Eight- and nine-week-old Hungarian Landrace pigs were tested with halothane as described by Laky et al. (1985). Immediately after the test blood samples were taken for determination of the activity of serum creatine kinase (CK), creatine kinase MB (CK-MB) isoenzyme, aldolase (ALD), lactate dehydrogenase (LDH) and alpha-hydroxybutyrate dehydrogenase (alpha-HBDH). Elevated creatine kinase, creatine kinase MB isoenzyme and aldolase activities indicating enhanced susceptibility to stressors were found in 92% of the halothane reacted and 16% of the halothane non-reacted animals. In these individuals the activities of lactate dehydrogenase and alpha-hydroxybutyrate dehydrogenase were also high. Data of the literature show a close relationship between enhanced susceptibility to stressors and halothane reaction in pigs. It was suggested, therefore, that determination of the activity of appropriate serum enzymes might be used for detecting this enhanced susceptibility.  相似文献

Five Swedish Landrace pigs with a mean weight of 51 +/- 5 kg performed an exercise test on a treadmill at a speed of 1.8 m/s and a duration of 10 min. Hypophosphatemia was then induced in these pigs by addition of aluminium hydroxide (liquid antacid) to the normal feed. After 3 weeks, the exercise test was repeated when the mean weight of the pigs was 65 +/- 9 kg. Five other Swedish Landrace pigs with a mean weight of 72 +/- 4 kg performed a similar exercise test. Muscle biopsies from M. biceps and blood samples were taken from all pigs 3-5 days before and immediately after each exercise test. Hypophosphatemic pigs had significantly lower serum phosphate and higher aluminium levels than normophosphatemic pigs. In all pigs, glycogen content in muscle decreased significantly (-108 to -135 mmol/kg muscle) with exercise while no changes were seen in adenosine triphosphate, creatine phosphate or inorganic phosphate concentrations. In normophosphatemic pigs, glucose-6-phosphate and lactate concentrations increased significantly during exercise by 2-4 mmol/kg and 12.8-14.4 mmol/kg, respectively. However, in hypophosphatemic pigs, glucose-6-phosphate concentrations decreased significantly during exercise by 4.4 mmol/kg and lactate levels were unchanged. These results indicate that low serum inorganic phosphate levels influence muscle metabolism and glycolysis in connection with physical exercise.  相似文献

The distribution of lactic dehydrogenase, aldolase and creatine kinase in various horse tissues was determined. Using polyacrylamide gel electrophoresis the lactic dehydrogenase and creatine kinase isoenzyme composition of horse serum, taken before and after exercise, was studied. Horse tissue isoenzyme patterns were also obtained. By comparing tissue and serum patterns, skeletal muscle was found to be the tissue of origin of the increase in serum lactic dehydrogenase and creatine kinase observed after exercise.  相似文献

Pannon White growing rabbits (a group of 8) were exposed to treadmill exercise (3-9 m/s, 1.2-1.6 km/day) twice a day for 4 weeks, while additional 8 animals, kept inactive, were assigned as the control group. Weekly, 12 hours after exercise, venous blood was taken for serum metabolite and enzyme activity measurements. Total serum protein, albumin and creatinine levels significantly increased during the second half of the training, as compared to the control group. Triacylglycerol levels in the exercised group as compared to controls, however, were higher only after the first and the fourth weeks of the experiment. Resting non-esterified fatty acid (NEFA) concentration of the trained rabbits was lower at the end of the trial. On the other hand, there were no significant differences, as compared to the respective controls, in serum urea, total and HDL cholesterol levels. At the end of the exercise alkaline phosphatase activity was higher and total lactate dehydrogenase activity was lower in the trained rabbits. Serum alanine aminotransferase, aspartate aminotransferase and gamma-glutamyl transpeptidase activities were not changed, while creatine kinase activity was slightly lower in the trained group. The serum cortisol concentration was not different in the trained and control rabbits.  相似文献

