Fecal samples from suckling (n=153) and weaned (n=116) piglets with diarrhea in Japan were examined for shedding of viral, bacterial, and parasitic pathogens using culture, microscopic, and polymerase chain reaction methods. In suckling piglets, diarrhea was attributed to infection with a single etiologic agent in 60.8% of cases and with combinations of agents in 22.2%. In weaned piglets, diarrhea was attributed to a single etiologic agent in 43.1% and to combinations of agents in 47.4% of cases. Rotavirus was the most prevalent agent in suckling (67.3%) and weaned (65.5%) piglets. The detection of other pathogens was associated with age of the animals examined. Coccidia were predominantly isolated from suckling piglets, whereas Escherichia coli was found predominantly in weaned piglets. Although a relationship was not observed between detection rate of rotavirus and age of piglets, a single group of rotavirus was detected in 87.5% of suckling piglets whereas multiple groups were detected in 51.6% of weaned piglets. The results of this study confirm that diarrhea in piglets can, to a variable degree, be causally associated with multiple agents. Additionally, these results suggest reasons why this syndrome can be difficult to control.  相似文献

A cross-sectional survey was performed on 20 pig breeding farms in southern Hesse, central Germany, to evaluate the prevalence and age-dependent occurrence of intestinal protozoan parasites in unweaned piglets. Faecal samples of 514 clinically unaffected piglets of different age (< 1 to 5-7 weeks) were examined using the sodium acetate-acetic acid-formalin (SAF) concentration technique. Infections with the following protozoan species were detected: Balantidium coli (16 of 20 farms), Entamoeba sp. (15), Jodamoeba sp. (14), Isospora (I.) suis (9), Chilomastix sp. (6) and Eimeria spp. (6). The protozoan species differed in the start and course of (oo)cyst excretion. I. suis oocysts and Jodamoeba cysts were detected already in the first week of life whereas shedding of the other parasites started later on. The prevalence of Isospora oocyst excretion increased to a maximum (18%) in 2-3 weeks old animals followed by a sharp decline. The proportion of Balantidium, Entamoeba or Jodamoeba positive suckling piglets continously increased until the age of 5-7 weeks to 60%, 52% and 22%, respectively, whereas that of Chilomastix positive animals remained on a low level of 8-12% independent of the age. Mixed fecal samples of 264 litters from five piglet production farms (155-238 sows/farm) were investigated three times during the suckling period for the occurrence of Isospora suis over the period of 1 year. On all five farms Isopora suis was found to be a common endoparasite with infection rates being highest in litters of 3-4 weeks of age. By the end of the third investigation period the cumulative infection rate was 53.8% of the litters ranging from 20.0% to 81.5% for the single farms. During the suckling period the infection rate increased from 18.6% to 32.6% and then to 37.7%. Diarrhea was present in 66.3% of the sampled litters with the highest rates at the end of the suckling period. 63.4% of the litters which showed diarrhea and 34.8% of those without diarrhea excreted I. suis within the study period. Diarrhea was recorded for 78.2% of the I. suis-positive litters and for 52.5% of the Isospora-negative litters. In summer and fall the occurrence of I. suis was higher (66.3% and 61.0%, respectively) than in spring and winter (47.7% and 37.9%, respectively). In litters with diarrhea and pathogenic E. coli I. suis often occurred simultaneously. Above-average hygiene measures and mainly perforated pen floors seemed to lower the risk of isosporosis. With the exception of Strongyloides ransomi other parasites were not found in the fecal samples of suckling piglets. Two specialized piglet rearing farms, a conventional large-scale rearing unit and a farm managed according to the segregated early weaning (SEW) system were examined three times during the 6-7 week rearing period. In both units I. suis was common, but was not correlated with diarrhea. In the SEW unit the infection rates decreased from 37.5% to 20.2% and to 4.1%, while the infection rate in the conventional unit slightly increased from the first (17.2%) to the second (21.9%) investigation and stayed at this level at the third sampling.  相似文献

