以愈创木酚、α-萘酚、没食子酸、L-酪氨酸和单宁酸等5种酚类物质为底物,采用鉴别培养基筛选法从14株茶树内生真菌中初筛获得4株产多酚氧化酶的真菌。根据变色圈的大小、颜色深浅和摇瓶发酵的结果复筛获得产多酚氧化酶能力较强的菌株CSN-13。对菌株CSN-13产酶营养条件进行初步分析,结果表明,在供试的6种碳源物质中,以麸皮对菌株CSN-13产多酚氧化酶的促进作用最为明显;供试的5种氮源物质中,以硝酸铵的促进作用最为明显;在发酵培养基中添加茶水,对产酶有明显的促进作用。采用正交设计对CSN-13产酶发酵培养基进行初步优化,优化后的培养基配方为:麸皮(40βg·L-1)、硝酸铵(15βg·L-1)、茶水(4βg·L-1)、KH2PO4(2βg·L-1)、MgSO4·7H2O(0.5βg·L-1)、无水CaCl2(0.075βg·L-1)、CuSO4·5H2O(0.01βg·L-1)。采用优化后的培养基,菌株CSN-13在28℃下培养5βd,酶活力达到241βU·mL-1·min-1,比优化前提高8.5倍。茶树内生真菌菌株CSN-13及其发酵产酶培养基的研究为多酚氧化酶的进一步开发打下了基础。  相似文献

观察普洱茶水提物对硝苯地平降压效果的影响。采用雄性SHR大鼠50只,按照平均尾动脉压随机分为模型组(M组)、硝苯地平3.1βmg·kg-1组(1/2临床剂量)(N.L组)、硝苯地平6.2βmg·kg-1组(临床剂量)(N.H组)、硝苯地平3.1βmg·kg-1+普洱茶水提物0.5βg·kg-1组(N.L+D.L组)、硝苯地平3.1βmg/kg+普洱茶水提物1.0βg·kg-1组(N.L+D.H组),每组10只,另设10只动物作为正常对照组(C组)。分组次日灌胃给予相应药物,每天2次,上午给予硝苯地平,下午给予普洱茶水提物,灌胃间隔3~4βh,每周对各组动物血压进行检测,持续4周。结果表明,大鼠给药4周后,硝苯地平6.2βmg·kg-1组大鼠尾动脉平均血压为(139±6)βmmHg(1βmmHg=0.133βkPa),与M组的(157±13)βmmHg比较显著降低(P<0.05);N.L+D.H组大鼠尾动脉平均血压为(136±11)βmmHg,与M组的(157±13)βmmHg比较显著降低(P<0.05),与硝苯地平6.2βmg·kg-1组的(139±6)βmmHg相比,降压效果相当(P>0.05)。研究表明,普洱茶水提物配合硝苯地平使用,可增强硝苯地平对SHR大鼠的降压功效,其降压效果与临床剂量硝苯地平单独使用相当,且对心脏具有一定程度的保护作用。  相似文献

为探索EGCG对Nicotine诱导肺癌细胞增殖的抑制作用,本研究通过MTT实验筛选出EGCG、Nicotine对肺腺癌细胞H1299的最佳作用浓度,利用实时荧光定量PCR技术检测EGCG、Nicotine对H1299细胞中Bax、Bcl-2、Jak2和Stat3基因mRNA相对表达量的变化。实验结果表明,EGCG对H1299细胞的半抑制浓度IC50值约为32βμmol·L-1(24βh)、15βμmol·L-1(48βh);1βμmol·L-1的Nicotine对H1299细胞促增殖作用明显;以15βμmol·L-1的EGCG预处理H1299细胞24βh可显著下调1βμmol·L-1 Nicotine的促增殖作用(P<0.05)。1βμmol·L-1的Nicotine处理H1299细胞可明显降低JAK2/STAT3信号通路中Bax基因mRNA的表达,增加Bcl-2、Jak2、Stat3基因的mRNA表达;15βμmol·L-1 EGCG预处理H1299细胞可反向调控Nicotine诱导所致JAK2/STAT3信号通路中Jak2、Stat3和Bax、Bcl-2基因表达量的变化,结果具有显著性差异(P<0.05)。由此可知,EGCG对Nicotine诱导的肺腺癌H1299细胞增殖及JAK2/STAT3信号通路中促增殖基因mRNA的表达起抑制作用。  相似文献

