This study aimed to investigate the ability of single nucleotide polymorphism (SNP) haplotypes in chicken mtDNA for presumption of the origins of chicken meat. We typed five SNPs of the D‐loop region in mtDNA by allele‐specific PCR (AS‐PCR) in 556 hens, that is 233 White Leghorn (WL), 50 Dekalb‐TX35 (D‐TX), 140 Barred Plymouth Rock (BPR) and 133 Rhode Island Red (RIR) kept in the National Institute of Livestock and Grassland Science (NILGS, Tsukuba, Japan). Five haplotypes were observed among those chickens by AS‐PCR. WL, D‐TX, BPR and RIR displayed three, two, one and four SNP haplotypes, respectively. By a combination of the haplotypes by AS‐PCR and the breeds, these chickens were classified into 10 groups. After the D‐loop was sequenced in two chickens from every group (20 individuals), 15 SNP sites (including one insertion) and eight sequence haplotypes were observed. In conclusion, haplotype variation was observed in and among the layer breeds of the NILGS. This study demonstrates that SNP haplotypes in mtDNA should be appropriate for the presumption of the origins of chicken meat.  相似文献

The D‐loop region in mitochondrial DNA (mtDNA) was sequenced and compared among six chickens. Eleven single nucleotide polymorphisms (SNP) were observed. For six of the SNP sites, polymerase chain reaction (PCR) primers that had each base (A, C, G and T) as the penultimate base at the 3′ end (N2 base) and bases specific to both alleles at the 3′ end were produced to type the polymorphisms. Twenty‐one out of 96 primers succeeded in distinguishing the SNP by the presence or absence of PCR product. This method provides an easy way to discriminate SNP in chicken mtDNA.  相似文献

The effects of the 5‐HT1A receptor agonist 8‐OH‐DPAT on food and water intake in male broiler chickens were investigated. The injection of 25 or 50 μg/kg of 8‐OH‐DPAT 15 min before refeeding in fasted animals produced a decrease in food intake. No effect was observed in drinking. The injection of 25 or 50 μg/kg of the 8‐OH‐DPAT 60 min after the start of refeeding did not produce any significant modification in food intake. No effect on drinking was recorded. The agonist 8‐OH‐DPAT injected 15 min before water presentation in water‐deprived chickens, produced an increased drinking 60 min after the presentation of water. No effect on food intake was observed. The results show that the effect on food intake of the agonist 8‐OH‐DPAT in fasted–refed broiler chickens was similar to those observed in mammals and layer‐strain chickens. However, the agonist did not alter significantly the food intake when the broilers were fed 60 min before the injection. These results are contrary to the observed effects in mammals and in layer‐strain chickens. Probably, the selection for rapid growth rate in broilers causes modifications in the feeding control pattern. The comparison between broilers and layers strain may be a useful tool to elucidate the complex mechanisms involved in food and water intake regulation in chickens.  相似文献

Ascites is a cardiovascular metabolic disease characterized by accumulation of fluid around the heart and in the abdominal cavity that eventually leads to death. This syndrome is the end‐point result of a series of metabolic incidents that are generally caused by impaired oxygen availability. Mitochondria are the major sites of oxygen consumption, therefore major contributors to oxidative stress. Genetic, metabolic and dietary factors can influence variations in mitochondrial biogenesis (mitochondrial size, number and mass) that might have an effect on oxygen consumption and reactive oxygen species production. This study evaluated the effect of genotype on PGC‐1α mRNA gene expression and mitochondrial biogenesis. These parameters were examined in male broiler chickens at 22 weeks of age from the SUS and RES lines divergently selected for ascites phenotype. From each line, five birds were sampled for right ventricle and breast muscle. Gene expression and mtDNA copy number were assessed by quantitative PCR. Results showed that birds from SUS had significantly higher PGC‐1α mRNA gene (p = .033) and mitochondrial DNA copy number (p = .038) in breast muscle. There was no difference in right ventricle PGC‐1α expression or mitochondrial DNA copy number between the two lines. These findings indicate that mitochondrial biogenesis and PGC‐1α mRNA gene expression differ between male broiler chickens from RES and SUS lines in a tissue‐specific manner.  相似文献

