The aim of the present study was to determine the effects of dietary supplements of vitamins on vitamin status, libido, and semen characteristics in young boars under normal and intensive semen collection. Sixty Landrace, Yorkshire, and Duroc boars were allocated randomly from 6 to 10 mo of age to one of the following diets: 1) basal diet (industry level) for minerals and vitamins (Control, n = 15); 2) basal diet supplemented with vitamin C (ASC, n = 15); 3) basal diet supplemented with fat-soluble vitamins (FSV, n = 15); and 4) basal diet supplemented with water-soluble vitamins (WSV, n = 15). After puberty (approximately 12 mo of age), semen was collected at a regular frequency (three times every 2 wk) for 5 wk. Thereafter, all boars were intensively collected (daily during 2 wk). A recovery period (semen collection three times every 2 wk) followed and lasted for 10 wk. Sperm quality (percentage of motile cells and percentage of morphologically normal cells) and quantity (sperm concentration, semen volume, and total sperm number) were recorded as well as direct and hormone related measurements of boar libido. Blood and seminal plasma samples were taken to monitor vitamin status. High concentrations of B6 (P < 0.05) and folic acid (P < 0.05) were observed in the blood plasma of WSV boars, whereas greater concentrations of vitamin E (P < 0.01) were obtained in FSV boars. In the seminal plasma, folic acid concentrations tended to be greater in WSV boars (P < 0.08). During the intensive collection period, there was a tendency (P < 0.06) for semen production to be greater in WSV boars, the effect being less pronounced (P < 0.10) in FSV boars. During the recovery period, the percentage of motile sperm cells was greater in WSV boars (P < 0.03) and, to a lesser extent, in FSV boars (P < 0.10) compared with Control boars. Sperm morphology and libido were not affected by treatments. These results indicate that the transfer of vitamins from blood to seminal plasma is limited and the dietary supplements of water-soluble and fat-soluble vitamins may increase semen production during intensive semen collection.  相似文献

In swine artificial insemination, several dose regimens are applied, ranging from 1.5 x 10(9) to 6.0 x 10(9) spermatozoa per intra-cervical insemination dose. A lower sperm dose is more profitable for artificial insemination centres and offers a more effective use of superior boars. To evaluate fertility, 50 boars were used for a total of 10 773 homospermic first inseminations at a dose of 2 billion spermatozoa. In addition, 96 boars were used at a dose of 3 billion spermatozoa for 34 789 homospermic first inseminations. Fertility was determined by a 60-day non-return rate (NR%) of first inseminations. Litter size was registered by total number of piglets born separately in primiparous and multiparous farrowings. On average, a sow was inseminated 1.5 times. A significant decrease was observed in all three fertility parameters (NR%, litter size of both primiparous and multiparous farrowings) with a dose of 2 billion spermatozoa compared with a dose of 3 billion spermatozoa. The NR% was 75.8% and 84.0% (p < 0.001), the mean litter size of primiparous farrowings 10.1 and 10.7 (p < 0.001) and the mean litter size of multiparous farrowings 11.7 and 12.1 (p < 0.001) for 2 and 3 billion spermatozoa/dose, respectively. The proportion of normal spermatozoa in the sperm morphology analysis correlated significantly with NR% in both insemination regimens: p < 0.001, r = 0.604 and p < 0.05, r = 0.223 for 2 and 3 billion spermatozoa/dose, respectively. These results confirm that quantity can at least partly compensate for poor sperm quality. When the boars with <70% normal spermatozoa in the morphology evaluation were excluded from the data there were no correlation between the sperm morphology and NR%. However, the difference between the NR% and litter size remained statistically significant (p < 0.001) in favour for the bigger insemination dose. In conclusion, a decrease in sperm dose from 3 to 2 billion spermatozoa on commercial farms will severely decrease prolificacy at least under field conditions, where a sow is inseminated an average of 1.5 times/heat, and the semen is typically used within 3 days after collection. We recommend that under commercial circumstances the homospermic semen doses contain no <3 billion spermatozoa/dose.  相似文献

