Eighteen suckled Brahman cows were allotted randomly to treatments arranged in a three-period crossover design according to calving date and prior treatment such that each cow received 30, 150 and 300 IU oxytocin (OT) i.v. on d 10, 20 or 30 postpartum. Blood was collected via an indwelling jugular catheter every 15 min for 195 min. Samples collected before OT administration were used to determine basal plasma 13,14-dihydro-15-keto prostaglandin F2 alpha (PGFM) concentration. Day, time and the day X dose interaction affected PGFM (P less than .0001). All doses of OT elevated PGFM on all days postpartum (P less than .0001). Basal PGFM was greater (P less than .0001) on d 10 (252.2 +/- 51.2 pg/ml) than on d 20 (78.2 +/- 14.8 pg/ml) or on d 30 (64.8 +/- 7.4 pg/ml). The rise in PGFM in response to OT was greatest on d 10 and decreased (P less than .001) with increasing days postpartum. On d 10, 150 IU of OT caused a greater (P less than .0007) rise in PGFM than either 30 or 300 IU. On d 20, the 300-IU dose raised PGFM more (P less than .005) than either 30 or 150 IU, whereas on d 30 no differences among doses were detected. Cows had higher basal PGFM and a greater response to OT on d 10 postpartum than on d 20 or 30; cows were more responsive on d 20 than on d 30. Uterine infections (i.e., endometritis) can have a major economic impact on dairy production. Identifying cows that are susceptible to endometritis and improving the diagnosis of endometritis could lead to a reduction in the impact of such infections. Thus, we used Holstein cows to determine whether postpartum changes in 13, 14-dihydro-15-keto-PGF2alpha (PGFM), a metabolite of PGF2alpha, could be used to identify cows that are susceptible to endometritis and to improve the diagnosis of endometritis. Cows were assigned to three treatments. 1) Control (n = 10) had no clinical or bacteriological signs of endometritis during the study. 2) Treated (n = 11) developed endometritis spontaneously and were treated i.m. with 25 mg of PGF2alpha immediately after clinical diagnosis (d 17.6 +/- 0.8 postpartum; mean +/- SEM). 3) Untreated (n = 10) developed endometritis spontaneously and were not treated after diagnosis (d 20.0 +/- 0.5). Examinations of external and internal genitalia and bacteriological data were used to diagnose endometritis. From d 0 (calving) until approximately d 63 postpartum, jugular blood was collected three times weekly. Progesterone and PGFM were quantified in plasma. For PGFM, the treatment x day interaction was significant (P < 0.01). Overall PGFM profiles for Control and Treated differed (P < 0.05), but the Untreated profile did not differ from either Control or Treated. To better understand the interaction, PGFM data from d 0 to 35 postpartum were partitioned into consecutive 7-d periods, and d-36 and greater data were partitioned into one period. Effects of treatment, day, and the treatment x day interaction were then evaluated within period. Except for the d-15 to -21 period, PGFM was greater (P < 0.03) in Control than in Treated and Untreated. In Treated and Untreated, PGFM increased during the d-15 to -21 period. For progesterone, treatment did not affect the profiles, but day was significant (P < 0.001). Progesterone concentrations were basal from d 0 until approximately d 12, and they generally increased after d 12. Onset of endometritis was associated with increased progesterone concentrations. Treatment did not affect the interval from calving to first detected estrus (29.5 +/- 4.9 d) or from calving to AI (73.3 +/- 8.7 d). We conclude that PGFM measures have the potential to be used to identify cows that are more likely to develop endometritis and that PGFM may aid in the diagnosis of endometritis.  相似文献

Effects of heat stress during the last third of gestation on reproductive changes postpartum were studied in Holstein cows. Cows and heifers 160 to 190 d of gestation were assigned in June 1978 to shade (n = seven cows and two heifers) or no shade (n = eight cows and two heifers) management treatments. After parturition, all cows (n = 19) were moved to the milking herd and managed uniformly. On the day of calving and on each Monday, Wednesday and Friday thereafter until d 50 postpartum, jugular blood samples were collected. Beginning approximately 7 d postpartum, the reproductive tract of each cow was examined rectally after collection of blood samples. Estrus was monitored twice