本文分别在温室和田间条件下利用人工接种的方法研究了马铃薯纺锤块茎类病毒(PSTV)和PVY在克新4号及Kennebec两品种上的相互作用.试验结果表明:在温室条件下PSTV和PVY的复合接种均能在二品种上不同程度地产生条型坏死症状,而在田间未出现明显症状.PSTV与PVY混合接种或PSTV接种一周后再接PVY会促进PVY增殖,植株内PVY的浓度显著高于单独接种PVY,但PVY侵染后再接PSTV对PVY的浓度无显著影响.PSTV和PVY的工作对PSTV的浓度无显著影响.PSTV与PVY复合侵染的三个接种类型都明显地降低平均单株产量和商品薯率,产量损失最大的处理为PSTV与PVY混合接种,可导致克新4号减产45.4%。  相似文献

Foliar treatment of potato (Solanum tuberosum L.) with metribuzin at 0.57–1.0 lb/A caused a necrotic reaction in leaflets of plants infected with potato virus Y (PVY), but not in plants infected with potato virus X (PVX) or potato leafroll virus. Necrosis symptoms resulting from metribuzin-PVY interaction were distinct from symptoms of either PVY infection or of metribuzin injury. This reaction was similar in the Russet Burbank, Lemhi Russet, and Pioneer cultivars. Russet Burbank infected with PVY and PVX, alone or in combination, was treated with metribuzin to study herbicidevirus interaction effects on yield. Secondary PVY infection alone caused a 57% yield reduction, and when combined with PVX caused a 71% yield reduction. Although leaflet necrosis was induced by the metribuzin-PVY combination, there was no significant yield interaction. Results suggest that post-emergence application of metribuzin can be used as an aid for detecting and removing potato virus Y infected plants from potato seed fields  相似文献

马铃薯纺锤块径类病毒(Potato spindle tuber viroid)是危害黑龙江省马铃薯生产的一种重要病害。为了查明不同类病毒变株对主栽品种克新1号和4号所引起的产量损失和症状反应,应用往复聚丙烯胺凝胶电泳法(Peturn-Polyacmdegel electophoresis)鉴定类病毒。试验结果表明,接种当年,接种类病毒的处理平均减产37%,按商品产量计算减产达47%。当年表现症状的植株平均为59.6%,块茎产生症状的植株平均为57.4%。用于接种的3个马铃薯纺锤块茎类病毒变株的致病力,以当地的类病毒弱系为最强,其次为北美的类病毒强系,北美的类病毒弱系的致病力较弱。类病毒对产量的影响与马铃薯品种和类病毒变株的组合有极为密切的关系。克新1号接种当地的类病毒弱系时薯块严重畸变,商品产量只为不接种对照的21%。用往复电泳法检测各个试验处理被类病毒侵柴的情况,芽接种处理,于出苗后1周即可检测到类病毒,叶接种处理,于接种后2周即能检测到类病毒。出苗后7周,植株中类病毒浓度达到高峰,随即急剧下降,到出苗后10周,大部分接种植株已检测不到类病毒。块茎直径达1厘米时,即能检测到类病毒,随着块茎膨大,类病毒浓度也相应增高,到收获前,块茎中的类病毒含量只略低于植株含类病毒高峰期的浓度。用于本试验的克新1号、4号无类病毒和主要马铃薯病毒的核心材料,在1988年已交付给省内外的良种场繁殖使用。于当年秋季抽样检测克山第二良种场连续种植二年的种薯,当年种植试管苗生产的种薯,以及试管苗等,均未检测到类病毒。  相似文献