An investigation involving seven boars, active in artificial insemination, and 1,350 multiparous sows was conducted at a private farm and aimed at examining the relationship between sperm quality traits and boar fertility in terms of farrowing rate and litter size. This experiment was done for 6 months. The semen samples were evaluated for subjective sperm motility and concentration. Ejaculates with at least 1 × 108 sperm/mL and 70% sperm progressive motility were extended with a commercial medium to 30 × 106 sperm/mL and used for artificial insemination (AI). AI dose was 100 mL semen containing 3 × 109 spermatozoa. Aliquots of diluted semen were assessed for live morphologically normal spermatozoa (LMNS, eosin-nigrosin stain exclusion assay) and sperm chromatin instability (SCI, acridine orange assay). Farrowing rates according to different boar sperm varied (p < 0.001) from 59.3 to 88.92%. The mean values of LMNS (47.2~76.5%) and SCI (0.16~4.67%) differed significantly among boars. LMNS (r = 0.79, p < 0.05) and SCI (r = -0.90, p < 0.02) accounted for 62.2 and 81.7% of the variability in farrowing rates, respectively. After the combination of sperm traits, the relationship between percentage of LMNS with stable chromatin structure and farrowing rate was significant (r = 0.86, p < 0.05). The number of live piglets per parturition was not significantly correlated with sperm quality attributes. In conclusion, boar fertility after AI with freshly diluted semen can be predicted based on the evaluation of sperm morphology and chromatin integrity.  相似文献

Three experiments evaluated the effects of dietary Se and vitamin E on the ultrastructure of spermatozoa, ATP concentration of spermatozoa, and the effects of adding sodium selenite to semen extenders on subsequent sperm motility. The experiment was a 2 x 2 arrangement of treatments in a randomized complete block design. A total of 10 mature boars were fed from weaning to 18 mo of age diets fortified with two levels of supplemental Se (0 or .5 ppm) or vitamin E (0 or 220 IU/kg diet). The nonfortified diets contained .06 ppm Se and 4.4 IU vitamin E/kg. In Exp. 1, the spermatozoa from all boars were examined by electron microscopy. Vitamin E had no effect on structural abnormalities in the spermatozoa. When the low-Se diet was fed the acrosome or nuclei of the spermatozoa was unaffected, but the mitochondria in the tail midpiece were more oval with wider gaps between organelles. The plasma membrane connection to the tail midpiece was not tightly bound as when boars were fed Se. Immature spermatozoa with cytoplasmic droplets were more numerous when boars were fed the low-Se diet, but the occurrence of midpiece abnormalities occurred in boars fed diets with or without Se or vitamin E. Our results suggest that Se may enhance spermatozoa maturation in the epididymis and may reduce the number of sperm with cytoplasmic droplets. In Exp. 2, the concentration of ATP in the spermatozoa was evaluated in the semen of all treatment boars. When the low-Se diet was fed, ATP concentration was lower (P < .01), whereas vitamin E had no effect on ATP concentration. Experiment 3 investigated the effect of diluting boar semen with a semen extender with sodium selenite added at 0, .3, .6, or .9 ppm Se. Three ejaculates from each boar were used to evaluate these effects on sperm motility to 48 h after dilution. Sperm motility declined (P < .01) when Se was added to the extender, and this decline was exacerbated as the concentration of added Se increased (P < .01). The added Se was demonstrated to be tightly adhered to the spermatozoa. Overall, these results suggest that low Se-diets fed to boars resulted in abnormal spermatozoal mitochondria, a lower ATP concentration in the spermatozoa, and a loose apposition of the plasma membrane to the helical coil of the tail midpiece, but no effect from inadequate vitamin E was demonstrated. The concentration of testosterone, 17-beta oestradiol and aflatoxin B1 were studied in the semen plasma of 21 boars of four breeds for the period of twelve months. The following spermiological parameters were investigated: semen volume, sperm concentration, percentage of abnormal spermatozoa, and survival of spermatozoa. The fertilizing capacity of ejaculates was evaluated according to the conception rate of sows and gilts after the first insemination, according to the average number of piglets per litter and average number of live-born piglets per litter. The highest aflatoxin B1 residues in sperm were recorded in March to May and were related with aflatoxin concentration in feed ration. The group of boars with fertility disorders had more aflatoxin in their sperm (up to 100 pmol . l-1), lower sperm concentration, lower survival of spermatozoa, and a larger proportion of abnormal spermatozoa. The year season had a significant influence on the concentration of the hormones. The highest average value of testosterone (10.2 +/- 1.28 nmol) was obtained in autumn and lower values were recorded in winter. The changes in 17-beta estradiol concentration were similar to the changes in testosterone content, with the maximum value in November (0.249 nmol X 1(-1]. The boars with reproduction disorders had a significantly lower concentration of 17-beta oestradiol. Significant correlations were found between the concentration of the hormones, semen volume, and sperm concentration. 17-beta oestradiol also had a significant positive correlation to abnormal spermatozoa and to the activity of aspartate aminotransferase.  相似文献

