抗菌肽饲料添加剂对小鼠巨噬细胞吞噬功能的影响   总被引:3,自引:0,他引:3  
对试验小鼠饲喂1‰和3‰抗菌肽饲料添加剂,鸡红细胞吞噬试验检测小鼠腹腔巨噬细胞吞噬功能,研究抗菌肽饲料添加剂对小鼠免疫功能的影响.结果表明,抗菌肽饲料添加剂可以显著提高小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬率和吞噬指数,增强小鼠非特异性免疫功能.  相似文献

[目的]探讨羊胎盘转移因子注射液对动物细胞免疫的影响。[方法]以家兔和昆系小鼠为试验对象,应用E玫瑰花环试验和淋巴细胞转化试验,检测羊胎盘转移因子注射液对家兔T淋巴细胞数量与活性的作用,及其对小鼠淋巴细胞的活化作用。[结果]羊胎盘转移因子注射液具有显著增加淋巴细胞数量与活性的作用,可明显地提高淋巴细胞的转化率。[结论]羊胎盘转移因子注射液具有改善和提高受试动物特异性细胞免疫的功能。  相似文献

本实验测定了中药复方病菌净Ⅱ对小鼠腹腔巨噬细胞吞噬鸡红细胞功能的影响。测试结果,病菌净Ⅱ能显著提高小鼠腹腔巨噬细胞吞噬鸡红细胞的功能。其吞噬百分率和吞噬指数都显著高于对照组(P〈0.01)。结果表明,中药复方病药净Ⅱ有增强机体非特异性免疫功能的作用。  相似文献

为明确中药与甲氧苄啶复方在动物体内抗感染作用的机制,本试验研究了其对小鼠血液学和免疫学指标的影响。将受试药物按1、5、10 g/kg体重连续腹腔注射15 d,通过Coulter-JT血细胞分析仪对血液中的红细胞、白细胞、中性粒细胞、单核细胞和淋巴细胞进行了计数。采用脏器系数测定法、腹腔巨噬细胞吞噬鸡红细胞法、比浊法和MTT法分别对小鼠免疫器官指数、腹腔巨噬细胞吞噬功能、血清溶菌酶含量、血清溶血素水平和淋巴细胞增殖能力进行测定。结果表明中药与甲氧苄啶复方可显著增加血液中的红细胞数、单核细胞比例等(P<0.05),显著增加免疫器官指数,增强腹腔巨噬细胞吞噬功能和血清溶菌酶活性,提高血清溶血素水平和淋巴细胞增殖能力(P<0.05)。因此中药与甲氧苄啶复方对小鼠非特异性免疫和特异性免疫功能均有增强作用。  相似文献

芪蓝四君子汤对雏鸡部分免疫指标的影响   总被引:1,自引:1,他引:0  
选用370只1日龄雏鸡,随机分成5组,Ⅰ~Ⅲ组在新城疫(ND)La Sota系冻干苗免疫时分别按低(0.125 g/mL)、中(0.25 g/mL)、高(0.5 g/mL)的3个浓度灌服芪蓝四君子汤1 mL,连续10 d。Ⅳ组和Ⅴ组分别为免疫对照组和非免疫对照组。各组分别于20、30、40、50、60日龄采血测定新城疫血凝抑制(HI)抗体水平,随后剖杀计算胸腺、脾脏、法氏囊的器官指数;并于21、35、49日龄时用碳粒廓清试验测定外周血巨噬细胞的吞噬功能。试验结果表明,高剂量芪蓝四君子汤能显著提高新城疫HI抗体效价(P0.01)和免疫器官指数(P0.05),促进外周血巨噬细胞的吞噬功能(P0.01)。中,低剂量的芪蓝四君子汤对抗体、免疫器官指数、外周血巨噬细胞的吞噬功能均有不同程度的增强作用。芪蓝四君子汤能有效提高雏鸡的特异性免疫以及非特异性免疫功能。  相似文献

为探讨亚硒酸钠染毒对小鼠免疫功能的影响,将试验动物分为阴性对照组(0.00mg/kg体重.d)、亚硒酸钠低剂量组(0.14mg/kg体重.d)、中剂量组(0.54mg/kg体重.d)和高剂量组(2.16mg/kg体重.d)。连续灌胃30d后处死小鼠,分别用巨噬细胞吞噬鸡红细胞试验与溶血空斑试验检测小鼠腹腔巨噬细胞的吞噬功能与抗体形成细胞的数量。结果显示,亚硒酸钠染毒使小鼠巨噬细胞的吞噬功能与抗体形成细胞的数量显著降低,且与染毒剂量具有相关性。表明亚硒酸钠染毒可抑制小鼠非特异性免疫功能,呈现明显剂量效应关系。  相似文献