Potato spindle tuber viroid (PSTV) caused albinism in the new growth of Rutgers tomato under continuous light. The albinism response was obtained with 4 isolates causing severe symptoms in tomato and with 1 mild isolate which is normally symptomless. Rutgers tomato inoculated with the 4 severe strains of PSTV developed typical ‘bunchy top’ symptoms in 12 to 16-hr day lengths. Symptoms developed slower and much less distinctly on plants incubated in 6-hr days. New growth in PSTV-infected plants was green in short days (12-hr illumination) and chlorotic to white in continuous light. Infectivities of extracts from white portions of plants were from 3 to 10 times greater than those from green portions of infected plants. Albino symptoms also developed in other tomato varieties when inoculated with PSTV and kept under continuous light. Albinism was most pronounced under continuous light at 30°C, somewhat less intense at 24°, and at 16°, new growth of infected leaves and stems remained green. Albinism did not develop in otherSolanum species which are symptomless hosts of PSTV nor in seedling potato plants grown in continuous light.  相似文献

Potato plants grown from true seed were naturally infected with the Solanum apical leaf curling virus (SALCV) at the high jungle site near San Ramon, Peru. Tubers from these plants were greenhouse indexed. Symptoms of SALCV infected potato plants include apical rolling, stunting, delayed emergence, weak plants, hair sprouts, and tuber dormancy. These symptoms superficially resemble certain aspects of unusually severe current season potato leafroll virus (4) infection or those of the aster yellows and stolbur mycoplasma diseases (5). Up to 12 percent of the indexed plants were naturally infected with SALCV and SALCV was demonstrated in approximately half of the tubers that produced hair sprouts.  相似文献

Six insect pests of potato, green peach aphid,Myzus persicae (Sulzer); potato leafhopper,Empoasca fabae (Harris); Colorado potato beetle,Leptinotarsa decemlineata (Say); tarnished plant bug,Lygus lineolaris (Palisot de Beauvois); red-legged grasshopper,Melanoplus femurrubrum (DeGeer); and southern army worm,Prodenia eridania (Cramer), were tested for their ability to transmit potato spindle tuber viroid (PSTV). PSTV infection was determined by a gel electrophoretic assay made 6 weeks after transfer of insects from infected to non-infected plants of the cultivar, ‘Katahdin.’ Plants grown from tubers of the original non-infected hosts were also assayed. PSTV was detected in only 2 of 183 test plants; these 2 plants had been infested with the tarnished plant bug. These results suggest that the insect species studied are relatively insignificant as vectors of PSTV.  相似文献

A total of 258 Plant Introductions (PI) belonging to 69Solarium species were evaluated in the greenhouse for their reaction to the tobacco veinal necrosis strain of potato virus Y (PVY N). One hundred and thirty-one (50.7%) of the PI accessions produced mosaic symptoms ranging from mild to severe. Local lesion and veinal necrosis symptoms were observed in 19 PI accessions (7.3%) and a variety of other symptoms were observed in another 11 PI accessions (4.2%). Only 97 PI accessions (37.5%) were symptomless carriers of PVYN. PI accession 473505 ofS. sparsipilum and PI accession 498021 ofS. brachycarpum developed local lesions and veinal necrosis with PVYN, but necrotic spots and mosaic with PVYo. Common mechanically-transmitted potato viruses A, S, M, and X did not interfere with PVY symptom development inS. sparsipilum andS. brachycarpum. Thus, PI 473505 and PI 498021 can be used as indicator plants for specific identification of PVYN. PI accession 472819 ofS. chacoense developed local lesions with systemic spread in PVYo, but without systemic spread in PVYN. Thus, this can be used as a differential host plant for PVY strains. Two PI accessions ofS. stoloniferum, PI 160372 and 161171 were immune to PVYN.  相似文献

