试验旨在观察鸡传染性支气管炎病毒(infectious bronchitis virus,IBV)和鸡新城疫病毒(newcastle disease virus,NDV)之间的增殖干扰现象,分析在免疫过程中两种疫苗存在的免疫干扰,为确定疫苗的免疫程序提供依据。采用完全随机试验设计,选取10日龄鸡胚尿囊腔接种不同浓度的IBV、NDV以及不同浓度混合的IBV和NDV,收集尿囊液测定其血凝效价(HA);用不同浓度的IBV、NDV以及不同浓度混合的IBV和NDV分别免疫BALB/c小鼠及SPF雏鸡,收集血清,间接酶联免疫吸附法(ELISA)和血凝抑制(HI)试验测定IBV和NDV的抗体效价及抑制效价。结果表明,同胚培养时,无论先接种IBV后接种NDV还是先接种NDV后接种IBV或IBV和NDV同时接种,IBV对NDV均有干扰作用,而NDV对IBV没有干扰作用;接种IBV和NDV的小鼠,其产生IBV和NDV的抗体效价均低于单独免疫组,免疫的次序及免疫间隔时间对IBV和NDV的抗体效价均有影响,IBV对NDV的免疫效果具有较大的干扰作用。不同针混合免疫方式能提高IBV和NDV的抗体效价;接种IBV和NDV的雏鸡,免疫次序及免疫相隔时间对IBV和NDV的抗体效价均有影响,IBV对NDV的免疫效果具有较大的干扰作用;雏鸡和小鼠免疫血清HI试验数据表明,免疫次序及免疫间隔时间对IBV和NDV抗体的产生均有影响,但对NDV抗体的产生影响更明显,随着免疫间隔时间的增加,IBV和NDV的抗体水平呈增加趋势,雏鸡比小鼠更能敏感地反映出IBV对NDV的免疫干扰作用。混合注射时IBV和NDV的抗体水平均降低,IBV对NDV的免疫效果有干扰作用。因此,在同胚培养、小鼠及雏鸡的免疫中,IBV对NDV有干扰作用,而NDV对IBV无干扰作用。混合接种时IBV和NDV抗体效价均下降,IBV对NDV的免疫效果有干扰作用。免疫次序及免疫间隔时间对IBV和NDV抗体的产生均有影响,但随着免疫间隔的增加,IBV和NDV的抗体水平呈增加趋势。  相似文献

This experiment aimed to investigate the proliferation interference effects of the infectious bronchitis virus (IBV) and Newcastle disease virus (NDV),analyze the immune interference of two vaccines in the immune process,and provide a basis for determining vaccine immune program.This experiment was in a complete randomized design,10-day-old allantoic cavity of the chick embryo was inoculated with different concentrations of IBV and NDV and with a mixture of different concentrations of IBV and NDV,and then the allantoic fluid was collected for determination of the titer of hemagglutination (HA).Furthermore,BALB/c mice and SPF chicken were immunized with different concentrations of IBV and NDV and also with the mixed IBV and NDV with different concentrations,the blood of mice and chicken were collected,and antibody titer and inhibitory titer of IBV and NDV were determined by indirect enzyme linked immunosorbent assay (ELISA) and hemagglutination inhibition (HI) test.Experimental results showed that in the homeomorphic cultivation research,whether IBV and NDV were inoculated in a different order or inoculated at the same time,the inoculation of IBV behaved interference to the NDV,while the inoculation of NDV had no interference effects to IBV;Mice were inoculated with IBV and NDV,IBV and NDV antibody titers were lower than single immunized group,immune procedures and immune intervals often had effect on the titer of IBV and NDV antibody,IBV had interference on the immune effect of NDV,different needle mixed immunization can improve the antibody titer of IBV and NDV.In the group of SPF chicken inoculated with IBV and NDV,the order of the inoculation and the immune interval had effect on the titer of IBV and NDV antibody,IBV had a interference on the immune effect of NDV;In the immune serum inhibitory test,the data showed that the IBV and NDV antibody production were affected by the immune order of the inoculation and the interval,the effect on the NDV antibody was more obvious,but with the increase of the immune interval,the antibody level of IBV and NDV showed an increasing trend,the chicken was more sensitive than the mice to reflect the immune interference of IBV on NDV.IBVand NDV antibodies titer was also reduced in the groups that was inoculated with the mixed IBV and NDV,IBV had a interference on the immune effect of NDV.In the homeomorphic cultivation research and animal research,IBV behaved interference to the NDV,while NDV had no interference effects to IBV.IBV and NDV antibody titers decreased when mixed immunization,IBV had a interference on the immune effect of NDV.The order of the inoculation and the immune interval had effect on the antibody titer of IBV and NDV,but with the increase of the immune interval,the antibody level of IBV and NDV showed an increasing trend.  相似文献

