Channel catfish virus (CCV) causes an acute haemorrhagic disease in channel catfish, Ictalurus punctatus (Rafinesque), fry and fingerlings. The present study describes a polymerase chain reaction (PCR)-based assay for detection of CCV DNA in the tissues of acutely infected juvenile catfish. The assay is rapid, sensitive and specifically detects CCV DNA derived from epidemiologically distinct viral isolates. The use of two independent PCR primers sets, each specific for particular CCV genes (open reading frames 8 and 59), provides a means to confirm the results and minimize false-positive results. The method identifies CCV DNA in several tissues of acutely infected fish, including the brain, blood, intestine, kidney and liver. The CCV PCR assay is useful for the diagnosis of acute CCV disease and for studies to investigate the molecular basis of CCV pathogenesis.  相似文献

为构建基于杆状病毒表达系统的CCV新型亚单位疫苗,将CCV的囊膜蛋白基因ORF6克隆至杆状病毒转座载体pFastBacTM 1质粒中,并将阳性重组转座质粒转化进含穿梭载体Bacmid的感受态细胞DH10Bac中,获得重组子rBacmid-ORF6。在脂质体介导下将该重组子转染sf9昆虫细胞,获得重组杆状病毒AC-ORF6。AC-ORF6感染的sf9昆虫细胞,经超薄切片电镜观察,可见重组杆状病毒呈多粒包埋,经间接免疫荧光、Western-blotting检测,CCV的ORF6蛋白可以在感染了AC-ORF6的sf9细胞中表达。研究表明,获得了插入ORF6基因的重组杆状病毒,并且该基因可以在重组杆状病毒介导下在昆虫细胞中表达,从而为基于CCV ORF6的杆状病毒亚单位疫苗研究奠定了基础。  相似文献

Abstract. Five species of fingerling fish, channel catfish, Ictalurus punctatus , tilapia, Sarotherodon aureus , golden shiner, Notemigonus crysoleucas , largemouth bass, Micropterus salmoides and bighead carp, Aristichthys nobilis , were tested to determine their susceptibility to the bacterium, Edwardsiella ictaluri , at 26°C, Channel catfish demonstrated high susceptibility to E. ictaluri as 100% of those fish injected with 1.5 × 103 cells died within 10 days. Tilapia demonstrated slight susceptibility to the pathogen while golden shiner, bighead carp and largemouth bass were not susceptible. E. ictaluri was isolated from a higher percentage of peritoneal cavities, livers and kidneys of channel catfish than of other species. Sequential growth of E. ictaluri in the liver of channel catfish is described.  相似文献

Abstract Two groups of European catfish, Silurus glanis L., fingerlings were infected with channel catfish virus (CCV) by either intraperitoneal injection with 105 TCID50 of CCV, or bathing in water containing 105 TCID50 of CCV per 1·0 ml. The virus was isolated from spleen, intestine and brain of CCV-injected fish at day 1 and the titres ranged from 102·1 to 103·3 TCID50/g. However, the tissue distribution of CCV was irregular and no virus was isolated after day 3 post-exposure. In CCV-bathed fish, the virus was isolated only from the liver of one specimen at day 3 post-exposure. No clinical signs of CCV disease developed in any of the fish. Specimens in each regime from all sampling periods showed some minor histopathological changes, but there were no differences between treatments. Lesions included oedema and focal haemorrhage in the liver and the spleen was congested. Electron micrographs of tissue samples showed the presence of a few virus particles around the nuclei of kidney, spleen and intestinal cells, and in or around a myelinated nerve within the optic lobes of infected fish during the first 4 days of infection.  相似文献

ABSTRACT:   Prey fish selection by Far Eastern catfish and largemouth bass were examined using bluegill, Japanese dace and crucian carp as food fish. In both aquaria (1.2 m × 0.45 m) and ponds (2.8 m × 1.2 m), bluegill was not preyed on by catfish and bass more than dace and crucian carp. In aquaria, there was no significant difference in the consumption of dace and crucian carp between catfish and bass, but in ponds, catfish preyed on dace and crucian carp more and less than bass, respectively. In the case that only bluegill and catfish were introduced in ponds, catfish consumed 4–15 g of bluegill per day. The introduction of catfish into ponds and lakes for the purpose of eradicating bluegill is considered appropriate for areas with few native fish species.  相似文献

