An 8‐week growth trial was conducted to investigate the effects of non‐genetically modified (nGM) soybean (Youchun 06‐1) and genetically modified (GM) soybean (Roundup Ready®) with and without a heat treatment on the growth and health of three Cyprinidae species with different feeding habits (grass carp Ctenopharyngodon idellus, gibel carp Carassius auratus gibelio, and black carp Mylopharyngodon piceus; body weight: 283.0 ± 2.0 g, 60.5 ± 0.7 g, and 261.4 ± 3.1 g). Five diets (FM, nGMS, hnGMS, GMS, and hGMS) were made with fishmeal and full‐fat soybean (Youchun 06‐1, heat‐treated Youchun 06‐1, Roundup Ready®, and heat‐treated Roundup Ready®). The FM diet contained fishmeal as the sole protein source. The experimental diets contained full‐fat soybean meal as 60% of dietary protein. Both temperature (60, 80, 100, and 120°C) and duration (1 and 2 hr) of heating influenced trypsin inhibitor activity (TIA) and protein solubility of nGM soybean and GM soybean. The TIA in GM soybean was higher than that in nGM soybean. After heating at 120°C for 2 hr, each amino acid of soybean treatment showed a slight decline. Neither source of soybean nor heat treatment affected the growth performance, feed utilization, chemical composition of the whole fish body and muscle, and lipase or amylase activity of the three Cyprinidae species (p > 0.05). Heat treatment of soybean meal slightly increased the plasma antioxidant capacity of the three fish and plasma cholecystokinin of black carp and grass carp. The unheated soybean treatment adversely affected the height and density of black carp intestinal villi, and all the soybean treatments caused disruption of the grass carp intestinal epithelium.  相似文献

To understand the actual production of fish culture about the utilization of dietary protein and excreta impact on the environment between mandarin fish (Siniperca chuatsi) and grass carp (Ctenopharyngodon idellus), the study to investigate the effect of temperatures (19 ± 0.5°C, 24 ± 0.5°C and 29 ± 0.5°C) on ammonia‐N excretion in mandarin fish and grass carp under fed and fasted states was conducted. These two species were fed a practical diet containing 325.2 g kg?1 crude protein at 3% body weight per day. The ammonia‐N excretion rate was significantly increased when temperature increased from 19 to 29°C, and a linear relationship between ammonia‐N excretion rate and temperature. The maximum ammonia‐N excretion levels of mandarin fish and grass carp were observed at 4–8 h and 2–4 h after feeding, respectively, and the minimum values for both species were observed at 24 h after feeding. Under the feeding condition, mandarin fish had a lower ammonia‐N excretion level compared to grass carp at 24°C and 29°C. The average amount of ammonia‐N excreted by mandarin fish at 24 h is significantly higher than grass carp under fasting conditions, except 19 ± 0.5°C. These results indicated that mandarin fish might make better use of protein at higher temperature than grass carp when fed practical diets in commercial production. These results of this study suggested that mandarin fish had a lower ammonia‐N excretion level compared with grass carp, making a less contribution to environmental loading in an intensive fish culture.  相似文献

ABSTRACT

Changes in volatile compounds in dark and ordinary muscle of yellowtail during 2 days of refrigerated storage at 5°C were investigated. Twenty-seven compounds were identified in ordinary muscle and 39 in dark muscle during 2 days of storage at 5°C. Thirteen compounds at Day 0 of storage and 29 compounds at Day 2—such as 2,3-pentadione, 1-penten-3-ol, (Z)-2-penten-1-ol, and (E,Z)-2,4-heptadienal—identified in the dark muscle showed significantly higher values compared to ordinary muscle. Levels of thiobarbituric acid-reactive substances (TBARS) in dark muscle were significantly higher than those in ordinary muscle throughout 2 days of storage period, and a significant increase in TBARS occurred in just dark muscle at Day 2 of storage. In ordinary muscle, viable cells remained at the same order of magnitude as their initial values for 2 days. Eight aldehydes in ordinary muscle and 25 volatile compounds in dark muscle increased significantly without microbial action prior to increase in TBARS during short-term cold storage. Principal component analysis of the volatile compounds in dark and ordinary muscle was able to differentiate between different storage time samples of same muscle type as well as different muscle samples of same storage time.  相似文献