采用超高效液相色谱-静电场轨道阱高分辨质谱,建立了茶叶中21种农药残留量的检测方法。茶叶样品经甲醇和水(V甲醇V=1∶1)提取后,采用乙二胺-N-丙基硅烷和强阳离子交换剂作为混合吸附剂进行萃取净化。以C18色谱柱进行色谱分离,采用Full Scan扫描模式进行定性、定量分析。结果显示,21种农药在0.5~200βμg·L-1范围内均呈良好的线性关系,相关系数(r2)大于0.99。在10、50、100βμg·kg-1 3个加标水平下,平均回收率为70%~125%之间,相对标准偏差(RSD)小于15%,定量限为10βμg·kg-1。本方法操作简单快速,灵敏度高,适用于茶叶中21种农药残留检测。  相似文献

简单序列重复(Simple sequence repeats, SSR)在茶树遗传与育种研究中具有重要的作用,基于4300 DNA分析系统的SSR发掘具有通量高、准确和灵敏等特点,已被用于许多物种的分子标记研究,但在茶树的相关研究中尚未见报道。本研究应用单因素试验和L9(34)正交设计试验对影响茶树SSR-PCR的主要参数进行优化,建立了适合于4300 DNA分析系统的茶树SSR-PCR反应体系:1.0 μL DNA(25 ng·μL-1),0.2 μL M13F-F、0.2 μL R和0.4 μL IR-M13F,0.8 μL dNTPs(25 mmol·L-1),1.0 μL 10×Buffer(含Mg2+),0.1 μL Ex-Taq聚合酶(5 U·μL-1),无菌水定容至10 μL。所有引物浓度均为1 μmol·L-1。同时,本研究还证明,可以以自行配制的6.5%聚丙烯酰胺凝胶溶液(acry:bis=29:1)替代4300 DNA分析系统指定凝胶溶液,检测SSR位点。  相似文献

茶树发根在LG0培养基和添加1βmg/L IBA的1/2MS培养基上生长迅速,但只在一个培养于LG0培养基的发根克隆中检测到高含量的茶氨酸,含量达到23.12βmg/g鲜重,高于未转化根的18.77βmg/g鲜重。除茶氨酸外,该发根克隆中也检测到高含量的天门冬氨酸、谷氨酸、天门冬酰胺、谷氨酰胺和精氨酸。发根克隆培养于添加1βmg/L IBA的1/2MS培养基,茶氨酸的含量则显著降低,说明培养基组成也是影响发根中茶氨酸的含量的因素之一。茶树发根中也含有少量的儿茶素类单体,主要为儿茶素和表儿茶素。  相似文献

为了抑制冠突散囊菌发酵绿茶液态饮料中儿茶素的降解,本研究采用植物乳杆菌和冠突散囊菌对绿茶液态饮料进行联合发酵,试验通过响应面法优化了联合发酵绿茶液态饮料工艺,并采用高效液相色谱(HPLC)法和顶空固相微萃取串联气质联用(HS-SPME/GS-MS)法分别检测了联合发酵绿茶液态饮料中儿茶素含量和香气成分。结果表明,在干茶添加量10βg·L-1、冠突散囊菌添加量10βmL·L-1的前提下,联合发酵绿茶液态饮料的最佳工艺条件为植物乳杆菌添加量20βmL·L-1、蔗糖添加量75βg·L-1、30℃下静置发酵3βd。在此工艺下联合发酵绿茶液态饮料中总儿茶素含量为1β419.94βμg·mL-1,与冠突散囊菌发酵绿茶液态饮料(848.72βμg·mL-1)相比,显著增加(P<0.05);且醇类化合物(30.27%)、醛类化合物(15.25%)、烃类化合物(11.35%)、酯类化合物(9.86%)和酮类化合物(9.01%)含量与未发酵绿茶液态饮料相比均显著增加(P<0.05)。  相似文献

