Summary An indirect sandwich enzyme-linked immunosorbent assay (ELISA) was used to monitor potato stems of three cultivars for the presence of the ring rot pathogen,Clavibacter michiganensis subsp.sepedonicus, during the growing season and in progeny tubers after harvest. The highest ELISA values were obtained with the highest concentration of bacteria used to inoculate seed pieces in all cultivars tested. Low ELISA values were obtained for stems and progeny tubers selected from plants grown from seed inoculated at lower bacterial concentrations. Estimates of bacterial densities in stems and progeny tubers by immunofluorescence indicated that low ELISA values were most probably caused by low bacterial numbers. It is suggested that the sensitivity of ELISA for detecting the ring rot pathogen in potato stems and progeny tubers is a function of the concentration of bacteria in individual seed pieces.  相似文献

The presence ofClavibacter michiganensis subsp.sepedonicus was detected by immunofluorescence and ELISA in diluent in which cores from 200 potato tubers had been incubated on a rotary shaker. By this procedure the ring rot pathogen was detected in composite samples from naturally infected tubers and tuber samples “spiked” with infected tissue or a pure culture of the pathogen. Populations of saprophytic bacteria increased about 100-fold in the diluent during 18 h of incubation but did not interfere with serological detection of C.m. sepedonicus. Some of the results suggest thatC. m. sepedonicus cells multiply during incubation of tuber samples. Extraction of ring rot bacteria from potato tuber tissue by incubation on a rotary shaker requires little work and minimizes the amount of plant tissue in the extract compared to the standard extraction procedure involving maceration and differential centrifugation.  相似文献

Summary Plant-to-plant transmission of the bacterial ring rot (BRR) pathogen,Clavibacter michiganensis subsp.sepedonicus (Spieck. et Kott.) Skapt. et Burkh. of potato (Solanum tuberosum L.), was studied in field trials over a three year period. Healthy and infected seed tubers were planted 35 cm apart. In one treatment, a subsurface barrier was placed between the healthy and infected seed tubers separating the root systems of neighbouring plants. In this treatment, none of 216 plants grown from healthy seed tubers was infected at harvest, as determined by indirect immunofluorescence antibody staining (IFAS) with monoclonal antibodies. In the other treatment, no subsurface barrier was used. In this treatment, two of 368 plants (0.5%) grown from healthy seed tubers were infected at harvest. It is concluded that plant-to-plant transmission may occur but at very low frequency, and is unlikely to play a significant role compared with the potential of transmission by shared potato handling equipment.  相似文献

Summary In seven years of field experiments, there were significant differences in the numbers of stems per plant and the number of harvestable tubers (>3 cm) at maturity, per plant and per stem, averaged for two cultivars and three seed treatments (Table 1). There was only a weakly positive correlation between the typical stem number for each year and the number of harvestable tubers per plant (r= +0.2, Table 2), whilst the correlation with the number of tubers initiated was much closer (r= +0.77*, Table 2). The negative influence of number of stems on number of tubers per stem is shown byr= −0.39 and −0.47 (Table 2). Identical stem populations in the wet, cold spring of 1987, and the warm, dry season of 1983 initiated fewer tubers per stem than in 1989 with its average temperatures and modest precipitation (Table 1). Tuber set per plant and per stem correlated positively with the number of harvested tubers per plant and per stem (r= +0.53 and +0.63) and accounted for some of the yearly variation. A much stronger effect on the number of harvestable tubers came from the annual reduction of tubers (r= −0.72 and −0.91*, Table 2). This was high if the availability of water deteriorated in the period of growth succeeding tuber formation (Table 1, Fig. 1). The same applied to the seed treatments planted at different stages of development. Tuber development occurred earlier with chitted seed, often at times when there was an adequate water supply. A large number of tubers were initiated and these were in large part reduced in the short dry spells which followed (Figs 2 and 4). Irrigation programmes must take account of this relationship.

Summary A sampling strategy was evaluated in the Andean highlands of Peru to optimise the detection ofRalstonia solanacearum in seed tubers harvested from symptomless crops. A sensitive and specific serological method developed at CIP was used to detect the pathogen in latently infected tubers. Optimum sample size was evaluated for symptomless crops after analysing various numbers of composite samples and using a binomial distribution model to calculate the detection probabilities.R. solanacearum was detected in all lots from fields with visible symptoms, so validating the detection technique. About half of the seed lots from apparently healthy fields at altitudes of up to 3,100 m were found positive for the pathogen.R. solanacearum was detected with 99% probability in samples of 350 tubers from seed lots from symptomless crops. This number of seed tubers could feasibly be processed in a seed-health test without incurring too high a cost for labour and materials.  相似文献

