Marssonina brunnea is an important pathogen that causes Marssonina leaf spot disease of poplar (MLDP) in various poplar species. Resistance breeding is considered as the main method for preventing this disease and requires information on genetic diversity and population structure. However, molecular markers that may be utilized in the identification of this fungus are limited. This study investigated the distribution of microsatellites in the M. brunnea genome. A total of 15,356 microsatellite markers (excluding mononucleotide repeats) were isolated from 50.1 Mb of genomic sequence. Eight M. brunnea isolates were evaluated in terms of 102 loci, followed by the selection of markers that could be utilized in investigating the population structure of 47 M. brunnea isolates from two populations. Twenty‐four polymorphic microsatellite markers were developed for M. brunnea. The number of alleles per locus ranged from two to eight (average: 3.75). The polymorphism information content (PIC) of these loci ranged from 0.0408 to 0.6492. The average Shannon's information index of these loci in the two populations was 0.3819 and 0.5351, respectively. Using these markers, M. brunnea isolates were mainly divided into two distinct clusters based on the relatedness of the sampling sites. These results indicate that the selected markers could be effectively utilized in investigating the population genetic structure of M. brunnea. This is the first report on microsatellite markers in M. brunnea.  相似文献

Segregation distortion of molecular markers has been reported in a broad range of organisms. It has been detected in an interspecific BC1 Populus pedigree established by controlled crossing between clone “LM50” (Populus tomentosa) and its hybrid clone “TB01” (P. tomentosa×P. bolleana). The study with a total of 150 AFLP markers (approximately 18.9% of the total loci) exhibited significant deviation from the Mendelian ratio (1:1) (p<0.01). Twenty-five percent of the markers were mapped on the pa-rental specific genetic linkage maps of clones “LM50” and “TB01” with a pseudo-test-cross mapping strategy. Twelve linkage groups had markers with skewed segregation ratios, but the major regions were on linkage groups TLG2, TLG4 and TLG6 in the linkage map of clone “LM50”. We also analyzed the association between distorted loci and expression of complex traits with Map-maker/QTL software. A total of 16 putative QTLs affecting 12 traits were identified in the distorted regions on seven linkage groups. Therefore we could detect the distribution of skewed loci along the entire genome and identify the association between quantitative traits and segregation loci via genetic mapping in an interspecific BC1 P. tomentosa family. Furthermore, the genetic nature and pos-sible causes of these segregation distortions for differentiation between female and male parents were also discussed.  相似文献

Liriodendron are ideal plants for research on mating system, systemic evolution and population genetics. Here we report the development and application of a set of simple sequence repeat (SSR) markers derived from expressed sequence tags (ESTs) of L. tulipifera. In total, 176 primer pairs were developed and yielded 132 EST-SSR markers that amplified clear SSR bands with genomic DNA of L. tulipifera. Transferability of these EST-SSR markers to related species and genera was further examined. Twelve highly informative loci screened from 132 EST-SSR markers were applied to explore the effective pollen dispersal patterns and male reproductive success of Liriodendron. A provenance trial population of Liriodendron comprised 12 L. chinense (Hemsl.) Sarg provenances and 5 L. tulipifera provenances. The four maternal trees were randomly sampled as tested maternal trees in the population. Paternities of 220 open-pollination progenies from 4 maternal trees above were potentially identified by means of paternity analysis, of which 138 progenies (accounting for 62.7%) were assigned to 49 male parents at the 95% confidence level. The cumulative exclusion probability of 12 EST-SSR loci was 98.5%. The average effective pollen dispersal distance of pollen donors ranged from 15 to 35 m, with the maximum dispersal distance of 77 m. There were obvious male reproductive success differences among individuals, ranging from 0 to 10.9%. The cumulative male reproductive success of individuals within 35 m radius around the known maternal trees reached 51.4%. Diversiform mating patterns, including selfing, intra-species mating and interspecies mating, coexisted in the occasion of open-pollination in Liriodendron.  相似文献