Abstract. Two populations of channel catfish were examined for the presence of channel catfish virus (CCV) by use of a nucleic acid probe. In one population of 22 fish with no history of CCV, viral DNA was found in every liver. These fish had previously been examined by a technique involving co-cultivation of their leucocytes with catfish tissue culture cells. The co-cultivation method had identified virus in 10 of these fish. The second fish population consisted of 14 adults that had survived a CCV outbreak in 1980. Of the 14 fish, 11 showed positive indication of CCV DNA. The tissue distribution of the CCV differed from fish to fish. All fish from the first group and one fish from the second group showed some alterations in the DNA banding patterns expected from pure CCV DNA. This might be indicative of modifications in the genomic structure of the CCV DNA when the virus is latent in a fish.  相似文献

Abstract  Channel catfish, Ictalurus punctatus (Rafinesque), are commonly stocked into small impoundments that contain bluegill, Lepomis macrochirus Rafinesque, and these species may compete for food resources because both prey on macroinvertebrates. Prey selectivity and diet overlap of sympatric bluegill and channel catfish were evaluated in two small impoundments. Both fish species fed primarily on macroinvertebrates; but diet overlap between the species generally was not extensive because channel catfish consumed a more diverse array of foods, including more crayfish, fish and plant material. The use of foods other than macroinvertebrates increased as channel catfish grew larger. Bluegill also ingested large quantities of plant material at times, and ingestion of plants increased with fish size. Results of this study suggest that food competition between bluegill and channel catfish may occur when small channel catfish are abundant because, like bluegill, they feed almost entirely on macroinvertebrates and may reduce macroinvertebrate abundance. Thus, overstocking channel catfish in small impoundments managed for bluegill should be avoided.  相似文献

A continuous cell line [goldfish tail fin (GFTF)] derived from a goldfish tail fin, Carassius auratus, was established and characterized. GFTF cells predominantly consist of fibroblast-like cells that were maintained and subcultured more than 50 times over a period of 15 months. Cells grew at temperatures between 15 and 37°C, with an optimum temperature of 25°C. The growth rate of GFTF cells increased proportionally with the foetal bovine serum (FBS) concentration (5-20%), with optimum growth at 20% FBS. The chromosome numbers were 88-112, with a modal peak of 104 chromosomes. Five known fish viruses were tested to determine susceptibility. Results demonstrated that GFTF is susceptible to snakehead rhabdovirus, spring viraemia of carp virus and channel catfish virus (CCV). In addition, GFTF demonstrated a higher sensitivity to, and increased viral production of, CCV than that observed in the control cell line, channel catfish ovary cells. This suggests that GFTF cells would be useful as a diagnostic tool for viral diseases in this fish species, as well as for the isolation and study of goldfish viruses in the future. Furthermore, these cells were transfected with pEGFP-N1 vector DNA and some fluorescent signals were observed, suggesting that GFTF cells could be a useful tool for transgenic and genetic manipulation studies.  相似文献

With a well‐developed olfactory system and taste buds located inside the mouth and on the external body surface, catfishes are excellent subjects to investigate nonconsumptive predator effects. Juvenile channel catfish Ictalurus punctatus are often eaten by largemouth bass Micropterus salmoides, and we tested the hypothesis that largemouth bass fed catfish would adversely affect foraging and growth in channel catfish. Groups of catfish were placed in tanks partitioned by a mesh screen: experimental tanks had a largemouth bass on the other half of the tank, and control tanks had an empty chamber. Experimental catfish exhibited a long‐term decrease in spontaneous motion, feeding and growth. Feeding catfish to the bass caused the experimental catfish to freeze after a multi‐second latency suggesting transport of an alarm cue. Thus, there were long‐term effects from the continuous presence of the bass and additional short‐term cues from the bass consuming a catfish.  相似文献

A nucleic acid probe for channel catfish virus (CCV) was constructed using recombinant DNA techniques. This probe consisted of a specific viral DNA fragment generated by digestion of CCV DNA with the restriction enzyme EcoRI. The probe was used to examine DNA isolated from tissues of fish that had been injected with CCV. Viral DNA was detected in some tissues of various injected fish. The sensitivity limit of detection was determined to be one viral DNA per cell.  相似文献