An observational study was conducted to determine the prevalence of Isospora suis oocysts in fecal samples from suckling piglets in Ontario, and to evaluate the relationship between the presence of I. suis oocysts and diarrhea. Fifty farms and 709 litters of piglets were included in the study. Oocysts were detected on 70% of farms, with 187 litters infected. A litter of pigs that was positive for oocysts was significantly more likely to exhibit diarrhea than a litter that was negative [odds ratio (OR) = 4.0; 95% confidence interval (CI) = 2.8 to 5.8; P < 0.001]. Management and housing factors were examined with respect to risk factors for the presence of I. suis. Farms that did not use a detergent when cleaning farrowing crates were 10-times more likely to be positive for I. suis than those that used a detergent (P = 0.007). It was concluded that coccidiosis is a common problem on Ontario swine farms.  相似文献

Porcine coccidiosis caused by Isospora suis is one of the leading causes of neonatal diarrhea in suckling piglets. Currently the only registered drug for metaphylaxis is toltrazuril. To evaluate the effect of treatment on piglets from 7 Austrian farms without and 8 Austrian farms with toltrazuril application we examined oocyst excretion (including determination of oocysts per gram of feces; OPG), diarrhea (fecal score FS 1-4 with 3 and 4 being diarrhea), and general health (health score HS 1-4 with 3 and 4 describing poor health). Both groups included farms with different levels of hygiene. Samples from 265 litters without treatment, comprising 1588 individual samples, and 1548 samples from 258 treated litters were taken twice (around the 14th and the 21st day of life, respectively), examined by autofluorescence and, if positive, by McMaster counting. In both groups animals had less diarrhea and lower health scores during the second sampling but the treated piglets were always significantly healthier and had less diarrhea. The percentage of weaned piglets was higher in treated animals although this was not significant (p=0.052). In the first round of sampling 17.8% of the individual samples from untreated piglets were positive for oocysts (with a maximum prevalence on the 12-15th day of life) while in the treated piglets only 0.4% shed oocysts p<0.001). At the second sampling only 2.1% of the untreated animals and none of treated piglets excreted I. suis (p=0.083). Positive animals shed up to 8 × 10(3)OPG. There was an increased risk for infected piglets to develop diarrhea (odds ratio, OR 4.73) and poor health (OR 5.05) in untreated piglets, and poor hygiene without disinfection was identified as a risk factor for poor health (OR 1.90), diarrhea (OR 1.42) and oocyst excretion (OR 1.73). The risk of poor health (OR 2.89) and diarrhea (OR 1.44) was also increased for piglets under poor hygienic conditions receiving toltrazuril, so both metaphylaxis of coccidiosis and good hygiene are necessary to effectively control neonatal diarrhea. The costs of treatment are considerably lower than the estimated financial production losses. Therefore, treatment is recommended for farms where clinical coccidiosis is diagnosed.  相似文献

In different age groups endoparasites are of varying importance. Sows are predominantly infected with gastrointestinal strongyles (mainly Oesophagostomum spp.), Ascaris suum and Eimeriae, to a lesset extent with Trichuris suis, Hyostrongylus rubidus, Strongyloides ransomi or Isospora suis, while in suckling piglets I. suis and occasionally S. ransomi frequently occur. Weaners and fatteners are infected with coccidia and gastrointestinal strongyles, later with A. suum. Whipworms are also found occasionally. I suis causes diarrhoea in suckling piglets, while in older animals the infection is not correlated with disease. Damage induced by helminths is mainly due to reduced performance, in the case of ascarosis to reduced carcass value. Therefore planned antiparasitic measures should be taken at all stages of pig keeping. Hygienic measures can reduce infection pressure; however parasite elimination is difficult to obtain under conventional management conditions. Consequently the control of parasitic infection at the early stage of piglet production is desirable.  相似文献