The objective of this study was to characterize the effect of an experimental infection with the classical swine fever (CSF) virus on libido and ejaculate parameters of adult boars. Four boars 10 month old were infected with a CSF field isolate (Visbek/Han95). Semen was collected every second day after infection and daily during the pyrexic phase. The only clinical signs in the boars were an increase in body temperature, but never above 39.9 degrees C and a temporally reduction of food intake. The libido was always good, so semen collection was performed in three boars without difficulty and the semen quality was always in the range of the minimum requirements for sperm that is used for artificial insemination. Although one boar had a good libido only a sperm free ejaculate could be collected on one day. The results show that a CSF virus infection of adult boars hardly causes any clinical symptoms and that sperm can be obtained despite fever. Insemination boars may thus be of special epidemiological relevance for the dissemination of the CSF virus.  相似文献

The effect of porcine reproductive and respiratory syndrome virus (PRRSV) on semen quality was examined in a group of 11 spontaneously infected boars in a commercial boar stud. Semen samples were collected 4 weeks prior to 4 weeks post-infection (wpi). Infection with PRRSV of the European genotype subtype 1 (EU-1) was verified by specific quantitative real-time polymerase chain reaction (RT-PCR) in 36% of the serum samples. All boars seroconverted before 4 wpi and remained in normal condition throughout the study. Comparison of the percentage of morphologically intact spermatozoa revealed an increase of acrosome-defective spermatozoa (P = 0.012) between −4 and 4 wpi. Significant deleterious effects on semen quality were detected for membrane integrity when semen had been stored for 2 days after sampling. Analysis of sperm subpopulations in a thermoresistance test on day 7 after sampling revealed alterations in the percentage of circular, progressively motile spermatozoa (P = 0.013), in the percentage of non-linear, progressively motile spermatozoa (P = 0.01), and on the amplitude of lateral sperm head displacement (P = 0.047). There was no difference in the incidence of mitochondrially active spermatozoa (P = 0.075). Investigation of routine production data between pre- and post-infection status showed no differences on ejaculate volume (P = 0.417), sperm concentration (P = 0.788), and percentage of motile spermatozoa (P = 0.321). This case report provides insights into a potential control strategy for PRRSV outbreaks in boar studs.  相似文献

