Worldwide, foodborne illness is a significant public health issue in both developed and developing countries. Salmonellosis, campylobacteriosis and shigellosis are common foodborne gastrointestinal illnesses caused by the bacteria Salmonella spp., Campylobacter spp. and Shigella spp. respectively. These zoonotic diseases are frequently linked to eggs and poultry products. The aim of this study was to investigate the presence of these pathogens in Australian backyard poultry flocks and to determine risk factors for these pathogens. Poultry faeces samples were collected from 82 backyards and screened for Salmonella spp., Campylobacter spp. and Shigella spp. using qPCR. A questionnaire was administered to the backyard poultry owners to assess their knowledge regarding management of poultry and eggs and to identify potential risk factors that may contribute to the presence of zoonotic pathogens in the flocks. One composite faecal sample was collected from each backyard (82 samples). Composite sampling here means taking one or more grab samples from a backyard to make up approximately 10 grams. Four per cent of samples, that is 4% backyards tested, were positive for Salmonella spp., 10% were positive for Campylobacter spp. and none were positive for Shigella spp. A higher infection rate was seen in multi-aged flocks (24%) compared with the single-aged flocks (3%). The survey found that many participants were engaging in risky food safety behaviours with 46% of participants responding that they washed their eggs with running water or still water instead of wiping the dirt off with a damp cloth to clean the eggs and 19% stored their eggs at room temperature. This study demonstrated that backyard poultry may pose a potential risk for salmonellosis and campylobacteriosis. Additionally, Australian public health and food safety regulations should be modified and effectively implemented to address the risks associated with backyard poultry husbandry.  相似文献

Spermatozoa in semen samples from 8 individual male domestic fowls were shown to have a differential and characteristic ability to hydrolyse holes in the inner perivitelline layer from laid eggs in an in vitro assay. 2. The number of holes produced by samples of spermatozoa per unit area of inner perivitelline layer in vitro was linearly correlated with sperm ATP content ( r = 0.85) and motility (r=0.76). 3. The number of holes formed in the inner perivitelline layer in vitro was also linearly correlated with the numbers of holes formed in the inner perivitelline layer of eggs fertilised in vivo , in inseminated hens (r=0.90); and was correlated logarithmically with the proportion of fertile eggs laid by these hens.  相似文献

Salmonella enterica serovar Enteritidis (S. Enteritidis) is one of the most prevalent causes for human gastroenteritis and is by far the predominant Salmonella serovar among human cases, followed by Salmonella Typhimurium. Contaminated eggs produced by infected laying hens are thought to be the main source of human infection with S. Enteritidis throughout the world. Although previous studies have looked at the proportion of infected eggs from infected flocks, there is still uncertainty over the rate at which infected birds produce contaminated eggs. The aim of this study was to estimate the rate at which infected birds produce contaminated egg shells and egg contents. Data were collected from two studies, consisting of 15 and 20 flocks, respectively. Faecal and environmental sampling and testing of ovaries/caeca from laying hens were carried out in parallel with (i) for the first study, testing 300 individual eggs, contents and shells together and (ii) for the second study, testing 4000 eggs in pools of six, with shells and contents tested separately. Bayesian methods were used to estimate the within‐flock prevalence of infection from the faecal and hen post‐mortem data, and this was related to the proportion of positive eggs. Results indicated a linear relationship between the rate of contamination of egg contents and the prevalence of infected chickens, but a nonlinear (quadratic) relationship between infection prevalence and the rate of egg shell contamination, with egg shell contamination occurring at a much higher rate than that of egg contents. There was also a significant difference in the rate of egg contamination between serovars, with S. Enteritidis causing a higher rate of contamination of egg contents and a lower rate of contamination of egg shells compared to non‐S. Enteritidis serovars. These results will be useful for risk assessments of human exposure to Salmonella‐contaminated eggs.  相似文献

The poultry red mite (Dermanyssus gallinae) is a serious problem for the poultry industry worldwide. However, the relationship between the mite population and the damage that they cause is still unclear. In this study, the mite population in poultry houses was examined using an established trap method, and the risk of blood‐stained eggs caused by the mites was assessed. Traps were placed once a week outside the egg channels and/or on the floor in two poultry farms in Fukuoka Prefecture, Japan, from April 2012 to July 2014. The numbers of blood‐stained eggs and total eggs were counted at weekly intervals. The results showed that the number of mites increased from April to May, and reached a peak around the beginning of June when the average temperature and humidity were >24°C and 70–90%, respectively. In the segmented model, the correlation between the proportion of blood‐stained eggs and the number of mites or temperature was positive over a threshold. In conclusion, our established trap method is useful for monitoring mites and can be used to predict when poultry farms should be treated to prevent appearance of blood‐stained eggs.  相似文献