daily and cows were inseminated after d 45 postpartum. Prepartum heat stress increased 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) concentrations postpartum and increased the rate of uterine involution. Regardless of prepartum treatment, progesterone concentrations indicated that luteal phases had begun by d 12.4 +/- 1.3 postpartum, which was about 3 d before PGFM was basal. The first luteal phase lasted only 10.7 +/- .9 d. First estrus was not detected until d 32.3 +/- 4.8 postpartum. The previously gravid uterine horn had a negative effect on ovarian volume, diameter of the largest follicle and percentage of ovaries with a corpus luteum. However, prepartum heat stress attenuated this effect. This study indicates that heat stress prepartum had residual effects on postpartum reproductive changes, and that the previously gravid uterine horn exerted some control, which was attenuated by heat stress, over ovarian recrudescense. Even though heat stress prepartum affected sensitive measures of postpartum reproductive function, it did not alter days to first estrus, days open (102.3 +/- 13.1) or services/conception (2.5 +/- .3).  相似文献

Two experiments were conducted with the opioid antagonist naloxone to determine the effect of opioid receptor blockade on hormone secretion in postpartum beef cows. In Exp. 1, nine anestrous postpartum beef cows were used to measure the effect of naloxone on serum luteinizing hormone (LH), cortisol and prolactin concentrations. Cows received either saline (n = 4) or 200 mg naloxone in saline (n = 5) iv. Blood samples were collected at 15-min intervals for 2 h before and after naloxone administration. Serum LH concentrations increased (P less than .01) in naloxone-treated cows from 1.8 +/- .04 ng/ml before treatment to 3.9 +/- .7 ng/ml and 4.2 +/- .5 ng/ml at 15 and 30 min, respectively, after naloxone administration. In contrast, LH remained unchanged in saline-treated cows (1.6 +/- .3 ng/ml). Serum cortisol and prolactin concentrations were not different between groups. In Exp. 2, 12 anestrous postpartum beef cows were used to examine the influence of days postpartum on the serum LH response to naloxone. Four cows each at 14 +/- 1.2, 28 +/- .3 and 42 +/- 1.5 d postpartum received 200 mg of naloxone in saline iv. Blood samples were taken as in the previous experiment. A second dose of naloxone was administered 2 h after the first, and blood samples were collected for a further 2 h. This study tested the hypothesis that the increased glucose requirement of lactation had effects that were independent of the suckling-dependent inhibition of postpartum endocrine function in beef cows. Mature Hereford cows were either suckled ad libitum and infused with saline iv (n = 9) from d 2 through 4 (d 0 = jugular catherization on d 32 +/- 3 postpartum); were nonsuckled and infused with saline from d 2 through 4 (n = 10); or were nonsuckled and infused with phlorizin (3 g/d) from d 2 through 4 (n = 10). Nonsuckled cows infused with phlorizin had lower (P less than .05) plasma concentrations of glucose and amino acid nitrogen (AAN) on d 2 compared with pre-infusion levels (d 1), but their metabolic profile returned to levels similar to the suckled cows by d 3 and 4. Nonsuckled cows infused with saline had elevated glucose and insulin and lower AAN and free fatty acids (FFA) on d 3 and 4 compared with pre-weaning (d 1) levels (P less than .05). Nonsuckled cows infused with phlorizin did not show this weaning-induced elevation in glucose and insulin. The number of luteinizing hormone (LH) pulses was not affected by treatment. However, in contrast to the large LH pulses observed in the nonsuckled cows infused with saline, both the suckled cows and the nonsuckled cows treated with phlorizin had more small and fewer large amplitude pulses (P less than .01). Treatment did not affect serum concentrations of follicle stimulating hormone, gonadotropin release in response to gonadotropin releasing hormone (25 micrograms) or the number of cows ovulating by 55 d after calving. We conclude that the increased glucose clearance caused by phlorizin infusion or lactation results in depression of LH pulse amplitude in suckled postpartum beef cows.  相似文献