Viruses that infect naturally an important Sudanese potato stock Zalinge were detected using enzyme-linked immunosorbent assay, immunosorbent electron microscopy and sap-inoculation to test plants. All of the 19 plants of Zalinge tested were infected with potato leaf roll virus (PLRV) and potato virus S (P VS), and five plants also with potato virus X (PVX). No potato virus Y (PVY), A (PVA) nor M (PVM) were found. The viruses were eradicated with thermo and chemotherapy using standard procedures. The combination of both therapies did not result in any virus-free plants, but resulted in poor plant survival. Thermotherapy reduced the incidence of PLRV and PVS by 45% and 50%, respectively, and one virus-free plant was obtained. It grew vigorously in the greenhouse, was symptomless and had a significantly increased tuber yield compared to the virus-infected plants. Following sap-inoculation with PVYO, Zalinge showed mosaic symptoms, developed necrosis in the leaves and stem and died 14 days post-inoculation. However, the plants of Zalinge infected with PVYN remained symptomless, which suggested that hypersensitivity was specific to PVYO. The fast development of lethal necrosis following infection with PVYO may contribute to the low incidence of PVY in Zalinge in the field in Sudan.  相似文献

Seed transmission of potato spindle tuber virus (PSTV) has special significance for potato breeding programs. Transmission of PSTV has been reported to occur via true seed. Test families in the Frito-Lay breeding program were obtained from self- and cross-pollinations to determine the incidence of infected seedlings from healthy and diseased parents. Diseased seedlings from PSTV-infected Katahdin selfed were scored by visual and tomato index procedures as 100% and 87% infected, respectively. A similar result was obtained among seedlings from the cross of PSTV-infected Sebago x PSTV-infected Katahdin. Symptoms of PSTV infection were not detected by either method among seedlings from healthy Katahdin, selfed. The Frito-Lay, Inc., potato breeding program employs a number of phyto-sanitary procedures to exclude or reduce the possibility of PSTV contamination and spread.  相似文献

Summary Conditions necessary for the detection of potato leafroll virus (PLRV) and potato virus Y (PVY) in tubers from primary and secondary infected plants were investigated. Tubers were analysed before and after breaking dormancy by rindite treatment. PLRV was reliably detected indormant tubers whereas PVY was readily detected only when tubers had been rindite-treated and held for two to three weeks at 22°C and high humidity in the dark. PLRV occurred in higher concentration at the heel end than at the rose end of infected tubers and the concentration remained nearly unchanged during the experimental period of 35 days, whereas PVY was found to be more concentrated at the rose end and was rapidly accumulating in the tubers after the break of dormancy. In dormant tubers PVY concentration dropped during storage at 22°C. The use of ELISA for tuber indexing is discussed.  相似文献

Maintaining potato viruses X, S, Y, A, M and leafroll in tissue culture plantlets is a convenient, cost and space effective alternative to the use of greenhouse plants. Of these six viruses, only certain strains of PVX induced symptoms in tissue culture plantlets. Nevertheless, all infected tissue culture plants were found to be more reliable than greenhouse grown plants as virus-infected controls in enzyme-linked immunosorbent assay (ELISA) testing. Another advantage of maintaining viruses in tissue culture plantlets was the elimination of contamination by other viruses or other pathogens. Leaves, stems, and roots of virus infected plantlets were tested separately for antigen levels by ELISA. In these tests, the stems and leaves of all but PVA infected tissue culture plants consistently gave positive ELISA values. In contrast, root tissue from PVY infected tissue culture plantlets was not reliable for PVY detection. In all cases, the viruses detected in the original source material were detected in the resulting tissue culture plantlets.  相似文献

A diagnostic test for the potato spindle tuber viroid (PSTV) that is based on hybridization of highly radioactive, recombinant DNA complementary to PSTV with PSTV bound to a nitrocellulose membrane and autoradiographic detection of the resulting DNA-RNA hybrids has been evaluated with tubers from 20 potato clones maintained at the International Potato Center (CIP) and with true seed obtained from healthy or PSTV-infected potato plants. The nucleic acid spot hybridization test detected PSTV not only in tuber sprouts from 10 clones that had previously tested positive in polyacrylamide gel electrophoretic (PAGE) analysis, but also in tuber sprouts from 9 clones that had previously tested negative in PAGE analysis. Further tests confirmed the presence of PSTV in these clones. The spot hybridization test detected PSTV in mixtures of seed extracts equivalent to as few as one seed from a PSTV-infected plant and 80 seeds from healthy plants. The spot hybridization test was shown to be more sensitive and reliable than PAGE analysis; it is suitable for the testing of large numbers of samples with a minimum expenditure of labor and materials.  相似文献