以闽中某县8个代表性茶园为研究对象,采集茶园土壤和茶树各器官样品,分析镉在茶园土壤-茶树中积累和分布规律,并探讨其受土壤理化性质的影响;同时测定茶汤中镉含量并算出茶叶中镉的溶出率,利用美国国家环保署(USEPA)推荐的健康风险评价模型进行人体致癌健康风险评价。结果表明,茶园土壤全镉含量均值为112.74βμg·kg-1,是福建省土壤背景值的2.06倍,茶园土壤镉积累明显;茶园土壤有效镉含量均值为26.44βμg·kg-1,镉活化率均值为24.86%,有效程度较高。土壤pH和有机质是影响土壤镉及其有效性的主要因素,土壤全磷和速效磷是影响镉活化率的主要因子;茶树主根和侧根与土壤全镉、有效镉和有机质呈显著正相关(P<0.05),新叶镉含量与土壤有效镉和全磷呈显著正相关(P<0.05)。茶树各器官镉含量分布规律为:侧根(1β253.89βμg·kg-1)>主根(382.20βμg·kg-1)>主茎(167.25βμg·kg-1)≈侧茎(154.65βμg·kg-1)>老叶(30.60βμg·kg-1)≈新叶(27.13βμg·kg-1),茶树根部镉富集系数显著大于其他器官(P<0.05),新叶和老叶镉富集系数较低,镉大部分被根和茎固定,向叶的迁移能力较低。茶汤中镉含量均值为192.28βng·L-1,远低于《生活饮用水卫生标准》中镉含量,茶叶中镉溶出率均值为15.29%;茶汤和茶叶中镉致癌健康年风险分别为6.33×10-7和4.42×10-6,比国际辐射防护委员会推荐的化学有害物最大可接受水平(5×10-5)低约1~2个数量级,说明可以安全饮用。  相似文献

采用色差计、MDA、T-AOC和沉积量的研究方法,测定了鸭腿肉在不同儿茶素处理在贮藏期的色差光学指标、MDA、T-AOC和儿茶素沉积量的变化。其结果表明:添加儿茶素的处理组会明显改变L*光学指标,儿茶素浓度与L*、a*、b*光学指标分别呈现不同的相关性,以添加500βmg/kg的EGCg和复合儿茶素的效果为佳;降低了MDA值,提高了T-AOC,EGCg和复合儿茶素分别以800βmg/kg、1β000βmg/kg添加量和500βmg/kg、1β000βmg/kg添加量为佳,与儿茶素在鸭腿肉中的沉积有很明显的量效关系,上述指标的改善均以复合儿茶素优于EGCg。  相似文献

安全性毒理学评价是研究食品安全的重要手段。本文综述了与普洱茶有关的安全性毒理学评价研究,为认识普洱茶安全性提供参考。急性经口毒性实验显示普洱茶的半致死率(LD50)大于5β000βmg·kg-1,属于实际无毒级别;28βd或90βd经口毒性试验发现普洱生、熟茶提取物无明显不良效应的剂量分别是1β250和5β000βmg·kg-1·d-1;遗传毒性研究表明普洱茶对原核细胞、真核细胞和生殖细胞均无致突变性;普洱茶的生殖毒性和发育毒性无效应剂量水平是700βmg·kg-1·d-1;普洱茶对小鼠肝细胞的毒性极小或无;人体急性、亚急性毒性研究均未观察到病理改变。综上,已有安全性毒理学评价研究显示普洱茶具有较高的饮用安全性。  相似文献