Sphaeropsis sapinea is an important latent pathogen of Pinus spp., outbreaks of which have a considerable impact on plantations. This study considers the population diversity and distribution of S. sapinea in northern Spain at different spatial scales from single plantations to a wide area covered by Pinus radiata trees. Estimation of genotypic diversity is an important component of the analysis of the genetic structure of plant pathogen populations. Ten simple sequence repeat (SSR) markers were used, together with vegetative compatibility tests, to study the genetic diversity among S. sapinea isolates. Polymorphism analysis at SSR loci is a simple and direct approach for estimating the genetic diversity of S. sapinea isolates. From a total of 86 isolates collected from four different areas, 14 microsatellite haplotypes and 13 vegetative compatibility groups (VCGs) were identified. The percentage of maximum genotypic diversity, based on Stoddart and Taylor's index, for microsatellites of the northern Spain population ranged from 14.6% to 38.1% and from 8.0% to 29.4% for VCGs. Analysis of these markers and vegetative compatibility groups confirmed that S. sapinea reproduces mainly asexually due to its reduced genotypic diversity in spatially close populations. Isolates of S. sapinea from northern Spain populations were predominantly monomorphic at the tested SSR loci. Vegetative compatibility groups also indicate a low level of genetic variability in these samples, which appear to be clonal.  相似文献

Summary Two highly variable DNA markers were developed for Cronartium flaccidum and Peridermium pini. The markers were used to study the genetic composition of aecia of single cankers of both species. All five cankers of P. pini were homogenic and contained only one allele in both loci studied, indicating that the aecia were homozygous. The homogeneity of the cankers suggested that either genetic exchange via spermatization does not occur in autoecious P. pini populations, or that the pairing is homothallic. Two C. flaccidum cankers contained heterozygous loci. However, one canker contained genetically variable aecia indicating heterothallism, whereas the aecia of the other canker were homogenic.  相似文献

The “algarrobo” [Prosopis chilensis (Molina) Stuntz] is a tree species that represents an important natural resource in arid and semi-arid regions of Argentina. In this paper, we analysed and compared the variability of 46 RAPD (Random Amplified Polymorphic DNA) loci with previous estimates obtained from 12 isozyme markers in nine Argentinean populations of P. chilensis representative of the whole range of this species in Argentina. We evaluated the population structure and the existence of genetic variants associated with environmental variables. Expected heterozygosity (H e) estimated from RAPD varied significantly among populations and regions. Hierarchical analysis of genetic variability (AMOVA) showed that most (88.1%) of the total diversity occurs within populations, the component among populations within regions (9.3%) was intermediate, while the between-region component was the lowest (2.6%). All three variance components were highly significant. The MDS plot from pair-wise Φ ST matrix was consistent with the highly significant among-region differentiation indicated by the AMOVA. All 12 variable isozyme loci and 26 out of 46 RAPD loci showed significant or highly significant association with at least one geographic/climatic variable according to the stepwise multiple regression analysis. These results imply that the genetic differentiation among populations is better explained by environmental or biogeographical grounds than by geographical distances, suggesting gametic disequilibrium with loci responsible for the adaptation to particular environmental conditions. The information from RAPD markers would provide a relevant criterion to preserve genetic diversity in programmes for conservation and rationale use of this species.  相似文献

The dynamics of a recent outbreak of the spruce bark beetle (Ips typographus) in Switzerland was ruled by a devastating winter storm in 1999 and the drought and heat of the summer 2003. Starting from a similar level of population sizes, estimated as the rate of infested growing stock, beetle populations increased differently in magnitude and time among different regions in Switzerland. Accordingly, we expected local or regional genetic differentiation as a result of such repeated population expansion/breakdown dynamics. We analyzed 5 nuclear microsatellites of spruce bark beetles sampled from pheromone traps at 30 locations distributed over Switzerland. Our genetic results did not indicate any sign of population differentiation, structure, isolation by distance, or recent bottlenecks. This complete lack of genetic structure suggests that spruce bark beetles are highly mobile, precluding the formation of a spatial structure at neutral molecular markers. Thus, this molecular–genetic approach does not allow us to discriminate among regional gene pools and to identify the origin of expanding beetle populations.  相似文献

Raffaelea quercus‐mongolicae is a fungus associated with oak wilt and deemed to cause extensive oak mortality in South Korea. Since the discovery of this fungus on a dead Mongolian oak (Quercus mongolica) in 2004, the mortality continued to spread southwards in South Korea. Despite continued expansion of the disease and associated significant impacts on forest ecosystems, information is lacking about the origin and genetic diversity of Rquercus‐mongolicae. Restriction‐site‐associated DNA (RAD) sequencing was used to assess genetic diversity and population structure among five populations (provinces) of Rquercus‐mongolicae in South Korea. In total, 179 single nucleotide polymorphisms (SNPs) were identified among 2,639 RAD loci across the nuclear genome of the 54 R. quercus‐mongolicae isolates (0.0012 SNPs per bp), which displayed an overall low expected heterozygosity and no apparent population structure. The low genetic diversity and no apparent population structure among South Korean populations of this ambrosia beetle‐vectored fungus support the hypothesis that this fungus was introduced to South Korea.  相似文献