High intensity exercise is associated with production of energy by both aerobic and anaerobic metabolism. Conditioning by repeated exercise increases the maximal rate of aerobic metabolism, aerobic capacity, of horses, but whether the maximal amount of energy provided by anaerobic metabolism, anaerobic capacity, can be increased by conditioning of horses is unknown. We, therefore, examined the effects of 10 weeks of regular (4-5 days/week) high intensity (92+/-3 % VO2max) exercise on accumulated oxygen deficit of 8 Standardbred horses that had been confined to box stalls for 12 weeks. Exercise conditioning resulted in increases of 17% in VO2max (P<0.001), 11% in the speed at which VO2max was achieved (P = 0.019) and 9% in the speed at 115% of VO2max (P = 0.003). During a high speed exercise test at 115% VO2max, sprint duration was 25% longer (P = 0.047), oxygen demand was 36% greater (P<0.001), oxygen consumption was 38% greater (P<0.001) and accumulated oxygen deficit was 27% higher (P = 0.040) than values before conditioning. VLa4 was 33% higher (P<0.05) after conditioning. There was no effect of conditioning on blood lactate concentration at the speed producing VO2max or at the end of the high speed exercise test. The rate of increase in muscle lactate concentration was greater (P = 0.006) in horses before conditioning. Muscle glycogen concentrations before exercise were 17% higher (P<0.05) after conditioning. Exercise resulted in nearly identical (P = 0.938) reductions in muscle glycogen concentrations before and after conditioning. There was no detectable effect of conditioning on muscle buffering capacity. These results are consistent with a conditioning-induced increase in both aerobic and anaerobic capacity of horses demonstrating that anaerobic capacity of horses can be increased by an appropriate conditioning programme that includes regular, high intensity exercise. Furthermore, increases in anaerobic capacity are not reflected in blood lactate concentrations measured during intense, exhaustive exercise or during recovery from such exercise.  相似文献

The stability and storage characteristics were studied of 11 bovine enzymes of potential clinical significance, namely, aldolase, alkaline phosphatase, alanine aminotransferase, aspartate aminotransferase, acetylcholinesterase, creatine kinase, gamma glutamyltransferase, glutathione peroxidase (GSH-Px), alpha-hydroxybutyrate dehydrogenase, lactate dehydrogenase and superoxide dismutase (SOD). Enzyme activities in fresh serum were compared with those in plasma containing various anticoagulants including lithium heparin, EDTA and oxalate/fluoride. The same preservatives were assessed for their effects on the whole blood activities of GSH-Px and SOD. Stabilities of enzymes in plasma and serum stored at room (+20 degrees C), refrigerator (4 degrees C) or deep freeze (-20 degrees C) temperatures were also compared. In addition, SOD and GSH-Px activities in samples stored, at the same temperatures, as whole blood or aqueous lysates were monitored.  相似文献

Serum enzyme concentrations were determined in 319 Hampshire pigs (approx 2 months of age). Significant positive correlations were found when creatine kinase (CK) concentrations were compared with lactate dehydrogenase and malate dehydrogenase values, but not when compared with aspartate aminotransferase or alanine aminotransferase values. In an additional study, a litter from a sow with high serum CK concentrations was observed at 2, 3, 4, and 5 months of age for serum CK concentrations. The CK concentration increased as the pigs aged, and soft exudative pork was found upon slaughter of the pigs at 5 months of age. It is believed that CK, lactate dehydrogenase, and malate dehydrogenase determinations in weanling pigs would have value for reducing the frequency of the porcine stress syndrome and soft exudative pork in breeding stock. If only a portion of the genetic carriers was removed, the effect would be considerable within a few generations.  相似文献

Five Siberian Husky Dogs participated in an initial study to determine their physiological response to three types of exercise. Blood samples were taken prior to, and three minutes following, a 7.5 km free run and 6 km team sled run for the determination of hemoglobin, hematocrit, red and white blood cell counts, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, lactate dehydrogenase, creatine phosphokinase and serum glutamic pyruvic transaminase. Samples were also taken following a 90s sprint run. Heart rate was taken immediately after each run by palpation of the femoral pulse. Average heart rates following the 90s, 7.5 km and 6 km runs were 190 bpm, 211 bpm and 166 bpm, respectively. Mean lactate concentrations following the 90s, 7.5 km and 6 km runs were 1.74 mMol · 1−1, 0.70 mMol · 1−1, and 3.06 mMol · 1−1, respectively. Elevation of lactate dehydrogenase and creatine phosphokinase was greatest following the 6 km sled run. Three of the above dogs were then studied before and after a 12 week interval training program, while three other dogs served as controls. The animals completed a three stage, submaximal treadmill test prior to and following the program. Pre and post blood samples were taken, and rectal temperature and heart rate were recorded continuously throughout the test. An analysis of variance was used to examine the significance of differences between and within groups. Although the response of heart rate, lactate, temperature and serum enzymes to submaximal exercise did not change with training significant differences between groups suggested that a more prolonged program may have resulted in such adaptations.  相似文献

