热应激对荷斯坦种公牛精液品质的影响及机理   总被引:10,自引:0,他引:10  
用5头荷斯坦种公牛研究了热应激对其生理常值,精液品质,血清生化指标,精清生化指标的影响。结果表明:(1)热应激可造成种公牛精液品质显著下降,使原精活力,冻精活动,精子密度,活精子百分数和顶体完整率分别比春季下降3.13%(P<0.01),1.11%(P<0.01),34.8%(P<0.01),15.0%(P<0.01)和17.8%(P<0.01),精子畸形率比春季上升25.5%(P<0.01)。(2)夏季慢性热应激使荷斯坦种公牛的直肠温度和呼吸频率极显著高于春,米,立3个非热应激季节(P<0.01)。(3)夏季荷斯坦种公牛血清睾酮,T3,皮质醇和促黄体素的含量分别为3.41ng/L,1.29ng/L,5.05ng/L和45.11ng/L,与非热应激季节相比,均处于较低的水平。(4)热应激可降低血糖和血清钠,钾,钙,镁水平(P<0.01),使血清GPT,GOT处于最高水平(P<0.01)。(5)夏季荷斯坦公牛精清睾酮含量仅为4.31ng/L,极显著低于非热应激季节(P<0.01)。(6)热应激造成荷斯坦种公牛精清钾,钙,镁处于最低水平(P<0.01),精清ALP,ACP处于最高水平(P<0.01),使精清钠由春季至夏季呈显著下降趋势。上述结果显示:热应激使体内微环境发生了显著变化,抑制了下丘脑-垂体-睾丸轴的功能,从而影响了精子的生成和贮存。  相似文献

本试验选择16头荷斯坦种公牛随机分为2组,对照组饲喂基础日粮,试验组在基础日粮中添加胡萝卜,试验期为75d,观察对荷斯坦种公牛原精产量、精子密度、精子活力和制冻精数量的影响。结果表明:在荷斯坦种公牛日粮中添加胡萝卜后,试验组牛的原精产量、鲜精精子活力和制冻精数量都极显著高于对照组(P〈0.01);精子密度与对照组间差异不显著(P〉0.05)。本试验结果验证了在荷斯坦种公牛日粮中添加胡萝卜可显著提高精液产量、精子活力和制冻精数量,提高精液品质。  相似文献

研究按季节和品种对种公牛采精记录进行方差分析,分别比较了季节对夏洛来、西门塔尔、利木赞、红安格斯、荷斯坦种公牛和弗莱维赫6个品种鲜精液品质的影响以及相同季节下的6个品种精液品质间的差异。分析了在季节和品种的双重条件下,精液品质间的相关性。结果表明:(1)西门塔尔、夏洛来和荷斯坦种公牛的单次射精量随季节变化排序均依次是:夏季秋季春季冬季。(2)西门塔尔、夏洛来、利木赞和红安格斯的鲜精子密度随季节变化排序依次是:春季夏季冬季秋季,且这四个品种鲜精子密度在四季的排序均为:西门塔尔夏洛来利木赞红安格斯。(3)各品种种公牛四季鲜精子活力排序均依次是:冬季春季秋季夏季;各品种种公牛鲜精子活力在四季的排序均为:利木赞夏洛来弗莱维赫、西门塔尔荷斯坦红安格斯;各品种夏季鲜精子活力均极显著低于其它三个季节(P0.01)。(4)荷斯坦种公牛鲜精子活力夏季占春季活力比率最高为(0.54/0.59),利木赞次之(0.5/0.63),弗莱维赫第三(0.47/0.6),安格斯最低(0.34/0.49)。(5)红安格斯的单次射精量、精子密度和精子活力在相同季节下都低于其它5个品种。(6)在不同季节不同品种下,种公牛的射精量与鲜精子密度和鲜精子活力的相关系数分别是0.006和~0.019,鲜精子密度和鲜精子活力的相关系数是0.253。  相似文献

