为探寻一种油料蛋白提取方法,该研究以双低冷榨菜籽饼为原料,碱性电解水为溶剂,通过单因素与响应面试验确定了碱性电解水提取冷榨菜籽饼分离蛋白(Alkaline electrolyzed water extracted Rapeseed Protein,ARP)的优化条件,并与相同条件下碱溶酸沉法提取菜籽分离蛋白(Ultrapure water extracted Rapeseed Protein,URP)的结果进行对比。结果表明: 提取工艺各环节因素对菜籽蛋白提取率影响的主次顺序分别为:碱性电解水pH值、温度、料液比、时间;提取优化工艺条件为温度45 ℃、碱性电解水pH值11.5、料液比1:10 g/mL和提取时间60 min,蛋白质提取率为59.34%。与相同条件下碱溶酸沉法相比,碱性电解水提取的菜籽分离蛋白游离巯基和二硫键含量高、表面疏水性强、粒径小、ζ电位绝对值大,同时其提取率、溶解性、持油性、乳化性和起泡性均显著得到改善(P<0.05);此外,ARP内源荧光光谱强度更高,二级结构更有序。由此可知,该研究结果为碱性电解水提取冷榨菜籽饼蛋白提供参考,不仅提高了提取率,而且最大程度地减少了对菜籽蛋白结构和功能特性的破坏。  相似文献

该文首次提出了以低温预榨花生饼为原料,经Viscozyme酶预处理后再碱溶酸沉提取蛋白质的新工艺,研究了酶处理条件对花生蛋白提取率的影响,通过中心组合试验设计和响应面分析优化的试验结果是:当花生饼浓度为15%,温度为45℃,酶用量1.26%,pH值为4.3,反应时间为134 min时,蛋白提取率为79.38%,而未用酶处理者蛋白提取率为58.35%,表明酶法预处理花生饼可以显著改善花生蛋白的提取效果。  相似文献

模糊评判优化水酶法提取膨化大豆油脂和蛋白   总被引:11,自引:2,他引:9  
针对水酶法同时提取大豆油与蛋白时,两种目标对工艺参数的优化有差别,因此应用模糊综合评判法对工艺参数进行双目标综合优化,得到同时兼顾高油脂提取率与高蛋白提取率的酶解工艺参数。其结果为:加酶量1.85%,酶解温度50℃,酶解时间3.6 h,料液比1︰6,pH值为9。此时,油脂提取率为92.76%左右,蛋白提取率为93.81%左右。利用扫描电子显微镜、能谱分析以及显微切片分别就蛋白水解状态对油脂释放影响机理进行了研究。通过研究可知,挤压膨化再粉碎后的大豆粉水解过程中油脂的释放取决于蛋白水解状态,经过挤压膨化后大豆细胞结构已经被较充分破坏,而大豆油脂与蛋白的作用没有被完全破坏,所以需要把蛋白充分水解才能使油脂释放聚集,因此挤压膨化后水酶法可以兼顾得到高油脂提取率和高蛋白提取率。  相似文献

脱脂麦胚蛋白粉是一种优质植物蛋白资源,采用均匀设计方法研究了pH值(4.0~8.0)、加热温度(20~70℃)和加热时间(10~30min)对其持水能力和蛋白溶解度的影响。随着温度的升高,持水能力显著增强,pH值的影响相对较小,而温度的影响可以忽略。pH值为4.0,温度为70℃时,持水能力最强。pH值对蛋白溶解度的影响显著,时间的影响可以忽略。pH值为8.0,温度为20℃时,蛋白溶解度最大。研究结果还表明,脱脂麦胚蛋白粉可作为食品添加剂以改善食品的营养和品质。  相似文献

该文通过中心组合设计及相应面分析,研究了碱法制备蚕蛹蛋白的浸提条件对蚕蛹蛋白浸提率的影响,得到了蚕蛹蛋白提取率的数学模型。优化了蚕蛹蛋白的浸提的条件,提高了碱法制备蚕蛹蛋白的蛋白提取率。结果表明NaOH浓度和液固比(V/m)对蚕蛹蛋白提取率影响显著(P<0.05)。蚕蛹蛋白浸提的最佳工艺条件为:NaOH浓度1.03%、液固比(V/m)70︰1、浸提温度50℃、NaCl浓度0.070 mol/L。在此最佳工艺条件下蚕蛹蛋白的实际提取率达78.69%。利用优化工艺条件制备的桑蚕蛹蛋白,其蛋白含量为96.70%,必需氨基酸占总氨基酸含量的47.1%,必需氨基酸与非必需氨基酸的比值为0.9,具有较好的应用价值。  相似文献