The development of broiler chickens over the last 70 years has been accompanied by large phenotypic changes, so that the resulting genomic signatures of selection should be detectable by current statistical techniques with sufficiently dense genetic markers. Using two approaches, this study analysed high‐density SNP data from a broiler chicken line to detect low‐diversity genomic regions characteristic of past selection. Seven regions with zero diversity were identified across the genome. Most of these were very small and did not contain many genes. In addition, fifteen regions were identified with diversity increasing asymptotically from a low level. These regions were larger and thus generally included more genes. Several candidate genes for broiler traits were found within these ‘regression regions’, including IGF1, GPD2 and MTNR1AI. The results suggest that the identification of zero‐diversity regions is too restrictive for characterizing regions under selection, but that regions showing patterns of diversity along the chromosome that are consistent with selective sweeps contain a number of genes that are functional candidates for involvement in broiler development. Many regions identified in this study overlap or are close to regions identified in layer chicken populations, possibly due to their shared precommercialization history or to shared selection pressures between broilers and layers.  相似文献

利用原子吸收分光光度法和焦锑酸钾组化电镜技术,测定肉鸡右心室壁总钙和右心室组织细胞内游离Ca2+含量和分布。结果显示肉鸡腹水综合征患鸡右心室壁总钙含量为(71.2±6.5)mg/kg,显著高于低温未发病和健康肉鸡(分别为58.0±4.4,46.8±3.1,P<0.01)。电镜观察发现Ca2+沉淀颗粒主要散在分布于患鸡心肌胞浆、肌浆网、线粒体等部位,腹水综合征患鸡Ca2+颗粒的沉积量比健康肉鸡显著增多。结果提示心肌细胞内Ca2+的异常可能与肉鸡腹水综合征的发生发展有关。  相似文献

The utilization of copra meal as a feed ingredient is limited because it contains a high level of mannan. However, recent findings indicate that the effect of copra meal on growth performance in broiler chickens can be improved by the supplementation of mannanase in the diet. In the present study, we examined the effect of mannanase‐hydrolyzed copra meal (MCM) on growth performance and muscle protein metabolism in growing broiler chickens (Gallus gallus domesticus). Forty 8‐day‐old male broiler chicks were assigned to two groups (four birds in each pen, five replicates) and fed either a commercial diet (as a control diet) or a diet containing MCM at 0.2% until 22 days of age. Dietary MCM significantly increased the weights of body, breast muscle, and thighs in chickens, whereas the weights of abdominal adipose tissue and liver were not affected. Cumulative feed intake was significantly increased by MCM. Dietary MCM significantly decreased plasma 3‐methylhistidine level. The messenger RNA and protein levels of muscle protein metabolism‐related factors were not altered by MCM. These findings suggest that the growth‐promoting effect of MCM is related to the suppression of muscle proteolysis in growing broiler chickens.  相似文献

This research was conducted to study the performance and carcass parameters of broiler chickens fed diets supplemented with heat‐treated non‐starch polysaccharide degrading enzyme. A total of 432 one‐day old Ross 308 broiler chickens were allocated to five treatments: (i) CON (basal diet), (ii) E1: CON + 0.05% multi‐enzyme, (iii) E2: CON + 0.1% multi‐enzyme, (iv) E3: CON + 0.05% thermo‐resistant multi‐enzyme and (v) E4: CON + 0.1% thermo‐resistant multi‐enzyme, each treatment consisted of six replications and 12 chickens in each replication. The chickens were housed in three floor battery cages during 28‐day experimental period. On days 1–7, gain in body weight (BWG) improved by feeding the diets supplemented with thermo‐resistant multi‐enzyme. On days 7–21 and 1–28, chickens fed the diets containing thermo‐resistant multi‐enzyme showed improved (p < 0.05) BWG and feed conversion ratio (FCR) compared to CON group. Supplementing the diets with multi‐enzyme or thermo‐resistant multi‐enzyme affected the percentage of drip loss on d 1 (p < 0.05). Drip loss percentage on days 3 and 5 and also meat colour were not affected significantly. Supplementing the diets with multi‐enzyme or thermo‐resistant multi‐enzyme did not affect the relative weights of organs but compared to CON group, relative weight of breast muscle increased and abdominal fat decreased (p < 0.05). Among measured blood constituents, chickens fed supplemented diets with thermo‐resistant multi‐enzyme showed higher (p < 0.05) IgG. Counts of red and white blood cells and lymphocyte percentage were not affected. In conclusion, the results demonstrated that supplementing pelleted diets with thermo‐resistant multi‐enzyme improved performance of broiler chickens.  相似文献