Serosurvey of five viruses in chickens on smallholdings in Bangladesh   总被引:1,自引:0,他引:1  
A serologic survey was undertaken in chickens in smallholdings in Bangladesh for avian influenza A virus (AIV), egg drop syndrome '76 virus (EDS'76V), infectious bronchitis virus (IBV), Newcastle disease virus (NDV) and reovirus (RV) in three phases: January 2002-May 2003, September 2003-August 2004, and August 2005-March 2006. Four hundred thirty-six sera collected in the 2nd phase, 295 in the first phase, 755 in the 1st plus 2nd phases and 295 in the 1st phase were investigated for AIV, EDS'76V, IBV and RV, respectively, using enzyme linked immunosorbent assays. All 854 sera collected in the three phases were screened for NDV using hemagglutination inhibition test. In chickens 20% were seropositive to AIV, 3% to EDS'76V, 74% to IBV, 88% to NDV, and 47% to RV. The seroprevalence in flocks was 23% to AIV, 6% to EDS'76V, 79% to IBV, 89% to NDV and 56% to RV. Twenty-five percent chickens had > or = 10log(2)HI titers to NDV.  相似文献

采用以野菊花、穿心莲、柴胡等10味中药提取的中药制剂,连续给22日龄SPF鸡口服5 d后,以禽流感病毒H9N2、新城疫病毒F4、传染性支气管炎病毒强毒株M41和支原体S6强毒分别攻击,观察SPF鸡临床症状、发病率、病理学变化和分离病毒。结果表明,此中药制剂75 mg/只口服组,新城疫病毒、禽流感病毒和传染性支气管炎病毒攻击后发病率分别为12.5%,0%,10%;37.5 mg/只剂量口服组发病率分别为50%,10%,10%,且低剂量组病毒分离阳性。攻击支原体后,以上两个剂量组SPF鸡病变指数分别3.5和5.5,研究证实本中药制剂可以控制以上4种病原所致的家禽呼吸道病的发生。  相似文献

A multivalent in ovo vaccine (MIV) was tested for safety and efficacy in a commercial broiler complex. The MIV comprised five replicating live viruses including serotypes 1, 2, and 3 of Marek's disease virus (MDV), an intermediate infectious bursal disease virus (IBDV) and a recombinant fowl poxvirus (FPV) vector vaccine containing HN and F genes of Newcastle disease virus (NDV). The performance of MIV-vaccinated broilers was compared with that of hatchmates that received turkey herpesvirus (HVT) alone (routinely used in ovo vaccine in the broiler complex). The chickens that hatched from the MIV-injected and HVT-injected eggs were raised under commercial conditions in six barns. Barn 1 housed 17,853 MIV-vaccinated chickens and each of the barns 2-6 housed 18,472-22,798 HVT-vaccinated chickens. The HVT-vaccinated chickens were given infectious bronchitis virus (IBV) and NDV vaccines at hatch and at 2 wk of age. The MIV-vaccinated chickens received IBV vaccine at hatch and IBV + NDV at 2 wk of age. The relative values of hatchability of eggs, livability and weight gain of chickens, and condemnation rates at processing were comparable between the MIV and the HVT groups (P > 0.05). Chickens from the MIV- and the HVT-vaccinated groups were challenged with virulent viruses under laboratory conditions. The resistance of vaccinated chickens against Marek's disease could not be assessed because of high natural resistance of unvaccinated commercial broilers to virulent MDV. The relative resistances of the MIV- and the HVT-vaccinated groups, respectively, against other virulent viruses were as follows: IBDV, 100% for both groups; NDV, 81% vs. 19%; FPV, 86% vs. 0%. The successful use of MIV under field conditions expands the usefulness of the in ovo technology for poultry.  相似文献