采用盆栽方法研究了水分处理对专用小麦各营养器官氮含量积累、运转和子粒蛋白质的影响。结果表明,在不同的水分处理中,开花后随着时间的推移小麦叶鞘和茎中氮含量逐渐降低,而根中氮含量先下降后上升。(1)80%FC提高了各专用小麦叶鞘氮含量;豫麦34茎中氮含量在各处理下差异不大,豫麦49在60%FC下提高茎中氮含量,豫麦50茎氮含量在40%FC下最高。(2)各营养器官氮转运量品种间表现为豫麦50〉豫麦49〉豫麦34,各品种间不同水分处理下各营养器官氮转运量均为80%FC〉60%FC〉40%FC。(3)各品种叶鞘氮转运效率为80%FC〉60%FC〉40%FC;但不同品种间各水分处理下茎氮转运效率因存有差异,豫麦34为:40%FC〉60%FC〉80%FC,豫麦49为:60%FC〉40%FC〉80%FC,豫麦50为:60%FC〉80%FC〉40%FC。(4)各营养器官及总转运氮对籽粒的贡献率为80%FC〉60%FC〉40%FC,各品种器官转运量和贡献率均为:叶鞘〉茎,器官转运效率为:茎〉叶鞘。(5)对豫麦34,蛋白质含量和产量均以60%FC为好,且蛋白产量达显著性差异;豫麦49和豫麦50,蛋白质含量以40%FC最高,蛋白质产量以80%FC最高。豫麦49差异均达到显著水平,豫麦50蛋白质产量达极显著水平。  相似文献

超声辅助提取冻干番茄粉番茄红素的工艺优化   总被引:1,自引:1,他引:0  
为了提高番茄原料中番茄红素的提取率,增加番茄深加工的利用价值,该文研究了冻干番茄粉粒度范围、液料比、超声温度、超声比功率和超声处理时间5个因素对番茄红素提取率的影响,通过响应面分析优化出超声辅助提取(ultrasound assisted extraction, UAE)冻干番茄粉(<74 μm)中番茄红素的最佳工艺,并与常规溶剂提取(conventional solvent extraction, CSE)进行了比较。确定的最优工艺条件为:液料比41:1 mL/g、超声温度55℃、超声比功率18 W/g,超声处理时间15 min。响应面验证试验番茄红素提取率为1.82±0.05 mg/g(超声时间10 min),与优化模型预测值1.83 mg/g基本一致,验证了模型的可靠性。UAE的番茄红素提取率较CSE增加了6.04%,提取时间节省了92.47%。该文为番茄红素的高效提取及番茄的综合利用提供了参考。  相似文献

为优化藜麦淀粉碱法提取工艺,以藜麦为试验材料,采用岭脊分析法研究料液比、NaOH 质量分数和浸提时间对淀粉提取率的影响,并对藜麦淀粉的颗粒形貌、化学结构及淀粉糊化特性进行研究。结果表明,碱法提取藜麦淀粉的优化工艺参数为料液比1:5 g/mL、NaOH质量分数0.2%和浸提时间5.5 h,淀粉提取率为98.94%±0.26%;藜麦淀粉为限制性膨胀淀粉,形态多呈不规则形,具有-OH、-CH2、-CHO、C-O-C和吡喃环典型淀粉分子官能团;随静置时间增加,淀粉透光率降低,凝沉体积增加,12 h后基本稳定;藜麦淀粉糊第四次冻融循环后达到稳定状态;对淀粉凝胶质构特性分析,硬度、内聚性、弹性、胶黏性和咀嚼性5个指标都随藜麦淀粉糊浓度增加而增大;流变性分析表明藜麦淀粉糊为假塑性流体,其弹性优于黏性。该文系统研究藜麦淀粉提取工艺和糊化特性,拓展了新的淀粉资源,同时也为藜麦淀粉的生产和应用提供一定的借鉴和参考。  相似文献

小麦胚芽资源的开发利用对于缓解中国十分紧缺的蛋白和油脂资源,提高人类膳食营养与健康水平具有重要的现实意义。该文在分析中国小麦胚芽资源分布与品质状况、小麦胚芽的利用现状以及存在问题的基础上,从小麦胚芽高效提取、稳定化、小麦胚芽油萃取、小麦胚芽微生物转化等方面,论述了关于小麦胚芽深加工技术与装备的国内外最新发展趋势。最终提出了从心磨系统提取高纯度小麦胚芽、采用微波在线稳定化小麦胚芽、亚临界流体萃取小麦胚芽油、小麦胚芽粕进一步发酵制备抗肿瘤功能食品的技术集成方案,旨在为面粉加工企业进一步深入开发利用小麦胚芽资源提供参考。  相似文献