The effect of L‐carnitine supplementation on mitogen (concanavalin A, Con A) induced proliferation of mononuclear cells (MNC) in the spleen was investigated in broiler chickens at different ages. Day‐old chickens were fed a diet supplemented with or without L‐carnitine (100 ppm) for 24 days. The carnitine‐supplemented group showed greater proliferation of MNC in the spleen in response to Con A than that of the control group at 24 days of age. In addition, at 24 days of age the carnitine‐supplemented group showed higher expression of interleukin (IL)‐2 and interferon (IFN)‐γ mRNA, but lower expression of inducible nitric oxide synthase (iNOS) in the Con A‐stimulated splenic MNC than the control group. The enhancement effect of L‐carnitine on MNC proliferation and IL‐2 mRNA expression was not found in chicks at 14 days of age. Addition of L‐carnitine (50 nmol/mL) to the culture medium enhanced proliferation and IL‐2 mRNA expression of splenic MNC obtained from 24‐day‐old but not from 14‐day‐old broiler chickens. The results suggest that L‐carnitine is capable of enhancing MNC proliferation in broiler chickens at 24 days of age partly through increasing IL‐2 and IFN‐γ production and decreasing NO production.  相似文献

Krüppel‐like factor 7 (KLF7) has been extensively studied in mammalian species, but its role in birds is still unclear. In the current study, cloning and sequencing showed that the full‐length coding region of chicken KLF7 (Gallus gallus KLF7, gKLF7) was 891 bp long, encoding 296 amino acids. In addition, real‐time RT‐PCR analysis showed that gKLF7 was broadly expressed in all 15 chicken tissues selected, and its expression was significantly different in spleen, proventriculus, abdominal fat, brain, leg muscle, gizzard and heart between fat and lean broilers at 7 weeks of age. Additionally, one novel single nucleotide polymorphism (SNP), XM_426569.3: c. A141G, was identified in the second exon of gKLF7. Association analysis showed that this locus was significantly associated with fatness traits in Arbor Acres broiler random population and the eighth generation of Northeast Agricultural University broiler lines divergently selected for abdominal fat content (NEAUHLF) population (P < 0.05). These results suggest that gKLF7 might be a candidate gene for chicken fatness traits.  相似文献

Objective Fowl adenoviruses (FAdVs) cause inclusion body hepatitis (IBH) in chickens. In this study, clinical cases of IBH from Australian broiler flocks were screened for the presence and genotype of FAdVs. Methods Twenty‐six IBH cases from commercial poultry farms were screened. Polymerase chain reaction (PCR) coupled with high‐resolution melt (HRM) curve analysis (PCR/HRM genotyping) was used to determine the presence and genotype of FAdVs. For comparison, field isolates were also assessed by virus microneutralisation and nucleotide sequence analysis of the hexon loop 1 (Hex L1) gene. PCR detection of chicken anaemia virus (CAV) and infectious bursal disease virus (IBDV) was also employed. Results FAdV‐8b and FAdV‐11 were identified in 13 cases each. In one case, FAdV‐1 was also identified. Cross‐neutralisation was observed between the FAdV‐11 field strain and the reference FAdV‐2 and 11 antisera, a result also seen with the type 2 and 11 reference FAdVs. Field strains 1 and 8b were neutralised only by their respective type antisera. The FAdV‐8b field strain was identical to the Australian FAdV vaccine strain (type 8b) in the Hex L1 region. The Hex L1 sequence of the FAdV‐11 field strain had the highest identity to FAdV‐11 (93.2%) and FAdV‐2 (92.7%) reference strains. In the five cases tested for CAV and IBDV, neither virus was detected. The evidence suggested the presence of sufficient antibodies against CAV and IBD in the parent flocks and there was no indication of immunosuppression caused by these viruses. Conclusion These results indicate that PCR/HRM genotyping is a reliable diagnostic method for FAdV identification and is more rapid than virus neutralisation and direct sequence analysis. Furthermore, they suggest that IBH in Australian broiler flocks is a primary disease resulting from two alternative FAdV strains from different species.  相似文献

To evaluate the effect of maternal leptin on muscle growth, we injected 0 μg (control, CON), 0.5 μg (low leptin dose, LL) or 5.0 μg (high leptin dose, HL) of recombinant murine leptin dissolved in 100 μl of PBS into the albumen of broiler eggs prior to incubation. The newly hatched chicks were all raised under the same conditions until 21 days of age (D21), when body weight was measured and samples of gastrocnemius muscle were collected and weighed. Myosin ATPase staining was applied to identify myofibre types and measure the cross‐sectional area (CSA) of myofibres. Real‐time PCR was performed to quantify leptin receptor (LEPR), insulin‐like growth factor 1 (IGF‐1), IGF‐1 receptor (IGF‐1R), growth hormone receptor (GHR) and myostatin (MSTN) mRNA expression in the gastrocnemius muscle. The activity of calpains (CAPNs) in the gastrocnemius muscle was measured using a quantitative fluorescence detection kit. Male chickens treated with both high and low doses of leptin had significantly higher (p < 0.05) body weight on D21. The high leptin significantly increased the CSA (p < 0.05) of gastrocnemius muscle in male chickens, which coincided with a 93% increase (p < 0.05) in IGF‐1 mRNA expression. Likewise, the LL dose increased the weight of gastrocnemius muscle in male chickens (p < 0.05), which was accompanied by a 41% down‐regulation (p < 0.05) of MSTN mRNA expression and a decreased activity of CAPNs. However, all these changes were not observed in female chickens. The proportion of myofibre types did not altered. No significant change was detected for LEPR and GHR mRNA expression. These results indicate that in ovo leptin treatment affects skeletal muscle growth in chickens in a dose‐dependent and sex‐specific manner. The altered expression of IGF‐1, MSTN mRNA and activity of CAPNs in skeletal muscle may be responsible for such effects.  相似文献