In 2010, a West Nile virus (WNV) epidemic was reported in Central Macedonia, Northern Greece, with 197 neuroinvasive disease (WNND) cases in humans. The following 3 years, WNV spreads to new areas of Greece and human cases reoccurred during the transmission periods. After the initial outbreak, a WNV surveillance system using juvenile backyard chickens was established in Central Macedonia (after the 2011 outbreak) and Eastern Macedonia‐Thrace (after the 2012 outbreak). Sera were screened for the presence of antibodies against WNV using cELISA and serum neutralization test, to monitor the spread of WNV and to assess the correlation between the WNV point seroprevalence in chickens and the incidence rates of human WNND cases in the aforementioned areas. WNV seroprevalence in chickens was 10.4% (95% CI: 7–15) in Central Macedonia (2011) and 18.1% (95% CI: 14–23) in Eastern Macedonia‐Thrace (2012). Seroprevalence in chickens and incidence rates of human WNND cases in Eastern Macedonia‐Thrace were strongly positively correlated (ρ = 0.98, P = 0.005) at the regional unit level, with the incidence of WNND in humans increasing with increasing WNV point seroprevalence in chickens. In Central Macedonia, the correlation was weaker (ρ = 0.68, P = 0.20), apparently due to small number of reported human WNND cases. Another study was also conducted using juvenile backyard chickens in Central Macedonia, aiming to detect early WNV enzootic circulation, before the onset of human cases during 2011 and 2013. The first seroconverted chickens were detected about 1.5 months before the laboratory diagnosis of any human WNND cases in Central Macedonia, for both years. WNV surveillance, using juvenile backyard chickens, was reliable for the identification of areas with WNV enzootic and silent transmission, and for early warning. Timely diffusion of information to public health authorities facilitated the successful implementation of preparedness plans to protect public health.  相似文献

以2种剂量的低致病性禽流感病毒(lowly pathogenic avian influenza virus,LPAIV),传染性支气管炎病毒(infectious bronchitis virus,IBV)疫苗株H120和H52,新城疫病毒(Newcastle disese viurs,NDV)Lasota株分别于气管内注射10日龄易感鸡,2d后,气管注射禽病原性大肠杆菌O37株(O78),连续观察5d,结果,除LPAIV单独感染组有6.25%的死亡率外,其余各病毒单独接种组均健活;大肠杆菌O37株单独接种组的死亡率为62.50%,较高剂量的LPAIV,IBV H120和H52,NDV Lasota株与大肠杆菌O37株有效强的协同致病作用,死亡率分别达到81.25%,100.00%,93.75%和87.50%,而较低剂量的上述病毒则无明显的协同作用,IBV,NDV疫苗株与大肠杆菌联合接种组的多数死亡鸡病程推迟。  相似文献

将鸡新城疫病毒(NDV)和鸡传染性支气管炎病毒(IBV)按一定的稀释比例等量混合接种9~10日龄SPF鸡胚,让其在同一鸡胚中增殖(简称同胚培养)。试验结果表明,用5倍稀释的ND-Lasota病毒液与 5000倍稀释的IBH120病毒液等体积混合后尿囊腔接种鸡胚,能在同一鸡胚中正常增殖,接种后96 h收毒,用红细胞凝集试验(HA)测得NDV、IBV两种病毒的血凝价分别为11 log2和12 log2,不低于各自病毒单独增殖的HA滴度,病毒的增殖基本趋于平衡。同时也证实了在合适的培养条件下,IBV对NDV的复制不会产生干扰。  相似文献