为探讨季节对不同品种种公牛精液品质的影响,实验采集了中国荷斯坦牛、西门达尔牛和日本和牛的种公牛精液周年生产数据,进行分析比较。结果表明:荷斯坦牛春、冬季原精活力达标率显著高于夏季(P0.05),冬季极显著高于夏季(P0.01);冬、秋季冻精活力达标率显著高于夏季(P0.05),秋季极显著高于春、夏(P0.01),春、冬季射精量显著高于秋季(P0.05),春季极显著高于秋季(P0.01);春、秋、冬季精液密度显著高于夏季(P0.05);西门达尔牛在春季原精活力达标率呈现最高,显著高于夏、秋季(P0.05);日本和牛冬季原精活力和冻精活力均最高,射精量四季之间无显著差异(P0.05),夏季精液密度显著高于春季(P0.05),极显著高于秋、冬季(P0.01)。春、冬季种公牛原精和冻精的达标率最佳;肉牛品种精液品质优于奶牛品种,肉牛品种中的西门达尔种公牛的精液品质优于和牛。  相似文献

本试验选择亚热带气候条件下广州地区的娟姗公牛和荷斯坦公牛各5头,比较两个品种公牛的精液品质(采精量、原精密度、原精活力、细管精液产量、冻后活力、低渗膨胀率及穿透率)。研究表明,荷斯坦公牛每次采精的采精量(16.14±0.06 mL)和细管精液产量(189.17±3.11支)都极显著地高于娟姗公牛(4.74±0.05 mL,158.46±2.64支)(P<0.01);娟姗公牛的原精密度(8.95±0.08亿/mL)极显著地高于荷斯坦公牛(8.32±0.07亿/mL;P<0.01);娟姗公牛原精活力(0.731±0.004)高于荷斯坦公牛(0.729±0.003),但两者差异不显著(P<0.05);娟姗公牛精液的冻后活力(0.355±0.003)极显著高于荷斯坦公牛(0.339±0.003;P<0.01);娟姗公牛冷冻精液的低渗膨胀率(34.50%±0.49%)显著高于荷斯坦公牛(31.21%±0.59%;P<0.01);娟姗公牛冷冻精液对去透明带仓鼠卵的穿透率(84.51%±13.83%)显著高于荷斯坦公牛(81.52%±6.13%;P<0.05)。  相似文献

在秋季驱虫季节,选择荷斯坦(Holstein)、夏洛来(Charolais)、利木赞(Limousin)、西门塔尔(Simmental)4个品种种公牛各10头,将其分为两组,分别采用阿维菌素背部浇泼和伊维菌素肌肉注射两种给药方法,研究不同驱虫药和方法对种公牛精子质量的影响。结果与投药前相比,伊维菌素注射法驱虫可使种公牛精液活力、密度、活精子百分数、顶体完整率平均下降14.3%(p<0.01)、28.4%(p<0.01)、21.0%(p<0.01)和19.5%(p<0.01),精子畸形率上升30.8%(p<0.01),总冻精产量由给药前2个月的69820支下降到给药后2个月的65540支;阿维菌素浇泼法驱虫可使种公牛精液活力、密度、活精子百分数、顶体完整率平均下降1.6%(p>0.05)、5.4%(p>0.05)、9.4%(p<0.05)和5.0%(p>0.05),精子畸形率上升15.8%(p<0.01),总冻精产量由投药前2个月的70370支上升到投药后2个月的71870支。两种驱虫方法对种公牛精液品质的影响因品种不同而异,从总的趋势看,无论是阿维菌素浇泼法还是伊维菌素注射法,对荷斯坦种公牛精液品质的影响小于西门塔尔、夏洛来和利木赞种公牛。  相似文献

奶牛育种是一项基础性、长期性的工作,但效益非常显著。根据美国农业部数据,育种在饲料营养、繁殖育种、疾病控制、饲养管理等主要技术因素中,对奶牛生产的贡献比例最大,达到40%。种公牛耐寒、怕热,最适宜生活的环境温度为11~18 ℃,外界气温的变化对种公牛的影响较大,特别是上海夏季高温、高湿严重,极易造成种公牛采食量下降、体温升高、精液品质和产量下降。本研究对2014年上海奶牛育种中心公牛站10 头荷斯坦种公牛精液精子畸形测定资料与上海地区气温变化情况进行分析,得出高温对种公牛冷冻精液精子畸形率影响较大,气温越高,精子畸形率越高的结论。春季和夏季公牛精子畸形率差异显著(P<0.01),种公牛冷冻精子畸形率在2—4月最低,7—9月最差,因此采精最佳季节为10月至翌年6月。  相似文献