以小鼠为实验动物研究家蚕茧壳水解物的免疫调节功能。分别给正常雄性BALB/c小鼠灌胃低剂量(575 mg/kg)、中剂量(1 150 mg/kg)和高剂量(2 300 mg/kg)家蚕茧壳水解物,30 d后测定小鼠胸腺、脾脏质量计算脏器/体质量比值,并通过脾淋巴细胞增殖试验、腹腔巨噬细胞吞噬鸡红细胞试验、乳酸脱氢酶法和血清溶血素法等,分别检测细胞免疫功能、巨噬细胞吞噬能力、NK细胞活性和体液免疫功能。与对照组相比,中剂量家蚕茧壳水解物灌胃组小鼠的脾指数显著增加(P0.05),低剂量组和高剂量组小鼠胸腺指数显著增加(P0.05);中、高剂量家蚕茧壳水解物灌胃组小鼠的脾淋巴细胞增殖能力、NK细胞杀伤活性、腹腔巨噬细胞对鸡红细胞的吞噬指数等显著(P0.05)或极显著提高(P0.01)。此外,家蚕茧壳水解物对小鼠体质量和血清溶血素含量没有显著影响。研究结果表明,家蚕茧壳水解物能促进正常小鼠的免疫器官质量增加,对小鼠的细胞免疫和非特异性免疫功能也有明显的促进作用。  相似文献

通过冷冻透析法制备奶牛隐性乳房炎特异性转移因子,并对其免疫活性进行检测。方法为采用匀浆、冻融、透析被免疫猪的脾脏,制备特异性转移因子,运用理化性质检验、安全试验、无菌试验检验所制备转移因子的纯度及安全性,并运用小鼠巨噬细胞吞噬功能试验及E玫瑰花环试验对特异性转移因子免疫活性进行检测。试验结果为:所制备的奶牛隐性乳房炎特异性转移因子多肽含量为1.3 p g/mL,核酸含量为24μg/mL,OD_(260 nm)/OD_(280 nm)值为2.08,且安全、无菌。巨噬细胞吞噬功能试验及E玫瑰花环形成试验表明转移因子可明显增加巨噬细胞吞噬能力并能增加E玫瑰花环的形成数。因此,本品可作为奶牛隐性乳房炎的治疗药物,亦可作为细胞免疫增强剂使用。  相似文献

为明确中药增效抗菌复方的抗感染作用机制,本实验进行其对小鼠免疫功能影响的研究.将受试药物按每千克体质量1 g、5 g和10 g的剂量对小鼠连续腹腔注射15 d,采用脏器系数测定法、腹腔巨噬细胞吞噬鸡红细胞法、乳酸脱氢酶释放法和MTT法分别对小鼠免疫器官指数、腹腔巨噬细胞吞噬功能、NK细胞活性、血清溶血素水平和淋巴细胞增殖能力进行测定.结果表明,该药物可以显著增加小鼠免疫器官指数,增强腹腔巨噬细胞吞噬功能和NK细胞活性,也可以明显提高血清溶血素水平和淋巴细胞增殖能力.因此,中药增效抗菌复方对小鼠非特异性免疫和特异性免疫功能均有增强作用.  相似文献

为研究复方白毛藤注射剂对小鼠免疫调节作用 ,采用常规免疫学方法进行了 con A诱导的小鼠淋巴细胞转化增殖试验 ,硝基氟苯诱导迟发型变态反应试验 ,腹腔巨噬细胞吞噬功能试验及小鼠碳粒廓清试验。结果表明 :该制剂中 ,高剂量组均具有明显增强 con A诱导的小鼠脾淋巴细胞增殖能力 ,能显著增强硝基氟苯诱导的小鼠迟发型变态反应 ,明显增强小鼠腹腔巨噬细胞吞噬鸡红细胞能力及小鼠碳粒廓清能力。与对照组相比差异极显著 (P <0 .0 1 ) ,中、高剂量组差异不显著 (P >0 .0 5)。  相似文献