We have examined the pattern of random amplified polymorphic DNA (RAPD) variation among six populations of Crataegus monogyna from northern Italy, extending over an area of about 20,000 km2. The aim was to evaluate local differentiation in relation to geographical distance. Of the 73 loci analysed, 65 were polymorphic, and there were no bands at complete fixation at the population level. The mean genetic diversity was 0.291, with the values for individual populations ranging between 0.252 and 0.333. According to the analysis of molecular variance most of the genetic variation was found within populations (about 80%), with a significant proportion being attributable to genetic differences between populations. No evidence for isolation by distance was found in the set of populations sampled. The efficacy of RAPD markers in analysing genetic variation, and the contribution of the results in terms of the preservation of biodiversity and defining the most appropriate strategies for collecting forest reproductive material, are discussed.  相似文献

Raffaelea quercus‐mongolicae is a mycangial symbiont of a wood‐boring ambrosia beetle, Platypus koryoensis (Coleoptera: Curculionidae: Platypodidae), which is believed to be associated with oak mortality in South Korea. Since the first discovery of the disease in 2004, oak mortality has continued to spread across the country, and the pathogen has had a significant impact on forest ecosystems in the country; however, little is known regarding the biology of the fungus. The aim of this study was to develop markers to determine the mating types of individual isolates of R. quercus‐mongolicae collected across the country and then determine the mating‐type ratio in natural populations. To achieve this, partial sequences of MAT1‐1–3 and MAT1‐2–1 genes were amplified using degenerate primers and cloned. The new primer sets, which were specific to each mating‐type idiomorph, identified isolates as MAT1‐1 and MAT1‐2 mating types. The roughly equal frequencies of the two mating types suggest that sexual reproduction might have occurred in natural populations of Rquercus‐mongolicae in South Korea.  相似文献

The molecular variation and genetic relationships among five populations of the fall webworm (Hyphantria cunea Drury) in China were assessed using microsatellite technology. Ten microsatellite primers, producing polymorphic bands, were used across 300 samples. The percentage of polymorphic loci (PPB) was 98.36%; the percentage of polymorphic loci in five populations ranged from high to low in the following order: Cangzhou population, Yantai population, Qinhuangdao population, Dandong population, and Shijiazhuang population. The results showed that 34.38% of the total genetic variation of the fall webworm (GST) occurs among populations, while most variation (65.62%) exists within populations. Nei’s genetic distances ranged from 0.1386 to 0.3224. Using the unweighted pair group method with arithmetic mean (UPGMA), Nei’s genetic distances were analyzed by a clustering technique and the dendrogram shows that population differentiation is closely related to the time and geographic origin of the invasion. The major factors impacting genetic diversity of fall webworm populations are longitude, the plain area ratio, annual precipitation, latitude and time of invasion. The formation of genetic structure is correlated with characteristics of the life history and invasion ecology of the species.  相似文献

Hepatacodium miconioides is the Class II protected plant species in China. This paper studies the genetic diversity and differentiation of its nine natural populations in Zhejiang Province by using random amplified polymorphic DNA (RAPD) technique. Twelve random primers were selected in the amplification, and 164 repetitive loci were produced. The percentage of polymorphic loci in each H. miconioides population ranged from 14.60% to 27.44%, with an average of 20.73%. Among the test populations, Kuochangshan had the highest percentage of polymorphic loci, Simingshan took the second place, and Guanyinping had the lowest percentage. As estimated by Shannon index, the genetic diversity within H. miconioides populations accounted for 27.28% of the total genetic diversity, while that among H. miconioides populations accounted for 72.72%. The genetic differentiation among H. miconioides populations as estimated by Nei index was 0.715,7. This figure was generally consistent with that estimated by Shannon index, i.e., the genetic differentiation among populations was relatively high, but that within populations was relatively low. The gene flow among H. miconioides populations was relatively low (0.198,7), and the genetic similarity ranged from 0.655,7 to 0.811,9, with an average of 0.730,6. The highest genetic distance among populations was 0.422,9, while the lowest was 0.208,3. All the results showed that there was a distinct genetic differentiation among H. miconioides populations. The genetic distance matrix of nine test populations was calculated using this method, and the clustering analysis was made using the unweighted pair group method with arithmetic mean (UPGMA). Gmelina arborea is an important timber-yielding tree that grows naturally in the tropical and subtropical regions of Southeast Asia and has also been introduced as a plantation species outside these regions. Genetic diversity in this tree species was observed in stone/seed-related traits and in vitro responses of cultured nodal segments from plants of eight different populations representing natural forests, fragmented forests and plantations. Variance analysis showed significant differences between populations for these traits. However, it was not possible to separate the different populations using these traits by multivariate analysis, even after environmental variation was reduced over six subcultures. Genetic diversity was therefore analysed using molecular markers. Inter-simple sequence repeat (ISSR) primers yielded 95% polymorphic loci among the eight populations and UPGMA analysis enabled separation of these populations on the basis of their genetic distances. Diversity was also analyzed using population genetics parameters like Nei’s genetic diversity and gene differentiation. Nei’s genetic diversity was 0.29 between populations and 0.11 within populations. AMOVA analysis indicated 41 and 59% within- and between-population genetic diversity, respectively. Mantel test suggested that genetic differentiation between six Indian populations was positively correlated to geographic distance (r = 0.626, P = 0.029). Assessment of the genetic variation in G. arborea populations is an important step in selection of conservation strategies for this species since diversity forms the basis for species adaptation.  相似文献