The response of broiler chickens to 3 levels of sunflower meal and 2 levels of NSP‐ase enzyme combination (with and without) was investigated in 3 × 2 factorial arrangement under complete randomized design (CRD). A total of 240 Hubbard broiler chicks were fed on practical mash diets having 2950 kcal of ME and 21% CP from 1 to 42 days of age. The BW gain was not significantly reduced when 25% SFM was added in the diets during 1 to 42 days of age. Supplementation of NSP‐ase in broiler diets (day 1–42 overall) demonstrated non‐significant differences (p < 0.05) across the treatments in terms of FI and BWG. The difference in feed:gain at 15% or 20% SFM was observed to be non‐significant. Replacement of SBM with SFM or inclusion of SFM at higher level (25%) increased/deteriorated FCR. The addition of exogenous NSP‐ase showed a significant improvement (p < 0.01) in feed:gain. The improvement was clearly demonstrated when SFM was added to the experimental diet at 15% or even 20%. Supplementation of NSP‐ase at the 25% inclusion level could not, however, sustain the beneficial effect, which was possibly due to excessively high dietary CF. No difference was noted across the treatments regarding carcass response. Relative gizzard weight and intestinal weight were observed to be improved in birds consuming higher levels of SFM (p = 0.00). The digestibility of CF was observed to improve when SFM was used at 20% and 25% in the diets. No improvement in the digestibility of CF was observed with NSP‐ase supplementation, which meant other factors were clearly involved. Supplementation of NSP‐ase improved FCR up to 20% SFM. At 25% SFM, no improvement in the digestibility of CF was observed with NSP‐ase supplementation.  相似文献

This study aims to assess the association of polymorphisms and mRNA expression of adipocyte‐type fatty acid‐binding protein (A‐FABP) with intramuscular fat (IMF) in the breast muscle (BM) and leg muscle (LM) of Baicheng‐You chickens (BYCs). A total of 180 chickens, including sixty black Baicheng‐You chickens (BBYCs), sixty silky Baicheng‐You chickens (SBYCs) and sixty white Baicheng‐You chickens (WBYCs), were reared from 1 to 120 day. A polymerase chain reaction–single‐strand conformation polymorphism strategy (PCR‐SSCP) was used to detect the polymorphism of the A‐FABP gene in the first exon, and the C51T silent mutational site was found. The IMF content with the AA genotype was significantly higher than that with the AG genotype (p = 0.0473) in the LM of WBYC. Thus, this site could be taken as a molecular marker in selecting a higher IMF content of LM in WBYC. A‐FABP gene mRNA expression in the BM and LM of BYCs was detected, and a significant positive correlation was observed in the LM of WBYC. These findings provide fundamental data that might be useful in further study of the role of the A‐FABP gene in IMF content and fatty metabolism in chickens.  相似文献

The genetic relationship between 195 Mangalica and 79 non‐Mangalica pigs was studied using mitochondrial D‐loop SNP genotyping. Altogether, 35 polymorphic sites and 27 haplotypes were identified. Of the haplotypes, eight and 16 are Mangalica and non‐Mangalica specific, respectively, while three contain both Mangalica and non‐Mangalica individuals. Genetic distance values and phylogenetic analysis indicate that Mangalica individuals are very closely related, and five haplotypes represent approximately 92% of the Mangalica pigs involved in the study, thus determining the major maternal lineages. In contrast to previous microsatellite studies, individuals of Mangalica could not be distinguished as three separate breeds using mtDNA genotyping. Comparing modern and archaeological mtDNA sequences revealed that present day Mangalica is related to pigs that lived in the Carpathian basin where postulated ancestors of Mangalica also lived. This is the first DNA‐based genetic evidence to support the described breeding history of Mangalica.  相似文献