According to the European Food Safety Authority, salmonellosis is still one of the main causes of infectious foodborne gastroenteritis in humans. Broilers are an important source of salmonellosis after eggs and pork. Between 1987 and 1999 the trend of human salmonellosis incidence in Belgium increased constantly. However, from 2000 until 2005 a decrease in human cases was observed, probably following the sanitary measures implemented in the poultry breeder and laying sector. In order to decrease human infections it is essential to tackle the problem at the farm level to minimize cross-contamination from farm to fork. This paper seeks to answer two questions: (i) given the Salmonella status of the farm at a certain occasion (equal to the sampling time of the flock), what are the risk factors that the farm will be Salmonella positive at a following occasion? And (ii) what are the risk factors for a farm to be persistently positive for two consecutive flocks? We used surveillance data on 6824 broiler flocks studied for Salmonella infectivity from 2005 to 2006 in Belgium. The farms were tested regularly (3 weeks before slaughter of each broiler flock) for the presence of Salmonella based on multiple faecal samples per flock on a farm yielding clustered data. Generalized estimating equations, alternating logistic regression models, and random-intercept logistic regression models were employed to analyse these correlated binary data. Our results indicated that there are many factors that influence Salmonella risk in broiler flocks, and that they interact. Accounting for interactions between risk factors leads to an improved determination of those risk factors that increase infection with Salmonella. For the conditional analysis, the risk factors found to increase the risk of Salmonella infection on a farm at a current occasion given the previous Salmonella status included: Salmonella infection of day-old chicks (of the current flock); a previously infected flock even though the farm was equipped with a hygiene place to change clothes prior to entering the broiler house; having temporary workmen when there was a separation between birds of different species; and separating birds of different species in the Walloon region relative to the Flanders region. Sanitary measures such as a cleaning and disinfecting procedure conducted by an external cleaning firm, applying the all-in all-out procedure, and hand washing decreased the risk despite their interaction with other factors. From the joint analysis, the most important factors identified for increased risk for persistent Salmonella on a farm involved the interaction between having temporary workmen when there were poultry or farmers in contact with foreign poultry or persons, and the interaction between having temporary workmen when there were poultry or farmers in contact with external poultry or persons.  相似文献

Plasmodium juxtanucleare is one of the agents that cause avian malaria in chickens and little is known about the treatment of the infection. The aim of this study was to compare the antimalarial activities of rutin and chloroquine in chickens infected with P. juxtanucleare after immunossuppression with corticosteroid. Antimalarial activity was verified in 33 fowls by evaluation of parasitemia levels in Giemsa-stained blood smears taken to 30 days. Rutin did not exert a reduction in parasitemia from P. juxtanucleare when compared to chloroquine treatment that kept the parasites at a low level, demonstrating its antimalarial effect. During the course of the infection, the trophozoite stage predominated (80%), followed by schizont (17%) and gametocyte (3%). Maximum parasitemia levels were recorded on day 12 in control and rutin groups. There were no significant differences in the hematocrit values, weight body or the temperature of the fowls among the groups evaluated (p > 0.05).  相似文献

Salmonella enterica remains one of the most important food-borne pathogens of humans and is often acquired through consumption of infected poultry meat or eggs. Control of Salmonella infections in chicken is therefore an important public health issue. Infection with S. enterica serovar Typhimurium results in a persistent enteric infection without clinical disease in chickens of more than 3 days of age, and represents a source for contamination of carcass at slaughter and entry into the human food chain. Data presented indicate a profound effect of age at initial exposure on the persistence of infection and a lesser effect on the development of effective immunity to re-challenge. The percentage of birds positive for Salmonella was high until 8–9 weeks of age, regardless of the age at which the birds were infected (1, 3 or 6 weeks). The birds infected at 3 and 6 weeks of age produced a more rapid and higher antibody response (IgY and IgA) than those infected at 1 week of age, but in all cases infection persisted for a considerable period despite the presence of high antibody levels. Following a re-challenge infection with S. Typhimurium, all three previously-infected groups had fewer bacteria in the gut, spleen and liver compared with age-matched birds receiving a parallel primary infection. However, the birds primary infected at 3 and 6 weeks of age cleared infection more rapidly than those infected at a younger age. Interestingly older-primed birds had higher specific T lymphocyte proliferative responses and specific circulating levels of IgY antibody at time of re-challenge. Although birds initially infected at 1 week of age and those that were previously uninfected produced a stronger antibody response following re-challenge, they were slower to clear Salmonella from the gut than the older-primed groups which expressed a stronger T lymphocyte response. The data presented indicate that clearance of Salmonella from the gut is age-dependent and we propose that this relates to the increased competence of the enteric T cell response. The findings that Salmonella persists beyond 8–9 weeks, irrespective of age at exposure, has implications for the broiler sector and indicates the need to remain Salmonella free throughout the rearing period. Moreover, the re-challenge data demonstrates that infection at a young age is less effective in producing protective immunity than in older chickens. This feature of the development of protective immunity needs to be considered when developing vaccines for the broiler sector of the poultry industry.  相似文献