Thirty-nine strains of Gremmeniella abietina were isolated between 1980 and 2005 from diseased or symptomless Pinus sylvestris and Pinus nigra, or sporadically Pinus jeffreyi and Pinus armandii from four regions of Poland with varying climatic conditions. Thirty-five of the strains were genetically similar to the G. abietina A type found in Scandinavia, whereas four were similar to the B type. On the basis of random amplified microsatellite markers, very high intrapopulation and interpopulation genetic variation was detected among the strains. The average value of the Jaccard genetic similarity coefficients among strains was 0.56 for A type and 0.47 for B type, and the monomorphic loci partition was small, at 14% of all analysed loci for the A type. The degree of genetic variation of the fungal strains within each of the four regions was similar, showing no significant differences. The Jaccard coefficient for the strains isolated from P. sylvestris and P. nigra differed significantly, at 0.57 and 0.54 respectively. The genetic distance among A type strains from the investigated regions, expressed using Nei’s coefficient, was not correlated with geographic distance; however, it was highly dependent on elevation (correlation coefficient r = 0.92).  相似文献

The transmission ratio of the pinewood nematode (PWN), Bursaphelenchus xylophilus, to the emerging adult Japanese pine sawyer (JPS), Monochamus alternatus, in its pupal chamber is a determinant of the number of the nematodes carried by JPS beetles. To investigate the factors affecting the transmission ratio, we counted the number of the nematodes carried by 36 newly emerged JPS beetles and the number remaining in and around their pupal chambers, and then estimated the transmission ratio (the number of nematodes carried by a JPS adult as a percentage of the total number of nematodes aggregating in and around its pupal chamber). The total number of nematodes aggregating in and around a pupal chamber ranged from 0 to 19,041, and the number of nematodes carried by a beetle ranged from 0 to 18,920. The transmission ratio correlated with neither the water content of the wood around the pupal chamber nor the degree of wood discoloration caused by blue-stain fungus. The transmission ratio varied with the abundance of the nematodes aggregating in and around the pupal chamber. In pupal chambers with more than 1,000 nematodes, almost all the nematodes were transmitted to the beetle. However, in pupal chambers with fewer than 1,000 nematodes, the transmission ratio varied greatly, from 0 to 100%. These results suggest that aggregation of many PWNs in the pupal chamber might stimulate transmission of the PWNs to the JPS adult and that this abundance-related transmission might contribute to the large variation in the number of PWNs carried by the JPS beetle.  相似文献

利用SSR标记对17个湿加松亲本无性系进行遗传多样性分析,进而分析亲本遗传距离与子代杂种优势的相关性。结果表明:14个微卫星标记在湿加松亲本群体中均呈现高度多态,共检测到57个等位基因,多态位点百分率为87.72%,平均杂合度为0.144 4 0.884 1,以SSR标记遗传距离聚类,将参试亲本材料划分为2大类群,可以用于湿加松亲本遗传多样性的评价。树高、胸径、材积的杂种优势与亲本遗传距离相关性达显著或极显著水平,相关系数分别为0.542 0、0.641 2、0.639 3,决定系数分别为0.293 8、0.411 0、0.408 7。分子标记遗传距离与树高、胸径、材积的杂种优势相关程度较高,利用SSR标记预测湿加松杂种优势是可行的,为湿加松良种选育及亲本选配提供参考。  相似文献