Newcastle disease virus isolated from an outbreak in racing pigeons in India was found to be velogenic, based on the mean time to death in 10-day-old embryonated hen's eggs, the intravenous pathogenicity index in 6-week-old chickens and the pathogenesis in chickens and pigeons. The virus induced disease in chickens without prior adaptation in chickens. The virus was antigenically unusual since it could not be grouped with the available panel of monoclonal antibodies at the World Reference Laboratory for Newcastle disease, UK. However, commercially available lentogenic and mesogenic vaccines provided 100% protection to chickens against this antigenically unusual NDV.  相似文献

传染性喉气管炎新城疫鸡痘重组病毒免疫效力的研究   总被引:1,自引:2,他引:1  
在表达鸡传染性喉气管炎病毒(ILTV)糖蛋白gB基因和新城疫病毒(NDV)F基因的重组鸡痘病毒(rF-PV-gB-F)安全性检验合格后,以5.0×101~5.0×104PFU不同含量按0.1mL/鸡的剂量免疫100只30日龄SPF鸡,30d后分组分别用ILTVWG株和NDVF48E9株强毒进行攻击。免疫鸡抗鸡痘病毒抗体都转为阳性,痘反应和接种剂量有关,重组疫苗的最小反应剂量为50PFU。重组疫苗可以诱发对新城疫和传染性喉气管炎的保护,0.1mL/鸡的接种量在500~5000PFU浓度范围内的免疫效果最好,对于ILTV攻击的发病保护率在70%以上,对NDV强毒攻击的抗死亡保护率可以达到80%,这为进一步考察疫苗的免疫效力试验以及进行田间试验奠定了基础。  相似文献

Avian metapneumovirus (aMPV), Newcastle disease virus (NDV), and infectious bronchitis virus (IBV) are important respiratory pathogens of chickens. To achieve early posthatch protection against all three diseases it would be helpful to deliver live aMPV, IBV, and NDV vaccines simultaneously at 1 day of age. However, previous work has indicated that the efficacy of aMPV vaccines may be affected when codelivered with IBV or NDV vaccines. The efficacy of an aMPV vaccine when codelivered to chickens in a trivalent combination with an NDV and an IBV vaccine was examined. The serological antibody response to the aMPV vaccine given with the IBV and NDV vaccine was significantly lower than when the aMPV vaccine was given alone. However, the aMPV vaccine did not affect the serological response to the IBV and NDV vaccines. Irrespective, the efficacy of the aMPV vaccine was not affected based on clinical signs postchallenge. This is the first report showing aMPV, IBV, and NDV vaccines can be codelivered without affecting the efficacy of the aMPV vaccine.  相似文献

应用多重RT-PCR反应(mRT-PCR)结合变性高效液相色谱(DHPLC)技术建立禽产品中禽流感病毒和新城疫病毒的快速检测方法。以禽流感病毒NP基因和新城疫病毒F基因各设计1对引物,经优化单相RT-PCR(sRT-PCR)反应体系,建立多重RT-PCR(mRT-PCR)同时检测禽流感和新城疫,其PCR扩增产物经DHPLC技术进行快速检测分析后,检测极限分别达10-0.5ELD50/0.1mL和10-1.5ELD50/0.1mL。与普通凝胶电泳比较,敏感性高2个数量级。特异性试验表明,mRT-PCR-DHPLC仅检测到禽流感病毒与新城疫病毒,而对其他5种禽类病原体和SPF鸡胚尿囊液均未检测到阳性吸收峰。所建立的方法检测不同亚型禽流感病毒29株、不同来源及不同毒力的新城疫病毒16株,结果与实时荧光定量PCR检测结果完全一致;与病原分离法同时检测人工感染鸡的组织脏器,两种方法检测禽流感的符合率为94.4%(34/36),检测新城疫的符合率为95.4%(21/22)。病原分离法的检出率虽高于DHPLC方法,但经统计学分析两者差异不显著,两种方法同时对27份进口鸡胗样品与90份棉拭子样品进行检测,阴性符合率为100%,可适用于进出口禽肉产品的检测。  相似文献