In order to evaluate the prevalence of Isospora suis in conventional piglet production in Germany, pooled faecal samples from 327 pig litters from 18 pig production units (20-320 sows each) were examined. At least 10 litters from each farm were investigated. I. suis was present on 83% of the farms and 42.5% of the litters, the infection rate being highest in the third week of age (48.2%). I. suis was found more frequently in samples of diarrhoea than in firm faeces (49.2% compared to 22.2%). Twenty naturally infected piglets from six of these farms underwent examination post mortem, including histology, virology and bacteriology. Histological examination revealed atrophy of the villi in various degrees, mild crypt hyperplasia, fusion of the villi, metaplastic epithelium, erosions and necrosis, especially in the medium and the posterior jejunum and in the ileum. Asexual and sexual developmental stages of the parasite were found in varying numbers in the epithelium of the whole of the small intestine. Bacteria and viruses were mostly excluded as the cause of diarrhoea, and it was concluded that I. suis was the primary pathogen inducing distinct changes and clinical symptoms of diarrhoea.  相似文献

The aim of this investigation was to determine the prevalence of coccidian infections in suckling piglets and sows in Poland. The research was carried out in 14 out of 16 Polish provinces in the years 2003-2005. The investigation was conducted on three types of farms: large farms (>100 sows), medium farms (25-100 sows) and small farms (<25 sows). Diarrhoea of unweaned piglets was observed on all the examined farms. Overall, 780 litters of suckling piglets from 104 farms and 267 mother sows were examined. The faeces were analyzed with the modified McMaster method. Isopsora suis was found in 217 (27.8%) litters from 70 (66.7%) farms. Eimeria spp. was detected only in 20 (2.6%) litters from 12 (11.5%) farms. On the large farms I. suis infection was detected in 31.7% of litters whereas Eimeria spp. in 1.4% of them. On the medium sized farms I. suis was found in 18.1% of litters and Eimeria spp. in 0.6%. On the small farms I. suis was detected in only 13.2% of litters, whereas Eimeria spp. in as many as 28.9%. I. suis and Eimeria spp. oocysts were found in 18 (6.7%) and 16 (6%) sows respectively. From 72 sows producing I. suis infected piglets only 12 (16.7%) shed I. suis oocysts and as little as 4 (5.6%) shed Eimeria oocysts. In the remaining 56 sows (77.8%) no cases of coccidian infections were detected. The results of this investigation demonstrate the high prevalence of I. suis in suckling piglets on the large swine farms in Poland.  相似文献

Intestinal contents of suckling (n?=?45) and newly weaned (n?=?45) piglets, suffering from diarrhea in the province of Villa Clara in Cuba, were tested for viral, bacterial, and parasitic enteropathogens from May to June 2008. At least one enteropathogen was identified in 53.3 % of piglets and enterotoxigenic Escherichia coli (ETEC; 25.6 %) was the major pathogen; mostly STa+/STb+ or F4+/STa+/STb+ ETEC were isolated. The overall occurrence of the rest of pathogens was 10 % for transmissible gastroenteritis virus (TGEV) and Cryptosporidium parvum, 6.7 % for rotavirus A and Isospora suis, 5.6 % for α-toxigenic Clostridium perfringens, 3.3 % for verotoxigenic E. coli (VTEC), and 2.2 % for Salmonella enterica subspecies enterica serovar Newport. TGEV and α-toxigenic C. perfringens were only identified in suckling piglets, while Salmonella Newport and VTEC were only detected in weaned pigs. Porcine epidemic diarrhea virus (PEDV), β-toxigenic C. perfringens, Eimeria spp., and helminths were not identified. Eight kinds of mixed infections were detected in 25 % of enteropathogen positive piglets. ETEC was present in 10 of 12 mixed infections, and TGEV infections were never combined. This survey demonstrates that several enteropathogens are circulating in piggeries located in the province of Villa Clara in Cuba, and that is necessary to improve surveillance, prevention, and control of enteric infections in order to increase production efficiency.  相似文献