麦粒姿态的自动判别,是近红外高光谱成像系统自动检测多姿态麦粒内部虫害的前提。依据麦粒目标在最优波长图像中的坐标、重心等信息,从高光谱数据立方体中自动分割出单个完整麦粒的子图像。利用麦粒胚部端粗糙度较大的原理,依据纹理、不变矩、均值等13个可能胚部区域特征的判别正确率,确定不变矩4为判别麦粒胚部区域的有效特征。针对麦粒胚部区域,提取梯度图像和二值图像的26个特征,利用人工鱼群算法选择出延伸率、胚部区域对称度、延伸率等13个特征。选取1 200个样本进行训练,600个样本进行检验,利用最大离差法自动确定13个特征的模糊权重,麦粒3个姿态可拓分类的正确识别率为94.5%,证实了基于局部区域特征的麦粒姿态自动识别的可行性。  相似文献

Defatted wheat germ protein (DWGP) was isolated by alkaline extraction at pH 9.5 and subsequent isoelectric precipitation at pH 4.0, and its nutritional and functional properties were studied. The results showed that the amino acid content of defatted wheat germ was as high as 26.793 g/100 g, and the contents of eight essential amino acids were all relatively high. The isoelectric point of DWGP was 4.0. When pH >6.0, the DWGP had high solubility with a nitrogen solubility index of 70%. The emulsifying activity and emulsifying stability of DWGP were similar to those of bovine serum albumin and a little higher than those of casein. DWGP had good foaming capacity, but its foaming stability (FS) was not very good. However, the FS of DWGP can be improved through physical, chemical, or enzymatic methods. Moreover, DWGP had excellent water retention (WR); especially at pH 8.0 and a temperature of 70 degrees C, the WR of DWGP was the highest at 229.4%. DWGP offers is a potential source of functional protein isolate for possible food applications.  相似文献

粮食加工副产物的深加工与综合利用是中国粮食工业发展的重要方向,为了准确获得中国小麦资源总量、分布以及品质状况,在调查中国小麦胚芽资源分布基础上,从中国小麦主产区的大型面粉企业采集小麦胚芽样品,分析商用小麦胚芽的主要营养组成以及微生物污染状况。结果表明,中国每年实际可利用小麦胚芽约20万t,资源主要集中在山东、河南、河北等省;商用小麦胚芽的平均酸价(以KOH计)22.8mg/g,夏季最高达60.4mg/g;其平均含水率为12.1%、蛋白质质量分数为33.70%(其中赖氨酸质量分数为2.10%)、油脂质量分数为11.12%(其中亚油酸质量分数为58.34%),α-生育酚质量分数为0.23%;其菌落总数平均为2.65×104cfu/g。小麦胚芽资源调查与品质评估结果,对中国粮食工业发展规划和粮食副产物深加工企业决策将具有重要的参考价值。  相似文献

为深入探究小麦胚芽中的脂质成分,该研究采用不同溶剂分别提取小麦胚芽中的磷脂和中性脂质,利用核磁共振技术 (nuclear magnetic resonance,NMR)对小麦胚芽中的脂质成分进行分析,结果表明:31P NMR在小麦胚芽中检测到6种磷脂和相对应的5种溶血性磷脂,以及非脂质含磷化合物甘油磷脂酰胆碱(GPC);磷脂中含量最高的是磷脂酰胆碱(PC),其摩尔浓度为0.42 μmol/g,摩尔分数为28.50%,质量浓度为0.31 mg/g,质量分数为30.2%;1H NMR测定小麦胚芽中的多种中性脂质组成及含量,包括甘油三酯(TG)、甘油二酯(DG)、甘油单酯(MG)和游离脂肪酸(FA),其中含量最高的甘油酯是TG,占比为77.25%,含量最低的是2-甘油单酯(2-MG),占比为0.03%;小麦胚芽的甘油三酯和磷脂中检测出亚油酸(L)、油酸(O)和亚麻酸(Ln)等6种不饱和脂肪酸,其中亚油酸的含量最高,在甘油三酯中占比56.26%,在磷脂中占比45.37%。溶血性磷脂和GPC是磷脂的水解产物,DG、MG和FA是甘油三酯的水解产物,这些物质可以反映样品中脂质的水解程度。研究结果表明,利用NMR不仅能够对小麦胚芽脂质的组分进行定性定量分析,而且可以监测小麦胚芽脂质水解程度的变化。NMR技术在小麦胚芽相关产品的脂质分析研究具有重要作用。  相似文献