Sixteen countries, including Bangladesh, have reported the presence of tilapia lake virus (TiLV), an emerging tilapia pathogen. Fish polyculture is a common farming practice in Bangladesh. Some unusual mortalities reported in species co-cultivated with TiLV-infected tilapia led us to investigate whether any of the co-cultivated species would also test positive for TiLV and whether they were susceptible to TiLV infection under controlled laboratory experiments. Using 183 samples obtained from 15 farms in six districts across Bangladesh, we determined that 20% of the farms tested positive for TiLV in tilapia, while 15 co-cultivated fish species and seven other invertebrates (e.g. insects and crustaceans) considered potential carriers all tested negative. Of the six representative fish species experimentally infected with TiLV, only Nile tilapia showed the typical clinical signs of the disease, with 70% mortality within 12 days. By contrast, four carp species and one catfish species challenged with TiLV showed no signs of TiLV infection. Challenged tilapia were confirmed as TiLV-positive by RT-qPCR, while challenged carp and walking catfish all tested negative. Overall, our field and laboratory findings indicate that species used in polycultures are not susceptible to TiLV. Although current evidence suggests that TiLV is likely host-specific to tilapia, targeted surveillance for TiLV in other fish species in polyculture systems should continue, in order to prepare for a possible future scenario where TiLV mutates and/or adapts to new host(s).  相似文献

The koi herpesvirus (KHV) has spread worldwide since its discovery in 1998 and causes disease and mortality in koi and common carp populations with a high impact on the carp production industry. Many investigations have been conducted to examine ways of distribution and to identify possible transmission vectors. The answers, however, raise many new questions. In the present study, different wild fish species taken from carp ponds with a history of KHV infection were examined for their susceptibility to the virus. In the tissue of these fish, the virus load was determined and it was tested whether a release of the virus could be induced by stress and the virus then could be transferred to naive carp. Wild fish were gathered from carp ponds during acute outbreaks of virus‐induced mortality in summer and from ponds stocked with carp carrying a latent KHV infection. From these ponds, wild fish were collected during the harvesting process in autumn or spring when the ponds were drained. We found that regardless of season, temperature variation, age and infection status of the carp stock, wild fish from carp ponds and its outlets could be tested positive for the KHV genome using real‐time PCR with a low prevalence and virus load. Furthermore, virus transfer to naive carp was observed after a period of cohabitation. Cyprinid and non‐cyprinid wild fish can therefore be considered as an epidemiological risk for pond carp farms.  相似文献

Abstract  Standard methods for comparing population characteristics within and among fish populations greatly enhance communications among fisheries scientists, improve the efficiency of data analysis, and provide insight that helps guide management actions. Although standard methods are available for comparing some fish population characteristics (e.g. length structure, body condition), similar methods are lacking for comparing growth. The purpose of this study was to provide standards (i.e. percentiles and a standard growth model) for nine ecologically and recreationally important fish species. Percentile distributions of mean back-calculated length at age were estimated using data obtained from the published literature and from data solicited from state and federal agencies throughout North America. Percentiles of growth were estimated for bluegill, Lepomis macrochirus Rafinesque, common carp, Cyprinus carpio Linnaeus, flathead catfish, Pylodictis olivaris (Rafinesque), largemouth bass, Micropterus salmoides (Lacepède), sauger, Sander canadensis (Griffith & Smith), smallmouth bass, Micropterus dolomieu Lacepède, white bass, Morone chrysops (Rafinesque) and yellow perch, Perca flavescens (Mitchill). Standard growth models (i.e. von Bertalanffy models) were developed for these species and for channel catfish, Ictalurus punctatus (Rafinesque). These results provide tools that will help scientists compare growth of fishes across North America.  相似文献

Channel catfish, Ictalurus punctatus (Rafinesque), populations in six lakes in northern Idaho, USA, were sampled to describe their population characteristics. During the summers of 2011 and 2012, 4864 channel catfish were sampled. Channel catfish populations had low to moderate catch rates, and length structure was dominated by fish <400 mm. Channel catfish were in good body condition. All populations were maintained by stocking age‐1 or age‐2 fish. Growth of fish reared in thermally enriched environments prior to stocking was fast compared to other North American channel catfish populations. After stocking, growth of channel catfish declined rapidly. Once stocked, cold water temperatures, prey resources and (or) genetic capabilities limited growth. Total annual mortality of age 2 and older channel catfish was generally <40%. Tag returns indicated that angler exploitation was low, varying from 0 to 43% among lakes. This research provides insight on factors regulating channel catfish population dynamics and highlights important considerations associated with their ecology and management.  相似文献