Thirty‐two 15‐day old broiler chicks (Chunky strain ROSS 308) were randomly divided into four treatments in a 2 × 2 factorial design. The main factors were diet (basal diet or basal diet supplemented with 0.15% astaxanthin‐rich dried cell powder (Panaferd‐P [astaxanthin 30 ppm]) and ambient temperature (thermo‐neutral [25 ± 1°C] or high [35 ± 1°C for 6 hr]). Dietary supplementation with Panaferd‐P did not affect growth performance, though high ambient temperature decreased feed intake and the weight of breast tender muscle, liver, and heart. High ambient temperature also decreased redness in both breast and leg muscles of chickens, while Panaferd‐P increased redness and yellowness of breast and leg muscles of chickens. Panaferd‐P increased Paracoccus carotinifaciens‐derived pigments (i.e., adonixanthin, astaxanthin, adonirubin, and cantaxanthin) as well as corn‐derived pigments such as zeaxanthin and lutein in breast and leg muscles. High ambient temperature increased the malondialdehyde (MDA) concentration in breast muscle, while Panaferd‐P decreased the MDA concentration in breast muscle under both temperature conditions. Our results suggest that dietary supplementation with Panaferd‐P increases muscle carotenoid content, the redness and yellowness of meat and decreases the muscle MDA concentration in broiler chickens kept under thermo‐neutral or high ambient temperature conditions.  相似文献

Effect of dietary supplementation of astaxanthin (Ax) from Phaffia rhodozyma on lipopolysaccharide‐induced inflammatory responses was investigated in male broiler chickens fed a corn‐based diet. Birds (1 week of age) were fed a corn‐enriched diet containing either 0 or 100 ppm Ax for 2 weeks and were intraperitoneally injected with lipopolysaccharide (LPS, 1 mg/kg body weight). Inflammatory responses were evaluated by determining changes in expression of messenger RNA (mRNA) in cytokines and mediators related to inflammatory responses (interleukin (IL)‐1 beta and ‐6, inducible nitrite synthase (iNOS), interferon (IFN)‐ γ and cyclooxygenase (Cox)‐2 in the liver and spleen after 2 h of LPS injection and plasma ceruloplasmin concentration as an acute phase protein. Birds fed Ax showed significantly higher iNOS mRNA expression in the liver and spleen compared to that of control birds. Ax‐fed birds also showed greater increase in mRNA expression in the liver of IL‐1, IL‐6 and IFN‐γ compared to that of control birds. The enhancing effect of Ax was further progressed when LPS was injected. No difference was found in plasma ceruloplasmin concentration between the Ax‐fed group and control group. The results suggest that feeding supplementation of Ax (100 ppm) to a corn‐enriched diet possibly does not have anti‐inflammatory effect in male broiler chickens.  相似文献

Foot pad dermatitis (FPD), a condition of inflammation and necrotic lesions on the plantar surface of the footpads, is commonly observed in fast‐growing broiler chickens and turkeys. FPD negatively affects the welfare of birds, performance indices and the economic profit of poultry meat production. Nutrition is an important factor affecting water intake, excreta moisture and litter quality and, in this way, the occurrence and intensity of FPD in birds. This article reviews and discusses the recent results published in the literature on the effects of nutritional factors on litter quality and FPD severity in broiler chickens and turkeys. Literature data on the efficacy of nutritional methods on the litter quality and FPD occurrence are not consistent. However, the results of several experiments indicate that the optimal level of crude protein, biotin and electrolytes (Na, K) in the diet, as well as feed additives such as feed enzymes hydrolysating non‐starch polysaccharides and organic sources of microelements (zinc), may reduce the litter moisture as well as FPD incidence and severity in broiler chickens and turkey.  相似文献

The role of birds as sources of Shiga toxin-and intimin-producing Escherichia coli was studied. Fecal samples from live gulls (n=86), pigeons (n=33) and broiler chickens (n=199) from 23 flocks were analyzed for stx and eae by PCR. No stx positive samples were detected. In contrast, eae E. coli were highly prevalent among gulls (40%), and was also found in pigeons (7%) and chickens (57% of the flocks contaminated). The eae positive isolates were analyzed genetically and O-serogrouped. One isolate from a pigeon was found to have stx (2f). The isolates of gulls differed from those of pigeons and chickens, and all eae E. coli isolates from birds differed from human pathogenic strains by the lack of EHEC-hlyA and bfp/EAF as well as distribution of O-serogroups. Thus, birds cannot be regarded as important carriers of zoonotic stx or eae E. coli in Finland.  相似文献