The aim of this study was to investigate the effect of season, temperature, humidity, age of the boar, and semen collection interval on sperm morphology in Duroc boars in Thailand, kept either in a conventional open air system (CONV) or in an evaporative cooling system (EVAP). In total, 1176 ejaculates from 110 sexually mature boars in six CONV herds and five EVAP herds were morphologically examined during a one-year period. Analysis of variance was applied to the data. Minor differences in the sperm morphology traits analyzed were found between the housing systems. The objective of this research was to determine the effect of vaccination against porcine circovirus type 2 (PCV2) on ejaculate characteristics, PCV2-specific antibody titers in serum, viremia, and viral shedding in the semen of PCV2-positive boars. Before vaccination, all boars were confirmed by PCR to be naturally infected with PCV2. The boars were vaccinated with a commercial killed vaccine against PCV2 (n = 5) or served as controls and received 2 mL of 0.9% saline (n = 5). Semen and blood samples were collected before vaccination at wk 0 and at 7-d intervals thereafter until wk 8. Sperm concentration and characteristics of sperm motility were assessed using a computer-assisted sperm analysis system, and sperm morphology was evaluated using light microscopy after staining. The PCV2 antibody titers were determined in serum using an ELISA, and the genomic copy numbers of PCV2 DNA in serum and semen were determined by real-time PCR. In general, there were no effects of treatment or treatment × week on semen or sperm characteristics (P > 0.10). An effect of treatment × week was detected for serum antibody titers (P < 0.01). Compared with controls, PCV2 antibody titers in vaccinated boars were less (P < 0.01) at wk 7 (1.01 ± 0.05 titer/mL vs. 1.23 ± 0.05 titer/mL) and tended (P = 0.07) to be less at wk 8 (1.05 ± 0.05 titer/mL vs. 1.17 ± 0.05 titer/mL). There were no effects of treatment or treatment × week for serum and semen genomic copy numbers of PCV2 DNA (P > 0.10). There was a tendency (P = 0.09) for an effect of week on serum viral load. It was evident that during this experiment, boars experienced reoccurring PCV2 infection, and the detection of an increased PCV2 DNA load in serum preceded that in semen; the duration of reoccurring infection appeared to be less in vaccinated boars compared with controls. In summary, vaccination against PCV2 can reduce antibody titers when given postinfection and has no dramatic effect on indicators of semen quality. Vaccination against PCV2 in naturally infected boars can also decrease the length of reoccurring infection and decrease the duration of viral shedding in semen.  相似文献

The objective was to determine the effects of repeated injections of PGF2alpha (Lutalyse; Pfizer) on semen and libido characteristics in terminal-line boars. Semen was collected once weekly from wk 0 to 15 and on four consecutive days during wk 16. Boars received an i.m. injection of 10 mg of PGF2alpha (n = 11) or 2 mL of vehicle (n = 11) immediately before entering the collection room. For the weekly collections, semen volume (220.3 +/- 3.2 mL; mean +/- SE), gel weight (38.7 +/- 0.7 g), total sperm cells (65.4 +/- 1.2 billion), motile sperm cells (67.4 +/- 0.6 %), and sperm velocity (125.9 +/- 1.2 microm/s), were affected by time (P < 0.01) but not by treatment (P > 0.10). Sperm concentration (0.31 +/- 0.01 billion/mL) was not affected (P > 0.10) by time or treatment. The percentage of morphologically normal sperm cells, assessed at wk 16, did not differ (P = 0.39) between groups (80.8 +/- 1.0). Libido was evaluated from wk 0 to 16. There were no effects of treatment or time (P > 0.10) on the period from injection to the start of ejaculation (225.6 +/- 9.1 s). Duration of ejaculation was affected by treatment (P < 0.01; 472.0 +/- 43.1 s and 280.4 +/- 43.1 s, for PGF2alpha-treated and control boars, respectively) and time (P < 0.01). During the intensive collection period (wk 16), semen volume (200.1 +/- 7.1 mL), gel weight (39.2 +/- 1.5 g), sperm concentration (0.19 +/- 0.01 billion/ mL), total sperm cells (39.5 +/- 3.6 billion), motile sperm cells (65.6 +/- 2.2%), and sperm velocity (117.8 +/- 3.7 microm/ s) were affected by time (P < 0.10) but not by treatment (P > 0.10). The period from injection to the start of ejaculation tended to decrease (by 44%) during the intensive collection period in PGF2alpha-treated boars, but not in controls (treatment x time, P = 0.07). Regardless, the period from injection to the start of ejaculation did not differ (P = 0.63) between groups on d 4 of the intensive collection period. Duration of ejaculation was affected by treatment (P < 0.01; 459.1 +/- 24.1 s for PGF22alpha-treated boars vs. 303.1 +/- 24.1 s for controls) but not by time (P > 0.10). Overall, there were no exceptional positive or negative effects of long-term treatment with PGF2alpha on semen characteristics and libido in boars.  相似文献