The absorption, distribution and elimination of carnosic acid, the main antioxidant found in rosemary was studied, in vivo, in rats. Therefore, carnosic acid was administrated in a single dose, intravenously (20.5 ± 4.2 mg/kg) and orally (64.3 ± 5.8 mg/kg), to four and nine rats, respectively. Blood samples were collected at different time points, and plasma concentrations of carnosic acid were determined using LC-MS. Furthermore, total collection of urine and feces was done during 4 h and 24 h for the intravenous and oral administrations, respectively. After euthanizing the rats, intestinal content, liver and muscle tissue were sampled to determine carnosic acid concentrations. The bioavailability of carnosic acid, after 360 min, was 40.1%. Traces of carnosic acid were found in the rats intestinal content, liver and muscle tissue of abdomen and legs. The recovery of carnosic acid in the feces, 24 h after oral administration, was 15.6 ± 8.2%. Carnosic acid is absorbed into the bloodstream after oral administration in rats and is therefore bioavailable. It was found that carnosic acid in vivo is present in its free form and that its main elimination route is the fecal route.  相似文献

研究了一种茶叶复合物的解酒作用。通过观察小鼠灌胃白酒后翻正反射消失与恢复时间,急性乙醇中毒的死亡率,自发活动的变化以及小鼠血液中乙醇浓度,以评价茶叶复合物缓解醉酒症状的作用。结果显示:低、中、高剂量茶叶复合物均能延长翻正反射消失时间,缩短小鼠翻正反射恢复时间(P<0.01);降低急性乙醇中毒小鼠的死亡率;抑制乙醇致中枢兴奋引起的小鼠自主活动次数增多(P<0.01);高剂量茶叶复合物能显著降低灌胃乙醇后30、60、90、120、150min小鼠血液中的乙醇浓度(P<0.05或P<0.01),各时间点小鼠血浆乙醇质量浓度分别为(523±129)、(671±175)、(954±203)、(1012±326)、(634±139)mg/L。本研究为茶叶解酒产品的开发提供了一定的理论基础。  相似文献

Curcumin, the active constituent of Curcuma longa L. (family Zingiberaceae), has gained increasing interest because of its anti-cancer, anti-inflammatory, anti-diabetic, and anti-rheumatic properties associated with good tolerability and safety up to very high doses of 12 g. Nanoscaled micellar formulations on the base of Tween 80 represent a promising strategy to overcome its low oral bioavailability. We therefore aimed to investigate the uptake and transepithelial transport of native curcumin (CUR) vs. a nanoscaled micellar formulation (Sol-CUR) in a Caco-2 cell model. Sol-CUR afforded a higher flux than CUR (39.23 vs. 4.98 μg min?1 cm?2, respectively). This resulted in a higher Papp value of 2.11 × 10?6 cm/s for Sol-CUR compared to a Papp value of 0.56 × 10?6 cm/s for CUR. Accordingly a nearly 9.5 fold higher amount of curcumin was detected on the basolateral side at the end of the transport experiments after 180 min with Sol-CUR compared to CUR. The determined 3.8-fold improvement in the permeability of curcumin is in agreement with an up to 185-fold increase in the AUC of curcumin observed in humans following the oral administration of the nanoscaled micellar formulation compared to native curcumin. The present study demonstrates that the enhanced oral bioavailability of micellar curcumin formulations is likely a result of enhanced absorption into and increased transport through small intestinal epithelial cells.  相似文献