用氢化可的松注射液制造免疫功能抑制小鼠模型,以左旋咪唑及黄芪多糖为阳性对照,设计茯苓散高(200mg/kg)、中(100mg/kg)、低(50mg/kg)3个剂量组(以茯苓多糖计)。给各组分别灌注生理盐水或药物,连续14d。试验从3个方面6个试验进行茯苓散药效学方面的考察,即巨噬细胞功能测定(小鼠巨噬细胞吞噬鸡红细胞试验、茯苓散对小鼠非特异性免疫功能的影响一碳粒廓清法)、细胞免疫功能测定(茯苓散对小鼠免疫器官的影响、茯苓散对二硝基氟苯诱导小鼠迟发型变态反应的影响)、体液免疫功能测定(抗体生成细胞检测、血清溶血素的测定)。结果显示,茯苓散3个剂量组均能显著提高免疫低下小鼠吞噬细胞的吞噬活性(P〈0.05),吞噬细胞功能结果为阳性;茯苓散中、高剂量组能刺激脾脏、胸腺质量的增加(P〉0.05),也能显著促进免疫低下小鼠的迟发型变态反应(P〈0.05),细胞免疫功能结果为阳性;茯苓散3个剂量组均能显著提高小鼠的抗体生成细胞数(P〈0.05),且显著增强小鼠血清凝集素水平(P〈0.05),体液免疫功能结果为阳性。其中中剂量组效果最好,表明茯苓散对免疫低下小鼠的免疫功能有一定的增强作用。  相似文献

Density-separated feline RBC were analyzed for surface immunoglobulin and in vitro susceptibility to macrophage phagocytosis before and after introduction of a sterile abscess. Prior to abscess induction, least dense cells did not have detectable surface immunoglobulin G (IgG) as a direct antiglobulin reaction was negative. Two of four intermediate and three of four most dense cell fractions had detectable surface IgG as indicated by a positive direct antiglobulin reaction. On Days 3 and 5 after induction of sterile abscesses all three RBC fractions were positive and the size of the RBC clumps tended to increase. In vitro macrophage phagocytosis of all three RBC fractions increased significantly after abscess induction. The phagocytosis could be partially blocked by preincubation of macrophages with feline IgG.  相似文献

Ochratoxin A (OA) was incorporated in the diets of growing gilts (mean body weight, 20.1 kg) at a concentration of 2.5 mg of OA/kg of feed and was fed continuously for 35 days. Humoral and cell-mediated immunologic measurements were evaluated to determine the effects of OA on immune function in swine. Cutaneous basophil hypersensitivity to phytohemagglutinin (PHA), delayed hypersensitivity to tuberculin, PHA-induced lymphocyte blastogenesis, interleukin-2 production, total and isotype immunoglobulin concentrations, antibody response to chicken RBC, and macrophage activation were used to evaluate immune function. Gilts treated with OA had reduced cutaneous basophil hypersensitivity response to PHA, reduced delayed hypersensitivity to tuberculin, decreased stimulation index for lymphoblastogenesis, decreased interleukin-2 production when lymphocytes were stimulated with concanavalin A, and decreased number and phagocytic activity of macrophages. Differences were not observed for total and isotype immunoglobulin concentrations, or humoral hemagglutination (chicken RBC) titer. These data indicate that OA may suppress cell-mediated immune response in growing swine.  相似文献

Salmonella Enteritidis (SE) is one of the leading causes of food-borne salmonellosis, and macrophages play an essential role in eliminating this pathogen. Among the interventions to improve Salmonella clearance in chickens are the use of prebiotics and direct fed microbials (DFM) in animal feed as they have immunomodulatory effects. Therefore, we tested the influence of a prebiotic fructooligosaccharide (FOS)-inulin on the ability of the chicken macrophage HD11 cell line to phagocytose and kill SE, and express selected inflammatory cytokines and chemokines in an in vitro model. There were significantly fewer viable intracellular SE in HD11 cells treated with FOS-inulin than the untreated cells. However, SE phagocytosis, nitric oxide expression or production were not influenced by the prebiotic treatment. Among the inflammatory markers tested, IL-1β expression was significantly lower in HD11 cells treated with FOS-inulin. These results suggest that FOS-inulin has the ability to modulate the innate immune system as shown by the enhanced killing of SE and decreased inflammasome activation.  相似文献