采用三个不同品种公牛研究了青海气候条件下热应激对其精液品质的影响。结果表明:三个不同品种公牛在热应激状态下,新西兰荷斯坦公牛(HS)原精精液量、精子密度显著降低(P〈0.05)、原精活力及冻后活力极显著降低(P〈0.01);德国西门塔尔公牛(XM)的原精精液量、精子密度极显著降低(P〈0.01),原精活力及冻精品质无明显变化(P〉0.05);澳洲矮脚公牛(LL)的原精精液量、精子密度、原精活力、冻精品质却显著提高(P〈0.05)。  相似文献

本研究比较了经免疫注射口蹄疫疫苗后两个品种共13头种公牛的鲜精精液品质。结果表明,荷斯坦牛和西门塔尔牛免疫期的精液量和精液密度指标无明显变化(P>0.05);活力指标显著降低(P<0.05);精子畸形率显著增加(P<0.05)。尾部畸形增加是影响精子直线运动和冲力的主要原因。  相似文献

以利木赞和夏洛来种公牛为试验牛,研究不同季节对其精液品质、冻精产量、生理常值、血清及精清生化指标的影响。结果表明:(1)与其它季节相比,夏季肉用种公牛的原精活力、冻精活力、活精于百分数、顶体完整率均显著降低,而精于畸形率显著升高(P〈0.05)。精子密度以秋季最低。(2)冻精产量夏季处于最低水平,平均每月仅为7388支。(3)夏季肉用种公牛的血清LH、睾酮、T3、皮质醇的含量分别为48.12、4.15、1.11、4.89ng/L,显著低于春、秋、冬季(P〈0.01)。(4)夏季肉用种公牛血清钠、钾、钙、镁均处于最低水平,显著低于其它季节(P〈0.01)。(5)夏季肉用种公牛精清睾酮水平明显下降,显著低于春季、秋季和冬季。(6)精清钠、钾、钙夏季含量最低,分别为83.12mmol/L、18,77mmol/L和4.63mmol/L;精清镁含量由春季至秋季呈显著下降趋势;精清磷含量夏季最高,为4.65mmol/L,显著高于春季、秋季和冬季。(7)肉用种公牛精清ALP夏季含量为1784.3U/L,显著高于春、秋两季,但低于冬季。(8)精清睾酮与精液品质各指标存在显著相关;血清生化指标与精清生化指标存在显著相关;直肠温度、呼吸频率与精液品质及血清生化指标存在显著相关性,与精清生化指标相关性不大。  相似文献

The study was designed to evaluate the influence of season on semen characteristics and seminal plasma protein profile of buffalo bull semen. Thirty‐six ejaculates were collected in three seasons (winter, summer and rainy) from six adult Bhadawari bulls, and semen characteristics were evaluated immediately after collection. The seminal plasma was harvested by centrifugation and protein profiling, and percentage protein fractions were analysed by SDS‐PAGE. The significant effect of season was observed on ejaculate volume, sperm concentration, progressive motility, percentage live spermatozoa, hypo‐osmotic swelling test (HOST) and acrosomal integrity. The electrophoretogram of seminal plasma proteins revealed 20 protein bands in winter, 23 bands in rainy and 25 bands in summer seasons, illustrating the significant effect of seasons on seminal plasma proteins. Among these protein bands, 18 bands were observed common in semen samples of all three seasons while protein bands of 46, 55, 58, 144 and 160 kDa were found in rainy and summer seasons. The protein bands of 48 and 60 kDa were observed only in winter season, whereas 184 and 200 kDa were reported in summer season only. The protein fractions (protein%) of common protein bands observed in three seasons revealed a significant effect of season on protein bands of 24.5, 66, 70, 72, 84 and 86 kDa. From the study, it was pertinent that bull seminal plasma contains specific proteins in particular season, which may be associated with some of the semen characteristics, and these proteins could be used as markers of the semen quality of buffalo bulls.  相似文献

