Vasoactive intestinal peptide (VIP) is a neuropeptide with broad tissue distribution. Although its precise function is unknown, it is thought to exert its effect, at least in part, by interacting with cell surface receptors. Nuclear receptors for VIP have now been identified by specific binding of 125I-labeled VIP to nuclei of a human colonic adenocarcinoma cell line (HT29) and by cross-linking of 125I-labeled VIP to its receptor on intact nuclei. In contrast, 125I-labeled transferrin shows only background binding to nuclei but significant binding to intact cells. Purity of the isolated nuclei was further substantiated by electron microscopy. The apparent molecular sizes of the VIP--cross-linked nuclear and cell surface receptors are similar but not identical.  相似文献

There is growing evidence that the neuropeptides oxytocin and vasopressin modulate complex social behavior and social cognition. These ancient neuropeptides display a marked conservation in gene structure and expression, yet diversity in the genetic regulation of their receptors seems to underlie natural variation in social behavior, both between and within species. Human studies are beginning to explore the roles of these neuropeptides in social cognition and behavior and suggest that variation in the genes encoding their receptors may contribute to variation in human social behavior by altering brain function. Understanding the neurobiology and neurogenetics of social cognition and behavior has important implications, both clinically and for society.  相似文献

Remarkable similarities in the intracellular and genetic events occur when lymphoid and hematopoietic cells are exposed to their specific growth factors. The interleukin-2 (IL-2) receptor, whose cell-surface expression is an absolute requirement for the growth and differentiation of lymphoid cells, was detected on various nonlymphoid hematopoietic cell types in this study. Cell lines consisting either of granulocyte-macrophage precursors or mast cells, which are dependent on interleukin-3 (IL-3) for their growth, expressed high levels of the IL-2 receptor on their surface. Analysis of the binding characteristics of these receptors with 125I-labeled recombinant IL-2 revealed that only receptors with low affinity for IL-2 were present on these cells. Addition of purified recombinant IL-3 to these cell lines led to an increase in IL-2 receptor gene expression within 1 hour in isolated nuclei. This IL-3--induced increase in the number of IL-2 receptors on the cell surface is maximal within 24 hours. Addition of 10,000 units of IL-2 to these cells had no apparent effect on their growth or differentiation. The presence of the receptor with only low affinity for IL-2 on hematopoietic cells and the regulation by IL-3 suggest that this receptor is involved in some important metabolic event in hematopoiesis.  相似文献

Vasopressin and oxytocin strongly modulate autonomic fear responses, through mechanisms that are still unclear. We describe how these neuropeptides excite distinct neuronal populations in the central amygdala, which provides the major output of the amygdaloid complex to the autonomic nervous system. We identified these two neuronal populations as part of an inhibitory network, through which vasopressin and oxytocin modulate the integration of excitatory information from the basolateral amygdala and cerebral cortex in opposite manners. Through this network, the expression and endogenous activation of vasopressin and oxytocin receptors may regulate the autonomic expression of fear.  相似文献

Dynorphin and vasopressin: common localization in magnocellular neurons   总被引:15,自引:0,他引:15  
The opioid peptide dynorphin is widely distributed in neuronal tissue of rats. By immunocytochemical methods, it was shown previously that dynorphin-like immunoreactivity is present in the posterior pituitary and the cells of the hypothalamic neurosecretory magnocellular nuclei which also are responsible for the synthesis of oxytocin, vasopressin, and their neurophysins. By using an affinity-purified antiserum to the non-enkephalin part of the dynorphin molecule it has now been demonstrated that dynorphin and vasopressin occur in the same hypothalamic cells of rats, whereas dynorphin and oxytocin occur in separate cells. Homozygous Brattleboro rats (deficient in vasopressin) have magnocellular neurons that contain dynorphin separate from oxytocin. Thus dynorphin and vasopressin, although they occur in the same cells, appear to be under separate genetic control and presumably arise from different precursors.  相似文献

Mating behavior in ovariectomized female rats treated with estrogen can be greatly enhanced by subcutaneous injections of progesterone. Application of potassium chloride to the cortex of females previously treated with estrogen can also induce greatly increased sexual receptivity as indicated by the lordosis response. This facilitation of mating behavior by a treatment known to cause functional decortication suggests that mechanisms mediating female mating behavior are under tonic inhibition by an inhibitory system which involves the cerebral cortex.  相似文献

AMPA glutamate receptors (AMPARs) mediate fast excitatory synaptic transmission. Upon fast consecutive synaptic stimulation, transmission can be depressed. Recuperation from fast synaptic depression has been attributed solely to recovery of transmitter release and/or AMPAR desensitization. We show that AMPAR lateral diffusion, observed in both intact hippocampi and cultured neurons, allows fast exchange of desensitized receptors with na?ve functional ones within or near the postsynaptic density. Recovery from depression in the tens of millisecond time range can be explained in part by this fast receptor exchange. Preventing AMPAR surface movements through cross-linking, endogenous clustering, or calcium rise all slow recovery from depression. Physiological regulation of postsynaptic receptor mobility affects the fidelity of synaptic transmission by shaping the frequency dependence of synaptic responses.  相似文献