Summary Soil in a potato field naturally infested with black dot (Colletotrichum coccodes) was fumigated with methyl bromide at 126 g m−2 or left unfumigated. Potato seed tubers (cv. BP1) uninfected, lightly infected (1–25% surface affected), severely infected (26–100% surface affected) and severely infected withC. coccodes but dusted with prochloraz manganese chloride as Octave 2.5% DP at 750 g per 100 kg seed were planted in fumigated and unfumigated soil. When harvested, the incidence of black dot on the progeny of infected seed planted in unfumigated soil was twice that of progeny in fumigated soil, with progeny of uninfected seed having a 68.5 times higher disease incidence in unfumigated soil. Black dot on progeny tubers was reduced by pre-treatment of seed with prochloraz in fumigated soil only. With black dot infested fields, planting disease-free seed or treating seed with fungicides would not decrease disease on progeny tubers.  相似文献

Summary Three different seed treatments and two cultivars were studied over seven years of field experiments (Table 1). Differences were detected between the cultivars in all characteristics studied: numbers of stems, maximum number of tubers (tuber initials), the number of harvestable tubers (>3 cm) at maturity per plant and per stem, and tuber reduction (percentage of initiated tubers that did not develop). Apart from the number of harvestable tubers, there were also differences between seed treatments (Tables 2, 3 and 4). Chitted seed of ‘Mariella’ and sprouted seed of ‘Adretta’ gave the greatest number of tubers per plant (Tables 3 and 4). In both cases desprouting reduced the numbers of stems and tubers per plant but increased the number of harvestable tubers per stem. Chitted seed set the most tubers per plant and per stem but had the highest levels of tuber reduction (Table 4). Due to intra- and inter-stem competition in both cultivars the percentage tuber reduction is significantly correlated with the number of tuber initials (maximum tuber number, Table 5). Smaller numbers of tubers per stem (Table 3) and significantly negative correlations between the number of stems and the maximum and harvestable number of tubers per stem (Table 5) highlight the effect of strong inter-stem competition in ‘Adretta’. Moreover, in this cultivar intra-stem competition seems to play an important role too. The rate of tuber reduction was significantly higher than in ‘Mariella’ (Table 3), and the final number of tubers significantly depended on the percentage of tuber reduction during the period of tuber growth (r=0,57*, Table 5). In contrast, the number of harvestable tubers per plant in ‘Mariella’ was significantly correlated with the maximum tuber number during the period of tuber set (r=0,61*, Table 5).

Summary Methods for the estimation of amylolytic activity are reviewed. A procedure for the routine extraction of amylolytic activity from freeze-dried powder prepared from potato tubers is described. The extraction medium is buffered at pH 7.0 and contains glycerol, dithiothreitol, calcium chloride and the non-ionic detergent, nonidet P-40. α-Amylase activity and exoamylolytic activity were estimated in crude extracts of potato tubers using the substrates, blockedp-nitrophenyl-maltoheptaoside andp-nitrophenyl-maltopentaoside respectively. These substrates are included in kits supplied by Biocon, (UK) Ltd to measure the α- and ?-amylase activity in cereals. The validity of using these kits for the determination of α- and ?-amylase activity in potato tuber tissue is discussed.  相似文献

Zusammenfassung Die physiologische Alterung der Pflanzknollen durch Lagerung bei überoptimalen Temperaturen (jahresunterschiedlich 16–29,5°C, 1988–90 vor dem Pflanzen abgekeimt, 1991 vorgekeimt wie Kontrolle) führte im Vergleich zur vorgekeimten Kontrolle im Mittel von 25 Sorten in allen Versuchsjahren zu einer Verminderung der Bestandesdichte und des Ertrages. Eine Gruppe von Sorten erwies sich in allen Jahren als relativ tolerant gegen die Alterung, eine andere als signifikant empfindlicher, dazwischen lag ein breites Mittelfeld mit zum Teil erheblichen Schwankungen in den Jahren. Die Reaktion der Sorten zeigte keine Beziehung zu ihrer Reifegruppe und ihrer Keimintensit?t, dagegen war die sortentypische Ertragsdepression nicht sehr eng, aber signifikant mit dem Schwund w?hrend der Lagerung korreliert.
Summary Seed tubers were stored at above optimal temperatures. Mean temperatures varied during test years from 16 to 29.5°C (in 1988–1990 tubers desprouted before planting; in 1991 chitted tubers as in control; Table 1). This procedure of physiological ageing of seed tubers resulted in reduced plant density and lower yield compared to the chitted control in every year of the test period (average of 25 cultivars). The emergence was significantly delayed as compared to the control in all years (Table 2). Assessed for yield, the cultivars Désirée, Sanetta, Astilla and Andra showed a significantly higher degree of tolerance to warm storages (>80%, when compared to the control) than the cultivars Arkula, Dorisa, Karlena and Koretta (<25%; Table 3). Between these two extremes there was a large number of intermediate levels of tolerance, a result of fluctuating reactions of various cultivars to changing conditions in the years of the test period (Fig. 1). No correlation could be found between the cultivars' reactions and their maturity group or their sprouting behaviour. There was, however, for each cultivar a significant negative correlation between loss in weight through transpiration and respiration and plant density and yield (r=−0.55 and −0.54; Table 4). Plant density and yield were closely correlated (Table 4). The regressions between loss of plants and reductions in yields show that after seed tuber storage at higher temperatures a reduced yield must be expected even when plant density is not reduced (loss of plant 0%; Fig. 2). With complete stands, the use of physiologically aged seed tubers that were desprouted before planting (1988–1990) resulted in a reduction of yield of 17–20%; when old seeds were chitted as in control a loss in yield of 15% was calculated.