Abstract. The synthetic LH-RH analogue, des-GLY10[D-Ala6] LRH ethylamide was shown to be effective for induced spawning of Chinese carps. Ovulation was induced in 23 of 34 female grass carp, Ctenopharyngodon idella Val., receiving a single injection of synthetic LH-RH analogue. Spawning success was 92% for 3-year and older fish at optimum temperatures (22–26°C). Reduced spawning success was noted for 2-year-old female fish, and when temperature exceeded 27°C. No mortality of study fish was observed following a single injection. Hybrid carp, grass carp x bighead carp, Aristichthys nobilis Val., and grass carp x silver carp, Hypophthal-michthys molitrix Val., were produced using synthetic LH-RH analogue. Male grass carp, bighead carp and silver carp exhibiting pearl organs were stimulated to produce milt regardless of the presence or absence of free flowing milt at the time of injection. The potential importance of synthetic LH-RH analogue in fish culture and mechanisms of hormonal action are briefly discussed.  相似文献

This study explored the pharmacokinetic characteristics of bronopol in grass carp (Ctenopharyngodon idella) under different temperatures. The concentrations of bronopol in the plasma and tissues (kidney, liver and muscle) of grass carp after soaking in bronopol (6.75 µg/ml) at 15 and 20°C were determined using liquid chromatography–mass spectrometry. The concentrations of bronopol in both plasma and tissue first increased and then decreased with soaking time at both tested temperatures. The peak bronopol concentrations at 15°C (plasma: 8.934 µg/ml; kidney: 9.23 µg/g; liver: 9.47 µg/g; and muscle: 7.98 µg/g) were slightly lower than those at 20°C (plasma: 9.654 µg/ml; kidney: 10.83 µg/g; liver: 13.40 µg/g; and muscle: 8.68 µg/g). In an analysis of pharmacokinetic parameters, the bronopol concentration–time profiles for plasma were best described by a two‐compartment, open pharmacokinetic model with first‐order absorption. The t1/2β for plasma at 15 and 20°C was 82.933 and 92.041 hr, respectively, indicating that the elimination of bronopol was faster at 15°C than at 20°C. In addition, AUC0‐t and AUC0‐∞ values of bronopol were 477.892 and 495.809 µg/L hr at 15°C and 495.809 and 589.859 µg/L hr at 20°C, indicating that the content of bronopol in the organism was higher at higher temperature.  相似文献

为探究种青养鱼养殖模式对草鱼肌肉营养成分和品质特性的影响,随机选取以种青喂草为主养殖模式下养殖的草鱼(生态草鱼)和投喂人工配合饲料进行养殖的草鱼(饲料草鱼)各16尾,测定其肌肉系水力和质构特性指标,以及肌肉常规营养成分、矿物元素、氨基酸和脂肪酸含量。结果显示,生态草鱼和饲料草鱼肝体比和空壳率无显著性差异;生态草鱼肌肉系水力指标中滴水损失显著低于饲料草鱼,冷冻渗出率不显著地低于饲料草鱼,失水率不显著地高于饲料草鱼,p H值无显著性差异;肌肉的硬度、弹性、凝聚性、胶黏性和回复性均无显著性差异;生态草鱼粗脂肪含量显著低于饲料草鱼,水分、灰分、粗蛋白含量均无显著性差异;生态草鱼P和Fe含量均显著高于饲料草鱼,Mg、Mn和Cr含量均极显著高于饲料草鱼;生态草鱼和饲料草鱼肌肉氨基酸组成基本一致,均含有17种氨基酸,其中人体必需氨基酸总量分别为6.85%和6.27%。生态草鱼必需氨基酸指数(EAAI)为76.07,而饲料草鱼为77.29,饲料草鱼略高于生态草鱼。生态草鱼和饲料草鱼肌肉均含19种脂肪酸,其中棕榈酸、花生四烯酸(ARA)、亚油酸(LA)、油酸、二十二碳六烯酸(DHA)和硬脂酸含量较高,为主要脂肪酸,而花生五烯酸(EPA)+DHA含量分别为8.95%和10.70%,且差异显著。研究表明,生态草鱼和饲料草鱼在肌肉质构特性方面无显著性差异,与饲料草鱼相比,生态草鱼具有肌肉系水力强、低脂肪和矿物元素含量高的特点。  相似文献