The activities of serum creatine kinase (CK), lactate dehydrogenase (LD) and LD isoenzymes were studied in 14 Prestice black pied pigs from a herd affected with congenital tremor. Mean CK activity was 19.57 +/- 3.56 mu kat.l-1 for 6 adult pigs, and it was 21.03 +/- 1.33 mu kat.l-1 and 20.42 +/- 1.23 mu kat.l-1 for the affected (n = 5) and control (n = 3) piglets, respectively. No significant differences were demonstrable between the groups in CK activity. Total serum LD and LD-4 as well as LD-5 isoenzyme activities were higher in sows. Piglets affected with congenital tremor showed an increase in total LD enzyme and LD-5 isoenzyme activity. It is concluded that no relationship exists between congenital tremor and serum CK activity in piglets. At the same time, there is a positive relationship between congenital tremor and significantly (P < 0.01) elevated LD enzyme and LD-5 isoenzyme activity. The results allow us to suggest that total lactate dehydrogenase and LD-5 isoenzyme activities could be used as biological markers of congenital tremor in piglets.  相似文献

Samples of blood were collected (by jugular venipuncture) from 10 homozygote halothane-reacting pigs and 7 homozygote nonreactors. Antioxidant status was assessed by measurement of blood glutathione peroxidase, glucose-6-phosphate dehydrogenase, and superoxide dismutase activities and plasma concentrations of vitamin E. Significant differences in these values were not apparent between halothane-reacting pigs and nonreactors. Halothane-reacting pigs had tissue damage as indicated by significantly increased plasma activities of the enzymes creatine kinase and pyruvate kinase. The latter showed better discrimination between the 2 populations than did creatine kinase and could prove to be a more effective marker for identification of stress-susceptible pigs.  相似文献

Using simple techniques, the neutrophil function, in its phagocytosis and oxidative metabolism stages, was evaluated in horses. This was done before and after moderate exercise at the aerobic-anaerobic threshold (standardized heart rate 150 beats/min and lactate level of 3.07 +/- 0.21 mmol/L). The objective was to determine whether regular training and moderate exercise improved the neutrophil function. A group of 19 horses was used; 11 of these were untrained and the remainder trained for national jumping events. The exercise test consisted of a 5 min trot followed by a 3 min gallop on a long lunge. Blood samples were taken for analysis before, immediately after and 15 min after exercise. The results showed that (a) there is a difference in the internalization of particles (PI, PP and PE) by neutrophils from trained and untrained horses at a single time point during active recovery, and PP is higher in trained horses immediately after exercise; and (b) oxidative metabolism is significantly lower in untrained animals before and 1 min after exercise. The moderate exercise at the aerobic-anaerobic threshold did not have any influence on the peripheral blood neutrophil function of the phagocytosis and oxidative metabolism of particles.  相似文献

A group of clinically normal horses was subjected to controlled strenuous exercise. Elevated serum concentrations of lactic dehydrogenase, aldolase and creatine kinase were observed after exercise but no significant change in serum glutamic-oxalacetic transaminase was noted. These changes were reduced by repeated exposure to exercise suggesting that measurement of serum enzyme elevations, particularly creatine kinase, might be a useful index of fitness in the horse. Administration of prednisolone prior to exercise also reduced these changes. Since the serum enzyme concentrations had returned to normal within 72 hours of exercise, and since the cytoplasmic enzyme glutamic oxalacetic transaminase was not released from the tissues this supported the hypothesis that efflux of intracellular enzymes into the circulation was due to a temporary selective change in cell membrane permeability rather than to tissue necrosis.  相似文献

The objective of the present study was to compare changes of blood parameters induced by the same work and performed in the same conditions in two racehorse breeds, Thoroughbred and Purebred Arabian. The effect of moderate-intensity exercise was studied in 20 stallions--ten Thoroughbreds, aged 2-3 years and the same number of Purebred Arabians, 3-4 years old. All the horses were administrated the same effort test consisting in 1200 m gallop at a speed typical of the daily training sessions. Three jugular venous blood samples were collected for each horse: at rest, just after the end of the gallop and after 30-minute rest. In the gathered blood, a hemoglobin (Hb) concentration was determined as well as plasma level of glucose (Glc), triacylglycerols (TG), glycerol, free fatty acids (FFA), total plasma proteins (TP) and the activity of creatine kinase (CK), lactate dehydrogenase (LDH) and aspartate aminotransferase (AST). In the Arabian horses, an increase in levels of TP, glycerol, FFA and CK activity measured just after exercise was higher than that in Thoroughbreds. Similarly, after a 30-minute rest, a post-exercise rise of TP, AST, glycerol and FFA proved to be higher in the Arabian horses compared to that in the Thoroughbreds. Only TG plasma concentration measured 30 minutes following the effort was significantly lower in the Arabian horses than in Thoroughbreds. It can be concluded that the Thoroughbred horses adapted better to the effort test applied in this study as compared to the Purebred Arabian horses. The parameters related to lipid metabolism proved to be the most sensitive indicators of breed differences in relation to moderate-intensity exercise.  相似文献