One of the species that causes avian malaria is Plasmodium juxtanucleare. It is commonly found in poultry, especially when the birds receive food free of coccidiostats. Since industrial and organic poultry breeding is increasing in the world and few studies have been conducted examining the clinical parameters of both healthy and infected birds, this work evaluated whether the infection caused by P. juxtanucleare in Gallus gallus provokes alterations in the birds' hepatic profile. We analyzed the activity of ALT and AST and carried out histological analyses of liver sections of infected fowls by intracelomic inoculation with infected blood from a donor fowl with a parasite load of around 7%. The infected birds' parasite load was evaluated during 45 days by means of blood smears. There was a positive correlation between the increase in parasite load and higher ALT activity in the infected fowls, but there was no significant variation of the AST activity between the control and infected groups, possibly because of the non-specificity of this enzyme as an indicator of hepatic lesion. The results show that infection caused by P. juxtanucleare in G. gallus provokes hepatic alterations, indicated by the increase in the ALT enzyme activity and by the inflammatory infiltrates found in the liver sections of the infected fowls.  相似文献

Larvae of the house fly (Musca domestica) grew and developed normally in fresh poultry manure where the proper environmental conditions were maintained. A temperature of 27 °C and a moisture content ranging from 60 to 75% in fresh poultry manure was determined to be optimum for larval development. A moisture content of 80% or greater created anaerobic problems for larvae.

Optimum production of house fly pupae was obtained by inoculating 0.50 to 0.75 g of fly eggs per kg of fresh poultry manure. Five to eleven days were required for the eggs to be converted to pupae depending on the environmental conditions. After digestion, the poultry manure, was reduced to about half and was granular in texture. The digested residue readily dried, had less odour and contained approximately 15% protein.  相似文献

Active serologic surveillance programs to detect avian influenza viruses (AIVs) in table egg-laying chickens have been initiated by several states as a response to the economic threat posed by these viruses. Most outbreaks of avian influenza in domestic poultry are caused by mildly pathogenic AIVs. In the study reported here, infection by an H6N2 AIV was used as a model of mildly pathogenic AIV infections in egg-type chickens. The total number of eggs laid by 5 control hens was 619 or 0.904 eggs/day/hen, whereas the total number laid by 10 infected hens was 1,018 or 0.743 eggs/day/hen. The difference in egg production between the 2 groups was not statistically significant (P = 0.38). Anti-influenza antibodies were monitored by use of an agar gel immunodiffusion test and an ELISA for a period of 20 weeks after inoculation. Antibodies in serum developed sooner, peaked at higher levels, and remained at higher levels than did antibodies found in egg yolk, as indicated by ELISA results. For infected chickens, the correlation between serum and egg yolk ratios was 0.66. Serum samples would appear to be preferable to egg yolk samples for surveillance programs intended to identify chicken flocks that may have been infected by an AIV weeks or months before samples are collected.  相似文献

Increasingly large numbers of poultry are held in production systems with access to outdoor areas. In these systems intestinal helminths are found with flock prevalences of up to 100%. Helminth infections influence chicken health negatively, which is why the following investigation has been performed.In the present experiment, 20 chickens of two inbred chicken lines containing the major histocompatibility complex (MHC) haplotypes, B14 and R5, were inoculated with 500 embryonated Ascaridia galli eggs. The A. galli-specific IgG titres of serum samples and the excretion of A. galli eggs in chicken faeces were measured for a period of 81 weeks.The level of excreted A. galli eggs measured as eggs per gram chicken faeces (EPG) varied greatly between chickens in each line. Significant differences were found between the two lines and with the R5 chickens reaching the highest levels. Likewise, the A. galli-specific IgG titres in serum differed significantly between the two lines, and an inverse relationship between infection level (EPG) and antibody titres was found. Although this inverse relationship suggests that humoral immunity may be involved in protection against A. galli infection, the high antibody titres did not prevent continued infection.  相似文献

