以动物实验的方法分析裂褶菌改善人体疲劳的情况,检测裂褶菌子实体干粉和发酵液对小鼠肝糖原含量、肌糖原含量、琥珀酸脱氢酶(succinate dehydrogenase,SDH)活力、乳酸脱氢酶(1actic dehydrogenase,LDH)活力、血清尿素氮(blood urea nitrogen,BUN)含量及血乳酸(1actic acid,LD)含量的影响。结果表明,试验Ⅰ组、试验Ⅱ组和试验Ⅲ组小鼠的肝糖原含量、SDH活力及LDH活力极显著高于对照组(饲喂100%基础饲料)(P<0.01);试验Ⅰ组和试验Ⅱ组的肌糖原含量极显著高于对照组(P<0.01);运动后试验Ⅰ组、试验Ⅱ组和试验Ⅲ组小鼠的BUN增量和LD的水平极显著低于对照组(P<0.01),说明裂褶茵具有明显解除运动疲劳的作用。  相似文献

为了探讨和筛选皱环球盖菇(Stropharia rugosoannulata)抗疲劳作用和活性部位,将160只昆明种小鼠分为2批,每批8组(80%乙醇提取物、石油醚萃取物、氯仿萃取物、乙酸乙酯萃取物、正丁醇萃取物、水层、阳性对照和空白对照组),每组10只,以红景天胶囊为阳性对照,每日灌胃1次,给药30 d。一批小鼠用来观察记录负重力竭游泳时间;另一批小鼠游泳30 min后,摘除眼球取血测定血清中尿素氮(blood urea nitrogen,BUN)、乳酸(lactic acid,LA)、乳酸脱氢酶(lactate dehydrogenase,LDH)、肌酸激酶(creatine kinase,CK)的值;取血后摘出左后肢股四头肌和肝脏,计算肝脏系数,分别测定左后肢股四头肌中肌糖原(muscle glycogen,MG)含量与肝脏中肝糖原(liver glycogen,LG)、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的值。结果表明:皱环球盖菇氯仿萃取物提高了52.93%的小鼠负重力竭游泳时间,也可明显提高LDH和SOD活力,提高肌肉组织与肝脏中糖原含量,小鼠体内LG含量达(11. 10±1.41) mg/g,运动后BUN、LA、CK、MDA水平不同程度下降,小鼠肝脏中MDA含量与空白对照组相比下降17.58%。皱环球盖菇具有抗疲劳作用,发挥抗疲劳作用的活性成分主要分布于氯仿分部。  相似文献

目的:探讨红菇抗运动性疲劳的功效。方法:以容县红菇子实体为材料,昆明小鼠为试验对象,采用不同剂量的红菇子实体提取液对试验组小鼠连续灌胃21d后,对试验组与对照组的小鼠游泳力竭时间、乳酸脱氢酶、肝糖原含量等指标进行检测,研究容县野生红菇子实体提取液抗运动性疲劳作用。结果:经21d连续灌胃红菇子实体提取液的小鼠负重游泳时间与对照组相比显著延长;肝糖原含量比对照组显著提高,乳酸脱氢酶活力比对照组显著降低。结论:红菇提取液均能增强机体对运动负荷的适应能力、抵抗疲劳的产生和加速疲劳的消除,具有一定的抗疲劳作用。  相似文献

研究灌胃灵芝(Ganoderma lingzhi)三萜白蛋白纳米体TbSAN(每次运动训练前2h按300mg/kg的剂量灌胃)与运动(每周进行4次训练,每次训练游泳45min)结合对小鼠体重、肥胖小鼠骨骼肌中游离脂肪酸(FFA)和肉毒碱棕榈酰转移酶-1(CPT-1)含量及脂肪酸转移酶CD36(FAT/CD36)蛋白表达量的影响。结果表明:实验28d后,Ⅱ组(运动)、Ⅲ组(灌胃灵芝三萜白蛋白纳米体)体重显著低于Ⅰ组(肥胖模型),Ⅳ组(灌胃灵芝三萜白蛋白纳米体结合运动)体重极显著低于Ⅰ组;Ⅱ组、Ⅲ组和Ⅳ组的骨骼肌FFA含量极显著低于Ⅰ组;Ⅱ组和Ⅲ组的骨骼肌中CPT-1含量和FAT/CD36蛋白表达量均显著低于Ⅰ组,而Ⅳ组极显著低于Ⅰ组。  相似文献