为观察鸡传染性支气管炎病毒(IBV)HN99株对新城疫病毒(NDV)增殖的干扰作用,该试验采用不同浓度的IBV标准株M41和地方株HN99与鸡新城疫病毒(NDV)分别按不同接种顺序同胚增殖,利用病毒血凝试验(HA)测定NDV的效价,观察IBV对NDV的干扰作用,从而为检测IBV地方株HN99提供方法,也为同胚增殖两种病毒提供一系列的数据参考。试验结果表明,鸡传染性支气管炎病毒地方株HN99对NDV的干扰作用与其浓度和接种顺序有关。  相似文献

Two types of live attenuated vaccines have been used worldwide for the control of infectious laryngotracheitis virus (ILTV): 1) chicken embryo origin (CEO) vaccines; and 2) tissue culture origin vaccines (TCO). However, the disease persists in spite of extensive use of vaccination, particularly in areas of intense broiler production. Among the factors that may influence the efficiency of ILTV live attenuated vaccines is a possible interference of Newcastle Disease virus (NDV) and infectious bronchitis virus (IBV) vaccines with the protection induced by ILTV vaccines. The protection induced by CEO and TCO vaccines was evaluated when administered at 14 days of age alone or in combination with the B1 type strain of NDV (B1) and/or the Arkansas (ARK) and Massachusetts (MASS) serotypes of IBV vaccines. Two weeks after vaccination (28 days of age), the chickens were challenged with a virulent ILTV field strain (63140 isolate, group V genotype). Protection was evaluated at 5 and 7 days postchallenge by scoring clinical signs and quantifying the challenge virus load in the trachea using real-time PCR (qPCR). In addition, the viral load of the vaccine viruses (ILTV, NDV, and IBV) was quantified 3 and 5 days postvaccination also using qPCR. The results of this study indicate that the NDV (B1) and IBV (ARK) vaccines and a multivalent vaccine constituted by NDV (B1) and IBV (ARK and MASS) did not interfere with the protection induced by the CEO ILTV vaccine. However, the NDV (BI) and the multivalent (B1/MASS/ARK) vaccines interfered with the protection induced by the TCO vaccine (P < 0.05). Either in combination or by themselves, the NDV and IBV vaccines decreased the tracheal replication of the TCO vaccine and the protection induced by this vaccine, since the ILTV-vaccinated and -challenged chickens displayed significantly more severe clinical signs and ILTV load (P < 0.05) than chickens vaccinated with the TCO vaccine alone. Although NDV and IBV challenges were not performed, the antibody responses elicited by NDV and/or the IBV vaccinations were significantly reduced (P < 0.05) when applied in combination with the CEO vaccine.  相似文献

本研究对分离自上海某养鸡场的疑似鸡传染性支气管炎病毒(Infectious bronchitis virus,IBV)进行了分离和鉴定.所采用的方法有:鸡胚致病性试验、对鸡新城疫病毒(Newcastle disease virus,NDV)的干扰试验、动物回归试验以及RT-PCR鉴定等.试验结果表明,该病原接种鸡胚后,48 h可引起鸡胚死亡,胚体呈侏儒样变化,对NDV感染有明显干扰作用;用分离的病毒接种SPF雏鸡可产生明显呼吸道症状,肾脏表现肿大和大量尿酸盐沉积等病理变化;用IBV特异性引物可以从分离物中扩增出IBV的N基因序列,测序结果表明,该毒株属于肾型IBV.综上所述,本研究在上海流行区域成功分离到一株IBV,将其命名为SH11株.  相似文献

利用PALL滤器将灭活后的新城疫、禽流感、传染性支气管炎、产蛋下降综合征等病毒分别进行10倍浓缩,经检验合格后,分别按照一定比例将抗原浓缩液与灭菌的医用白油配比,经胶体磨制备成三联或四联油乳剂灭活疫苗,均通过了实验室的各项指标检测。经过在隔离器进行实验室安全和效力检验,以及田间试验证明,该疫苗安全有效,抗体水平高,维持时间长,能够保护鸡群抵抗新城疫、禽流感、传染性支气管炎等强毒的攻击。  相似文献