Participatory research on vaccination of village poultry against Newcastle disease (ND) was carried out in the village of Disaneng, in the North West Province of South Africa. Three application methods for ND Inkukhu vaccine were shown to induce sufficient levels of immunity in back-yard poultry when correctly administered. These are eye-droplet administration to individual fowls, in-feed and in-water administration to small flocks. After a community meeting and group discussion to select methods of vaccination, only 2 of the 3 methods were chosen; the individual administration of droplets into the eyes was considered to be too impractical because back-yard fowls are difficult to catch. Visual and practical training material was prepared and presented to volunteer vaccinators (n = 23). Vaccinators were then required to register all the poultry owners in their ward who wished to have poultry vaccinated. Once an indication of the number of chickens to be vaccinated had been made available, ND Nobilis Inkukhu vaccine was supplied to vaccinators free of charge. Community vaccinators were responsible for the organisation of the vaccination campaign, including storage and preparation of vaccine for application. All 9 wards in the village were initially involved with a total of 482 households, owning 6141 chickens, participating. This represented slightly in excess of 60 % of the fowls in the area. Involvement in a 2nd round of vaccinations, 1 month later, was far poorer with only 211 households owning a total of 1636 chickens participating. Serum samples were collected from vaccinated fowls using systematic random sampling and tested for circulating antibodies. The levels of protection varied, with no significant difference found between in-feed and in-water vaccine administration. Volunteer vaccinators were found to be unreliable, easily demotivated, did not keep good records and left the project when offered permanent employment. Contacting them to make arrangements for delivering vaccine was difficult and time consuming. Structured interviews indicated that deaths in poultry and the attitude of the owners probably contributed to the demotivation of the volunteers used as community vaccinators. It was concluded that volunteers are not the ideal choice for vaccination of village poultry against Newcastle disease.  相似文献

1. The daily food intake of captive red grouse was measured throughout the year. It was positively related to body weight and to daylength, and negatively to air temperature. Grouse ate more during their main summer moult, and the hens ate most while producing eggs. 2. The total weight of eggs laid was positively related to daily food intake during the 5 weeks before laying, but not to daily intake during laying. The rate at which eggs were laid was related to daily intake during laying as well as before laying. The possible significance of these results for poultry management is discussed.  相似文献

Backyard poultry has become increasingly popular in industrialized countries. In addition to keeping chickens for eggs and meat, owners often treat the birds as pets. However, several pathogenic enteric bacteria have the potential for zoonotic transmission from poultry to humans but very little is known about the occurrence of zoonotic pathogens in backyard flocks. The occurrence and the antimicrobial resistance of Salmonella enterica, Campylobacter spp., Listeria monocytogenes and enteropathogenic Yersinia spp. was studied in 51 voluntary backyard chicken farms in Finland during October 2012 and January 2013. Campylobacter isolates were further characterized by pulsed‐field gel electrophoresis (PFGE), and the occurrence of ESBL/AmpC‐producing E. coli was investigated. The findings from this study indicate that backyard chickens are a reservoir of Campylobacter jejuni strains and a potential source of C. jejuni infection for humans. Backyard chickens can also carry L. monocytogenes, although their role as a primary reservoir is questionable. Campylobacter coli, Yersinia pseudotuberculosis and Salmonella enterica were only found sporadically in the faecal and environmental samples of backyard poultry in Finland. No Yersinia enterocolitica carrying the virulence plasmid was isolated. All pathogens were highly susceptible to most of the antimicrobials studied. Only a few AmpC‐ and no ESBL‐producing E. coli were found.  相似文献

S. Typhimurium is one of the 2 most common salmonella serotypes causing human salmonellosis in Denmark. In order to illustrate the significance of different production animals as a source of infection, 1461 isolates were characterized by phage typing. The isolates originated from human patients and from cattle, pigs and poultry. By phage typing the isolates could be separated in 35 different phage types. Five types (10, 12, 66, 110 and 135) predominated and comprised 78.8% of the isolates. In humans, 57.3% of the isolates were phage type 12. This phage type was also predominant in pig herds and, to a lesser degree, in cattle. Phage types 110, 120, 135 and 193 constituted 86.5% of the poultry isolates while these phage types only made up 12.9% of the human isolates. The investigation showed that pigs are probably a major source of S. Typhimurium infection in humans in Denmark today.  相似文献