Summary Using a modified procedure of Return-polyacrylamide gel electrophoresis (R-PAGE) mild strains of potato spindle tuber viroid (M-PSTV) were detected reliably from dormant tubers. The sensitivity of R-PAGE detection of M-PSTV was equivalent to that of nucleic acid hybridization. Both methods detected M-PSTV when infected tissue was mixed with healthy tissue in a ratio of 1 to 100. When extracted nucleic acid was diluted with buffer, R-PAGE detected PSTV at a dilution of 1:256 and nucleic hybridization only up to 1:64 PSTV was readily detected from 18 potato cultivars. In addition, mild, ‘intermediate’ and severe strains were separated by R-PAGE, on the basis of their mobility on the electrophorogram.  相似文献

AC Novachip is a smooth, white-skinned potato cultivar with oval to oblong tubers and a maturity similar to Kennebec. It has excellent chip quality, especially after long storage and reconditioning. The boil and bake quality is similar to Kennebec; the specific gravity is higher than Kennebec but lower than Atlantic. AC Novachip has moderate resistance to common scab and PVY.  相似文献

The transmission of PVS by the cutting knife in potato cvs Norland, Kennebec and Russet Burbank was studied in the field and greenhouse. Field studies showed transmission to Norland via tuber infection was significantly less than for Kennebec and Russet Burbank (P = 0.01). However, significant cultivar differences were not found in the greenhouse (P = 0.05). Contamination of the cutting knife by cutting through sprouts resulted in significantly greater numbers of plants infected for cv Kennebec than when sprout contact was avoided (P = 0.05). Although the same trends were observed for cv Norland and Russet Burbank the differences were not significant (P = 0.05). Infectious PVS particles were readily recovered from materials commonly contacted during potato cultivation, storage and processing. Retention time of infectious PVS was usually greater when materials were held at 4°C and 100% relative humidity than at ambient greenhouse conditions. Retention times ranged from 0 hr (PVS not recovered) to 180 hr on unpainted wood. Results demonstrated the need for strict indexing and sanitation procedures during the production of PVS-free potato seed.  相似文献

Summary The symptomatology of Andean potato latent virus (APLV) was studied in several wild potato species and potato cultivars using isolates Caj, Hu and Ay from Peru. In greenhouse tests, the virus was latent in most isolate/cultivar combinations with primary infection but secondary infection commonly caused symptoms. Wild species infected with the different isolates normally reacted with symptoms with either type of infection. The main symptoms were mosaics, chlorotic netting of minor leaf veins and, with wild species, leaf deformation; the netting symptom was typical only with Caj. In infected cultivars growing in the field, an additional symptom, rugosity, sometimes also developed and environmental conditions at high altitude seemed to enhance symptom formation. Very severe symptoms were found in a cultivar jointly infected with APLV and Andean potato mottle virus. Transmission of APLV from an infected plant to its tubers was erratic.  相似文献

Summary The infection pressure of two viruses, potato leafroll (PLRV) and potato virus Y (PVY), both common in seed potatoes grown in Cyprus, was determined in three experiments in 1982–83. Virus-free bait plants, of potato and four other species, were exposed weekly to field infection during the growing season (March–June), and then returned to an aphid-free glasshouse for symptom expression. Only tobacco plants produced clear symptoms enabling reliable assessment of PVY infection pressure. When assessed with ELISA or by tuber indexing, the potato plants were efficient baits for both viruses whose infection period commenced at emergence (mid March to early April) and ended within 6–7 weeks. The seasonal trend of aphid populations, determined with Moericke traps or 100-leaf counts, correspond to that of virus spread. Correlation and regression analysis of aphid and virus data implicated the alate form ofMyzus persicae as the principal vector of both viruses.  相似文献