Nuclear estrogen receptor from MCF-7 cells undergoes a time-dependent, hormone-inducible transformation to a form that is less extractable from nuclei and less exchangeable with ligand. This receptor-modifying, intranuclear event is independent of receptor loss (processing) and appears associated with hormone responsiveness (progesterone-receptor induction) in these cells. The magnitude of receptor loss, however, is variable and apparently not a prerequisite for hormone action to induce progesterone receptor.  相似文献

在室内养虫笼观察并分析松墨天牛Monochamus alternatus在以24h为周期内的交配行为及其规律。松墨天牛交配行为包括2个步骤:一是雌虫接近雄虫,在其周围活动;二是雄虫抱握雌虫,发生交配行为。交配日节律观察结果显示:松墨天牛在24h内有2次交配高峰期,10:00—14:00和18:00—24:00,它在这2个时间段以外很少或几乎不交配。松墨天牛交配行为具有以下特点:普遍存在与同一个体或不同个体的多次交配现象,交配次数为2次f含)以上的松墨天牛个体约占全部个体的77.5%,每次交配时间平均为50s;除正常雄雌交配外,还会出现雄雄和雌雌“交配”以及2只雄虫争夺雌虫交配权的现象。图1表1参15  相似文献

Coordinate hormonal and synaptic regulation of vasopressin messenger RNA   总被引:3,自引:0,他引:3  
Previous studies have shown that adrenalectomy augments arginine vasopressin (AVP) messenger RNA levels in the adult paraventricular nucleus. It is now demonstrated that unilateral lesions in the lateral septal nucleus enhance the adrenalectomy-induced expression of AVP mRNA. This effect was entirely ipsilateral to the lesion and most prominent in the rostral paraventricular nucleus and related nuclei. Moreover, AVP and AVP mRNA were found to be colocalized with oxytocin in a few neurons. These results indicate that mRNA expression is modulated by synaptic influences and raise the possibility that synaptically mediated selection of neuronal phenotypes is a dynamic feature of the mature central nervous system.  相似文献

Central amygdala (CeA) projections to hypothalamic and brain stem nuclei regulate the behavioral and physiological expression of fear, but it is unknown whether these different aspects of the fear response can be separately regulated by the CeA. We combined fluorescent retrograde tracing of CeA projections to nuclei that modulate fear-related freezing or cardiovascular responses with in vitro electrophysiological recordings and with in vivo monitoring of related behavioral and physiological parameters. CeA projections emerged from separate neuronal populations with different electrophysiological characteristics and different response properties to oxytocin. In vivo, oxytocin decreased freezing responses in fear-conditioned rats without affecting the cardiovascular response. Thus, neuropeptidergic signaling can modulate the CeA outputs through separate neuronal circuits and thereby individually steer the various aspects of the fear response.  相似文献

Polypeptide receptors on mammalian plasma membranes can be categorized on the basis of function. The binding of ligand by class I receptors results in changes in cell metabolism or behavior. Hormone-receptor interactions typify this group. The binding of ligand by class II receptors in ligand internalization. Although changes in cellular activity may result from metabolism of the internalized ligand, the interaction between ligand and class II receptor does not itself lead to alterations in cell behavior. Class II receptors include those for low-density lipoproteins and for alpha-macroglobulin-protease complexes. Although receptors within each category are chemically disparate, they show striking similarities in behavior. Analysis of the behavioral patterns of receptors in each category reveals insights into receptor physiology and allows for a prospective analysis of receptor characteristics.  相似文献

Molecular cloning of the chicken progesterone receptor   总被引:16,自引:0,他引:16  
To define the functional domains of the progesterone receptor required for gene regulation, complementary DNA (cDNA) clones encoding the chicken progesterone receptor have been isolated from a chicken oviduct lambda gt11 cDNA expression library. Positive clones expressed antigenic determinants that cross-reacted with six monospecific antibodies derived from two independent sources. A 36-amino acid peptide sequence obtained by microsequencing of purified progesterone receptor was encoded by nucleotide sequences in the longest cDNA clone. Analysis of the amino acid sequence of the progesterone receptor deduced from the cDNA clones revealed a cysteine-rich region that was homologous to a region found in the estrogen and glucocorticoid receptors and to the avian erythroblastosis virus gag-erb-A fusion protein. Northern blot analysis with chicken progesterone receptor cDNA's indicated the existence of at least three messenger RNA species. These messages were found only in oviduct and could be induced by estrogens.  相似文献