为探讨鸡转移因子脂质体(chicken transfer factor liposome,CTFL)对小鼠腹腔巨噬细胞及消化吸收的影响,本试验选取了大小相近的18~22 g SPF级雌性小鼠30只,随机分成3组,每组10只,分别腹腔注射鸡转移因子(chicken transfer factor,CTF)、CTFL和生理盐水2 mL/只,隔日注射1次,连续注射5次,第11和21天检测小鼠腹腔巨噬细胞对鸡红细胞的吞噬率,同时分别取十二指肠、空肠和回肠固定到10%福尔马林液中、石蜡切片、苏木素—伊红染色,显微镜下观察,测量各段小肠的绒毛长度。结果显示,CTF和CTFL组对小鼠腹腔巨噬细胞吞噬率的影响与对照组相比均差异极显著(P<0.01),CTF和CTFL组相比差异不显著(P>0.05)。小鼠饲养11和21 d时,CTF和CTFL组的十二指肠绒毛平均长度均显著长于对照组(P<0.05),且CTFL组的十二指肠绒毛平均长度比CTF组长,而空肠和回肠平均绒毛长度均无显著差异(P>0.05)。结果表明,CTFL和CTF均能显著提高机体免疫功能,CTFL对小鼠消化吸收的促进作用比CTF强。  相似文献

The effect of a single oral dose of 4 mg of T-2 toxin/kg of body weight on in vivo phagocytosis of sheep RBC by peritoneal macrophages was evaluated in nonsensitized mice and in mice sensitized with sheep RBC. T-2 toxin treatment had no effect on the viability or phagocytic activity of resident peritoneal macrophages in nonsensitized mice. However, a significant (P less than 0.005) increase in phagocytic activity occurred in cells from mice treated with toxin and subsequently sensitized with sheep RBC. In contrast, phagocytosis of sheep RBC was significantly (P less than 0.05) suppressed in cells from mice treated with toxin after sensitization. Toxin treatment induced necrosis of lymphocytes and significant decreases in thymus and spleen weights. Seemingly, T-2 toxin, administered at a dose that caused marked lymphoid depletion, suppressed or enhanced in vivo macrophage phagocytic activity in antigenically sensitized mice, and enhancement or suppression of phagocytosis was a function of the time of toxin treatment in relation to antigenic stimulation.  相似文献

研究了鹿胎及胎盘制剂对老年雄性大鼠免疫器官指数、血清球蛋白含量和巨噬细胞吞噬率及吞噬指数的影响。结果表明,鹿胎及胎盘制剂对老年雄性大鼠胸腺和脾脏指数影响不显著(P>0.05);但提高了老年雄性大鼠血清球蛋白含量,其中鹿胎制剂Ⅰ号组最高,比对照组高出62.64%,与其他各组相比有显著差异(P<0.05);鹿胎制剂Ⅱ号组和人胎盘粉处理组大鼠巨噬细胞吞噬率和吞噬指数与对照组相比有极显著提高(P<0.01),说明鹿胎及胎盘制剂对老年雄性大鼠具有提高免疫机能的作用。  相似文献

This study investigated possible immunotoxic effects of Senna occidentalis (So) seeds incorporated in broiler chicken rations at different concentrations (0.0%, 0.25%, 0.50% and 0.75%), for 28 or 42 days. We evaluated innate immune function (macrophage activities of spreading, phagocytosis, peroxide and nitric oxide production) and acquired immune function (humoral and cellular immune responses), as well as lymphoid organ weights and pathology. There was enhanced macrophage activity, as hydrogen peroxide production increased (P < 0.05) in cells of birds given 0.75%So, but there were no other pro-inflammatory effects. Birds receiving 0.75% of So in ration for 42 days gained less weight (P < 0.01), and showed a decrease in relative weight of the bursa of Fabricius (P < 0.05) and spleen (P < 0.01). In addition, morphological changes were also noted in these lymphoid organs, with depletion of lymphoid cells on the spleen and bursa of Fabricius, resulting in lower relative weight of both lymphoid organs. No impairment of humoral immune response against Newcastle disease and in cellular immune response after a phytohaemagglutinin challenge was found. It is probable that mitochondrial damage and related apoptosis may be responsible for the enhanced peroxide production and the reduced relative weight of the bursa of Fabricius and spleen.  相似文献