为探讨荷斯坦牛和西门塔尔牛冻精的精液品质及体外受精后胚胎发育能力的差异,利用目测法、低渗膨胀法和考马斯亮蓝染色法评估了荷斯坦牛和西门塔尔牛冻精的活力、质膜完整率和顶体完整率,并比较了二者冻精体外受精后胚胎的卵裂率和囊胚率。结果表明,荷斯坦牛和西门塔尔牛冻精的活力(30.4%和27.2%)、质膜完整率(41.96%和36.22%)和顶体完整率(77.02%和73.02%)均无显著差异(P>0.05),但荷斯坦牛冻精体外受精后的卵裂率(57.5%和48.6%)和囊胚率(30.3%和23.2%)显著高于西门塔尔牛冻精(P<0.05)。提示,不同品种公牛精液体外受精后的发育能力有显著差异(P>0.05)。  相似文献

Peroxidation damage to spermatozoa and seminal plasma has an important role in sperm quality. Thus, the objective of this study was to determine the levels of lipid and protein oxidation in spermatozoa and seminal plasma of Asian elephants (Elephas maximus) with varying percentage of progressive motility. Lipid and protein oxidation was measured by the thiobarbituric acid‐reactive species (TBARS) assay and the 2, 4‐dinitrophenylhydrazine (DNPH) carbonyl groups assay, respectively. Fresh semen samples were collected from Asian elephants and classified according to the percentage of motile spermatozoa into good (>60%) and poor (≤20%) motility. Results revealed that seminal plasma malondialdehyde (MDA) and seminal plasma protein carbonyls (PCs) were significantly higher in poor motility than in good motility (p < .05). The MDA and PC levels in seminal plasma were negatively correlated with the percentages of progressive motility (p < .05). In addition, the negative correlation between sperm concentration and seminal plasma MDA level was investigated (p < .05). The sperm viability was also negatively correlated with sperm PC level (p < .05). This study indicated that lipid and protein oxidation has deleterious effect on semen quality of Asian elephants.  相似文献

人工授精技术是目前通过冷冻精液进行牛遗传改良而应用最为广泛的生物技术。虽然如此,由于精液产品的质量而导致人工授精的遗传影响被限制,在精液的冷冻-解冻过程中40%~50%的活精子失去完整性或功能受到损伤。作者在参阅国内外相关文献的基础上,通过阐述冷冻-解冻对精子细胞的损伤作用及机理、精浆对牛冷冻精液质量的影响、冷冻-解冻过程中精子的过氧化损伤,以及冷冻-解冻后精子相关基因与蛋白质的变化等,以期为牛高质量冷冻精液的研究提供一定参考,进一步提升冷冻精液质量。  相似文献

本试验旨在研究不同精液品质特征长白公猪血清和精浆中元素含量的差异,分析血清和精浆中元素含量对精液品质的影响。基于107头长白公猪的1402次精液品质记录,将公猪群按照精液可利用率划分为3组:低利用率组(利用率<60%,n=21)、中等利用率组(60%≤利用率≤80%,n=27)和高利用率组(利用率>80%,n=59)。采集每头长白公猪的血清和精浆样品,采用电感耦合等离子体质谱法检测血清和精浆中营养元素和毒性元素含量。结果表明:1)低利用率组长白公猪的有效精子数和精子活力显著低于中等利用率和高利用率组(P<0.01),精子畸形率显著高于中等利用率和高利用率组(P<0.01)。2)不同利用率组间长白公猪的血清和精浆元素含量无显著差异(P>0.05);但在血清和精浆元素含量与精液品质参数相关性分析中发现,精浆铅元素含量与精子活力呈显著负相关(P<0.05),与精子畸形率呈显著正相关(P<0.05)。3)对精浆铅含量分组进一步分析发现,精浆铅含量为0μg/L时长白公猪的精子畸形率显著低于精浆铅含量>10.0μg/L时(P<0.05),精子畸形率降低约6.11%。总的来说,长白公猪精浆中毒性元素铅的存在会通过损害精子活力和形态,影响公猪精液品质。  相似文献