This study investigated the effect of prolonged BCAA supplementation on metabolic response to a 1600m run on treadmill in Standardbred trotters. Four trained Standardbreds were divided into two groups and assigned in a 2×2 Latin square design. Both groups were fed and exercised similarly: one group received an oral amino acids supplement (12 g leucine, 9 g isoleucine and 9 g valine) 30 minutes prior to exercise and immediately after, the other group received a placebo. The horses received the supplement 3 days per week for 5 weeks. In the last week horses performed an anaerobic exercise test on an inclined (3.5%) high-speed treadmill. The exercise consisted of a 15 minute warm-up phase immediately followed by a 1600 m run at maximal speed (heart rate > 200 beats/min). Blood samples were collected pre-exercise, after exercise and during recovery (10 min, 30 min, and 24 h), and analyzed for lactate, ammonia, total protein, urea, uric acid, creatinine, free fatty acids (FFA), creatine kinase (CK), lactate dehydrogenase (LDH), aspartate amino transferase (AST). Heart rate was continuously recorded during exercise and recovery. No statistical differences between the groups were observed for all the considered parameters. Nevertheless, BCAA supplementation resulted in a higher plasma ammonia and urea concentrations as reported in previous studies in humans and rats. These data suggest that a BCAA supplementation are not effective in enhancing performance in healthy and well-fed horses.  相似文献

The normal activities of lactate dehydrogenase and creatine kinase isoenzymes in the tissues and serum of clinically normal five-and-a-half-month-old lambs are presented. The percentage distribution of the lactate dehydrogenase isoenzymes in serum, liver, heart, lung and skeletal muscle were in agreement with previous studies but the distribution in ovine abomasum, small intestine, large intestine and red blood cells has not been previously described. Up to five creatine kinase isoenzymes were detected in ovine tissues and four in serum. One creatine kinase isoenzyme present in serum and tissues was thought to represent a mitochondrial isoenzyme which is absent from the normal serum of other species. Estimations of both lactate dehydrogenase and creatine kinase isoenzymes allowed all the tissues examined to be distinguished and were therefore more likely to allow tissue-specific isoenzyme patterns to be detected in serum than the estimation of the isoenzymes of either enzyme alone.  相似文献

Effects of an 18 min exercise test, on three separate occasions during a one year jump-training programme, was studied in seven horses. Determinations were carried out on venous blood for packed cell volume, haemoglobin, total protein, lactate and pyruvate, glucose, free fatty acids, insulin, glucagon, blood gases, bicarbonate, pH, aldolase, aspartate aminotransferase and alanine amino-transferase. Exercise caused a slight increase in lactate and pyruvate, total protein, aldolase, alanine aminotransferase, pO2, bicarbonate and pH. Glucose, free fatty acids and pCO2 levels decreased. Training caused no significant difference in these changes. However, during the year, increases in lactate and decreases in pH (resting levels) were observed.  相似文献

The aim of this study was to determine the cardiotoxic potency of tulathromycin. Tulathromycin (10 mg/kg, SC) was administered to ten adult male rabbits, and blood samples were obtained before and after drug administration (0 and 6 hours). Serum cardiac damage markers (troponin I, creatine kinase-MB, myoglobin, lactate dehydrogenase, aspartate aminotransferase), routine serum biochemical values (alkaline phosphatase, alanine aminotransferase, gamma-glutamyltransferase, creatinine, blood urea nitrogen, cholesterol, triglyceride, high-density lipoprotein, amylase, total protein, albumin, glucose, calcium, ionised calcium, sodium, potassium), white blood cell (WBC) and red blood cell (RBC) counts, arterial blood gas parameters (pH, partial carbon dioxide pressure, partial oxygen pressure, actual bicarbonate, standard bicarbonate, total carbon dioxide, base excess in vivo, base excess in vitro, oxygen saturation, packed cell volume, haemoglobin) and serum oxidative status (malondialdehyde, nitric oxide, superoxide dismutase, retinol, β-carotene) were measured. Increased levels of troponin I, creatine kinase-MB and creatinine, and decreased WBC counts, ionised calcium and potassium levels were observed after drug administration. Tulathromycin treatment may cause cardiotoxicity, but its effects may be less dramatic than those of other macrolide antibiotics frequently used in veterinary medicine.  相似文献

The degree of development of rigor mortis in the carcases of slaughter pigs was assessed subjectively on a three-point scale 35 minutes after they were exsanguinated, and related to the levels of cortisol, lactate and creatine kinase in blood collected at exsanguination. Earlier rigor development was associated with higher concentrations of these stress indicators in the blood. This relationship suggests that the mean rigor score, and the frequency distribution of carcases that had or had not entered rigor, could be used as an index of the degree of stress to which the pigs had been subjected.  相似文献