Two trials were conducted in which Angus x Hereford first-calf cows were assigned randomly at calving to one of two treatments: exposure to mature penile-blocked bulls (BE) or isolation from bulls (NE). In Trial 1 (BE, n = 38; NE, n = 37), cow to bull ratio increased from 12:1 to 19:1 over a 14-d period; in Trial 2 (BE, n = 25; NE, n = 24), this ratio was maintained at 13:1. In both trials, blood samples were collected weekly for progesterone and ovaries and uteri of cows were examined rectally. Cows were observed for estrus twice daily (am:pm) beginning 10 d after calving. In Trial 2, intensive blood sampling for LH began 10 d after calving (eight cows per treatment) and continued at weekly intervals until estrus or the end of the trial. Postpartum weight change, condition score change and time to uterine involution did not differ (P greater than .10) between treatments in either trial. Interval to estrus was shorter (P less than .05) for BE cows than for NE cows in both trials. A greater proportion (P less than .05) of BE cows exhibited estrus by 60 and 90 d after calving and showed an increase in progesterone before first estrus. Mean and baseline LH concentrations and amplitude, frequency and duration of LH pulses were not altered (P greater than .10) by bull exposure. In conclusion, exposing first-calf suckled beef cows to bulls after calving hastened resumption of estrous cycles. Bull exposure did not alter patterns of LH concentrations but did increase proportions of cows that showed increased progesterone before first estrus.  相似文献

Uterine and cervical involution after calving was measured sequentially in 25 parous commercial Friesian cows by using electronic calipers and real-time ultrasound imaging, transrectally. Ovarian activity was monitored by using real-time ultrasound imaging and by the assay of milk progesterone concentrations. Peripheral plasma prostaglandin F2 alpha metabolite concentrations were assayed during this period. Five groups of five cows were treated intramuscularly, approximately 48 hours after calving, with either 100 mg progesterone in oil, 25 mg dinoprost tromethamine, 5 mg oestradiol benzoate, 1.2 mg of the long-acting oxytocin analogue carbetocin or 5 ml sterile water. There were no statistically significant differences between the intervals from calving to the completion of involution or between the intervals from calving to the first ovulation for the cows in the different groups. The mean PGFM concentrations in the peripheral circulation were significantly higher in the carbetocin-treated group, presumably owing to the stimulation of endogenous prostaglandin F2 alpha secretion. This study provides no evidence that a single treatment with any of the four hormones significantly affected the reproductive function of the cows after calving.  相似文献

Five commonly practised veterinary procedures were studied: Palpation per rectum of the reproductive tract, intramuscular injection, single venepuncture, repeated venepuncture and jugular vein catheterisation. Plasma cortisol concentrations increased from baseline values of approximately 2 ng/ml to maximum mean values between 6.5 +/- 2.5 ng/ml and 13.8 +/- 5.6 ng/ml approximately 13 to 27 minutes after each manipulation. Baseline values occurred approximately 80 minutes later. In the control bleeding periods unacclimatised cows initially had high values of plasma cortisol (5 to 10 ng/ml) which returned to baseline after two hours, ie, before beginning any procedure. There were no statistically significant changes in luteinising hormone concentrations. The concentration of 13,14 dihydro, 15-keto prostaglandin F2 alpha (PGFM) increased from 61.0 +/- 4.6 pg/ml to 209.8 +/- 152.1 pg/ml in three out of five cows palpated on days 16 and 19 of the oestrous cycle. Increases did not occur in five other cows palpated during the follicular phase, nor in five cows palpated on day 12. However, after palpation on day 8, one animal did have concentrations of PGFM similar to those occurring during spontaneous release on days 18 to 20 of the oestrous cycle.  相似文献