Folate deficiency is a global health problem related to neural tube defects, cardiovascular disease, dementia, and cancer. Considering that folic acid (FA) supply through industrialized foods is the most successful intervention, limitations exist for its complete implementation worldwide. Biofortification of plant foods, on the other hand, could be implemented in poor areas as a complementary alternative. A biofortified tomato fruit that accumulates high levels of folates was previously developed. In this study, we evaluated short-term folate bioavailability in rats infused with this folate-biofortified fruit. Fruit from tomato segregants hyperaccumulated folates during an extended ripening period, ultimately containing 3.7-fold the recommended dietary allowance in a 100-g portion. Folate-depleted Wistar rats separated in three groups received a single dose of 1 nmol of folate/g body weight in the form of lyophilized biofortified tomato fruit, FA, or synthetic 5-CH3-THF. Folate bioavailability from the biofortified tomato was comparable to that of synthetic 5-CH3-THF, with areas under the curve (AUC0–∞) of 2,080?±?420 and 2,700?±?220 pmol?·?h/mL, respectively (P?=?0.12). Whereas, FA was less bioavailable with an AUC0–∞of 750?±?10 pmol?·?h/mL. Fruit-supplemented animals reached maximum levels of circulating folate in plasma at 2 h after administration with a subsequent steady decline, while animals treated with FA and synthetic 5-CH3-THF reached maximum levels at 1 h. Pharmacokinetic parameters revealed that biofortified tomato had slower intestinal absorption than synthetic folate forms. This is the first study that demonstrates the bioavailability of folates from a biofortified plant food, showing its potential to improve folate deficiency.  相似文献

应用诊断施肥综合法(DRIS)对低产园和高产园槟榔进行了叶片营养诊断。结果表明,相对于高产园,低产园元素间关系较不平衡。槟榔叶片N、P、K、Ca、Mg以及Fe、Mn、Cu、Zn的适宜含量分别为(20.53±0.67)、(1.90±0.01)、(13.30±0.46)、(7.14±0.57)、(3.68±0.41)g/kg及(109.50±4.16)、(112.50±9.02)、(6.07±0.64)、(31.23±0.15)mg/kg。低产园N、P、K、Ca、Mg、Fe、Mn、Cu、Zn元素DRIS诊断指数分别为-44.58、-1.60、-17.16、0.09、-31.94、52.78、31.03、23.12、-11.74,需求强度较大的元素为N、Mg、K,其次是Zn、P。  相似文献

Small fruits like bilberry, blackberry and mulberry are rich sources of anthocyanins and other phenols, compounds with a certified antioxidant activity and spectacular effects in some chronic diseases. Romanian bilberry, blackberry and mulberry extracts were tested as anti-hyperglycemic agents on diabetic rats. Anthocyanins extraction was carried out with 80 % acidified ethanol in ultrasonically conditions at 23?±?2 °C and 40 kHz. Monomeric anthocyanins content was determined by pH differential method and varied between 1200 and 2800 mg/L. The analyses of anthocyanins were achieved using high performance liquid chromatography and mass spectrometry. Phenolics content was determined by Folin-Ciocalteu procedure and values varied between 2320 and 4250 mg/L gallic acid. Antioxidant activities of extracts were estimated by DPPH scavenging method and the values varied between 8 and 16 miliequivalents Trolox. In order to evaluate the toxicology of the extracts, the heavy metals concentration and pesticides content were analyzed. The extracts were administrated to diabetic rats in drinking water for five weeks. The administration of bilberry extract offered no satisfactory results. Treatment with blackberry extract determined a significant decrease of glucose level from 360 to about 270 mg/dL (p?<?0.05). The mulberry extract administration determined a significant decrease of glucose level from 252 mg/dL at the start day to 155 mg/dL at the final of experiment (p?<?0.05).  相似文献

The aim of this research was to evaluate the antioxidant capacity and physical-chemical characteristics of commercial white myrtle berry (Myrtus communis L. var. leucocarpa DC) liqueur (WMBL). The total phenolic (TP) content was measured spectrophotometrically, applying a modified Folin-Ciocalteu’s method, and phenolic compounds were identified by high-performance liquid chromatography (HPLC) coupled with electrospray mass spectrometry, and quantified by HPLC coupled with ultraviolet/visible detection. The antioxidant capacities were evaluated by FRAP, CUPRAC, DPPH?, and ABTS?+ assays. The volatiles were assessed by gas chromatography and mass spectrometry (GC-MS/FID) after headspace solid-phase microextraction (HS-SPME) and liquid-liquid extraction (LLE). WMBL showed lower TP levels (636.3 ± 39.2 mg GAE/L) than in purple myrtle berry liqueur (PMBL). Nevertheless, WMBL exhibited better antioxidant capacities, potentially due to high concentrations of gallic acid (294.2 ± 14.2 mg/L) and its derivatives (58.3 ± 2.1 mg/L). Other phenolic compounds detected by HPLC-DAD and LC-MS/MS were flavonols like myricetin and its derivatives (myricetin-3-O-galactoside and myricetin-3-O-rhamnoside) with concentrations similar to those found in PMBL. GC-MS/FID analysis revealed 44 compounds (terpenes, higher aliphatic compounds and shikimic acid pathway derivatives). 1,8-Cineole was the most abundant terpene in the liqueur (26.5% (HS-SPME) and 9.6% (LLE)).  相似文献