Summary The intercellular volume within tuber tissue increases with tuber maturation. At the same time, both tuber injury after mechanical load and lentical perforation after water uptake by tubers in wet peat decreases. The degree of tuber injury is correlated with intercellular space (r=−0.667,n=16) and with water saturation of tissue (r=0.697,n=16). The tubers lose water during storage. In wet soil wilted tubers take up significantly higher amounts of water than turgid tubers, and lenticel perforation is much more frequent. Tuber maturation in autumn and low water loss during storage enhance the cushioning effect of the intercellular gas volume of tuber tissue against outside mechanical load and internal tension after water uptake.  相似文献

Summary In eight experiments in 4 years, inoculating seed tubers withR. solani caused stem canker and in seven experiments decreased the total yield and the yield of tubers 40–70 mm. Yields of larger tubers were usually increased. In most experiments inoculating increased the reducing sugar content of the tubers and darkened the colour of crisps, but did not consistently affect the amounts of sucrose. Treating soil with aldicarb or oxamyl before planting also slightly increased the total reducing sugars.  相似文献

Summary Potato tubers were inoculated with two biotypes ofPhytophthora infestans then stored at 3,7, 10 and 15°C. Image analysis quantified average reflective intensity (ARI) of diseased tissue from cut surfaces of sample tubers. Tuber tissue infection and infection rate were measured by calculating Mean ARI of samples. Average tuber tissue infection and infection rate was minimal at 3°C (P.i.-US8 orP.i.-US1). Tuber tissue infection increased at temperatures >3°C, from 220 Mean ARI seven days after inoculation (dai) to 190–150 Mean ARI 50 dai (depending on cultivar and biotype ofP. infestans). Rate of tuber tissue infection caused byP.i.-US1 at 7°C was about zero in cv. Snowden but greater than −0.2 ARI day−1 (cvs Russet Burbank and Superior). Rate of late blight infection in tuber tissue generally increased with temperature from −0.2 ARI day−1 (at 7°C) in all cultivars to a maximum of −0.8 ARI day−1 (10°C).  相似文献

Summary In each of the five years 1978 to 1982, seed lots of cv. Bintje, variously infested byPhoma foveata Foister andFusarium solani var.coeruleum (Sacc.) Booth, as found by wound and cut tests, respectively, were planted in a series of field experiments at six sites in Sweden. A statistically significant relationship between seed and progeny infestation was found forP. foveata each year, and in three years forF. solani. Benomyl and thiabendazole mist treatments of seed tubers at grading in spring controlled the development of gangrene and dry rot in treated seed tubers, but did not consistently reduce the levels ofP. foveata andF. solani infestation in progeny from treated seed tubers. The site of cultivation markedly influenced mean levels ofP. foveata infestation, which were found to be related to temperature and rainfall during a 40 day pre-harvest period at the trail sites; however, mean levels ofF. solani infestation were not so related.  相似文献

Summary The viability of five pathogens was decreased by treatment with hot water when tested in vitro.Polyscytalum pustulans was most sensitive andRhizoctonia solani least sensitive. Potato tubers were exposed to 55°C for 5 min in a commercial continuous hot water treatment plant using naturally contaminated seed tubers and tubers which had been inoculated by dipping in comminuted cultures. The frequency of eyes colonised byP. pustulans, Helminthosporium solani, andR. solani was reduced to virtually zero and the effect persisted on tubers subsequently stored at 4°C and at 15°C for up to 16 wk. Results withColletotrichum coccodes were inconclusive. Treatment suppressedPenicillium spp. which, however, rapidly recolonised the eyes during storage, leading to higher contamination levels in the treated than in the untreated tubers. With tubers inoculated withPhoma foveata, good control was achieved when the incubation period before treatment was 10 d but not when the period was 42 d.  相似文献