Plasma progesterone, oestradiol, 13,14-dihydro, 15-keto, prostaglandin F2 alpha (PGFM) and luteinising hormone were measured in daily samples from eight dairy cows for 15 days before and after spontaneous calving, and compared to patterns obtained from 15 cows during and after induction of parturition with dexamethasone +/- prostaglandin. The average (+/- sd) length of pregnancy in the control cows was 285 +/- 3.04 days compared to 264 +/- 7.4 days in the induced group. Only nine cows required prostaglandin on day 11 after dexamethasone, which resulted in all nine calving 1.7 +/- 0.3 days later. Eight of the 15 treated cows retained the fetal membranes; however, the calving to first heat interval for all treated cows was 46.7 +/- 10 days (range 30 to 66 days) and calving to conception 52.4 +/- 17 days (range 30 to 85 days) compared to 42.1 +/- 15 days (range 28 to 64 days) and 76.5 +/- 24 days (range 54 to 115 days) respectively for the eight cows which calved without induction. The pre-partum hormone profiles were similar in all cows. Hormone profiles in the post partum period were also similar in all cows, irrespective of previous history. Baseline values for progesterone and oestradiol were not reached until 4.6 +/- 1.7 days and 2.75 +/- 0.7 days post partum respectively, whereas PGFM values increased for two or three days, reaching baseline levels 8.87 +/- 2.10 days later. Two experiments were designed to examine whether hormonal profiles were related to luteal life span in pluriparous postpartum anestrous beef cows. Cows (Exp. 1, n = 34; Exp. 2, n = 23) received norgestomet (N) for 9 d or served as controls (C). Each cow received 1,000 IU human chorionic gonadotropin (hCG) 48 h after removal of N (d 0). Blood samples collected every 15 min for 8 h on d -5, 3 and 5 (Exp. 1) or on d -10 and -1 (Exp. 2) were assayed for luteinizing hormone (LH) and follicle stimulating hormone (FSH). Cortisol was determined in hourly samples collected on d -5 and in samples collected every 2 min during suckling on the same day (Exp. 1). Concentrations of 15-keto-13,14-dihydro-PGF2 alpha (PGFM) were determined in samples collected at 15-min intervals for 2 h on d -5, 3, 5 and 10 (Exp. 1). Estradiol-17 beta was measured in samples collected on d -5 (Exp. 1) or on d -10 and -1 (Exp. 2). Life span of induced corpora lutea was longer (P less than .05) in N than C cows. Percentages of N cows in which corpora lutea, formed in response to hCG, exhibited a normal life span were 83% on farm 1 and 25% on farm 2 (Exp. 1), and 90% (Exp. 2), compared with 0% in C cows. Concentrations of FSH were not affected by N but were lower (P less than .05) on d -5 in cows on farm 2 (.6 +/- .1 ng/ml) than in cows on farm 1 (.8 +/- .1 ng/ml). On d -5, a treatment X farm interaction (P less than .05) for mean LH was observed and frequency of pulses of LH was higher (P less than .01) in N than C cows (2.7 +/- .4 vs. .8 +/- .8 pulses/8 h). Neither cortisol nor PGFM was affected by N. Estradiol was increased in d -1 (6.1 +/- .5 vs 2.6 +/- .8 pg/ml; P less than .01) by N. It is suggested that pre-treatment with N enhanced life span of induced corpora lutea, in part, by influencing secretion of LH and development of follicles, but a threshold concentration of FSH was required for N to exert this effect.  相似文献

A possible role for endogenous opioid peptides (EOP) in the control of luteinizing hormone (LH) and prolactin (PRL) secretion was studied by injecting the opioid antagonist, naloxone (NAL), into postpartum ewes and cows. Twelve ewes that lambed during the fall breeding season and nursed their lambs were injected iv with NAL (1.0 mg/kg) on d 10, 14, 18, 22 and 26 postpartum. Blood samples were collected at 15-min intervals from 2 h before to 2 h after NAL, and serum concentrations of LH and PRL were quantified. Following treatment on d 10, suckling lambs were removed from 6 of the 12 ewes, creating non-suckled (NS) and suckled (S) treatment groups for subsequent study on d 14 through 26. On d 10, NAL treatment increased LH (P less than .01) but concentrations of PRL were not affected. When averaged across d 14 to 26, post-NAL concentrations of LH were greater (P less than .001) than pre-NAL concentrations (6.5 +/- .7 vs 1.9 +/- .4 ng/ml). In contrast, concentrations of PRL in the post-NAL period were lower (P less than .001) than pre-NAL concentrations (129 +/- 15 vs 89 +/- 10 ng/ml). Compared with S ewes over d 14 to 26, those in the NS group had similar pre-NAL concentrations of LH, tendencies for higher (P less than .10) post-NAL concentrations of LH, lower (P less than .001) mean serum concentrations of PRL (pre- and post-NAL) and similar pre-NAL vs post-NAL differences in serum PRL. Six suckled beef cows on d 24 to 35 were injected iv with either saline or NAL (.5 mg/kg) in a replicated crossover design. Injections of NAL increased serum concentrations of LH (P less than .05), when averaged over all 12 injections in the six cows, but serum PRL was not changed. However, three of six cows did not respond to NAL with increases in serum LH. These non-responding cows were similar to the responding cows in their pre-injection concentrations of LH and PRL, but they tended (P = .10) to have higher serum concentrations of cortisol than responding cows.  相似文献