The interference phenomenon of infectious bronchitis virus (IBV) with growth of Newcastle disease virus (NDV) in embryonating chicken eggs (ECE) was used as a diagnostic method. Fifteen field isolates obtained from presumptively infectious-bronchitis-affected chickens were analyzed by the IBV-NDV interference test. Eight isolates were capable of interfering with the growth of the La Sota strain of NDV, as measured by hemagglutination (HA) activity when IBV was inoculated 10 hr before NDV into ECE. The interference was considered specific for IBV, because it could be eliminated by adding homologous anti-IBV serum. The sensibility of this method could be demonstrated, because in some cases low-passage levels of IBV isolates showing HA interference ability were not capable of producing lesions in ECE. Furthermore, serologically negative IBV samples did not interfere with NDV growth. From these results, the IBV-NDV interference test appears to be a potential diagnostic alternative for identifying IBV field isolates.  相似文献

Inactivated Newcastle disease virus (NDV), avian influenza virus (AIV), and infectious bronchitis virus (IBV) antigens were evaluated for immunological efficacy in monovalent and polyvalent vaccines. Vaccinated broilers were bled for hemagglutination-inhibition (HI) tests at 1- or 2-week intervals. Half of the chickens were challenged with the Largo isolate of velogenic viscerotropic (VV) NDV at 8 weeks post-vaccination, and the remainder were challenged with the Massachusetts 41 strain IBV at 9 weeks post-vaccination. Newcastle disease HI titers were reduced significantly (P less than 0.05) from those of monovalent control vaccine groups when IBV antigen was emulsified in mixtures with low (1-3x) concentrated NDV or NDV and AIV antigens. Avian influenza HI titers were significantly (P less than 0.05) lower than those of the control monovalent groups when highly concentrated NDV was part of the polyvalent vaccine. Infectious bronchitis HI titers were higher than those of control monovalent groups in 13 of 15 vaccine groups when IBV antigen was in polyvalent formulations. VV NDV challenge killed all non-NDV vaccinates and induced increased HI titers in NDV vaccinates but no morbidity or mortality. Sixty of 80 IBV vaccinates experienced a fourfold or greater HI titer increase following challenge. All non-IBV vaccinates seroconverted at 1 week post-challenge.  相似文献

Infectious bronchitis virus (IBV) causes respiratory disease in chickens all over the world. IBV has many serotypes that do not confer cross protection against each other. Hemagglutination inhibition (HI) test has been used to determine the serotypes of IBV as a substitute to the more laborious virus neutralization test and the more sophisticated restriction endonuclease digestion or sequencing of the S1 gene. In Jordan, no previous studies have been carried out to determine the involvement of IBV in respiratory disease in chickens, or the serotypes of IBV that possibly exist. In this study, serum from different chicken flocks (n = 20) that suffered from respiratory disease were tested for IBV antibodies using commercial IBV antibody ELISA at time of the initial signs of the respiratory disease and repeated on serum samples from the same flocks 10–14 days later. ELISA titer for IBV increased in 14 out of 20 flocks (70%) after 10–14 days of the initial signs of the respiratory disease and this indicates a recent exposure to IBV. The second serum samples from these 14 flocks were further examined against a panel of five IBV antigens (Ark, Conn, DE-072, JMK, and Mass) by HI test to determine the serotype(s) of IBV they have been exposed to. The HI test results indicated that the exposure of some of these flocks were to Ark, DE-072, and Mass like serotypes. However, the HI titers against the antigens used in this study were relatively similar in 10 out of the 14 flocks (71%) and the serotype of IBV that these flocks were exposed to could not be determined and the possible causes of this are discussed.  相似文献

An investigation of the disease status of village poultry in Mauritania   总被引:2,自引:0,他引:2  
Three contagious poultry diseases in Mauritania were studied. Serum samples and tracheal swabs were taken from 80 chickens in village poultry flocks in each of three different regions. Antibodies against Newcastle disease virus (NDV) were detected in 4.8% of chickens, antibodies against Gumboro disease virus in 15.8% of chickens and antibodies against Salmonella pullorum in 6.2% of chickens. Six isolates of NDV were made, of which four formed plaques on chicken embryo fibroblast monolayers. Seropositivity was highest in the Trarza region, bordering the River Senegal, and four out of six of the NDV isolates were made from chickens in this region.  相似文献