珊瑚状猴头菌多糖对大鼠肝抗氧化及代谢调节   总被引:1,自引:0,他引:1  
探讨珊瑚状猴头菌多糖的抗氧化作用和对大鼠肝脏代谢的相关基因表达的影响。将大鼠随机分为正常对照组、多糖干预组和模型组。对照组喂饲基础饲料,多糖干预组(剂量1组、剂量2组)和模型组喂饲高胆固醇饲料,同时多糖干预组(剂量1组、剂量2组)给予多糖灌胃5周,5周后处死大鼠取肝脏。分别测定大鼠肝脏的丙二醛(MDA)含量,总超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的活力。使用实时荧光定量PCR分别检测大鼠肝脏相关基因表达量:酰辅酶A:胆固醇酰基转移酶2(ACAT2)、卵磷脂胆固醇酰基转移酶(LCAT)。与模型组比较:多糖剂量1组、剂量2组大鼠肝脏的MDA含量显著下降(P〈0.01),T-SOD活力显著升高(P〈0.01),GSH-Px活力显著升高(P〈0.01),CAT活力显著提高(P〈0.01);多糖剂量1组、多糖剂量2组大鼠肝脏的基因表达量ACAT2mRNA显著下降(P〈0.01);LCATmRNA显著升高(P〈0.01)。珊瑚状猴头菌多糖具有显著的抗氧化作用,对大鼠肝脏的代谢具有调节作用。  相似文献

采用掌状玫耳(Rhodotus palmatus)粗多糖(低、高剂量分别为100、200 mg·kg-1)腹腔注射环磷酰胺所致免疫低下小鼠,测定脾脏、胸腺指数,血清中TNF-α、IL-1β、IL-6、溶血素含量,耳肿胀度,血液中白细胞、红细胞、血红蛋白、血小板含量,血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,血清中尿素氮(BUN)、肌酸酐(CRE)含量,并测定脾脏组织NF-κB通路相关蛋白表达量,研究掌状玫耳粗多糖对免疫低下小鼠免疫功能的影响。结果表明:与模型组相比,掌状玫耳粗多糖高、低剂量组小鼠胸腺、脾脏指数均显著增加;高剂量组TNF-α、IL-1β和IL-6含量显著增加;高、低剂量组小鼠耳肿胀度均显著增加;高剂量组小鼠血清溶血素含量显著增加;高剂量组小鼠血液中白细胞、红细胞、血红蛋白含量显著增加,高、低剂量组小鼠血液中血小板含量均显著增加;高剂量组小鼠血清中ALT活性显著降低,高、低剂量组小鼠血清中AST活性和CRE、BUN含量均显著降低;高、低剂量组TLR-4、IKKα、NF-κBp50表达量均显著升高,IκB-α表达量显著降低;高剂量组NF-κB p65表达量极显著升...  相似文献

为探讨茶树菇菌丝体孢内多糖的体内抗氧化活性,分别用水提、酸提、碱提菌丝体粗多特对小鼠灌胃给约(2000mg/kg),检测小鼠血清中超氧化物歧化酶(SOD)的活性以及肝组织中丙二醛(MDA)的含量。结果表明,与对照组相比,菌丝体多糖灌胃各组小鼠血清SOD活力显著增高(P〈0.05),同时肝组织中MDA的含量显著降低(P〈0.05)。  相似文献

用D-半乳糖建立小鼠衰老模型,将试验小鼠随机分8组,喂予不同剂量的虫草发酵产物,测定体重、脾脏指数、虹清中丙二醛含量、超氧化物歧化物酶活性以及小鼠疲劳耐受能力等。研究表明:虫草真菌发酵产物能显著提高小鼠SOD活性,降低MDA含量(P〈0.01),小鼠脾脏指数得到显著提高,耐疲劳能力也明显增强。结论:虫草发酵产物具有明显延缓衰老作用,并能提高小鼠的免疫力和抗疲劳能力。  相似文献

采用改良酚红(PSP)含量测定法,分别以正常、新斯的明负荷、肾上腺素负荷小鼠为实验研究模型,探索白灵菇对小鼠胃排空的影响。结果表明:白灵菇提取液(5.00g.kg^-1)1次性ig给药可显著抑制正常小鼠的胃排空(P〈0.01);对肾上腺素负荷小鼠引起的胃排空抑制没有明显的促进作用(P〉0.05);对新斯的明负荷小鼠引起的胃排空亢进也没有明显的拮抗作用(P〉0.05)。  相似文献

研究了转化桑叶的毛头鬼伞(Coprinus comatus)子实体对四氧嘧啶诱导的糖尿病小鼠体重、血糖、血清胰岛素、肝糖原、果糖胺、血脂代谢、体内氧化压力、肝脏中糖代谢酶活性等的影响.转化桑叶的鸡腿菇子实体提高了单一毛头鬼伞子实体或桑叶的降血糖作用,促进了胰岛素分泌,降低了体内氧化压力,提高了糖代谢的酶活性,但对血脂调节作用不显著.  相似文献

