control poults. They also responded more favorably to hemorrhagic enteritis and fowl cholera vaccination.  相似文献

1. The experiment was conducted in order to determine the effect of the direction of turkey crossing on quality traits of their thigh muscle.

2. In total, 1358 turkeys of slow- (SG) and fast-growing (FG) lines as well as SF crosses (SG × FG) and FS crosses (FG × SG) were reared with access to free range to 21 weeks of age in the case of males and 15 weeks of age in the case of hens.

3. After slaughter of 15 males and 15 hens from each genetic line, their thigh muscles were examined and the following traits were measured after 5-min, 45-min, 2-h and 24-h post-mortem: temperature, pH, glycogen content (G), lactate content (L) and electrical conductivity (EC). Quality attributes were evaluated based on chemical composition, water holding capacity, cooking loss (%) and colour.

4. The rate of post-mortem changes in temperature, G, L and pH in muscles differed among the 4 lines of turkeys, with the highest metabolic rate determined for muscles of SG turkeys, followed by muscles of SF, FS and FG birds. A more beneficial muscle water holding capacity of both sexes of turkeys and better results of cooking loss in male muscles were found in SG and SF turkeys. The thigh muscles of the crosses were characterised by a higher content of protein and a lower content of fat compared to the muscles of FG turkeys, and in the case of the males also by a higher protein content compared to the muscles of SG birds.

5. Owing to the faster post-mortem metabolism, better water holding capacity, lower cooking loss and fat content at a similar content of protein, the thigh muscles of SF crosses raised in the free range system represent a better quality of meat for consumers compared to the thigh muscles of FS turkeys.  相似文献

However, cervical dislocation was used to collect tissues, a method that results in significant involuntary muscle convulsions. In this study, cervical dislocation alone was found to reduce muscle glycogen by 23%, therefore, barbiturate overdose was used to collect tissue samples before and after FW, at the end of refeeding, and from continuously fed controls at each interval. Additionally, plasma samples from 6-wk-old male chickens were taken at the initiation and end of a 24-hr feed withdrawal, and at various times during refeeding. After 24 hr of FW, liver glycogen decreased markedly (77%; P < 0.05), whereas muscle glycogen decreased slightly and transiently, such that it returned to and remained at control levels, even after prolonged (72 hr) FW. Plasma glucose was decreased, whereas glucagon was elevated after a 24-hr feed withdrawal (P < 0.05), when compared with control concentrations. Muscle glycogen levels were not significantly increased over control levels after refeeding, but liver glycogen was increased by 380% (P < 0.05). Feed deprivation followed by refeeding resulted in increased circulating insulin and glucose levels when compared with control levels. Therefore, by using methods of tissue collection that ensure that muscle glycogen determinations are not confounded by artifactual degradation, these results verify that regulation of avian muscle glycogen stores is similar to that in mammals.  相似文献