A study was performed to determine the role formalin fixation plays in flow cytometric (FCM) analysis of DNA in nucleated blood cells. Blood was frozen and stored at −80°C in a DMSO-citrate-sucrose buffer. When FCM analysis was needed the sample was thawed and added to a large volume of propidium iodide (PI) stain and analyzed after not less than 10 min of staining. No centrifugation steps were necessary. This protocol has been used by us for the discrimination of triploid from diploid Chinese grass carp (Ctenophary ngodon idella). Fixation of PI-staned nucleated RBCs with a final concentration of 1–2% phosphate-buffered formalin significantly lowered the amount of fluorescence recorded from the sample even when the fixed and stained RBCs were washed free of the fixative. Fixation with formalin prior to staining with PI yielded similar results. Our results show that formalin fixation of samples prior to or after staining with PI may lead to problems in data interpretation not previously appreciated. A FCM study was done to select the most appropriate biological DNA reference standard for grass carp RBCs. In this study DNA histograms of red blood cells from chicken, rainbow trout and diamondback watersnake as well as human nucleated white blood cells were compared to grass carp DNA histograms. This study examined how muscle metabolic organization varied during an annual cycle in which rainbow trout (Oncorhynchus mykiss) were held in outdoor holding ponds in which they were exposed to natural changes in temperature (range 0.2 to 15.6°C) and photoperiod. We examined the activities of glycolytic and mitochondrial enzymes in red and white muscle to evaluate whether trout enhance their capacity for lipid and carbohydrate oxidation during cold-acclimization. When assayed at habitat temperature, the enzyme activities generally increased in spring to reach a maximum in summer followed by a decrease in the fall. This led to significantly higher activities at warm than cold periods for all enzymes measured in red muscle and all but one in white muscle. The activities at 10°C provided little evidence for compensatory adjustments of aerobic capacity. Particularly in red muscle, enzyme levels at 10°C were generally lower during cold than warm periods. The variation of enzyme activities throughout the cycle was not due to changes in protein concentration, as the same responses were observed when activities were expressed per g wet mass or per mg protein. Although the aerobic capacity did not increase with cold-acclimatization, the relative capacity for lipid oxidation was higher in winter than in summer trout. In contrast, the relative capacity for aerobic glycolysis was higher in summer than in winter trout. Thus, the metabolic capacities of trout muscle undergo seasonal reorganization.  相似文献

研究了等静超高压对草鱼肌肉超微结构与质构特性的影响,并与猪肌肉用对照。结果表明:1.加压处理的草鱼肌肉,其超微结构中出现较明显的变化,如肌原纤维中A带和I带的细丝均被破坏,粗丝相互聚集,出现间隙,Z线变粗,不连续,H带和M线消失等,但肌原纤维的外形仍保持完整。2加压处理的草鱼肌肉,其外观无显著变化,色泽稍白,略有汁液流出,但质构特性与处理前无明显差别。  相似文献