Multiparous Angus and crossbred Angus cows were used to determine the effect of induced endometritis on plasma concentrations of 13,14-dihydro-15-keto-prostaglandin F2 alpha (PGFM) and progesterone (P4) and on duration of the estrous cycle of treatment. Beginning on the day of calving (d 0), blood samples were collected on alternate days. On three consecutive days, ranging from d 8 to 14 of the first postpartum estrous cycle, uterine horns were inoculated transcervically with either 3 x 10(9) colony forming units (cfu) of Actinomyces pyogenes and 1.5 x 10(9) cfu of beta-hemolytic Escherichia coli (treated; n = 9) in sterile PBS or with sterile PBS alone (control; n = 9). Samples of uterine fluid were collected by transcervical aspiration twice weekly from just before the start of each series of inoculations until the end of the experiment. Endometrial biopsies were collected transcervically between d 4 to 6 and 11 to 13 after inoculation. Based on clinical observations and results of bacterial cultures, all treated cows developed acute uterine infections. Controls did not develop uterine infections. Endometrial biopsies indicated that there were no significant diffuse or focal cellular reactions in response to the infection. The interestrous interval was greater (P less than .0003) for treated (27.7 +/- 1.0 d) than for control (20.6 +/- 1.0 d) cows, but P4 concentrations were similar between the two groups. Mean PGFM concentration and PGFM profiles were similar (P greater than .10) between treated and control cows before bacterial infusions. The purpose of this study was to evaluate the influence of animal, breed and parity on postpartum reproductive functions in dairy cows. A total of 141 cows were included in the experiment, which was carried out as part of a study on traits affecting longevity in Swedish dairy cows. The cows belonged to 4 different breed-groups and were 1st to 5th calvers. The duration of the study was 3 years and 44 cows were followed during 1 postpartum period, 49 cows during 2 consecutive periods, 43 cows during 3 periods and 5 cows during 4 periods.The cows were clinically examined, by rectal palpation, at 10-day intervals between calving and first AI, which was at first normal oestrus more than 50 days after calving. External signs of heat were checker and recorded three times daily by the herdsmen. Blood samples for progesterone assay were taken at days 10, 15 and 20 after calving and thereafter every 10th day until first AI. Samples taken at days 10, 15 and 20 were also assayed for content of 15-keto-13,14-dihydro-PGF.Heat detection records, records from clinical examinations and plasma progesterone assays were chronologically compiled for each postpartum period and based on this, intervals between calving and postpartum ovulations, recorded uterine involution, 1st and subsequent oestrus and regular reproductive functions were estimated. Least-squares methods were used for the statistical evaluation of data.The results indicate a large variation within and between cows in postpartum reproductive performance. In the total material 1st ovulation occurred before recorded uterine involution and there was a close relationship between 1st ovulatory oestrus and the onset of regular reproductive functions. The interval between calving and 1st ovulation significantly influenced the length of the first cycle in the sense that a large proportion of the early ovulating cows had a short interval between 1st and 2nd postpartum ovulations. The large variations were also evident in the plasma levels of 15-keto-13,14-dihydro-PGF. There was a marked decline between days 10 and 15 postpartum and most cows were close to basal levels at 20 days postpartum.The individual cow had a significant influence on intervals from calving to recorded uterine involution, 1st ovulatory oestrus, regular reproductive functions and conception. The breed influence was significant for intervals between calving and 1st ovulation and recorded uterine involution whereas the parity of the cow only influenced the interval between calving and recorded uterine involution.  相似文献