Screening and identification of Salmonella enteritidis in commercial poultry flocks have assumed principal roles in preventing transmission of this pathogen to humans from hen eggs. Serologic diagnosis of S. enteritidis infection in commercial flocks currently relies on laboratory-based tests for detection of antibodies to the lipopolysaccharide, whole flagella, and bacteria. We amplified a sequence from the g,m flagellin of S. enteritidis, followed by cloning, expression, and purification of the protein. The recombinant protein was first characterized by western blot and subsequently evaluated as enzyme-linked immunosorbent assay (ELISA) antigen for detection of S. enteritidis infection. A total number of 49 positive sera and 40 negative sera were tested for ELISA validation. A cutoff value of 0.14 was shown to be sufficient to discriminate the negative and positive sera. Results obtained by testing sera raised against different bacterial strains/serotypes further confirmed that this recombinant flagellin-based ELISA was indeed specific for the detection of S. enteritidis. Both sensitivity and specificity of the developed ELISA test were comparable with a commercially available test, indicating that it is a highly promising and reliable diagnostic tool for S. enteritidis infection.  相似文献

An increase in the rate of human infections with Salmonella enteritidis (SE) occurred between 2007 and 2010 in British Columbia (BC). During the same time period, an increase in SE from poultry‐sourced isolates and increased clinical severity in poultry were also observed in BC. This article describes a multi‐sectoral collaboration during a 3‐year investigation, and the actions taken by public health and animal health professionals. Human cases were interviewed, clusters were investigated, and a case–control study was conducted. Environmental investigations were conducted in food service establishments (FSE). Suspect foods were tested. Laboratory data from poultry‐sourced isolates were analysed. Five hundred and eighty‐four human cases of SE with the same pulsed‐field gel electrophoresis pattern were identified between May 2008 and August 2010. Seventy‐three percentage of cases reported consumption of eggs. The odds of egg consumption were 2.4 times higher for cases than controls. Implicated FSE were found to use ungraded eggs, which had been distributed illegally. Investigation suggested that there were multiple suppliers of these eggs. Collaboration between public health and animal health professionals led to data sharing, improved understanding of SE, engagement with the poultry industry and public communication. Multi‐disciplinary, multi‐sectoral and multi‐pronged investigations are recommended to identify the likely source of illness in large, protracted foodborne outbreaks caused by commonly consumed foods.  相似文献

The objective of this study was to develop a multiplex polymerase chain reaction (PCR) to detect and differentiate food‐borne pathogens of the three genera Campylobacter, Arcobacter and Helicobacter in a single step procedure. One common reverse primer and three genus‐specific forward primers were designed by hybridizing to the 16S rRNA of selected reference strains. Besides the species with significance as food‐borne pathogens isolated from poultry meat –Campylobacter jejuni, Campylobacter coli, Arcobacter butzleri and Helicobacter pullorum– several other members of these genera were tested to determine the specificity of the designed multiplex PCR. In total, 20 ATCC and NCTC reference strains of Campyobacter, Arcobacter and Helicobacter were used to evaluate the PCR. Specific amplificates were obtained from all thermophilic species of Campylobacter as well as from species of Arcobacter and Helicobacter. No amplification product was obtained from the non‐thermophilic Campylobacter, C. hyointestinalis and C. fetus. Furthermore, a total of 43 field strains of the three genera isolated from poultry, pigs, cattle and humans were investigated using this PCR. To confirm the classification of 10 H. pullorum strains the 16S rRNAs were sequenced. The developed PCR is a helpful diagnostic tool to detect and differentiate Campylobacter, Arcobacter and Helicobacter isolated from poultry and poultry products.  相似文献

A sero-epidemiological survey of EDS '76 antibodies was carried out in chickens, guinea fowls, ducks and pigeons in various locations in Nigeria with the haemagglutination-inhibition test. Infection rates of 67.43, 79.68, 73.91 and 86.66% were obtained for chickens, guinea fowls, ducks and pigeons respectively. The high prevalence rates observed in these non-chicken species suggest that they should be considered as important in the epidemiology of EDS '76 in poultry in Nigeria. An extensive survey is recommended together with vaccination.  相似文献