以商业配方为基础,研究不同糖源(小麦、大麦、玉米、白高粱、木薯)膨化饲料对大规格草鱼[初始体质量(400.77±7.45) g]生长、体成分、越冬及部分生理生化指标的影响。共设置5个处理,每处理3个重复,每个重复30尾鱼,养殖实验于水库网箱(2.0 m×2.0 m×2.0 m)中进行,饲养时间为16周,养殖实验结束后,每网箱保留15尾进行越冬实验,时间为16周。结果显示,玉米组草鱼增重率(WGR)和蛋白质效率(PER)显著低于其他各组,饲料系数(FCR)显著高于其他各组,其他4组差异不显著;肥满度(CF)小麦组最高,脏体比(VSI)玉米组最高,肝体比(HSI)和肠体比(ISI)木薯组最高;小麦组血糖(GLU)含量显著高于其他各组,大麦和白高粱组总胆固醇(T-CHO)含量显著高于其他3组,木薯组甘油三酯(TG)含量显著低于其他各组。小麦、大麦和玉米组的肌肉粗脂肪含量显著高于白高粱和木薯组,木薯组的粗蛋白含量显著低于其他4组。白高粱和木薯组的肝糖原显著高于其他组,大麦和白高粱组的肌糖原显著高于其他组。越冬后,大麦组草鱼体质量下降幅度显著高于其他各组,HSI下降幅度显著小于其他各组,肝糖原含量显著升高,GLU下降幅度显著低于其他各组,肌肉粗蛋白含量下降幅度最高,其他各组越冬后体质量下降幅度差异不显著,小麦组CF下降比例最高,玉米和木薯组HSI下降幅度显著大于其他各组,玉米组的VSI下降比例最大。研究表明,在膨化工艺条件下,以生长、增重成本和越冬为评价指标,小麦、木薯和白高粱是大规格草鱼适合的糖源,木薯增重成本最低。  相似文献

为研究草鱼MSTN-1基因多态性及与早期生长性状和肌肉成分的相关性,本研究扩增出全长为3824 bp的草鱼MSTN-1基因,用长江选育草鱼群体对MSTN-1基因多态性进行筛选和验证。结果共发现3个多态性位点(Locus 1:C1799T,野生型EE/突变型EF;Locus2:C1842T,野生型HH/突变型HI;Locus 3:TGAAGCGCTGGTTCT/2585-,野生型BB/缺失型BD)。利用一般线性模型分析3个位点及其组合型(剔除个体数少于3的组合)与生长性状和肌肉成分的相关性,发现2个位点对幼鱼生长性状表型差异有显著影响,但3个位点对肌肉成分差异均无显著影响。多重比较发现,单倍型HI突变组的体长和体质量显著高于HH野生组,BD突变组的体长和体质量显著性低于BB野生组;多倍型中存在HI突变组合的体长、体质量均显著高于其他组,存在BD突变的组合在体长性状方面显著低于其他组。表明草鱼MSTN-1基因3个SNPs中,HI突变是对草鱼生长性状的有利突变,BD突变是不利突变,而EF突变无显著影响,可将MSTN-1基因作为分子标记辅助草鱼选育的候选基因。  相似文献

Gel properties of blends of surimi from silver carp and white croaker set for a range of times and at different temperatures were evaluated for breaking force, breaking distance, and whiteness. Total protein content was 12% in all samples, and the concentration of white croaker surimi protein in the blends was designed at 0, 10, 20, 30, and 40%. The blended surimi had greater gel strength than either of the two types of surimi alone. Breaking force and breaking distance after setting were also significantly higher than that of the blended surimi cooked directly without setting. The blended surimi exhibited less modori than either of the two types of surimi alone heated to 50°C. The whiteness values of the blended surimi were significantly higher than that of white croaker alone. These results indicate there are advantages to be gained in utilizing surimi from freshwater silver carp in blends with surimi from marine captured white croaker which is becoming limited in supply.  相似文献