The objective of this study was to determine whether an antiestrogen (enclomiphene) would shorten the interval to first estrus and conception in postpartum beef cows. Sixty postpartum Angus beef cows were stratified by age, body condition, and calving date and were randomly assigned to one of two treatment groups. Group 1 cows (n = 24) received three silastic implants, each containing 150 mg of enclomiphene, on d 20 postpartum. Implants were removed on d 30 postpartum. Group 2 cows (n = 28), received empty implants and served as controls. Cows were artificially inseminated at first detected estrus. Estrus detection and ovulation were further verified by increased serum progesterone. Concentrations and pulse frequencies of LH were determined from blood samples collected at 15-min intervals for 6 h on d 20, 25, 30, and 40 postpartum. Hypothalami and pituitaries were collected from four cows in each treatment group on d 30 postpartum and analyzed for concentrations of estradiol receptors. Concentrations of total and unoccupied hypothalamic and pituitary estradiol receptors were reduced by enclomiphene. Neither concentrations nor pulse frequencies of LH differed significantly between treatment groups on any of the 4 d. Days to first estrus did not differ (P greater than .05) between enclomiphene-treated (57 +/- 6; n = 24) and control (56 +/- 4; n = 28) cows. Days to conception did not differ between treated (81 +/- 9) and control (79 +/- 8) cows. The dose of enclomiphene used in this study reduced hypothalamic and pituitary estrogen receptors but did not alter secretion of LH or days to first estrus in the postpartum beef cow.  相似文献

Cortisol and luteinizing hormone (LH) were measured in serum after the administration of adrenocorticotropic hormone (ACTH) to suckled (S) and nonsuckled (NS) beef cows. Blood was sampled on 2 consecutive days every 2 weeks for four bleeding periods starting 14 days after calving. Cows were injected with 200 IU ACTH or saline in a 2-day switchback design. Serum was collected before ACTH or saline injection and at 30-min intervals thereafter for 8 hours. Average cortisol concentrations in serum were similar in S and NS cows (6.4 +/- .6 and 6.1 +/- .8 ng/ml, respectively) after saline. Average cortisol concentrations in serum collected during an 8-hr period after ACTH on days 14, 28, 42 and 56 postpartum were 24.7 +/- 2.4, 31.8 +/- 3.5, 36.4 +/- 4.2 and 40.7 +/- .5 ng/ml, respectively, for S cows, and 31.1 +/- 2.9, 44.7 +/- 5.2, 45.0 +/- 5.7 and 46.0 +/- 5.4 ng/ml, respectively, for NS cows. Cortisol response to ACTH, measured as area under the response curve, was greater (P less than .05) in NS than in S cows. Amount of cortisol released by 200 IU ACTH was maximal by days 28 to 29 postpartum in NS cows, but the response increased gradually between days 14 to 15 and days 56 to 57 in S cows. overall, LH in serum averaged .55 +/- .08 ng/ml for S cows and .92 +/- .06 ng/ml for NS cows after saline, and .49 +/- .07 ng/ml for S cows and .94 +/- .06 ng/ml for NS cows after ACth. Fourteen anovulatory postpartum (38.0 +/- 1.9 d) beef cows that ovulated after an injection of 250 micrograms gonadotropin releasing hormone (GnRH) in saline were used to examine the influence of indomethacin on luteal function. Beginning the d after GnRH, 6 cows were given intrauterine infusions of indomethacin for 14 d and the other eight cows received vehicle. After GnRH treatment, concentrations of progesterone in serum were elevated longer (P less than .01) for indometacin-treated cows than for vehicle-treated cows. At the same time prostaglandin metabolite (PGFM) concentrations were lower (P less than .01) in indomethacin-treated cows than in vehicle-treated cows. In summary, indomethacin suppressed PGFM concentrations and enhanced function of corpora lutea induced in postpartum suckled beef cows.  相似文献