Pollen protoplasts provide a sexual and haploid system for haploid production, cell fusion and mutation studies used in plant improvement. Due to the multiploidy, heterozygosity, and often self-incompatibility in tetraploid genotypes, haploid potatoes are desirable for breeding schemes via ploidy manipulations. In this study, two tetraploid varieties and two dihaploid lines of potato were used for pollen tetrad protoplast isolation and culture. The meiotic tetrad buds were first pre-treated at 5 °C for 0–12 days, then the tetrads were transferred into enzyme solutions containing different concentrations of snailase (0.5–1.5%), 0.3 M osmolites (sucrose, mannitol, glucose or sorbitol), 1.0% Cellulose, 0.5% Hemicellulase, 0.5% Pectolyase, 0.3% Sucrose, 3 mM 2-(N-Morpholino) ethane sulfonic acid, 1% polyvinyl pyrrolidone, 0.01% casein hydrolysate and K3 medium compositions. Among the four donor materials, tetraploid cv. Gannongshu No. 3 (‘GNS No.3’) showed the greatest protoplast yield (74.6 ± 2.4%). In this variety, most of the tetrad protoplasts regenerated a cell wall and continued cell divisions were observed when they were inoculated in K3 basic medium supplemented with (0.5–1.0) mg/L 2,4-D + (0.1–0.5) mg/L KT + 0.4 mg/L 6-BA +800 mg/L glutamine +100 mg/L serine. ‘GNS No.3’ also showed the greatest first division frequency (21.6 ± 1.5%) and sustained division to form multicellular structures. The study findings suggested that cultured tetrad pollen protoplasts could reverse the gametophytic developmental pattern programmed in vivo to a sporophytic pathway leading to multicellular microspore-derived colonies.  相似文献

The bioavailability and interactive effects of calcium, iron and zinc fortified whole wheat flour chapattis on female Sprague–Dawley albino rats were determined. The whole wheat flour was fortified with CaCO3, FeSO4 and ZnSO4 at 1000, 40 and 20 mg/kg, respectively and fed to 64 female Sprague–Dawley rats for 4 weeks. Retention of Ca, Fe and Zn was determined in plasma, femur and liver tissues. Ca levels in plasma and liver did not alter significantly while the Ca retention in femur changed significantly with the change in fortificants. The calcium retention was higher in the femur of rats fed on the diet supplemented with Ca + Zn (221 ± 13.61 mg/g) than the rats fed on the diet supplemented with Ca + Fe (219 ± 18.92 mg/g). The iron in plasma, liver and femur ranged from 2.19 ± 0.11 to 2.77 ± 0.10 μg/ml, 203.5 ± 13.24 to 251.5 ± 17.19 μg/g and 43.5 ± 1.05 to 52 ± 2.07 μg/g while zinc content ranged from 1.31 ± 0.089 to 1.55 ± 0.114 μg/ml, 89.5 ± 3.41 to 132 ± 6.10 μg/g and 175 ± 8.29 to 191 ± 10.85 μg/g, respectively. The minerals interacted with each other resulting in decrease of bioavailability among each other. The absorption was higher in the rats fed on fortified diet than those fed on unfortified chapatti diet. Hence, it is concluded that fortification of whole wheat flour is feasible to overcome the Ca, Fe and Zn malnutrition in the vulnerable groups.  相似文献