The study was to investigate effects of dietary chlorogenic acid (CGA) on growth performance, flesh quality and serum biochemical indices of grass carp (95.1 ± 0.3 g) (Ctenopharyngodon idella) fed seven different diets, including control diet, Eucommia ulmoides (EU)‐supplemented diet (20 g kg–1) and CGA‐supplemented diets containing 100, 200, 400, 600 and 800 mg/kg CGA. Contents of collagen and alkaline‐insoluble collagen in muscle and skin were significantly increased by dietary CGA and EU (< .05). Total essential amino acids (TEAA) and total amino acids (TAA) in muscle of grass carp fed EU diet or 400, 600 and 800 mg/kg CGA diet were significantly higher than those of fish fed control diet and 100 and 200 mg/kg CGA diet (< .05). Fish fed 200–800 mg/kg CGA showed significantly lower muscle crude lipid content than EU, control and 100 mg/kg CGA groups (< .05). Fish fed CGA‐supplemented diets (100–800 mg/kg) had significantly higher muscle fibre density and lower muscle fibre diameter than control group (p < .05). In conclusion, supplementation of CGA improved flesh quality of grass carp, and supplemental level of CGA for improving flesh quality and growth was estimated to be 400 mg/kg diet.  相似文献

In this study, two experiments were performed to explore the function of silymarin in adipogenesis in grass carp (Ctenopharyngodon idellus) using in vitro and in vivo models. In experiment 1, differentiated grass carp pre-adipocytes were treated with silymarin for 6 days. Treatment with 100 μg mL?1silymarin (SM100 group) significantly reduced triglyceride accumulation at day 6. The adipogenic gene expression levels of PPARγ, C/EBPα, SREBP1c, FAS, SCD1, and LPL, and the protein expression level of PPARγ were significantly down-regulated in the SM100 group. Additionally, the SM100 group had significantly lower reactive oxygen species production and reduced glutathione contents compared with the control in vitro. In experiment 2, the juvenile grass carp (mean body weight= 27.4 ± 0.17 g) were fed six isonitrogenous and isocaloric diets in a factorial design containing 0, 100, or 200 mg kg?1 silymarin (SM0, SM100, SM200) associated with either 4 or 8% lipid levels (low lipid, LL, and high lipid, HL, respectively) for 82 days. The results demonstrated that dietary silymarin supplementation significantly reduced the elevated intraperitoneal fat index in grass carp fed with high-lipid diets, and the gene expression of adipogenesis (PPARγ, FAS) when supplemented with dietary silymarin was notably lower than when no silymarin was supplemented under the high-lipid diets. Thus, our data suggest that silymarin suppressed lipid accumulation in grass carp both in vitro and in vivo, and the effect might be due to an influence on the expression of adipogenesis factors and ROS production partly associated with effects on antioxidant capability.  相似文献

The effect of different thawing methods—running water, microwave, slack water (25°C), air (25°C), and refrigerator (4°C)—and multiple freeze-thaw cycles (0, 1, 3, and 5) on the quality of common carp were investigated. Microwave thawing showed the lowest score for sensory assessment and the highest cooking loss (p < 0.05), while 4°C refrigerator thawing showed less thawing loss and cooking loss but required the longest thawing time. Ca2+-ATPase activity and sulfhydryl content of air thawing were lower (p < 0.05), while no significant differences were observed among other thawing methods. Water loss and the thiobarbituric acid reactive substances (TBARS) value increased (p < 0.05) when freeze-thaw cycles increased. The sensory scores and hardness decreased significantly when the freeze-thaw cycles increased (p < 0.05). A significant decrease (p < 0.05) was observed in salt-extractable protein content after five freeze-thaw cycles. Different thawing methods affected the quality of common carp. Frozen common carp subjected to freeze-thaw cycles resulted in accelerated lipid and protein oxidation and caused mechanical damage to muscle tissues.  相似文献