This study was designed to determine whether an anti-estrogen can block the negative effect of estrogens on luteinizing hormone (LH) release and therefore decrease the postpartum interval in suckled beef cows. In Exp. I, eight suckled postpartum beef cows were randomly assigned to treatment and control groups. Treatment cows received 1 g/d clomiphene citrate (im) from d 21 to 28 postpartum, while control cows were injected with saline. On d 28 postpartum, there was no difference (P greater than .05) in mean total and basal LH concentrations or LH pulse frequency between treatment and control cows. All control cows exhibited estrus on d 52 +/- 3; treatment cows exhibited estrus on d 134 +/- 12 (P less than .05). In Exp. II, 17 suckled cows were randomly assigned to three treatment groups: 1) control group (n = 6) receiving one empty implant, 2) 10-cm enclomiphene implant group (n = 5) and 3) 30-cm enclomiphene implant group (n = 6). The silastic implants were placed sc on d 20 and removed on d 29 postpartum. Mean total LH concentrations during d 24 to 29 postpartum in the 30-cm enclomiphene implant group were higher than the 10-cm implant (P less than .05) and control group (P less than .05). Thirteen multiparous (M; 451 kg, 6.1 body condition score) and 11 primiparous (P) Brahman cows (408 kg, 6.3 body condition score) were assigned randomly within parity to receive either 2-min uterine manipulation (UM) per rectum 35 d after calving or no UM (C). This resulted in four groups: MUM (n = 8), MC (n = 5), PUM (n = 5) and PC (n = 6). All animals received a jugular cannula on d 34. Blood samples were collected at 10-min intervals from 30 min prior to UM until 120 min after UM and at 20-min intervals through 300 min after UM. Plasma was harvested immediately and stored at -20 degrees C until RIA for 13, 14-dihydro-15 keto prostaglandin F2 alpha (PGFM). Number of PGFM peaks in 330 min and amplitude of these peaks were similar (P greater than .10) in MC (2.0 peaks, 445.6 pg/ml) and PC (1.8 peaks, 446.9 pg/ml). Response to UM differed (P less than .02) by parity. The PGFM response of UM and C primiparous cows did not differ. Mean PGFM concentrations between 70 and 300 min after UM were greater (P less than .05) in MUM than in MC cows. Area of the PGFM curve did not differ (P greater than .10) in primiparous groups but was greater (P less than .01) in MUM than in MC cows. Uterine manipulation 35 d after parturition increased plasma PGFM in M but not in P cows.  相似文献

Twenty-eight Hereford x Angus cows (4 yr of age) were used to determine the effects of pre- and postpartum dietary energy on performance and reproductive function in suckled beef cows. The experiment was designed as a 2 x 2 factorial with cows receiving either 70 (L) or 150% (H) of NRC recommended level of dietary energy before and(or) after parturition, resulting in four treatment combinations (L-L, L-H, H-L, H-H). Prepartum diets were fed for approximately 110 d, and postpartum diets were fed until either 10 d after the second postpartum ovulation or 150 d postpartum for those cows that failed to ovulate. Cows receiving low compared with high levels of energy before calving lost more (P less than .01) weight, body condition, subcutaneous fat, and longissimus muscle area before parturition and had calves with lighter (P less than .01) birth weights. Cows receiving low compared with high levels of energy postpartum lost more (P less than .01) weight, body condition, and longissimus muscle area after parturition. Low levels of energy before and after parturition decreased (P less than .01) milk production and calf weight at 70 d of age. Rates of cervical and uterine involution were unaffected by dietary energy treatments. Cows receiving high levels of energy prepartum had increased (P less than .01) mean concentrations and pulse frequency of LH in serum after parturition, and cows receiving high levels of energy postpartum had increased (P less than .05) pulse frequency of LH. Low levels of energy postpartum decreased (P less than .01) appearance rate of small (5.0 to 7.9 mm) and large (greater than or equal to 10 mm) follicles, and low levels of energy prepartum decreased (P less than .02) appearance rate of large follicles based on transrectal ultrasonography. Cows receiving high levels of energy prepartum had shorter (P less than .02) intervals from parturition to ovulation, and a higher (P less than .01) percentage of the cows that received high levels of energy postpartum ovulated by 150 d postpartum. In summary, prepartum level of dietary energy influenced birth weight and weight gain of calves, milk production, concentrations and pulse frequency of LH in serum, appearance rate of large follicles, and the interval to first ovulation. Postpartum level of dietary energy influenced milk production, weight gain of calves, pulse frequency of LH, appearance rate of small and large follicles, and the percentage of cows that ovulated after parturition.  相似文献