Sperm morphology and the fertilizing capacity of ejaculated spermatozoa were examined in 6 Swedish Landrace boars before and after heat stress. The boars were exposed to 35° C during 100 h in a climatic room. Fertility was measured by insemination of gilts before and at various times after heat stress. Each gilt (n = 44) was inseminated with a total of 5×109 spermatozoa diluted to 10O ml with EDTA-glucose diluent and fertilization was assessed by examining recovered ova 2 days after insemination.Changes in semen quality varied among the boars from a very weak response in 2 boars to pronounced semen alterations occurring 2–6 weeks after heat stress in the other boars. A close relationship was found between seminal changes and fertilization rates, all ejaculates which had high fertilization rates being of the same quality as the pre-exposure ejaculates. The ejaculates that had poor fertility were characterized by lowered sperm motility and increased numbers of spermatozoa with abnormal heads, proximal cytoplasmic droplets and nuclear pouch formations.  相似文献

Phospholipase C zeta (PLCz) and cyclooxygenase isoenzyme type 2 (COX-2) are important in spermatogenesis, but their effect has not yet confirmed in pigs. Therefore, this study was aimed to analyse their association with sperm quality and fertility and to identify the mRNA and protein expression in boars reproductive tissues. DNA samples from 231 Pietrain (PI) and 109 Pietrain × Hampshire (PIHA) pigs with records of sperm quality [sperm concentration (SCON), motility, semen volume, plasma droplet and abnormal spermatozoa rate] and fertility (non-return rate and number of piglet born alive) traits were available. A SNP in non-coding region of PLCz g.158 A > C was associated with SCON (p < 0.05) in PIHA population while the polymorphism of COX-2 g.68 G > A in 3' UTR was not associated with any traits. For mRNA and protein expression study, a total of six boars were divided into two groups with G-I and G-II, where G-I was characterized for relatively better sperm quality. Both genes expressed higher in reproductive tissues compared with non-reproductive tissues. Phospholipase C zeta mRNA expressed higher in testis (p < 0.01), all parts of epididymis and spermatozoa from G-I, while COX-2 expressed higher in testis (p < 0.05), head and body of epididymis (p < 0.01), and spermatozoa from G-II boar. Both proteins were localized in Leydig cells and spermatozoa. These results might shed light on roles of these genes in spermatogenesis as candidate for boar sperm quality and fertility, but still the lack of association across populations should be considered.  相似文献

为研究不同品种、采精月份、采精月龄和采精间隔等因素对加系公猪精液品质的影响,以及品种、初次采精周龄对精液质量稳定性的影响,本研究以江西某种公猪站79头加系大白猪、长白猪、杜洛克猪种公猪为试验群体,收集2018年12月至2020年12月3 921条精液采集与精液质量数据,通过混合线性模型与方差分析探究各因素对精液量、精液密度、精子活力、总精子数及其稳定性的影响。结果显示,从不同品种对精液质量的影响来看,长白猪精液量和总精子数均高于大白猪、杜洛克猪,但杜洛克猪精液密度高于长白猪、大白猪,杜洛克猪精子活力最低;从不同月份来看,1~3月采精精液密度最高,4~6月采精精子活力最高,10~12月采精精液量和总精子数最高,精液量呈现秋冬多、春夏少的季节变化规律。公猪不同月龄采精,精液质量指标也存在差异,月龄越小精液量越低,但精液密度偏高,精子活力相对较好,在19~24月龄黄金期总精子数最高。不同采精间隔对精液质量有较大影响,采精间隔越长,精液量、精液密度、精子活力和总精子数相对较好,采精间隔为5 d时综合性能最佳,但过长的采精间隔导致精子活力降低。品种影响总精子数稳定性,长白猪、大白猪总精子数稳定性显著优于杜洛克猪(P<0.05)。本研究结果表明,品种、采精月份、采精月龄和采精间隔均会影响公猪精液质量,关注这些因素有助于公猪站制定更完善的生产计划,提高公猪利用率。  相似文献