Juvenile mirror carp were fed diets containing 303.4, 321.7, 341.2, 361.0 and 379.1 g kg?1 proteins, respectively, and reared at different water temperatures (18, 23 and 28°C) for 60 days. Gene expression of heat shock protein gene (Hsp70) and the warm temperature acclimation‐related 65 kDa protein gene (Wap65), immunity and antioxidant status in the carp were investigated. Results indicated that the contents of serum complement 3 (C3), complement 4 (C4) and immunoglobulin M (IgM), as well as activities of liver superoxide dismutase (SOD) and lysozyme (LSZ) were significantly enhanced with increasing dietary protein (< 0.05), while content of malondiadehyde (MDA) decreased. Gene expression level of Wap65 in the liver significantly increased with dietary protein, while gene expression of Hsp70 decreased. The contents of C3, C4 and IgM, the activities of SOD and LSZ and gene expression level of Wap65 in the liver significantly increased with temperature. These results suggest that: Serum immune parameter, antioxidant enzymes and Hsp70 and Wap65 expression interact in fish to improve ability to adapt to the environment; and the optimal conditions for the immunity of carp are 348.1?354.5 g kg?1 protein at 18°C, 352.3?364.9 g kg?1 at 23°C and 360.2?364.3 g kg?1 at 28°C, and the optimum temperature for carp is 23°C.  相似文献

Histamine is the main causative agent of scombroid poisoning. However, unlike scombroid fish, histamine poisoning due to consumption of flying fish has never been reported. In this study, the white muscle of flying fish had high levels of free histidine at approximately 423.9 mg/100 g, and was inoculated with Staphylococcus xylosus Q2 isolated from dried flying fish at 5.0 log CFU/g and stored at ?20 to 35°C to investigate histamine-related quality. The histamine contents quickly increased to higher than 50 mg/100 g in samples stored at 25 and 35°C within 12 h as well as stored at 15°C within 48 h. However, bacterial growth and histamine formation were controlled by cold storage of the samples at 4°C or below. Once the frozen flying fish samples stored at ?20°C for 2 months were thawed and stored at 25°C after 24 h, histamine started to accumulate rapidly (>50 mg/100 g of fish). Therefore, flying fish muscle was a good substrate for histamine formation by bacterial histidine decarboxylation at elevated temperatures (>15°C) when it is contaminated with S. xylosus. In conclusion, since the improperly contaminated flying fish muscle with S. xylosus could lead to production of hazardous levels of histamine over time when stored at temperatures >15°C, the flying fish should be stored below 4 °C or below to control proliferation of S. xylosus, and TVBN and histamine production.  相似文献

Salted grass carp fillets were dried by hot air drying (HD, at 35 and 45°C) and vacuum microwave drying (VMD, at 1, 4, and 7 W/g) to a final moisture content. Compared with hot air drying, the vacuum microwave drying greatly reduced drying time. The rehydration rate constant of salted grass carp fillets dried by microwave vacuum drying at 7-W/g microwave intensity was significantly higher than that of the hot air drying at 35 and 45°C. Compared with hot air drying, the vacuum microwave dried samples had a higher crude fat content (dry basis). The lightness of the samples dried by vacuum microwave drying was higher, and the yellowness was lower than those dried by hot air drying. The drying methods had no significant effect on the hardness and springiness.  相似文献

The effects of temperature on Pagellus bogaraveo muscle cellularity were investigated, by morphometry, throughout the endogenous feeding stage, testing two rearing temperatures: 14 and 18 °C. The following parameters were estimated in transversal body sections at post‐opercular and post‐anal body levels: the total cross‐sectional muscle area, the total number of fibres and the mean cross‐sectional fibre area. At hatching, no significant influence of the temperature was observed on the morphometric parameters measured in the white muscle. At mouth opening, an increase in the number of post‐opercular white fibres was promoted by the highest temperature. During embryonic development, the red muscle fibre number in the post‐anal part of the larvae increased with higher temperature, but it appears that the difference was no longer present at mouth opening. An increase in the fibre area and in the total cross‐sectional area of red muscle at the post‐anal level was promoted by a 4 °C increase in the temperature during the vitelline phase. In conclusion, the axial musculature of blackspot seabream embryos/larvae reacted differently to temperature influence according to the body location, strongly supporting the need to look at and account for different body locations when evaluating muscle cellularity in fish, namely in growth/aquaculture‐related studies.  相似文献