The prevalence of rotavirus infection was studied in 1090 litters from 26 sow herds. Samples of normal, semifluid and watery stools were examined for rotavirus by an ELISA-test on faeces. Rotavirus was detected in 77% of the herds and in 30.5% of the litters (prevalence rates). The highest prevalence rate was seen in piglets between 21 and 41 days of age. Gilts' litters had a very high prevalence during the first week of life. Apart from this, no difference was found between litters from gilts and older sows. Rotavirus was detected more frequently in semiliquid, loose stools than in normal or watery stools, and an association between virus detection and diarrhoea could not be demonstrated. However, litters which shedded rotavirus during the suckling period had lower weight gains and higher incidence rates of respiratory diseases than virus-free litters. Litters weaned at 2 weeks in battery cages had slightly increased risk of shedding rotavirus compared to litters weaned in more traditional systems. The study revealed that rotavirus is widespread in Danish swine herds. The findings give evidence to suggest that the type of mild diarrhoea in 3-week-old piglets known as steatorrhoea or "white scours" may be associated with rotavirus infection, possibly in combination with E. coli and other agents. The high prevalence in piglets weaned at 2 weeks plus the higher morbidity and mortality among such piglets sustain the conclusion that piglets should not be weaned before 3 weeks of age or below a body weight of 6-7 kg.  相似文献

Summary

Faecal samples from suckling piglets from 113 litters on 25 farms in the Netherlands were examined to study the prevalence of gastrointestinal parasites, in particular Strongyloides ransomi and Isospora suis. S. ransomi was not found but I. suis was demonstrated in 17 of the 25 farms and in 41 out of 77 litters of these farms (53%). No other parasites were found. On the basis of these results, a longitudinal study on the incidence and significance of I. suis was performed on 10 farms (5 litters/farm). I. suis was found on 9 out of 10 farms and in 56% of the litters. Farms differed in infection levels, measured as the numbers of patent litters, and in the onset of patency. Other parasites were only found in faecal samples from the sows and from farrowing pens. These were, in order of prevalence, Balantidium coli, Ascaris suum, Eimeria spinosa, E. debliecki and E. suis.  相似文献

A total of 142 stool specimens from pigs on 24 farms from the province of Zaragoza (northeastern Spain) were screened for Cryptosporidium spp. Samples were first analysed by routine techniques (formalin-ethyl acetate sedimentation method and modified Ziehl-Neelsen stain) selecting those microscopically positive for genetic characterization. Cryptosporidium species and genotypes were determined by a nested PCR-RFLP technique at the 18S ribosomal DNA locus and sequencing of the PCR-positive secondary products. Cryptosporidium oocysts were microscopically identified in the faeces of 32 pigs (22.5%) from 15 farms (62.5%). Infected animals included 23 weaned piglets (30.7%), 5 fattening pigs (11.9%) and 4 sows (16%). Diarrhoea was not detected in any of the infected pigs. The molecular characterization was successfully performed in 26 samples from 14 farms. Cryptosporidium suis was found in 10 specimens from 7 farms (nine weaned piglets and one sow) and the Cryptosporidium pig genotype II in 16 samples from 10 farms (13 weaned piglets and 3 fattening pigs). Both C. suis and the pig genotype II were concurrently detected on three farms.  相似文献