AIM: The aim of this research is to study the earlier enzyme activity changes of the diaphragm in diabetic rats. METHODS: An enzyme histochemical method was used to observe the changes in the enzyme activities of dehydrogenases,hydrolases and oxidases in 4th week diabetic rat diaphragm. RESULTS: The activites of enzymes including SDH(Succinate dehydrogenase),MDH(Malate dehydrogenase), GDH(Glutamate dehydrogenase), ICDH(Isocitrate dehydrogenase), NADHD(NADH diaphorase), G-6-PD(Glucose-6-phosphate dehydrogenase), ACP(Acid phosphatase) and ANAE(Acid α-naphtyl acid esterase) were increased in diabetic diaphragm compared with the control. LDH (Lactate dehydrogenase)and CCO(Cytochrome oxidase) activities were decreased, whereas NADPHD(NADPH diaphorase) showed no changes in diabetic rats. Eleven kinds of enzyme were analysed with image analysis.Optical density (A) of SDH, MDH, GDH, ICDH, NADHD, G-6-PD, ACP and ANAE in diaphragm of diabetic rats were significantly higher than that of control rats (P<0.01). A value of LDH and CCO in diaphragm from diabetic rats were significantly lower than that of control rats (P<0.01). A value of NADPHD in diaphragm from diabetic rats showed no apparent alteration compared with the control rats(P>0.05). CONCLUSION: Increase in the aerobic capacity, decrease in the glycolytic capacity, and disturbance of lipid and energy metabolism were found in diaphragm of 4th week diabetic rats.  相似文献

AIM: To observe the effect of simvastatin on myocardial tissue after renal ischemia-reperfusion injury and its mechanism. METHODS: A rat model of renal ischemia-reperfusion injury was prepared by clamping the bilateral renal arteries for 45 min. The rats (n=36) were randomly divided into sham operation group, renal ischemia-reperfusion (I/R) group and simvastatin group with 12 rats in each group. The content of serum creatinine (SCr), blood urea nitrogen (BUN) and myocardial tissue malondialdehyde (MDA), the myocardial activity of lactate dehydrogenase (LDH), creatine kinase (CK) and superoxide dismutase (SOD), and the myocardial protein expression of Bcl-2 and Bax were detected. RESULTS: Compared with sham operation group, the content of SCr, BUN and myocardial MDA, and the myocardial activity of LDH and CK in I/R group were significantly increased (P<0.05), and the activity of SOD was significantly decreased (P<0.05). Compared with I/R group, the content of SCr, BUN and myocardial MDA, and the myocardial activity of LDH and CK in simvastatin group were significantly decreased (P<0.05), while SOD activity was enhanced (P<0.05). The protein expression of Bcl-2 and Bax in sham operation group was less than that in I/R group (P<0.05), and the protein level of Bax in simvastatin group was significantly lower than that in I/R group (P<0.05), while the protein level of Bcl-2 was increased (P<0.05). CONCLUSION: Simvastatin has a protective effect on the myocardium of the rats with renal ischemia-reperfusion injury, and the protective mechanism may be related to the elimination of free radicals by simvastatin, increase in the protein expression of Bcl-2 and decrease in the protein expression of Bax.  相似文献

AIM: To determine the beneficial effects of icariin on streptozotocin (STZ)-induced diabetic testopathy in rats. METHODS: The diabetic animal model was induced in male Sprague-Dawley rats by an injection of streptozotocin (40 mg/kg, iv). The rats were randomly divided into 3 groups: control group, model group and icariin (80 mg/kg, ig) group. Twelve weeks after injected with streptozotocin, all rats were anaesthetized and killed to remove the testes from scrotum. Serum concentrations of glucose and testosterone, and the levels of succinate dehydrogenase (SDH), acid phosphatase (ACP), γ-glutamyl transpeptidase (γ-GT) and lactate dehydrogenase (LDH) in testes were measured. The morphology of the testicular tissues was observed under light microscope. Immunohistochemistry was employed to determine the protein levels of TGF-β1 and type Ⅳ collagen. RESULTS: Compared with control group, the content of serum glucose increased while the serum level of testosterone and the activitiy of SDH, ACP, γ-GT and LDH in testis decreased in model group (P<0.01). The histopathological examination showed that the diameters of seminiferous tubules and various grades of spermatocytes in the testis were markedly decreased. Compared with control group, the expression of TGF-β1 and collagen Ⅳ was significantly increased in model group. These alterations were significantly attenuated in icariin group (P<0.01). CONCLUSION: Icariin evidently relieves testicular damage in rats with diabetic testopathy by improving the secretion of testosterone and reducing the expression of TGF-β1 and collagen Ⅳ at protein level.  相似文献