以四类异质(粘果、易变、偏凸和二角山羊草)异核(224,90-110)小麦雄性不育系为母本,与生产上一系列小麦品种(系)进行杂交,系统调查了杂种F1代减数分裂中期Ⅰ单价体频率及后期出现落后染色体及染色体桥频率,并对中、后期染色体变异率与杂种F1自交结实率进行了相关分析。结果表明:①其杂种F1自交结实率与其减数分裂中期的单价体频率及后期染色体变异率有一定的负相关,但未达到显著水平。而中期单价体频率及后期染色体变异率存在着正相关,且达显著水平;②异源细胞质对染色体配对的影响因父本遗传背景不同而不同;③粘类小麦雄性不育系杂种F1育性不仅与染色体配对有关,亦受环境及基因型的影响。  相似文献

研究了复合蛋氨酸螯合铜、蛋氨酸螫合铁、蛋氨酸螫合锰、蛋氨酸螫合锌和蛋氨酸硒对母猪血清微量元素、相关酶、激素的含量以及繁殖性能的影响,结果表明:使用复合微量元素母猪血清铜、铁、锌的水平有提高的趋势,血清锰、硒的水平则降低;配后期(配种后30d)对比于配前期(断奶后3d),血清超氧化物歧化酶(SOD)活性有显著提高,亚铁氧化酶和谷胱甘肽过氧化物酶(GSH-PX)活性则显著降低;复合微量元素显著提高了配后期血清中亚铁氧化酶和配前期血清GSH-PX活性,降低了配后期GSH-PX的活性;有机微量元素对血清碱性磷酸酶(AKP)活性没有影响,有机微量元素对配前母猪血清中促黄体素(LH)水平没有影响,显著提高了母猪配后30d时血清中孕酮(P)水平;使用复合有机微量元素母猪的窝产活仔数提高了0.6头,平均窝质量提高了0.78k。  相似文献

Metkephamid is an analog of methionine enkephalin that retains high affinity for the delta receptor and is a systemically active analgesic. Since it is at least 100 times more potent than morphine as an analgesic when placed directly into the lateral ventricles, and is 30 to 100 times more potent on the delta receptor and yet is roughly equipotent on the mu receptor in vitro, it is concluded that it probably produces analgesia by action on delta receptors as well as, or rather than, on mu receptors. It has less tendency to produce respiratory depression, tolerance, and physical dependence than standard analgesics, and it is presently undergoing clinical trial.  相似文献

Partially purified lipid extracts of Saccharomyces cerevisiae contain a substance that displaces tritiated estradiol from rat uterine cytosol estrogen receptors. The yeast product induces estrogenic bioresponses in mammalian systems as measured by induction of progesterone receptors in cultured MCF-7 human breast cancer cells and by a uterotrophic response and progesterone receptor induction after administration to ovariectomized mice. The findings raise the possibility that bakers' yeast may be a source of environmental estrogens.  相似文献

Odorant receptor (OR) is crucial for insects to detect and recognize external chemical cues closely related to their survival. The insect OR forms a heteromeric complex composed of a ligand-specific receptor and a ubiquitously odorant receptor coreceptor (Orco). This study used the CRISPR/Cas9 technique to knock out (KO) Orco and reveal its essential role in acting on OR-meditated olfactory behavior in a critical invasive agricultural pest, the fall armyworm (FAW), Spodoptera frugiperda. Electroantennogram (EAG) results suggested that the Orco mutants of both male and female moths severely reduced their electrophysiological responses to the eight tested plant volatiles and two sex pheromones. However, the Orco gene played distinct roles in mating behavior between sexes: the mating behavior was fully disrupted in mutated males but not in mutated females. The oviposition result indicated that the Orco KO females displayed reduced egg laying by 24.1% compared with the mated wild type (WT) females. Overall, these results strongly suggest that Orco is an excellent target for disrupting FAW’s normal behavior and provides a feasible pest control approach.  相似文献

以长白母猪为对照,分析了1月龄、发情期和间情期姜曲海母猪子宫细胞质雌二醇受体(CER)和细胞核雌二醇受体(NER)以及细胞质孕酮受体(CPR)和细胞核孕酮受体(NPR)。3个时期该猪子宫CER分别为351、410和367fmol/mgDNA,其Kd值分别为7.1、4.8和4.0nmol;子宫NER分别为245,1390和620fmol/mgDNA,Kd值为3.7、6.6和4.4nmol;CPR分别为251、364和147fmol/mgDNA;NPR分别为232、337和184fmol/mgDNA。在发情期和间情期,姜曲海母猪子宫NER以及与细胞质受体的比值均比长白母猪高,说明此时姜曲海母猪子宫对雌二醇具有较高的敏感性。性成熟后,试验猪的雌二醇受体和孕酮受体均受它们配体激素的调节。  相似文献