本试验以表达猪乳铁蛋白肽的重组屎肠球菌(pNZ8112-PLFcin/Ef)饲喂断奶仔猪,研究其对仔猪生长性能的影响和抗产肠毒素大肠杆菌(enterotoxigenic Escherichia coli,ETEC)感染的效果。选取28日龄体重相近的健康断奶仔猪36头,随机分为3组(重组屎肠球菌组、空载体组和培养基组),每组3个重复,每个重复4头仔猪。重组屎肠球菌组和空载体组分别饲喂添加pNZ8112-PLFcin/Ef (6×1012 CFU/kg)和pNZ8112/Ef (6×1012 CFU/kg)的基础日粮,而培养基组饲喂含有相同体积的GM17液体培养基的基础日粮。试验期26 d。结果显示,与培养基组相比,重组屎肠球菌组断奶仔猪的平均日增重极显著提高(P<0.01);料重比显著降低(P<0.05);腹泻率明显降低,肠道菌群的均匀度和多样性指数均下降。为进一步探究表达猪乳铁蛋白肽的重组屎肠球菌对断奶仔猪抵抗ETEC感染的保护作用,在连续饲喂21 d后,每个重复中随机挑选1头体况相近的断奶仔猪灌服ETEC。结果发现,与培养基组相比,攻菌后重组屎肠球菌组断奶仔猪血清白细胞介素-2(IL-2)、免疫球蛋白G (IgG)含量、肠黏液中分泌型免疫球蛋白A (sIgA)水平均显著升高(P<0.05);脾脏指数显著提高(P<0.05),但胸腺指数、肠段长度及重量则均无显著差异(P>0.05)。综上所述,表达猪乳铁蛋白肽的重组屎肠球菌能够起到促进断奶仔猪生长及抗ETEC感染的保护作用。  相似文献

The modification of flotation method for the examination of diarrhoeic piglet faeces for the detection of Isospora suis oocysts was elaborated. The method was based on removing fractions of fat from the sample of faeces by centrifugation with a 25% Percoll solution. The investigations were carried out in comparison to the McMaster method. From five variants of the Percoll flotation method, the best results were obtained when 2ml of flotation liquid per 1g of faeces were used. The limit of detection in the Percoll flotation method was 160 oocysts per 1g, and was better than with the McMaster method. The efficacy of the modified method was confirmed by results obtained in the examination of the I. suis infected piglets. From all faecal samples, positive samples in the Percoll flotation method were double the results than that of the routine method. Oocysts were first detected by the Percoll flotation method on day 4 post-invasion, i.e. one-day earlier than with the McMaster method. During the experiment (except for 3 days), the extensity of I. suis invasion in the litter examined by the Percoll flotation method was higher than that with the McMaster method. The obtained results show that the modified flotation method with the use of Percoll could be applied in the diagnostics of suckling piglet isosporosis.  相似文献

To prevent enterotoxigenic Escherichia coli (ETEC) induced postweaning diarrhoea, the piglet needs an active mucosal immunity at the moment of weaning. In the present study, the feasibility of oral vaccination of suckling piglets against F4+ETEC infection with F4 fimbriae was studied. Furthermore, oral vaccination with enteric-coated pellets of F4 fimbriae was compared to vaccination with F4 fimbriae in solution. Therefore, piglets were orally administered 1mg F4 fimbriae in pellets or in solution during three successive days at the age of 7 and 21 days, whereas control piglets were not vaccinated. Five days postweaning (33 days of age), all animals were orally challenged with F4+ETEC. Despite the induction of an immune response upon oral administration of both F4 fimbriae in pellets as in solution, the colonisation of the small intestine by F4+ETEC upon oral challenge could not be prevented. However, a marginal but significant reduction in F4+ E. coli faecal excretion was found in the piglets vaccinated with F4 fimbriae in pellets, indicating that the use of an enteric-coat which protects the F4 fimbriae against inactivation by milk factors and degradation by enzymes and bile improves vaccination.  相似文献