Mycobacteriosis in fish can result in ulcers, emaciation, and in some cases death. Mycobacteria have been previously isolated from a variety of Chesapeake Bay fish species, and the current study was designed to identify potential host specificity and location fidelity of mycobacterial isolates. Mycobacteria were isolated from wild fish of the Chesapeake Bay collected from the Upper Bay, the Choptank River, Herring Bay, the Chicamacomico River, the Pocomoke River and the Potomac River in 2003–2006. Mycobacterial isolates were recovered from striped bass, Morone saxatilis , Atlantic menhaden, Brevoortia tyrannus , white perch, Morone americana , summer flounder, Paralichthys dentatus , spot, Leiostomus xanthurus , largemouth bass, Micropterus salmoides , channel catfish, Ictalurus punctatus , common carp, Cyprinus carpio carpio , spotted seatrout, Cynoscion nebulosus , killifish, Fundulus sp., blueback herring, Alosa aestivalis , American gizzard shad, Dorosoma cepedianum and American silver perch, Bairdiella chrysoura . Twenty-nine well-defined mycobacterial groups resulted from gas chromatography dendrogram clustering of isolates. The majority of groups included more than one host species and more than one site of collection. However, four groups contained only striped bass isolates, three of which were similar to M. shottsii . Therefore, multiple Chesapeake Bay fish species are colonized with multiple mycobacterial isolates, of which few appear to be host or location specific.  相似文献

Abstract Predation is an important force structuring aquatic communities, but predator–prey interactions are complex and regulated by multiple factors. Invasive fishes may interact with native fishes to alter predator–prey preferences and community dynamics. For example, common carp, Cyprinus carpio L., is an invasive species that can become abundant and negatively affect aquatic ecosystems. Juvenile common carp are occasionally found in predator diets, but predator preferences for common carp compared with alternative prey remains unknown. Prey selection and feeding behaviour of five piscivores (flathead catfish, Pylodictis olivaris (Rafinesque); largemouth bass, Micropterus salmoides (Lacepède); smallmouth bass, M. dolomieu Lacepède; walleye, Sander vitreus (Mitchill); and northern pike, Esox lucius L.) foraging on juvenile common carp and two alternative prey (fathead minnow, Pimephales promelas Rafinesque, and yellow perch, Perca flavescens Mitchill) at variable densities and habitats were evaluated. Common carp and fathead minnow were generally selected for or neutrally selected across predator species, habitat types and prey assemblages. By contrast, yellow perch was generally selected against. Common carp were easily captured but difficult to manipulate and ingest compared with other prey. These results reveal that common carp are vulnerable to a variety of predators, suggesting control of this detrimental invader may be possible through biomanipulation.  相似文献

Channel catfish, Ictalurus punctatus (Rafinesque), goldfish, Carassius auratus (L.), striped bass, Morone saxatilis (Walbaum), and Gulf sturgeon, Acipenser oxyrinchus desotoi Vladykov, were acclimatized to fresh water or salinities of 9.0‰ or less and then exposed to Flavobacterium columnare (formerly known as Flexibacter columnaris ), the bacterial pathogen that causes columnaris disease. None of the fish acclimatized to 3.0 or 9.0‰ salinity died, and all deaths in lower salinities occurred between 1 and 5 days after exposure to F. columnare . Mortality was 97.7% in fresh water and 67.1% in 1.0‰ salinity for channel catfish (model SE, 1.8) and 66.5% in fresh water and 40.8% in 1.0‰ salinity for goldfish (model SE, 1.2); and 96.9% in fresh water and 61.7% in 1.0‰ salinity for striped bass (model SE, 1.8). After exposure to F. columnare , none of the Gulf sturgeon died. Flavobacterium columnare was isolated from the skin and gills of all fish dying during the experiments, but was not isolated from survivors in fresh water and 1.0‰ salinity 21 days after bacterial exposure. In vitro growth of bacteria was significantly higher in 1.0 or 3.0‰ salinity than in control medium (0.3‰ salinity). However, in vitro adhesion of bacteria was reduced with increasing salinity, which could explain the lower mortality of fish at higher salinities.  相似文献