The present study compared the quality of sperm collected by artificial vagina or pharmacologically induced ejaculation from a 10-year-old thoroughbred stallion with seminal vesiculitis. The pharmacological protocol involved intravenous administration of detomidine (0.01 mg/kg) and oxytocin (20 IU) and successfully induced ejaculation in all attempts of semen collection. Sperm motility, plasma membrane and acrosome integrity (PMAI), reactive oxygen species (ROS) levels, polymorphonuclear neutrophil (PMN) percentage, and bacterial profiles of fresh and cooled semen (5°C for 24 hr) were evaluated. Semen obtained by the pharmacological method presented reduced seminal volume, decreased PMN percentage and superior sperm motility in cooled samples. Moreover, higher PMAI and lower ROS levels were observed in semen collected by the pharmacological method. Therefore, pharmacologically induced ejaculation is an alternative to obtain semen with minimal contamination and with sperm of superior quality and longevity from stallions with seminal vesiculitis.  相似文献

Basic characteristics of European bison (Bison bonasus) semen were described and the efficacies of two extenders-Triladyl, containing egg yolk, and a synthetic extender, containing soybean lipids-were tested for semen cryopreservation. Seven ejaculates were collected by electroejaculation from a 10-yr-old, European bison bull. Each ejaculate was diluted at 37 degrees C to a final concentration of 200 x 10(6) sperm/ml with Triladyl or the synthetic extender. Extended semen samples were frozen according to a standard bull semen freezing protocol. After 2 wk of storage, one straw from each extender and ejaculate was thawed, and postthaw quality was evaluated by individual sperm motility and movement rate, numbers of sperm morphologic abnormalities and intact acrosomes, functional integrity of the sperm membranes determined by hypoosmotic swelling test (HOST), viability (live-dead, eosin-nigrosin stain), and a heterologous in vitro sperm penetration assay (SPA). A total of 600 in vitro-matured bovine oocytes were inseminated with 1 X 10(6) spermatozoa of Holstein semen frozen-thawed in Triladyl (control) or of European bison semen frozen in Triladyl or the synthetic extender. Nuclear status of the oocytes was determined after 18 h of sperm-oocyte coincubation. Extender had no effect on any evaluated parameters of semen after dilution and cooling (4 hr at 5 degrees C) or in postthaw individual motility, quality of movement, and sperm morphology. However, significantly (P < 0.05) higher numbers of spermatozoa with intact acrosomes, intact membranes (HOST), and viable sperm (P < 0.01) were in semen frozen in Triladyl than in the synthetic extender. Mean values for heterologous SPA for bull (control) and for bison semen frozen in the synthetic extender were very much alike-63.3+/-10.6% and 63.1 +/- 15.9%, respectively; bison semen frozen in Triladyl was lower, 43.0+/-24.2% but not significantly different. Cumulative results from a variety of viability assays of diluted/cooled and frozen-thawed semen, including the heterologous SPA, suggest that European bison semen can be successfully frozen in both extenders tested in this study.  相似文献

Kjxstad, H., E. Ropstad and K. Andersen Berg: Evaluation of spermatological parameters used to predict the fertility of frozen bull semen. Acta vet. scand. 1993,34,299-303.– Post-thaw motility, velocity and acrosome integrity of frozen semen were determined in 18 bulls with varying fertility (average non-return rates: 71.3 (± 2.8) - range: 65.2-75.7). Five semen straws were investigated from each bull. The average values for sperm motility (percentage motile spermatozoa), sperm velocity (graded from 0-3) and acrosome integrity (proportion of spermatozoa with intact acrosome) were 67.5%, 2.5 and 79.3%, respectively. Significant correlations were found between sperm motility and velocity, but not between sperm motility and acrosome integrity. Both sperm motility and velocity were significantly related to bull fertility. It was concluded that of the post-thaw semen characteristics investigated in this study these 2 parameters provided a reliable basis for prediction of bull fertility.  相似文献