As a N2-fixing tree species, sea buckthorn (Hippophae rhamnoides) is well adapted to arid regions and is utilized for multiple purposes in China. Current knowledge of genetic variability of H. rhamnoides is limited in terms of rangewide distributions. Eleven natural populations of sea buckthorn in northeastern and northwestern China were analyzed to detect genetic variation among and within populations, by use of ISSR (inter-simple sequence repeats) markers. Using eight primers, 207 polymorphic loci were observed, ranging in size from 250 to 2500 bp. The coefficient of gene differentiation (Gst = 0.0679) showed that the total molecular variance of 11 populations was mainly existed within populations. The genetic variation within and among the 11 populations was 93.21 and 6.79%, respectively. No significant correlation between genetic and geographic distances of the populations was found using ISSR markers. Our study provides a population-level genetic profile for further investigation and conservation of genetic diversity of sea buckthorn.  相似文献

In order to obtain information on the genetic structure ofAbies concolor and the genetic variation among 11 populations introduced from America to China, allozyme analysis based on starch gel electrophoresis technology was used. 24 loci of 10 allozyme systems were mensurated, and the genetic structure and genetic diversity of the 11 populations of A. concolor evaluated. The results show that the genetic variation among is significant, and the genetic variation within A. concolor populations is more important. In contrast with other conifers, the variation of A. concolor is above the average level of conifers, and higher than the same level of Abies. The percentage of polymorphic loci (P) was 62.5%, the number of alleles per locus (A) 2.08, the number of effective alleles per locus (Ae) was 1.37, the expected heterozygosity (H) 0.204, and the Shannon information index (/) 0.351 7. There is a short genetic distance (D=0.061) and a low gene flow (Nm=0.839 4) among the 11 introduced populations ofA. concolor with high genetic variation. The genetic differentiation coefficient (Gst) was 0.229 5, which is higher than that of the mean in Abies or Pinus.  相似文献

Abstract

The English yew (Taxus baccata L.) is an endangered rare tree species in Austria. More than 600 bud samples were collected from seven populations in three regions of the Eastern Alpine mountains for estimating their genetic structures by using isozyme gene markers. Nine gene loci encoding for seven enzyme systems were investigated. The revealed 32 alleles indicate a higher multiplicity of the east alpine gene pool compared with other European regions. English yew showed a high level of genetic variation, with a mean number of alleles per locus (A/L) of 2.7. A total of 76.4% of the loci were polymorphic, the average expected heterozygosity was (H e) 0.274 and the mean observed heterozygosity (H o) was 0.238. The relative proportion of genetic differentiation among the regions was found to be 6.2%, the level of gene flow was high (Nm=3.78) and the level of inbreeding (0.130) among the regions was medium. The obligatory out crossing (dioecious plant), the long generation time, the ability to regenerate from stump and the seed dispersal by birds and rodents might contribute to the high level of genetic variation found within the population. The conservation propositions are discussed with respect to the genetic structure and the poor recruitment of this species.  相似文献

To test how efficiently plantations and seed orchards captured genetic diversity from natural Anatolian black pine (Pinus nigra Arnold subspecies pallasiana Holmboe) seed stands, seed sources were chosen from 3 different categories (seed stands (SS), seed orchards (SO) and plantations (P)) comprising 4 different breeding zones of the species in Turkey. Twenty-five trees (mother trees) were selected from each SS, SO and P seed sources and were screened with 11 Random Amplified Polymorphic DNA (RAPD) markers. Estimated genetic diversity parameters were found to be generally high in all Anatolian black pine seed sources and the majority of genetic diversity is contained within seed sources (94%). No significant difference in genetic diversity parameters (numbers of effective alleles, % of polymorphic loci and heterozygosity) among seed source categories was found, except for a slight increase in observed heterozygosities in seed orchards. For all seed source categories, observed heterozygosity values were higher (Ho = 0.49 for SS, 0.55 for SO and 0.49 for P) than expected ones (He = 0.40 for SS, 0.39 for SO and 0.38 for P) indicating the excess of heterozygotes. In general, genetic diversity in seed stands has been transferred successfully into seed orchards and plantations. However, the use of seeds from seed orchards can increase the amount of genetic diversity in plantations further. The study also demonstrated that number of plus-tree clones (25–38) used in the establishment of seed orchards was adequate to capture the high level of diversity from natural stands.  相似文献