旨在评价酵母培养物(YC)对公猪生产性能的影响。将90头体重约为80kg的公猪随机分为3组,即对照组、试验Ⅰ组和试验Ⅱ组;对照组饲喂基础日粮,试验Ⅰ组和试验Ⅱ组基础日粮中分别添加5kg/t和7kg/t的酵母培养物,预试期5d,正试期45d,试验期间,3组饲养管理条件一致;试验结束时,评定各组公猪精液的颜色、气味,测定并比较各组公猪的精液量、精液pH值、精子活力、精子密度、精子畸形率、抗力指数。结果表明,试验Ⅰ组和试验Ⅱ组公猪的精液颜色和精液气味与对照组相比没有明显差异;3组公猪的精子抗力指数一致;试验Ⅰ组和试验Ⅱ组的精液量、精子活力、精子密度均显著大于对照组(P<0.05),精子畸形率均显著小于对照组(P<0.05),但这4项指标在2个试验组之间差异均不显著(P>0.05);3组公猪的精液pH值差异均不显著(P>0.05)。综上提示,基础日粮中添加一定量的酵母培养物能够提高公猪部分生产性能指标。  相似文献

Chinese Erhualian pigs are known for prolificacy with distinct reproductive traits compared with Western commercial breeds. In this study, a four‐generation intercross resource population was constructed using White Duroc boars and Chinese Erhualian sows as founder animals, and a total of 14 male reproductive traits were recorded in 411 F2/F3 boars including the testis and epididymis weights, the seminiferous tubular diameter and spermatogenesis at 60, 90 and 300 days of age, semen characteristics, serum testosterone concentration and libido level at 300 days of age. The White Duroc–Erhualian boars showed remarkable segregations in the traits measured except for the seminiferous tubular diameter and had high ratio (13.9%) of the abnormality of spermatogenesis, providing a good experimental population for detecting quantitative trait loci affecting these male reproductive traits. Furthermore, the correlations among nine male reproductive traits at 300 days of age indicated that the testis weight and the body weight were strongly correlated with the sperm production, supporting the two traits as important parameters for boar selection to increase sperm production and ultimately improve boar fertility. The libido level in the White Duroc–Erhualian boars that was evaluated by a new and easily recorded scoring system showed a significant correlation with serum testosterone concentration. Yet, both libido and serum testosterone concentration were not correlated with the sperm production. Results of this study provided new information on the male reproductive physiology and genetics in Chinese Erhualian and White Duroc boars.  相似文献

Retinol-binding protein 4 (RBP4), androgen receptor (AR), relaxin (RLN), acrosin (ACR) and osteopontin (polymorphism in intron 6 named OPNin6; polymorphism in promoter region named OPNprom) were addressed as functional candidate genes for sperm quality and boar fertility and investigated for their association with sperm concentration, motility, semen volume per ejaculate, plasma droplets rate, abnormal spermatozoa rate as well as non-return rate and number of piglets born alive. Therefore 356 AI boars of the purebred Pietrain (PI) and crossbred Pietrain x Hampshire (PI x HA) were genotyped at these loci. Analysis of variance revealed significant associations of RBP4 (p < 0.05), ACR (p < 0.01), and OPNin6 (p < 0.05) with sperm motility. OPNin6 (p < 0.05) was also associated with number of piglets born alive. Moreover, AR (p < 0.05) and OPNprom (p < 0.05) were significantly associated with abnormal spermatozoa rate. For RLN (p < 0.01) there was evidence for effects on sperm volume and ACR significantly affected sperm concentration (p < 0.05) as well as non-return rate (p < 0.05). No significant effects of any locus on plasma droplets rate were observed.  相似文献

本试验旨在研究牛磺酸(Tau)对种公猪性欲、精液品质、血清激素含量及精浆抗氧化能力的影响。选用24头年龄、体重相近的健康成年大约克夏种公猪,随机分为4个组,每组6个重复,每个重复1头猪。各组分别饲喂牛磺酸添加水平为0(对照组)、2、4、6 g/kg的饲粮,试验期90 d,分为1~45 d和46~90 d 2个阶段。结果表明:1)试验46~90 d时,添加6 g/kg牛磺酸显著提高了种公猪的性欲(P0.05)、采精量(P0.01)、精子活力(P0.05),4 g/kg牛磺酸改善了精子密度和精子畸形率(P0.05);2)与对照组相比,6 g/kg牛磺酸水平组极显著提高了种公猪血清促黄体素(LH)含量(P0.01),显著提高了睾酮(T)含量(P0.05);3)试验46~90 d时,6 g/kg牛磺酸显著降低了种公猪精浆丙二醛(M DA)含量(P0.05),显著提高了超氧化物歧化酶(SOD)活力(P0.05),4 g/kg牛磺酸显著提高了谷胱甘肽过氧化物酶(GSH-Px)活力(P0.05)。由此可见,饲粮中长期添加牛磺酸可以调控种公猪血清激素含量,增加精浆抗氧化能力,进而增强种公猪性欲,提高种公猪精液品质,本试验条件下牛磺酸适宜添加水平为6 g/kg。  相似文献