Summary Potato seed tubers may suffer from premature sprouting during storage, thus limiting their suitability for cultivation. Commonly used sprout suppressant treatments negatively affect but viability and therefore a reliable method to inhibit bud development must still be found for seed tubers. The monoterpene carvone ((S)-(+)-carvone) was tested in small scale experiments. The vapour of this compound fully inhibited bud growth of tubers cv. Monalisa stored at 23°C without affecting bud viability throughout 6 months of treatment. The most effective range of carvone vapour concentrations was between 0.34 and 1.06 μmol mol−1. With these qualities we can expect carvone to become a suitable sprout suppressant for seed tubers.  相似文献

Summary Seed tubers, cv. Désirée, derived from stocks treated annually with thiabendazole were infected with thiabendazole-resistant strains ofHelminthosporium solani andPolyscytalum pustulans. Samples of seed tubers were either untreated or immersed for 5 min in fungicide suspensions of thiabendazole, imazalil or thiabendazole plus imazalil and planted on four farms in 1988 and 1989. After harvest, tuber samples from each treatment were treated with thiabendazole and stored for 6 months. Applying imazalil or thiabendazole plus imazalil to seed tubers decreased the severity of silver scurf and skin spot on stored progeny tubers. Thiabendazole applied to seed tubers or to progeny tubers after harvest did not affect the severity of either disease, but post-harvest treatment decreased the incidence of black scurf after storage.  相似文献

Summary Multiplication factors and progeny yield variation in crops from minitubers of five weight classes (ranging from 0.13–0.25 g to 2.00–3.99 g) and conventional seed tubers were studied in field experiments in three years. Multiplication factors were calculated as the number and weight of progeny tubers produced per planted tuber or per unit planted tuber weight. They were lower for the lighter minitubers when calculated per tuber and higher when calculated per weight. Yield variation was described by coefficients of variation for the number and weight of progeny tubers produced. Variation over individual plants of a crop was higher in stands from the lighter minitubers. Variation over plots within a field was sometimes higher for the lighter minitubers, but variation over years was similar for all minituber classes. Variation over plots in progeny tuber weight was higher for minitubers than for conventional tubers.  相似文献

Summary The effect of water stress on yielding capacity of potatoes was studied in 3 years using four different levels of soil water potential (0.7, 1.9, 3.4 and 7.8 atm.) in experiments in specially adapted structures that excluded uncontrolled water supply. Seed tubers from these crops were planted in the following years to investigate effects of water stress on their subsequent performance. Limited soil moisture availability decreased yield and the number and size of tubers. The growing period was shortened by 1–4 weeks and dormancy period by 2–8 weeks. In the following year, seed produced under conditions of moisture stress gave plants with 20% fewer stems. 24–33% less yield, 18–22% fewer tubers and 19–22% fewer large tubers than plants from seed produced under abundant water supply. It is concluded that the yield potential of seed tubers can be improved by careful attention to the availability of soil moisture during their production.  相似文献

Summary In five field trials with the cultivars Gloria (early), Bintje (mid-early) Désirée (mid-late) and Morene (late), crops grown from conventional 28–35 or 35–45 mm seed were compared with crops grown from microtubers, of various sizes and from various sources, that had been producedin vitro. The following means to quicken initial crop development from the micropropagated material were tested: large microtubers, plastic soil cover, and transplanting of plantlets grown from microtubers pre-planted in a glasshouse. Crops grown from microtubers weighing less than half a gram yielded much less than crops grown from conventional seed crops but their yields were increased by each treatment. With the later-maturing cultivars, which generally produce few tubers per plant, the yields within seed grades from plants grown from transplanted microtuber plants were comparable with those of conventional crops. Reasons are given, why direct planting of microtubers, with or without plastic foil, is not a practical option.  相似文献

Summary In three experiments seed tubers of cvs Jaerla, Bintje and Alpha, stored at 12 °C in the dark, were planted in glasshouses on various dates at soil temperatures varying from 8 °C to 28 °C to study the phenomenon of ‘non-emergence’ with ageing seed tubers. In general the tubers were desprouted before planting. At all plantings cv. Alpha produced normal plants. The cvs Jaerla and Bintje, however, showed high proportions of the ‘little potato’ phenomenon when planted in cold soil, and high proportions of decayed tubers more frequently in warm soil. When using physiologically older seed at later planting dates, both the range of soil temperatures became wider and the proportions of seed tubers experiencing both ‘little potato’ and decay increased.  相似文献