Residual malathion in wheat was estimated at a lower value when analysis was performed by extraction with acetone after addition of water to swell the wheat, according to the Japanese Bulletin Method. The supernatant of the wheat homogenate showed degradation not only of malathion but also of phenthoate. Malathion and phenthoate were not degraded by the boiled supernatant of the wheat homogenate. It was presumed for this reason that glutathione reductase (GR; EC 1.6. 4.2) in the wheat degraded malathion. The following results were obtained: (1) GR originating in wheat could degrade malathion and phenthoate. (2) The degradation of malathion by the GR was inhibited by excessive GSSG. (3) There was a high correlation between GR activity and malathion degradation activity of the supernatant of wheat homogenates. It is likely that GR acted on the specific structure of malathion and phenthoate, the S=P-S bond, and the blanch structure bonding with the sulfur atom. Following the above, extraction with acetone after addition of water (the Japanese Bulletin Method) should be replaced by extraction with pure organic solvent and without addition of water for swelling.  相似文献

The incubation conditions of wheat germ for angiotensin I-converting enzyme inhibitory activity (ACEI) elevation and peptide accumulation were investigated, and five ACE inhibitory peptides were obtained. The effect of individual factors such as incubation time, temperature, initial pH, and liquid to solid ratio on ACEI and peptide concentration of incubation medium was evaluated, respectively. The combinations of four factors were further optimized using a Box-Behnken design. Under the best incubation condition (pH 4.4 with a liquid to solid ratio 8.14 mL/g at temperature 47 °C, for 7 h), maximum ACEI (92.16%) and peptide concentration (88.12 mg/g) were obtained, which were 6.2- and 2.4-fold, respectively, as compared to the unincubated wheat germ. After they were purified, five ACE inhibitory peptides, VEV, W, NPPSV, QV, and AMY, were identified by liquid chromatography/tandem mass spectrometry. The IC(50) were 115.20, 94.87, 40.56, 26.82, and 5.86 μM, respectively.  相似文献

An efficient supercritical fluid extraction (SFE) process with carbon dioxide (SFE-CO(2)) was developed for the extraction of natural vitamin E (V(E)) from wheat germ. Both the pretreatment of extracted wheat germ and extraction conditions were optimized to ensure maximal V(E) yield. The extraction was undertaken at the extracting pressure of 4000-5000 psi, the extracting temperature of 40-45 degrees C, and the carbon dioxide flow rate of 2.0 mL/min for 90 min. An optimized pretreatment of wheat germ was usually necessary with a particle size of 30 mesh and a moisture content of 5.1%. A yield comparison of V(E) and its isomers extracted by supercritical CO(2) with those by conventional solvent extraction suggested that this SFE process was a practical process prospectively superior to conventional solvent extraction to prepare V(E) from wheat germ.  相似文献

The oxidation of ferulic acid (FA) or 5-O-(trans-feruloyl)-L-arabinose (EFA) by a purified wheat germ peroxidase was followed by UV spectrophotometry and high-performance liquid chromatography using an electrochemical detection. Wheat peroxidase (POD) exhibits a ping-pong bireactant mechanism forming phenoxy radicals more rapidly from FA than from EFA in routine assay conditions. When both the free and the esterified forms of FA are present, the reverse was found. This result could be due to a nonenzymatic cooxidation of FA by the phenoxy radicals of EFA leading to the formation of phenoxy radicals of FA and the EFA regeneration. Addition of ascorbic acid (AA) provokes a delay of FA consumption. AA reduced very rapidly the phenoxy radicals formed by POD back to initial phenol avoiding the formation of ferulate dimers until it was completely oxidized in dehydroascorbic acid. Conversely, cysteine addition slowed but did not delay the FA consumption. The thiol reduced a fraction of the phenoxy radicals produced by wheat POD and was oxidized into cystine, while the other part of phenoxy radicals formed ferulate dimers. These results could be of interest to understand the POD effect on the wheat dough rheological properties.  相似文献

在粮食质量等级评定中,黑胚粒存在的多少影响小麦的整体等级划分。该文以产自新疆和河南不同品种的710粒有黑胚的小麦、627粒破损小麦和1 167粒正常小麦为研究对象,采用彩色线阵CCD动态获取小麦图像,对原始图像进行多余背景去除、色偏校正、亮度变换以及形态学处理后,再对小麦图像单通道图像进行自适应双阈值分割,能有效的实现对小麦黑胚特征的分割。试验选择三种小麦进行特征提取以及模式识别,得到最佳分辨率分别为95.1%,96.0%和98.3%。结果表明该方法对小麦黑胚的识别具有良好的效果。  相似文献