Little is known about the sources and kinetics of enterotoxigenic Escherichia coli colonization in pigs during the pre-and post-weaning period. In this study, farrowing pens, sows, and piglets were tested for the presence of E. coli O149 by real-time polymerase chain reaction (PCR) after bacterial culture pre-enrichment on 2 farms, one with a history of post-weaning diarrhea (problem farm - PF) and the other without such a history (non-problem farm - NPF). Unlike those on the PF, the sows from the NPF did not carry E. coli O149 before parturition, although they were colonized to frequencies similar to animals on the PF soon afterwards. Most piglets from the NPF were colonized within a week after birth, whereas only a small proportion of those on the PF were colonized during that period. No difference was observed in the frequency of piglet colonization at the 2 farms either at weaning or during the following week. Post-weaning diarrhea (PWD), which is caused by enterotoxigenic E. coli (ETEC), is a multifactorial disease. The presence of ETEC alone is not always sufficient for the disease to develop. Many other factors are considered to be associated with the occurrence of PWD, including feed type (1,2), feeding regimen (1,3,4), the presence of other infectious agents (3,5), weaning age, and weight (6). Weaning, which is considered to be a major physiological and psychological stress factor, is critical for the disease to occur (7). Although piglets are already colonized with ETEC before weaning (4,8), on many farms, clinical disease occurs only after weaning (1). Both sows (9,10) and the environment (6) could be possible sources of infection for piglets, but results from previous studies have not resolved this issue because of the low sensitivity of ETEC detection methods. This study provides preliminary data based on a sensitive detection method for E. coli O149 in pigs and their environment. The results demonstrate the potential of real-time PCR for future studies on this topic.  相似文献

51 gnotobiotic and 63 conventional, one-, or two-days-old piglets were divided into five groups and infected orally either with Isospora suis or rotavirus alone, or with both agents simultaneously or successively with alternative sequences and various intervals. 15 gnotobiotic and 10 conventional piglets served as controls. The development of small intestinal lesions after infection with I. suis was biphasic. The dominant alteration resulting from rotavirus infection was villus atrophy, considerably more pronounced and extensive in gnotobiotic than in conventional piglets. Synergistic action of I. suis and rotavirus was manifested both clinically, and morphologically. This action culminated at the time of the actual, or presumed development of merogony of I. suis, i.e. on DPI 3 to 5. The action develops only if the intestinal epithelium is damaged functionally and morphologically by a preceding rotavirus infection. It is concluded that the synergistic action is based on a competition of rotavirus and I. suis for mature, enzymatically active absorptive cells.  相似文献

In two studies carried out in 1974-75 on 10 eight and 25 two week old piglets obtained from a SPF-herd the endoparasitic status was determined. Samples of faeces were examined by microscopy which revealed the presence of Isospora suis, a finding that was quite unexpected in a SPF-herd. During days 17-23 pp. the 25 piglets were not found to excrete oocysts, the earliest findings of oocysts being made 24-35 days p.p. The 10 piglets being kept isolated in individual pens excreted oocysts up until 88 days p.p. The epidemiology of Isospora suis is discussed.  相似文献

Laboratory surveillance data from the Animal Health Laboratory, University of Guelph, on the etiological diagnoses of neonatal diarrhea in piglets were analyzed to determine the relative importance and trends of different enteric pathogens in Ontario. A total of 237 cases, including live and dead 1- to 7-day-old piglets, were submitted for diagnosis of gastrointestinal illness between 2001 and 2010. The combined frequencies for cases of gastrointestinal illness involving Escherichia coli, Clostridium perfringens type A, rotavirus, and Clostridium difficile, either as single pathogens or a complex of pathogens, accounted for 56% of the total cases. In a total of 33% of cases of gastrointestinal illness, an etiological agent was not identified. The frequency of cases diagnosed with enterotoxigenic E. coli was decreased from 2007. Cases submitted in 2010 were more likely to be diagnosed with C. perfringens type A compared to cases submitted in 2002 to 2007 (P < 0.05). There was a significant trend for cases submitted in the winter to be diagnosed with C. perfringens type A, enterotoxigenic E. coli, rotavirus, and Cystoisospora suis (formerly Isospora suis) (P < 0.05). Enterotoxigenic E. coli was less likely diagnosed if C. difficile, C. perfringens, or rotavirus were detected (P < 0.05). Younger piglets were more likely to be diagnosed with C. perfringens type A (P < 0.05) and C. difficile (P < 0.05) than older piglets. This study shows that E. coli, C. perfringens type A, rotavirus, and C. difficile are enteric pathogens of concern for Ontario swine farrowing operations and further research is required to understand the reasons for the cases that are not diagnosed.  相似文献