This study was carried out to assess the in vitro quality of canine semen frozen in an ultrafreezer at -152 degrees C and to evaluate the male-to-male variation of frozen semen in five male dogs of the Canarian Mastiff breed. Four ejaculates of each dog were processed individually (5% glycerol and 0.5% Equex) to reach a final concentration of 100 x 10(6) spermatozoa/ml. Then, two freezing techniques were tested to assess the seminal quality (sperm motility, live spermatozoa and abnormal sperm cell percentages) at 1, 30, 60, 120 and 360 days after freezing: (i) semen was frozen and stored in liquid nitrogen; (ii) semen was frozen and stored in the ultrafreezer at -152 degrees C. After freezing-thawing, both freezing protocols showed no significant differences in sperm motility and the percentages of live and abnormal spermatozoa. On the other hand, the microscopic characteristics of spermatozoa in fresh semen were practically similar among males; however, after the semen processing and freezing, significant differences were observed (p < 0.05) among males, especially as regards sperm motility. This inter-individual variability was detected in both freezing protocols, showing that the male-to-male variation in the seminal quality post-freezing was independent of the freezing technique used. The in vitro results obtained in the Canarian Mastiff breed confirmed that the use of ultra-freezers at -152 degrees C is a potential alternative to liquid nitrogen for storing canine semen for long periods of time.  相似文献

Background

The aim of the present study was to investigate the efficacy of the Improvac on testosterone concentration in blood serum, sexual behavior and sperm quality in matured AI boars. A total of nine Danish Landrace AI boars were included in the analysis.

Methods

The trial period lasted for 15 weeks and was divided into four periods: Control period: three weeks before vaccination; Period I – four weeks after first vaccination; Period II – four weeks after second vaccination, Period III – four weeks after third vaccination. Blood and sperm samples were collected at weekly intervals. Freshly collected sperm samples were analyzed.

Results

Testosterone concentration correlated with libido (r = 0.531; p < 0.001), volume of ejaculate (r = 0.324; p < 0.001) and the percentage of morphologically normal spermatozoa (r = 0.207; p < 0.05). Testosterone concentration rised significantly (p < 0.05) in 5–6 week of trial, e. i. after the first dose of Improvac and after this peak the level of testosterone further progressively decreased (p < 0.05).

Conclusions

Results from this study indicate that active immunization of sexually matured boars against GnRH has negative impact on testosterone concentration, sexual behavior, volume of ejaculate and total number of normal spermatozoa in ejaculate.  相似文献

The aim of the study was to determine the relation between the semen quality, frequency of sperm defects, sperm dimensions and shape, and the ejaculate volume of Large White and Landrace boars. A total of 648 ejaculates collected from 31 Large White and 30 Landrace boars were divided into three groups according to the criterion of the ejaculate volume. In this study Landrace boars produced ejaculates with higher volume, sperm concentration, and total numbers of spermatozoa than Large White boars. Landrace boars also showed a lower frequency of sperm with morphological abnormalities (P < 0.05). Landrace boars sperm had larger heads, which were by 0.15 μm longer, and by a larger perimeter and area (P < 0.05). Landrace boar spermatozoa also had a longer flagellum and were generally larger and by 2.07 μm longer than Large White boar sperm (P < 0.05). Significant differences were also found in the shape of sperm of the two breeds (P < 0.05). Landrace boars sperm had more elongated heads, and the ratio of head size to flagellum length was lower than in Large White boars sperm (P < 0.05). Sperm from ejaculates with low volume had a shorter flagellum and a greater head length/flagellum length ratio than sperm from medium- and high-volume ejaculates (P < 0.05).  相似文献