以棉籽壳培养料为对照,测定了烟叶渣培养料中糙皮侧耳(Pleurotus ostreatus)的菌丝满袋时间及子实体中氨基酸、无机盐、重金属和烟碱的含量.结果表明,当配方为79%烟叶渣、10%谷壳、10%麸皮,辅料添加量为1%(其中石膏:碳酸钙:过磷酸钙=2:2:1)时,糙皮侧耳的采收时间比棉籽壳组缩短了3d,且子实体中有15种氨基酸的含量高于棉籽壳组,重金属汞、铅、镉、砷的含量均符合《中华人民共和国农业部行业标准》(绿色食品食用菌NY/T 749-2003),烟碱含量<7.8mg/kg.  相似文献

采用麦芽汁液体培养基培养蜜环菌获得菌丝体,采用麦芽汁半固体培养基培养蜜环菌获得蜜环菌菌素、皮壳状菌丝和子实体。以高效液相色谱-二极管阵列检测法(High Performance Liquid Chromatography-Diode Array detector,HPLC—DAD)比较四种菌体的甲醇提取物的成分差异。液体培养的蜜环菌菌丝体与固体培养的蜜环菌菌素、皮壳状菌丝和子实体的蜜环菌素类成分有明显差别。图谱分析表明,菌素与皮壳状菌丝的成分接近,与子实体和液体培养菌丝体之间有较大差别。  相似文献

AIM: To study the effect of chronic corticosterone (CORT) injection on the depression-like behaviors and the brain glycogen level in mice. METHODS: Male C57BL/6N mice (n=40) were randomly divided into normal control group and model group. The mice in model group were subcutaneously consecutively injected with CORT for 4 weeks. The mouse model of chronic stress depression was constructed. The forced swim test and open field experiment were conducted to prove chronic stress model. The serum level of CORT in the mice was measured by radioimmunoassay. The protein levels of hippocampal synaptophysin (SYP) and brain-derived neurotrophic factor (BDNF) were detected by Western blot. Hippocampus glycogen, glycogen synthase and glycogen phosphorylase were determined by indirect fluorescence measurement. RESULTS: Compared with normal control group, the immobility time of the forced swim test in model group was significantly lengthened (P<0.01), and the ability of spontaneous activity was reduced (P<0.01), indicating that chronic CORT injection induced depression-like behaviors in mice. The CORT level increased significantly (P<0.01) in model group. CORT injection decreased the protein expression of hippocampal SYP and BDNF (P<0.01), reduced hippocampal glycogen level (P<0.05) and glycogen synthase activity (P<0.05), and increased glycogen phosphorylase activity (P<0.05). CONCLUSION: Chronic CORT injection causes hippocampal neuron damage and induces the depression-like behaviors of mice, which may be associated with decreasing hippocampal glycogen level by CORT.  相似文献

采用超高效液相色谱(UPLC)-二极管阵列检测器(PDA)和液相二级质谱(LC/MS/MS)方法测定多菌灵在添加浓度范围分别为0.025~1 mg/kg和0.05~10 mg/kg 时在糙皮侧耳(Pleurotus ostreatus)子实体和培养料中的残留量,结果表明:多菌灵在子实体和培养料中的平均添加回收率分别为93%~99%、93%~101%,相对标准偏差(RSD)均为3%~6%。在0.1%、0.3%多菌灵有效成分用量下拌料施用后,糙皮侧耳第一和第五潮菇(菇柄、菇盖和全菇)中多菌灵残留量分别为0.089~0.077、0.533~0.485 mg/kg,低于欧盟关于多菌灵在新鲜菇类中的最大残留限量标准。多菌灵在子实体各个部位的富集能力由强到弱为菇脚>菇柄>全菇>菇盖。培养料中多菌灵的消解符合一级动力学模型,菌袋部位、菌丝和灭菌对培养料中多菌灵消解均有影响:菌袋上部(距离栽培袋底部22 cm处)的培养料中的多菌灵消解快,半衰期33 d短于中部(距离栽培袋底部11 cm处)的36.47 d和下部的63 d;培养料中菌丝生长有利于多菌灵消解,高压灭菌(0.1 Mpa 压力下,121℃灭菌2 h)可以有效地促进农药消解。灭菌后降解率达91.8%和85.9%,未灭菌时培养料中多菌灵含量基本稳定。  相似文献

用气相色谱-质谱法对双孢蘑菇(Agaricus bisporus)和香菇(Lentinula edodes)中67种农药进行检测。样品经乙腈均质提取,盐析除水,氨基柱净化后用气相色谱-质谱仪分析,在选择离子监测模式下以特征离子的保留时间定性定量测定67种农药。在添加农药标准品浓度为0.05、0.10和0.25 mg/kg水平上,大多数农药的平均回收率在70%~120%之间,检出限在0.01~0.5μg/kg之间。  相似文献
